Drinking in Atlantic salmon (Salmo salar) juveniles was investigated in fresh water and following transfer to sea water. There was a significant effect of fish size on drinking, and smolts (20–30 g) imbibed about ten times less water than alevins of 0.2–0.3 g. Freshwater smolts drank at a rate of 0.15 ± 0.03 ml · kg−1 · h−1 and administration of doses of 10 or 20 mg · kg−1 of papaverine (stimulator of the renin- angiotensin system RAS) or [Asn1, Val5]-Angiotensin II (0.4 μmol · kg−1) resulted in significant increases in drinking, while administration of the angiotensin converting enzyme inhibitor, enalapril (50 mg · kg−1) had no effect on drinking. Transfer of Atlantic salmon smolts to 1/3, 2/3 and full strength sea water resulted in significant increases in drinking to 1.06 ± 0.12, 1.24 ± 0.0.16 and 3.89 ± 0.28 ml · kg−1 · h−1, respectively. In sea water, stimulation of the endogenous RAS by administration of papaverine (20 mg · kg−1) resulted in a 20% increase in drinking, while administration of enalapril to doses of 50 and 200 mg · kg−1 lowered drinking to 1.99 ± 0.48 and 0.32 ± 0.06 ml · kg−1 · h−1, respectively. All treatments were without effect on blood plasma levels of Na+ and Cl in fresh water, while in sea water smolts both stimulation and inhibition of drinking resulted in hemoconcentration of Na+ and Cl. Many populations of shortnose sturgeon, Acipenser brevirostrum, in the southeastern United States continue to suffer from poor juvenile recruitment. High summer water temperatures, which may be exacerbated by anthropogenic activities, are thought to affect recruitment by limiting available summer habitat. However, information regarding temperature thresholds of shortnose sturgeon is limited. In this study, the thermal maximum method and a heating rate of 0.1°C min−1 was used to determine critical and lethal thermal maxima for young-of-the-year (YOY) shortnose sturgeon acclimated to temperatures of 19.5 and 24.1°C. Fish used in the experiment were 0.6 to 35.0 g in weight and 64 to 140 days post hatch (dph) in age. Critical thermal maxima were 33.7°C (±0.3) and 35.1°C (±0.2) for fish acclimated to 19.5 and 24.1°C, respectively. Critical thermal maxima significantly increased with an increase in acclimation temperature (p < 0.0001). Lethal thermal maxima were 34.8°C (±0.1) and 36.1°C (±0.1) for fish acclimated to 19.5 and 24.1°C, respectively. Lethal thermal maxima were significantly affected by acclimation temperature, the log10 (fish weight), and the interaction between log10(fish weight) and acclimation temperature (p < 0.0001). Thermal maxima were used to estimate upper limits of safe temperature, thermal preferences, and optimal growth temperatures of YOY shortnose sturgeon. Upper limits of safe temperature were similar to previous temperature tolerance information and indicate that summer temperatures in southeastern rivers may be lethal to YOY shortnose sturgeon if suitable thermal refuge cannot be found.  相似文献

Relatively little is known about salinity acclimation in the primitive groups of fishes. To test whether physiological preparative changes occur and to investigate the mechanisms of salinity acclimation, anadromous green sturgeon, Acipenser medirostris (Chondrostei) of three different ages (100, 170, and 533 dph) were acclimated for 7 weeks to three different salinities (<3, 10, and 33 ppt). Gill, kidney, pyloric caeca, and spiral intestine tissues were assayed for Na+, K+-ATPase activity; and gills were analyzed for mitochondria-rich cell (MRC) size, abundance, localization and Na+, K+-ATPase content. Kidneys were analyzed for Na+, K+-ATPase localization and the gastro-intestinal tract (GIT) was assessed for changes in ion and base content. Na+, K+-ATPase activities increased in the gills and decreased in the kidneys with increasing salinity. Gill MRCs increased in size and decreased in relative abundance with fish size/age. Gill MRC Na+, K+-ATPase content (e.g., ion-pumping capacity) was proportional to MRC size, indicating greater abilities to regulate ions with size/age. Developmental/ontogenetic changes were seen in the rapid increases in gill MRC size and lamellar length between 100 and 170 dph. Na+, K+-ATPase activities increased fourfold in the pyloric caeca in 33 ppt, presumably due to increased salt and water absorption as indicated by GIT fluids, solids, and ion concentrations. In contrast to teleosts, a greater proportion of base (HCO3 and 2CO3 2−) was found in intestinal precipitates than fluids. Green sturgeon osmo- and ionoregulate with similar mechanisms to more-derived teleosts, indicating the importance of these mechanisms during the evolution of fishes, although salinity acclimation may be more dependent on body size.  相似文献

The salinity tolerance, and hydromineral regulation capabilities of three size groups (small 110–170 g; medium 230–290 g, large 460–700 g; n=48 for each group) of 13-month-old juvenile Gulf of Mexico sturgeon were investigated. Fish (n=6 for each salinity) were transferred directly from freshwater (FW) to a series of experimental salinity treatments (0, 5, 10, 15, 20, 25, 30, and 35 parts per thousand (ppt)). Fish were also acclimated in brackish water (20 ppt) for 2 weeks and transferred to a salinity of 34 ppt. In this condition juvenile Gulf of Mexico sturgeon adapted to saltwater (SW) and maintained their hydromineral balance. FW adapted sturgeon (n=6) had an average blood hemotocrit of 28.2±0.8%, plasma osmolality of 260.7±1.6 mOsm kg−1 H2O, and plasma ion concentrations of 135.7±1.2 mM l−1 Na+, 106.9±1.9 mEq l−1 Cl, and 2.9±0.1 mM l−1 K+. In SW adapted sturgeon (n=8) blood parameters averaged 26.9±0.7% for hematocrit, 294.2±2.3 mOsm kg−1 H2O for osmolality, 152.0±1.7 mM l−1 Na+, 149.2±1.4 mEq l−1 for Cl, and 3.1±0.1 mM l−1 K+. The method of transfer (abrupt or slow acclimation) directly affected fish survival and the time they took to achieve ionic and osmotic regulation. This SW adaptation appears to be related to body size, the larger the fish the easier the adaptation process. A threshold size of about 170 g was apparent for the fish to adapt to saltwater after 2 weeks of acclimation. Chloride cells were present in both FW and SW adapted sturgeon with SW and brackish water fish having chloride cells significantly (P<0.05) more numerous (561±53 and 598±45 cells mm−2) and larger in size (41.0±3.85 and 34.2±4.49 μm2) than FW adapted sturgeon (10±1.0 cells mm−2 and 22±2.53 μm2). Few chloride cells were observed in the opercular membrane, however, none were found in the pseudobranch and spiracle.  相似文献

Antarctic marine organisms are considered to have extremely limited ability to respond to environmental temperature change. However, here we show that the Antarctic notothenioid fish Pagothenia borchgrevinki is an exception to this theory. P. borchgrevinki was able to acclimate its resting metabolic rate and resting ventilation frequency after a 5°C rise in temperature. Acute exposure to 4°C resulted in an elevation in metabolic rate (57.8 ± 4.79 mg O2 kg−1 h−1) and resting ventilation rate (40.38 ± 1.61 breaths min−1) compared with fish at −1°C (metabolic rate 34.45 ± 3.12 mg O2 kg−1 h−1; ventilation rate 29.88 ± 3.72 breaths min−1). However, after a 1-month acclimation period, there was no significant difference in the metabolic rate (cold fish 29.52 ± 3.01; warm fish 31.13 ± 2.30 mg O2 kg−1 h−1), or the resting ventilation rate (cold fish 28.75 ± 0.98; warm fish 34.25 ± 2.28 breaths min−1) of cold and warm acclimated fish. Acclimation changes to the rate of oxygen consumption following exhaustive exercise were complex. The pattern of oxygen consumption during recovery from exhaustive exercise was not significantly different in either cold or warm acclimated fish.  相似文献

The physiology of hyper-salinity tolerance in teleost fish: a review   总被引:3,自引:0,他引:3  
Hyper-saline habitats (waters with salinity >35 ppt) are among the harshest aquatic environments. Relatively few species of teleost fish can tolerate salinities much above 50 ppt, because of the challenges to osmoregulation, but those that do, usually estuarine, euryhaline species, show a strong ability to osmoregulate in salinities well over 100 ppt. Typically, plasma Na+ and Cl concentrations rise slowly or not at all up to about 65 ppt. At higher salinities ion levels do rise, but the increase is small relative to the magnitude of increase in concentrations of the surrounding water. A number of adjustments are responsible for such strong osmoregulation. Reduced branchial water permeability is indicated by the observation that with the exposure to hyper-salinities drinking rates rise more slowly than the branchial osmotic gradient. Lower water permeability limits osmotic water loss and greatly reduces the salt load incurred in replacing it. Still, increased gut Na+/K+-ATPase (NAK) activity is necessary to absorb the larger gut salt load and increased HCO3 secretion is required to precipitate Ca2+ and some Mg2+ in the imbibed water to facilitate water absorption. All Na+ and Cl taken up must be excreted and increased branchial salt excreting capacity is indicated by elevated mitochondrion-rich cell density and size, gill NAK activity and expression of chloride channels. Excretion of Na+ and Cl occurs against a larger gradient than in seawater and calculation of the equilibrium potential for Na+ across the gill epithelium indicates that the trans-epithelial potential required for excretion of Na+ climbs with salinity up to about 65 ppt before leveling off due to the increasing plasma Na+ levels. During acute transition to SW or mildly hyper-saline waters, some species have shown the ability to upregulate branchial NAK activity rapidly and this may play an important role in limiting disturbances at higher salinities. It does not appear that the opercular epithelium, which in SW acts in a way that is functionally similar to the gills, continues to do so in hyper-saline waters. Little is know about the hormones involved in acclimation to hyper-salinity, but the few studies available suggest a role for cortisol, but not growth hormone and insulin-like growth factor. Despite the increased transport capacity evident in both the gill and gut in hyper-saline waters there is no clear trend toward increased metabolic rate. These studies provide a general outline of the mechanisms of osmoregulation in these species, but significant questions still remain.  相似文献

Physical, biogeochemical and photosynthetic parameters were measured in sea ice brine and ice core bottom samples in the north-western Weddell Sea during early spring 2006. Sea ice brines collected from sackholes were characterised by cold temperatures (range −7.4 to −3.8°C), high salinities (range 61.4–118.0), and partly elevated dissolved oxygen concentrations (range 159–413 μmol kg−1) when compared to surface seawater. Nitrate (range 0.5–76.3 μmol kg−1), dissolved inorganic phosphate (range 0.2–7.0 μmol kg−1) and silicic acid (range 74–285 μmol kg−1) concentrations in sea ice brines were depleted when compared to surface seawater. In contrast, NH4 + (range 0.3–23.0 μmol kg−1) and dissolved organic carbon (range 140–707 μmol kg−1) were enriched in the sea ice brines. Ice core bottom samples exhibited moderate temperatures and brine salinities, but high algal biomass (4.9–435.5 μg Chl a l−1 brine) and silicic acid depletion. Pulse amplitude modulated fluorometry was used for the determination of the photosynthetic parameters F v/F m, α, rETRmax and E k. The maximum quantum yield of photosystem II, F v/F m, ranged from 0.101 to 0.500 (average 0.284 ± 0.132) and 0.235 to 0.595 (average 0.368 ± 0.127) in the sea ice internal and bottom communities, respectively. The fluorometric measurements indicated medium ice algal photosynthetic activity both in the internal and bottom communities of the sea ice. An observed lack of correlation between biogeochemical and photosynthetic parameters was most likely due to temporally and spatially decoupled physical and biological processes in the sea ice brine channel system, and was also influenced by the temporal and spatial resolution of applied sampling techniques.  相似文献

Transepithelial potentials (TEP) were measured in killifish, acclimated to freshwater (FW), seawater (SW), 33% SW or cycling salinities relevant to tidal cycles in an estuary, and subsequently subjected to salinity changes in progressive or random order. Random compared to progressive salinity changes in an upward or downward direction in FW- and SW-acclimated fish, respectively, did not greatly influence responses to salinity change. Fish acclimated to SW or 33% SW as well as those acclimated to cycling salinities behaved similarly (TEP more positive than +15 mV in 100% SW, decreasing to ~0 mV at 20–40% SW, and more negative than −30 mV in FW). In contrast, FW-acclimated fish displayed a less pronounced TEP response to salinity (0 mV in FW through 20% SW, increasing thereafter to values more positive than +10 mV at 100% SW). We conclude that when evaluated under estuarine tidal conditions, the killifish gill exhibits adaptive electrical characteristics, opposing Na+ loss at low salinity and favouring Na+ extrusion at high salinity, changes explained at least in part by the Cl to Na+ permeability ratio. Thus animals living in the estuaries can move to lower and higher salinities for short periods with little physiological disturbance, but this ability is lost after acclimation to FW.  相似文献

Feeding and exhaustive exercise are known to elevate metabolism. However, acid–base status may be oppositely affected by the two processes. In this study, we first investigated the acid–base response of Chinese catfish to feeding (the meal size was about 8% of body mass) to test whether an alkaline tide (a metabolic alkalosis created by gastric HCl secretion after feeding) would occur. We then determined the combined effects of feeding and exhaustive exercise on excess post-exercise oxygen consumption and acid–base status to determine whether the alkaline tide induced by feeding protects against acid–base disturbance during exhaustive exercise and affects subsequent recovery. Arterial blood pH increased from 7.74 ± 0.02 before feeding to 7.88 ± 0.02 and plasma [HCO3 ]pl increased from 5.42 ± 0.29 to 7.83 ± 0.37 mmol L−1 6 h after feeding, while feeding had no significant effect on P\textCO2 P_{{{\text{CO}}_{2} }} . Exhaustive exercise led to a significant reduction in pH by 0.46 units and a reduction of [HCO3 ]pl by ~3 mmol L−1. Lactate concentrations in white muscle and plasma increased by 2.4 mmol L−1 and 13.4 μmol g−1, respectively. Fed fish had a higher pH and [HCO3 ]pl than fasting fish at rest, and the reductions in pH (0.36 units) and [HCO3 ]pl (~2 mmol L−1) were thus lower after exhaustive exercise. However, the recovery of acid–base status and metabolites were similar in digesting and fasting fish. Overall, a significant alkaline tide was found in Chinese catfish after feeding. The alkaline tide elicited by feeding significantly prevented the decreases in pH and [HCO3 ]pl immediately after exhaustive exercise, but recovery from exhaustive exercise was not affected by digestion.  相似文献

The green sturgeon is a long-lived, highly migratory species with populations that are currently listed as threatened. Their anadromous life history requires that they make osmo- and ionoregulatory adjustments in order to maintain a consistent internal milieu as they move between fresh-, brackish-, and seawater. We acclimated juvenile green sturgeon (121 ± 10.0 g) to 0 (freshwater; FW), 15 (estuarine; EST), and 24 g/l (SF Bay water; BAY) at 18°C for 2 weeks and measured the physiological and biochemical responses with respect to osmo- and ionoregulatory mechanisms. Plasma osmolality in EST- and BAY-acclimated sturgeon was elevated relative to FW-acclimated sturgeon (P < 0.01), but there was no difference in muscle water content or abundance of stress proteins. Branchial Na+, K+-ATPase (NKA) activity was also unchanged, but abundance within mitochondrion-rich cells (MRC) was greater in BAY-acclimated sturgeon (P < 0.01). FW-acclimated sturgeon had the greatest NKA abundance when assessed at the level of the entire tissue (P < 0.01), but there were no differences in v-type H+ATPase (VHA) activity or abundance between salinities. The Na+, K+, 2Cl co-transporter (NKCC) was present in FW-acclimated sturgeon gills, but the overall abundance was lower relative to sturgeon in EST or BAY water (P < 0.01) where this enzyme is crucial to hypoosmoregulation. Branchial caspase 3/7 activity was significantly affected by acclimation salinity (P < 0.05) where the overall trend was for activity to increase with salinity as has been commonly observed in teleosts. Sturgeon of this age/size class were able to survive and acclimate following a salinity transfer with minimal signs of osmotic stress. The presence of the NKCC in FW-acclimated sturgeon may indicate the development of SW-readiness at this age/size.  相似文献

In an experimental site for reforestation of degraded area, three-year-old plants of Bertholletia excelsa Humb. & Bonpl. were subjected to different fertilization treatments: T0 = unfertilized control, T1 = green fertilization (branches and leaves) and T2 = chemical fertilization. Higher net photosynthetic rates (P N) were observed in T1 [13.2±1.0 μmol(CO2) m−2 s−1] compared to T2 [8.0±1.8 μmol(CO2) m−2 s−1] and T0 [4.8±1.3 μmol(CO2) m−2 s−1]. Stomatal conductance (g s), transpiration rate (E) and water use efficiency (WUE) of individuals of T1 and T2 did not differ significantly, however, they were by 88, 55 and 63%, respectively, higher in T1 than in the control. The mean values of variable fluorescence (Fv), performance index (P.I.) and total chlorophyll [Chl (a+b)] were higher in T1. Our results indicate that green fertilization improves photosynthetic structure and function in plants of B. excelsa in young phase.  相似文献

The effects of feeding on both acid–base and ion exchange with the environment, and internal acid–base and ion balance, in freshwater and seawater-acclimated flounder were investigated. Following voluntary feeding on a meal of 2.5–5% body mass and subsequent gastric acid secretion, no systemic alkaline tide or respiratory compensation was observed in either group. Ammonia efflux rates more than doubled from 489 ± 35 and 555 ± 64 μmol kg−1 h−1 under control conditions to 1,228 ± 127 and 1,300 ± 154 μmol kg−1 h−1 post-feeding in freshwater and seawater-acclimated fish, respectively. Based on predictions of gastric acid secreted during digestion, we calculated net postprandial internal base gains (i.e., HCO3 secreted from gastric parietal cells into the blood) of 3.4 mmol kg−1 in seawater and 9.1 mmol kg−1 in freshwater-acclimated flounder. However, net fluxes of ammonia, titratable alkalinity, Na+ and Cl indicated that branchial Cl/HCO3 and Na+/H+ exchange played minimal roles in counteracting these predicted base gains and cannot explain the absence of alkaline tide. Instead, intestinal Cl/HCO3 exchange appears to be enhanced after feeding in both freshwater and seawater flounder. This implicates the intestine rather than the gills as a potential route of postprandial base excretion in fish, to compensate for gastric acid secretion.  相似文献

In frog red blood cells, K-Cl cotransport (i.e., the difference between ouabain-resistant K fluxes in Cl and NO3) has been shown to mediate a large fraction of the total K+ transport. In the present study, Cl-dependent and Cl-independent K+ fluxes via frog erythrocyte membranes were investigated as a function of external and internal K+ ([K+] e and [K+] i ) concentration. The dependence of ouabain-resistant Cl-dependent K+ (86Rb) influx on [K+] e over the range 0–20 mm fitted the Michaelis-Menten equation, with an apparent affinity (K m ) of 8.2 ± 1.3 mm and maximal velocity (V max ) of 10.4 ± 1.6 mmol/l cells/hr under isotonic conditions. Hypotonic stimulation of the Cl-dependent K+ influx increased both K m (12.8 ± 1.7 mm, P < 0.05) and V max (20.2 ± 2.9 mmol/l/hr, P < 0.001). Raising [K+] e above 20 mm in isotonic media significantly reduced the Cl-dependent K+ influx due to a reciprocal decrease of the external Na+ ([Na+] e ) concentration below 50 mm. Replacing [Na+] e by NMDG+ markedly decreased V max (3.2 ± 0.7 mmol/l/hr, P < 0.001) and increased K m (15.7 ± 2.1 mm, P < 0.03) of Cl-dependent K+ influx. Moreover, NMDG+ Cl substitution for NaCl in isotonic and hypotonic media containing 10 mm RbCl significantly reduced both Rb+ uptake and K+ loss from red cells. Cell swelling did not affect the Na+-dependent changes in Rb+ uptake and K+ loss. In a nominally K+(Rb+)-free medium, net K+ loss was reduced after lowering [Na+] e below 50 mm. These results indicate that over 50 mm [Na+] e is required for complete activation of the K-Cl cotransporter. In nystatin-pretreated cells with various intracellular K+, Cl-dependent K+ loss in K+-free media was a linear function of [K+] i , with a rate constant of 0.11 ± 0.01 and 0.18 ± 0.008 hr−1 (P < 0.001) in isotonic and hypotonic media, respectively. Thus K-Cl cotransport in frog erythrocytes exhibits a strong asymmetry with respect to transported K+ ions. The residual, ouabain-resistant K+ fluxes in NO3 were only 5–10% of the total and were well fitted to linear regressions. The rate constants for the residual influxes were not different from those for K+ effluxes in isotonic (∼0.014 hr−1) and hypotonic (∼0.022 hr−1) media, but cell swelling resulted in a significant increase in the rate constants. With an emphasis on the tight junction protein occludin, the response of goldfish following abrupt exposure (0–120 h) as well as long-term acclimation (14 and 28 days) to ion-poor water (IPW) was examined. Both abrupt and long-term exposure to IPW lowered serum osmolality, [Na+] and [Cl], and elevated serum glucose. After abrupt exposure to IPW, gill tissue exhibited a prompt and sustained decrease in Na+–K+–ATPase activity, and a transient increase in occludin expression that returned to control levels by 6 h. Following 14 and 28 days in IPW, gill occludin expression was markedly elevated, while Na+–K+–ATPase activity was only significantly different (elevated) at day 14. Kidney tissue exhibited an elevation in both Na+–K+–ATPase activity and occludin expression after 28 days; however, in the intestine, occludin expression declined at day 14 but did not differ from FW fish at day 28. These studies demonstrate that goldfish can tolerate abrupt as well as sustained exposure to ion-poor surroundings. Data also suggests that occludin may play an adaptive role in fishes acclimated to ion-poor conditions by contributing to the modulation of epithelial barrier properties in ionoregulatory tissues. A series of metallopeptides based on the amino terminal copper/nickel (ATCUN) binding motif have been evaluated as classical inhibitors and catalytic inactivators of both rabbit and human angiotensin-converting enzyme (hACE), and human endothelin-converting enzyme 1 (hECE-1). The cobalt complex [KGHK–Co(NH3)2]2+, where KGHK is lysylglycylhistidyllysine, displayed similar K I and IC50 values to those found for [KGHK–Cu]+, in spite of the enhanced charge, and so either the influence of charge is offset by the steric influence of the axially coordinated ammine ligands, or binding is dominated by contributions from the amino acid side chains, especially the C-terminal lysine that mimics the binding pattern observed for lisinopril. Moreover, the inhibition observed for [KGHK–Co(NH3)2]2+ contrasts with the activation of hACE by Co2+(aq), reflecting the stimulation of enzyme activity following replacement of the catalytic zinc cofactor by cobalt ion at each of the two active sites. Quantitative analysis of the dose-dependent stimulation of activity by Co2+(aq) yielded apparent affinities of 1.3 ± 0.2 and 56 ± 8 μM for the two sites in the presence of saturating Zn2+ (10 μM). Catalytic inactivation of hACE by [KGHK–Cu] + at subsaturating concentrations had previously been characterized, with k obs = 2.9 ± 0.5 × 10−2 min−1. Under similar conditions, the same complex is found to catalytically inactivate hECE-1, with k obs = 2.12 ± 0.16 × 10−2 min−1, demonstrating the potential for dual-action activity against two key drug targets in cardiovascular disease. The purpose of the present study was to ascertain the tissue-specific expression of the water channel protein, aquaporin 3 (AQP3), during salinity acclimation and larval development of silver sea bream (Sparus sarba). A cDNA fragment encoding aquaporin 3 (aqp3) from silver sea bream gill was cloned and from the deduced amino acid sequence a polyclonal antibody was prepared. AQP3 was found to be present in gill, kidney, liver, brain, heart, and spleen but not in whole blood. The abundance of AQP3 was significantly highest in gills of hypoosmotic (6 ppt) and isoosmotic (12 ppt) acclimated sea bream when compared to seawater (33 ppt) and hypersaline (50 ppt)- acclimated sea bream. Spleen tissue also displayed significantly high levels of AQP3 protein in hypoosmotic and isoosmotic salinities whereas the AQP3 abundance in brain, liver, heart, and kidney remained unchanged across the range of salinities tested. The ontogenetic profile of AQP3 was also investigated from developing sea bream larvae and AQP3 was first detected at 14 days posthatch (dph) and increased steadily up to 28–46 dph. In conclusion, this study has demonstrated that AQP3 expression is modulated in gill and spleen tissue of salinity acclimated sea bream and that it can be detected relatively early during larval development.  相似文献

Plasma and urine of toadfish (Opsanus tau) in sea water and 10% sea water were analyzed to assess responses of an aglomerular fish to hypoosmotic challenge. Following transfer to 10% sea water, plasma osmotic pressure decreased slowly from 318 to 241 mmol · kg H2O−1, over a period of 10–15 days. Urine osmotic pressure decreased in parallel from 299 to 207 mmol · kg H2O−1, leaving urine/plasma ratios of osmotic pressure essentially unchanged. In contrast, the volume and composition of urine changed rapidly following transfer to 10% sea water. Urine flow rate increased 110% from 3.0 to 6.3 μl · 100g−1 · h−1 and Na+ excretion increased 346%, while excretion of Mg2− and SO4 2− decreased 81% and 90%, respectively. Excretion rates for Cl were low in seawater toadfish and decreased further in 10% sea water. An unknown sulfur-containing anion, present in the urine of seawater toadfish, contributed significantly to the composition and ionic balance in urine of toadfish in 10% sea water. These results suggest that the inability to produce strongly dilute urine obliges toadfish to lose salt in order to excrete water, in hypoosmotic media. The decrease in plasma osmotic pressure may be both a strategy to reduce osmotic and ionic gradients in dilute media and a consequence of the kidney's inability to excrete water without salt. The effect of ANG II on pHi, [Ca2+]i and cell volume was investigated in T84 cells, a cell line originated from colon epithelium, using the probes BCECF-AM, Fluo 4-AM and acridine orange, respectively. The recovery rate of pHi via the Na+/H+ exchanger was examined in the first 2 min following the acidification of pHi with a NH4Cl pulse. In the control situation, the pHi recovery rate was 0.118 ± 0.001 (n = 52) pH units/min and ANG II (10−12 M or 10−9 M) increased this value (by 106% or 32%, respectively) but ANG II (10−7 M) decreased it to 47%. The control [Ca2+]i was 99 ± 4 (n = 45) nM and ANG II increased this value in a dose-dependent manner. The ANG II effects on cell volume were minor and late and should not interfere in the measurements of pHi recovery and [Ca2+]i. To document the signaling pathways in the hormonal effects we used: Staurosporine (a PKC inhibitor), W13 (a calcium-dependent calmodulin antagonist), H89 (a PKA inhibitor) or Econazole (an inhibitor of cytochrome P450 epoxygenase). Our results indicate that the biphasic effect of ANG II on Na+/H+ exchanger is a cAMP-independent mechanism and is the result of: 1) stimulation of the exchanger by PKC signaling pathway activation (at 10−12 – 10−7 M ANG II) and by increases of [Ca2+]i in the lower range (at 10−12 M ANG II) and 2) inhibition of the exchanger at high [Ca2+]i levels (at 10−9 – 10−7 M ANG II) through cytochrome P450 epoxygenase-dependent metabolites of the arachidonic acid signaling pathway.  相似文献

The aim was to study osmotic and ionic regulation in alien ctenophore, Mnemiopsis leidyi (total length of 1.5-3 cm), acclimated gradually to artificial sea water of different salinities (8, 13, 18 and 23 ppt) in laboratory conditions (24 ± 2 °C) with the salt composition and proportion of the Caspian Sea. Results showed that this species is hyper-osmoconformer (maintaining internal osmolarity 2-22 mOsmol l− 1 above external) in non-lethal salinities ranging from 8 to 23 ppt. The results of ionic regulatory investigations revealed that this species can regulate Ca2+ and SO42− so that the concentration of Ca2+ in the internal fluid and SO42− in the external fluid were significantly more in 8 and 13 ppt, respectively. No regulation was observed about other ions such as Cl, Mg2+ and K+ in the mentioned salinities.  相似文献

The biodegradation potential of an innovative enclosed tubular biofilm photobioreactor inoculated with a Chlorella sorokiniana strain and an acclimated activated sludge consortium was evaluated under continuous illumination and increasing pretreated (centrifuged) swine slurry loading rates. This photobioreactor configuration provided simultaneous and efficient carbon, nitrogen, and phosphorous treatment in a single-stage process at sustained nitrogen and phosphorous removals efficiencies ranging from 94% to 100% and 70–90%, respectively. Maximum total organic carbon (TOC), NH4 +, and PO4 3− removal rates of 80 ± 5 g C mr −3 day−1, 89 ± 5 g N mr −3 day−1, and 13 ± 3 g P mr −3 day−1, respectively, were recorded at the highest swine slurry loadings (TOC of 1,247 ± 62 mg L−1, N–NH4 + of 656 ± 37 mg L−1, P–PO4 3+ of 117 ± 19 mg L−1, and 7 days of hydraulic retention time). The unusual substrates diffusional pathways established within the phototrophic biofilm (photosynthetic O2 and TOC/NH4 + diffusing from opposite sides of the biofilm) allowed both the occurrence of a simultaneous denitrification/nitrification process at the highest swine slurry loading rate and the protection of microalgae from any potential inhibitory effect mediated by the combination of high pH and high NH3 concentrations. In addition, this biofilm-based photobioreactor supported efficient biomass retention (>92% of the biomass generated during the pretreated swine slurry biodegradation).  相似文献
