The CO2-, H2O- and 16O2/18O2 isotopic-gas exchange and the fluorescencequenching by attached leaves of the wild-type and of the phytochrome-deficienttomato aurea mutant was compared in relation to water stressand the photon fluence rate. The chlorophyll content of aurealeaves was reduced and the ultra-structure of the chloroplastswas altered. Nevertheless, the maximum rate of net CO2 uptakein air by the yellow-green leaves of the aurea mutant was similarto that by the dark-green wild-type leaves. However, less O2was produced by the leaves of the aurea mutant than by leavesof the wild-type. This result indicates a reduced rate of photosyntheticelectron flux in aurea mutant leaves. No difference in bothphotochemical and non-photochemical fluorescence quenching wasfound between wild-type and aurea mutant leaves. Water stresswas correlated with a reversible decrease in the rates of bothnet CO2 uptake and transpiration by wild-type and aurea mutantleaves. The rate of gross 16O2 evolution by both wild-type andaurea mutant leaves was fairly unaffected by water stress. Thisresult shows that in both wild-type and aurea leaves, the photochemicalprocesses are highly resistant to water stress. The rate ofgross 18O2 uptake by wild-type leaves increased during waterstress when the photon fluence rate was high. Under the sameconditions, the rate of gross 18O2 uptake by aurea mutant leavesremained unchanged. The physiological significane of this differencewith respect to the (presumed) importance of oxygen reductionin photoprotection is discussed. This research examined the hypothesis that as cacti evolve tothe leafless condition, the stem epidermis and cortex becomemore leaflike and more compatible with a photosynthetic role.All cacti in the relict genus Pereskia have non-succulent stemsand broad, thin leaves. All members of the derived subfamilyCactoideae are ‘leafless’, having an expanded cortexthat is the plant's only photosynthetic tissue. In Pereskia,leaves have a high stomatal density (mean: 50.7 stomata mm–2in the lower epidermis, 38.1 mm–2 in the upper epidermis),but stems have low stomatal densities (mean: 11.3 mm 2, threeof the species have none). Stems of Cactoideae have a high stomataldensity (mean: 31.1 mm–2, all species have stomata). Theouter cortex cells of stems of Cactoideae occur in columns,forming a palisade cortex similar to a leaf palisade parenchyma.In this palisade cortex, the fraction of tissue volume availablefor gas diffusion has a mean volume of 12.9%, which is identicalto that of Pereskia leaf palisade parenchyma. Pereskia stemcortex is much less aerenchymatous (mean: 5.3% of cortex volume).Cactoideae palisade cortex has a high internal surface density(0.0207 cm2 cm–2 which is higher than in Pereskia stemcortex (0.0150 cm2 cm–3) but not as high as Pereskia leafpalisade parenchyma (0.0396 cm2 cm–3). Pereskia stem cortexhas no cortical bundles, but Cactoideae cortexes have extensivenetworks of collateral vascular bundles that resemble leaf veins. The pattern for primary products of CO2-fixation and the chloroplaststructure of Amaranthus retrqflexus L., a species which incorporatescarbon dioxide into C4 dicarboxylic acids as the primary productof photosynthesis, were compared in various chlorophyll containingtissues,i.e., foliage leaves, stems, cotyledons and pale-greencallus induced from stem pith. Despite some morphological differencesin these assimilatory tissues, malate and aspartate were identifiedas the major compounds labelled during a 10 sec fixation of14CO2 in all tissues. Whereas, aspartate was the major componentin C4-dicarboxylic acids formed in foliage leaves, malate predominatedas the primary product in stems, cotyledons and the pale-greencallus. The contribution of individual vascular bundles of the stemto the flow of assimilates from a selected source leaf to thesink regions was investigated inUrtica dioica L., a plant witha decussate leaf arrangement. Two homologous sets of eight vascularstrands were recognized, arranged in mirror symmetry in thestem internodes. In each set, three of the bundles were identifiedas traces of one leaf merging into the vascular system of thestem one node below the origin of the leaf. The main bundleof a stem-half bifurcates at each end of the internode intotwo subdominant bundles, which combine in the next but one nodeto form the dominant bundle again. Each set of vascular strandsalso contains two minor bundles which pass more or less withoutinterruption through the whole stem. The uppermost mature source leaf (leaf number 5 as counted fromthe tip) was exposed to14CO2in a closed gas circuit. The concentrationof the carbon-labelled CO2was maintained at the ambient CO2levelto maintain the natural source strength of the leaf. By theend of the usual nocturnal dark phase, carbon from the sourceleaf had been imported predominantly by sink leaves of the sameorthostichy. Lesser, but significant amounts of radiocarbonwere also incorporated into the sink leaves of the adjacenttwo orthostichies via the marginal leaf traces. In spite ofthe junction of the vascular strands in the nodes and an interfascicularconnection of the stem bundles, randomization of the photosynthatesfrom individual leaves was minimal in the vascular system ofthe stem in the upward direction, and also low in the flux tothe roots. Substantial amounts of radioactivity were also foundin the lately-formed xylem elements of the vascular strandsand their interfascicular connections, indicating active secondarygrowth. Three-month-old Carrizo citrange (hybrid of Citrus sinensisL. OsbeckxPoncirus trifoliata Blanco) seedlings were grown incontrolled environment chambers in pots of fine sand. Plantswere irrigated with either non-saline or saline solutions overa 3-week period. After these treatments, plants were transferredto vessels containing a 5 m M15NO3K (96% atom excess15N) solution,and transpiration as well as concentration of15N and Cl-in roots,stem and leaves were measured after 24 h. Transpiration and15NO3-uptakerates were inhibited after exposure to NaCl and the concentrationof salt pre-treatment determined the intensity of this inhibitoryeffect. To determine the effect of transpiration on NO3-absorption,net15NO3-uptake rate was measured in salt stressed and non-stressedplants exposed to different light intensities or relative humiditiesand also in detached roots. Reduction in NO3-uptake was moreclosely related to Cl-antagonism from salt stress than to reducedtranspiration rate. Leaves of three C4 plants, Setaria italica, Pennisetum typhoides,and Amaranthus paniculatus possessed five- to ten-fold higheractivities of a (Na+-K+)-dependent ATPase than those of twoC3 plants, Oryza sativa and Rumex vesicarius. Na+-K+ ATPasefrom leaves of Amarathus exhibited an optimal pH of 7?5 andan optimal temperature of 35 ?C. It required 40 mM K+ and 80mM Na+ for maximal activity. Ouabain partially inhibited (Na+-K+)-dependentATPase activity in leaves of C4 plants. Ouabain also blockedthe movement of label from initially formed C4 acids into endproducts in leaves of only C4 plants, Setaria and Amaranthusbut not in a C3 plant, Rumex. We propose that Na+-K+ ATPasemay mediate transfer of energy during active transport of C4acids from mesophyll into the bundle sheath.  相似文献

Populus plants were grown in a medium lacking calcium and exposedto 14CO2. In contrast to plants in the complete nutrient medium,the percentage amount of 14C-assimilates increased in the leavesof calcium-deficient plants and decreased in the stem and theroots. When plants were grown without potassium or magnesiumno differences in the amount of 14C-label occurred in comparisonwith plants in the complete nutrient medium. Translocation wasrecorded by microautoradiography. It was observed that considerableamounts of labelled photoassimilates were unloaded from thephloem in the middle part of the stem in plants of the completenutrient medium. In contrast, during calcium starvation 14C-labelwas restricted to the phloem of the stem. In addition, the concentrationsof magnesium and phosphorus showed a remarkable increase instem sieve tubes of calcium-deficient plants. When sieve tubesof source leaves from Populus, barley and maize were comparedwith those of sink leaves, the latter showed higher calciumconcentrations. The results suggest that calcium is a necessaryfactor in the regulation of phloem translocation. The activities of Hill reaction and photosynthetic 14CO2 fixationin bundle sheath strands enzymatically isolated from millet(Panicum miliaceum) were 3–15 times as high as those observedin corn (Zea mays). In both preparations, 3-phosphoglyceratewas the initial 14CO2 fixation product and the radioactivitywas incorporated into sucrose and insoluble compounds (glucose-polymers)during the later period. After 20 sec of photosynthetic 14CO2fixation, the percent of 14C incorporated into sugar phosphatesin millet was about 3 times as high as that in corn, while incorn, the percent of 14C in 3-phosphoglycerate was higher thanthat observed in millet throughout the experimental period.When 14C-phosphoglycerate was added to the isolated bundle sheathstrands, the rates of transfer of the radioactivity to dihydroxyacetonephosphate and sugar diphosphates in millet were significantlyhigher than those in corn. These results indicate that in thebundle sheath strands isolated from corn in which photosystemII activity is deficient, the reductive pentose cycle is impairedat the reduction step of 3-phosphoglycerate to glyceraldehydephosphate due to the limited supply of NADPH through the photoelectrontransport system. Differences in premature leaf abscission and in visible steminjury in genetic lines of poplar followed continuous fumigationswith air pollutant levels of SO2 (90–100 nl l–1)and O3 (70–80 nl l–1) either separately or together.The clones used were: Populus deltoides var. missiouriensisMarsh., P. nigra cv. ‘italicd’ L., and the hybridsP. nigra cv. ‘italica’ xP. deltoides (He-X/3) andP. nigra cv.‘italica’ x P. nigra cv. ‘Serres’(He-K/7). While most leaf abscission occurred within 20 d fromthe start of fumigation, stem lesions (intumescences), appearedonly after 72 d. Their anatomical characteristics include theformation of lysigenous aerenchyma in the lower parts of theintumescence, the sloughing of superficial cells from the injuredarea, and the development of crystalline formations on the surfaceof the lesions. P. deltoides exhibited the least morphologicalresponse to the gases. Ethylene released from fumigated leaves was determined at thesame gas concentration of SO2 (100 nl l–1), O3 (75 nll–1) and their mixture. Leaves of P. deltoides consistentlyshowed the lowest ethylene production after the gas treatments.P. ‘italica’ production was higher but was littlealtered by the treatments. The two hybrids He-X/3 and He-K/7showed the greatest increases in ethylene evolution with time.With He-K/7 exposed to the gas mixture the production of ethylenedecreased after the initial sharp rise during days 1–2,and reflected the considerable leaf damage observed after day3. The results suggest that sensitivity to air pollution, (as shownby leaf abscission and the formation of stem intumescences)can be correlated with the level of pollutant-induced ethyleneevolution from leaves. Initially high levels could induce abscission,whilst prolonged production could be responsible for intumescenceinitiation. The discussion proposes a series of events fromSO2 and/or O3 entry into the leaf and the physiological reasonsfor the clonal differences. Microswards of white clover (Trifolium repens L.) were grownin controlled environments at 10/7, 18/13 and 26/21 °C day/nighttemperatures. The vertical distribution of leaves of differentages and their rates of 14CO2-uptake in situ were studied. Extending petioles carried the laminae of young leaves throughthe existing foliage. A final position was reached within 1/4to 1/3 of the time between unfolding and death. Newly unfoldedleaves had higher rates of 14CO2-uptake per leaf area than olderones at the same height in the canopy. At higher temperatures,the decrease with age was faster. However, the light-photosynthesisresponse of leaves which were removed from different heightsin the canopy varied much less with leaf age than did the ratesof 14CO2-uptake in situ. The comparison of the rates of 14CO2-uptake in situ with thelight-photosynthesis response curves suggests that young leavesreceive more light than older ones at the same height in thecanopy. This would imply that young white clover leaves havethe ability to reach canopy positions having a favourable lightenvironment. This ability may improve the chances of survivalof white clover in competition with other species. Changes in growth and yield parameters, and 14CO2 and (U-14C)sucrose incorporation into the primary metabolic pool, and essentialoil have been investigated under Mn-deficiency and subsequentrecovery in Mentha piperita, grown in solution culture. UnderMn-deficiency, CO2 exchange rate, total chlorophyll, total assimilatoryarea, plant dry weight, and essential oil yield were significantlyreduced, whereas chlorophyll a/b ratio, leaf area ratio andleaf stem ratio significantly increased. In leaves of Mn-deficientplants, 14CO2 incorporation into the primary metabolic pool(ethanol-soluble and -insoluble) and essential oil were significantlylower, whereas (U-14C) sucrose incorporation into these componentswas significantly higher as compared to the control. Among theprimary metabolites, the label was maximum in sugars, followedby organic acids and amino acids. A higher label in these metaboliteswas, in general, observed in stems of Mn-deficient plants ascompared to the control. Mn-deficient plants supplied with completenutrient medium for 3 weeks exhibited partial recovery in growthand yield parameters, and essential oil biogenesis. Thus, underMn-deficiency and subsequent recovery, the levels of primaryphotosynthetic metabolites and their partitioning between leafand stem significantly influence essential oil biogenesis. The photosynthetic capacity of newly expanded leaves of vernalizedor non-vernalized plants of S24 perennial ryegrass (Lolium perenneL.), grown in long or short photoperiods, was measured in twoexperiments. In the first, leaves were protected from shadingduring development, while in the second, the natural shade ofneighbouring tillers in a sward was allowed. In the first experiment there was little effect of vernalization,day length or flowering, and leaves in all treatments had photosyntheticrates at 250 W m–2 of between 28 and 32 mg CO2 dm–2h–1.In the second experiment the photosynthetic rate of successiveleaves fell as sward leaf area increased. This downward trendwas reversed, however, in flowering tillers in the vernalizedlong-day treatment, while in the other treatments, which didnot flower, photosynthetic capacity continued to fall. It isconcluded that the leaves of reproductive tillers have highphotosynthetic capacities because stem extension carries themto the top of the canopy where they are well illuminated duringexpansion. The influence of a water stress or foliar ABA spraying pretreatmenton stomatal responses to water loss, exogenous ABA, IAA, Ca2+,and CO2 were studied using excised leaves of Solanum melongena.Both pretreatments increased stomatal sensitivity of water loss,in the presence and absence of CO2, but decreased stomatal sensitivityto exogenous ABA. CO2 greatly reduced the effect of exogenouslyapplied ABA. IAA decreased leaf diffusion resistance for controland ABA sprayed leaves, but did not influence the LDR of previouslywater-stressed leaves. The possible role in drought resistance played by sclerophyllywas studied in the Mediterranean oaks Quercus ilex, Q. suberand Q. pubescens. Studies were conducted on leaves at 30, 50and 80% of their final surface area, as well as on mature leavesof the current year's growth in June and September and on 1-year-oldleaves. Leaves of different ages of the three species showed quite differentdegrees of sclerophylly (DS). Q. ilex leaves reached the definitiveDS of 1.75 g dm–2 during leaf expansion; Q. pubescensleaves hardened at the end of their expansion, with a finalDS of 0.93 g dm–2; Q. suber showed the lowest DS of 0.76g dm–2. Leaf conductance to water vapour (g1) of 1-year-old leaves ofQ. ilex, measured in the field, showed a duration of the g1peak values about twice that of the other two species. The minimumleaf relative water content (RWC), however, was near the samein the three species, indicating that water loss was recoveredpartly by Q. ilex leaves. Agrostis capillaris L.5, Festuca vivipara L. and Poaalpina L.were grown in outdoor open-top chambers at either ambient (340 3µmol mol–1) or elevated (6804µmol mol–1)concentrations of atmospheric carbon dioxide (CO2) for periodsfrom 79–189 d. Photosynthetic capacity of source leaves of plants grown atboth ambient and elevated CO2 concentrations was measured atsaturating light and 5% CO2. Dark respiration of leaves wasmeasured using a liquid phase oxygen electrode with the buffersolution in equilibrium with air (21% O2, 0.034% CO2). Photo-syntheticcapacity of P. alpina was reduced by growth at 680 µmolmol–1 CO2 by 105 d, and that of F. vivipara was reducedat 65 d and 189 d after CO2 enrichment began, suggesting down-regulationor acclimation. Dark respiration of successive leaf blades ofall three species was unaltered by growth at 680 relative to340 µmol mol–1 CO2. In F. vivipara, leaf respirationrate was markedly lower at 189 d than at either 0 d or 65 d,irrespective of growth CO2 concentration. There was a significantlylower total non-structural carbohydrate (TNC) concentrationin the leaf blades and leaf sheaths of A. capillaris grown at680µmol mol–1 CO2. TNC of roots of A. capillariswas unaltered by CO2 treatment. TNC concentration was increasedin both leaves and sheaths of P. alpina and F. vivipara after105 d and 65 d growth, respectively. A 4-fold increase in thewater-soluble fraction (fructan) in P. alpina and in all carbohydratefractions in F. vivipara accounted for the increased TNC content. In F. vivipara the relationship between leaf photosyn-theticcapacity and leaf carbohydrate concentration was such that therewas a strong positive correlation between photosynthetic capacityand total leaf N concentration (expressed on a per unit structuraldry weight basis), and total nitrogen concentration of successivemature leaves reduced with time. Multiple regression of leafphotosynthetic capacity upon leaf nitrogen and carbohydrateconcentrations further confirmed that leaf photosynthetic capacitywas mainly determined by leaf N concentration. In P. alpina,leaf photosynthetic capacity was mainly determined by leaf CHOconcentration. Thus there is evidence for down-regulation ofphotosynthetic capacity in P. alpina resulting from increasedcarbohydrate accumulation in source leaves. Leaf dark respiration and total N concentration were positivelycorrelated in P. alpina and F. vivipara. Leaf dark respirationand soluble carbohydrate concentration of source leaves werepositively correlated in A. capillaris. Changes in source leafphotosynthetic capacity and carbohydrate concentration of plantsgrown at ambient or elevated CO2 are discussed in relation toplant growth, nutrient relations and availability of sinks forcarbon. Trends in several photosynthetic parameters and their responseto changed growth light were followed for 15 d in leaves ofyoung birch saplings using a rapid-response gas exchange measuringequipment. These in vivo measurements were compared to biochemicalassays that were made from the same leaves after the gas exchangestudies. The measurements were made on leaves that were selectedprior to the study and were at that time of similar age. Forthe first 7 d the photosynthetic parameters were followed fromthe growth conditions of moderate light (200 µmol m–2s–1; referred to as controls later in the text). On day7 some of the saplings were transferred to grow either underhigh (450 µmol m–2 s–1; referred to as highlight plants) or low (75 µmol m–2 s–1; referredto as low light plants) light and the capability of the preselectedleaves for acclimation was followed for 6 d. For comparison,at the end of the experiment the measurements were made on bothcontrols and on young leaves that had developed under high andlow light. Generally the in vivo measured rate of CO2 uptake (gross photosynthesis)both at 310 ppm CO2 and 2000 ppm CO2 corresponded very wellto the biochemically determined CO2 fixation capacity in vitroafter rapid extraction (measured as the initial and total activityof Rubisco, respectively). However, if the flux of CO2 intothe chloroplasts was limited by the closure of the stomata,as was the case of the high light plants, then the in vitromeasured Rubisco activity was greater than the in vivo measuredCO2 uptake. Vmax, calculated from the mesophyll conductanceat 1% O2, exceeded the initial activity of Rubisco (assayedat saturating RuBP and CO2) constantly by 60%. The catalyticactivity of Rubisco in birch leaves was overall very low, evenwhen calculated from the total activity of Rubisco (Kcat 0.63–1.18 s–1), when compared to herbaceous C3 species. Signs of light acclimation were not observed in most of thephotosynthetic parameters and in chloroplast structure whenmature birch leaves were subjected to changes in growth lightfor 6 d. However, the change of the growth light either to highor low light caused day-to-day fluctuations in most of the measuredphotosynthetic parameters and in the case of the high lightplants signs of photoinhibition and photodestruction were alsoobserved (decrease in the amount of chlorophyll and increasein chlorophyll a/b ratio). As a result of these fluctuationsthese plants achieved a new and lower steady-state conditionbetween the light and dark reactions, as judged from the molarratio of RuBP to Rubisco binding site. Mistletoes usually have slower rates of photosynthesis thantheir hosts. This study examines CO2assimilation, chlorophyllfluorescence and the chlorophyll content of temperate host–parasitepairs (nine hosts parasitized by Ileostylus micranthus and Carpodetusserratus parasitized by Tupeia antarctica). The hosts of I.micranthus had higher mean annual CO2assimilation (3.59 ±0.41 µmol m-2 s-1) than I. micranthus(2.42 ± 0.20µmol m-2 s-1), and C. serratus(2.41 ± 0.43 µmolm-2 s-1) showed higher CO2assimilation than T. antarctica(0.67± 0.64 µmol m-2 s-1). Hosts saturated at significantlyhigher electron transport rates (ETR) and light levels thanmistletoes. The positive relationship between CO2assimilationand electron transport suggests that the lower CO2assimilationrates in mistletoes are a consequence of lower electron transportrates. When photosynthetic rates, ETR and chlorophyll a /b ratioswere adjusted for photosynthetically active radiation, hostsdid not have significantly higher CO2assimilation (3.21 ±0.37 µmol m-2 s-1) than mistletoes (2.54 ± 0.41µmol m-2 s-1), but still had significantly higher ETRand chlorophyll a / b ratios. The electron transport rates,saturating light and chlorophyll a / b ratios of sun leavesfrom mistletoes were similar to host shade leaves. These responsesindicate that in comparison with their hosts, mistletoe leaveshave the photosynthetic characteristics of the leaves of shadeplants.
