Newly moulted females of the insect Oncopeltus fasciatus were exposed to cadmium (Cd) dissolved in the drinking water (50-400 mg l(-1) Cd) for 5 days. Cd exposure delayed ovarian maturation and inhibited egg production. Exposure to Cd, moreover, decreased hemolymph levels of the two major vitellogenin polypeptides of O. fasciatus, VG1 and VG2, in a concentration-dependent way, probably by a reduction in their synthesis. The ovarian levels of VG1 and VG2 were also decreased in Cd-exposed females. It was next investigated whether Cd effects might be a consequence of the endocrine disruption of vitellogenin synthesis, which is controlled by juvenile hormone (JH). JH replacement therapy did not restore VG1 or VG2 levels in Cd-exposed females, but did so in starved females. Our results do not therefore support a disturbance of JH production or a reduction in feeding as the cause of the reduced vitellogenin polypeptide levels, but rather point to the site of action of JH, the JH receptor, as the target of Cd effects.  相似文献

Progress in ecotoxicological research documents that crustaceans are highly vulnerable to diverse chemicals and toxicants in the environment. In particular, pollutants affecting endocrine homeostasis in crustaceans (i.e., endocrine disruptors) are intensively studied, and serious reproductive disorders have been documented. In this review, current knowledge about the endocrine regulation of the crustacean reproduction is put together with the published ecotoxicological data with an attempt to summarize the potential of xenobiotics to affect crustacean reproduction. Following gaps and trends were identified: (1) Studies are required in the field of neurohormone (serotonin and dopamine) regulation of the reproduction and possible modulations by environmental toxicants such as antidepressant drugs. (2) Molting-related parameters (regulated by ecdysteroid hormones) are closely coordinated with the development and reproduction cycles in crustaceans (cross-links with methyl farnesoate signalling), and their susceptibility to toxicants should be studied. (3) Other biochemical targets for xenobiotics were recently discovered in crustaceans and these should be explored by further ecotoxicological studies (e.g., new information about ecdysteroid receptor molecular biology). (4) Some sex steroid hormones known from vertebrates (testosterone, progesterone) have been reported in crustaceans but knowledge about their targets (crustacean steroid receptors) and signalling is still limited. (5) Determination of the sex in developing juveniles (affecting the sex ratio in population) is a sensitive parameter to various xenobiotics (including endocrine disruptors) but its modulation by general environmental stress and non-specific toxicity should be further studied.  相似文献

Cadmium (Cd) is a ubquitous element and a significant inorganic pollutant that has previously been found to bioaccumulate in reproductive organs of fish and disrupt important endocrine processes, especially those involved in synthesis, release and metabolism of hormones. Clearly, there is potential for reproductive effects in fish populations exposed to Cd, however, few studies have investigated the non-lethal consequences of Cd in fish. To this extent, adult male and female Japanese medaka were exposed to 0-10 ppb Cd for 7 weeks. Reproductive endpoints were monitored during weeks 6 and 7 of exposure and compared to physiological responses along the hypothalamus-pituitary-gonadal (HPG) axis, including plasma vitellogenin (VTG), hepatic estrogen receptor (ER), plasma steroids, gonadal-somatic indices (GSI), and gonadal steroid release. There were no observed effects on VTG and ER by long-term Cd exposure. However, gonadal steroid release was significantly decreased in males and females at all exposure concentrations and female plasma estradiol levels were significantly altered at concentrations higher than 5 ppb Cd. Overall, responses along the HPG axis were more sensitive to Cd exposure than the reproductive and developmental endpoints, which were not affected in this study, indicating that higher level impairment in fish might be relatively protected.  相似文献

The objectives of this study were to derive a useful case definition of delayed resumption of ovarian activity, based on factors associated with reduced fertility, and to assess its impact on subsequent reproductive performance in Holstein cows (Bos taurus). Milk samples were collected twice weekly from 219 cows from four commercial herds, and whole-milk progesterone concentrations were determined with an enzyme-linked immunosorbent assay. Ovulation was considered to have occurred 5 d before the first rise of milk progesterone concentration above the basal level. Survival analysis was used to derive a case definition of delayed resumption of ovarian activity postpartum based on factors that were predictive of reduced pregnancy rate. First postpartum ovulation occurring beyond 35 d postpartum was associated with a reduced pregnancy rate (hazard ratio [HR] = 0.50; P < 0.001) and was defined as delayed resumption of ovarian activity; overall, 75 (34.9%) cows were in this category. These cows were more likely not to conceive on first artificial insemination (odds ratio [OR] = 2.85; P = 0.01) and more likely not to become pregnant within 100 d (OR = 3.30; P = 0.001) and 210 d (OR = 3.20; P < 0.001) postpartum compared with cows with normal resumption of ovarian activity. Furthermore, 13 (6%) cows that ovulated within 35 d postpartum had a prolonged (≥14 d) interval between either first and second or second and third luteal phases postpartum. A prolonged interluteal interval was also associated with a reduced pregnancy rate (HR = 0.35; P = 0.02). Days open (mean ± SEM) were greater (P = 0.0002) in cows with delayed resumption of ovarian activity (213 ± 13 d) and in cows with prolonged interluteal interval (220 ± 37 d) than in cows with normal resumption of ovarian activity (152 ± 9 d). In conclusion, first ovulation occurring beyond 35 d postpartum was defined as delayed resumption of ovarian activity, and the first ovulation occurring within 35 d postpartum but the absence of luteal activity ≥14 d between two consecutive luteal phases was defined as a prolonged interluteal interval; both abnormalities adversely affected the subsequent reproductive performance of Holstein cows.  相似文献

Cadmium (Cd) is a ubiquitous element and an important anthropogenic metal contaminant. A series of assays were modified or developed for Japanese medaka (Oryzias latipes), and used to compare the effects of Cd exposure on indicators of endocrine function in adult animals previously exposed in ovo or as hatchlings. Adults were raised either from eggs produced during a 2 week exposure to 0-10 microg/l Cd or from fry exposed for 2 weeks beginning 2 days after hatching. The reproductive capacity of the resulting adults was determined during a 2 week period during which half of the animals were re-exposed to Cd. Two week Cd exposure did not result in reproductive impairment despite producing some changes in circulating steroid concentration. In addition, 1 microg/l cadmium exposure in ovo elevated male hepatic vitellogenin (VTG) relative to controls. Hence, steroid parameters were a better biomarker of cadmium exposure than changes in VTG. However, reproductive impairment was not correlated to change in VTG or plasma steroids.  相似文献

Synopsis Changes in serum steroid hormones were studied during the reproductive cycle of a viviparous rockfish,Sebastiscus marmoratus. Serum levels of estradiol-17β (E2) and testosterone (T) were moderately high throughout the spawning period from December until February (E2), and until post-spawning in April (T). Serum progesterone (prog) fluctuated but remained low throughout the annual reproductive cycle; 17α,20β-dihydroxy-4-pregnen-3-one (17α, 20β-diOHprog), on the other hand, was relatively high during the spawning period. During the spawning period, 7 of 12 females reared under laboratory conditions spawned twice at 10-to 16-day intervals. Histological observations indicated that oocytes developed gradually during gestation of the preceding brood and; after parturition, developed more quickly towards the end of vitellogenesis and subsequent fertilization. In repeat spawners, E2 and female-specific serum proteins remained high several days after the first parturition, then gradually decreased. Prog showed no significant changes over the period. The 17α, 20β-diOHprog, however, was low immediately after parturition, then rapidly increased, remained elevated during the middle of the period and then decreased. These results indicate that E2 is involved in vitellogenesis, and 17α, 20β-diOHprog may have some important roles in gestation in the multiple spawnerS. marmoratus.  相似文献

Honey bee (Apis mellifera L.) workers are essentially sterile females that are used to study how complex social behavior develops. Workers perform nest tasks, like nursing larvae, prior to field tasks, like foraging. Despite worker sterility, this behavioral progression correlates with ovary size: workers with larger ovaries (many ovary filaments) start foraging at younger ages on average. It is untested, however, whether the correlation confers a causal relationship between ovary size and behavioral development. Here, we successfully grafted supernumerary ovaries into worker bees to produce an artificial increase in the amount of ovary tissue. We next measured fat body mRNA levels for the yolk precursor gene vitellogenin, which influences honey bee behavioral development and can correlate with ovary size. Vitellogenin was equally expressed in surgical controls and bees with supernumerary ovaries, leading us to predict that these groups would be characterized by equal behavior. Contrary to our prediction, bees with supernumerary ovaries showed accelerated behavioral development compared to surgical controls, which behaved like reference bees that were not treated surgically. To explore this result we monitored fat body expression levels of a putative ecdysteroid-response gene, HR46, which is genetically linked to ovary size in workers. Our data establish that social insect worker behavior can be directly influenced by ovaries, and that HR46 expression changes with ovary size independent of vitellogenin.  相似文献

The fathead minnow (Pimephales promelas) has been proposed as a model species for assessing the adverse effects of endocrine-disrupting chemicals (EDCs) on reproduction and development. The purpose of these studies was to develop baseline reproductive biology and endocrinology data for this species to support interpretation of tests with potential EDCs. Pairs of reproductively-active fathead minnows (n=70) were evaluated with respect to reproductive cyclicity in terms of spawning interval and fecundity. The mode and mean (+/-SE) spawning intervals for the fish in this study were 3.0 and 3.7+/-0.1 days, respectively. The mean number of eggs produced per spawn was 85+/-2.8. Animals were sacrificed at periodic intervals during the established spawning cycle and measurements made of gonadal condition (gonadosomatic index [GSI], histopathology) and plasma concentrations of vitellogenin and sex steroids (beta-estradiol, testosterone, 11-ketotestosterone). The GSI in females varied significantly as a function of spawning interval, with the largest values occurring day 2 post-spawn, just prior to the interval of maximum spawning activity. Plasma beta-estradiol concentrations in females also varied significantly relative to peak values in the GSI and spawning activity. Vitellogenin concentrations in the female, and male GSI and steroid concentrations did not vary significantly relative to position in the spawning cycle. Concentrations of beta-estradiol in females and 11-ketotestosterone in males were positively correlated with testosterone concentrations.  相似文献

The eurohaline fish, Fundulus heteroclitus, adapts rapidly to enhanced salinity by increasing the ion secretion by gill chloride cells. An increase of approximately 70 mOsm in plasma osmolarity was previously found during the transition. To mimic this in vitro, isolated opercular epithelia of seawater-adapted Fundulus mounted in a modified Ussing chamber were exposed to an increase in NaCl and/or osmolarity on the basolateral side, which immediately increased I(SC). Various Cl(-) channel blockers as well as the K(+) channel blocker Ba(2+) added to the basolateral side all inhibited the steady-state as well as the hypertonic stimulation of I(SC). The exists -agonist isoproterenol stimulates I(SC) in standard Ringer solutions. In contrast, when cell volume was kept at the larger value by simultaneous addition of water, the stimulation with isoproterenol was abolished, suggesting that the key process for activation of the Na(+), K(+), 2Cl(-) cotransporter is cell shrinkage. The protein kinase C (PKC) inhibitor chelerythrine and the myosin light chain kinase (MLCK) inhibitor ML-7 had strong inhibitory effects on the mannitol activation of I(SC), thus both MLCK and PKC are involved. The two specific protein kinase A (PKA) inhibitors H-89 and KT 5720 had no effect after mannitol addition whereas isoproterenol stimulation was completely blocked by H-89. This indicates that PKA is involved in the activation of the apical Cl(-) channel via c-AMP whereas the shrinkage activation of the Na(+), K(+), 2Cl(-) cotransporter is independent of PKA activation. The steady-state Cl(-) secretion was stimulated by an inhibitor of serine/threonine phosphatases of the PP-1 and PP-2A type and inhibited by a PKC inhibitor but not by a PKA inhibitor. Thus, it seems to be determined by continuous phosphorylation and dephosphorylation involving PKC but not PKA. The steady-state Cl(-) secretion and the maximal obtainable Cl(-) secretion were measured in freshwater-adapted fish and in fish retransferred to saltwater. No I(SC) could be measured in freshwater-adapted fish or in the fish within the first 18 h after transfer to saltwater. As evidenced from Western blot analysis using antiserine-antibodies, a heavily serine phosphorylated protein of about 190 kDa was consistently observed in the saltwater-acclimated fish, but was only weakly present in freshwater-acclimated fish. This observation indicates that acclimatization to saltwater stimulates the expression of this 190-kDa protein and/or a serine/threonine kinase, which subsequently phosphorylates the protein.  相似文献

The current models for branchial acid excretion in fishes include Na(+)/H(+) exchange and the electrogenic excretion of H+ via H+-ATPase. The predominant route of acid excretion in some freshwater fishes is thought to be via the H+-ATPase/Na+ channel system. The euryhaline Fundulus heteroclitus may not fit this profile even when adapted to freshwater (FW). We hypothesize that the Na+/H+ exchanger (NHE) in this species may play a predominant role in acid-base regulation for both marine and FW adapted animals. Acidosis induced by ambient hypercapnia (1% CO2 in air), resulted in an increase in net H+ excretion to the water in F. heteroclitus pre-adapted to FW, brackish (isoosmotic; BW) and seawater (SW). Both FW and SW adapted mummichogs were tested for NHE protein expression using mammalian NHE antibodies, and we identified NHE-like immunoreactive proteins in gill membrane preparations from both groups. Hypercapnia induced a approximately three-fold elevation in gill NHE2-like protein in FW animals but SW adapted fish showed inconsistent NHE3-like protein expression. There was no change in NHE-1 levels in FW fish. In contrast, SW animals demonstrated a significant increase in both NHE1 and NHE3-like proteins following hypercapnia but limited expression of the NHE2 protein. We hypothesize that different isoforms of NHE may be preferentially expressed depending on the salinity to which the animals are adapted. Net H+ transfers during acidosis may be driven, at least in part by the action of these transporters.  相似文献

Teleost fish experience passive osmotic water influx in fresh water (FW) and water outflux in salt water, which is normally compensated by water flow driven by active ion transport mechanisms. Euryhaline fish may also minimize osmotic energy demand by "behavioral osmoregulation", seeking a medium isotonic with their body fluids. Our goal was to evaluate the energy requirement for osmoregulation by the euryhaline fish Fundulus heteroclitus, to determine whether it is of sufficient magnitude to favor behavioral osmoregulation. We have developed a method of weighing small fish repetitively for long periods without apparent damage, which was used to assess changes in water content following changes in external salinity. We found that cold (4 degrees C) inhibits osmoregulatory active transport mechanisms in fish acclimated to warmer temperatures, leading to a net passive water flux which is reversed by rewarming the fish. A sudden change of salinity at room temperature triggers a transient change in water content and the initial slope can be used to measure the minimum passive flux at that temperature. With some reasonable assumptions as to the stoichiometry of the ion transport and ATP-generating processes, we can calculate the amount of respiration required for ion transport and compare it to the oxygen uptake measured previously under the same conditions. We conclude that osmoregulation in sea water requires from 6% to 10% of the total energy budget in sea water, with smaller percentages in FW, and that this fraction is probably sufficient to be a significant selective driving force favoring behavioral osmoregulation under some circumstances.  相似文献

Larval and juvenile fishes were collected at low tide from the surface of an intertidal salt marsh on Sapelo Island, Georgia every 6 days from 25 May through 20 December 1982. Larval fishes were present in shallow puddles of tidal water on the marsh from the beginning of the sampling period until the end of October. Juveniles were present throughout the year. Fundulus heteroclitus (Linnaeus) and F. luciae (Baird) accounted for 96.3% (67.0% and 29.3%, respectively) of the 4355 fishes collected. These fishes exhibited synchronous, temporal pulses in larval abundance, suggesting that reproduction was discontinuous and controlled by the same factor(s) in both species. Larval F. heteroclitus inhabited aquatic micro-habitats near the upland edge of the intertidal marsh, but as the larvae grew to juvenile size (≈ 10 mm standard length) they moved to lower elevations near a tidal creek. Large juveniles and adults of F. heteroclitus infrequently occurred in the samples, presumably because they leave the intertidal marsh as the tide ebbs. F. luciae were rarely found in low marsh areas, but all age classes (including adults) occurred at higher elevations, supporting previous suggestions that this species prefers high marsh habitats. The vegetated, intertidal salt marsh appears to be the principal nursery habitat for both of these cyprinodontid species.  相似文献

Exposure to estrogenic substances during a time window, the so-called critical period, in perinatal life causes an irregular development of the genital tract that leads to ovary-independent proliferation and cornification in the vaginal epithelium in mice. We have previously demonstrated that retinol inhibits the irreversible effects of estrogen on the vagina. Here, mice kept in a vitamin-A-deficient condition during perinatal life were shown to be more sensitive to the harmful effects of estrogen. In addition, expression of mRNA for retinol binding protein type 2 (CRBP2), a small intestine-specific cytosolic protein that captures intracellular retinal and retinol, was detected in the vaginal epithelium. Induction of increased expression of CRBP2 mRNA by estrogen was also evident in the uterus and epididymis. Both estradiol-17 and diethylstilbestrol markedly increased the tissue content of CRBP2 mRNA in the vagina and uterus during the neonatal critical period but not after 15 days of age. These results taken together imply that estrogen disrupts the local vitamin A balance by an induction of CRBP2 gene expression in the epithelium in the developing mouse genital tract, and that retinoid imbalance may contribute to the genesis of irreversible effects of estrogen on the vagina.This work was supported by Grants-in-Aid for Priority Area B (to T.M. and M.M.) and for Young Scientists (to M.M.) from the Ministry of Culture, Sports, Science and Technology, Japan  相似文献

Purpose

This study aims to analyze and quantify the environmental impacts associated with the production of testliner paper using 100?% recovered paper as fiber raw material, by applying the life cycle assessment principles. A simulation of advanced sorting technology was done to prepare and use batches of raw materials with different levels of contaminants. Comparative studies of environmental impact assessment were focused on the quality of recovered paper, which is decisively influenced by the efficiency of the sorting process. The particularity of the study is that so far it is the only one that analyzes the environmental impact generated by recovered paper quality.

Methods

To analyze the environmental impacts in the scenarios, life cycle assessment methodology was considered. Potential environmental impacts were assessed by using the CML 2009, Dec.07 method developed by the Centre for Environmental Science from the University of Leiden.

Results and discussion

In this study, acidification potential, abiotic resources depletion potential, eutrophication potential, global warming potential, photochemical ozone creation potential, and human toxicity potential were the impact categories analyzed. Considering that the system boundaries refer only to the paper mill that was obtained, all unitary processes involved in the manufacturing of product system influence in varying proportions the impact categories chosen for evaluation. A higher concentration of contaminants leads to a higher amount of energy and water used, and thus, a significant amount of waste and emissions generated. Simulations performed have highlighted the importance of sorting technology that influences the quality of raw material that will be used.

Conclusions

Utilization of recovered paper batches with a low quality contributes to an increased environmental impact associated with the testliner paper manufacturing stage. A low quality of recovered paper will influence energy consumption in different modules of the system (recycled fiber pulp preparation, paper machine, and wastewater treatment), the volume of waste generated, and consequently the emissions released both in air and water.  相似文献

The effect of ovariectomy on feeding, mating, Juvenile Hormone (JH) production, and maternal behaviour was assessed in female ring-legged earwigs, Euborellia annulipes (Lucas) (Dermaptera: Carcinophoridae), during the first 16 days of adult life (the first gonadotrophic cycle and early brooding). Ovariectomy of 2-day-old adults did not affect weight gain, nor did it alter mating behaviour on day 7. Similarly, ovariectomy did not prevent the increase in JH biosynthesis that accompanies vitellogenesis in this species, which suggests a cycle of JH production that is not dependent on the presence of the ovaries. Both ovariectomy and mating status affected feeding behaviour. Most introduced eggs were consumed (i.e. disappeared) within 24 h, and younger (7-day-old) females consumed more eggs than did older ones. However, 12-day-old intact virgins and 16-day-old ovariectomized, mated females consumed fewest eggs, and allowed some hatching. Thus, ovariectomy did not abolish changes in feeding behaviour that normally accompany reproduction but, instead, appeared to delay the reduction in feeding that normally accompanies the completion of the cycle of egg development. By contrast, mating enhanced the reduction in feeding late in the reproductive cycle. Mating significantly enhanced maternal behaviour in both ovariectomized and sham-operated females. Hatching success from egg clutches introduced to day 16 virgin or mated females that had been ovariectomized or sham-operated on day 2, was significantly greater in the mated groups.  相似文献

Estuarine fish, such as the Atlantic killifish (Fundulus heteroclitus), are constantly and rapidly exposed to changes in salinity. Although ion transport in killifish gills during acclimation to increased salinity has been studied extensively, no studies have examined the role of aquaglyceroporin 3 (AQP3), a water, glycerol, urea, and ammonia transporter, during acclimation to increased salinity in this sentinel environmental model organism. The goal of this study was to test the hypothesis that transfer from freshwater to seawater decreases AQP3 gene and protein expression in the gill of killifish. Transfer from freshwater to seawater decreased AQP3 mRNA in the gill after 1 day, but had no effect on total gill AQP3 protein abundance as determined by western blot. Quantitative confocal immunocytochemistry confirmed western blot studies that transfer from freshwater to seawater did not change total AQP3 abundance in the gill; however, immunocytochemistry revealed that the amount of AQP3 in pillar cells of secondary lamellae decreased in seawater fish, whereas the amount of AQP3 in mitochondrion rich cells (MRC) in primary filaments of the gill increased in seawater fish. This response of AQP3 expression is unique to killifish compared to other teleosts. Although the role of AQP3 in the gill of killifish has not been completely elucidated, these results suggest that AQP3 may play an important role in the ability of killifish to acclimate to increased salinity.  相似文献
