Ca2+ (0.1-1.0 millimolar) accelerated dark-induced stomatal closure and reduced stomatal apertures in the light in epidermal peels of Commelina communis L. In contrast, ethyleneglycol-bis-(β-aminoethyl ether) N,N′tetraacetic acid (EGTA) (2 millimolar), a Ca2+ chelator, prevented closure in the dark and accelerated opening in the light. EGTA did not promote significant opening in the dark. It is therefore concluded that EGTA does not increase ion uptake into guard cells, but rather prevents ion efflux. Addition of EGTA to incubating solutions with 10 millimolar KCl resulted in steady state apertures of 15.6 micrometers, whereas in the absence of EGTA similar apertures required 55 millimolar KCl and 150 millimolar KCl was needed in the presence of 1 millimolar CaCl2. The results demonstrate the importance of Ca2+ in the regulation of stomatal closure and point to a role of Ca2+ in the regulation of K+ efflux from stomatal guard cells.  相似文献

The effects of anti-calmodulin (CaM) serum, the CaM antagonist W7-agarose, the Ca2+ chelator ethyleneglycol-bis-(β-aminoethyl)-N,N,N′,N′-tetraacetic acid (EGTA) and exogenous pure CaM on pollen germination and tube growth of Hippeastrum rutilum Herb were studied. Pollen germination and tube growth were inhibited or completely stopped by anti-CaM serum in a dose-dependent manner, while the same amount of preimmune serum had no effect on either process. Pollen germination and tube growth were also inhibited or completely stopped by the CaM antagonist W7-agarose and the Ca2+ chelator EGTA. The addition of exogenous pure CaM enhanced pollen germination and tube growth, whereas the same amount of bovine serum albumin had no effect. The inhibitory effects caused by anti-CaM serum, W7-agarose and EGTA-washing could be reversed completely by the addition of exogenous pure CaM. These results indicate that extracellular CaM initiates pollen germination and tube growth, whereas exogenous CaM enhances the above processes, and may provide a novel view for understanding the control of pollen germination and tube growth. The Ca2+ content of Globodera rostochiensis egg-shells was investigated by X-ray microanalysis. When intact eggs were treated with 5 mM sodium 1, 2-di (2-aminoethoxy) ethane-N,N,N',N',-tetra-acetate (EGTA) only part of the egg-shell Ca2+ was removed. Similarly treated opened egg-shells lost almost all their Ca2+. We think that Ca2+ of intact egg-shells which is accessible to EGTA is in the outer part of the shell and that the inaccessible Ca2+ is in the inner lipoprotein layer. Much Ca2+ was removed from opened egg-shells by the hatching agents Zn2+, La3+ and decationised potato-root exudate, but none by dilute acetic acid or Mg2+. Hatching agents, by binding to or replacing Ca2+, may change lipoprotein membrane structure. Eggshells treated with potato-root exudate contained about 3 times as much Ca2+ as untreated shells, because the treatment makes additional binding sites available. Our results suggest that three types of Ca2+ -binding sites occur in the egg-shell.  相似文献

We investigated the interaction of Ca2+ and auxin on root elongation in seedlings of Zea mays L. The seedlings were raised either in the presence of Ca2+ (high calcium; HC = imbibed and raised in 10 millimolar CaCl2), in the absence of additional Ca2+ (intermediate calcium; IC = imbibed and raised in distilled H2O, calcium supply from seed only), or without additional Ca2+ and subsequently depleting them of Ca2+ (low calcium; LC = imbibed and raised in distilled H2O and subsequently treated with 1 millimolar ethyleneglycol-bis-[β-aminoethylether]-N,N,N′,N′ -tetraacetic acid [EGTA]). Exposure of roots of either HC or IC seedlings to auxin concentrations from 0.1 to 10 micromolar resulted in strong inhibition of elongation. In roots of LC seedlings, on the other hand, auxin concentrations as high as 10 micromolar caused only slight inhibition of elongation. Adding 0.5 millimolar Ca2+ to LC roots in the presence of IAA allowed normal expression of the inhibitory action of the hormone. Inhibition of elongation in IC roots by indoleacetic acid was reversible upon treatment of the roots with 1 millimolar EGTA. The inhibitory action of auxin could then be re-established by supplying 0.5 millimolar Ca2+. The data indicate that Ca2+ may be necessary to the growth-regulating action of auxin. The significance of this finding is discussed with respect to the potential role of Ca2+ as a second messenger of auxin action and the relevance of this model to recent evidence for gravi-induced redistribution of Ca2+ and its role in establishing gravitropic curvature.  相似文献

The decline in ethylene production in apple (Pyrus malus L. cv. Golden Delicious) tissue slices during 24 hours incubation in 600 millimolar sorbitol and 10 millimolar 2-(N-morpholino)ethanesulfonic acid buffer (pH 6.0) is recognized as a senescent phenomenon. The inclusion of very high concentrations (100 millimolar) of Ca2+, Mg2+, or Ca2+ plus Mg2+ severely inhibited ethylene production during the first 6 hours of incubation. However, after 6 hours and up to 24 hours the ethylene-forming system was stablized. These high concentrations of Ca2+, Mg2+, or Ca2+ plus Mg2+ virtually eliminated lipid peroxidation and protein leakage from these slices. Also conversion of 1-aminocyclopropane-1-carboxylic-1-acid to ethylene and the influence of indoleacetic acid on ethylene production was stabilized after 24 hours of incubation by these high concentrations of Ca2+, Mg2+, and Ca2+ plus Mg2+. Addition of divalent ionophores severely inhibited ethylene production, but this inhibition was prevented by Ca2+ in concentrations greater than the ionophore. These data suggest that the loss of ethylene production by aging tissue slices results from degradation of membranes. They support previous work that indicates that the ethylene-forming system, perhaps the segment of the pathway from 1-aminocyclo-propane-1-carboxylic-1-acid to ethylene, resides in the plasma membrane.  相似文献

The objective of this work was to test whether Ca2+, a second messenger in stress response, is involved in ABA-induced antioxidant enzyme activities in Stylosanthes guianensis. Plants were sprayed with abscisic acid (ABA), calcium channel blocker, LaCl3, calcium chelator, ethylene glycol-bis(β-amino ethyl ether)-N,N,N′, N′-tetraacetid acid (EGTA), and ABA in combination with LaCl3 or EGTA. Their effects on superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities and chilling resistance were compared. The results showed that ABA decreased electrolyte leakage and lipid peroxidation but increased maximum photochemical efficiency measured as variable to maximum fluorescence ratio (Fv/Fm) under chilling stress. Treatment with LaCl3 or EGTA alone and in combination with ABA increased electrolyte leakage and lipid peroxidation, decreased Fv/Fm, suggesting that the block in Ca2+ signalling decreased chilling resistance of S. guianensis and the ABA-enhanced chilling resistance. ABA-induced SOD and APX activities were suppressed by LaCl3 or EGTA. The results suggested that Ca2+ is involved in the ABA-enhanced chilling resistance and the ABA-induced SOD and APX activities in S. guianensis.  相似文献

Glutamate synthase (EC 2.6.1.53) has been examined in developing endosperms and roots of maize. KCl is required for maximum activity in each tissue. The effect with KCl is seen with buffer strength of 25 to 100 millimolar in the assay. The optimum concentration for the enzyme from endosperm is 20 millimolar and for the enzyme from root tissue the saturating concentration is about 20 millimolar. In root material the enzyme is labile but activity can be restored if KCl is added to the assay. Divalent cations such as Mg2+ or Mn2+ also activate the enzyme to some extent.  相似文献

The purpose of the study was to determine inhibitory effect of calcium chelator; ethylene glycol-bis(2-aminoethylether)-N,N,N′,N′-tetraacetic acid (EGTA) on flowering of a short-day (SD) plant Pharbitis nil. It was found that 20 mM solution of EGTA applied on cotyledons of 5-d-old P. nil seedlings four hours before the start of 16-h-long induction night decreased the flowering response by 55% compared to the control plants not treated with this Ca2+ chelator. It also caused a very significant decrease of photosynthesis rate, transpiration rate and stomatal conductance both in light and darkness conditions. The results of this study confirm earlier hypothesis suggesting the effect of Ca2+ and its modulators on P. nil flowering is due to their influence on the stomata.  相似文献

The divalent cation requirements of NOS activity in bovine retina homogenate supernatant were investigated. Supernatants were assayed under standard conditions (in mM: EDTA 0.45, Ca2+ 0.25, Mg2+ 4.0). In order to investigate the enzyme's dependence on divalent cations, the tissue homogenate was depleted of di- and trivalent cations by passing it over a cation-exchange column (Chelex 100). Surprisingly, NOS activity was 50-100% higher in this preparation. However, addition of either EDTA (33 M) or EGTA (1 mM) almost fully inhibited NOS activity, suggesting a requirement for residual divalent metal cation(s). Phenanthroline or iminodiacetic acid at low concentrations had little effect on activity, suggesting no requirement for Fe2+, Zn2+ or Cu2+. Ca2+ had a moderate stimulatory effect, with an optimum activity around 0.01 mM. Mg2+ or Mn2+ had little effect at concentrations < 0.25 mM. However, in the presence of EDTA, Mn2+ or Ca2+ markedly stimulated NOS activity with the optimum at 0.1 mM. At high concentrations (> 0.1-0.2 mM), all divalent cations tested (Ba2+, Zn2+, Co2+, Mn2+, Mg2+, Ca2+), as well as La3+, dose-dependently inhibited NOS activity. We propose that retinal NOS requires low concentrations of naturally occurring divalent metal ions, most probably Ca2+, for optimal activity and is inhibited by high di- and trivalent metal concentrations, probably by competition with Ca2+.  相似文献

The adsorption of 5′-AMP onto precipitated calcium phosphate (CaPi) requires the presence of soluble calcium and this dependence exhibits a Michaelian-like behavior. This result suggests that the formation of a complex between 5′-AMP and free Ca2+ (CaAMP) is a prelude to the adsorption of the nucleotide in the solid matrix. At concentrations one order of magnitude higher, Mn2+ and Mg2+ can substitute for soluble Ca2+ in the adsorption of 5′-AMP onto solid CaPi. However, when added simultaneously with 5′-AMP to a heterogeneous mixture that contains CaPi and soluble Ca2+, Mn2+ and Mg2+ inhibit the adsorption of 5′-AMP in a concentration-dependent manner. This suggests the formation of complexes that are much less effective for 5′-AMP adsorption than the CaAMP complex. On the other hand, Mn2+ and Mg2+ cannot promote desorption of the nucleotide attached to the precipitate in the presence of soluble Ca2+ if they are added after adsorption has attained equilibrium. Although desorption of 5′-AMP can be obtained by a sequential dilution of the soluble phase with buffer and no nucleotide in a process that obeys a Langmuir equation, the lack of effect of Mn2+ or Mg2+ when adsorption has attained its maximal value suggests strong interactions between the CaAMP complex and the solid matrix when adsorption equilibrium is reached. The divalent cations present in the matrix also participate with different selectivity in the attachment of the CaAMP complex, indicating that a cation-exchange mechanism could have acted in the modulation of adsorptive/desorptive processes involving biomonomers and phosphate surfaces in primitive aqueous environments. Tripositive-pyrophosphate [M(III)-PPi] complexes were used to investigate the role of free divalent cations on the membrane-bound pyrophosphatase. Divalent cations remain free and the M(III)-PPi complexes were employed as substrates. Formation of a La-PPi complex was studied by fluorescence, and the fact that Zn2+ and Mg2+ remain free in the solution was validated. Hydrolysis of La-PPi is stimulated by the presence of fixed concentrations of free Mg2+ or Zn2+ and this stimulation depends on the concentration of the cations when the La-PPi complex is fixed. The divalent cation stimulation order is Zn2+ > Co2+ > Mg2+ > Mn2+ > Ca2+ (at 0.5 mm of free cation). With different M(III)-PPi complexes, Zn2+ produces the same K m, for all the complexes and Mg2+ stimulates with a different K m. The results suggest that both Mg2+ and Zn2+ activate the membrane-bound pyrophosphatase but through different mechanisms.  相似文献

Experiments examined the metabolic basis of Ca2+-induced conidiation during the 12-h period following the addition of Ca2+ to 40-h vegetative cultures ofPenicillium notatum. Vegetative mycelium had enzymic capacity for three routes of glucose catabolism viz. the Embden-Meyerhof-Parnas (EMP), pentose phosphate (PP) and the Entner-Doudoroff (ED) sequences. Inhibitors of EMP enzymes restricted vegetative growth more than that associated with conidiation whilst arsenate augmented the limited capacity of lower levels of Ca2+ to promote conidiation. Arsenite (5.6 mmol · 1–1) partially blocked the metabolism of pyruvate and caused its accumulation, which was also promoted by Ca2+ alone. Arsenite did not induce conidiation in vegetative cultures but when combined with Ca2+ it enhanced conidiation. Radiorespirometry and the analysis of accumulated pyruvate, promoted by arsenite, indicated that approximately 54% of carbon was catabolized via combined EMP/ED routes and 46% by the PP pathway and subsequently via a weakly functional TCA cycle. Calcium-induced cultures swung to a primarily ED (25%) and PP (75%) based catabolism with low substrate level phosphorylation, including a facility for a non-phosphorylative ED route, and further diminished oxidative TCA capacity. Pyruvate accumulation in Ca2+-induced cultures coincided with the decline in activity of pyruvate dehydrogenase and a reduced capacity for gluconeogenesis, with other enzymes of pyruvate metabolism showing altered activities. These changes in enzyme activities, pyruvate accumulation and its subsequent metabolism were related to growth rate and the developmental cycle, and are discussed in conjunction with the regulatory role of calcium.  相似文献

Summary The toxicity of chromium and tin on growth, photosynthetic carbon-fixation, oxygen evolution, heterocyst differentiation and nitrogenase activity ofAnabaena doliolum and its interaction with bivalent cations has been studied. Some interacting cations, viz. Ca2+, Mg2+ and Mn2+, substantially antagonised the toxic effects of chromium and tin with reference to growth, heterocyst differentiation and nitrogenase activity in the following hierarchal sequence: Ca2+ > Mg2+ > Mn2+. However, the sequence of hierarchy was Mg2+ > Ca2+ > Mn2+ for carbon fixation and Mn2+ > Mg2+ > Ca2+ for photosynthetic oxygen evolution. Synergistically inhibitory patterns were noticed for all the parameters, viz. growth,14CO2 uptake, oxygen evolution, heterocyst differentiation and nitrogenase activity ofA. doliolum when Ni2+, Co2+ and Zn2+ were combined with the test metals in the growth medium. These cations followed the following sequence of synergistic inhibition: Ni2+ > Co2+ > Zn2+. Among all the interacting cations, Ca2+, Mg2+ and Mn2+ exhibited antagonistic effects which relieved the test cyanobacterium from metal toxicity. In contrast to this, Ni2+, CO2+ and Zn2+ showed synergistic inhibition which potentiating the toxicity of test metals in the N2-fixing cyanobacteriumA. doliolum. It is evident from the present study that bivalent cations, viz. Ca2+, Mg2+, Mn2+, Ni2+, Co2+ and Zn2+, may appreciably regulate the toxicity of heavy metals in N2-fixing cyanobacteria if present in aquatic media.  相似文献

The presence of calcium is essential for chloroplast movement induced by blue light in Lemna trisulca L. The regulatory role of calcium was confirmed by the inhibition of chloroplast movement by cytochalasin B and trifluoperazine. The calcium concentration in tissues was modified by ethylene glycol-bis(2-aminoethylether)-N,N,N, N-tetraacetic acid (EGTA), the calcium ionophore A23187 and La3+. Only a long period of incubation (12h) in EGTA or La3+ caused distrubances in chloroplast movement. This indicates that calcium influx is not essential for chloroplast movement. Those conditions that dramatically changed the internal calcium concentration, either applications of calcium ionophore A23187 and EGTA, or ionophore and La3+, markedly decreased the amplitude of response to blue-light pulses. This demonstrates that disturbances of chloroplast movement are observable only when internal stores of calcium are affected by Ca2+-antagonists. We suggest that the calcium involved in blue-light-induced chloroplast movement is derived from intracellular stores. After acid-treatment of spinach (Spinacia oleracea) chloroplasts, various partial electron transport reactions are inactivated from 25 to 75%. Divalent cations in concentrations from 10 to 50 millimolar can partially restore electron transport rates. Two cation-specific sites have been found in photosystem II: one on the 3-(3,4-dichlorophenyl)-1, 1-dimethylurea-insensitive silicomolybdate pathway, which responds better to restoration by Mg2+ than by Ca2+ ions, the other on the forward pathway to photosystem I, located on the 2,5-dimethylbenzoquinone pathway. This site is selectively restored by Ca2+ ions. When protonated chloroplasts are treated with N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)aziridine, a carboxyl group modifying reagent, presumed to react with glutamic and aspartic acid residues of proteins, restoration of electron transport at the Ca2+-selective site on the 2,5-dimethylbenzoquinone pathway is impaired, while no difference in restoration is seen at the Mg2+ site on the 3-(3,4-dichlorophenyl)-1,1-dimethylurea-insensitive silicomolybdate pathway.

Trypsin treatment of chloroplasts modifies the light-harvesting pigment-protein complex, destroys the dibromothymoquinone-insensitive 2,5-dimethyl-benzoquinone reduction, but does not interfere with the partial restoration of activity of this pathway by Ca2+ ions, implying that the selective Ca2+ effect on photosystem II (selective Ca2+ site) is different from its effects as a divalent cation on the light-harvesting pigment-protein complex involved in the excitation energy distribution between the two photosystems.

Cytosolic ATP-phosphofructokinase (PFK) from spinach leaves (Spinacia oleracea L.) was inhibited by submillimolar concentrations of free Mg2+. The free Mg2+ concentration required for 50% inhibition of PFK activity was 0.22 millimolar. Inhibition by free Mg2+ was independent of the MgATP2− concentration. Inorganic phosphate (Pi) reduces the inhibition of PFK activity by Mg2+. Free ATP (ATP4−) also inhibits PFK activity. For free ATP the inhibition of PFK activity was dependent on the MgATP2− concentration. Fifty percent inhibition of PFK activity requires 1.2 and 3.7 millimolar free ATP at 0.1 and 0.5 millimolar MgATP2−, respectively. It was proposed that free ATP competes for the MgATP2− binding site, whereas free Mg2+ does not. Pi diminished the inhibitory effect of free ATP on PFK activity. Free ATP and Pi had substantial effects on the MgATP2− requirement of cytosolic PFK. For half-maximum saturation of PFK activity 3 and 76 micromolar MgATP2− was required at 0.007 and 0.8 millimolar free ATP in the absence of Pi. At 5 and 25 millimolar Pi, half-maximum saturation was achieved at 9 and 14 micromolar MgATP2−. PFK activity was inhibited by Ca2+. The inhibition by Ca2+ depends upon the total Mg2+ concentration. Fifty percent inhibition of PFK activity required 22 and 32 micromolar Ca2+ at 0.1 and 0.2 millimolar Mg2+, respectively. At physiological concentrations of about 0.5 millimolar free Mg2+, Ca2+ would have little effect on cytosolic PFK activity from spinach leaves. PFK is not absolutely specific for the nucleoside 5′-triphosphate substrate. Besides MgATP2−, MgUTP2−, MgCTP2−, and MgGTP2− could be used as a substrate. All four free nucleotides inhibit PFK activity. The physiological consequences of the regulatory properties of cytosolic PFK from spinach leaves will be discussed. A model will be introduced, in an attempt to describe the complex interaction of PFK with substrates and the effectors Mg2+ and Pi.  相似文献

Stomatal movements depend on both ion influx and efflux; attainment of steady state apertures reflects modulation of either or both processes. The role of Ca2+ in those two processes was investigated in isolated epidermal strips of Commelina communis, using the Ca2+ chelator EGTA to reduce apoplastic [Ca2+]. The results suggest that a certain concentration of Ca2+ is an absolute requirement for salt efflux and stomatal closure. EGTA (2 millimolar) increased KCl-dependent stomatal opening in darkness and completely inhibited the dark-induced closure of initially open stomata. Closure was inhibited even in a KCl-free medium. Thus, maintenance of stomata in the open state does not necessarily depend on continued K+ influx but on the inhibition of salt efflux. Opening in the dark was stimulated by IAA in a concentration-dependent manner, up to 15.4 micrometer without reaching saturation, while the response to EGTA leveled off at 9.2 micrometer. IAA did not inhibit stomatal closure to the extent it stimulated opening. The response to IAA is thus consistent with a primary stimulation of opening, while EGTA can be considered a specific inhibitor of stomatal closing since it inhibits closure to a much larger degree than it stimulates opening. CO2 causes concentration-dependent reduction in the steady state stomatal aperture. EGTA completely reversed CO2-induced closing of open stomata but only partially prevented the inhibition of opening.  相似文献

The (K+,Mg2+)-ATPase was partially purified from a plasma membrane fraction from corn roots (WF9 × Mol7) and stored in liquid N2 without loss of activity. Specific activity was increased 4-fold over that of the plasma membrane fraction. ATPase activity resembled that of the plasma membrane fraction with certain alterations in cation sensitivity. The enzyme required a divalent cation for activity (Co2+ > Mg2+ > Mn2+ > Zn2+ > Ca2+) when assayed at 3 millimolar ATP and 3 millimolar divalent cation at pH 6.3. When assayed in the presence of 3 millimolar Mg2+, the enzyme was further activated by monovalent cations (K+, NH4+, Rb+ Na+, Cs+, Li+). The pH optima were 6.5 and 6.3 in the absence and presence of 50 millimolar KCl, respectively. The enzyme showed simple Michaelis-Menten kinetics for the substrate ATP-Mg, with a Km of 1.3 millimolar in the absence and 0.7 millimolar in the presence of 50 millimolar KCl. Stimulation by K+ approached simple Michaelis-Menten kinetics, with a Km of approximately 4 millimolar KCl. ATPase activity was inhibited by sodium orthovanadate. Half-maximal inhibition was at 150 and 35 micromolar in the absence and presence of 50 millimolar KCl. The enzyme required the substrate ATP. The rate of hydrolysis of other substrates, except UDP, IDP, and GDP, was less than 20% of ATP hydrolysis. Nucleoside diphosphatase activity was less than 30% of ATPase activity, was not inhibited by vanadate, was not stimulated by K+, and preferred Mn2+ to Mg2+. Activity of membrane-bound phospholipase D in microsomes from bark tissues of black locust tree (Robina pseudoacacia L.) was demonstrated to be regulated by a competitive binding of divalent cations. Binding of Ca2+ at high concentrations (1 to 50 millimolar) modified the pH activity profile, shifting the optimum pH by 0.5 unit toward neutral and increasing the activity in the neutral pH. Mg2+, on the other hand, inhibited the reaction of membrane-bound phospholipase D without added Ca2+, and competitively inhibited the Ca2+ stimulation. The regulatory effects of those ions were dependent on pH. Reduction in pH resulted in a decrease in the apparent dissociation constant for Ca2+ and an increase in that for Mg2+. From Lineweaver-Burk double reciprocal plots of Ca2+ and the initial velocity, it was suggested that the binding of Ca2+ in the higher concentration resulted in nearly the same conformational change of enzyme as reduction in pH. Mg2+, on the other hand, counteracted those effects of Ca2+ and lower pH on the enzyme conformation in such a manner as to inactivate. The membrane-bound phospholipase D because more sensitive to Ca2+ and less sensitive to Mg2+ as the hardiness of the tissues decreased. This fact may indicate that some qualitative changes in membranes are involved in the hardiness changes and also in the susceptibility of phospholipid to degradation by phospholipase D in plant cells.  相似文献
