Theoretical analyses of the genetic organization of pioneer species have postulated two very different scenarios. Some models have predicted that such species would show strong population substructuring, whereas other models have suggested that extinction and recolonization can augment gene flow and reduce interpopulation differentiation. We tested these alternative scenarios by analyzing the genetic structure of eight loci from populations of the pioneer dioecious tree, Cecropia obtusifolia, in the tropical rain forest region of Los Tuxtlas, México. The populations studied exhibit low overall FST values, no clear pattern of isolation by distance, and high estimates of gene flow. These results suggest either that the species is not at a genetic equilibrium under present levels of gene flow with populations derived from each other in the recent past, or that pollen and seed dispersal in this species occur over long distances (up to more than 100 km). Mating among relatives appears higher than expected by chance based on significantly positive fixation indices (F) and FIS values at some loci. However, no direct evidence for biparental inbreeding was found. The multilocus and single-locus outcrossing rates for C. obtusifolia were estimated at tm = 0.974 (SE = 0.024) and ts = 0.980 (SE = 0.035), respectively. These are not significantly different from 1, and the difference, tmts = — 0.006 (SE = 0.018), is not significantly different from 0. These estimates, however, could be biased because in all enzymes, except PGM-1, we found statistically significant departures from the mixed-mating model used to estimate them. Two rare alleles were found only in seeds collected from the soil, and the greatest number of different alleles were found also in soil seeds. It is hypothesized that the seed bank may play an important role in the genetic buffering of C. obtusifolia. Significantly positive or negative fixation indices in adults at some loci and significantly different heterozygosities among different life stages (from seeds to adults) suggest the action of selection at some loci.  相似文献

The mating system and allozyme variation at 20 loci in three Klamath Mountains and two Sierra Nevada populations of Jeffrey pine (Pinus jeffreyi Grev. & Balf.) were investigated. On average, multilocus estimates of the proportion of viable progeny due to outcrossing (tm) were high in all populations (mean tm = 0.935, range 0.881 to 0.971). Despite differences in stand structure, tm did not differ (P > 0.05) between the Klamath (mean tm = 0.933) and Sierra Nevada (mean tm = 0.937) populations. At all but one locus in one population and at two in another, genotype frequencies fit (P > 0.05) Hardy-Weinberg expectations. Mean estimates of observed heterozygosity in Klamath (0.182) and Sierra Nevada (0.327) populations were comparable to values reported for other conifers.  相似文献

Acacia cyclops, or Ngaamarur, is a common coastal shrub or small tree of the southwest of Western Australia and South Australia used for restoration in these landscapes and elsewhere. Knowledge of genetic diversity and mating systems of restoration populations is often lacking but can help inform likely restoration success. We compared genetic diversity and mating system parameters at three restoration populations of A. cyclops to nearby remnant reference populations. Mean levels of genetic diversity in restoration populations (Na = 1.806, Ne = 1.560, He = 0.270, Ho = 0.359) were not significantly different from those in the reference populations (Na = 1.833, Ne = 1.574, He = 0.269, Ho = 0.352) suggesting diverse seed collections were made for restoration. Allelic genetic divergence among restoration and reference populations was low (DST ≤ 0.04) suggesting that seeds were of local provenance. Mating system parameters were similar among restoration (tm = 0.989, tm–ts = 0.010, rpm = 0.392) and reference populations (tm = 0.868, tm–ts = 0.039, rpm = 0.410), suggesting functionality of the mating system and restitution of generalist insect pollinator services in restored populations at 7, 10, and 12 years after planting. Lower levels of heterozygosity in progeny compared to adults suggest post‐germination or late‐acting selection against inbred seedlings. Evidence for selection and seedling attrition further emphasizes the benefits of collections from many individuals in well‐connected outcrossed source populations. Overall, results suggest available local genetic diversity was captured and restoration populations have mating systems equivalent to those of reference populations.  相似文献

Seasonal and annual respiration of a ponderosa pine ecosystem   总被引:2,自引:0,他引:2  
The net ecosystem exchange of CO2 between forests and the atmosphere, measured by eddy covariance, is the small difference between two large fluxes of photosynthesis and respiration. Chamber measurements of soil surface CO2 efflux (Fs), wood respiration (Fw) and foliage respiration (Ff) help identify the contributions of these individual components to net ecosystem exchange. Models developed from the chamber data also provide independent estimates of respiration costs. We measured CO2 efflux with chambers periodically in 1996–97 in a ponderosa pine forest in Oregon, scaled these measurements to the ecosystem, and computed annual totals for respiration by component. We also compared estimated half-hourly ecosystem respiration at night (Fnc) with eddy covariance measurements. Mean foliage respiration normalized to 10 °C was 0.20 μmol m–2 (hemi-leaf surface area) s–1, and reached a maximum of 0.24 μmol m–2 HSA s–1 between days 162 and 208. Mean wood respiration normalized to 10 °C was 5.9 μmol m–3 sapwood s–1, with slightly higher rates in mid-summer, when growth occurs. There was no significant difference (P > 0.10) between wood respiration of young (45 years) and old trees (250 years). Soil surface respiration normalized to 10 °C ranged from 0.7 to 3.0 μmol m–2 (ground) s–1 from days 23 to 329, with the lowest rates in winter and highest rates in late spring. Annual CO2 flux from soil surface, foliage and wood was 683, 157, and 54 g C m–2 y–1, with soil fluxes responsible for 76% of ecosystem respiration. The ratio of net primary production to gross primary production was 0.45, consistent with values for conifer sites in Oregon and Australia, but higher than values reported for boreal coniferous forests. Below-ground carbon allocation (root turnover and respiration, estimated as Fs– litterfall carbon) consumed 61% of GPP; high ratios such as this are typical of sites with more water and nutrient constraints. The chamber estimates were moderately correlated with change in CO2 storage in the canopy (Fstor) on calm nights (friction velocity u* < 0.25 m s–1; R2 = 0.60); Fstor was not significantly different from summed chamber estimates. On windy nights (u* > 0.25 m s–1), the sum of turbulent flux measured above the canopy by eddy covariance and Fstor was only weakly correlated with summed chamber estimates (R2 = 0.14); the eddy covariance estimates were lower than chamber estimates by 50%.  相似文献

Populations of many species are isolated within narrow elevation bands of Neotropical mountain habitat, and how well dispersal maintains genetic connectivity is unknown. We asked whether genetic structure of an epiphytic orchid, Epidendrum firmum, corresponds to gaps between Costa Rican mountain ranges, and how these gaps influence pollen and seed flow. We predicted that significant genetic structure exists among mountain ranges due to different colonization histories and limited gene flow. Furthermore, we predicted that pollen movement contributes more to gene flow than seeds because seeds are released into strong winds perpendicular to the narrow northwest–southeast species distribution, while the likely pollinators are strong fliers. Individuals from 12 populations and three mountain ranges were genotyped with nuclear microsatellites (nDNA) and chloroplast sequences (cpDNA). Genetic diversity was high for both markers, while nDNA genetic structure was low (FSTn = 0.020) and cpDNA structure was moderate (FSTc = 0.443). Significant cpDNA barriers occurred within and among mountain ranges, but nDNA barriers were not significant after accounting for geographic distance. Consistent with these contrasting patterns of genetic structure, pollen contributes substantially more to gene flow among populations than seed (mp/ms = 46). Pollinators mediated extensive gene flow, eroding nDNA colonization footprints, while seed flow was comparatively limited, possibly due to directional prevailing winds across linearly distributed populations. Dispersal traits alone may not accurately inform predictions about gene flow or genetic structure, supporting the need for research into the potentially crucial role of pollinators and landscape context in gene flow among isolated populations.  相似文献

The genetic diversity in the wild and semi-domestic populations of Daba ecorace of Antheraea mylitta was studied to ascertain the distribution of variability within and among populations of semi-domestic bivoltine (DB), trivoltine (DT) and nature grown wild populations (DN) with inter-simple sequence repeat (ISSR) markers. A total of 138 markers were produced among 56 individuals of the three populations, of which 98% were polymorphic. For the individual populations, the percentage polymorphism was 58.69, 52.9 and 77.54 for DB, DT and DN, respectively. Average number of observed (1.791 ± 0.408) and effective alleles (1.389 ± 0.348) was also high in the wild populations in comparison to the bivoltine and trivoltine semi-domestic populations. Genetic diversity (Ht) in DB, DT and DN was 0.180 ± 0.033, 0.153 ± 0.032 and 0.235 ± 0.033, respectively and within-population genetic diversity (Hs) ranged from 0.166 to 0.259 with a mean of 0.189. Mean gene differentiation (GST) was found to be 0.25. Shanon’s diversity index was 0.278, 0.237 and 0.361 for DB, DT and DN and overall it was 0.391. Gene flow (Nm) among the populations was 1.509. The dendrogram produced by UPGMA with Dice’s genetic distance matrices resulted in the formation of three major clusters separating the three populations. Considerable intra- and inter-population variability is found in all three populations. The population structure analysis further suggests that the semi-domestic populations of Daba ecorace are at the threshold of differentiating themselves. The high genetic variability present within wild Daba population of A. mylitta is of much importance for conservation as well as utilization in systematic breeding program.  相似文献

Boltonia decurrens(Asteraceae), a federally listed, threatened floodplain species, requires regular flooding for suitable habitat and seed dispersal. Flood suppression and habitat destruction have resulted in fewer than 25 populations remaining throughout its 400 km range. Because individual populations are widely interspaced (>10 km) and subject to frequent extinction and colonization, seed dispersal along the river, not pollen flow, is likely the primary determinant of population genetic structure. We used neutral genetic markers (isozymes) assayed for fourteen populations to determine which demographic processes contribute to the genetic structure of B. decurrens. Significant genetic differentiation was detected among populations (F ST=0.098, P< 0.05) but not among regions (F RT=0.013, P> 0.05), suggesting that long-distance dispersal events occur and involve seed from a small number of populations. Correspondingly, we found no evidence of isolation by distance, and admixture analyses indicate that colonization events involve seed from 3 to 5 source populations. Individual populations exhibited high levels of fixation (mean F IS=0.192, P< 0.05), yet mean population outcrossing rates were high (t m=0.87–0.95) and spatial autocorrelation analyses revealed no fine-scale within population structure, indicating that inbreeding alone cannot explain the observed fixation. Rather, genetic bottlenecks, detected for 12 of 14 populations, and admixture at population founding may be important sources of fixation. These observations are consistent with a metapopulation model and confirm the importance of regular flooding events, capable of producing suitable habitat and dispersing seed long distances, to the long-term persistence of B. decurrens.  相似文献

Theobroma cacao L. expressed sequence tags (ESTs) were converted into useful genetic markers for fingerprinting individuals and genetic linkage mapping. Primers were designed to microsatellite‐containing ESTs. Twenty‐two T. cacao accessions, parents of various mapping populations segregating for disease resistance and crop yield characteristics, were tested. Twenty‐seven informative loci were discovered with 26 primer pairs. The number of detected alleles ranged from two to 11 and averaged 4.4 per locus. All 27 markers could be mapped into at least one of the existing F1 or F2 populations segregating for agronomically important traits.  相似文献

Knowledge of mating systems is required in order to understand the genetic composition and evolutionary potential of plant populations. Outcrossing in a population may co-vary with the ecological and historical factors influencing it. However, literature on the outcrossing rate is limited in terms of wild sorghum species coverage and eco-geographic reference. This study investigated the outcrossing rates in wild sorghum populations from different ecological conditions of Kenya. Twelve wild sorghum populations were collected in four sorghum growing regions. Twenty-four individuals per population were genotyped using six polymorphic simple sequence repeat (SSR) markers to compute their indirect equilibrium estimates of outcrossing rate as well as population structure. In addition, the 12 populations were planted in a field in a randomised block design with five replications. Their progeny (250 individuals per population) were genotyped with the six SSR markers to estimate multi-locus outcrossing rates. Equilibrium estimates of outcrossing rates ranged from 7.0 to 75.0%, while multi-locus outcrossing rates (t m) ranged from 8.9 to 70.0% with a mean of 49.7%, indicating that wild sorghum exhibits a mixed mating system. The wide range of estimated outcrossing rates in wild sorghum populations indicate that environmental conditions may exist under which fitness is favoured by outcrossing and others under which selfing is more advantageous. The genetic structure of the populations studied is concordant with that expected for a species displaying mixed mating system.  相似文献

The endangered annual plant Limnanthes floccosa ssp. californica Arroyo is restricted to vernal pools in Butte County, California. To identify populations with unique genetic resources, guide reintroduction efforts, and design seed collection scenarios for long-term ex situ seed storage we determined extant genetic diversity and structure by surveying 457 individuals from 21 distinct populations using nine polymorphic microsatellite markers. We found low within population genetic diversity: low allelic diversity (1.9 [0.06 SE] alleles/locus); low heterozygosity (H obs = 0.10 ± 0.018, H exp = 0.19 ± 0.015), and a high fixation index (0.556 ± 0.044). The number of polymorphic loci ranged between 11 and 89%. Bayesian ordination determined 20 distinct populations and we found high genetic structure among these (F st = 0.65, P < 0.0001). We identified notable gene flow barriers across populations, confirming regional structuring between three previously defined population density centers and two outlying populations (F st = 0.21, P < 0.0001). Population size estimates ranged between ~50 and >5,000 extant plants per site. Our study confirms previous isozyme-based results and suggests that the loss of any population would represent a significant loss in the species’ genetic diversity. Recovery requires active restoration of existing populations and permanent habitat protection. We recommend close comparison of microhabitats of declining populations with genetically similar populations, to determine the potential for human assisted gene flow via seed movement to recover declining populations.  相似文献

The levels and partitioning of genetic diversity and inbreeding depression were investigated in Tupistra pingbianensis, a narrow endemic of South-east Yunnan, China, characterized by a naturally fragmented distribution due to extreme specialization on a rare habitat type. Here genetic diversity and patterns of genetic variation within and among 11 populations were analyzed using AFLP markers with 97 individuals across its whole geographical range. High levels of genetic variation were revealed both at the species level (P99 = 96.012%; Ht = 0.302) and at the population level (P99 = 51.41%; Hs = 0.224). Strong genetic differentiation among populations was also detected (FST = 0.2961; ⍬= 0.281), which corresponded to results reported for typical animal-pollinated, mixed selfing and outcrossing plant species. This result was consistent with mating patterns detected by our pollination experiments. The indirect estimate of gene flow based on ⍬ was low (Nm = 0.64). Special habitat and its life history traits may play an important role in shaping the genetic diversity and the genetic structure of this species. A pollination experiment also fail to detect significant inbreeding depression upon F1 fruit set, seed weight and germinate rate fitness-traits. Since naturally rare species T. pingbianensis is not seriously genetically impoverished and likely to have adapted to tolerating a high level of inbreeding early in its history, we propose this species need only periodic monitoring to ensure their continued persistence but not intervention to remain viable.  相似文献

We present a study of the genetic variation, genetic structure, and outcrossing rate of two wild populations of Phaseolus coccineus ssp. formosus (both from Central Mexico), two cultivated populations of P. coccineus ssp. coccineus (one from Central Mexico, the other from Chiapas), and one cultivated population of P. vulgaris (from Central Mexico), using seven electrophorectic loci. All the populations of P. coccineus showed high and similar levels of genetic variation, without differences among wild and cultivated populations (H ranged between 0.187 to 0.271). In contrast, P. vulgaris has low levels of genetic variation (H = 0.041). The three P. coccineus populations from Central Mexico were genetically similar, while the cultivated P. coccineus from Chiapas and the cultivated P. vulgaris from Central Mexico were very different. In all the loci and all the populations for both species, the fixation indices (F) were positive and differed significantly from random mating expectation in 59% of the cases for P. coccineus. The highest F was the estimated for P. vulgaris. For P. coccineus, the multilocus outcrossing rate estimates ranged from 0.592 to 0.698, and these did not differ significantly among populations. For P. coccineus, we also estimated the outcrossing rate for each of 60 different families, showing great variation, with a majority of families with a t between 0.8 and 1, but a substantial fraction of the families with a t as low as 0.12–0.20. The domestication process has neither eroded the levels of genetic variation of the cultivated populations of P. coccineus nor changed its mating system.  相似文献

Caridina gracilipes, an atyid shrimp of Taiwan, inhabits the fresh and brackish waters. Its life history can be classified into amphidromous and landlocked types. Recent human’s transportation has caused concerns of genetic contamination between these long evolved lineages and ecological instabilities. Molecular markers with high sensitivities are required for detecting the possible introgression due to human disturbance. In the study, we described the development of 15 polymorphic microsatellite loci in C. gracilipes for genetic and ecological studies. These new markers were tested in 20 individuals of three populations. The number of alleles ranged from 6 to 17. Expected (H E) and observed (H O) heterozygosities ranged from 0.653 to 0.954, averaged at 0.865, and from 0.000 to 0.4, averaged at 0.146, respectively. All loci are significantly deviated from Hardy–Weinberg expectations due to the heterozygote deficiency. Significant genetic differentiation among populations was detected based on F ST values, which were estimated from 0.82 to 0.88. The genetic variability and population genetic structure of six populations of Praecereus euchlorus and Pilosocereus machrisii were investigated. The genetic variability in single populations of Pilosocereus vilaboensis, Pilosocereus aureispinus, and Facheiroa squamosa was also examined. All of these cacti species have a patchy geographic distribution in which they are restricted to small areas of xeric habitats in eastern Brazil. An analysis of genetic structure was used to gain insights into the historical mechanisms responsible for the patchy distribution of P. euchlorus and P. machrisii. High genetic variability was found at the populational level in all species (P=58.9–92.8%, Ap=2.34–3.33, He=0.266–0.401), and did not support our expectations of low variability based on the small population size. Substantial inbreeding was detected within populations (FIS=0.370–0.623). In agreement with their insular distribution patterns, P. euchlorus and P. machrisii had a high genetic differentiation (FST=0.484 and FST=0.281, respectively), with no evidence of isolation by distance. Accordingly, estimates of gene flow (Nm) calculated from FST and private alleles were below the level of Nm=1 in P. machrisii and P. euchlorus. These results favored historical fragmentation as the mechanism responsible for the patchy distribution of these two species. The genetic distance between P. machrisii and P. vilaboensis was not compatible with their taxonomic distinction, indicating a possible local speciation event in this genus, or the occurrence of introgression events.  相似文献

Eucalyptus breeding is typically conducted by selection in open-pollinated progenies. As mating is controlled only on the female side of the cross, knowledge of outcrossing versus selfing rates is essential for maintaining adequate levels of genetic variability for continuous gains. Outcrossing rate in an open-pollinated breeding population of Eucalyptus urophylla was estimated by two PCR-based dominant marker technologies, RAPD and AFLP, using 11 open-pollinated progeny arrays of 24 individuals. Estimated outcrossing rates indicate predominant outcrossing and suggest maintenance of adequate genetic variability within families. The multilcous outcrossing rate (tm) estimated from RAPD markers (0.93±0.027), although in the same range, was higher (α>0.01) than the estimate based on AFLP (0.89±0.033). Both estimates were of similar magnitude to those estimated for natural populations using isozymes. The estimated Wright’s fixation index was lower than expected based on tm possibly resulting from selection against selfed seedlings when sampling plants for the study. An empirical analysis suggests that 18 is the minimum number of dominant marker loci necessary to achieve robust estimates of tm. This study demonstrates the usefulness of dominant markers, both RAPD and AFLP, for estimating the outcrossing rate in breeding and natural populations of forest trees. We anticipate an increasing use of such PCR-based technologies in mating-system studies, in view of their high throughput and universality of the reagents, particularly for species where isozyme systems have not yet been optimized. Maxipiñon (Pinus maximartinezii Rzedowski), which is confined to a single population of approximately 2000 to 2500 mature trees, covers about 400 ha in southern Zacatecas, Mexico. Genetic diversity measured by expected heterozygosity was 0.122, which is moderate for pines. However, percentage polymorphic loci was low, 30.3%. The fixation index (F) of 0.081 indicated only slight heterozygote deficiency. Mating system analysis indicated a significant but low level of selling; the multilocus outcrossing rate, tm, was 0.816. The mean of single locus estimates, ts, was smaller (0.761), perhaps suggesting mating among relatives, although the difference between tm and ts was not statistically significant. The most striking features of maxipiñon's genetic structure were that no polymorphic locus had more than two alleles and most alleles at polymorphic loci were at intermediate frequencies. This is in contrast to other pines, which often have three to five or more alleles at some loci and in which the distribution of allele frequencies is U-shaped, most alleles being present at frequencies less than 10% or greater than 90%. A population with only two alleles per locus and at intermediate frequencies could occur if the population had been reduced to an extreme bottleneck and then expanded rapidly before random drift modified allele frequencies. A novel origin from a hybridization event would also explain the results. Significant gametic disequilibrium was detected at several pairs of loci in both maternal and paternal gametes. The presence of disequilibrium is in agreement with an origin from an extreme bottleneck, perhaps even a single seed. Furthermore, it demands that the event be relatively recent. The number of generations, as calculated from the observed mean disequilibrium, suggested that maxipiñon derived from an extreme bottleneck four to five generations ago, which is less than 1000 years in this species.  相似文献

Summary A multilocus procedure was used to estimate outcrossing rates in ten roadside populations of Trifolium hirtum in California. Three groups of populations were studied: cultivars, hermaphroditic, and gynodioecious (sexually dimorphic) populations. The multilocus outcrossing rate (tm) varied from 0.05 to 0.43 among populations. Population level tm estimates were significantly correlated with the observed heterozygosity in gynodioecious populations but not in hermaphroditic populations. The outcrossing rate of hermaphrodites and females was estimated in three gynodioecious populations; the estimates of tm varied from 0.09 to 0.23 for hermaphrodites and from 0.73 to 0.80 for females. The distribution of outcrossing rates in gynodioecious populations is bimodal. Our results indicate that for the levels of selfing observed among hermaphrodites, inbreeding depression is likely to be a major factor in the maintenance of females in gynodioecious populations.  相似文献

Microsatellites were used to evaluate the mating system of the remaining trees in a logged population of Swietenia macrophylla, a highly valuable and threatened hardwood species, in the Brazilian Amazon. A total of 25 open pollinated progeny arrays of 16 individuals, with their mother trees, were genotyped using eight highly polymorphic microsatellite loci. Genotypic data analysis from the progeny arrays showed that 373 out of the 400 seedlings (93.25%) were unambiguously the result of outcrossed matings and that the remaining 6.75% had genotypes consistent with self-fertilisation. Apomixis could be ruled out, since none of the 400 seedlings analysed had a multi-locus genotype identical to its mother tree. The high estimate of the multi-locus outcrossing rate (t m = 0.938 ± 0.009) using the mixed mating model also indicated that the population in this remnant stand of S. macrophylla was predominantly allogamous. The relatively large difference between the multi-locus and single-locus outcrossing estimates (t m −t s = 0.117 ± 0.011) provides evidence that, in spite of the high outcrossing rate, a considerable degree of biparental inbreeding has contributed to the genetic structure of this population. Levels of outcrossing were not evenly distributed among maternal trees (t m ranging from 0.39 to 1.00), suggesting the occurrence of a variable degree of self-incompatibility and/or dichogamy among individual trees of this monoecious species. Due to its generalist pollination system and some level of tolerance for selfing, S. macrophylla seems to be resilient to environmental disturbances such as those caused by logging, since it sets fruits with predominantly outcrossed seeds even at low stand densities. Therefore, the remaining individuals in logged areas or in relict fragments may be very important for long-term population recovery and genetic conservation programmes.  相似文献

The Caesalpinia hintonii complex is formed by five endemic species (C. hintonii, C. laxa, C. macvaughii, C. melanadenia and C. epifanioi) occurring in central Mexico. This species complex is under incipient genetic divergence as by-product of local adaptations in reproductive and morphological traits to different habitats. We estimate the genetic variation and structure of populations of this species complex to assess the extent of genetic differentiation among populations and related species along its geographic distribution. Estimations of genetic diversity and structure were done based on ten enzymes and 18 loci. Mean number of alleles per locus ranged from 1.5 to 1.9. Polymorphic loci ranged from 42.1 to 68.4. Observed (Ho: range 0.191–0.275) and expected (He: range 0.205–0.317) heterozygosities in this complex were higher compared with other endemic and legume species. Neis genetic diversity estimates showed that most genetic variation was found within (HS = 0.325) rather than among populations (DST = 0.085). Populations of the species C. hintonii showed a considerable genetic differentiation (FST = 0.207). The results of genetic diversity and structure within and among populations are in accord with the great morphological differentiation described for this species complex.  相似文献

European grayling populations in Bavaria have shown steady declines during the last 10–20years. In order to provide guidelines for conservation strategies and future management programs, we investigated the genetic structure of 15 grayling populations originating from three major Central European drainages (the Danube, the Elbe and the Rhine/Main) using 20 microsatellite loci. Genetic divergence between the three drainage systems was substantial as illustrated by highly significant heterogeneity of genotype frequencies, high number of drainage-specific private alleles, high between-drainage F ST values, high assignment success of individuals to their drainage of origin and the high bootstrap support for the genetic distance based drainage-specific population clusters. In agreement with earlier studies, microsatellites revealed relatively low levels of intrapopulational genetic diversity in comparison to the overall level of variation across populations. Maximum likelihood methods using the coalescent approach revealed that the proportion of common ancestors was generally high in native populations and that the estimates of N e were correlated with the genetic diversity parameters in all drainages. The number of effective immigrants per generation (N e m) was less than one for all pairwise comparisons of populations within the drainages, indicating restricted interpopulational gene flow. Based on these findings we recommend a drainage and sub-drainage specific conservation of grayling populations in order to preserve their overall genetic diversity and integrity. For large-scale stocking actions to supplement declining or to restore extinct populations, creation of separate broodstocks for major conservation units (ESUs and MUs) is warranted.
