The effect of iron supply on the growth and nodulation ofLupinus angustifolius L. (Gungurru),Lupinus luteus L. (R-1171) andLupinus pilosus Murr. (P20957) was studied in acid solutions. Plants of the three species were grown together in the same solution and inoculated withBradyrhizobium (Lupinus) WU 425. Plants were then grown with or without applied NH4NO3. The lupin species differed greatly in their sensitivity to low iron concentrations in solution withL. pilosus being most tolerant andL. luteus most sensitive.L. pilosus had the highest iron concentration in tissues and had a higher ratio of iron concentration in the youngest fully expanded leaf blades (YEB) to that in roots than the other two species.L. luteus had higher iron concentrations in roots but lower iron concentration in YEB and shoots than didL. angustifolius. The requirements of internal iron for the maximal chlorophyll synthesis in YEB were 65 μg g-1 forL. angustifolius andL. luteus, and 52 μg g-1 forL. pilosus. In contrast to effects on growth, the three species had similar external iron requirements for nodule formation in roots and for maximal nitrogen concentrations in shoots. The results indicate that iron tolerant lupin species require lower internal and external iron supply and have a greater ability than sensitive species to translocate iron from roots to shoots.  相似文献

Spread of necrotic and non‐necrotic strains of Bean yellow mosaic virus (BYMV) was compared when aphid vectors moved both types from external or internal virus sources to plots of Lupinus spp. (lupin). Regardless of whether virus sources were internal or external, removed or left in place, and spread was within plots with homologous sources or across buffers to plots containing the opposite type of virus source, non‐necrotic BYMV always spread faster than necrotic BYMV in plots of L. angustifolius (narrow‐leafed lupin). When necrotic BYMV spread from external sources into plots sown with two L. angustifolius genotypes differing in their necrosis responses to different BYMV strain groups and one genotype of L. luteus (yellow lupin) giving only non‐necrotic responses, differing symptom reactions in the two L. angustifolius genotypes revealed presence of two distinct necrotic BYMV strain groups and overall virus spread was greater in this species than in L. luteus. Spread of non‐necrotic BYMV in L. angustifolius was always polycyclic in nature. However, when it came initially from external sources, spread of necrotic BYMV was largely monocyclic. This work demonstrates how temporal virus spread can be diminished when hypersensitive (necrotic) resistance is deployed and the limitations associated with employing hypersensitivity that is strain specific.  相似文献

We examined the capacity of several Old-World lupin species (Lupinus luteus L., L. hispanicus Boiss. et Reuter and L. angustifolius L.) and one species of a New-World lupin (L. mutabilis Sweet) to form cluster roots under a range of conditions in solution culture. The effect of the synthetic auxin, IBA (indole-3-butyric acid), on cluster-root development in L. luteus and L. albus L. provided with an adequate phosphorus (P) supply was also investigated. In addition, the effect of a high nitrate-N (NO3-N) supply on the efflux of citrate and malate from roots of L. angustifolius was examined to determine if specific regions of the root system exuded these organic anions. When P-deficient, L. hispanicus, L. luteus and L. mutabilis formed cluster roots that secreted organic anions. Citrate was generally the dominant organic anion exuded, although succinate was also exuded in large quantities from L. luteus. Citrate efflux by L. hispanicus and L. luteus was at least comparable to that reported for P-deficient L. albus[up to 1.092 nmol g–1 fresh weight (FW) s–1], but was over an order of magnitude lower in L. mutabilis (0.036 nmol g–1 FW s–1). Citrate and malate were not detected in significant amounts from either the lateral roots or the root tips of any species grown under P-sufficient or -deficient conditions. Citrate efflux from cluster roots of L. luteus showed a diurnal pattern, similar to that reported for L. albus, with maximum efflux during the day, and declining to a minimum before dawn. IBA added to the nutrient solution induced cluster-root formation on both L. albus and L. luteus at concentrations of P that would normally suppress the production of these roots. However, the IBA-induced cluster roots did not exude significant amounts of citrate. Although L. angustifolius did not produce cluster roots when P-deficient, it produced cluster-like root structures that exuded citrate (0.053 nmol g–1 FW s–1) when grown at a high nitrate-N (NO3-N) supply. L. angustifolius did not exude significant citrate or malate from lateral roots or root tips when grown at either high or low NO3-N supply. Our findings for L. hispanicus and L. luteus are the first reports of cluster-root formation in response to P deficiency for these Old-World species, and for L. mutabilis, it is the first report of cluster roots for a New-World lupin species. These reports indicate that evolutionary and biogeographical aspects of cluster-root formation in the genus Lupinus need to be revised. Furthermore, investigation is warranted to determine the capacity of species of the large group of New-World lupins to form cluster roots in soils of their native habitats.  相似文献

Two molecular cytogenetics methods, PRINS (primed in situ DNA labeling) and C-PRINS (cycling PRINS), were optimized for the physical mapping of several types of DNA sequences on the mitotic chromosomes of the narrow-leafed lupin (Lupinus angustifolius L.). The fragment of the FokI element from Vicia faba was localised by indirect PRINS reaction. Two other sequences, fragments of the coding sequences of L. luteus and of L. angustifolius, were localised by indirect C-PRINS. These techniques are faster and more sensitive than FISH, and they allowed the mapping of short DNA fragments. The data obtained shows that both types of PRINS are valuable tools for chromosome identification in lupin.  相似文献

The weevils Sitona gressorius and Sitona griseus are specialist herbivores on lupins in Europe. The adult weevils feed on the leaves, and the larvae on the root nodules of the plants. This causes severe damage to lupin crops. In the present study, the feeding preferences of lupin weevil adults on different lupin genotypes were examined with respect to a possible effect of lupin alkaloids on host selection. A total of 12 genotypes from the species Lupinus albus, L. angustifolius, L. luteus, and L. nanus were grown in a field experiment and the feeding damage on the leaves caused by naturally occurring lupin weevil adults was estimated. Additionally, a feeding choice test with S. gressorius adults was performed to examine feeding preferences under laboratory conditions. A gas chromatographic analysis provided information on the alkaloid content and profiles in the leaves of the tested lupin genotypes. In the field experiment, significant differences in the extent of the feeding damage within the 12 lupin genotypes were observed. The dual-choice feeding bioassay did not show discrimination of lupin species, but two L. angustifolius genotypes were significantly less affected than the standard L. luteus “Bornal”. The alkaloid analysis revealed large contrasts in alkaloid concentrations and profiles in the leaves of the tested genotypes. Correlation analysis with the results from the field and laboratory did not indicate a significant influence of the total foliar alkaloid content on the extent of weevil feeding.  相似文献

The feeding behavior of cowpea aphid, Aphis craccivora Koch (Homoptera: Aphididae) was examined on seedlings of narrow leafed lupin, Lupinus angustifolius L., and yellow lupin, L. luteus L., using electronic monitoring of insect feeding behavior (EMIF). Aphid feeding behavior was first compared between resistant (cv. Kalya) and susceptible (cv. Tallerack) varieties of narrow-leafed lupin. Aphids spent significantly more time in non- penetration and stylet pathway activities, and significantly less time in the sieve element phase on Kalya than on Tallerack, suggesting that feeding deterrence is an important component of aphid resistance in Kalya. Aphid feeding on a susceptible yellow lupin variety (cv. Wodjil) was then compared with that on two resistant lines, one (Teo) with high and the other (94D024-1) with low seed alkaloid content. There were no consistent differences in aphid feeding behavior between Wodjil and Teo. Total, mean and percentage sieve element phase times were significantly lower, and total and percentage times in non-phloem phase were greater on 94D024-1 than on Wodjil, suggesting the possibility of phloem-based deterrence in 94D024-1.  相似文献

Experiments were conducted to investigate the potential induction of plant defenses by Myzus persicae Sulzer (Homoptera: Aphididae) feeding on five lupin, Lupinus spp. (Leguminosae), varieties with well‐characterized levels of aphid resistance. Myzus persicae feeding on L. angustifolius and L. luteus varieties induced genotype‐specific changes in their host that were not consistent with the level of aphid resistance or the plant species. The plant responses were systemically detected by apterous and alate forms of the aphids. Chemical assays revealed no induction of oxidizing enzyme (catalase, peroxidase, or polyphenol oxidase) activity, serine or cystein proteinase inhibitors, or soluble phenolics in any of the five varieties tested following 3 days of feeding by 10 or 30 aphids. However, there were significant differences among the five lupin varieties in the levels of peroxidase and polyphenol oxidase activity, proteinase inhibitors, and soluble phenolics.  相似文献

Forty three Bradyrhizobium strains isolated in Poland from root nodules of lupin species (Lupinus albus, L. angustifolius and L. luteus), and pink serradella (Ornithopus sativus) were examined based on phylogenetic analyses of three housekeeping (atpD, glnII and recA) and nodulation (nodA) gene sequences. Additionally, seven strains originating from root-nodules of yellow serradella (O. compressus) from Asinara Island (Italy) were included in this study. Phylogenetic trees revealed that 15 serradella strains, including all yellow serradella isolates, and six lupin strains grouped in Bradyrhizobium canariense (BC) clade, whereas eight strains from pink serradella and 15 lupin strains were assigned to Bradyrhizobium japonicum (BJ1). Apparently, these species are the two dominant groups in soils of central Europe, in the nodules of lupin and serradella plants. Only three strains belonged to other chromosomal lineages: one formed a cluster that was sister to B. canariense, one strain grouped outside the branch formed by B. japonicum super-group, and one strain occupied a distant position in the genus Bradyrhizobium, clustering with strains of the Rhodopseudomonas genus. All strains in nodulation nodA gene tree grouped in a cluster referred to as Clade II, which is in line with earlier data on this clade dominance among Bradyrhizobium strains in Europe. The nodA tree revealed four well-supported subgroups within Clade II (II.1-II.4). Interestingly, all B. canariense strains clustered in subgroup II.1 whereas B. japonicum strains dominated subgroups II.2-II.4.  相似文献

Summary Interspecific hybridization has a role to play in the domestication of wild species through the introgression of desirable genes from related domesticated species. Almost complete genetic isolation among the Mediterranean-African rough-seeded lupin species has hitherto ruled out introgression within that group. Recent work in Western Australia with L. atlanticus, L. cosentinii, L. pilosus and L. digitatus has nevertheless suggested that hybrid sterility may be overcome if specially selected lines of each are used in crossing. We have now selected F2-F5 plants and backcrosses (Bc1; Bc2) from L. atlanticus/L. cosentinii, some with improvement in seed fertility combined with domestication genes from either species. Relatively better F1-F2 plant fertility in subsequent crosses of L. atlanticus/L. digitatus than in L. atlanticus/L. cosentinii or L. digitatus/L. cosentinii indicate closer relationship between L. atlanticus and L. digitatus than in the other cross combinations. Use of embryo culture may help to overcome L. pilosus/L. atlanticus F1 plant sterility. Some of the interspecific selections could act as genetic bridges between L. cosentinii and L. pilosus.  相似文献

PAGE and SDS-PAGE electrophoretic analysis of 31 accessions of 10 Old-WorldLupinus species, 5 smooth-seeded and 5 rough-seeded, covered total seed albumins and 2S albumins isolated by a solid-phase extraction. PAGE albumin patterns showed a distinct difference between the smooth- and rough-seeded lupins. SDS-PAGE analysis of seed albumins revealed interspecific differences, mainly due to the 2S albumins. The differences were especially marked in the smooth-seeded species. In the rough-seeded lupins the following subgroups were distinguished: (1)L. atlanticus, (2)L. cosentinii andL. digitatus, (3)L. palaestinus andL. pilosus. Evidence was provided that the 2S albumin class contains conglutin , so far classified as a globulin. The results are discussed with reference to taxonomic relationships of the Old-World lupins and characterization of the lupin seed albumin fraction.  相似文献

The complete protocols for long-term micropropagation of some cultivars of four lupin species: Lupinus luteus, L. albus, L. angustifolius and L. mutabilis were elaborated. The shoots were regenerated in vitro via induction of axillary buds development. Plantlets were multiplicated on lowered salts MS-derived media containing BAP in diverse and generally low concentrations. Significant differences in regeneration capacity between species and cultivars were observed. The highest multiplication ratio revealed L. mutabilis and L. luteus. Regenerated shoots were rooted in vitro on low-salts MS-derived media with B5 vitamins. Media were supplemented with different auxins that affected roots formation of particular species and cultivars. Rooting ability of regenerated shoots decreased rapidly through in vitro culture. For that reason, grafting was applied as an alternative method of transfer of shoots to in vivo conditions. This method turned out to be successful for the majority of studied species and cultivars.  Transgenic yellow lupin (Lupinus luteus L.) plants have been generated by meristem co-cultivation with Agrobacterium tumefaciens. The binary plasmid pPZBNIa contains the bar gene under the control of a CaMV 35 S promoter. The transformation method involves inoculation of embryonic axis explants with A. tumefaciens, flooding the meristem with glufosinate, and initial culture on non-selective medium. Shoots were transferred to culture medium containing 20 mg/l glufosinate. Following subculture, shoots were grafted onto non-transgenic narrow-leafed lupin (L. angustifolius L.) seedling rootstocks, or rooted in vitro. The overall transformation efficiency, as determined at the T1 generation, was 0.05%–0.75%. The transgenic nature of plants grown to the T6 generation was confirmed by phosphinothricin acetyl transferase, PCR and Southern analyses. Defining plant adaptation to soil conditions is critical for the successful introduction of grain legume species into farming systems. This nutrient solution study examined the effects of pH (4, 5, 6, 7 and 8) and bicarbonate (5 mM KHCO3) on the growth and nodulation of 14 grain legume species supplied with N or reliant on N2-fixation. Species includedPisum sativum L.,Cicer arietinum L.,Lens culinaris Med., and a range ofLupinus, Vicia andLathyrus species. Species differed greatly in response to solution pH. For both N-fertilized and N2-fixing plants, shoot growth ofL. culinaris was very sensitive to low pH (pH < 7), whereas shoot growth ofLupinus angustifolius L. andLupinus albus L. was sensitive to higher pH (pH ≥ 6). Other species had a broader optimal pH range for growth when supplied with N, but were generally sensitive to low pH (pH < 7 forC. arietinum andVicia sativa L., pH < 6 forP. sativum, Vicia faba L.,Lathyrus sativus L. andLathyrus cicera L., and pH < 5 forVicia benghalensis L. andVicia narbonensis L.) when reliant on N2-fixation. For these other species, symbiotic N2-fixation appeared to be more sensitive than host plant growth to low pH. This finding was supported by lower nodule numbers and mass, and lower N concentrations in shoots of sensitive species at low pH relative to higher pH. ForL. culinaris, nodule numbers and mass were relatively unaffected by pH 5–8, N concentrations in shoots were high at low pH and plants developed symptoms relating to H+ toxicity at pH as high as 7. These results indicate that host plant growth ofL. culinaris is more sensitive to low pH than theRhizobium symbiosis. ForL. albus andL. angustifolius, both host plant growth and symbiotic N2-fixation appeared to be equally sensitive to pH ≥ 6.Lupinus pilosus Murr. was more tolerant of high pH than the otherLupinus species. At pH 4, two genotypes ofC. arietinum had better early nodulation than other species.Vicia ervilia L. nodulated poorly at all levels of solution pH, indicating that the commercial Group E inoculum (Rhizobium leguminosurum bv.viceae SU303) may not be effective for this species in solution culture. Addition of bicarbonate decreased shoot growth, nodulation and N concentrations in shoots of most species. Early nodulation (nodule number) ofLathyrus ochrus (L.) Summary A genomic DNA library of Lupinus luteus cv. Ventus was constructed in the phage vector EMBL3 using Mb oI-digested DNA. Screening with a 1070 bp labelled repetitive unit from L. luteus yielded several DNA clones. The repetitive family is composed of elements whose length is at least 16 kb. The average copy number of the cloned fragments is 5.0 × 104 per haploid genome and constitutes approximately 3% of the total L. luteus genome. The homologous repeats were found in all ten cultivars of L. luteus tested but were not detected in two cultivars each of the closely related species Lupinus albus and Lupinus angustifolius. The EcoRI family fragments could thus be considered as species-specific DNA elements. These fragments may be useful as molecular markers in the genetic manipulation of L. luteus.  相似文献

We report the first gene-based linkage map of Lupinus angustifolius (narrow-leafed lupin) and its comparison to the partially sequenced genome of Medicago truncatula. The map comprises 382 loci in 20 major linkage groups, two triplets, three pairs and 11 unlinked loci and is 1,846 cM in length. The map was generated from the segregation of 163 RFLP markers, 135 gene-based PCR markers, 75 AFLP and 4 AFLP-derived SCAR markers in a mapping population of 93 recombinant inbred lines, derived from a cross between domesticated and wild-type parents. This enabled the mapping of five major genes controlling key domestication traits in L. angustifolius. Using marker sequence data, the L. angustifolius genetic map was compared to the partially completed M. truncatula genome sequence. We found evidence of conserved synteny in some regions of the genome despite the wide evolutionary distance between these legume species. Conglutin δ, a 2S globulin, was purified and compared in six species or varieties of lupin seeds. A common pattern is suggested, present in all species, corresponding to a protein which could exist as a monomer or a dimer. The first form contains one subunit, from 11 to 16.2 kDa, according to the species. It possesses a quaternary structure closely related to conglutin δ1 and was previously described in the narrow-leaved lupin. The second form contains two similar subunits (23 to 26 kDa) and could be the conglutin δ2. These two subunits are associated even when SDS is used and are probably disulfide-linked subunits. Each subunit is composed of two disulfide-linked polypeptides. One is acidic with molecular weight from 14 to 17.3 kDa and the second is acidic to neutral, from 2.4 to 4.5 kDa. Three species (L. luteus, L. arboreus and L. pilosus) present a supplementary subunit, with different molecular weight and p than that previously described and which never associates in a dimer form. It has been purified in L. luteus. When native, this protein is oligomeric. The subunit of 12 kDa in this species is composed of a polypeptide of 9 kDa (pl 4.5) disulfide-linked to one of 3 kDa (pl 6.5). This supplementary protein remains partly associated with the first in the yellow lupin (L. luteus). It probably corresponds to a new protein, different from conglutin δ.  相似文献

A collection of 51 bean yellow mosaic virus (BYMV) isolates was transmitted from infected Trifolium subterraneum (subterranean clover) to Lupinus angustifolius (narrow‐leafed lupin) by Myzus persicae (green peach aphid). Depending on isolate and L. angiistifolius genotype used, two distinct responses developed in L. angustifolius plants, either systemic necrosis and plant death or non‐necrotic reactions of varying severity. Ten isolates caused necrosis and plant death in cv. Danja. However, when nine of these were inoculated to breeding line 90L423‐07‐13, seven induced non‐necrotic reactions, while two caused necrosis and plant death. Thirty seven isolates always produced non‐necrotic reactions regardless of genotype of L. angustifolius inoculated. Non‐necrotic and necrotic isolates originally came both from lupins and other species, and the non‐necrotic isolates were no less efficiently transmitted by M. persicae than the necrotic ones. When one isolate of each type was inoculated together to T. subterraneum and nine months later this culture was used as an acquisition source for aphid transmission to L. angustifolius, only the necrotic type was detected. Previous infection of L. angustifolius plants with a non necrolic isolate prevented subsequent infection by a necrotic one. All necrotic and non‐necrotic isolates reacted with BYMV antiserum in ELISA but only two cross‐reacted with antiserum to clover yellow vein virus (CYVV). When selected necrotic and non‐necrotic isolates were inoculated to differential hosts, all behaved like BYMV and not CYVV. When three isolates of each type were transmitted to 11 other cool season grain legume species, except in Cicer arietinum (chickpea), there were no necrotic reactions, but symptom severity varied with the isolate and species inoculated. The two isolates that caused necrosis in C. arietinum did not do so in L. angustifolius. The six isolates from Vicia faba (faba bean) all caused non‐necrotic reactions in L. angustifolius cv. Danja and 90L423‐07‐13. These and two necrotic isolates readily infected five genotypes of V. faba always causing severe symptoms. However, three non‐necrotic isolates from L. angustifolius and a further necrotic isolate were poorly infectious on V. faba in which they generally induced mild symptoms. These results show that at least three strain groups of BYMV can be distinguished by their reactions in different L. angustifolius genotypes, one causing necrosis and death in cv. Danja and 90L423‐07‐13, one causing necrosis and death in Danja but not 90L423‐07‐13, and one causing non‐nccrotic reactions in both. These strain groups could not be distinguished when representative necrotic and non‐necrotic isolates were inoculated to other grain legume species. However, inoculation to V. faba distinguished two other BYMV strain groupings differing in severity of symptoms and ability to infect this species.  相似文献

Amino acid composition and fatty acid composition were determined on seed samples of a range of white lupin (Lupinus albus) cultivars and accessions grown in either of two environments.Variability between genotypes was found for lysine, arginine and glutamic acid content, but not for the concentrations of other amino acids. The deficiency in sulphurcontaining amino acids, typical of legume proteins, was evident, with methionine and cyst(e)ine totalling only 2.2% of the protein. Variability was limited, indicating that improvement by breeding would be impracticable. Lupinus albus differed slightly from other lupin species in amino acid composition, having higher levels of threonine, tyrosine and isoleucine, but a lower level of glutamic acid than both L. angustifolius and L. luteus. Four low-alkaloid lines of L albus each had higher lysine content than the high-alkaloid line, but ‘Kiev Mutant’, despite earlier claims, had a lysine level no higher than the other three low-alkaloid lines.Fatty acid composition of the seed oil varied considerably between genotypes. Oleic acid ranged from 43.6 to 54.4% and linolenic acid from 6.7 to 15.2%, these two fatty acids being negatively correlated at one site. Linoleic acid content varied between 17.2 and 26.9% and was not correlated with other fatty acids. Total oil content averaged 9.6% with little variability between lines.It is concluded that, relative to other lupin species, L. albus has a more favourable amino acid profile for its utilisation in cereal-based diets for animals, particularly if the energy source is wheat, which is deficient in threonine. The higher oil content would be an important energy benefit to such diets and may allow their protein/energy balance to be maintained at higher levels of incorporation of L. albus seed meal than is possible with other lupin species.  相似文献

The aim of this study was to investigate the productive traits and fatty acid profiles of eight sweet varieties of Lupinus albus, Lupinus angustifolius and Lupinus luteus cultivated in a Mediterranean environment (Sicily, southern Italy). This is being done to identify the most suitable species, from an agronomical and nutritional point of view, for supporting animal husbandry in this area. Seed yield, oil quantity and oil quality were considerably different depending on the genotype. L. albus showed the highest yield (2.0–2.4 t/ha), 100 seed weight (27.0 g, on average), oil content (70.4–75.5 g/kg), and n?3/n?6 polyunsaturated fatty acid ratio (0.39–0.51), L. angustifolius showed the highest content of saturated fatty acids (21.03–26.66 g/100 g) and L. luteus showed the highest levels of n?6 polyunsaturated fatty acids (47.56–50.85 g/100 g), unsaturated/saturated (4.93–5.62) and polyunsaturated/saturated (3.33–3.99) fatty acid ratios and the best atherogenic (0.06) and thrombogenic (0.09–0.11) indices. The data obtained suggests L. albus as the most interesting lupin species for aiding the crop-livestock food chain and L. luteus as a promising crop due to its high nutritive traits for the Mediterranean environment.  相似文献
