Most statistical methods for censored survival data assume there is no dependence between the lifetime and censoring mechanisms, an assumption which is often doubtful in practice. In this paper we study a parametric model which allows for dependence in terms of a parameter delta and a bias function B(t, theta). We propose a sensitivity analysis on the estimate of the parameter of interest for small values of delta. This parameter measures the dependence between the lifetime and the censoring mechanisms. Its size can be interpreted in terms of a correlation coefficient between the two mechanisms. A medical example suggests that even a small degree of dependence between the failure and censoring processes can have a noticeable effect on the analysis.  相似文献

Current status data arise due to only one feasible examination such that the failure time of interest occurs before or after the examination time. If the examination time is intrinsically related to the failure time of interest, the examination time is referred to as an informative censoring time. Such data may occur in many fields, for example, epidemiological surveys and animal carcinogenicity experiments. To avoid severely misleading inferences resulted from ignoring informative censoring, we propose a class of semiparametric transformation models with log‐normal frailty for current status data with informative censoring. A shared frailty is used to account for the correlation between the failure time and censoring time. The expectation‐maximization (EM) algorithm combining a sieve method for approximating an infinite‐dimensional parameter is employed to estimate all parameters. To investigate finite sample properties of the proposed method, simulation studies are conducted, and a data set from a rodent tumorigenicity experiment is analyzed for illustrative purposes.  相似文献

To account for the correlation between failure and censoring, we propose a new frailty model for clustered data. In this model, the risk to be censored is affected by the risk of failure. This model allows flexibility in the direction and degree of dependence between failure and censoring. It includes the traditional frailty model as a special case. It allows censoring by some causes to be analyzed as informative while treating censoring by other causes as noninformative. It can also analyze data for competing risks. To fit the model, the EM algorithm is used with Markov chain Monte Carlo simulations in the E-steps. Simulation studies and analysis of data for kidney disease patients are provided. Consequences of incorrectly assuming noninformative censoring are investigated.  相似文献

When modeling survival data, it is common to assume that the (log-transformed) survival time (T) is conditionally independent of the (log-transformed) censoring time (C) given a set of covariates. There are numerous situations in which this assumption is not realistic, and a number of correction procedures have been developed for different models. However, in most cases, either some prior knowledge about the association between T and C is required, or some auxiliary information or data is/are supposed to be available. When this is not the case, the application of many existing methods turns out to be limited. The goal of this paper is to overcome this problem by developing a flexible parametric model, that is a type of transformed linear model. We show that the association between T and C is identifiable in this model. The performance of the proposed method is investigated both in an asymptotic way and through finite sample simulations. We also develop a formal goodness-of-fit test approach to assess the quality of the fitted model. Finally, the approach is applied to data coming from a study on liver transplants.  相似文献

Multivariate recurrent event data are usually encountered in many clinical and longitudinal studies in which each study subject may experience multiple recurrent events. For the analysis of such data, most existing approaches have been proposed under the assumption that the censoring times are noninformative, which may not be true especially when the observation of recurrent events is terminated by a failure event. In this article, we consider regression analysis of multivariate recurrent event data with both time‐dependent and time‐independent covariates where the censoring times and the recurrent event process are allowed to be correlated via a frailty. The proposed joint model is flexible where both the distributions of censoring and frailty variables are left unspecified. We propose a pairwise pseudolikelihood approach and an estimating equation‐based approach for estimating coefficients of time‐dependent and time‐independent covariates, respectively. The large sample properties of the proposed estimates are established, while the finite‐sample properties are demonstrated by simulation studies. The proposed methods are applied to the analysis of a set of bivariate recurrent event data from a study of platelet transfusion reactions.  相似文献

Right- and interval-censored data are common special cases of coarsened data (Heitjan and Rubin, 1991, Annals of Statistics19, 2244-2253). As with missing data, standard statistical methods that ignore the random nature of the coarsening mechanism may lead to incorrect inferences. We extend a simple sensitivity analysis tool, the index of local sensitivity to nonignorability (Troxel, Ma, and Heitjan, 2004, Statistica Sinica14, 1221-1237), to the evaluation of nonignorability of the coarsening process in the general coarse-data model. By converting this index into a simple graphical display one can easily assess the sensitivity of key inferences to nonignorable coarsening. We illustrate the validity of the method with a simulated example, and apply it to right-censored data from an observational study of cardiac transplantation and to interval-censored data on time to detectable viral load from a clinical trial in HIV disease.  相似文献

This article describes a general class of factor analytic models for the analysis of clustered multivariate data in the presence of informative missingness. We assume that there are distinct sets of cluster-level latent variables related to the primary outcomes and to the censoring process, and we account for dependency between these latent variables through a hierarchical model. A linear model is used to relate covariates and latent variables to the primary outcomes for each subunit. A generalized linear model accounts for covariate and latent variable effects on the probability of censoring for subunits within each cluster. The model accounts for correlation within clusters and within subunits through a flexible factor analytic framework that allows multiple latent variables and covariate effects on the latent variables. The structure of the model facilitates implementation of Markov chain Monte Carlo methods for posterior estimation. Data from a spermatotoxicity study are analyzed to illustrate the proposed approach.  相似文献

In cancer drug development, demonstrating activity in xenograft models, where mice are grafted with human cancer cells, is an important step in bringing a promising compound to humans. A key outcome variable is the tumor volume measured in a given period of time for groups of mice given different doses of a single or combination anticancer regimen. However, a mouse may die before the end of a study or may be sacrificed when its tumor volume quadruples, and its tumor may be suppressed for some time and then grow back. Thus, incomplete repeated measurements arise. The incompleteness or missingness is also caused by drastic tumor shrinkage (<0.01 cm3) or random truncation. Because of the small sample sizes in these models, asymptotic inferences are usually not appropriate. We propose two parametric test procedures based on the EM algorithm and the Bayesian method to compare treatment effects among different groups while accounting for informative censoring. A real xenograft study on a new antitumor agent, temozolomide, combined with irinotecan is analyzed using the proposed methods.  相似文献

AIDS Clinical Trial Group (ACTG) randomized trial 021 compared the effect of bactrim versus aerosolized pentamidine (AP) as prophylaxis therapy for pneumocystis pneumonia (PCP) in AIDS patients. Although patients randomized to the bactrim arm experienced a significant delay in time to PCP, the survival experience in the two arms was not significantly different (p = .32). In this paper, we present evidence that bactrim therapy improves survival but that the standard intent-to-treat comparison failed to detect this survival advantage because a large fraction of the subjects either crossed over to the other therapy or stopped therapy altogether. We obtain our evidence of a beneficial bactrim effect on survival by artificially regarding the subjects as dependently censored at the first time the subject either stops or switches therapy; we then analyze the data with the inverse probability of censoring weighted Kaplan-Meier and Cox partial likelihood estimators of Robins (1993, Proceedings of the Biopharmaceutical Section, American Statistical Association, pp. 24-33) that adjust for dependent censoring by utilizing data collected on time-dependent prognostic factors.  相似文献

We discuss how to apply the conditional informative missing model of Wu and Bailey (1989, Biometrics 45, 939-955) to the setting where the probability of missing a visit depends on the random effects of the primary response in a time-dependent fashion. This includes the case where the probability of missing a visit depends on the true value of the primary response. Summary measures for missingness that are weighted sums of the indicators of missed visits are derived for these situations. These summary measures are then incorporated as covariates in a random effects model for the primary response. This approach is illustrated by analyzing data collected from a trial of heroin addicts where missed visits are informative about drug test results. Simulations of realistic experiments indicate that these time-dependent summary measures also work well under a variety of informative censoring models. These summary measures can achieve large reductions in estimation bias and mean squared errors relative to those obtained by using other summary measures.  相似文献

SUMMARY: In clinical studies, when censoring is caused by competing risks or patient withdrawal, there is always a concern about the validity of treatment effect estimates that are obtained under the assumption of independent censoring. Because dependent censoring is nonidentifiable without additional information, the best we can do is a sensitivity analysis to assess the changes of parameter estimates under different assumptions about the association between failure and censoring. This analysis is especially useful when knowledge about such association is available through literature review or expert opinions. In a regression analysis setting, the consequences of falsely assuming independent censoring on parameter estimates are not clear. Neither the direction nor the magnitude of the potential bias can be easily predicted. We provide an approach to do sensitivity analysis for the widely used Cox proportional hazards models. The joint distribution of the failure and censoring times is assumed to be a function of their marginal distributions. This function is called a copula. Under this assumption, we propose an iteration algorithm to estimate the regression parameters and marginal survival functions. Simulation studies show that this algorithm works well. We apply the proposed sensitivity analysis approach to the data from an AIDS clinical trial in which 27% of the patients withdrew due to toxicity or at the request of the patient or investigator.  相似文献

Multivariate current status data, consist of indicators of whether each of several events occur by the time of a single examination. Our interest focuses on inferences about the joint distribution of the event times. Conventional methods for analysis of multiple event-time data cannot be used because all of the event times are censored and censoring may be informative. Within a given subject, we account for correlated event times through a subject-specific latent variable, conditional upon which the various events are assumed to occur independently. We also assume that each event contributes independently to the hazard of censoring. Nonparametric step functions are used to characterize the baseline distributions of the different event times and of the examination times. Covariate and subject-specific effects are incorporated through generalized linear models. A Markov chain Monte Carlo algorithm is described for estimation of the posterior distributions of the unknowns. The methods are illustrated through application to multiple tumor site data from an animal carcinogenicity study.  相似文献

Summary Increasingly, estimations of health care costs are used to evaluate competing treatments or to assess the expected expenditures associated with certain diseases. In health policy and economics, the primary focus of these estimations has been on the mean cost, because the total cost can be derived directly from the mean cost, and because information about total resources utilized is highly relevant for policymakers. Yet, the median cost also could be important, both as an intuitive measure of central tendency in cost distribution and as a subject of interest to payers and consumers. In many prospective studies, cost data collection is sometimes incomplete for some subjects due to right censoring, which typically is caused by loss to follow-up or by limited study duration. Censoring poses a unique challenge for cost data analysis because of so-called induced informative censoring, in that traditional methods suited for survival data generally are invalid in censored cost estimation. In this article, we propose methods for estimating the median cost and its confidence interval (CI) when data are subject to right censoring. We also consider the estimation of the ratio and difference of two median costs and their CIs. These methods can be extended to the estimation of other quantiles and other informatively censored data. We conduct simulation and real data analysis in order to examine the performance of the proposed methods.  相似文献

Longitudinal clinical trials often collect long sequences of binary data. Our application is a recent clinical trial in opiate addicts that examined the effect of a new treatment on repeated binary urine tests to assess opiate use over an extended follow-up. The dataset had two sources of missingness: dropout and intermittent missing observations. The primary endpoint of the study was comparing the marginal probability of a positive urine test over follow-up across treatment arms. We present a latent autoregressive model for longitudinal binary data subject to informative missingness. In this model, a Gaussian autoregressive process is shared between the binary response and missing-data processes, thereby inducing informative missingness. Our approach extends the work of others who have developed models that link the various processes through a shared random effect but do not allow for autocorrelation. We discuss parameter estimation using Monte Carlo EM and demonstrate through simulations that incorporating within-subject autocorrelation through a latent autoregressive process can be very important when longitudinal binary data is subject to informative missingness. We illustrate our new methodology using the opiate clinical trial data.  相似文献
