Little is known about the permeability of plant cuticles to ionic molecules with hydration shells that render them lipid insoluble and limit their diffusion to narrow aqueous pores. Therefore, the permeation of cuticular membranes to ionised calcium salts with anhydrous molecular weights ranging from 111 to 755 g mol–1 was studied. Penetration was a first-order process and rate constants (k) (proportional to permeability) decreased exponentially with molecular weight. Plots of log k vs. molecular weight had slopes of –2.11×10–3 and –2.80×10–3, respectively, depending on the year in which the cuticular membranes were isolated. This corresponds to decreases in permeability by factors of about 7 to 13 when molecular weight increased from 100 to 500 g mol–1. This size selectivity is small compared to the dependence on molecular weight of solute mobility in Populus cuticles. A decrease in mobility of neutral molecules by more than 3 orders of magnitude has been reported [A. Buchholz et al. (1998) Planta 206:322–328] for the same range of molecular weights. Hence, discrimination of large ionic species diffusing in aqueous pores (polar pathway) is much smaller than that for neutral solutes diffusing in cutin and waxes (lipophilic pathway). Plant cuticles are lipid membranes with separate diffusion paths for lipophilic non-electrolytes and hydrated ionic compounds. Ions are lipid insoluble and require an aqueous pathway across cuticles. Based on experimental data, the aqueous pathway in cuticles has been characterized. Aqueous pores arise by hydration of permanent dipoles and ionic functional groups. They can be localized using ionic fluorescent dyes, silver nitrate, and mercuric chloride. Aqueous pores preferentially occur in cuticular ledges, at the base of trichomes, and in cuticles over anticlinal walls. Average pore radii ranged from 0.45 to 1.18 nm. Penetration of ions was a first order process as the fraction of the salt remaining on the cuticle surface decreased exponentially with time. Permeability of cuticles to ions depended on humidity and was highest at 100% humidity. Wetting agents increased rate constants by factors of up to 12, which indicates that the pore openings are surrounded by waxes. The pores in cuticular ledges of Helxine soleirolii allowed passage of berberine sulphate, which has a molecular weight of 769 g mol(-1). Increasing the molecular weight of solutes from 100 to 500 g mol(-1) decreased the rate constants of penetration by factors of 7 (Vicia faba) and 13 (Populus canescens), respectively. Half-times of penetration of inorganic salts and organic ions across Populus cuticles and Vicia leaf surfaces varied between 1 and 12 h. This shows that penetration of ionic compounds can be fairly rapid, and ions with molecular weights of up to 800 g mol(-1) can penetrate cuticles that possess aqueous pores.  相似文献

Penetration of calcium chloride across astomatous cuticular membranes (CMs) isolated from leaves of Pyrus communis L. has been studied. Penetration was a first-order process when calcium chloride concentrations ranged from 2 g l−1 to 10 g l−1. Rate constants were increased 10-fold by adding wetting agents but they did not depend on temperature. The accelerators tributyl phosphate and diethyl sebacate had no effect on rates of penetration. Increasing humidity over the salt residue on the CMs from 50 to 90% increased rate constants by about 2-fold. Extracting cuticular waxes from pear leaf CMs increased rate constants by factors of 2 to 3, depending on humidity. Leaf CMs from Malus domestica Borkh., Populus alba L., Stephanotis floribunda Brongn. and Schefflera actinophylla (Endl.) Harms were also permeable to CaCl2. Highest rate constants were observed with poplar CMs while Schefflera CMs exhibited the lowest permeability. By comparing these results with the well established transport properties of the lipophilic pathway it is concluded that calcium chloride hexahydrate penetrated cuticular membranes via aqueous pores. Summary Penetration of 2-14C abscisic acid (ABA) through enzymatically isolated cuticles from tomato fruit and from the upper epidermis of apricot, pear and orange leaves was assessed. Penetration was linear with time, greater as the undissociated than the dissociated ion, and greater through dewaxed than non-dewaxed cuticles. Significantly less (3–6 times) (2-14C)ABA penetrated the tomato fruit cuticle than NAA or 2,4-D. The leaf cuticles were less permeable than the tomato fruit cuticle. There was no evidence that the ABA was altered during transfer across the cuticle.  相似文献

Summary The three types of porin (matrix-proteins) fromSalmonella typhimurium with molecular weights of 38,000, 39,000 and 40,000 were reconstituted with lipid bilayer membranes either as a trimer or as an oligomer (complex I). The specific conductance of the membranes increased several orders of magnitude after the addition of the porins into the aqueous phase bathing the membranes. A linear relationship between protein concentration in the aqueous phase and membrane conductance was found. In the case of lower protein concentrations (10–12 m), the conductance increased in a stepwise fashion with a single conductance increment of 2.3 nS in 1m KCl. For a given salt the conductance increment was found to be largely independent of the particular porin (38 K, 39K or 40 K) and on the state of aggregation, although porin oligomers showed an up to 10 times smaller conductance increase in macroscopic conductance measurements. The conductance pathway has an ohmic current voltage characteristic and a poor selectivity for different alkali ions. Further information on the structure of the pores formed by the different porins fromSalmonella was obtained from the selectivity for various ions. From the permeability of the pore for large ions (Tris+, glucosamine+, Hepes_ a minimum pore diameter of 0.8 nm is estimated. This value is in agreement with the size of the pore as calculated from the conductance data for 1m KCl (1.4 nm for a pore length of 7.5 nm). The pore diameter may well account for the sugar permeability which has been found in reconstituted vesicles. The findings reported here are consistent with the assumption that the different porins form large aqueous channels in the lipid bilayer membranes and that the single condutance unit is a trimer. In addition, it is suggested that one trimer contains only one pore rather than a bundle of pores.  相似文献

A method for isolation of d-amino acid oxidase (DAAO) from disrupted Trigonopsis variabilis cells has been developed. In an aqueous two-phase system consisting of PEG6000 (220 g l–1), potassium phosphate (110 g l–1, K2HPO4 + KH2PO4 = 10.1:1, mol mol–1) and dl-methionine (11 g l–1), the major portion of cellular proteins (87%) was partitioned into the salt phase. By sequential extraction, 48% of DAAO was recovered in PEG phase, giving a yield of 211 U mg protein–1.  相似文献

Antibacterial activity of lyase-depolymerized products of alginate   总被引:1,自引:0,他引:1  
A series of mannuronic acid (M-block) and guluronic acid (G-block) fractions (M1–M5 and G1–G5) with different molecular weights were obtained by lyase depolymerization of alginate and evaluated for in vitro antibacterial activity against 19 bacterial strains. The antibacterial data revealed that both types of fractions generally showed activity against certain tested bacteria, whereas M-block fractions showed broader spectra and more potent inhibition than G-block fractions. Among these fractions, M3 (molecular weight 4.235 kDa) exhibited the broadest spectrum of inhibition and high inhibitory activity against Escherichia coli (minimal inhibitory concentration, MIC = 0.312 μg mL−1), Salmonella paratyphi B (MIC = 0.225 μg mL−1), Staphylococcus aureus (MIC = 0.016 μg mL−1) and Bacillus subtilis (MIC = 0.325 μg mL−1).  相似文献

The leaf surfaces on which mass-reared Trichogramma spp. are released for augmentative biological control may be an important factor effecting the success of these releases. We observed and recorded the walking pattern of Trichogramma nubilale females on leaves of Zea mays, Canna × generalis (Canna lilly), Silphium perfoliatum (cup plant), Abutilon theophrasti (velvetleaf), Schizachyruim scoparium (little bluestem), a smooth and a fuzzy polyester material, and waxed paper. For each surface type, a total of 10 mated, 1–2 days old, naive and fed females were observed and their walking path was traced. Females walked fastest on waxed paper and leaves of S. scoparium and slowest on leaves of A. theophrasti and the fuzzy material. Turning rates were high on Canna × generalis and waxed paper. In general, walking speed was negatively affected by the presence and density of trichomes and possibly the lack of leaf veins. The effect of surface structures, such as trichomes is likely to scale to the body size of the searching parasitoid. Parasitoids can walk over short trichomes, but short directionally pointed trichomes (as on S. scoparium) can guide the search paths in certain directions. The effects of long trichomes may depend on trichome density relative to the parasitoid body length. When trichome density is on the same order of magnitude as 1/L2 (Z. mays), wasps will stand between trichomes, and will frequently run into trichomes, which will retard walking speeds and increase turning.  相似文献

Winter wheat plants were grown in open top chambers either at 365 µmol mol–1 (AC) or at 700 µmol mol–1 (EC) air CO2 concentrations. The photosynthetic response of flag leaves at the beginning of flowering and on four vertical leaf levels at the beginning of grain filling were measured. Net photosynthetic rates (P N) were higher at both developmental phases in plants grown at EC coupled with larger leaf area and photosynthetic pigment contents. The widely accepted Farquhar net photosynthesis model was parameterised and tested using several observed data. After parameterisation the test results corresponded satisfactorily with observed values under several environmental conditions.  相似文献

The temperature-induced volume expansion of enzymatically isolated cuticular membranes of twelve plant species was measured. All cuticular membranes exhibited distinct second-order phase transitions in the temperature range of about 40 to 50° C. Increases in the volumes of fruit cuticles (Lycopersicon, Cucumis, Capsicum, Solanum and Malus) were fully reversible, while leaf cuticular membranes (Ficus, Hedera, Nerium, Olea, Pyrus, Picea and Citrus) underwent irreversible structural changes. Below the phase-transition temperature, volumetric expansion coefficients ranged from 0.39·10–6 m3·kg–1·K–1 to 0.62·10–6 m3·kg–1·K–1, and above from 0.60·106 m3·kg–1·K\-1 to 1.41· 10–6 m3·kg–1·K–1. Densities of cuticles at 25° C ranged from 1020 kg·m–3 to 1370 kg·m–3. Expansion coefficients and phase transitions were characteristic properties of the polymer matrix as a composite material, rather than of cutin alone. It is argued that the sudden increase of water permeability above the transition temperature, is caused by an increase of disorder at the interface between the polymer matrix and the soluble cuticular lipids. Solute mobilities of 28 compounds in isolated cuticular membranes (CM) from Capsicum annuum L. fruit, Citrus aurantium L. and Pyrus communis L. leaves were studied using unilateral desorption from the outer surface. First-order rate constants of desorption (k*), which are directly proportional to the diffusion coefficient in the waxy outer limiting skins of cuticles were measured. When log k* was plotted vs. molar volumes of test compounds linear graphs were obtained. The y-intercepts of these graphs (k*) represent the mobility of a hypothetical molecule having zero molar volume and the slopes of the graphs () represent the size selectivity of the barrier and are related to the free volume available for diffusion. Thus, solute mobilities in cuticles are composed of two independent terms which are subtractive. If k* and are known, k* can be estimated for any solute from its molar volume (Vx) using the equation log k*=log k* –Vx. These parameters were used to analyse the effects of plant species, extraction of cuticular waxes and molecular structure of solutes on solute mobilities in plant cuticles. For aliphatic solutes, k* was a factor of 10 smaller than for cyclic compounds, while was 0.011 and 0.012, respectively. The k*-values for CM of the three species were very similar, but was higher for bitter-orange CM (0.012) than for those of pepper fruits and pear leaves (0.009). This has the consequence that differences in solute mobilities (k*) among cuticles from different plan species increase with increasing molar volumes of solutes. Our data and our analysis provide evidence that constituents of cuticular waxes are mobile, at least in the solid amorphous wax fraction, but mobility decreases rapidly with increasing molar volume. For instance, if amounts to 0.01, mobilities of wax monomers decrease by a factor of 10 for every increase in molar volume of 100 cm3 · mol–1. Thus, hexadecanoic acid is quite mobile in the amorphous wax fraction of Citrus (k*=1.5×10–6·s–1), but for dotriacontane having twice the molar volume, k* was only 2.5×10–9·s–1, which is almost three orders of magnitude smaller. Wax esters have even higher molar volumes and their mobilities will be even smaller (about 4×10–12·s–1 for a C48-ester). Since low chain mobilities are a prerequisite for low mobilities and permeabilities, the selective advantage of high-molecular-weight wax monomers in plant cuticular waxes becomes obvious. Extracting cuticular waxes from pear leaf CM increased solute mobilities by a factor of 182, but it had no effect on size selectivity. Influence of soil temperature on methane emission from rice paddy fields   总被引:18,自引:2,他引:16  
Methane emission rates from an Italian rice paddy field showed diel and seasonal variations. The seasonal variations were not closely related to soil temperatures. However, the dieL changes of CH4 fluxes were significantly correlated with the diel changes of the temperature in a particular soil depth. The soil depths with the best correlations between CH4 flux and temperature were shallow (1–5cm) in May and June, deep (10–15cm) in June and July, and again shallow (1–5 cm) in August. Apparent activation energies (Ea) calculated from these correlations using the Arrhenius model were relatively low (50–150 kJ mol–1) in May and June, but increased to higher values (80–450 kJ mol–1) in August. In the laboratory, CH4 emission from two rice cultures incubated at temperatures between 20 and 38°C showed E . values of 41 and 53 kJ mol–1) Methane production in anoxic paddy soil suspensions incubated between 7 and 43°C showed E values between 53 and 132 kJ mol–1 with an average value of 85 kJ mol–1) and in pure cultures of hydrogenotrophic methanogenic bacteria E a values between 77 and 173 (average 126) kJ mol–1. It is suggested that diel changes of soil properties other than temperature affect CH4 emission rates, e.g. diel changes in root exudation or in efficiency of CH4 oxidation in the rhizosphere.  相似文献

Fick's second law has been used to predict the time course of electrical conductance change in isolated cuticles following the rapid change in bathing solution (KCI) from concentration C to 0.1 C. The theoretical time course is dependent on the coefficient of diffusion of KCI in the cuticle and the cuticle thickness. Experimental results, obtained from cuticles isolated from sour orange (Citrus aurantium), fit with a diffusion model of an isolated cuticle in which about 90% of the conductance change following a solution change is due to salts diffusing from polar pores in the wax, and 10% of the change is due to salt diffusion from the wax. Short and long time constants for the washout of KCI were found to be 0.11 and 3.8 hours, respectively. These time constants correspond to KCI diffusion coefficients of 1 × 10−15 and 3 × 10−17 square meters per second, respectively. The larger coefficient is close to the diffusion coefficient for water in polar pores of Citrus reported elsewhere (M Becker, G Kerstiens, J Schönherr [1986] Trees 1: 54-60). This supports our interpretation of the washout kinetics of KCI following a change in concentration of bathing solution.  相似文献

d-Ribose, a five-carbon sugar, is used as a key intermediate for the production of various biomaterials, such as riboflavin and inosine monophosphate. A high d-ribose-producing Bacillus subtilis SPK1 strain was constructed by the chemical mutation of the transketolase-deficient strain, B. subtilis JY1. Batch fermentation of B. subtilis SPK1 with 20 g l–1 xylose and 20 g l–1 glucose resulted in 4.78 g l–1 dry cell mass, 23.0 g l–1d-ribose concentration, and 0.72 g l–1 h–1 productivity, corresponding to a 1.5- to 1.7-fold increase when compared with values for the parental strain. A late-exponential phase was chosen as the best point for switching to a fed-batch process. Optimized fed-batch fermentation of B. subtilis SPK1, feeding a mixture of 200 g l–1 xylose and 50 g l–1 glucose after the late-exponential phase reduced the residual xylose and glucose concentrations to less than 7.0 g l–1 and gave the best results of 46.6 g l–1d-ribose concentration and 0.88 g l–1 h–1 productivity which were 2.0- and 1.2-fold higher than the corresponding values in a simple batch fermentation.  相似文献

Summary The grey alder compensates leaf area losses due to insect grazing by continuously producing new leaves throughout the vegetative period. Different degrees of defoliation were attained experimentally by a controlled release of the oligophagous beetle Agelastica alni on arbitrarily selected trees from a homogenous population of young alders. The reduction in leaf area per tree significantly influenced the density of leaf trichomes, assessed 10–30 days later, on newly sprouting leaves only. Cross-correlations between leaf area reduction and trichome density were strongest for leaves which completed unfolding 14–21 days after damage. Dualchoice assays suggested a negative influence of trichomes on oviposition rate of A. alni. Removal of trichomes by shaving demonstrated the highly significant effect of trichomes on feeding behavior of adults and larvae in dual-choice assays. The role of the induced increase in trichome density as a possible short-term defense reaction against herbivorous insects is discussed.  相似文献

During the hydrolysis of an exopolysaccharide (EPS) produced by Citrobacter sp., the maximum liberation of hexosamine was obtained with 6 m HCl at 115 °C in an autoclave for 1 h. The glycosidic bond energy and degree of acetylation of the hexosamine in EPS were approximately 77 kJ mol–1 and 61%, respectively. Thermal destruction of the hexosamines and the effect of salt on the hexosamine determination could be minimized under the optimized hydrolytic conditions. Counter diffusion of chloride, applied as NaCl at the inner side of isolated cuticles, and silver, applied as AgNO3 at the outer side, lead to the formation of insoluble AgCl precipitates in isolated cuticles. AgCl precipitates could be visualized by light and scanning electron microscopy. The presence of AgCl precipitates in isolated cuticles was verified by energy dispersive X-ray analysis. It is argued that insoluble AgCl precipitates formed in polar pores of cuticles and as a consequence, cuticular transpiration of 13 out of 15 investigated species was significantly reduced up to three-fold. Water as a small and uncharged but polar molecule penetrates cuticles via two parallel paths: a lipophilic path, formed by lipophilic cutin and wax domains, and a aqueous pathe, formed by polar pores. Thus, permeances P (m s−1) of water, which is composed of the two quantities P Lipid and P Pore, decreased, since water transport across polar pores was affected by AgCl precipitates. Cuticles with initially high rates of cuticular transpiration were generally more sensitive towards AgCl precipitates compared to cuticles with initially low rates of transpiration. Results presented here, significantly improves the current model of the structure of the cuticular transpiration barrier, since the pronounced heterogeneity of the cuticular transport barrier, composed of lipophilic as well as polar paths of diffusion, has to be taken into account in future.  相似文献

The time-dependence of Mn accumulation was confirmed in potato foliage (Solanum tuberosum. L.cv. Norland) grown in solution culture. Older leaves grown at 0.61 mM Mn had substantially higher Mn concentrations than younger leaves and stem samples. Levels of Mn in older leaves increased steadily from 4000 µg g–1 at one week to 8–10,000 µg g–1 at 6 weeks, but were relatively constant in the emerging leaves. Even foliage grown at low Mn levels (0.01 mM Mn) had 4 fold gradients in Mn concentration from younger (40 µg g–1) to older leaves (180 µg g–1).At 0.61 mM Mn, concentrations of 3–4000 µg g–1 in the youngest fully-developed leaves did not bring about any decline in yield, and levels of up to 5000 µg g–1 occurred in individual potato leaves before Mn toxicity symptoms were observed. Potato foliage grown at the high Mn had similar leaf numbers, but showed an increased stem length and smaller leaves than foliage grown at 0.01 mM Mn. In particular, the leaf area of the middle and lower leaf fractions were affected by the high Mn level.The ability of rapidly growing plants to withstand high concentrations of Mn is discussed in relation to the pattern of dry matter and Mn accumulation shown by potato foliage.  相似文献

Callus of Orthosiphon stamineus could be induced successfully from petiole, leaf and stem tissues but not roots when cultured on MS medium containing different concentration of NAA (0–4.0 mg l–1) and 2,4-D (0–2.0 mg l–1). Highest fresh weight callus production was obtained from leaf explants and those with best friability were obtained on MS medium plus 1.0 mg l–1 2,4-D plus 1.0 mg l–1 NAA. Cell suspension cultures were established from these cultures. The appropriate cell inoculum size for the best cell growth was 0.75 g of cells in 20 ml culture medium. Cell suspension culture using MS medium supplemented with 1.0 mg l–1 2,4-D promoted the best cell growth with maximum biomass of 8.609 g fresh weight and 0.309 g dry weight 24 days after inoculation. Cells that grew in MS medium supplemented with 1.0 mg l–1 2,4-D reached the stationary growth phase in 15 days as compared to the cells that grew in MS medium supplemented with 1.0 mg l–1 2,4-D + 1.0 mg l–1 NAA reached the stationary phase in 24 days. MS medium supplemented with 1.0 mg l–1 2,4-D was considered as the maintenance medium for maintaining the optimum cell growth of O. stamineus in the cell suspension cultures with 2-week interval subculture.  相似文献

A method is described that allows quantitative determination of polyethylene glycol (PEG) concentrations by spectrophotometric measurement of fluorescein dye absorbance after its partitioning into an aqueous two-phase system containing mPEG (Mr 5 kDa) in the upper phase and ammonium sulfate in the lower phase. The absorbance decrease of fluorescein in the lower phase is directly proportional to the mPEG concentration, with two proportionality constants equal to 4.42 × 105 and 2.84 × 105 M−1 cm−1 in the range of 0-0.4 and 0.4-1 μM, respectively. This experimental technique can be extended to PEGs of other molecular weights by means of calibration curves that give for each size of PEG the adequate proportionality constants. The results indicate that the quantitative determination is not affected by the presence of many substances such as proteins, reducing agents, and salts, at the usual concentrations.  相似文献
