Infection threads were observed abundantly in the root hairsof Lotus corniculatus L., but very rarely in L. hispidus, Desf.,in response to infection by Rhizobium strains 3001 and 3002.Numbers of infections differed between species and strains andwere also affected by temperature. In L. corniculatus all thenodules originated from infection threads, but in L. hispidusmost nodules appeared to originate by direct bacterial penetrationthrough the epidermis, and infected root hairs were very rarelyseen. Both species of Lotus were tolerant to cold temperatures,the minimum temperature for nodulation being 10 ?C. The optimumtemperature for nodulation of L. corniculatus was 20 ?C with3001 and between 27 and 30 ?C with 3002, a few nodules beingformed with both strains at 35 ?C. L. hispidus formed more nodulesthan L. corniculatus and the optimum temperature for both thestrains was between 25 and 27 ?C. No infection threads were seen in root hairs or nodules of Stylosanthesguyanensis (Aubl.) S. W. and S. humilis H.B.K. infected withRhizobium strain CB1552, and all the nodules were formed inthe axils of lateral roots. Optimum temperature for nodulationin S. guyanensis and S. humilis was around 27 ?C; nodulationwas completely inhibited at 15 ?C and very few nodules wereformed at 35 ?C. Both in Lotus and Stylosanthes the transfer of plants from suboptimalto optimal and supraoptimal temperatures increased nodulation.Delayed inoculation and excision of root tips increased nodulation.  相似文献

The water-relations parameters of Chara inflata cells were determineddirectly using the micro pressure probe technique. The turgorpressure of cells in artificial pond water (0 = 0.06 MPa) wasabout 0.65 MPa and the half-time (T1/2) for water exchange wasabout 6.5 s. The calculated values of the hydraulic conductivity(LP) were in the range 1–2 ? 10–6m s–1 (MPa)–1.The volumetric elastic modulus () was 32.8 MPa for turgor rangingfrom 0.77 to 0.82 MPa. Large changes in the water-relations parameters and the electricalproperties of the membrane occurred when the turgor was decreasedto low values. These changes included: (i) a decrease in theT1/2 for water exchange, (ii) an increase in LP and (iii) depolarizationof the membrane potential difference (Vm). The micro pressure probe, which enabled the turgor pressureof the cell to be altered, was used in combination with thevoltage-clamp technique to determine the relationship betweenK+ and Cl conductances of the plasmalemma and the cellturgor. The K+ conductance increased reversibly as the turgorwas reduced in the range 0 to 0.6 MPa and the Cl -conductanceincreased as the turgor was reduced in the range 0.1 to 0.5MPa. It is suggested that these pressure-dependent K+ and Clconductances may have a dual role in electrical events and thenon-electrical responses such as changes in the cell volume. Cytoplasmic pH (pHc) in Chara corallina was measured (from [14C]stribution)as a function of external pH (pH0)and temperature. With pH0near 7, pHc at 25?C is 7.80; pHcincreases by 0.005 pH units?C–1 temperature decrease, i.e. pHc at 5 ?C is 7.90. WithpH? near 5.5, the increase in pHc with decreasing temperatureis 0.015 units ?C–1 between 25 and 15?C, but 0.005 units?C–1 between 15 and 5?C. This implies a more precise regulationof pHc with variations in pHo at 5 or 15 ?C compared with 25?C. The observed dp Hc/dT is generally smaller than the –0.017units ?C–1 needed to maintain a constant H+/OH–1,or a constant fractional ionization of histidine in protein,with variation in temperature. It is closer to that needed tomaintain the fractional ionization of phosphorylated compoundsor of CO2–HCO3 The value of dpHc/dT has importantimplications for several regulatory aspects of cell metabolism.These include (all as a function of temperature) the rates ofenzyme reactions, the H+ at the plasmalemma(and hence the energy available for cotransport processes),and the mechanism for pHc regulation by the control of bidirectionalH+ fluxes at the plasmalemma.  相似文献

Glycogen synthetase, ADP-glucose-a (l4) glucan transglucosylase[E.C. 2.4.1.11 [EC] ] from a purple sulfur bacterium, Chromatium,was purified to a homogeneous state and its enzymic propertieswere studied. The molecular weight of the enzyme was 8.6?104dalton as determined by analytical gel filtration on a columnof Sephadex G-100. Since sodium dodecyl sulfate-polyacrylamidegel electrophoresis gave the molecular weight value of 8.4?104to the monomeric form of the enzyme, we concluded that Chromatiumglycogen synthetase is comprised of a single polypeptide chain.The optimal pH of teh transglucosylation reaction was between8.0–8.5. The enzyme molecule utilized only ADP-glucoseas the glucose donor. The km value was determined as 3.8?10-4M by the radioisotopic method of measuring the incorporationof 14C-glucose into the acceptor glycogen, and 6.1?10-5M bythe enzyme coupling method. The most effective glucose acceptor(primer) was proved to be a long-chain a (16) branched a (14)polyglucan, e.g. Chromatium and cow glycogen, whereas short-chainmalto-oligosaccharides were much less efficient in the chain-elongationreaction. 1 Part I of this series is Ref. For a single seed population of each of four species of grainlegume positive linear relationships were shown between temperatureand rate of germination for different fractions (G) of eachpopulation, from a base temperature, Tb(G), at which germinationrate is zero, to an optimum temperature, To(G) at which germinationrate is maximal. At constant temperatures warmer than To(G)there were negative relations (probably linear) between temperatureand rate of germination to the maximum temperature for germination,Tm(G), Within each population Tb(G) did not differ, but it didvary between species, viz.0.0?C, 0.25?C, 4.and 8.5?C for chickpea(Cicer arietinum L.), lentil (Lens culinaris Medic.), soyabean(Glycine max [ Merr.) and cowpea (Vigna unguiculata [L.] Walp.),respectively. In contrast, To(G) varied both within each populationand also between the four species: 80% of seeds in each populationhad To(G) values within the range 31.8?C to 33.8 ?C, 24.0?Cto 24.4?C, 34.0?C to 34.5?C and 33.2?C to >40?C, respectively.Values of Tm(G) were much more vanable: the 80% population rangewas 48 .0?C to 60.8?C for chickpea, 31.8?C to 34.4?C for lentiland 46.8?C to 55.2?C for soyabean; reliable estimates couldnot be made for cowpea, but the results suggest higher and morevariable values of Tm(G) than in the other three species. Atsub-optimal temperatures the distribution of thermal time forthe different fractions of each population was normal, exceptfor lentil where it was log-normal. A single equation is proposedto describe the influence of sub-optimal temperatures on ratesof germination for whole seed populations. At supra-optimaltemperatures, variation in thermal time for the different fractionsof each population was only slight. The implications of thesefindings for the adaptation of grain legume crops to differentenvironments, and for the screening of germplasm, are discussed. Leaf temperatures (T1) of the parasitic plant Striga hermonthicaare substantially below those of the air (Ta), [TaT1]reaching 7 ?C at Ta = 40 ?C. This results from high rates oftranspiration and the consequent evaporative cooling of theleaf. Application of an antitranspirant, which mechanicallyimpedes foliar loss of water vapour, reduced transpiration andstomatal conductance by 40% and 57%, respectively, and reduced[TaT1] to 2 ?C at Ta = 40 ?C. The temperature sensitivityof photosynthesis in the host-parasite association differed,the optima (Topt) being 37.2 and 40.1 ?C for S. hermonthicaand sorghum, respectively. Once Topt had been exceeded in S.hermonthica net photosynthesis declined rapidly, reaching thelethal limit (Tmax) at 42.6 ?C. S. hermonthica is particularlysensitive to high temperatures and antitranspirant-induced overheatingleads to blackening and shrivelling of the leaf after as littleas 4 h at Ta = 40 ?C. Application of an antitranspirant underfield conditions in the Sudan at Ta = 40 ?C resulted in 28%and 67% reductions in transpiration and stomatal conductance,together with a 5 ?C increase in T1, and subsequent leaf death.In addition to these short-term physiological responses, antitranspirantspraying of the arasite increased the grain and straw yieldof the crop by factors of 3.4 and 2.6, respectively. Antitranspirantsmay have potential use as a method of controlling Striga inthe field. Bot. 38: 1–12 Glasshouse grown, symbiotically-dependent seedlings of Acaciaalata R.Br., .A. extensa Lindl., and A. pulchella R.Br. wereexamined for acetylene reduction in closed assay systems usingundisturbed potted plants, excavated whole plants, nodulatedroots or detached nodules. Nitrogenase activity declined sharplyover the first hour after exposure of detached nodules to acetylene(10% v/v in air), less steeply or not at all over a 3 h periodin assays involving attached nodules. Using detached nodules,rates of acetylene reduction, nitrogen (15N2) fixation, andhydrogen evolution in air (15N2) and acetylene-containing atmosphereswere measured in comparable 30 min assays. Total electron flowthrough nitrogenase in air was determined from rates of nitrogen(15N2) fixation ( ? 3) plus hydrogen evolution, that in thepresence of acetylene from rates of acetylene reduction andhydrogen evolution in air: acetylene. Values for the ratio ofelectron flow in air: acetylene to that in air ranged from 0?43to 0?83 in A. pulcheila, from 0?44 to 0?66 in A. alala and from0?37 to 0?70 in A. extensa, indicating substantial inhibitionof electron flow through nitrogenase of detached nodules byacetylene. Relative efficiencies of nitrogenase functioningbased on hydrogen evolution and acetylene reduction were from0?15 to 0?79, those based on nitrogen (15N2) fixation and hydrogenevolution from 0?53 to 0?87. Molar ratios of acetylene reducedto nitrogen (15N2) fixed were 2?82 ? 0?24, 201 ? 0?15, and 1?91? 0?11 (?s.e.; n = 7) for A. pulcheila,A. extensa and A. alata respectively A standard 5–10 min acetylene reduction assay, conductedon freshly detached unwashed nodules in daytime (12.00–14.00h), was calibrated for field use by comparing total N accumulationof seedlings with estimated cumulative acetylene reduction overa 7-week period of glasshouse culture. Molar ratios for acetylenereduced: nitrogen fixed using this arbitrary method were 3?58for A. alata, 4?82 for A. extensa and 1?60 for A. pulchella.The significance of the data is discussed. The vapour pressures of aqueous solutions of polyethylene glycol6000 have been measured (by equilibration with sucrose solutions)up to the saturation point at 25 °C (1.45 g g–1 water).The reduced-osmotic-pressure (/c), when plotted versus concentration(c), rapidly and linearly increased up to a concentration ofabout 0.8 g g–1 (crossing the similar plot for sucrose).Above this concentration, the reduced-osmotic-pressure rosemore slowly, but still more rapidly than sucrose. The maximumosmotic pressure achieved at saturation was nearly 18 MPa. Usingthe virial equation: /c= RT/M + RTA2c, the calculated secondvirial coefficient (A2) for the linear part is 4.5 x 10–3mol g–1, a value slightly greater than most literaturevalues at 25 °C. Bot. 38: 67–76 The permeabilities of the amines, ammonia (NH3), methylamine(CH3NH2) and ethylamine (CH3CH2NH2) in the giant-celled charophyteChara corallina (C. australis) R.Br. have been measured andcompared. The permeabilities were corrected for uptake fluxesof the amine cations. Based on net uptake rates, the permeabilityof ammonia was 6?4?0?93 µm s–1 (n = 38). The permeabilitiesof methylamine and ethylamine were measured in net and exchangeflux experiments. The permeabilities of methylamine were notsignificantly different in net and exchange experiments, norto that of ammonia (Pmethylamine = 6?0?0?49 µm s–1(n = 44)). In net flux experiments the apparent permeabilityof ethylamine was slightly greater than that of ammonia andmethylamine (Pethylamine, net = 8?4?1?2 µm s–1 (n= 40)) but the permeability of ethylamine based on exchangeflux data was significantly higher (Pethylamine, exchange =14?1?2 µm s–1 (n = 20)). Methylamine can be validlyused as an ammonium analogue in permeability studies in Chara. The plasmalemma of Chara has acid and alkaline bands; littlediffusion of uncharged amines would occur across the acid bands.The actual permeability of amines across the alkaline bandsis probably about twice the values quoted above on a whole cellbasis i.e. the permeability of ammonia across the permeablepart of the plasmalemma is probably about 12 µm s–1. A strong correlation between the photosynthetic parameters Jmaxand Vcmax was found by Wullschleger (1993) in a survey of 109plant species. Measurements were made at various leaf temperatures,but the temperature dependence of Jmax and Vcmax differ. Oncevalues for Jmax and Vcmax in Wullschleger's analysis were adjustedto a common temperature, using an equation for the temperaturedependence of these parameters, the slope of the linear regressionfor Jmax versus Vcmax forced through the origin increased from1.97 to 2.68, and r2 increased from 0.79 to 0.87. The purpose ofthis study was to test the hypothesis that regulated body temperatureis decreased in the preovulatory phase in eumenorrheic women. Six womenwere studied in both the preovulatory phase (Preov-2;days 9-12), which was 1-2days before predicted ovulation when 17-estradiol(E2) was estimated to peak, andin the follicular phase (F; days2-6). The subjects walked on a treadmill (~225W · m2)in a warm chamber (ambient temperature = 30°C; dew-pointtemperature = 11.5°C) while heavily clothed.E2, esophageal temperature(Tes), local skin temperatures,and local sweating rate were measured. The estimate of when theE2 surge would occur was correctfor four of six subjects. In these four subjects,E2 increased(P  0.05) from 42.0 ± 24.5 pg/mlduring F to 123.2 ± 31.3 pg/ml during Preov-2. RestingTes was 37.02 ± 0.20°Cduring F and 36.76 ± 0.28°C during Preov-2(P  0.05). TheTes threshold for sweating wasdecreased (P  0.05) from 36.88 ± 0.27°C during F to 36.64 ± 0.35°C during Preov-2. Both meanskin and mean body temperatures were decreased during rest in Preov-2group. The hypothesis that regulated body temperature is decreasedduring the preovulatory phase is supported.

Label from 14CO2 and [2-14C]mevalonic acid was incorporatedinto resin components of Newcastelia viscida. About 25% of thelabel from [2-14C]MVA was incorporated into resin componentsafter 4 h. Approximately 80% of the label in the resin was ina TLC band coinciding with the major terpenoid components, with60% associated with the major tricyclic diterpene, a pimaradienewhich contributes 15% to the leaf resin dry weight. Experimentswith detached trichomes showed about 0?01% incorporation oflabel from [2-14C]MVA into pimaradiene, with most of the [abelbeing associated with ß-sitosterol. Smith, J. R. 1987. Potassium transport across the membranesof Chara. II. 42K fluxes and the electrical current as a functionof membrane voltage.—J. exp. Bot. 38: 752–777. The current required to clamp the trans-membrane voltage ofinternodal cells of Chara australis at different levels wasmeasured simultaneously with either the 42K influx or efflux.Examination of the voltage-dependence of the ratio of the electricalcurrent to the unidirectional tracer fluxes yielded no evidenceof any amplification of the electrical driving force on theK+ ions. There was thus no evidence for the interaction of K+ions with themselves or any other species during their passageacross the membrane. These measurements allow the determinationof , the fraction of the electrical current carried by K+ ions.When the external [K+] = 10 mol m–3, the average valueof was 0?85 for Vm > –125 mV and 07?5 for Vm <–150 mV. When the external [K+] = 0?1 mol m–3, was 0?6 for Vm < –80 mV and 0?1 for Vm > –250mV. It was also found that the conductance associated with K+transport was inhibited by hyperpolarization. We have isolatedciliated respiratory cells from the nasal epithelium of wild-type andcystic fibrosis (CF) null mice and used the patch-clamp technique toinvestigate their basal conductances. Current-clamp experiments onunstimulated cells indicated the presence ofK+ andCl conductances and, undercertain conditions, a small Na+conductance. Voltage-clamp experiments revealed three distinct Cl conductances.Itv-indep wastime and voltage independent with a linear current-voltage(I-V)plot; Iv-actexhibited activation at potentials greater than ±50 mV, giving anS-shapedI-Vplot; andIhyp-act wasactivated by hyperpolarizing potentials and had an inwardly rectifiedI-Vplot. The current density sequence was Ihyp-act = Iv-act  Itv-indep. Theseconductances hadCl-to-N-methyl-D-glucaminecation permeability ratios of between 2.8 and 10.3 and were unaffectedby tamoxifen, flufenamate, glibenclamide, DIDS, and5-nitro-2-(3-phenylpropylamino) benzoic acid but were inhibited byZn2+ andGd3+.Itv-indep andIv-act werepresent in wild-type and CF cells at equal density and frequency.However, Ihyp-actwas detected in only 3% of CF cells compared with 26% of wild-typecells, suggesting that this conductance may be modulated by cysticfibrosis transmembrane conductance regulator (CFTR).

The Effect of Temperature on Leaf Appearance in Rice   总被引:13,自引:3,他引:10  
Temperature is the principal environmental determinant of cropleaf appearance. The objective of this study is to analyse whetherthere are different effects of day temperature (TD) and nighttemperature (TN) on main-stem leaf appearance in rice (OryzasativaL.). Plants of 12 rice cultivars were grown at five constant temperatures(22, 24, 26, 28 and 32 °C) and four diurnally fluctuatingtemperatures (TD/TN: 26 /22, 30 /22, 22 /26 and 22 /30 °C)with a constant photoperiod of 12hd-1. The leaf appearance onthe main stem was measured. A constant change in leaf appearance rate was observed duringontogeny. The relation between the number of emerged leavesand days from seedling emergence was described by a power-lawequation with only one cultivar-specific parameter. Values forthis parameter were estimated for the five constant temperaturetreatments, and the relation between this parameter and temperaturewas quantified by a nonlinear model. Leaf appearance for thefour fluctuating temperature treatments could be accuratelypredicted on the basis of these relations in each cultivar.This indicated that there were no specific effects ofTDandTNonleaf appearance in rice, in contrast with phenological developmentto flowering. The optimum temperature for leaf development wasfound to be substantially higher than for development to flowering. The final main-stem leaf number differed with diurnal temperatureconditions. When a diurnal temperature delayed flowering, itincreased the leaf number as well. This might explain whyTDandTNhada different effect on development to flowering but not on leafdevelopment. The theory and practice of applying the thermodynamics of irreversibleprocesses to mass-flow theories is presented. Onsager coefficientswere measured on cut and uncut phloem and cut xylem strandsof Heracleum muntegazzimum. In 0.3 N sucrose + 1 mN KC1 theyare as follows. In phloem, LEE = 5 ? 10–4 mho cm–1,LpE = 9 ? 10–6 cm3 s–1 cm–2 volt–1 cm,and LPP = 0.16 cm3 s–1 cm–2 (J cm–3)–1cm. In uncut phloem strands LEE is about 1 ? 10–3 mhocm–1. In xylem in 2 x 10–3 N KCI, Lpp = 50 to 225,LPE = 2 ? 10–4, and LEE = 4 ? 10–3. The measurementsare tentative since the blockage of the sieve plates is an interferingfactor, but if they are valid they lead to the conclusion thatneither a pressure-flow nor an electro-kinetic mechanism envisaginga ‘long distance’ current pathway can be the majormotive ‘force’ for transport in mature phloem. Measurementsof biopotentials along conducting but laterally detached phloembundles of Heracleum suggest, nevertheless, that there may bea small electro-osmotic component of at least 0.1 mV cm–1endogenous in the phloem.  相似文献

The influence of temperature on photosynthesis and transpirationwas studied in ten varieties of Lolium perenne, L. multiflorum,Dactylis glomerata, and Festuca arundinacea from three climaticorigins grown in three different controlled environments (15?C, 72 W m-2 visible irradiation, 16-h photoperiod; 25 ?C, 72W m-2 visible irradiation, 16-h photoperiod; and 25 ?C, 180W m-2 visible irradiation, 16-h photoperiod) and in the glasshousein July/August. The optimum temperature for photosynthesis was influenced primarilyby growth environment; growth at low temperature (15 ?C) resultedin a low optimum temperature, which differed little from varietyto variety. The maximum CO2-exchange rate was influenced bygrowth environment and by variety. Within a variety, plantsgrown at higher light intensity or lower temperature had a greaterCO2-exchange rate. Seven varieties showed a negative correlationbetween the optimum leaf temperature and the maximum CO2-exchangerate. Activation energies for photosynthesis were influencedby growth environment only. There were marked varietal differences in the values of leafresistances (ra + rt) obtained from transpiration data at theoptimum leaf temperature for CO2 exchange. In Lolium, and Dactylisthe Mediterranean varieties had higher leaf resistances thanthe Northern varieties with the maritime varieties intermediate.In general the Dactylis varieties had higher resistances thanthe corresponding Lolium and Festuca varieties. Only at highgrowth temperatures was (ra+rl) insensitive to temperature;otherwise an activation energy of about 10 kcal/mole was observed.A negative correlation was found between mean varietal diffusionresistances (ra+rl), and corresponding maximum CO2-exchangerates.  相似文献

The effects of several treatments, normally used to inhibitelectrogenic proton transport, upon the potassium permeability(Pk) of the membranes of Chara were examined by means of simultaneousmeasurements of the 42K influx (inK) and the membrane electricalconductance (Gm). inK, PK and Gmwere found to be substantiallyunaffected when the external pH (pH?) was varied over the range5?0 to 85. However, when pH? was increased to 11 it was foundthat, although Gm increased considerably, both Pk and inK decreasedtypically by an order of magnitude. When cells were placed intotal darkness, PK decreased substantially only after one dayhad elapsed. For the particular experimental conditions used,the inhibitors DES, NaN3, and La3+ were found to alter PK, whereasDCCD left PK substantially unaffected. These results suggestthat care must be taken with some common procedures used toexamine the electrical properties of the electrogenic protonpump. The dependence of membrane potentials on changes in the extra-cellularK+ concentration [K+]e was investigated in potato tuber sliceswith dripping perfusion, and in growing Vigna hypocotyl segmentswith pressurized intra-organ perfusion methods. Only under anoxiawere the membrane potential of potato tuber slices and the electricpotential difference between the parenchyma symplast and xylem(Vpx) of Vigna hypocotyl segments depolarized markedly (46 mVand 42 mV/log[K+]e unit, respectively) with increasing [K+]eabove the critical values. The electric potential differencebetween the parenchyma symplast and organ surface (Vps of thehypocotyl segments remained nearly unchanged up to 30 mEq [K+]e.Under highly aerobic conditions the membrane potentials wererelatively independent of [K+]e except at very high K+ concentrations.Vps showed even hyperpolarization with the increasing KCl concentrationin the perfusion solution that is not in direct contact withthe surface membrane of the parenchyma symplast. The respiration-dependentelectrogenic components of the membrane potentials regularlyincreased with the increasing [K+]e. A voltage-dependent homeostaticcontrol of membrane potential is discussed. We studied the course of early leaf area expansion and specificleaf area (SLA) in potato (Solanum tuberosum L.) and wheat (Triticumaestivum L.) genotypes and tested whether air temperature explainsdifferences in these courses within different environments.Such knowledge can be used to improve crop growth modelling.The relative rate of leaf area expansion (RL) of potato andwheat decreased with thermal time, but was nearly linear upto a leaf area index (L) of 1.0. TheRL (L < 1; mean: 17.9x 10-3°C-1 d-1) of potato showed an interaction betweengenotype and environment, and varied with year. TheRL (L <1; mean: 7.1 x 10-3°C-1 d-1) of winter wheat was lower thanthat of spring wheat (mean: 10.9 x 10-3°C-1 d-1), and bothvaried with year. SLAof potato increased nearly linearly withthermal time from 5 to 15 m2 kg-1at 50% emergence, to 20 to25 m2 kg-1at 155°Cd, and then decreased slightly. The SLAofboth winter and spring wheat began at 16 to 23 m2 kg-1and inmost cases increased slightly with thermal time. In potato,regression parameters of SLAwith thermal time were affectedby environment (management conditions and year) and genotype;in wheat they were affected by environment (year and site).Treatment effects on RLof potato were not correlated with thoseon SLA , and were only partly correlated for wheat. Thereforewe conclude that the early foliar expansion of potato is associatedwith a strong increase in SLA , and not so for wheat.
