The surface area of pericardial sacs of the semiterrestrialcrab, Ocypode cordimana, is greater than that of pericardialsacs of the intertidal crabs, Ocypode macrocera and Ocypodeplatytarsis. Responses of the pericardial sacs with relationto conservation of water and molting vary with the species.In the semiterrestrial crab, Ocypode cordimana, the pericardialsacs store water during proecdysis which is used to stretchthe new cuticle after ecdysis. In contrast, the pericardialsacs of the intertidal crabs, Ocypode macrocera and Ocypodeplatytarsis, do not store water during proecdysis. But the sacsof these crabs swell for a brief period during ecdysis, apparentlyto accommodate excess fluid entering the body while the legsare being withdrawn from the old exoskeleton. Although the significanceremains unknown, deposits of uric acid are found in the pericardialsacs of Ocypode cordimana but not in the pericardial sacs ofOcypode macrocera and Ocypode platytarsis. Exposure of specimensof Ocypode cordimana to dry sand reduces the swelling of pericardialsacs, while proecdysial growth of the limbs remains unaffected.The pericardial sacs of Ocypode cordimana swell at a rapid rateif the crabs are maintained in darkness rather than in light.Implications of the present findings are discussed with relationto the habitat of the crabs.  相似文献

Results of histochemical tests for cytochrome oxidase activityin four species of Rauwolfia have been reported. The cells beneaththe terminal shoot meristem constitute the pith. Histochemicaltests showed intensified enzymatic activity in those cells ofthe pith which would differentiate as sclereid initials. Similarcytochrome oxidase activity also occurred in the sclereid initialsand the developing sclereids. The cytochrome oxidase activitywas associated with two types of particulate formation, thegranular and rod-shaped. The ground parenchymatous cells ofthe pith and the leaf-base showed very little enzymatic activityof cytochrome oxidase. The characteristic indophenol blue colorationdue to cytochrome oxidase activity did not appear in controlsections. Physiological changes correlated with morphogeneticexpression of some pith cells demonstrate that the physiologicalchanges occur before the initiation of sclereids in the morphologicallyhomogeneous parenchymatous cells of the pith. Intensified cytochromeoxidase activity was also recorded in the meristematic tissuesof shoot apex, procambium, axillary buds and the laticiferouscells of Rauwolfia.  相似文献

SYNOPSIS. Different species of larval trematodes that utilizethe same intermediate host characteristically exhibit distinctivepatterns of response to environmental fluctuations. Not onlyare the responses distinctive for each species, but also theycannot be correlated with that of the host. The physiologicalresponses of the molluscan host to physical changes in the environment,however, are modified significantly by the presence of a larvaltrematode infection. The thermal metabolic acclimation patternsof infected Nassarius obsoleta are quite distinct from thoseof non-infected N. obsoleta. Thex activity patterns during thermalacclimation of cytochrome c oxidase from digestive glands ofsnails infected with different species of larval trematodesindicate that each species of larval trematodes differentiallyinfluences the hosttissue, so that the cytochrome oxidase systemdoes not respond to temperature in the same way as does uninfectedhost tissue.  相似文献

Rice seeds were germinated for up to 5 days under water (submerged)and some for another day in air (air-adapted). Control seedswere germinated for 6 days throughout in air. Low-temperaturedifference spectra of shoot mitochondria were compared amongthese three types of seedlings. All cytochromes found in theaerobic seedlings were present in the submerged seedlings. However,there were some differences in the cytochromes b553 and c ofthese two types of seedlings. The cytochrome aa3 peak heightand cytochrome oxidase activity per mitochondrial protein increased1.6- and 2.8-fold, respectively, during air adaptation. Slightlyhigher concentrations of the b-type cytochromes than found inair-adapted mitochondria were already present in submerged mitochondria.The computed difference between the dithionite-reduced differencespectra of mitochondria from submerged seedlings before andafter air adaptation, showed that cytochromes aa3 and c hadincreased more than cytochrome b557 during air adaptation. Sulphite-cytochrome c reductase (sulphite: ferricytochrome coxidoreductase, EC 1.8.2.1 [EC] ) derived from Thiobacillus novelluswas purified by chromatography on a DEAE-cellulose column andby gel filtration with a Sephadex G-100 column. Although thereductase thus purified moved as a single band both in gel filtrationand in isoelectric focusing it was always split into two bandsby polyacrylamide gel electrophoresis; the one had the enzymaticactivity and showed absorption spectrum of cytochrome, whilethe other had no activity and was colourless, in contrast withthe results reported by Charles and Suzuki [(1966) Biochim.Biophys. Acta 128: 522]. The enzymatic properties of the purifiedreductase were almost the same as those of the enzyme obtainedby Charles and Suzuki. Cytochrome c-551 free of the reductase activity was obtained.Its molecular weight was determined to be 23,000 by polyacrylamidegel electrophoresis in the presence of sodium dodecyl sulphate.The cytochrome seemed to exist in the organism as a complexwith the reductase or a subunit of the enzyme. In the stateof the complex with the enzyme, the cytochrome was reduced veryquickly on addition of sulphite, while the cytochrome free ofthe reductase activity was hardly reduced by the enzyme withsulphite. Ditercalinium, the generic name for 7H-pyridocarbazole dimer,is a planar aromatic cation which exhibits strong inhibitoryeffects on the respiratory chain of plant mitochondria. By enzymaticassays and spectroscopic studies, the inhibition is shown tooccur at the level of the cytochrome c oxidase complex. It issuggested that the mechanism of inhibition could be due to thecomplexation of the aromatic cation with the diphosphatidylglycerolenvironment essential for cytochrome c oxidase activity ratherthan to a drug-enzyme direct interaction. 7H-pyridocar-bazolecan be used as an appropriate tool to study the role that negativelycharged acidic phospholipids, as diphosphatidylglycerol, playin the functioning of cytochrome c oxidase. Summary Structural and functional studies of the hemocyanin of the semi-terrestrial ghost crab,Ocypode quadrata, demonstrate a variety of differences in comparison to the hemocyanin of aquatic crabs. These differences may be related to the terrestrial habit of this crab. Unlike aquatic crabs, the major (56%) blood component is the hexamer; the remaining 44% is dodecamer. The hexamers and dodecamers are not in rapid equilibrium. Electrophoretic analysis of the subunit composition indicates three major components referred to as 1, 3, and 4, and one minor component referred to as component 2. These components, although electrophoretically distinct, are alike immunologically. Components 1 and 2 are essentially absent from purified hexamers, whereas they compose 1/3 of the subunits in dodecamers. These results suggest that they are involved in linking hexamers to form dodecamers, and that two, rather than one, subunits are involved in the bridge. Oxygen-binding measurements show a higher degree of cooperativity, and a much reduced allosteric effect ofl-lactate on the dialyzed hemocyanin as compared to the hemocyanin of aquatic crabs. Exercise rapidly, induces a large drop in hemolymph pH (0.5 units) and a corresponding increase in lactate concentrations (to 10 mM).  相似文献

Distribution of iron-containing oxidases in aging nodal rootsof rice and wheat was studied. Activities of cytochrome c oxidase(1.9.3.1 [EC] , cytochrome c : O2 oxidoreductase), catalase (1.11.1.6 [EC] ,H2O2: H2O2 oxidoreductase) and peroxidase (1.11.1.7 [EC] , donor:H2O2 oxidoreductase) in wheat roots were comparatively higherthan were those in rice roots at corresponding stages. Cytochromec oxidase in roots remained active throughout the lives of therice and wheat crops. In rice roots, catalase seemed to playa distinct role around the panicle formation stage. Decay ofcatalase activity took place earlier than did that of peroxidaseand cytochrome c oxidase activities. In wheat roots similarenzyme activity changes were not observed. Mitochondria prepared from young Arum spadix have enoughcytochrome c oxidase to account for the rate at which succinateis oxidized, but succinoxidase activity increases markedly asthe plants mature so that in old material cytochrome oxidaseactivity is only 10 per cent, of succinoxidase. 2. Disintegration of the mitochondria by vibration with ballotini,treatment with digitonin or incubation in the warm reveals anintra-mitochondrial cytochrome c oxidase probably active enoughto account for the fastest rates of succinate oxidation. 3. Succinic dehydrogenase activity is demonstrated and experimentswith p-chloromercuribenzoate indicate that it plays a part inthe oxidation of succinate. 4. Cyanide completely inhibits both external and internal cytochromeoxidase but even at the earliest stages it only reduces succinoxidaseby about 50 per cent. Purified chromatophore membranes and spheroplast membrane vesicleswere prepared from Rhodopseudomonas sphaeroides by the methodof Michels and Konings (1978). The cytochrome c oxidase activityof the spheroplast membrane vesicles was about 15-fold higheron a bacteriochlorophyll basis than that of the chromatophoremembranes. The cytochrome c oxidase activity of the chromatophoremembranes was greatly stimulated (about 10-fold) by an additionof Triton X-100 (0.1%), but there was less stimulated (about1.5-fold) in the case of spheroplasts. The pH optimum of theoxidase activities of the spheroplast membrane vesicles andof chromatophore membranes treated with 0.1% Triton X-100, andthe salt dependencies of the activity of both preparations werethe same. These results show that the membrane-bound b-type cytochromeoxidase of this bacterium accepts electrons from ferrocytochromec on the periplasmic side of the membrane (on the outer surfaceof the spheroplast membrane vesicles). Food habits and ecological rôle of the ghost crab. Ocypode quadrata ( Fabricius) on a North Carolina barrier beach have been investigated in field and laboratory studies. Despite previous reports that they are scavengers, the crabs spent little time in ureas where drifted material accumulated. Dead material accounted for less than 10% of the food in the field. The crabs did. however, give evidence of being facultative scavengers, readily consuming virtually any form of organic matter. Live prey, consisting almost exclusively of mole crabs, Emerita talpoida (Say), and coquina clams, Donax variabilis Say, made up more than 90% of the diet. Handling times indicate that about equal weights of E. talpoida and D. variabilis are consumed. Because of its higher caloric content E. talpoida provides ≈60% of the energy and D. variabilis ≈25%.The effect of O. quadrata on the prey species was assessed by comparing the estimated rates of its feeding (based on resting metabolism) with estimated production of E. talpoida and D. variabilis. Calculations indicate that ghost crabs consume most of the production of both species.Ghost crabs have essentially no terrestrial competitors or predators on the beaches concerned and the stability of this simple food web in such a physically unstable environment may be attributed to the flexible feeding behavior of the predators and their ability to endure long periods of starvation, and to the prey having high biotic potential and dispersal rates. In order to distinguish between the regulatory effects of oxygen tension and light intensity on cytochrome c oxidase protein and enzymatic activity cells of Rhodobacter capsulatus were shifted from phototrophic (anaerobic, light) growth to aerobic-light, aerobic-dark and to anaerobic-dark conditions, respectively. During shift-experiments the formation of oxidase protein and regulation of oxidase activity was followed by immunological and enzymatic means. The results support the idea, that the formation of oxidase protein is regulated by oxygen tension and light intensity changes, whereas the regulation of oxidase activity seems only to be correlated to the oxygen tension. A DNA sequence involved in the oxygen-dependent regulation of cytochrome oxidase could be identified in the regulation-deficient oxidase mutant H41 of R. capsulatus. In order to survive and to grow in the presence of a high salinity(550 mM NaCl) Synechocystis PCC6803 increases its energeticcapacity. The salt-induced increase of electron transport ratesinvolves both cytochrome c oxidase and photosystem I. In contrast,electron transport rates measured through complexes I plus IIIof the respiratory chain, or through the photosystem II pluscytochrome b6f complexes of the photosynthetic chain, do notshow appreciable changes. The time at which changes in electrontransport rates occur in the photosystem I and cytochrome coxidase complexes after the onset of salt stress indicates similaritiesin the adaptation of dark respiration and (cyclic) photosyntheticelectron flow. Given an increase of whole cell respiration andof PSI cyclic electron flow larger than the neosynthesis ofcytochrome aa3 and PSI reaction centers would predict, it appearsthat both adaptations require more than just synthesis of thesetwo complexes. Investigations were made of the properties of diaphorase, cytochromec reductases, cytochrome c oxidase, and other components ofelectron transfer system in various fractions of leaf homogenateof Begonia semperflorens.
  1. All the fractions tested showed the existence of cytochromec oxidase, succinic- and reduced diphosphopyridine nucleotide-cytochromec reductases, and diaphorase. Activities of these enzymes werefound to be associated mainly with the particulate fractions.The particulate fractions showed, in particular, a capacityof reducing oxidized cytochrome c with fumarate, malate, -ketoglutarate,ß-hydroxy-butyrate, and citrate.
  2. Optimum pH foroxidation of cytochrome c by the particulatefractions was foundto be 5.5, while that for reduction was7.2.
  3. The activityof cytochrome c reductase was partially suppressedby malonate.Partial inhibition of cytochrome c oxidase wascaused by azideand cyanide, the inhibitory effects observedbeing strongerwith particulate fractions than with solublefractions.

The ghost crab Ocypode quadrata plays an important role in energy transfer between trophic levels, and has been widely used in evaluations of impacted environments. In order to provide data on the biology of this potential bioindicator species, the population structure and vertical distribution of individuals were studied on two beaches in southeastern Brazil. Each beach was divided into quadrants of 1000 m2 with boundaries of upper, middle and lower levels in relation to the waterline. Collected monthly by active searching through one year, the specimens of O. quadrata were sexed, measured for carapace width, and returned to the beach. Of the total of 1904 specimens collected, the largest proportion (46.2%) were males, followed by 31.4% juveniles. The vertical distribution of the ghost crabs differed among age groups: males mostly occupied the middle and upper levels; adult females, ovigerous or not, were more abundant in the lower level; and juveniles were evenly distributed in all levels, with a slight tendency toward the middle. The sex ratio favored males in a few months of the year and in the larger size classes. The abundance of O. quadrata is limited by low temperatures, and its spatial and temporal distribution is controlled by food availability and ease of reproduction. Knowledge of the biology of these crabs is essential in order to use them as a bioindicator species; the vertical distribution patterns may reflect changes in the beach hydrodynamics or other environmental factors.  相似文献

The electron transfer activity of an aa3-type two-subunit cytochromec oxidase of Nitrobacter agilis was inhibited by DCCD. Althoughthe activity of the purified cytochrome c oxidase dissolvedin 1% Triton X- 100 was not affected by DCCD even at a ratioof 1,000 mol DCCD per mol cytochrome aa3, the activity of theenzyme dissolved in 0.02% Tween 20 or 0.02% Triton X-100 wasinhibited by 60% or more at a ratio of 1,000 mol DCCD per molcytochrome aa3. The results of SDS polyacrylamide gel electrophoresisof the enzyme incubated with DCCD suggested that subunit IImight be a binding site for DCCD. Cytochrome b561 from Rhodopseudomonas sphaeroides had cytochromec (c2) oxidase activity and a pH optimum at 6.0 for this activity.The activity was affected by the ionic strength of the reactionmixture. The apparent Km and maximal velocity (Vmax) valuesin the absence of addea salts were 14 µM and 120 nmoloxidized per min per mg protein for horse heart cytochrome c.Reduced horse heart cytochrome c was reoxidized in first-orderkinetics by this cytochrome b561. The specific activity was0.7 s–1 per mg protein at 20°C at the concentrationof 30 µMM cytochrome c. Activity was inhibited by KCN and NaN3, but not by antimycin.The addition of a low concentration of KCN to the cytochromeb561 produced a change in the absorption spectrum, evidencethat KCN interacts with the heme moiety of cytochrome b561.Results of this and preceeding studies show that the cytochromeoxidase (cytochrome "o") described earlier (Sasaki et al. 1970)is cytochrome b561. Respiratory activities, cytochrome composition and cytochromec oxidase (EC 1.9.3.1 [EC] ) subunit II (COXII) levels of isolatedmitochondria in one euplasmic and four alloplasmic lines ofwheat (Triticum aestivum), having cytoplasms of Aegilops columnaris,Ae.crassa (4x),Ae. mutica and Ae. triuncialis, which show variousgrowth abnormalities, were studied. Cytoplasm-specific variationon the respiratory activities has been revealed among the cytoplasmsof five Triticum and Aegilops species. The cyanide-resistantrespiratory pathway does not seem to contribute to the variationin activity. The low level of state 3 and cytochrome oxidaseactivities were evident in lines having Ae. columnaris and Ae.triuncialis cytoplasms, which also show growth inhibition. Thelow cytochrome oxidase activity in both cytoplasms is consistentwith the low amount of cytochrome aa3 and the severe reductionin COXII levels. The relatively low amount of cytochrome aa3in Ae. mutica is also consistent with the reduced level of theCOXII. On the other hand, relatively low amounts of cytochromeb (b-557) are found in the Ae. crassa cytoplasmic line. Theseresults suggest that the growth abnormalities found in the alloplasmicwheat lines are related to the variation in respiratory activitiesand cytochrome composition. Nitrobacter agilis cytochrome c-550 was purified to an electrophoreticallyhomogeneous state, and some of its properties were determined.The cytochrome showed an absorption peak at 410 nm in the oxidizedform, and peaks at 416, 521 and 550 nm in the reduced form.Its isoelectric point was 8.1 at 5?C. Analysis of the aminoacid composition showed that the cytochrome molecule was composedof 108 amino acid residues, 16 of which were lysine residues. The cytochrome reacted rapidly with N. agilis cytochrome c oxidaseand yeast cytochrome c peroxidase and more slowly with Pseudomonasaeruginosa nitrite reductase and bovine cytochrome c oxidase.The reactivities with these redox enzymes suggested that thecytochrome might be an evolutionary stage between bacterialand eukaryotic cytochromes c. The primary structure of the cytochrome from the N-terminusto the 85th residue was determined. Nitric oxide (NO) is an intercellular signaling molecule; among its many and varied roles are the control of blood flow and blood pressure via activation of the heme enzyme, soluble guanylate cyclase. A growing body of evidence suggests that an additional target for NO is the mitochondrial oxygen-consuming heme/copper enzyme, cytochrome c oxidase. This review describes the molecular mechanism of this interaction and the consequences for its likely physiological role. The oxygen reactive site in cytochrome oxidase contains both heme iron (a3) and copper (CuB) centers. NO inhibits cytochrome oxidase in both an oxygen-competitive (at heme a3) and oxygen-independent (at CuB) manner. Before inhibition of oxygen consumption, changes can be observed in enzyme and substrate (cytochrome c) redox state. Physiological consequences can be mediated either by direct "metabolic" effects on oxygen consumption or via indirect "signaling" effects via mitochondrial redox state changes and free radical production. The detailed kinetics suggest, but do not prove, that cytochrome oxidase can be a target for NO even under circumstances when guanylate cyclase, its primary high affinity target, is not fully activated. In vivo organ and whole body measures of NO synthase inhibition suggest a possible role for NO inhibition of cytochrome oxidase. However, a detailed mapping of NO and oxygen levels, combined with direct measures of cytochrome oxidase/NO binding, in physiology is still awaited. mitochondria; cytochrome oxidase  相似文献
