The principal forms of amino nitrogen transported in xylem were studied in nodulated and non-nodulated peanut (Arachis hypogaea L.). In symbiotic plants, asparagine and the nonprotein amino acid, 4-methyleneglutamine, were identified as the major components of xylem exudate collected from root systems decapitated below the lowest nodule or above the nodulated zone. Sap bleeding from detached nodules carried 80% of its nitrogen as asparagine and less than 1% as 4-methyleneglutamine. Pulse-feeding nodulated roots with 15N2 gas showed asparagine to be the principal nitrogen product exported from N2-fixing nodules. Maintaining root systems in an N2-deficient (argon:oxygen, 80:20, v/v) atmosphere for 3 days greatly depleted asparagine levels in nodules. 4-Methyleneglutamine represented 73% of the total amino nitrogen in the xylem sap of non-nodulated plants grown on nitrogen-free nutrients, but relative levels of this compound decreased and asparagine increased when nitrate was supplied. The presence of 4-methyleneglutamine in xylem exudate did not appear to be associated with either N2 fixation or nitrate assimilation, and an origin from cotyledon nitrogen was suggested from study of changes in amount of the compound in tissue amino acid pools and in root bleeding xylem sap following germination. Changes in xylem sap composition were studied in nodulated plants receiving a range of levels of 15N-nitrate, and a 15N dilution technique was used to determine the proportions of accumulated plant nitrogen derived from N2 or fed nitrate. The abundance of asparagine in xylem sap and the ratio of asparagine:nitrate fell, while the ratio of nitrate:total amino acid rose as plants derived less of their organic nitrogen from N2. Assays based on xylem sap composition are suggested as a means of determining the relative extents to which N2 and nitrate are being used in peanuts.  相似文献

Xylem sap composition was examined in nodulated and nonnodulated cowpea (Vigna unguiculata [L.] Walp.) plants receiving a range of levels of NO3 and in eight other ureide-forming legumes utilizing NO3 or N2 as sole source of nitrogen. A 15N dilution technique determined the proportions of plant nitrogen derived from N2 in the nodulated cowpeas fed NO3. Xylem sap composition of NO3-fed, nodulated cowpea varied predictably with the relative extents to which N2 and NO3 were being utilized. The ratios of asparagine to glutamine (N/N) and of NO3 to ureide (N/N) in xylem sap increased with increasing dependence on NO3 whereas per cent of xylem nitrogen as ureide and the ratio of ureide plus glutamine to asparagine plus NO3 (N/N) in xylem sap increased with increasing dependence on N2 fixation. The amounts of NO3 and ureides stored in leaflets, stems plus petioles, and roots of cowpea varied in a complex manner with level of NO3 and the presence or absence of N2 fixation. All species showed higher proportions of organic nitrogen as ureide and several-fold lower ratios of asparagine to glutamine in their xylem sap when relying on N2 than when utilizing NO3. In nodulated (minus nitrate) cowpea and mung bean (Vigna radiata [L.] Wilczek) the percentage of xylem nitrogen as ureide remained constant during growth but the ratio of asparagine to glutamine varied considerably. The biochemical significance of the above differences in xylem sap composition was discussed.  相似文献

It is well established that nitrate is a potent inhibitor of nodulation and nitrogen fixation in legumes. The objective of this study was to demonstrate the relative insensitivity of these processes to nitrate with Calopogonium mucunoides, a tropical South American perennial legume, native to the cerrado (savannah) region. It was found that nodule number was reduced by about half in the presence of high levels of nitrate (15 mM) but nodule growth (total nodule mass per plant) and nitrogen fixation (acetylene reduction activity and xylem sap ureide levels) were not affected. Other sources of N (ammonium and urea) were also without effect at these concentrations. At even higher concentrations (30 mM), nitrate did promote significant inhibition (ca. 50%) of acetylene reduction activity, but no significant reduction in xylem sap ureides was found. The extraordinary insensitivity of nodulation and N2 fixation of C. mucunoides to nitrate suggests that this species should be useful in studies aimed at elucidating the mechanisms of nitrate inhibition of these processes.  相似文献

The composition of amino acids was determined in the xylem andphloem sap of symbiotic lupins grown under a variety of treatmentsdesigned to alter the rate of nitrogen fixation. Asparaginewas the major amino acid in both xylem and phloem with glutamine,glutamate and aspartate also major components. GABA had a highconcentration in the xylem while valine was a major componentin the phloem. Exposure to combined nitrogen in the form ofeither ammonium or nitrate caused a reduction in specific nitrogenaseactivity and was associated with subsequent changes in bothof the translocated saps. Inhibiting nitrogen fixation by exposingnodules to oxygen produced a lower amide to amine ratio in thexylem sap (1.3:1) compared with control and nitrate ratios (2.6:1)and ammonium ratios (7.1:1). Similar ratios for amide aminewere also observed in the phloem sap. Labelling studies using15N2 to follow nitrogen fixation, ammonium assimilation andamino acid transport have shown rapid accumulation of labelinto glutamine with subsequent enrichment in glutamate, aspartate,alanine, and GABA. Asparagine was found in high concentrationsin nodules and became slowly enriched. Labelled nitrogen fixedand assimilated in nodules was detected 40 min later in stemxylem extracts, largely as the amides glutamine and asparagine.These experiments provide evidence that large amounts of nitrogenouscompounds are cycled through the root nodules of symbiotic plants(contributing approximately 50% of xylem N) and that differencesin the composition of the phloem sap may influence nodule growthand activity. Utilising the 15N dilution technique the relationship between the proportion of N derived from N2 fixation and relative abundance of ureides in xylem sap was evaluated for Phaseolus vulgaris L. cv. Mokcham during vegetative and reproductive development. In order to establish calibration curves for time integrated estimates of N2 fixation, plants were raised in sand culture during the dry season in northern Thailand and continuously supplied with a N-free nutrient solution or the same solution amended with 0, 3, 6 or 9 mol m–3 nitrate. Large changes in plant dependence on N2 fixation were concomitantly reflected by corresponding alterations in N solutes in xylem sap. Regression analyses of the data suggested high correlations between relative ureide content and N2 fixation, but different slopes and line intercepts indicated the requirement for the use of calibration curves established for different phases of the development of the plant. Largest age related differences were noted between vegetative and reproductive development. Judging from 95% confidence limits, utilisation of appropriate calibrations can reduce errors of the technique to close to ±5%.A second experiment, involving similarly cultivated plants exposed to different sources of mineral N, indicated an effect of ammonium on xylem sap composition. This implies that calibrations, in which N2 fixation is regulated only by applications of various concentrations of nitrate, may lead to errors in situations where a major proportion of the plant available soil nitrogen fraction is present in the form of ammonium.  相似文献

Thein vivo 14CO2 fixation assay and xylem sap analysis showed that inSesbania rostrata the transport of fixed nitrogen from stem nodules was in the amide form. The majority of nitrogen was transported as asparagine. The close relationship between nodule phosphoenolpyruvate carboxylase and nitrogenase activities suggested that nodule CO2 fixation contributed directly to nitrogen assimilation in stem nodules ofS. rostrata.  相似文献

Rapid direct conversion of exogenously supplied [14C]aspartate to [14C] asparagine and to tricarboxylic cycle acids was observed in alfalfa (Medicago sativa L.) nodules. Aspartate aminotransferase activity readily converted carbon from exogenously applied [14C]aspartate into the tricarboxylic acid cycle with subsequent conversion to the organic acids malate, succinate, and fumarate. Aminooxyacetate, an inhibitor of aminotransferase activity, reduced the flow of carbon from [14C]aspartate into tricarboxylic cycle acids and decreased 14CO2 evolution by 99%. Concurrently, maximum conversion of aspartate to asparagine was observed in aminooxyacetate treated nodules (30 nanomoles asparagine per gram fresh weight per hour. Metabolism of [14C]aspartate and distribution of nodulefixed 14CO2 suggest that two pools of aspartate occur in alfalfa nodules: (a) one involved in asparagine biosynthesis, and (b) another supplying a malate/aspartate shuttle. Conversion of [14C]aspartate to [14C]asparagine was not inhibited by methionine sulfoximine, a glutamine synthetase inhibitor, or azaserine, a glutmate synthetase, inhibitor. The data did not indicate that asparagine biosynthesis in alfalfa nodules has an absolute requirement for glutamine. Radioactivity in the xylem sap, derived from nodule 14CO2 fixation, was markedly decreased by treating nodulated roots with aminooxyacetate, methionine sulfoximine, and azaserine. Inhibitors decreased the [14C]aspartate and [14]asparagine content of xylem sap by greater than 80% and reduced the total amino nitrogen content of xylem sap (including nonradiolabeled amino acids) by 50 to 80%. Asparagine biosynthesis in alfalfa nodules and transport in xylem sap are dependent upon continued aminotransferase activity and an uninterrupted assimilation of ammonia via the glutamine synthetase/glutamate synthase pathway. Continued assimilation of ammonia apparently appears crucial to continued root nodule CO2 fixation in alfalfa.  相似文献

Nodulated and denodulated roots of adzuki bean (Vigna angularis), soybean (Glycine max), and alfalfa (Medicago sativa) were exposed to 14CO2 to investigate the contribution of nodule CO2 fixation to assimilation and transport of fixed nitrogen. The distribution of radioactivity in xylem sap and partitioning of carbon fixed by nodules to the whole plant were measured. Radioactivity in the xylem sap of nodulated soybean and adzuki bean was located primarily (70 to 87%) in the acid fraction while the basic (amino acid) fraction contained 10 to 22%. In contrast, radioactivity in the xylem sap of nodulated alfalfa was primarily in amino acids with about 20% in organic acids. Total ureide concentration was 8.1, 4.7, and 0.0 micromoles per milliliter xylem sap for soybean, adzuki bean, and alfalfa, respectively. While the major nitrogen transport products in soybeans and adzuki beans are ureides, this class of metabolites contained less than 20% of the total radioactivity. When nodules of plants were removed, radioactivity in xylem sap decreased by 90% or more. Pulse-chase experiments indicated that CO2 fixed by nodules was rapidly transported to shoots and incorporated into acid stable constituents. The data are consistent with a role for nodule CO2 fixation providing carbon for the assimilation and transport of fixed nitrogen in amide-based legumes. In contrast, CO2 fixation by nodules of ureide transporting legumes appears to contribute little to assimilation and transport of fixed nitrogen.  相似文献

We investigated the synthesis and translocation of amino compounds in Parasponia, a genus of the Ulmaceae that represents the only non-legumes known to form a root nodule symbiosis with rhizohia. In the xylem sap of P. andersonii we identified asparagine. aspartate. glutamine, glutamated significant quantities of a non-protein amino acid. 4-methylglutamte(2-amino-4-methylpentanedioic acid). This identification was confirmed by two methods, capillary gas chromatography (GC) electron ionization (El) mass spectrometry (MS) and reverse phase high pressure liquid chromatography (HPLC) analysis of derivatized compounds. In leaf, root and nodule samples from P. andersonii and P. parviflora we also identified the related compounds 4-methyleneglutamate and 4-methyleneglulamine. Using 15N2 labelling and GC-Ms analysis of root nodule extracts we followed N2 fixation and ammonia assimilation in P. andersonii root nodules and observed Label initially in glutamine and subsequently in glutamate, suggesting operation of the glutamine synthetase/glutamine:2-oxoglutarate aminotransferase (GS/GOGAT) pathway. Importantly, we observed the incorporation of significant quantities of 15N into 4-methylglutamate in nodules, demonstrating the de nova synthesis of this non protein amino acid and suggesting a role in the translation of N in symbioticParasponia.  相似文献

Summary Comparisons were made of the levels of various solutes in xylem (tracheal) sap and fruit tip phloem sap of Lupinus albus (L.) and Spartium junceum (L.). Sucrose was present at high concentration (up to 220 mg ml-1) in phloem but was absent from xylem whereas nitrate was detected in xylem (up to 0.14 mg ml-1) but not in phloem. Total amino acids reached 0.5–2.5 mg ml-1 (in xylem) versus 16–40 mg ml-1 in phloem. Phloem: xylem concentration ratios for mineral nutrients (K, Na, Mg, Ca, Fe, Zn, Mn, Cu) spanned the range 0.7 to 20, the ratios generally reflecting an element's phloem mobility and its availability to the xylem from the roots.The accessibility of nitrate to xylem and phloem was studied in Lupinus. Increasing the nitrate supply to roots from 100 to 1000 mg NO3–Nl-1 increased nitrate spill over into xylem, but nitrate always failed to appear in phloem. However, phloem loading of small amounts of nitrate was induced by feeding 750 or 1000 mg NO3–Nl-1 directly to cut shoots via the transpiration stream. Transfer of reduced nitrogen to phloem was demonstrated by feeding 15NO3 to shoots and recovering 15N-enriched amides and amino acids in phloem sap. Increased nitrate supply to roots led to increased amino acid levels in xylem and phloem but did not alter markedly the balance between individual amino acids.The fate of xylem-fed 14C-labelled asparagine, glutamine and aspartic acid and of photosynthetically fed 14CO2 was studied in Spartium, with reference to phloem transport to seeds. Substantial fractions of the 14C of all sources appeared in non-amino compounds. [14C]asparagine passed largely in unchanged form to the phloem whereas the 14C from aspartic acid or glutamine appeared in phloem attached to other amino acids (e.g. asparagine and glutamic acid). Serine, asparagine and glutamine were the main amino compounds labelled in phloem sap after feeding 14CO2. The wide distribution of 14C amongst free and bound amino acids of seeds suggested that extensive metabolism of phloem-borne solutes occurred in the fruits.  相似文献

During vegetative regrowth of Medicago sativa L., soil N, symbiotically fixed N2 and N reserves meet the nitrogen requirements for shoot regrowth. Experiments with nodulated or non-nodulated plants were carried out to investigate the changes in N flows originating from the different N sources and in xylem transport of amino acids during regrowth. Exogenous N uptake, N2 fixation and endogenous N remobilization were estimated by 15N labelling and amino acids in xylem sap were analysed. Removal of shoots resulted in great declines of exogenous N flows derived either from N2 or from NH4NO3 during the first week of regrowth, thereafter recovery increased linearly. Mineral N uptake as well as N2 fixation occurred mainly between the 10th and 18th day after removal of shoots while exogenous N assimilation in intact plants remained at a steady level. Nitrogen remobilization rates in defoliated plants increased by at least three to five-fold, especially during the first 10 days following shoot removal. Compared to control plants, contents of amino acids in xylem sap, during the first 10 days of regrowth, were reduced by about 72% and 82% in NH4NO3 grown and in N2 fixing plants, respectively. Asparagine was the main amino acid transported in xylem sap of both treated plants. Its relative contents during this period significantly decreased from 75% to 59% and from 67% to 36% respectively in non-nodulated plants and in nodulated ones. This decline was accompanied by compensatory increase in the relative contents of aspartate and glutamine.  相似文献

Common bean,Phaseolus vulgaris L., is known to be ‘inefficient’ in nodulation and N2 fixation although it responds to applied nitrogen. An experiment was conducted to identify and to characterize bean cultivars nodulating in the presence of a high level of nitrogen. Sixteen cultivars and a check for inefficient nodulation, OAC Seaforth, were inoculated and grown for 40 days in replicated pots supplied with zero, 3.5 and 10.5 mM combined nitrogen as NO 3 and NH 4 + . Seven traits relating to nodulation and N2 fixation were all significantly affected by N level (N), cultivar (Cv) and N × Cv interactions (except for root dry weight), indicating that cultivars responded differently to the N treatments. Total dry weight (W) and shoot to root ratio (S/R) increased with the increased N levels. Nodule dry weight (Wn), visual nodulation score (Nv) and nodulation index (Nx) decreased as the N increased. Percent N and N content per plant increased with the increased N level. Plant weight (W) was positively correlated with Wn, Nv and N content and negatively correlated with %N. Nodulation score was positively associated with Wn and plant N content. Genotypes superior in nodulation and N2 fixation in the presence of N were identified. Cultivars Italian Barlotti, California Light Red Kidney, Kentucky Wonder A and Pueblo 152 were selected for further testing and use in improving the nitrate tolerant nodulating characteristic of bean.  相似文献

The use of the relative ureide content of xylem sap [(ureide-N/total N) × 100] as an indicator of N2 fixation in soybeans (Merr.) was examined under greenhouse conditions. Acetylene treatments to inhibit N2 fixation were imposed upon the root systems of plants totally dependent upon N2 fixation as their source of N and of plants dependent upon both N2 fixation and uptake of exogenous nitrate. Significant decreases in the total N concentration of xylem sap from plants of the former type were observed, but no significant decrease was observed in the total N concentration of sap from the latter type of plants. In both types of plants, acetylene treatment caused significant decreases in the relative ureide content of xylem sap. The results provided further support for a link between the presence of ureides in the xylem and the occurrence of N2 fixation in soybeans. The relative ureide content of xylem sap from plants totally dependent upon N2 fixation was shown to be insensitive to changes in the exudation rate and total N concentration of xylem sap brought about by diurnal changes in environmental factors. There was little evidence of soybean cultivars or nodulating strains affecting the relative ureide content of xylem sap. `Ransom' soybeans nodulated with Rhizobium japonicum strain USDA 110 were grown under conditions to obtain plants exhibiting a wide range of dependency upon N2 fixation. The relative ureide content of xylem sap was shown to indicate reliably the N2 fixation of these plants during vegetative growth using a 15N method to measure N2 fixation activity. The use of the relative ureide content of xylem sap for quantification of N2 fixation in soybeans should be evaluated further.  相似文献

Experiments were conducted to characterize the distribution of N compounds in the xylem sap of nodulated and nonnodulated soybean plants through development and to determine the effects of exogenous N on the distribution of N compounds in the xylem. Xylem sap was collected from nodulated and nonnodulated greenhouse-grown soybean plants (Glycine max [L.] Merr. “Ransom”) from the vegetative phase to the pod-filling phase. The sum of the nitrogen in the amino acid, nitrate, ureide (allantoic acid and allantoin), and ammonium fractions of the sap from both types of plants agreed closely with total N as assayed by a Kjeldahl technique. Sap from nodulated plants supplied with N-free nutrient solution contained seasonal averages of 78 and 20% of the total N as ureide-N and amino acid-N, respectively. Sap from nonnodulated plants supplied with a 20 millimolar KNO3 nutrient solution contained seasonal averages of 6, 36, and 58% of total N as ureide-N, amino acid-N, and nitrate-N, respectively. Allantoic acid was the predominant ureide in the xylem sap and asparagine was the predominant amino acid. When well nodulated plants were supplied with 20 millimolar KNO3, beginning at 65 days, C2H2 reduction (N2 fixation) decreased relative to nontreated plants and there was a concomitant decrease in the ureide content of the sap. A positive correlation (r = 0.89) was found between the ureide levels in xylem sap and nodule dry weights when either exogenous nitrate-N or urea-N was supplied at 10 and 20 millimolar concentrations to inoculated plants. The results demonstrate that ureides play a dominant role in N transport in nodulated soybeans and that the synthesis of ureides is largely dependent upon nodulation and N2 fixation.  相似文献

Nitrogen-fixing activity in two nodulated African legumes, Bambaragroundnut (Vigna subterraneaL.) and Kersting's bean (MacrotylomageocarpumL.), was assessed in the presence of nitrate (NO3-)ions in the rooting medium. Nitrogenase activity was unimpairedby the supply of 5 mol m-3NO3to both species. The ureides, allantoin and allantoic acid, represented major fractions of the soluble nitrogen pool of nodulated plants of cowpea (Vigna unguiculata [L.] Walp. cv. Caloona) throughout vegetative and reproductive growth. Stem and petioles were the principal sites of ureide accumulation, especially in early fruiting.

Labeling studies using 14CO2 and 15N2 and incubation periods of 25 to 245 minutes indicated that synthesis of allantoin and allantoic acid in root nodules involved currently delivered photosynthate and recently fixed N, and that the ureides were exported from nodule to shoot via the xylem. From 60 to 80% of xylem-borne N consisted of ureides; the remainder was glutamine, asparagine, and amino acids. Allantoin predominated in the soluble N fraction of nodules and fruits, allantoin and allantoic acid were present in approximately equal proportions in xylem exudate, stems, and petioles.

Extracts of the plant tissue fraction of nitrogen-fixing cowpea nodules contained glutamate synthase (EC 2.6.1.53) and glutamine synthetase (EC 6.3.1.2), but little activity of glutamate dehydrogenase (EC 1.4.1.3). High levels of uricase (EC 1.7.3.3) and allantoinase (EC 3.5.2.5) were also detected. Allantoinase but little uricase was found in extracts of leaflets, pods, and seeds.

Balance sheets were constructed for production, storage, and utilization of ureide N during growth. Virtually all (average 92%) of the ureides exported from roots was metabolized on entering the shoot, the compounds being presumably used as N sources for protein synthesis.

A sixth treatmentcomprised uninoculated plants supplied with 8–0 mol m-31513N-nitrate. For each species, more than 75% of the nitrogenwas exported from the nodules as the amides, asparagine andglutamine. In fully symbiotic plants, the amides also dominatednitrogen transport to the shoot When N2 fixation activity wasdecreased by the addition of nitrate to the rooting medium,the N-composition of xylem exudate and stem solutes changedconsiderably. The relative concentrations of asparagine tendedto increase in the xylem whilst those of glutamine were reduced;the levels of nitrate increased in both xylem exudate and thesoluble nitrogen pool of the stem with a rise in nitrate supply.The changes in relative nitrate contents reflected generallythe contributions of root and shoot to overall nitrate reductaseactivity at the different levels of nitrate used. The relationshipsbetween the relative contents of xylary or stem nitrate andamino nitrogen and the plants' reliance on N2 fixation (determinedby the 15N isotope dilution procedure) were examined. Nitrogen fixation in nine common bean (Phaseolus vulgaris L.) lines was estimated using the 15N isotope dilution method at two locations in two seasons. In the first season at one location no N2 fixation was detected while in the second season up to 51 kg N ha–1 were estimated. There were significant differences between lines and correlations between trials were significant for the amounts of N2 fixed, but not for total shoot nitrogen. The plants that fixed the most nitrogen nodulated rapidly after germination. Differences in maximum nodule mass, but not specific nodule activity, were detected also.  相似文献

Summary The nitrogenous compounds in the xylem (bleeding) sap of lines of field-grownPhaseolus vulgaris L., known to vary in N2 fixation and yield, were measured during growth with and without N fertilizer. Forty nine-67% of the total sap N was in the form of nitrate in fertilized plants, with low amounts of the ureides, allantoin and allantoic acid (4–12%). Ureides contributed between 17 and 38% to the total sap N of non-fertilized plants, with nitrate generally comprising less than 40%. Among the nine lines grown without fertilizer there were significant differences in the % of the sap N as ureides and also in the total mol N ml–1 sap.The ASN/GLN ratio (mol/mol) was greater in sap collected from the two parental lines given fertilizer compared with non-fertilized plants. However the actual ratio varied between the parental lines. Together the two amides generally comprised between 43–62% of the amino-N in sap samples from the two parental lines with or without N fertilizer.For each N treatment (fertilized or non-fertilized) there were no obvious differences in sap composition between the high N2 fixing lines and the low N2 fixing lines. However there was generally a positive relationship between the rate of N translocation (total N concn. ml–1 sap x rate of exudation) and the ranking of the lines on the basis of higher N2 fixation rates (acetylene reduction), which was to a large extent independent of the source of N available to the plant.  相似文献

Thirty one selected bean lines were evaluated in the field for ability to support N2 fixation when intercropped with maize which received 0, 30 and 60 kg N ha–1 as ammonium sulphate. The amount of fixed N2 was estimated using the natural variation of 15N and wheat as the standard non-fixing crop. Nitrogen as low as 15 kg N ha–1 at sowing suppressed nodule weight and activity (acetylene reduction activity) but not nodule number, suggesting that the main effect of mineral N was on nodule development and function. 15N data revealed a high potential of the bean genotypes to fix N2, with the most promising ones averaging between 50–60% of seed N coming from fixation. Bean lines CNF-480, Puebla-152, Mexico-309, Negro Argel, CNF-178, Venezuela-350 and WBR22-3, WBR22-50 and WBR22-55 were ranked as good fixers.  相似文献
