Survey of the year 2004 commercial optical biosensor literature   总被引:4,自引:0,他引:4  
The year 2004 represents a milestone for the biosensor research community: in this year, over 1000 articles were published describing experiments performed using commercially available systems. The 1038 papers we found represent an approximately 10% increase over the past year and demonstrate that the implementation of biosensors continues to expand at a healthy pace. We evaluated the data presented in each paper and compiled a 'top 10' list. These 10 articles, which we recommend every biosensor user reads, describe well-performed kinetic, equilibrium and qualitative/screening studies, provide comparisons between binding parameters obtained from different biosensor users, as well as from biosensor- and solution-based interaction analyses, and summarize the cutting-edge applications of the technology. We also re-iterate some of the experimental pitfalls that lead to sub-optimal data and over-interpreted results. We are hopeful that the biosensor community, by applying the hints we outline, will obtain data on a par with that presented in the 10 spotlighted articles. This will ensure that the scientific community at large can be confident in the data we report from optical biosensors.  相似文献

We took a different approach to reviewing the commercial biosensor literature this year by inviting 22 biosensor users to serve as a review committee. They set the criteria for what to expect in a publication and ultimately decided to use a pass/fail system for selecting which papers to include in this year's reference list. Of the 1514 publications in 2009 that reported using commercially available optical biosensor technology, only 20% passed their cutoff. The most common criticism the reviewers had with the literature was that "the biosensor experiments could have been done better." They selected 10 papers to highlight good experimental technique, data presentation, and unique applications of the technology. This communal review process was educational for everyone involved and one we will not soon forget.  相似文献

We identified 1219 articles published in 2006 that described work performed using commercial optical biosensor platforms. It is interesting to witness how the biosensor market is maturing with an increased number of instrument manufacturers offering a wider variety of platforms. However, it is clear from a review of the results presented that the advances in technology are outpacing the skill level of the average biosensor user. While we can track a gradual improvement in the quality of the published work, we clearly have a long way to go before we capitalize on the full potential of biosensor technology. To illustrate what is right with the biosensor literature, we highlight the work of 10 groups who have their eye on the ball. To help out the rest of us who have the lights on but nobody home, we use the literature to address common myths about biosensor technology.  相似文献

Optical biosensor technology continues to be the method of choice for label‐free, real‐time interaction analysis. But when it comes to improving the quality of the biosensor literature, education should be fundamental. Of the 1413 articles published in 2008, less than 30% would pass the requirements for high‐school chemistry. To teach by example, we spotlight 10 papers that illustrate how to implement the technology properly. Then we grade every paper published in 2008 on a scale from A to F and outline what features make a biosensor article fabulous, middling or abysmal. To help improve the quality of published data, we focus on a few experimental, analysis and presentation mistakes that are alarmingly common. We identified 1113 articles (103 reviews, 1010 primary research articles) published in 2005 that describe experiments performed using commercially available optical biosensors. While this number of publications is impressive, we find that the quality of the biosensor work in these articles is often pretty poor. It is a little disappointing that there appears to be only a small set of researchers who know how to properly perform, analyze, and present biosensor data. To help focus the field, we spotlight work published by 10 research groups that exemplify the quality of data one should expect to see from a biosensor experiment. Also, in an effort to raise awareness of the common problems in the biosensor field, we provide side-by-side examples of good and bad data sets from the 2005 literature.  相似文献

We have compiled 819 articles published in the year 2002 that involved commercial optical biosensor technology. The literature demonstrates that the technology's application continues to increase as biosensors are contributing to diverse scientific fields and are used to examine interactions ranging in size from small molecules to whole cells. Also, the variety of available commercial biosensor platforms is increasing and the expertise of users is improving. In this review, we use the literature to focus on the basic types of biosensor experiments, including kinetics, equilibrium analysis, solution competition, active concentration determination and screening. In addition, using examples of particularly well-performed analyses, we illustrate the high information content available in the primary response data and emphasize the impact of including figures in publications to support the results of biosensor analyses.  相似文献

Cell Cycle acknowledges the most outstanding cell cycle research of the past year and presents the list of the top ten papers, published in all journals between 2002 and mid 2003. Papers were put forward by and final selections were made by members of the Cell Cycle Editorial Board. More than 40 papers were recommended. Criteria for selection were: the paper will affect future basic research or research in therapy or describes a new technology. In 2003, the date for publication of the annual report for 2002 was postponed and it is therefore printed in this issue of the Journal . 2002 was, however, another very successful year during which we received 281 submissions and published 167 papers in volumes 256–258. The Journal continues to receive high-quality, original research papers and reviews from a wide range of taxa and a variety of disciplines. In this work, we have demonstrated that the exquisite optical properties based on localized surface plasmon resonance (LSPR) of Au2S/AuAgS-coated gold nanorods (Au2S/AuAgS-coated GNRs) can be utilized to develop a simple and sensitive biosensor, and goat anti-human IgG can be detected by the human IgG probe as low as 0.2 nM. Moreover, we introduce an integrated LSPR biosensor constructed by integrating Au2S/AuAgS-coated GNRs immobilized on glass slide and isolated Au2S/AuAgS-coated GNRs in the form of liquid. The detection of target binding was performed via direct spectral changes induced by changes of refractive index in the vicinity of individual particles. The integrated LSPR optical biosensor is label-free, cost-effective, and easy to fabricate and requires only a visible/near-infrared spectrometer for detection purposes. Additionally, the investigation on the mutual influence of the two types of nanorods in the integrated LSPR biosensor was performed. The results of separate experiments indicate that the nanorods in the form of isolate or in integrated exhibit a similar behavior.  相似文献

We have compiled a comprehensive list of the articles published in the year 2000 that describe work employing commercial optical biosensors. Selected reviews of interest for the general biosensor user are highlighted. Emerging applications in areas of drug discovery, clinical support, food and environment monitoring, and cell membrane biology are emphasized. In addition, the experimental design and data processing steps necessary to achieve high-quality biosensor data are described and examples of well-performed kinetic analysis are provided.  相似文献

For efficient generation of high-affinity protein-based binding molecules, fast and reliable downstream characterization platforms are needed. In this work, we have explored the use of staphylococcal cell surface display together with flow cytometry for affinity characterization of candidate affibody molecules directly on the cell surface. A model system comprising three closely related affibody molecules with different affinities for immunoglobulin G and an albumin binding domain with affinity for human serum albumin was used to investigate advantages and differences compared to biosensor technology in a side-by-side manner. Equilibrium dissociation constant (K(D)) determinations as well as dissociation rate analysis were performed using both methods, and the results show that the on-cell determinations give both K(D) and dissociation rate values in a very fast and reproducible manner and that the relative affinities are very similar to the biosensor results. Interestingly, the results also show that there are differences between the absolute affinities determined with the two different technologies, and possible explanations for this are discussed. This work demonstrates the advantages of cell surface display for directed evolution of affinity proteins in terms of fast postselectional, on-cell characterization of candidate clones without the need for subcloning and subsequent protein expression and purification but also demonstrates that it is important to be aware that absolute affinities determined using different methods often vary substantially and that such comparisons therefore could be difficult.  相似文献

The use of biosensor technology is described to address in real-time the production and subsequent purification of a bioactive recombinant protein product. The product, D1.3 Fv antibody fragment, was expressed in Escherichia coli and purified via two process routes, one for extracellular and one for intracellular product material. The cells were harvested by centrifugation in a solid bowl CARR Powerfuge and stored at –70°C. Clarification of the supernatant was performed by depth filtration, followed by affinity chromatography for final purification of the extracellular product. To purify the intracellular product the harvested cells were resuspended and homogenised. Removal of debris in the CARR Powerfuge was followed by depth filtration and affinity chromatography. In this work we have shown the rapid determination of bioactive product levels, and the impact this has on improved accountability and confidence is demonstrated in process mass balances on the product using the data acquired during process operation.  相似文献

A surface plasmon resonance (SPR) biosensor based on wavelength modulation was used for real-time detection of the interaction of three monoclonal antibodies and antigens of bovine cardiac troponin I (cTnI). In order to recognize antigenic epitopes of bovine cTnI, two experimental modes were applied. In the first experimental mode, three monoclonal antibodies were divided into three groups and three experiments were performed on biosensor surfaces prepared with protein A. In the second experimental mode, antigen was immobilized on the biosensor surface prepared by the amine-coupling method and three monoclonal antibodies were detected in turn. The results obtained by the two modes are consistent. In addition, the affinities of the monoclonal antibodies for the antigen were also determined by the association rate and the disassociation rate in real-time. These results validate the biosensor technology and illustrate how biosensors based on wavelength modulation can be used to study the interaction of monoclonal antibodies and antigens in real time.  相似文献

Journal of Plant Ecology (JPE) decided to establish awards of JPE Best Papers, in order to thank and encourage authors to submit their valuable research to JPE. This award will be given annually to the first author(s) of 2–5 papers selected by the editors, according to the citation data as well as the impact in the field of plant ecology. We will place extra emphasis on authors who had completed graduate studies not more than 5 years before submission of the paper, to encourage more early-career scientists to submit their work to JPE. Background

In contrast to Newton''s well-known aphorism that he had been able “to see further only by standing on the shoulders of giants,” one attributes to the Spanish philosopher Ortega y Gasset the hypothesis saying that top-level research cannot be successful without a mass of medium researchers on which the top rests comparable to an iceberg.

Methodology/Principal Findings

The Ortega hypothesis predicts that highly-cited papers and medium-cited (or lowly-cited) papers would equally refer to papers with a medium impact. The Newton hypothesis would be supported if the top-level research more frequently cites previously highly-cited work than that medium-level research cites highly-cited work. Our analysis is based on (i) all articles and proceedings papers which were published in 2003 in the life sciences, health sciences, physical sciences, and social sciences, and (ii) all articles and proceeding papers which were cited within these publications. The results show that highly-cited work in all scientific fields more frequently cites previously highly-cited papers than that medium-cited work cites highly-cited work.

Conclusions/Significance

We demonstrate that papers contributing to the scientific progress in a field lean to a larger extent on previously important contributions than papers contributing little. These findings support the Newton hypothesis and call into question the Ortega hypothesis (given our usage of citation counts as a proxy for impact).  相似文献

Whole-cell biosensors are mostly non-specific with respect to their detection capabilities for toxicants, and therefore offering an interesting perspective in environmental monitoring. However, to fully employ this feature, a robust classification method needs to be implemented into these sensor systems to allow further identification of detected substances. Substance-specific information can be extracted from signals derived from biosensors harbouring one or multiple biological components. Here, a major task is the identification of substance-specific information among considerable amounts of biosensor data. For this purpose, several approaches make use of statistical methods or machine learning algorithms. Genetic Programming (GP), a heuristic machine learning technique offers several advantages compared to other machine learning approaches and consequently may be a promising tool for biosensor data classification. In the present study, we have evaluated the use of GP for the classification of herbicides and herbicide classes (chemical classes) by analysis of substance-specific patterns derived from a whole-cell multi-species biosensor. We re-analysed data from a previously described array-based biosensor system employing diverse microalgae (Podola and Melkonian, 2005), aiming on the identification of five individual herbicides as well as two herbicide classes. GP analyses were performed using the commercially available GP software 'Discipulus', resulting in classifiers (computer programs) for the binary classification of each individual herbicide or herbicide class. GP-generated classifiers both for individual herbicides and herbicide classes were able to perform a statistically significant identification of herbicides or herbicide classes, respectively. The majority of classifiers were able to perform correct classifications (sensitivity) of about 80-95% of test data sets, whereas the false positive rate (specificity) was lower than 20% for most classifiers. Results suggest that a higher number of data sets may lead to a better classification performance. In the present paper, GP-based classification was combined with a biosensor for the first time. Our results demonstrate GP was able to identify substance-specific information within complex biosensor response patterns and furthermore use this information for successful toxicant classification in unknown samples. This suggests further research to assess perspectives and limitations of this approach in the field of biosensors.  相似文献

There is evidence of a significant increase in air temperature in the northern hemisphere over recent decades, with consequent changes for anemophilous pollen. In this work we present the effects of climatic change on Platanus spp. pollination in different areas of Italy and Spain, characterized by different climates. In particular, the historical series of pollen monitoring and meteorological data of two Italian stations, Perugia (1982 – 2003) and Torino (1985 – 2003), and two Spanish stations, Santiago de Compostela (1992 – 2003) and Vigo (1994 – 2003), were analysed. The changes recorded in all stations included the timing and behaviour of pollen release. However, no or minimal influence on the total pollen emission was found. Research has linked the changes in phenological events to an increase in temperature, moreover in this study temperature changes are believed to be mainly responsible for the variations recorded in the pollen season of Platanus. A previous start of pollination (?0.66; ?1.21; days/year) is reported in both Italian stations where the temperatures have significantly increased and a delay of 0.2 – 0.8 days/year in Spanish stations where a different trend of temperature is recorded. Other important data is given regarding the type of discharge of pollen grains during the pollen season. Pollination curves are examined by two statistical shape parameters (kurtosis and skewness) which show that pollen release is more gradual with higher temperatures or faster under colder conditions. A regression analysis links the atmospheric pollen presence to mean air temperature.  相似文献

Receptor activation by IL5 and GM-CSF is a sequential process that depends on their interaction with a cytokine-specific subunit alpha and recruitment of a common signaling subunit beta (betac). In order to elucidate the assembly dynamics of these receptor subunits, we performed kinetic interaction analysis of the cytokine-receptor complex formation by a surface plasmon resonance biosensor. Using the extracellular domains of receptor fused with C-terminal V5-tag, we developed an assay method to co-anchor alpha and betac subunits on the biosensor surface. We demonstrated that dissociation of the cytokine-receptor complexes was slower when both subunits were co-anchored on the biosensor surface than when alpha subunit alone was anchored. The slow-dissociation effect of betac had a similar impact on GM-CSF receptor stabilization to that of IL5. The effects were abolished by alanine replacement of either Tyr18 or Tyr344 residue in betac, which together constitute key parts of a cytokine binding epitope. The data argue that betac plays an important role in preventing the ligand-receptor complexes from rapidly dissociating. This slow-dissociation effect of betac explains how, when multiple betac cytokine receptor alpha subunits are present on the same cell surface, selective betac usage can be controlled by sequestration in stabilized cytokine-alpha-betac complexes.  相似文献
