Bacterial productivity in ponds used for culture of penaeid prawns   总被引:6,自引:0,他引:6
The quantitative role of bacteria in the carbon cycle of ponds used for culture of penaeid prawns has been studied. Bacterial biomass was measured using epifluorescence microscopy and muramic acid determinations. Bacterial growth rates were estimated from the rate of tritiated thymidine incorporation into DNA. In the water column, bacterial numbers ranged from 8.3×109 1–1 to 2.57×1010 1–1 and production ranged from 0.43 to 2.10 mg Cl–1 d–1. In the 0–10 mm zone in sediments, bacterial biomass was 1.4 to 5.8 g C m–2 and production was 250 to 500 mg C m–2 d–1. The results suggested that most organic matter being supplied to the ponds as feed for the prawns was actually being utilized by the bacteria. When the density of meiofauna increased after chicken manure was added, bacterial biomass decreased and growth rates increased.  相似文献

1. The microbial metabolism of organic matter in rivers has received little study compared with that of small streams. Therefore, we investigated the rate and location of bacterial production in a sixth‐order lowland river (Spree, Germany). To estimate the contribution of various habitats (sediments, epiphyton, and the pelagic zone) to total bacterial production, we quantified the contribution of these habitats to areal production by bacteria. 2. Large areas of the river bottom were characterized by loose and shifting sands of relatively homogenous particle size distribution. Aquatic macrophytes grew on 40% of the river bottom. Leaf areas of 2.8 m2 m?2 river bottom were found in a 6.6 km river stretch. 3. The epiphyton supported a bacterial production of 5–58 ng C cm?2 h?1. Bacterial production in the pelagic zone was 0.9–3.9 μg C L?1 h?1, and abundance was 4.0–7.8 × 109 cells L?1. Bacterial production in the uppermost 2 cm of sediments ranged from 1 to 8 μg C cm?3 h?1, and abundance from 0.84 to 6.7 × 109 cells cm?3. Bacteria were larger and more active in sediments than in the pelagic zone. 4. In spite of relatively low macrophyte abundance, areal production by bacteria in the pelagic zone was only slightly higher than in the epiphyton. Bacterial biomass in the uppermost 2 cm of sediments exceeded pelagic biomass by factors of 6–22, and sedimentary bacterial production was 17–35 times higher than in the overlying water column. 5. On a square meter basis, total bacterial production in the Spree was clearly higher than primary productivity. Thus, the lowland river Spree is a heterotrophic system with benthic processes dominating. Therefore, sedimentary and epiphytic bacterial productivity form important components of ecosystem carbon metabolism in rivers and shallow lakes. 6. The sediments are focal sites of microbial degradation of organic carbon in a sand‐bottomed lowland river. The presence of a lowland river section within a river continuum probably greatly changes the geochemical fluxes within the river network. This implies that current concepts of longitudinal biogeochemical relationships within river systems have to be revised.  相似文献

Phospholipid analyses were performed on water column particulate and sediment samples from Ace Lake, a meromictic lake in the Vestfold Hills, Antarctica, to estimate the viable microbial biomass and community structure in the lake. In the water column, methanogenic bacterial phospholipids were present below 17 m in depth at concentrations which converted to a biomass of between 1 and 7×108 cells/liter. Methanogenic biomass in the sediment ranged from 17.7×109 cells/g dry weight of sediment at the surface to 0.1×109 cells/g dry weight at 2 m in depth. This relatively high methanogenic biomass implies that current microbial degradation of organic carbon in Ace Lake sediments may occur at extremely slow rates. Total microbial biomass increased from 4.4×108 cells/ liter at 2 m in depth to 19.4×108 cells/liter at 23 m, near the bottom of the water column. Total nonarchaebacterial biomass decreased from 4.2 ×109 cells/g dry weight in the surface sediment (1/4 the biomass of methanogens) to 0.06×108 cells/g dry weight at 2 m in depth in the sediment. Phospholipid fatty acid profiles showed that microeukaryotes were the major microbial group present in the oxylimnion of the lake, while bacteria dominated the lower, anoxic zone. Sulfate-reducing bacteria (SRB) comprised 25% of the microbial population at 23 m in depth in the water column particulates and were present in the surface sediment but to a lesser extent. Biomass estimates and community structure of the Ace Lake eco-system are discussed in relation to previously measured metabolic rates for this and other antarctic and temperate ecosystems. This is the first instance, to our knowledge, in which the viable biomass of methanogenic and SRB have been estimated for an antarctic microbial community.  相似文献

Microbial biomass production has been measured to investigate the contribution of planktonic bacteria to fluxations in dissolved organic matter in marine and freshwater environments, but little is known about biomass production of thermophiles inhabiting geothermal and hydrothermal regions. The biomass production of thermophiles inhabiting an 85°C geothermal pool was measured by in situ cultivation using diffusion chambers. The thermophiles' growth rates ranged from 0.43 to 0.82 day?1, similar to those of planktonic bacteria in marine and freshwater habitats. Biomass production was estimated based on cellular carbon content measured directly from the thermophiles inhabiting the geothermal pool, which ranged from 5.0 to 6.1 μg C l?1 h?1. This production was 2–75 times higher than that of planktonic bacteria in other habitats, because the cellular carbon content of the thermophiles was much higher. Quantitative PCR and phylogenetic analysis targeting 16S rRNA genes revealed that thermophilic H2‐oxidizing bacteria closely related to Calderobacterium and Geothermobacterium were dominant in the geothermal pool. Chemical analysis showed the presence of H2 in gases bubbling from the bottom of the geothermal pool. These results strongly suggested that H2 plays an important role as a primary energy source of thermophiles in the geothermal pool.  相似文献

The composition and spatial distribution patterns of the macrobenthic faunal assemblages of an 8-ha tidal impoundment operated as a traditional shrimp pond at the Mai Po Marshes Nature Reserve, Hong Kong, were studied in relation to temporal changes in local environmental conditions. Species richness, abundance and biomass of macrobenthos across 5 different sub-habitats (seaward, middle, and landward parts of open water unvegetated areas, and Phragmites- and Kandelia-dominated, vegetated areas) within the pond were examined bimonthly between January 1997 and January 1998. Grab samples were collected randomly within the sub-habitats. Key physical environmental parameters of the sampling sites were also measured. A total of 46 species of macrobenthos was recorded: 11 polychaetes, 11 molluscs, 13 crustaceans and 11 insects. Mean species density in the five sub-habitats ranged from 0 to 3907 indm–2, with mean biomass ranging from 0 to 96.9 gm–2. The macrobenthos showed spatial and temporal differences among the sub-habitats and across sampling times. Species abundances of Polychaeta, Mollusca and Crustacea were significantly higher in the three open water areas than in the two vegetated (Phragmites- and Kandelia-dominated) areas, with an inverse pattern for Insecta. There were no clear temporal patterns although abundance and biomass generally increased in the cooler months. Results of a canonical correspondence analysis suggest that macrobenthic species richness, abundance and biomass in the open areas were positively correlated with salinity, while water depth, dissolved oxygen and sediment organic matter content had little relationship with the macrobenthic assemblage parameters. Ordination by multi-dimensional scaling suggests that different habitats supported distinct macrobenthic assemblages. The macrobenthic assemblage in the tidal pond was less species rich but denser than those in the neighboring tidal mangrove and mudflat, suggesting that conversion of these areas into extensively managed tidal aquaculture ponds results in reduced species richness in tropical mangrove habitats.  相似文献

Biomass, production and heterotrophic activity of bacterioplankton were determined for two weeks in the Great Astrolabe Reef lagoon, Fiji. Bacterial and Bacterial activities were distributed homogeneously throughout the water column (20 to 40 m deep) and varied little from site to site inside the lagoon. Bacterioplankton biomass and production also varied little over a diel period with coefficients of variation of 9 and 22%, respectively. On average, over the whole study, bacterial abundance was 0.77×109 cells l-1 and bacterial production averaged 0.36 μg-at. C l-1 d-1. Bacterial abundance and production were greater in the lagoon than in oceanic waters. Attachment to particles seems to provide an advantage for bacterioplankton growth because specific growth rates for attached bacterioplankton were, on average, significantly greater than that of the free community. Growth efficiency, determined by correlating the net increase of bacterial biomass and the net decrease of dissolved organic carbon (DOC) in dilution cultures, was very low (average 6.6%). Using carbon growth efficiency and bacterial production rates, heterotrophic activity was estimated to average 5.4 μg-at. C l-1 d-1. The turn-over rate of DOC (average 114 μg-at. C l-1) due to bacterial consumption was estimated to be 0.048 d-1 during the period of study. Accepted: 25 July 1998  相似文献

SUMMARY.
  • 1 The microbial heterotrophic utilization of dissolved organic matter (DOM) was determined experimentally in microcosms using stream water and stream-bed sediments from a third order reach of White Clay Creek (Pennsylvania, U.S.A.).
  • 2 Sources of DOM for the experiments included White Clay Creek water at baseflow and stormflow and cold water extracts of jewel weed (Impatiens capensis L.) and spicebush (Linderu henzoin (L.) Blume).
  • 3 The heterotrophic activity of the sediments was measured as uptake of the following: dissolved organic carbon (DOC), molecular weight fractions within the DOC pool, carbohydrates, amino acids and peptides, phenolics, and dissolved oxygen (DO), all in the overlying water.
  • 4 Concentrations of adenosine triphosphate (ATP), and direct microscopic counts of bacteria were used to estimate bacterial biomass in the surface sediments.
  • 5 The microcosm experiments showed that specific DOC molecular size classes and DOM functional groups were selectively removed from solution, exposure to one DOM source affected responses to a different DOM source and certain DOM sources were more readily utilized than others.
  • 6 Continued exposure to a DOM source increased microbial heterotrophic activity (a condition which persisted even after removal of the DOM source for several days).
  • 7 Rates of biotic DOC uptake ranged from 3.6 to 242.8 mg Cm-2h-1.
  • 8 Indirect estimates of biosynthesis calculated from DOC and DO data ranged from 1.6 at baseflow and 2.6–61.2 at stormflow to as high as 192.6 mg C m-2 h-1 when the community was repeatedly exposed to enriched DOM sources.
  • 9 The mean generation times of bacteria in sediments, determined from direct microscopy data, ranged from 12.5 to 46.2 h at 15°C.

The relationships between the biochemical composition of sediment organic matter and bacteria and microphytobenthic biomass distribution, were investigated along the coast of Northern Tuscany (Tyrrhenian Sea). Organic matter appeared to be of highly refractory composition. Among the three main biochemical classes, proteins were the major component (0.96 mg g-1 sediment d.w.) followed by total carbohydrates (0.81 mg g-1 sediment d.w.) and lipids (8.1 µg g-1 sediment d.w.). Bacterial number in surface sediments (0–2 cm) ranged from 1.7 to 24.5 × 108 cells g-1 of sediment dry weight showing a strong decrease with sediment depth. In surface sediments, significant correlations were found between bacterial biomass and protein concentration. Bacterial activity (measured by the frequency of dividing cells) was significantly related to lipid concentration. Bacterial and microphytobenthic biomass accounted for 3.1 and 18.1% respectively of the sediment organic carbon. In surface sediments bacterial lipids accounted, on average, for 27 % of total lipids, whereas bacterial proteins and carbohydrates accounted for 2.5 and 0.5% of total proteins and carbohydrates, respectively. The benthic degradation process indicated that lipids were a highly degradable compound (about 35% in the top 10 cm). Carbohydrate decreased for 25.6% in the top 10 cm, whereas proteins increased with depth, thus indicating that this compound may resist to diagenetic decomposition. These data suggest that specific organic compounds need to be measured rather than bulk carbon and nitrogen measurements in order to relate microbial biomass to the quality of organic matter.  相似文献

Fluxes of oxygen, inorganic nitrogen (DIN) and denitrification (isotope pairing) were measured from January 1997 to February 1998 via intact cores incubation in a shallow brackish area within the eutrophic Valli di Comacchio (northern Adriatic coast, Italy). Rates were measured in the light and in the dark in sediments colonized by the rooted macrophyte Ruppia cirrhosa and in adjacent sediments with benthic microalgae. Ruppia biomass (25-414 g DW m− 2) exhibited a seasonal evolution whilst that of microphytobenthos (12-66 mg chl a m− 2) was more erratic. Net (NP) and gross (GP) primary productivity was 1.15 and 6.89 mol C m− 2y− 1 for bare and 25.4 and 51.7 mol C m− 2y− 1 for Ruppia vegetated sediments. Nitrogen pools in Ruppia standing stock varied from 43.6 to 631.4 (annual average 201.2) mmol N m− 2; the macrophyte N content was correlated with DIN concentration in the water column. Estimated N pool in microphytobenthos was one order of magnitude lower (from 2.4 to 14.5 mmol N m− 2, annual average 7.2). Theoretical DIN assimilation calculated from NP was 127.8 and 1112.6 mmol N m− 2y− 1 whilst that calculated from GP was 765 and 2282 mmol N m− 2y− 1 for microphytobenthos and Ruppia respectively. Measured annual fluxes of DIN were 974.6 and − 577 mmol N m− 2y− 1 in bare and Ruppia vegetated sediments meaning that the two sites were a source and sink for DIN and that from 25 to 50% of Ruppia annual DIN requirements came from the water column. During the period of this study total denitrification was lower in the macrophyte colonized (92.3 mmol N m− 2y− 1) compared to bare sediments (163.3 mmol N m− 2y− 1) as a probable consequence of higher competition between denitrifiers and phanerogams. At both sites the ratio between denitrification of water column nitrate (DW) and denitrification coupled to nitrification (DN) was >1.6 due to little oxygen penetration in reducing sediments (< 1.2 mm) and scarce nitrification activity. DW (0-35 µmol N m− 2h− 1) was significantly correlated with water column NO3−  (2-16 µM). Theoretical DIN assimilation to denitrification ratio varied from 12.0 to 24.8 for Ruppia vegetated and from 0.8 to 4.7 for unvegetated sediments. At Valle Smarlacca, Ruppia may influence nitrogen cycling by incorporating large DIN pools in biomass which is scattered in surrounding areas and fuels intense bacterial activity. With increasing anthropogenic nutrient input and insignificant organic matter export in the open sea the already severe eutrophic conditions are enhanced and may accelerate the decline of the macrophyte meadow.  相似文献

Benthic bacterial biomass supported by streamwater dissolved organic matter   总被引:5,自引:0,他引:5
Bacterial biomass in surface sediments of a headwater stream was measured as a function of dissolved organic carbon (DOC) flux and temperature. Bacterial biomass was estimated using epifluorescence microscopic counts (EMC) and ATP determinations during exposure to streamwater containing 1,788g DOC/liter and after transfer to groundwater containing 693g DOC/liter. Numbers of bacteria and ATP concentrations averaged 1.36×109 cells and 1,064 ng per gram dry sediment, respectively, under initial DOC exposure. After transfer to low DOC water, biomass estimates dropped by 53 and 55% from EMC and ATP, respectively. The decline to a new steady state occurred within 4 days from ATP assays and within 11 days from EMC measures. A 4°C difference during these exposures had little effect on generation times. The experiment indicated that 27.59 mg/hour of natural DOC supported a steady state bacterial biomass of approximately 10g C/g dry weight of sediment (from EMC determinations). Steady state bacterial biomass estimates on sediments that were previously muffled to remove organic matter were approximately 20-fold lower. The ratio of GTPATP indicated differences in physiological condition or community composition between natural and muffled sediments.  相似文献

Viruses were found to be very abundant in the top layer of the sediments of Lac Gilbert, Québec. Viruses were extracted from the sediments using pyrophosphate buffer, and viruses from the diluted extracts were pelleted onto grids and enumerated using transmission electron microscopy. Viral abundance in the sediments ranged from 6.5 × 108 to 1.83 × 1010 ml–1, which is 10- to 1,000-fold greater than the number observed in the water column. This increase corresponds well with the 100- to 1,000-fold increase in bacterial abundance in the sediments. Viral abundance differed significantly among the surface sediment samples taken at different bottom depths and among samples taken at different depths of the water column. Viral abundance also varied significantly between the oxic and anoxic zones of the water column and the sediments. The virus-to-bacteria ratio varied greatly among the different sediment sites but not among depths in the water column. Viral abundance in the water column was related to bacterial abundance and chlorophyll concentration, whereas viruses in the sediments were most abundant in sediments with high organic matter content. Elevated viral abundance and their erratic distribution in the sediments suggest that viruses might play an important role in sediment microbial dynamics. Correspondence to: Roxane Maranger  相似文献

Rates of bacterial secondary production by free-living bacterioplankton in the Okefenokee Swamp are high and comparable to reported values for a wide variety of marine and freshwater ecosystems. Bacterial production in the water column of five aquatic habitats of the Okefenokee Swamp was substantial despite the acidic (pH 3.7), low-nutrient, peat-accumulating character of the environment. Incorporation of [3H]thymidine into cold-trichloroacetic acid-insoluble material ranged from 0.03 to 2.93 nmol liter−1 day−1) and corresponded to rates of bacterial secondary production of 3.4 to 342.2 μg of carbon liter−1 day−1 (mean, 87.8 μg of carbon liter−1 day−1). Bacterial production was strongly seasonal and appeared to be coupled to annual changes in temperature and primary production. Bacterial doubling times ranged from 5 h to 15 days and were fastest during the warm months of the year, when the biomass of aquatic macrophytes was high, and slowest during the winter, when the plant biomass was reduced. The high rates of bacterial turnover in Okefenokee waters suggest that bacterial growth is an important mechanism in the transformation of dissolved organic carbon into the nutrient-rich bacterial biomass which is utilized by microconsumers.  相似文献

Biomass, species composition and production of the planktoniccommunity were investigated during the rainy season in May andJune 1999 outside Zanzibar Island, Tanzania. In general, theplankton biomass of different organisms was uniform betweendepths as well as over time. The integrated water column primaryproduction ranged from 204 to 4142 mg C m–2 day–1.Bacterial production varied between 10 and 72 mg C m–2day–1, comprising ~5% of the total bacterial standingstock. The data obtained from these experiments are summarizedin a carbon budget. At the most 77% of the total primary productionchannelled through the heterotrophic flagellates, ciliates andheterotrophic dinoflagellates to higher trophic levels. Of theestimated carbon demand for mesozooplankton, 28% could potentiallybe met by ciliates and heterotrophic dinoflagellates.  相似文献

Disposal of fecally contaminated poultry litter by land application can deliver pathogens and fecal indicator bacteria (FIB) into receiving waters via runoff. While water quality is regulated by FIB enumeration, FIB testing provides inadequate information about contamination source and health risk. This microbial source tracking (MST) study compared the persistence of the Brevibacterium sp. strain LA35 16S rRNA gene (marker) for poultry litter with that of pathogens and FIB under outdoor, environmentally relevant conditions in freshwater, marine water, and sediments over 7 days. Salmonella enterica, Campylobacter jejuni, Campylobacter coli, Bacteroidales, and LA35 were enumerated by quantitative PCR (qPCR), and Enterococcus spp. and E. coli were quantified by culture and qPCR. Unlike the other bacteria, C. jejuni was not detectable after 48 h. Bacterial levels in the water column consistently declined over time and were highly correlated among species. Survival in sediments ranged from a slow decrease over time to growth, particularly in marine microcosms and for Bacteroidales. S. enterica also grew in marine sediments. Linear decay rates in water (k) ranged from −0.17 day−1 for LA35 to −3.12 day−1 for C. coli. LA35 levels correlated well with those of other bacteria in the water column but not in sediments. These observations suggest that, particularly in the water column, the fate of LA35 in aquatic environments is similar to that of FIB, C. coli, and Salmonella, supporting the hypothesis that the LA35 marker gene can be a useful tool for evaluating the impact of poultry litter on water quality and human health risk.  相似文献

Inorganic nitrogen and soluble reactive phosphate (o-P) concentrations were measured in the water of a marsh and in its interstitial water at two sites, and in the river water of a floodplain marsh of the Lower Paraná River. These values were compared with the N and P concentration in sediments and macrophyte biomass in order to assess nutrient availability, fate and storage capacity. High variability was found in the interstitital water using a 1 cm resolution device. Nitrate was never detected in the pore water. Depth averaged NH4 + concentrations in the upper 30 cm layer often ranged from N = 1.5 to 1.8 mg l-1, but showed a pronounced minimum (0.5–0.7 mg l-1), close to (March 95), or relatively soon after (May 94) the end of the macrophyte growing season. Soluble phosphate showed a large variation between P = 0.1–1.1 mg l-1 without any discernible seasonal pattern. NH4 + depletion in the pore water concentration and low N/P ratios (3.7 by weight) within the macrophyte biomass at the end of the growing period suggest that available N limits plant growth. NH4 + and o-P concentrations were 35 and 7 times higher, respectively, in the pore water than in the overlying marsh, suggesting a permanent flux of nutrients from the sediments. o-P accumulate in the marsh leading to higher concentrations than in the incoming river. NH4 + did not accumulate in the marsh, and no significant differences were observed between the river and the marsh water, while the NO3 - contributed by the river water was depleted within the marsh, caused probably by coupled nitrification-denitrification at the sediment–water interface. Although an order of magnitude smaller, the pore water pool can supply enough nutrients to build up the macrophyte biomass pool, but only if a fast turnover is attained. The Paraná floodplain marsh retains a large amount of nutrients being stored mainly in the sediment compartment.  相似文献

1. Phenotypic plasticity in resource allocation by Vallisneria natans was investigated in a greenhouse experiment, using three types of sediment [sandy loam, clay, and a 50 : 50 (by volume) mixture of the two sediments] and two levels of water‐column nutrient. The clay was collected from a highly eutrophic lake in Jiangsu Province, China, and the N and P concentrations applied in nutrient media were at the upper limits observed in most lakes of China. 2. Growth and biomass allocation were significantly affected by sediment type, rather than water‐column nutrients. Plant growth in clay and the mixture were similar, and 2.4–3.4 times higher than that in sandy loam. Compared with the plants grown in clay or the mixed sediments, the plants grown in sandy loam allocated relatively more biomass to root (11–17% versus 7–8% of total biomass), and relatively less to leaf (76–82% versus 86–87% of total biomass). Plastic variations in root area were induced by sediment type alone (P < 0.05), whereas the impacts of sediment type and water‐column nutrients on leaf area were insignificant (P > 0.05). 3. Plant N and P concentrations were significantly affected by both sediment type and water‐column nutrients. Increased nutrient availability in the water column enhanced plant N concentration by 3.5–20.2%, and plant P concentration by 19.1–25.8%. 4. Biomass accumulation and plant nutrient concentration in plants grown in different sediment types and water‐column nutrients indicate that sediment type had more significant impacts on growth and N and P concentrations of V. natans than did water‐column nutrients. Changes in phenotype are a functional response to nutrient availability in sediment, rather than to water‐column nutrients.  相似文献

Dynamics of bacterioplankton in a mesotrophic French reservoir (Pareloup)   总被引:1,自引:1,他引:0
Bacterioplankton abundance, biomass and production were studied at a central station (35 m depth) from April 1987 to September 1988 in a mesotrophic reservoir. Bacterial production was calculated by the (3H) thymidine method. For the water column, integrated estimates of bacterioplankton abundance ranged from 2.3 109 to 4.6 109 cells l–1, and carbon biomass from 0.037 to 0.068 mg C l–1; the thymidine incorporation rates ranged from 0.8 to 17.2 picomoles l–1 h–1, leading to net bacterial production estimates of less than 0.7 µg C l–1 d–1 in winter to 18 µg C l–1 d–1 in summer. About 55% of the production occurred in the euphotic layers.Over the year, the bacterial carbon requirement represented 90% of the autotrophic production for the whole lake. It was five times lower than autotrophic production in spring, but twice as high in summer. This important temporal lack of balance suggests that not all the spring primary production products are consumed immediately and/or that other carbon sources probably support bacterial growth in summer.  相似文献

The water chemistry, structure of sediment grain size, algal biomass, and patterns of nine bacteria physiological groups were studied monthly from May to October 1991 in an adjacent stream, ecotone, and reservoir zones (southern Poland). The distribution of five particulate organic carbon fractions was significantly different among the zones. Biomass of benthic algae demonstrated a maximum in the ecotone (22.74 g C m–2). Most of the bacterial groups were more numerous in the stream sediments where aerobic heterotrophs prevailed (1.2–6.5 × 1011 cells m–2). The significant differences in density were found for anaerobic heterotrophs, anaerobic nitrogen fixing, amylolytic, and cellulolytic bacteria among the zones. The small density of bacteria in the ecotone was probably an effect of the rapid water fluctuations, accompanied by movement of fine sediments or/and an inhibitory effect of compounds secreted by the periphyton. The principal component analysis showed that the ecotone zone was more similar to the reservoir open water than to the stream.  相似文献

Bacterial biomass, metabolic condition, and activity were measured over a 16-month period in the surface sediments of the following four field sites with differing dissolved organic matter regimes: a woodlot spring seep, a meadow spring seep, a second-order stream, and a third-order stream. Total bacterial biomass was measured by lipid phosphate and epifluorescence microscopic counts (EMC), and viable biomass was measured by 14C most probable number, EMC with 2-(p-iodophenyl)-3-(p-nitrophenyl)-5-phenyl tetrazolium chloride reduction, and ATP. Bacterial metabolic condition was determined from the percentage of respiring cells, poly-β-hydroxybutyrate concentrations, and adenylate energy charge. Activity measures included 14C-lipid synthesis, 32P-phospholipid synthesis, the rate of uptake of algal lysate dissolved organic carbon, and respiration, from which biosynthesis was calculated (dissolved organic carbon uptake corrected for respiration). Total bacterial biomass (from EMC) ranged from 0.012 to 0.354 μg of C/mg of dry sediment and was usually lowest in the third-order stream. The percentage of cells respiring was less than 25% at all sites, indicating that most bacteria were dormant or dead. Adenylate energy charge was measured only in the third-order stream and was uniformly low. Poly-β-hydroxybutyrate concentrations were greater in the woodlot spring seep than in the second- and third-order streams. Uptake of algal lysate dissolved organic carbon ranged from undetectable levels to 166 mg of C · m−2 · h−1. Little community respiration could be attributed to algal lysate metabolism. Phospholipid synthesis ranged from 0.006 to 0.354 pmol · mg of dry sediment−1 · h−1. Phospholipid synthesis rates were used to estimate bacterial turnover at the study sites. An estimated 375 bacterial generations per year were produced in the woodlot spring seep, and 67 per year were produced in the third-order stream.  相似文献

It is generally known that the water quality of shallow lakes can be influenced significantly by marginal wetlands. In order to study the efficacy of constructed littoral wetlands in the IJsselmeer area (The Netherlands) for water quality improvement, a field survey was carried out in 2003. Vegetation, soil, pore water and surface water characteristics were measured in spring and summer in two types of littoral zones: natural and constructed for 8–16 years. The study showed that constructed wetlands perform well and are suitable to enlarge the vegetated littoral zone in the IJsselmeer area. In both natural and constructed sites vegetation biomass varied between 2,200 g m−2 for helophyte vegetation and 1,300 g m−2 for low herbaceous vegetation. Nutrient concentrations in the pore water of constructed sites tended to be higher than in natural sites. and concentrations in pore water were much lower when vegetation was present, probably as a result of plant uptake. The N and P accumulation rate in the soil of constructed wetlands was 20 g N m−2 y−1 and 3 g P m−2 y−1 in vegetated plots; without vegetation the rate was much lower (8 g N m−2 y−1 and 1.8 g P m−2 y−1). We conclude that concerning their effect on water quality, constructed sites may replace natural sites, at least after 8–16 years. Principal component analysis showed a relationship between vegetation biomass and flooding, and nutrient concentrations in soil and pore water. Biomass was negatively correlated with extractable nutrients and positively with soil total N and P content. Flooding duration was negatively related to pore water salinity and positively to pore water nutrients. Due to their high biomass, helophyte stands retained significantly more nutrients than low pioneer vegetation and are therefore more suitable for improving water quality. Handling editor: S.
