Gibberellins GA1, GA4, GA8, GA9, GA19, GA20, GA29, GA44, GA81,indole-3-acetic acid (IAA) and abscisic acid (ABA) were identifiedin cambial region tissues of Eucalyptus globulus by comparingmass spectra and Kovats retention indices with those of authenticstandards. Using stable isotope labelled internal standardsGA19, GA20 and GA44 were quantified at levels of 2–7 ng(g fr wt)-1, other GAs were present at levels < 1 ng (g frwt)-1. Levels of IAA and ABA ranged from 417–1, 140 ng(g fr wt)-1 and 86–305 ng (g fr wt)-1 respectively. Abscisic acid (ABA) applied to the decapitated second internodeof runner bean plants enhanced outgrowth of lateral buds onlywhen internode stumps were no longer elongating. Applied toelongating internodes of slightly younger plants, ABA causesinhibition of bud outgrowth. Together with 10–4 M indol-3-ylacetic acid (IAA), ABA stimulated internode elongation and interactedadditively in the inhibition of bud outgrowth. A mixture of10–5 M ABA and 10–6 M gibberellic acid (GA3 ) causedsimilar effects on internode growth as IAA + ABA, but was mutuallyantagonistic in effect on growth of the lateral buds. The effects of ABA, isobutyric acid (IBA) and nicotine on K+and Cl+ efflux were studied in Arabidopsis thaliana seedlings,and the role of pHcyt, and Em in the regulation of the effluxof these ions was discussed. The data show that treatments withIBA and nicotine influenced in opposite directions the effluxof either K+ or Cl: K+ efflux was increased by nicotineand reduced in the presence of IBA, whereas Cl effluxwas stimulated by IBA and decreased by nicotine treatment. Underall the conditions tested ABA induced cytoplasmic acidificationand inhibition of K+ and Cl net efflux. Experiments aimedto estimate the individual contribution of pHcyt and Em in modulatingK efflux indicated that, within the range of acidic pHcytvalues, a regulation of K+ efflux was imposed by pHcyt on thecontrol exerted by Em, the efflux being inhibited by lower pHcytvalues. Conversely, in the alkaline side of pHcyt K+ effluxseemed linked only to the Em values. These results are consistentwith the hypothesis that the decrease in K+ efflux observedin non-stomatal tissues in the presence of ABA may be mediatedby the cytoplasmic acidification induced by the hormone. In the chilling sensitive (C.S.) species Phaseolus vulgarisit was found that at 22 ?C ABA induced stomatal closure butthis effect was dependent on the presence of CO2. In the absenceof CO2 the effect of ABA was completely lost. In contrast toABA, the effect of IAA at 22 ?C was to increase stomatal openingas the IAA concentration increased from 10–2 to 10 molm–3, and this effect was dependent upon the presence ofCO2. However, at 5 ?C the action of ABA was reversed and itwas found to induce stomatal opening when fed via the transpirationstream in excised leaves. Similarly, the CO2 response characteristicswere reversed at low temperatures as removal of CO2 from theatmosphere caused stomatal closure. However, the effect of IAAat 5 ?C in the presence of CO2 and with or without ABA was toincrease stomatal aperture with increasing IAA concentration.Significantly, ABA was found to have no effect upon aperturein the presence of CO2 when IAA was added. Wheat (Triticum aestivum L.) embryos form in dynamically-regulatedovular environments. Our objectives were to improve developmentof cultured immature wheat embryos by simulating, in vitro,abscisic acid (ABA) levels and O2 tensions as found in wheatovules during zygotic embryogenesis. We characterized from intactwheat kernels embryo respiration, embryo morphology and embryoand endosperm + ABA levels at 13, 19 and 25 d post-anthesis(DPA). Young (13 DPA) embryos were then excised and culturedin vitro, where they were exposed to 0·2 or 2·Ommol m–3 ±ABA and 2.·1, 2·5 or 7·4mol m–3 (6, 7 and 21%, respectively) gaseous O2. At 6and 12 d in culture, + ABA levels, embryo respiration and embryomorphology were characterized by treatment. Thirteen-day-oldembryos from two different plant populations differed by 17-foldin initial ABA content. However, this difference did not affectprecocious germination in vitro, nor did it affect the amountof exogenous ABA required to reduce precocious germination by40%. In this respect, embryos from both populations were equallysensitive to exogenous ABA. Cavity sap O2 levels (2·1to 2·5 mol m–3) were much more effective in preventingprecocious germination of cultured embryos than were cavitysap levels of ABA (0·2 to 2·0 mmol m–3).The combination of physiological levels of both ABA and O2 largelynormalized DW accumulation and embryo morphology without alteringendogenous + ABA levels. The influence of a water stress or foliar ABA spraying pretreatmenton stomatal responses to water loss, exogenous ABA, IAA, Ca2+,and CO2 were studied using excised leaves of Solanum melongena.Both pretreatments increased stomatal sensitivity of water loss,in the presence and absence of CO2, but decreased stomatal sensitivityto exogenous ABA. CO2 greatly reduced the effect of exogenouslyapplied ABA. IAA decreased leaf diffusion resistance for controland ABA sprayed leaves, but did not influence the LDR of previouslywater-stressed leaves. Ethylene and1-aminocyclopropane-l-carboxylic acid production in flacca,a wilty mutant of tomato, subjected to water deficiency andpretreatment with abscisic acid —J. exp. Bot. 37: 535–541. Plants of Lycoperstcon esculentum Mill. cv. Ailsa Craig wildtype and flacca (flc) were sprayed daily with H2O or 2?10–2mol m–3 abscisic acid (ABA). ABA treatment effected apartial phenotypic reversion of flc shoots; leaf areas wereincreased and transpiration rates decreased. Leaf expansionof wild type shoots was inhibited by ABA. Indoleacetic acid (IAA), ABA and l-aminocyclopropane-l-carboxylicacid (ACC) concentrations were determined by combined gas chromatography-massspectrometry using deuterium-labelled internal standards ABAtreatment for 30 d resulted in greatly elevated internal ABAlevels, increasing from 1?0 to 4?3 and from 0?45 to 4?9 nmolg–1 fr. wt. in wild type and flc leaves respectively.Endogenous IAA and ACC concentrations were much lower than thoseof ABA. IAA content ranged from 0?05 to 0?1 nmol g–1 andACC content from 0?07 to 0?24 nmol g–1 Ethylene emanationrates were similar for wild type and flc shoots. Wilting of detached leaves induced a substantial increase inethylene and ACC accumulation in all plants, regardless of treatmentor type. Ethylene and ACC levels were no greater in flc leavescompared to the wild type. ABA pretreatment did not preventthe wilting-induced increase in ACC and ethylene synthesis. In azuki bean (Azukia angularis = Vignia angularis) epicotylsections, 5 ? 10–4 M coumarin inhibited the incorporationof radioactivity from [U–14C]glucose into the cellulosefraction by 35% in the absence of indole-3-acetic acid (IAA)and by 40% in the presence of 1 ? 10–4 M IAA. There wasno inhibitory effect on the incorporation of radioactivity intothe other fractions. Coumarin at 5 ? 10–4 M reversed thepromoting effect of 1 ? 10–5 M gibberellin A3 (GA) andthe inhibitory effect of 1 ? 10–5 M kinetin on IAA-inducedelongation of sections with no significant effects on IAA-inducedelongation. Neither GA nor kinetin had any appreciable effectson cellulose synthesis. No inhibition of cellulose syntheiswas observed with 1 ? 10–3 M colchichine, which has beenreported to have effects similar to those of coumarin on GA-or kinetin-affected stem elongation. Coumarin at 5 ? 10–4M was ineffectual in breaking up wall microtubules, while adisrupting effect on wall microtubules was clearly demonstratedwith 3 ? 10–4M colchicine. Leaves from in vitro and greenhouse cultured plants of Malusdomestica (Borkh.) cv. Mark were subjected to 4 h of darkness;4 h of 1 M mannitol induced water stress; 1 h of 10–4M to 10–7 M cis-trans abscisic acid (ABA) treatment; 1h of 0.12% atmospheric CO2. Stomatal closure was determinedby microscopic examination of leaf imprints. In all treatments,less than 5% of the stomata from leaves of in vitro culturedplants were closed. The diameter of open stomata on leaves fromin vitro culture remained at 8 µm. In contrast, an averageof 96% of the stomata on leaves of greenhouse grown plants wereclosed after 4 h in darkness; 56% after 4 h of mannitol inducedwater stress; 90% after 1 h of 10–4 M ABA treatment; 61%after 1 h in an atmosphere of 0.12% CO2. Stomata of in vitroapple leaves did not seem to have a closure mechanism, but acquiredone during acclimatization to the greenhouse environment. Thelack of stomatal closure in in vitro plants was the main causeof rapid water loss during transfer to low relative humidity.  相似文献

Two-node explants from Sweet Orange cv. St Ives Valencia orangeshoots produced prolific callus and formed secondary abscissionzones within internodes when cultured in vitro with abscisicacid (ABA, 5 µM) or -naphthaleneacetic acid (NAA, 5 µM).Benzyladenine (BA, 1 µm) induced callus but had littleeffect on abscission. Secondary abscission zone formation wasassociated with ABA-induced and auxin-induced ethylene formation.Treatment of explants with inhibitors of ethylene synthesis[aminoethoxyvinyl glycine (AVG), Co2+, PO43–] preventedformation of secondary abscission zones but had variable effectson callus formation. Newly made explants contained high concentrationsof endogenous ABA (up to 6000 ng g–1 f.wt), as measuredby GC/MS/SIM. Long-term subculture of explants (two years) inmedia containing BA (1 µm) led to a reduction in endogenousABA level (40 ng g–1 f. wt) and to loss of capacity toform extensive callus and secondary abscission zones. The cell-membrane resistance (Rm) of Vigna hypocotyls was examined,and the effects of osmotic stress, ionic stress and IAA on Rmwere investigated. Rm decreased by 64 to 77% under osmotic stressin the presence of absorbable solutes (40 mM sorbitol, 15 mMKC1, 30 mM sucrose; or 40 mM sorbitol, 15 mM KC1, 30 mM sucroseplus 10–4 M IAA) or under ionic stress (50 mM NaCl or50 mM KC1). Rm was not changed by perfusion with 10–4M IAA. Therefore, the hyper-polarizations of the membrane potentialobserved in both cases should be ascribed totally to the activationof the electrogenic proton pump. Although Rm showed an increaseof 1.6 fold when the hypocotyls were subjected to osmotic stress(100 mM sorbitol or 100 mM sorbitol plus 10–4 M IAA),83.6% or 92.4% of the hyperpolarization of the membrane potential(Vpx was also the result of the activation of the pump. Theresults, calculated on the basis of the current source model,support the viewpoint that the hyperpolarization of the cellmembrane potential of Vigna hypocotyls under osmotic stress,ionic stress or in the presence of IAA is an expression of theactivation of the proton pump, and is not caused by an increasein Rm. 1 Present address: Researchers and Planners of Natural Environment, Yotsugi Bldg. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. An efficient medium for multiplication of Prunus cerasifera,J. F. Ehrh. cv. ‘Atropurpurea’, Pissardi plum, consistedof Linsmaier-Skoog basal medium (LS) supplemented with 3% sucrose,10mg 1–1 N4-(2-isopentenyl) adenine (2iP), and 162 mg1–1 phloroglucinol (Phl). Phl in the medium significantlyenhanced growth (P < 0.1 %) over cultures maintained on LSmedium with 10 mg 1–1 2iP and no Phl. Plantlets were rootedon a half-strength Murashige-Skoog (MS) medium supplementedwith 0.2 mg 1–1 indole-3-butyric acid (IBA). Epidermal strips from either well-watered or water-stressedplants of Commelina communis L. were subjected to a range ofABA concentrations (10–6–10–3 mol m–3)in the presence (330 parts 10–6 in air) or virtual absence(3 parts 10–6 in air) of CO2. The stomatal response toCO2 was greater in epidermis from water-stressed plants, althoughthere was a distinct CO2 response in epidermis from well-wateredplants. Additions of ABA via the incubation medium had littleeffect on the relative CO2 response. Stomata responded to ABAboth in the presence and virtual absence of CO2, but the relativeresponse to ABA was greatest in the high CO2 treatment. Whenwell-watered plants were sprayed with a 10–1 mol m–3ABA solution 1 d prior to use, the stomatal response of detachedepidermis to both CO2 and ABA was very similar to that of epidermisdetached from water-stressed leaves. It is hypothesized thata prolonged exposure to ABA is necessary before there is anymodification of the CO2 response of stomata.  相似文献

Stomatal responses of Vicia faba L. to indole acetic acid and abscisic acid   总被引:1,自引:0,他引:1  
Evidence is presented that stomata in isolated epidermal peelsof Vicia faba L. open in darkness in response to the externalpresence of indole acetic acid (IAA) in the incubation medium.The effect of IAA is found to be overcome completely in thepresence of either TRIS or MES buffers. In the absence of buffer,V. faba stomata are shown to be influenced by IAA in a concentration-dependenttrend which reached a maximum at an [IAA] of 10–3 molm–3. Further investigations reveal that stomata in thisspecies can be shown to respond to the presence of IAA and anotherphytohormone, abscisic acid (ABA). IAA and ABA are demonstratedto be antagonistic in their effects provided the incubationconditions are suitable. The data are discussed in relationto stomatal responses of other species in different treatmentconditions. Somatic embryogenesis can be induced in tissue cultures of Freesiarefracta either directly from the epidermal cells of explants,or indirectly via intervening callus. These two pathways ofsomatic embryogenesis can be controlled and regulated by varyingthe combinations and levels of exogenous hormones. When younginflorescence segments were cultured in vitro on modified N4(MN4) medium supplemented with 2 mg l–1 indoleacetic acid(IAA) and 3 mg l–1 6-benzylaminopurine (BAP), some ofthe epidermal cells began to exhibit the features of embryogeniccells. These cells produced embryoids and developed into newplants through direct somatic embryogenesis. If the same explantswere placed on Murashige and Skoog's (MS) medium containing2 mg l–1 IAA, 05 mg l–1 BAP and 05 mg l–1naphthaleneacetic acid (NAA), pale-yellow translucent nodularcalluses appeared on the surface of the explants. When thiskind of callus was transferred to MN6 medium with 2 mg l–1IAA and 3 mg l–1 BAP, embryoids formed which further developedinto plantlets. The regenerated plants were morphologicallynormal and possessed the normal diploid chromosome number of2n = 22. A similar result has also been obtained with youngleaf explants of this plant. The early segmentations of embryogeniccells and the development of embryoids were studied using histologicaland scanning electron microscopic techniques, and the resultshave been discussed in association with the ontogeny and originof the embryoids. Daucus carota tissues were grown on Murashige-Skoog medium (MS)at different concentrations with abscisic acid (ABA). Sevenbands of chloroplast fractions were obtained on a sucrose gradient.At 10–5M, ABA highly increased chlorophyll and proteinnitrogen content of medium density chloroplasts. With increasingage of the tissues, the most active chloroplasts according totheir 14CO2 fixation were found in smaller numbers. When treatedwith 10–5 M ABA, 34 day-old tissues cultivated in vitroshowed the chloroplast pattern of 110 day-old tissues. The effectof ABA—given to the tissues during a short pretreatmentor continuously present in the culture medium—on the ribulosediphosphate carboxylase activity was analysed. Astle, M. and Rubery, P. 1987. Carrier-mediated ABA uptake bysuspension-cultured Phaseolus coccineus L. cells: Stereospecificityand inhibition by ionones and ABA esters.—J. exp. Bot.38: 150–163. The substrate for the abscisic acid (ABA) carrier in Phaseoluscoccineus L. suspension-cultured cells is shown to be the (S)ABAenantiomer, Km = 1?0 mmol m–3. The methyl (MeABA) andphenyl (PheABA) esters of ABA inhibit carrier-mediated uptakeof ABA with half-maximal inhibition achieved at about 7?0 mmolm–3 and 10 mmol m–3 respectively: with (S)MeABAthis value is decreased to about 2?0 mmol m–3. There isno demethylation of radioactive MeABA by the cells during 5min incubations. Although MeABA reversibly inhibits the ABAcarrier, it is not a transport substrate: association of radioactiveMeABA with living cells is unaffected by non-radioactive MeABAor ABA and, by comparison with frozen-and-thawed cells, it isshown that the radioactivity remains extracellular. It is proposedthat MeABA binds to the carrier to form an abortive complexthat is not translocated. The terpenoid ABA analogue LAB 144143also inhibits carrier-mediated ABA uptake. At concentrationsup to about 20 mmol m–3 - and ß-ionone specificallyinhibit the ABA carrier with the half-maximal effect at about0?6 mmol m–3 ß-ionone. However, at higher iononeconcentrations, the uptake of ABA, indol-3-yl acetic acid andof 5,5-dimethyloxazolidine-2,4-dione (DMO) are all stimulated:this may reflect general permeabilization of the membrane toweak acids by ionone. Water and 1 mg1–1 each of IAA and IBA completely inhibitedon the cuttings of Phaseolus mungo obtained from the seedlingsraised under far-red light but rooting took place in the darkand under white and red lights. Sucrose, however, caused rootingunder far-red light and its effectiveness increased with theaddition of IAA and IBA to the sucrose medium IBA being moreeffective. Culturing in 1 and 5 mg 1–1 each of FudR, actinomycin-D,cycloheximide and chloramphenicol after pre-treatment with water,IAA, IBA, sucrose and IAA/IBA + sucrose inhibited rooting. Theeffect increased with the concentration of each inhibitor. Incontrast to this, the culturing in water, IAA, IBA, sucroseand IAA/IBA + sucrose after pre-treatment with these metabolicinhibitors produced varying effects. While the inhibition persistedin water, IAA and IBA and even in 5 mg 1–1 of each inhibitorthat in sucrose stimulated rooting and the effect increasedwith the addition of IAA and IBA to sucrose, the effect of IBA+ sucrose being more pronounced. This stimulation was irrespectiveof the inhibitor and light condition except that 1 mg 1–1actinomycin-D inhibited rooting in the dark and under far-redlight.  相似文献

In Gibberella fujikuroi and Penicillium notatum, IAA, 2,4-Dand GA3 promoted conidial germination and the elongation ofyoung hyphae. The promotive effects of IAA and GA3 were additive.In both fungi, the concentrations of endogenous auxin and gibberellinin the culture media were 10–10 to 610–12M.
