Responses of Vegetable Seeds to Controlled Hydration   总被引:1,自引:1,他引:0  
Leek, onion and carrot seeds were imbibed in water and in solutionsof polyethylene glycol (PEG) 6000 over the range –0.5to –4.0 MPa osmotic potential, for periods up to 28 dat 15 C. Seeds started to germinate after 7 and 14 d at –0.5MPa and –1.0 MPa PEG, respectively, but in the lattercase, germination did not exceed 5%. No germination occurredin solutions of lower (more negative) osmotic potential. Seedmoisture content increased with osmotic potential in all threespecies and the relationships were unaffected by the durationof imbibition in solutions of –1.0 to –4.0 MPa,though leek seeds had higher moisture contents than the otherspecies for any given osmotic potential. Linear relationships between response to priming (differencebetween mean germination times of primed and untreated seeds)and seed moisture content were obtained for each species, positiveresponses being obtained above 30–35% seed moisture contentwith optima at 46, 44.5 and 44% seed moisture contents in leek,onion and carrot, respectively. Priming had no effect on embryovolume or cell number per embryo in leek and onion. Carrot embryovolume increased by 43% and cell number per embryo doubled inprimed compared with untreated seeds, whereas seeds imbibedin water showed only a slight increase in cell number per embryoat the stage when radicles were beginning to penetrate the seedcoat. Seeds of Hancornia speciosa germinated best at a temperatureof 20–30 °C. The viability of the seeds during storagewas short and the best storage conditions for viability entailedkeeping the seeds in polyethylene bags. Seed viability was maintainedonly when the seeds were stored at a moisture content above30%; storage conditions which allowed dehydration resulted ina rapid loss of viability (the seeds showed recalcitrant behaviour). Low temperature during storage did not improve longevity. Arelationship between germination and moisture content was established,but when the moisture content fell below 25% there was a drasticreduction of germination. After 9 weeks of storage, even athigh moisture content, seeds lost viability. Loss of seed viability during seed dehydration was associatedwith increased leakage of electrolytes and organic solutes,and reduced tetrazolium staining during subsequent imbibition. Sweet pepper seeds were osmotically conditioned in 0.4 M mannitolsolution for 4 d (at 25 °C, in darkness) before or afterstorage at 35 °C for up to six months, and their germinationand viability was compared with that of untreated seeds storedunder the same conditions. Seeds that had been osmoconditionedprior to storage retained a high rate of germination and germinatedto a high final percentage (from 80 to 50 per cent) at both15 and 25 °C throughout the storage period. By contrast,both the rate and total level of germination of untreated pepperseeds declined rapidly at both germination temperatures, andby three months of storage the total level of seed viabilitywas already less than 10 per cent. Seeds that were first storedat 35 °C, and then osmoconditioned just prior to germination,showed a decline in germinability which when tested at 25 °Cwas the same as for untreated seeds, while tested at 15 °Coccurred at a slightly slower rate than for untreated seeds. It is evident that osmoconditioning prior to storage, in additionto the acceleration of germination, resulted in a dramatic delayof the ageing rate, thus increasing considerably the longevityof seeds. On the other hand, osmoconditioning after storagedid not seem to have any significant effect on seed viability,though it enhanced the germination rate. The rate and final percentage of field emergence (seed quality)was measured in 55 seed-lots of carrots (Daucus carota L.),45 seed-lots of onion (Allium cepa L.) and 32 seed-lots of cabbage(Brassica oleracea var. capitata), together with the followingbiochemical indices: rates of ATP-synthesis; reduction of tetrazoliumby seed powder; AMP and malate content. Differences in field performance of seed-lots (seed quality)showed some correlation with particular biochemical indices.Seed quality was most significantly correlated with AMP contentand ATP synthesis in carrot, with malate content, reductionof tetrazolium and ATP synthesis in onion and with malate andAMP content and reduction of tetrazolium in cabbage. Seeds of onion (Allium cepa) cv. Ailsa Craig were exposed to various doses of gamma radiation (0, 10, 20, 40, 80 and 100 krad) and subjected to accelerated ageing (RH 100%, 42°C) for 12 h. Radiation doses significantly affected the percentage of normal seedlings, abnormality types (%) and seedling growth. Seed viability, conductivities of seed leachates, final germination (%), germination speed and time to reach 50% germination (T 50) were not affected by the seed irradiation. Accelerated ageing after irradiation had significant influence on seed viability, conductivities of seed leachates, final germination (%) and percentage of normal seedlings. Germination speed, T 50 and seedling growth were not affected by the accelerated ageing. However, all the parameters studied were significantly influenced by the interaction of radiation doses and accelerated ageing. Accelerated ageing generally enhanced the damaging effects of irradiation on seeds. Therefore, it was concluded that onion seeds should not be exposed to adverse environmental conditions after irradiation.  相似文献

The influence of seed priming and ageing treatments on viabilityand rate of germination of tomato (Lycopersicon esculentum Mill.)seeds was examined under both long-term and controlled-deteriorationstorage conditions. Seeds of a single lot of tomato were eitherprimed or aged to increase or decrease the rate of germination(Argerich and Bradford, 1989). They were then stored at 6% moisturecontent (dry weight basis) at either 4 ?C or 30 ?C for 1 year.Both viability and germination rate were unaffected by eitherstorage temperature in control seeds, or by 4 ?C storage inprimed or aged seeds. At 30 ?C, however, viability and germinationrate of primed and aged seeds was markedly reduced after 6 monthsof storage. The temperature dependence of the germination rateand the spread of germination times within the population wasalso adversely affected by high temperature storage, particularlyfor primed seeds. Under controlled deterioration conditions(13.5% moisture content and 50 ?C), the rate of loss of viabilitywas greater for primed seed than for control or aged seeds.The relationship between seed viability and the mean germinationrate, however, was not influenced by the seed treatments. Thesedata are analysed in relation to current models of seed deteriorationduring storage and seed repair during priming. The results indicatethat enhancement of seed germination rates by priming treatmentssimultaneously lowers the resistance of seeds to deterioration.Primed tomato seeds must, therefore, be considered to be vigorousseeds with a reduced storage life. Effects of 2 °C chilling on the threshold moisture contentsand water potentials for various physiological processes wereestimated forAesculus hippocastanumL. seed. Seed harvested atthe time of maximum seed fall exhibited a dual response to drying:partial drying from near 50% to 32–40% moisture contentprogressively increased germination percentage (at 16 °C)up to various peak values; further desiccation was detrimental,confirming that the seeds are ‘recalcitrant’. Themoisture content for optimum germination was increased by atleast 10% as the chilling period was raised from 0 to 9 weeks.A negative linear relationship was found between log10mean timeto germinate and probit final germination, regardless of pre-treatment,indicating that partial desiccation and chilling are interchangeablein promoting germination of hydrated seed. For nearly fullyhydrated seeds, increasing the chilling period from 6 to 26weeks increased the viability-loss onset point for desiccationinjury from near 40% to about 48% moisture content without alteringthe drying rates of seed tissues. Extending moist chilling invarious seed lots from 0 to 26 weeks decreased subsequent longevityat 16 °C. For 26-week-chilled seeds longevity (the periodto lose one probit of germination) differed above and belowa threshold moisture content of 48%. It remained constant inthe moisture-content range 48–38%, but increased progressivelyas moisture content was raised above 48%. This threshold moisturecontent coincided with the value above which chilled seed pre-germinatedin storage. Seeds of Araucaria hunsteinii K. Schum. dried quicker at 29°Cthan at 19°C and quicker with the seed-coat removed thanwhen intact; seeds enclosed in polyethylene bags increased inmoisture content. At 15°C, seeds in a flow of air driedquicker than seeds in a box with silica gel, which in turn driedquicker than seeds in a box with no desiccant. No loss of germinationability occurred on drying fresh seed from 53 to about 32 percent moisture content (fresh weight basis); during further desiccationthe percentage germination was related to percentage moisturecontent in the form of a sigmoid curve, culminating in a completefailure to germinate at approximately 14 per cent moisture content.A consistent relationship was observed for all treatments andthe mean critical moisture content for seed death (failure togerminate) was near 23 per cent. Excised embryos grew on 1 percent agar but died if previously subjected to 14 h of desiccationat 15°C. In contrast, no relationship was found between germination andmoisture content of A. cunninghamii D. Don on desiccation from21 to 7 per cent moisture content. Possible causes for the observeddifference in response to desiccation are discussed and methodsfor seed storage are considered. Effects of dehydration, storage temperature and humidificationon germination of Salix alba andS. matsudana seeds were studied.Newly released seeds showed 100% germination before and afterdehydration to 11–12% moisture content. Germination ofthe high vigour lot (100% initial normal germination) was notaffected by dehydration to 6.7% moisture content but germinationdecreased with further dehydration to 4.3%. The lower vigourlot (75% initial normal germination) was more susceptible todehydration and germination decreased following dehydrationto 6.7% moisture content. Dry seeds of both species survivedimmersion in liquid nitrogen without loss of viability. Thegermination of seeds stored with 9% moisture content decreasedto 35–40% in 5 months at -20°C or in 2 months at 5°C.However, at 25°C seeds entirely lost viability within 2weeks. Seeds showed improved performance when stored at -70°C> - 20°C > 5°C > 25°C and tolerated dehydrationto a moisture content in equilibrium with 15% relative humidity.Results suggest that they are orthodox in storage behaviouralthough they are short-lived. Humidification treatment of lowvigour seed lots resulted in a remarkable increase in germinationpercentage. Leek and onion seed dry weight increased exponentially for thefirst 31 days after flowering (DAF) but thereafter the increasein dry weight was slower. Before maximum seed dry weight wasreached at 45 DAF in onion and 59 to 66 DAF in leek, seed moisturecontent, seed oxygen uptake and conductivity of the seed steepwater fell from initially high levels. Although some seeds germinated31 DAF in both species, full germination in both was not reacheduntil 66–80 DAF. Tolerance of the seed to artificial dryingimmediately after harvesting occurred 45 DAF in onion and 74DAF in leek. Free nuclear division continued in the endospermuntil 17–22 DAF in onion and until 31–35 DAF inleek but it was not until 45 DAF in onion and 66 DAF in leekthat the embryo and endosperm filled the cavity formed by thepericarp. After formation of cell walls in the endosperm thepattern of change in cell number in both species was similar.The shrunken appearance of the seed coat in leek, which occurredearly in seed development, was associated with the period offree nuclear division in the endosperm and, in addition, thepericarp was thinner than in onion. There was no evidence thatthe shrunken seed coat early in development was associated withself as opposed to open-pollination. Damage and degradation of cellular proteins is observed duringage-induced seed deterioration. L-Isoaspartyl protein methyltransferase(EC 2.1.1.77 [EC] ) is an enzyme hypothesized to play a role in limitingand repairing age-induced damage to proteins. Tomato (Lycopersiconesculentum Mill. ‘New Yorker’) seeds were assayedfor changes in L-isoaspartyl methyl-transferase activity duringaccelerated ageing and after osmotic priming. Accelerated ageingof seeds for 1–4 d at 45C and 100% relative humidityreduced germination from 94% to 71%, increased the mean timeof germination (MTG) from 2.4 to 5.8 d, and was accompaniedby a correlative decrease in L-isoaspartyl methyltransferaseactivity (r2=0.90). Aged and untreated seeds were primed for7 d at 20C in darkness using aerated solutions of 3% KNO3 orpolyethylene glycol 8000 (PEG) with equivalent osmotic potential(–1.25 MPa). Priming with KNO3 decreased the MTG, butdid not improve germination percentage for untreated seeds.Priming did not affect L-isoaspartyl methyltransferase activityin untreated seeds, but restored activity in aged seeds primedin KNO3 to levels near that of untreated seeds. Priming withPEG did not effectively improve the MTG or increase L-isoaspartylmethyltransferase activity. During germination, L-isoaspartylmethyltransferase activity remained constant for 48 h post-imbibitionand then declined, suggesting that the enzyme was developmentallyregulated and inactivated or degraded as radicle emergence occurred. Cucumber (Cucumis sativus L. cv. Mesa) and onion (Allium cepaL. cv. Rijnsburger Heldis) seeds were rapidly aged at 40 °Cand 74% relative humidity. Onion seeds were also slowly agedat 40 °C with 15% relative humidity for 11 months and onemore month at 28% relative humidity. Significant loss of totaland individual phospholipids was an early event during bothstorage treatments. With slow ageing of onion, loss of phosphatidylcholineoccurred several months before loss of viability and vigourwas detected. Phosphatidic acid, the lipid product of phospholipaseD action, increased during rapid ageing of both cucumber andonion. Phosphatidic acid was present in onion seeds before theageing treatments and its content remained unchanged in theslowly aged seeds. There was 1600 (cucumber) and 2000 (onion)times more phospholipase D activity (6 x 105 and 2·9x 105 nmol g–1 d–1 in cucumber and onion, respectively)in crude extracts from non-aged seeds than was required to accountfor the fastest fall in phospholipids (72, 372 and 144 nmolg–1 d–1 for cotyledons and radicles of cucumberand onion, respectively, over the first 9 d [cucumber] or 1d [onion] of ageing) and fastest increase in phosphatidic acid(7, 162 and 37 nmol g–1 d–1). How accurate a guidethe in vitro activity of phospholipase D was to the in vivoactivity was unclear. However, the considerable excess activityseen with the formation of phosphatidic acid supports the proposalthat hydrolysis of phospholipids by phospholipase D is a firststep in deterioration during ageing. Substantial lipoxygenaseactivity was also detected (58 x 103 and 54 x 103 nmol g–1d–1 respectively, for non-aged cucumber and onion seeds).However, the increase in conjugated dienes (an early productof peroxidation) in ageing cucumber seeds was comparativelysmall (90 nmol g–1 d–1 over 21 d ageing), and increasein malondialdehyde could not be detected, indicating that peroxidationmay not have been a major factor in cucumber. The increase inconjugated dienes during rapid ageing of onion seeds was larger(1·5 x 103 nmol g–1 d–1 over days 0–2of rapid ageing), much greater than the decrease in phospholipidacyl groups (260 nmol g–1 d–1 over days 0–2of rapid ageing) indicating the occurrence of peroxidation offatty acids released from reserve as well as from membrane lipids.This higher level of conjugated dienes during onion ageing wasthe main difference between cucumber and onion, indicating thatthe level of peroxidation could be an important difference betweenfast and slow ageing seeds. However, peroxidation is not theonly possible deleterious process since hydrolysis of the normalmembrane phospholipids to phosphatidic acid increased the contentof non-bilayer-forming lipids and this too could be a directmembrane-destabilizing consequence of phospholipase D actionduring ageing. The effect of matriconditioning, the physiological presowing seed technique, using Micro-Cel E on Allium cepa L. cv. Czerniakowska seed quality was studied. Several ratios of seeds, carrier, water and time of priming were tested. The most effective treatment for improving onion seed germination at most tested temperatures was priming to a ratio of 2 g seed:1 g Micro-Cel:3 g water for 5 days in light at 15 °C. Matriconditioning greatly improved the germination and emergence percentage, seedling fresh and dry weight and reduced electrolyte leakage compared to that of untreated seeds; this beneficial effect was especially evident at suboptimal temperatures. Matriconditioning improved the germinability of aged seeds, the effect being more pronounced in the more aged seeds. No significant differences in ethylene production by primed and non-primed seeds were observed in the absence of its precursor, 1-aminocyclopropane-1-carboxylic acid (ACC), but its presence during imbibition caused an increase in ethylene production; an enhanced activity of in vivo ACC oxidase in Allium cepa matriconditioned seeds in comparison to untreated seeds, indicates that the endogenous level of ACC is a limiting factor of ethylene production. Likewise, the activity of ACC oxidase isolated from matriconditioned seeds was higher than that from untreated seeds. Higher endo--mannanase and total dehydrogenase activities were observed in primed air-dried seeds in comparing to non-primed seeds.  相似文献

Onion seeds were osmotically primed in polyethylene glycol solution (342 g/kg water) either for 14 days before accelerated ageing at 40°C. 18% m.c. for 0, 24, 48, 72 or 96 h, or for 10, 14 or 17 days after ageing. Priming improved the rate of germination compared with non-primed seed. Priming before ageing delayed the loss of viability due to ageing, but priming after ageing had no effect on viability. Primed and dried onion seed was stored for 18 months at 10°C, 9% m.c. with no effect on viability; improvements in germination rate due to priming were maintained over the storage period. Conductivity measurements of seed leachates were not a consistently reliable indicator of germination performance.  相似文献

Peanut seeds (water content 6.21% ) were stored at 38–40 ℃ for artificial ageing storage with different gases. N2 or CO2 delayed peanut seed ageing in comparison with air. The rate of seed germination was not affected by storing for 26 weeks under N2 or CO2, but the vitality of seed lost when stored under air. Determination of peanut seed vigor (root length+ axial length)×percentage of germination may precisely show the change in seed quality during ageing. The results indicate the positive correlation between the respiration and subsequent growth in peanut seeds, and the significantly negative correlation between vigor and electrical conductivity or sugar content in the seed extract.  相似文献

The effects of storage conditions on the germination of developingmuskmelon (Cucumis melo L.) seeds were tested to determine whetherafter-ripening is required to obtain maximum seed vigour. Seedswere harvested at 5 d intervals from 35 (immature) to 60 (fullymature) days after anthesis (DAA), washed, dried, and storedat water contents of 3·3 to 19% (dry weight basis) at6, 20, or 30°C for up to one year. Germination was testedin water and in polyethylene glycol 8000 solutions ( –0·2to –1·2 MPa osmotic potential) at 15, 20, 25 or30°C. Germination percentages and rates (inverse of meantimes to radicle emergence) were compared to those of newlyharvested, washed and dried seeds. For 40 and 60 DAA seeds,one year of storage at 20°C and water contents <6·5%significantly increased germination percentages and rates at20°C, but had little effect on germination at 25 and 30°C.Storage reduced the estimated base temperature (Tb) and meanbase water potential (b) for germination of both 40 and 60 DAAseeds by approximately 5°C and 0·3 MPa, respectively.Immature 35 DAA seeds showed the greatest benefit from storageat 3 to 5% water content and 30°C, as germination percentagesand rates increased at all water potentials (). Storage underthese same conditions had little effect on the germination ofmature seeds in water, but increased germination percentagesand rates at reduced 's. Accelerated ageing for one month at30°C and water contents from 15 to 19° increased germinationrates and percentages of mature seeds at reduced 's, but longerdurations resulted in sharp declines in both parameters. Immatureseeds lost viability within one month under accelerated ageingconditions. An after-ripening period is required at all stagesof muskmelon seed development to expand the temperature andwater potential ranges allowing germination and to achieve maximumgerminability and vigour. Post-harvest dormancy is deepest atthe point of maximum seed dry weight accumulation and declinesthereafter, both in situ within the ripening fruit and duringdry storage. Germination and vigour of accelerated aged (AA) and naturally stored onion seeds were examined. Accelerated ageing was conducted at 40 °C and 100 % relative humidity (RH). Non aged seeds were stored for 34 months at 3 or 15 °C and 40, 60 or 90 % RH. To restore seed viability, stored and aged seeds were matriconditioned with Micro-Cel E. A distinct loss of germination was observed after 5 days of accelerated ageing. Naturally stored seeds maintained high viability for 34 months, when stored at 3 °C and 40, 60 and 90 % RH or at 15 °C and 40 %. An increase of RH to 60 and 90 % at 15 °C caused loss of germination and vigour. Matriconditioning improved germination and increased endogenic ethylene release and in vivo ACC oxidase activity of both aged and stored seeds.  相似文献

Samples of three seed lots of each of three cultivars of carrot, celery and onion were primed in polyethylene glycol solution for 2 weeks at 15 °C. Priming reduced the mean germination times (recorded at 15 °C) of all seed lots (compared to the untreated control) by 3–4 days in carrot, 6–10 days in celery and 3–5 days in onion. The largest reductions in mean germination time occurred in the slowest-germinating seed lots. There were highly significant interactions between priming and cultivars, and between priming and seed lots within cultivars for each species. Drying back the primed seeds at 15 °C increased the mean germination times (compared to primed seed which had not been dried) by 0·6 day in carrot and 1·4 days in celery, and there was no interaction with cultivars or seed lots. The corresponding increase for onion was either 1·0 or 1·8 days, according to the cultivar, but this variation was largely attributable to differences in time taken for the dried seeds to re-imbibe. Seeds dried back at 30 °C germinated 0·2·0·7 day (depending on the species) later than those dried at 15 °C. Percentage germination was not affected by either priming or drying back. Priming reduced the spread of germination times in all cultivars. For primed and dried-back seed, the spread of germination times was larger than that of primed seed in certain cultivars, but was always smaller than that of untreated seeds.  相似文献

The Effects of Priming and Ageing on Seed Vigour in Tomato   总被引:1,自引:0,他引:1  
A comparison was made of the effects of seed priming or ageingtreatments on the performance of tomato (Lycopersicon esculentumMill. cv. UC204C) seeds according to a number of indices ofseed vigour. A single lot of tomato seeds was primed in 120mol m–3 K2HPO4 + 150 mol m–3 KNO3 for 5 d at 20?C, or aged at 13% moisture content (dry weight basis) and 50?C for 6 d. Germination percentage (>98%) was unaffectedby priming and reduced to 85% by ageing. X-ray photographs andlongitudinal sections revealed the formation of free space surroundingthe embryo in dry primed seeds, which was not evident in controlor aged seeds. Priming increased the rate of germination atall temperatures above the base temperature (Tb), while ageingdecreased it. Tb was unaffected by priming and only slightlyincreased by ageing. The variation in individual times to germinationwas approximately doubled in both primed and aged seed comparedto the control, based upon the slopes of probit germinationpercentage versus log thermal time curves. Root growth aftergermination tests and seedling growth in both greenhouse andfield tests were not influenced by either priming or ageing.The conductivity test was found to be unreliable as a vigourtest for tomato seeds. The results identify several indiceswhich can be used to quantify seed vigour in tomato. They alsoillustrate that seed priming can enhance seed performance accordingto some criteria, while having no effect or decreasing qualityaccording to other criteria. Seed vigour can apparently be separatedinto various components which can be independently influencedby seed enhancement treatments.
