用激光扫描共聚焦显微镜研究了一般公认的耳蜗传出神经递质乙酰胆碱(ACh)和三磷酸腺苷(ATP)对豚鼠耳蜗外毛细胞(OHCs)胞内游离Ca^2+浓度(Ca^2+)的作用,OHCs用Ca^2+敏感荧光染料Fluo-3着色,胞内Ca^2+的分布以细胞底部稍强。ACh在OHC底部引起Ca^2+的缓慢上长并维持在一个较高水平。ATP在整个OHC引起一个急剧的Ca^2+升高,升高幅度在OHC顶部最大。随着AT  相似文献

牛磺酸占心肌游离氨基酸的50%以上,它对心肌细胞许多依赖于Ca^2+的生理两全是现象具有明显的调节作用。牛磺酸能增加高亲和力Ca^2+转驼系统转运Ca^2+的量和速度,对要和力Ca^2+转运系统的调节作用依胞外Ca^2+浓度的不同而不同,具有稳态调节细胞内的Ca^2+作用;另外,牛磺酸尚能抑制各种原因经起的胞内Ca^2+超负荷,显示其具有细胞保护作用。  相似文献

采用荧光分光光度计法检测维甲酸(RA)、1,25(OH)2VD3及佛波酯(PMA)诱导CCL229细胞分化后[Ca2+]i变化,并观察内质网(ER)特异的Ca2+-ATPase抑制剂Thapsigargin(TG)、IP3受体抑制剂Heparin对RA诱导[Ca2+]i变化的影响,从而探讨RA诱导[Ca2+]i变化与ER的关系。结果显示:RA和1,25(OH)2VD3在数秒内引起[Ca2+]i显著升高。在EGTA和Verapamil预处理细胞条件下,TG不能抑制RA引起Ca2+从细胞内钙池中外流,RA作用后TG仍能升高[Ca2+]i。另外,Heparin也不能完全抑制RA升高[Ca2+]i。提示RA诱导大肠癌细胞升高[Ca2+]i可能通过ER上IP3敏感性和非敏感性钙池,亦可能细胞内存在除ER外对RA敏感的钙池。  相似文献

本文观察了离体成年大鼠心室肌肌质网、线粒体Ca^2+-ATP酶活性和总钙含是到及原代培养乳腺心肌细胞^46Ca摄取、活性钙调素相对含量、胞浆内游离钙浓度在不同温度热暴露40min后的变化。结果表明:热暴露后,心肌奖浆、肌质网及线粒体中的Ca^2+-ATP酶活发表 所下降,心肌肌质网及线粒体中的总钙含量亦有降低趋势,心肌细胞活性钙调素相对含量显著下降,^46Ca摄取量及胞浆内游离浓度显著升高。提示,  相似文献

利用粘附式细胞仪 (ACAS -570)结合相应的荧光探针分别测定了外源性神经酰胺诱导NIH3T3细胞凋亡时胞浆游离Ca2 水平和UVB照射NIH3T3细胞所致细胞内 pH的变化以及神经酰胺的生成对这一变化的影响。结果表明 :1.神经酰胺能够导致NIH3T3细胞胞浆游离Ca2 升高 ;胞浆Ca2 升高既来源于胞外钙 ,又来源于胞内钙池 ,但外钙内流是引起和维持胞内Ca2 处于高水平的必要条件 ;NIH3T3细胞钙池上存在着两种受体 :IP3 受体和Ryanodine受体 ,其中IP3受体较占优势。2.UVB照射导致NIH3T3细胞凋亡时胞浆碱化并持续约2小时左右恢复正常 ,pH的变化参与了细胞凋亡的过程并受到神经酰胺生成的调控。这可能是UVB照射启动了磷脂肌醇通路激活磷脂酶C ,导致神经酰胺的生成、Ca2 动员和蛋白激酶C的活化 ,从而激活Na /H 对流引起胞浆碱化。所以 ,胞浆游离Ca2 的增加和 pH的升高不是两个孤立的事件。  相似文献

参照文献报道的方法建立了测定细胞神经酰胺的激酶催化法,以该法所做的标准曲线在一定浓度范围内有很好的线性关系。以UVB照射NIH 3T3细胞,可以使细胞神经酰胺的水平在1分钟之内迅速升高1.5倍以上,基本上达到了细胞所能生成神经酰胺的最高峰值。可细胞膜的外源性神经酰胺(C2-ceramide)作用于NIH 3T3细胞后能够导致细胞的凋亡和坏死,其凋亡率在一定剂量范围内随神经酰胺浓度的增加而增加;而在  相似文献

It is well-known that pH changes can influence a lot of cellular processes. In this work, we have specifically studied the influence of alkalinization, which can be developed in spinal cord neurons during hyperventilation (respiratory alkalosis) and chronic renal failure (metabolic alkalosis) on calcium homeostasis. Application of Tyrode solution with increased pH (pH = 8.8) to secondary sensory neurons isolated from rat spinal dorsal horn induced elevation of intracellular free calcium concentration in the cytosol ([Ca2+]i) if applied after membrane depolarization. Repetitive application of alkaline solution led to disappearance of such elevations. Depletion of endoplasmic reticulum (ER) calcium stores by 30 mM caffeine almost completely blocked the effect of elevated extracellular pH. If caffeine-induced [Ca2+]i transients were evoked during alkalinization, their amplitudes were decreased by 41%. Preapplication of 500 nM ionomycin resulted in disappearance of alkalinization-induced [Ca2+]i transients, whereas prolonged applications (for 20 min) of 200 nM thapsigargin, a blocker of Ca2+ ATPase of the endoplasmic reticulum, resulted in disappearance of the rapid phase of the [Ca2+]i transients induced by alkalinization. Preapplication of the mitochondrial protonophore CCCP (10 microM) also induced changes in the alkalinization-induced calcium response--it lost its peak and was transformed into an irregular wave terminating in several seconds. The data obtained indicate that alkalinization induces an increase of [Ca2+]i level in the investigated neurons via a combined action of both intracellular Ca2+-accumulating structures--the endoplasmic reticulum and mitochondria. This suggestion was supported by morphological data that both structures in these neurons are tightly connected and may interact during release of accumulated calcium ions.  相似文献

The role of acidic intracellular calcium stores in calcium homeostasis was investigated in the Drosophila Schneider cell line 2 (S2) by means of free cytosolic calcium ([Ca2+]i) and intracellular pH (pHi) imaging together with measurements of total calcium concentrations within intracellular compartments. Both a weak base (NH4Cl, 15 mM) and a Na+/H+ ionophore (monensin, 10 microM) evoked cytosolic alkalinization followed by Ca2+ release from acidic intracellular Ca2+ stores. Pretreatment of S2 cells with either thapsigargin (1 microM), an inhibitor of endoplasmic reticulum Ca(2+)-ATPases, or with the Ca2+ ionophore ionomycin (10 microM) was without effect on the amplitude of Ca2+ release evoked by alkalinization. Application of the cholinergic agonist carbamylcholine (100 microM) to transfected S2-DM1 cells expressing a Drosophila muscarinic acetylcholine receptor (DM1) emptied the InsP3-sensitive Ca2+ store but failed to affect the amplitude of alkalinization-evoked Ca2+ release. Glycyl-L-phenylalanine-beta-naphthylamide (200 microM), a weak hydrophobic base known to permeabilize lysosomes by osmotic swelling, triggered Ca2+ release from internal stores, while application of brefeldin A (10 microM), an antibiotic which disperses the Golgi complex, resulted in a smaller increase in [Ca2+]i. These results suggest that the alkali-evoked calcium release is largely attributable to lysosomes, a conclusion that was confirmed by direct measurements of total calcium content of S2 organelles. Lysosomes and endoplasmic reticulum were the only organelles found to have concentrations of total calcium significantly higher than the cytosol. However, NH4Cl (15 mM) reduced the level of total calcium only in lysosomes. Depletion of acidic Ca2+ stores did not elicit depletion-operated Ca2+ entry. They were refilled upon re-exposure of cells to normal saline ([Ca2+]o = 2 mM), but not by thapsigargin-induced [Ca2+]i elevation in Ca(2+)-free saline.  相似文献

Mitochondria trigger apoptosis by releasing caspase activators, including cytochrome c (cytC). Here we show, using a pH-sensitive green fluorescent protein (GFP), that mitochondria-dependent apoptotic stimuli (such as Bax, staurosporine and ultraviolet irradiation) induce rapid, Bcl-2-inhibitable mitochondrial alkalinization and cytosol acidification, followed by cytC release, caspase activation and mitochondrial swelling and depolarization. These events are not induced by mitochondria-independent apoptotic stimuli, such as Fas. Activation of cytosolic caspases by cytC in vitro is minimal at neutral pH, but maximal at acidic pH, indicating that mitochondria-induced acidification of the cytosol may be important for caspase activation; this finding is supported by results obtained from cells using protonophores. Cytosol acidification and cytC release are suppressed by oligomycin, a FoF1-ATPase/H +-pump inhibitor, but not by caspase inhibitors. Ectopic expression of Bax in wild-type, but not FoF1/H+-pump-deficient, yeast cells similarly results in mitochondrial matrix alkalinization, cytosol acidification and cell death. These findings indicate that mitochondria-mediated alteration of intracellular pH may be an early event that regulates caspase activation in the mitochondrial pathway for apoptosis.  相似文献

We made use of quin2 microfluorometry to determine the effects of endothelin (ET) on cytosolic free Ca2+ concentrations [Ca2+]i) in rat aortic smooth muscle cells in primary culture. In Ca2+-containing medium, ET induced a rapid and sustained elevation of [Ca2+]i. In the latter component, in particular, the elevation of [Ca2+]i was inhibited by diltiazem. In Ca2+-free medium, ET induced a rapid and transient [Ca2+]i elevation, which was not inhibited by diltiazem. When the caffeine-sensitive intracellular Ca2+ store was practically depleted by repeated treatment with caffeine in Ca2+-free media, ET did not elevate [Ca2+]i. Thus, it was suggested that ET induces [Ca2+]i elevation not only by extracellular Ca2+-dependent, mechanisms but also by releasing Ca2+ from the intracellular store, and that the ET-sensitive Ca2+ store may overlap with the caffeine-sensitive one, in cultured vascular smooth muscle cells.  相似文献

A natural polypeptide from marine Chlamys farreri (a kind of scallop) (PCF), has been recently been found to be an effective photoprotective agent against ultraviolet rays B (UVB)-induced mitochondria damage in normal human fibroblasts. To investigate whether PCF has the antiapoptotic effect on human keratinocytes, in the present study, we established an apoptotic model on HaCaT cell line by means of UVB radiance of 30 mJ/cm(2) and compared the effect of different PCF treatments on UVB-radiated cells. Flow cytometry analyses showed that PCF treatment before UVB-irradiation inhibited UVB-induced apoptosis, the loss of mitochondrial membrane potential (Deltapsim) and the increase of free Ca(2+) level in HaCaT cells. In parallel with these results, UVB-irradiation enhanced activities of caspases-3, 8, 9, while this enhancement was inhibited by PCF treatment prior to irradiation. PCF added after irradiation neither reduced UVB-induced activities of the three caspases nor synergized the effect of pre-added PCF. Cellular ultrastructural features obtained from transmission electron microscopy further confirmed the antiapoptotic effect of PCF pre-treatment. It is concluded that the antiapoptotic effect of PCF is not therapeutic but prophylactic. Caspases-3, 8, 9, Deltapsim and calcium are involved in UVB-induced apoptosis, while prophylactic PCF inhibits apoptosis of UVB-irradiated HaCaT cells by blocking the caspases activities, the Deltapsim lost and the elevation of intracellular free Ca(2+) level.  相似文献

Addition of serum growth factors or bombesin to quiescent NIH3T3-fibroblasts leads to a simultaneous mobilization of intracellular Ca2+ and an increase in cytosolic pH which is inhibitable by dimethylamiloride. The mobilization of intracellular Ca2+ is a pH-dependent process with an optimum at pH 7.1. In quiescent cells with a pHi greater than or equal to 6.8, inhibition of the Na+/H(+)-antiporter by dimethylamiloride or reduction of extracellular Na+ attenuates the growth factor induced Ca2(+)-response. It is concluded that the growth factor induced activation of the Na+/H(+)-antiporter facilitates the mobilization of Ca2+ by shifting the internal pH towards the optimum for the Ca2(+)-release.  相似文献

UVB irradiation induces apoptosis in several cell types. However, we report here that UVB irradiation prevents induction of apoptosis in cells detached from the extracellular matrix under serum-free conditions. NIH3T3 cells cultured in bovine serum albumin-coated dishes (detached from the extracellular matrix) underwent apoptosis under serum-free conditions, which was inhibited by UVB (<0.1 J/cm(2)) irradiation, keeping suspension conditions, as determined by chromatin condensation and the appearance of a subG1 DNA fraction. Furthermore, UVB irradiation decreased caspase-3/7, -8/6, and -9 activation and eliminated loss of mitochondrial inner transmembrane potential, suggesting suppression upstream of the caspase cascade. Treatment with PI3-kinase inhibitors, wortmannin, and LY294002 partly eliminated the UV-mediated inhibition of cell death and recovered the inhibited caspase-3/7 activity. Phosphorylation of Akt was observed from 15 min after UVB irradiation. These results suggested that UVB irradiation transduced a survival signal via PI3 kinase activation and phosphorylation of Akt, and induced some apoptosis inhibition factors upstream of the caspase cascade.  相似文献

The stimulation of different cell types with growth factors is often accompanied by a rapid intracellular alkalinization. By using mitogenic lectins, cluster of differentiation (CD)2 and CD3 mAb, as stimuli, we studied early changes of the intracellular pH in the activation process of resting human PBL. We found increases in free cytoplasmic Ca2+ levels and DNA synthesis but no intracellular alkalinization in the early activation phase upon stimulation with the mitogenic lectins, Con A, and PHA. Similarly stimulation with CD3 mAb led in most instances to no detectable pH shifts. Only in 7 out of 30 experiments was CD3 mAb-induced alkalinization observed. In contrast, stimulation with mitogenic combinations of anti-CD2 mAb led in all instances to rapid and clear-cut intracellular pH shifts very similar to those observed upon stimulation with PMA. In medium lacking sodium bicarbonate the intracellular alkalinization via the CD2 structure could be blocked by the amiloride analogue 5-(N-methyl-N-isobutyl)amiloride (MIA), which indicates that this increase in pH is mediated by the amiloride-sensitive Na+/H+ antiporter. Blockade of this antiporter had no negative effect, however, on T cell proliferation as measured by thymidine incorporation. In contrast, significantly enhanced proliferation rates were observed after stimulation with mitogenic combinations of anti-CD2 antibodies in the presence of MIA. No such effect of MIA could be observed in lectin induced T cell stimulation. These findings indicate that stimulation of the Na+/H+ antiporter via the CD2 structure is neither a prerequisite for T cell proliferation nor does it promote T cell growth. It rather seems to function in a regulatory role. In its absence, superinduction of proliferation can be achieved.  相似文献
