Optimal multivariate matching before randomization   总被引:1,自引:0,他引:1  
Although blocking or pairing before randomization is a basic principle of experimental design, the principle is almost invariably applied to at most one or two blocking variables. Here, we discuss the use of optimal multivariate matching prior to randomization to improve covariate balance for many variables at the same time, presenting an algorithm and a case-study of its performance. The method is useful when all subjects, or large groups of subjects, are randomized at the same time. Optimal matching divides a single group of 2n subjects into n pairs to minimize covariate differences within pairs-the so-called nonbipartite matching problem-then one subject in each pair is picked at random for treatment, the other being assigned to control. Using the baseline covariate data for 132 patients from an actual, unmatched, randomized experiment, we construct 66 pairs matching for 14 covariates. We then create 10000 unmatched and 10000 matched randomized experiments by repeatedly randomizing the 132 patients, and compare the covariate balance with and without matching. By every measure, every one of the 14 covariates was substantially better balanced when randomization was performed within matched pairs. Even after covariance adjustment for chance imbalances in the 14 covariates, matched randomizations provided more accurate estimates than unmatched randomizations, the increase in accuracy being equivalent to, on average, a 7% increase in sample size. In randomization tests of no treatment effect, matched randomizations using the signed rank test had substantially higher power than unmatched randomizations using the rank sum test, even when only 2 of 14 covariates were relevant to a simulated response. Unmatched randomizations experienced rare disasters which were consistently avoided by matched randomizations.  相似文献

Herbivory is an ecological process that is known to generate different patterns of selection on defensive plant traits across populations. Studies on this topic could greatly benefit from the general framework of the Geographic Mosaic Theory of Coevolution (GMT). Here, we hypothesize that herbivory represents a strong pressure for extrafloral nectary (EFN) bearing plants, with differences in herbivore and ant visitor assemblages leading to different evolutionary pressures among localities and ultimately to differences in EFN abundance and function. In this study, we investigate this hypothesis by analyzing 10 populations of Anemopaegma album (30 individuals per population) distributed through ca. 600 km of Neotropical savanna and covering most of the geographic range of this plant species. A common garden experiment revealed a phenotypic differentiation in EFN abundance, in which field and experimental plants showed a similar pattern of EFN variation among populations. We also did not find significant correlations between EFN traits and ant abundance, herbivory and plant performance across localities. Instead, a more complex pattern of ant–EFN variation, a geographic mosaic, emerged throughout the geographical range of A. album. We modeled the functional relationship between EFNs and ant traits across ant species and extended this phenotypic interface to characterize local situations of phenotypic matching and mismatching at the population level. Two distinct types of phenotypic matching emerged throughout populations: (1) a population with smaller ants (Crematogaster crinosa) matched with low abundance of EFNs; and (2) seven populations with bigger ants (Camponotus species) matched with higher EFN abundances. Three matched populations showed the highest plant performance and narrower variance of EFN abundance, representing potential plant evolutionary hotspots. Cases of mismatched and matched populations with the lowest performance were associated with abundant and highly detrimental herbivores. Our findings provide insights on the ecology and evolution of plant–ant guarding systems, and suggest new directions to research on facultative mutualistic interactions at wide geographic scales.  相似文献

Cebus monkeys (C. apella, C. albifrons, andC. capuchin) and rhesus monkeys (Macaca mulatta) were compared with respect to their performance on a battery of 6 tests. The battery consisted of tests of delayed response, oddity problems, matching problems, contingent response and delayed alternation. A suggestive trend was noted, with cebus performing relatively better on long delay problems and the oddity problem, and rhesus performing relatively better on the delayed alternation, matching and contingency problems. However, no statistically significant differences were found between the two groups on any of the tests. In everyday life contexts and work settings, monetary rewards are often contingent on future performance. Based on research showing that the anticipation of rewards causes improved task performance through enhanced task preparation, the present study tested the hypothesis that the promise of monetary rewards for future performance would not only increase future performance, but also performance on an unrewarded intermediate task. Participants performed an auditory Simon task in which they responded to two consecutive tones. While participants could earn high vs. low monetary rewards for fast responses to every second tone, their responses to the first tone were not rewarded. Moreover, we compared performance under conditions in which reward information could prompt strategic performance adjustments (i.e., when reward information was presented for a relatively long duration) to conditions preventing strategic performance adjustments (i.e., when reward information was presented very briefly). Results showed that high (vs. low) rewards sped up both rewarded and intermediate, unrewarded responses, and the effect was independent of the duration of reward presentation. Moreover, long presentation led to a speed-accuracy trade-off for both rewarded and unrewarded tones, whereas short presentation sped up responses to rewarded and unrewarded tones without this trade-off. These results suggest that high rewards for future performance boost intermediate performance due to enhanced task preparation, and they do so regardless whether people respond to rewards in a strategic or non-strategic manner.  相似文献

The QUEST system for quantitative analysis of two-dimensional gels   总被引:25,自引:0,他引:25  
The strategies and methods used by the QUEST system for two-dimensional gel analysis are described, and the performance of the system is evaluated. Radiolabeled proteins, resolved on two-dimensional gels and detected using calibrated exposures to film, are quantified in units of disintegrations per minute or as a fraction of the total protein radioactivity applied to the gel. Spot quantitation and resolution of overlapping spots is performed by two-dimensional gaussian fitting. Pattern matching is carried out for groups of gels called matchsets, and within each matchset every gel is matched to every other gel. During the matching process, spots are automatically added to each pattern at positions where unmatched spots were detected in other patterns. This results in enhanced accuracy for both spot detection and for matching. The spot fitting procedure is repeated after matching. Tests show that up to 97% of spots in each pattern can be matched and that fewer than 1% of the spots are matched inconsistently. Approximately 2000 proteins are detected from typical gels. Of these 1600 are high quality spots. Tests to measure the coefficient of variation of spot quantitation versus spot quality show that the average coefficient of variation for high quality spots is 21%. The intensities of the detected proteins range from 4 to 20,000 ppm of total protein synthesis. The QUEST analysis system has been used to build a quantitative database for the proteins of normal and transformed REF52 cells, as presented in the accompanying reports (Garrels, J., and Franza, B. Dynamics of neuronal interaction recorded by microelectrodes were examined in 90 arrays of cells of the human thalamic reticular nucleus (Rt) during stereotaxic surgical procedures. Cooperative interaction between adjacent neurons was found to occur in neuronal arrays after presentation of verbal (or sensory-cum-acoustic) functionally significant stimulus (FSS) as well as at stages of initiation and performance of goal-directed movement. Specialized dynamics were noted in the pattern of interaction between neuronal arrays of two types (A and B) with irregular background activity and 2–5 Hz bursting rhythm (types A and B respectively). This dynamic local neuronal interaction correlates with the stage of significant verbal stimulus presentation and that of performing goal-directed movements. Song-type matching during territorial contests may allow males of some bird species to direct their signal to a particular receiver. By matching the song-type and also responding immediately to a rival (temporal matching), a signalling male may indicate his willingness to escalate the contest. Black-capped chickadees ( Poecile atricapilla ) sing a single song-type, but are able to sing this song over a wide range of absolute frequencies. By using interactive playback to instigate and control the level of matching during trials, we investigated whether matching the frequency and the temporal patterning of song escalates contests. Males that were matched for both the frequency and the temporal pattern of their songs during trials escalated contests more than males that were not matched, while males that were only matched temporally had an intermediate response. During trials that consisted of temporal matching only, focal males often shifted frequency to match the playback. Our results confirm that frequency matching and temporal matching using a single song-type allows graded signalling during aggressive interactions in chickadees.  相似文献

Survival of patients with lung cancer could be significantly prolonged should the disease be diagnosed early. Growing evidence indicates that the immune response in the form of autoantibodies to developing cancer is present before clinical presentation. We used a phage-displayed antibody library to select for recombinant scFvs that specifically bind to lung cancer-associated IgM autoantibodies. We selected for scFv recombinant antibodies reactive with circulating IgM autoantibodies found in the serum of patients with early stage lung adenocarcinoma but not matched controls. Discriminatory performance of 6 selected scFvs was validated in an independent set of serum from stage 1 adenocarcinoma and matching control groups using two independent novel methods developed for this application. The panel of 6 selected scFvs predicted cancer based on seroreactivity value with sensitivity of 0.8 and specificity of 0.87. Receiver Operative Characteristic curve (ROC) for combined 6 scFv has an AUC of 0.88 (95%CI, 0.76–1.0) as determined by fluorometric microvolume assay technology (FMAT) The ROC curve generated using a homogeneous bridging Mesa Scale Discovery (MSD) assay had an AUC of 0.72 (95% CI, 0.59–0.85). The panel of all 6 antibodies demonstrated better discriminative power than any single scFv alone. The scFv panel also demonstrated the association between a high score - based on seroreactivity - with poor survival. Selected scFvs were able to recognize lung cancer associated IgM autoantibodies in patient serum as early as 21 months before the clinical presentation of disease. The panel of antibodies discovered represents a potential unique non-invasive molecular tool to detect an immune response specific to lung adenocarcinoma at an early stage of disease.  相似文献

Employers often make payment contingent on performance in order to motivate workers. We used fMRI with a novel incentivized skill task to examine the neural processes underlying behavioral responses to performance-based pay. We found that individuals' performance increased with increasing incentives; however, very high incentive levels led to the paradoxical consequence of worse performance. Between initial incentive presentation and task execution, striatal activity rapidly switched between activation and deactivation in response to?increasing incentives. Critically, decrements in performance and striatal deactivations were directly predicted by an independent measure of behavioral loss aversion. These results suggest that incentives associated with successful task performance are initially encoded as a potential gain; however, when actually performing a task, individuals encode the potential loss that would arise from failure.  相似文献

Propensity-score matching is frequently used in the medical literature to reduce or eliminate the effect of treatment selection bias when estimating the effect of treatments or exposures on outcomes using observational data. In propensity-score matching, pairs of treated and untreated subjects with similar propensity scores are formed. Recent systematic reviews of the use of propensity-score matching found that the large majority of researchers ignore the matched nature of the propensity-score matched sample when estimating the statistical significance of the treatment effect. We conducted a series of Monte Carlo simulations to examine the impact of ignoring the matched nature of the propensity-score matched sample on Type I error rates, coverage of confidence intervals, and variance estimation of the treatment effect. We examined estimating differences in means, relative risks, odds ratios, rate ratios from Poisson models, and hazard ratios from Cox regression models. We demonstrated that accounting for the matched nature of the propensity-score matched sample tended to result in type I error rates that were closer to the advertised level compared to when matching was not incorporated into the analyses. Similarly, accounting for the matched nature of the sample tended to result in confidence intervals with coverage rates that were closer to the nominal level, compared to when matching was not taken into account. Finally, accounting for the matched nature of the sample resulted in estimates of standard error that more closely reflected the sampling variability of the treatment effect compared to when matching was not taken into account.  相似文献

Earlier studies have shown variation among experimental attempts to establish whether human monozygotic twins that are genetically identical also have identical individual scents. In none of the cases were the dogs able to distinguish all the individual scents of monozygotic twins living in the same environment if the scents were presented to them separately. Ten specially trained police German Shepherd dogs of three Czech Republic Police Regional Headquarters were used for scent identification in our study. The dogs were supposed to match scents of two monozygotic pairs (5 and 7 years old) and two dizygotic twin pairs (8 and 13 years old). Scents were collected on cotton squares stored in glass jars. Dog handlers were blind to the experiment details. In each trial (line-up), one scent was used as a starting scent and the dog was then sent to determine if any of the 7 presented glass jars contained a matching scent. Scents of children of similar ages were used as distractors. In the matching procedure, the dogs matched correctly the scent of one twin with the other, as well as two scents collected from every single identical and non-identical twin to prove their efficacy and likewise, the presence of the matching twin scent in any given glass jar. All dogs in all trials distinguished correctly the scents of identical as well as non-identical twins. All dogs similarly matched positively two scents collected from the same individuals. Our findings indicated that specially trained German Shepherd dogs are able to distinguish individual scents of identical twins despite the fact that they live in the same environment, eat the same food and even if the scents are not presented to them simultaneously.  相似文献

Many species of territorial songbirds exhibit a behavior known as song-type matching, in which a male sings the same song type that his neighbor is singing. Song-type matching is associated with increased aggression, but researchers have not come to a consensus on its adaptive function. Building on studies that identify singing performance as a variable relevant to sexual selection, we hypothesize that higher-performance singers benefit from matching their opponent's song type because matching improves eavesdroppers' ability to compare the two males' performances. We present a model of song-type choice that predicts that males that can outperform their rivals benefit by matching. In contrast, lower-performance males should avoid both matching and being matched. Our hypothesis is compatible with some existing hypotheses of song-matching function, but it is not compatible with the hypothesis that song matching is a conventional signal of aggression. We offer unique predictions that could be used to test our idea. We speculate that lower-performance individuals might have driven the evolution of repertoire complexity because they stand to benefit from novel, unmatchable songs. The phenomenon that dissimilar signals are less accurately compared than similar signals may favor the evolution of multiple ornaments and of plastic signal development (e.g., song learning) in general.  相似文献

We tested whether squirrel monkeys have cross-modal representations of their human caretakers with a 0-delay symbolic matching-to-sample procedure. We first trained the monkeys to match photographs of two of their caretakers. After reaching criterion, they were exposed to two test sessions. In these sessions 32 all-reinforced test trials were interspersed among the training trials. In the test trials, a voice, either matching (congruent condition) or mismatching (incongruent condition) with the sample photographs, was played back after the sample stimulus disappeared. The monkeys' matching accuracies in the incongruent condition were lower than in the match condition. Post hoc analyses revealed that the presentation of the primary caretaker's voice interfered with performance in test trials where the secondary caretaker's face was presented (incongruent condition). This suggests that our subjects recalled their primary caretaker's representation upon hearing the appropriate voice.  相似文献

In recent years, electron tomography has improved our three-dimensional (3D) insight in the structural architecture of cells and organelles. For studies that involve the 3D imaging of stained sections, manual annotation of tomographic data has been an important method to help understand the overall 3D morphology of cellular compartments. Here, we postulate that template matching can provide a tool for more objective annotation and contouring of cellular structures. Also, this technique can extract information hitherto unharvested in tomographic studies. To evaluate the performance of template matching on tomograms of stained sections, we generated several templates representing a piece of microtubule or patches of membranes of different staining-thicknesses. These templates were matched to tomograms of stained electron microscopy sections. Both microtubules and ER-Golgi membranes could be detected using this method. By matching cuboids of different thicknesses, we were able to distinguish between coated and non-coated endosomal membrane-domains. Finally, heterogeneity in staining-thickness of endosomes could be observed. Template matching can be a useful addition to existing annotation-methods, and provide additional insights in cellular architecture.  相似文献

Fingerprint, as one of the most popular and robust biometric traits, can be used in automatic identification and verification systems to identify individuals. Fingerprint matching is a vital and challenging issue in fingerprint recognition systems. Most fingerprint matching algorithms are minutiae-based. The minutiae points are the ways that the fingerprint ridges can be discontinuous. Ridge ending and ridge bifurcation are two frequently used minutiae in most fingerprint matching algorithms. This article presents a new minutiae-based fingerprint matching using the onion peeling approach. In the proposed method, fingerprints are aligned to find the matched minutiae points. Then, the nested convex polygons of matched minutiae points are constructed and the comparison between peer-to-peer polygons is performed by the turning function distance. Simplicity, accuracy, and low time complexity of the onion peeling approach are three important factors that make it a standard method for fingerprint matching purposes. The performance of the proposed algorithm is evaluated on the database FVC2002. Since the fingerprints that the difference between the number of their layers is more than 2 and the a minutiae matching score lower than 0.15 are ignored, better results are obtained.KeywordsFingerprint Matching, Minutiae, Convex Layers, Turning Function, Computational Geometry.  相似文献

Unlike other vertebrates, humans cooperate in large groups with unrelated individuals. Many authors have argued that the evolution of such cooperation has resulted from reciprocity and other forms of contingent cooperation. This argument is not well supported by existing theory. The theory of contingent cooperation in pairs is well developed: reciprocating strategies are stable when common, and can increase when rare as long as population structure leads to modest levels of relatedness. In larger groups, however, it is not clear whether contingent cooperation can increase when rare. Existing work suggests that contingent strategies cannot increase unless relatedness is high, but depends on unrealistic assumptions about the effects of population structure. Here we develop and analyze a model incorporating a two level population structure that captures important features of human hunter–gatherer societies. This model suggests that previous work underestimates the range of conditions under which contingent cooperation can evolve, but also predicts that cooperation will not evolve unless (1) social groups are small, and (2) the relatedness within ethnolinguistic groups is at the high end of the range of empirical estimates.  相似文献

The present experiment examined the effects of several test manipulations on discrimination, accuracy and sensitivity to reinforcer frequency in a conditional discrimination. Four pigeons responded on a multiple schedule of matching to sample procedures in which the reinforcer-frequency ratio for correct comparison choice responding was varied across components within session from 1:9 to 9:1. Following stability, the effects of prefeeding, extinction, and distraction during sample and comparison presentation were assessed. Discrimination accuracy decreased under prefeeding, extinction, and distraction during sample presentation. Sensitivity to reinforcer frequency decreased under prefeeding and extinction. Decreases in sensitivity were positively related to decreases in discrimination accuracy. The decreases in discrimination accuracy and sensitivity under prefeeding and extinction are interpreted as being due to decreases in attending to the sample and comparison stimuli, respectively, possibly mediated by motivational effects of these manipulations. This interpretation is consistent with current conceptualizations of the contingencies that govern conditional-discrimination performance.  相似文献

This is a study of the ability of blindfolded human subjects to match the position of their forearms before and after eccentric exercise. The hypothesis tested was that the sense of effort contributed to forearm position sense. The fall in force after the exercise was predicted to alter the relationship between effort and force and thereby induce position errors. In the arms-in-front posture, subjects had their unsupported reference arm set to one of two angles from the horizontal, 30 or 60 degrees , and they matched its position by voluntary placement of their other arm. Matching errors were compared with a task where the arms were counterweighted, so could be moved in the vertical plane with minimal effort, and where the arms were moved in the horizontal plane. In these latter two tasks, the intention was to test whether removal of an effort sensation from holding the arm against gravity influenced matching performance. It was found that, although absolute errors for counterweighted and horizontal matching were no larger than for unsupported matching, their standard deviations, 6.1 and 6.8 degrees , respectively, were significantly greater than for unsupported matching (4.6 degrees ), indicating more erratic matching. The eccentric exercise led, the next day, to a fall in maximum voluntary muscle torque of >or=15%. This was accompanied by a significant increase in matching errors for the unsupported matching task from 2.7 +/- 0.5 to 0.8 +/- 0.7 degrees but not for counterweighted (1.4 +/- 0.2 to -0.2 degrees +/- 1.1 degrees ) or horizontal matching (-1.3 +/- 0.7 degrees to -1.8 +/- 0.7 degrees ). This, it is postulated, is because the reduced voluntary torque after exercise was accompanied by a greater effort required to support the arms, leading to larger matching errors. However, effort is only able to provide positional information for unsupported matching where gravity plays a role. In gravity-neutral tasks like counterweighted or horizontal matching, a change in the effort-force relationship after exercise leaves matching accuracy unaffected.  相似文献
