The configurations at C-24 of 24-alkylsterols of six samples of Clerodendrum species (Verbenaceae) - the aerial parts of C. fragrans, C. inerme, C. infortunatum, C. scandens, and C. siphonanthus, and the seeds of C. infortunatum - were examined by NMR. All samples contained 24 beta-ethylsterols possessing a delta 25-bond, clerosterol and 22-dehydroclerosterol, as the dominant sterol components. The other 24-ethylsterols lacking a delta 25-bond, 24-ethyl-22-dehydrocholestanol, 24-ethylcholesterol, and 24-ethyl-22-dehydrocholesterol, which were present as minor components, were shown to be mixtures of the 24 alpha- and 24 beta-epimers, with the 24 alpha-epimers predominating in all cases. Four minor 24-methyl-sterols, 24-methylcholestanol, 24-methylcholesterol, 24-methyl-22-dehydrocholesterol, and 24-methyllathosterol, were shown to be C-24 epimeric mixtures, whereas two others, 24-methyl-22,25-bisdehydrocholesterol and 24-methyl-22-dehydrolathosterol, were found to be present only as the 24 beta-epimers. This is the first report of the occurrence of 24 beta-ethyl-22-dehydrocholestanol in higher plants.  相似文献

The sterol composition of the warm-water brown alga Sargassum fluitans Børgesen of the Gulf of Mexico was determined by TLC, GLC and IR measurements. The presence of over ten sterols was suggested, of which four (fucosterol, cholesterol, 24-methylenecholesterol, and trans- 22-dehydrocholesterol) were identified and four (a 24-methylcholesterol, a 24-ethylcholesterol, a 24-methyl-trans-22-dehydrocholestero 1 and a 24-ethyl-trans-22-dehydrocholesterol) were recognized but not definitively identified. Saringosterol and 24-ketocholesterol were not found. The crude sterol mixture from S. fluitans was oxidized by osmium tetroxide to 24-ketocholesterol in poor yield.  相似文献

The free sterol mixture of the sponge Stylotella agminata contained a series of 3-hydroxymethyl-A-nor sterols of 5 alpha-cholestane, 5 alpha-cholest-22-ene, 24-methyl-5 alpha-cholestane, 24-ethyl-5 alpha-cholestane, and 24-ethyl-5 alpha-cholest-22-ene. A new cyclopropane-containing sterol was isolated and shown to be 3 epsilon-hydroxymethyl-A-nor-5 alpha-gorgostane. Sterols with conventional nuclei were also present as minor constituents;  相似文献

Free and esterified sterols of eggs of the root-knot nematodes Meloidogyne incognita races 2 and 3 and M. arenaria race 1 were isolated and identified by gas-liquid chromatography-mass spectrometry. The major sterols of eggs of each race were 24-ethylcholesterol (33.4-38.8% of total sterol), 24-ethylcholestanol (18.3-25.3%), 24-methylcholesterol (8.6-11.7%), 24-methylcholestanol (7.7-12.5%), and cholesterol (4.6-11.6%). Consequently, the major metabolic transformation performed by Meloidogyne females or eggs upon host sterols appeared to be saturation of the sterol nucleus. The free and esterified sterols of the same race did not differ appreciably, except for a slight enrichment of the steryl esters in cholesterol. Although the sterol composition of Meloidogyne eggs differed from that of other life stages of other genera of plant-parasitic nematodes, the three Meloidogyne races could not be distinguished from each other by their egg sterols. Ecdysteroids, compounds with hormonal function in insects, were not detected by radioimmunoassay in the Meloidogyne eggs either as free ecdysteroids or as polar conjugates.  相似文献

Sterols from cultured Dictyocha fibula Ehrenberg and Olisthodiscus luteus Carter were extracted and identified for comparison with sterols from other Chromophycota species. Although orientation at C-24 was not absolutely determined, the sterols of Dictyocha, a silicoflagellale, appeared to consist of only 24-methyl-22-dehydrocholesterol and 24-methylenecholesterol, a sterol composition not known in any other alga. This is in keeping with the taxonomic isolation Dictyocha has among algae. On the other hand, Olisthodiscus luteus contained 24-ethylcholesterol, 24-ethylcholestanol, 24-methylcholesterol, and cholesterol. This composition is in accord with sterols of members of the Xanthophyceae and Raphidophyceae.  相似文献

Sterols of both roots and nodules of Vicia faba were found to be mainly 24-methylcholesterol, 24-ethyl-5,22-cholestadien-3β-ol and 24-ethylcholesterol. Nodules contained nearly twice the quantity of sterols present in roots, mainly due to an increase in 24-ethylcholesterol from about 40–60% of the total sterol content of root extract to over 80% of nodule extracts. Insignificant amounts of sterol were detected in nodule bacteroid fractions and in free-living Rhizobium leguminosarum.  相似文献

Sterols from free sterol and steryl ester fractions from Heterodera zeae and from total lipids of Zea mays roots were analyzed by gas-liquid chromatography (GLC) and by GLC-mass spectrometry. The major free sterols of H. zeae were 24-ethylcholesterol (54.4% of total free sterol), 24-ethylcholesta-5,22-dien-3β-ol (13.3%), 24-methylcholesterol (12.5%), and cholesterol (7.2%). The same four sterols comprised 34.6%, 7.2%, 30.3%, and 18.6%, respectively, of the esterified sterols of H. zeae. Corn root sterols included 46.6% 24-ethylcholesta-5,22-dien-3β-ol, 16.7% methylcholesterol, 16.4% cycloartenol, 12.7% 24-ethylcholesterol, and 0.5% cholesterol. The sterol 24-composition of H. zeae differed greatly from that of the only other cyst nematode previously investigated, Globodera solanacearum.  相似文献

The Zygomycetes fungus Mortierella alpina was cultured to growth arrest to assess the phytosterol biosynthesis pathway in a less-advanced fungus. The mycelium was found to produce 13 sterols, but no ergosterol. The sterol fractions were purified to homogeneity by HPLC and their identifies determined by a combination of GC-MS and 1H NMR spectroscopy. The principal sterol of the mycelium was cholesta-5, 24-dienol (desmosterol) (83%), with lesser amounts of 24beta-methyl-cholesta-5,25(27)-dienol (codisterol) (2%), 24-methyldesmosterol (6%), 24(28)-methylene cholesterol (3%) and lanosterol (3%) and several other minor compounds (3%). The total sterol accounted for approximately 0.07% of the mycelial dry wt. Mycelium fed methionine-methyl-2H3 for 6 days, generated 3 2H-24-methyl(ene) sterols, [C28-2H2]24(28)-methylenecholesterol, [C28-2H3]24-methylcholesta-5,24-dienol and [C28-2H3]24beta-methyl-cholesta-5,25(27)-dienol. The formation of the 24-methyl sterols seems to be catalyzed by the direct methylation of a common Delta24-acceptor sterol thereby bypassing the intermediacy of an isomerization step for rearrangement of the Delta24(28)-bond to Delta25(25)-position as operates in Ascomycetes fungi and all plants.  相似文献

Several new 4α-methyl sterols with unusual unsaturation in the Δ8(14)-or Δ14-positions, 4α,24S-dimethyl-5α-cholest-8 (14)-en-3β-ol, 4α-methyl-24ξ-ethyl-5α-cholest-8(14)-en-3β-ol, 4α-methyl-24(Z)-ethylidene-5α-cholest-8(14)- en-3β-ol, 4α,23 (or 22),24ξ-trimethyl-5α-cholesta-8(14),22-dien-3β-ol, 4α,24S(or 23ξ)-dimethyl-5α-cholest-14-en-3β-ol and 14-dehydrodinosterol, have been isolated from extracts of the cultured marine dinoflagellates Amphidinium carterae, A. corpulentum and Glenodinium sp. 4α-Methyl-24ξ-ethyl-5α-cholestan-3β-ol was isolated from the steryl ester fraction of Glenodinium sp. The structures of these new sterols are based upon extensive 360 MHz 1H NMR and MS analyses.  相似文献

When Chlorella sorokiniana was grown in the presence of 4 ppm AY-9944 total sterol production was unaltered in comparison to control cultures. However, inhibition of sterol biosynthesis was shown by the accumulation of a number of sterols which were considered to be intermediates in sterol biosynthesis. The sterols which were found in treated cultures were identified as cyclolaudenol, 4α,14α-dimethyl-9β,19-cyclo-5α-ergost-25-en-3β-ol, 4α,14α-dimethyl -5α-ergosta-8,25-dien-3β-ol, 14α-methyl-9β,19-cyclo-5α-ergost-25-en-3β-ol, 24-methylpollinastanol, 14α-methyl-5α-ergost-8-en-3β-ol, 5α-ergost -8(14)-enol, 5α-ergost-8-enol, 5α-ergosta-8(14),22-dienol, 5α-ergosta-8,22-dienol, 5α-ergosta-8,14-dienol, and 5α-ergosta-7,22-dienol, in addition to the normally occurring sterols which are ergosterol, 5α-ergost-7-enol, and ergosta-5,7-dienol.The occurrence of these sterols in the treated culture indicates that AY-9944 is an effective inhibitor of the Δ8 → Δ7 isomerase and Δ14-reductase, and also inhibits introduction of the Δ22-double bond. The occurrence of 14α-dimethyl-5α-ergosta-8,25-dien-3β-ol and 14α-methyl-9β,19-cyclo-5α-ergost -25-en-3β-ol is reported for the first time in living organisms. The presence of 25-methylene sterols suggests that they, and not 24-methylene derivatives, are intermediates in the biosynthesis of sterols in C. sorokiniana.  相似文献

The complex sterol mixture isolated from A, nigra was found to contain a low level of Δ4-3-keto steroids, 5β-stanols and 4α-methyl sterols in addition to regular (4-demethyl) sterols. The following new marine sterols were isolated and identified using MS and 360 MHz NMR: 5β-cholest-22E-en-3β-ol, 24S-methyl-5β-cholest-22E-en-3β-ol, 24-methylene-5β-cholestan-3β-ol, both epimers at C-24 of 4α-methyl-24-ethyl-5α-cholest-22E-en-3β-ol, 4α, 22ξ, 23ξ-(or 24ξ-)trimethyl-5α-cholest-8(14)-en-3β-ol and (22S, 23S, 24S)-4α-24-dimethyl-22, 23-methylene-5α-cholestan-3β-ol. The latter sterol and 23-demethylgorqosterol have opposite configurations at C-22, C-23, and C-24; the Δ8(14) sterol has an unprecedented side chain.  相似文献

Two new sterols, epipolasterol and 22(23)-dihydroepipolasterol, have been isolated from the marine sponge Epipolasis sp. These are unusual metabolites as they both contain a t-butyl group in the sterol side chain. In addition, the presence of two degrees of unsaturation in the side chain of epipolasterol is rare. The known sterol, 22-dehydro-24-isopropylcholesterol was also found in this sponge.  相似文献

Fourteen 4α-methyl sterols have been isolated from the gorgonians Briareum asbestinum, Gorgonia mariae, Muriceopsis flavida and Pseudoplexaura wagenaari, including the following five new sterols: 4α-methyl-24-methylene-5α-cholestan-3β-ol, (24R)-4α, 24-dimethyl-5α-cholesta-7,22-dien-3,β-ol, 4α,24S(or 23ξ)-dimethyl-5α-cholest-7-en-3β-ol, (22E, 24R)-4α,23,24-trimethyl-5α-cholesta-7,22-dien-3β-ol and (24R)-4α,24-dimethyl-5α-cholesta-8(14),22-dien-3β-ol. There is strong evidence that these 4α-methyl sterols are synthesized by the algal (dinoflagellate) symbionts (zooxanthellae) of the gorgonians. It is suggested that analysis of 4Δ-methyl sterol mixtures isolated from a zooxanthellae-bearing invertebrate, collected in several different geographic locations, might give information on the specificity of the symbiotic association between a given animal species and a particular strain of zooxanthellae.  相似文献

Recent studies have led to the identification of an unusual class of dihydroxysterols (steroidal diols termed “pavlovols”)in a few species of microalgae from the genus Pavlova (family Pavlovaceae, class Haptophyceae = Prymnesiophyceae). These compounds have an additional hydroxyl group at G-4 in the sterol A ring, which appears to be very rare in sterol biosynthetic pathways. The sterol compositions of many other haptophytes from different orders have been analyzed, but to date all have lacked pavlovols. We now report the occurrence of these compounds in Diacronema vlkianum Prauser and two strains of Pavlova pinguis Green. This is the first report of the lipid composition of these species. Both microalgae contained “24-methylpavlovol” (4α, 24-dimethyl-5α-cholestan-3β, 4β-diol), P. pinguis also contained “24-ethylpavlovol” (4α-methyl-24-ethyl-5α-cholestan-3β, 4β-diol), and D. vlkianum contained a diol identified from its mass spectrum as 4α, 24β-dimethyl-5α-cholest-22E-en-3β,4β-diol. Both species contained structurally analogous 4-desmethyl sterols and 4-methyl sterols, although there were major differences in the proportions in each series. The major 4-desmethyl sterol in both species was 24-ethylcholesta-5, 22E-dien-3β-ol and the major 4-methyl sterol was 4α-methyl-24-ethyl-5α-cholest-22E-en-3β-ol. The presence of pavlovols in P. pinguis, combined with earlier data, suggests that all Pavlova species might have this distinguishing lipid feature. However, their identtjication in D. vlkianum extends the occurrence of these compounds to another genus and shows that they are not unique to the genus Pavlova. However, they are probably restricted to species from the order Pavlov ales. The modes of biosynthesis and functions of pavlovols remain unknown.  相似文献

The free sterol mixture of the sponge Psammaplysilla purpurea was shown to contain aplysterol as the major constituent. In addition to other sterols such as 5,7-cholestadien-3β-ol, cholesterol, 5α-cholestan-3β-ol, 24ε-methylcholesta-5,22-dien-3β-ol, 24ε-methylcholesterol, 24ε-ethylcholesta-5,22-dien-3β-ol and 24,28-dehydroaplysterol, a new minor sterol was isolated and shown by spectral analysis as well as partial synthesis to be 3β-hydroxy-26,27-bisnorcholest-5-en-24-one. The sterol mixture contains no other short side chain or 24-keto sterols except for small amounts of 3β-hydroxypregn-5-en-20-one and 3β-hydroxy-5α-pregnan-20-one.  相似文献

Free sterol fractions were isolated from the marine sponges Phyllospongia madagascarensis, Scalarispongia sp., Oceanapia sp., Monanchora clathrata and studied by GLC, GLC–MS, and spectroscopy NMR. P. madagascarensis and Scalarispongia sp. contained common Δ5-sterols; cholesterol was shown to be a main sterol of both the sponges. Oceanapia sp. contained stanols and minor Δ5-sterols with 24R-24,25-methylene-5α-cholestan-3β-ol as a main constituent. Many free sterols from M. clathrata were Δ7-series compounds, and latosterol was a main sterol. Δ4-3-Ketosteroids and Δ5-sterol esters were found in the Antarctic sponge Haliclona sp., but free sterols were practically absent except for trace amount of cholesterol. A chemotaxonomic application of sterols in relation to the genera Phyllospongia, Oceanapia and the family Crambeidae is provided. The known cases of the absence of sterols in sponges and probable reasons of the phenomenon are discussed.  相似文献

More than thirty monohydroxy sterols were detected in the marine sponge Petrosia ficiformis, ranging from the ubiquitous 24-norcholesta-5, 22-dien-3β-o1 to the C30 sterol, 24-propylidenecholesterol. New polyhydroxylated sterols, stylisterol A-C (1-3), and a novel 5,19-cyclosterol, hatomasterol (4) were isolated from the Okinawan sponge Stylissa sp. The biochemical compositions of seven strains of marine cryptomonad and a rhodophyte were determined in logarithmic phase batch (1.4 L flask) and semi-continuous (10 L carboy) culture. Lipid ranged from 13% to 28%, protein ranged from 53% to 68%, and carbohydrate ranged from 9% to 24% of the organic weight. The major lipid classes in the species examined were polar lipids (78-88% of total lipid). The major sterol in the Cryptophyceae and the Rhodophyceae was 24-methylcholesta-5,22E-dien-3beta-ol (62-99% of total sterols); which is also the major sterol in some diatoms and haptophytes. Smaller proportions of cholest-5-en-3beta-ol (1-17.7%) were also found in the Cryptophyceae. Most cryptomonads contained high proportions of the n-3 polyunsaturated fatty acids (PUFA), 18:3n-3 (20.7-29.9% of the total fatty acids), 18:4n-3 (12.5-30.2%), 20:5n-3 (7.6-13.2%) and 22:6n-3 (6.4-10.8%). However, the blue-green cryptomonad Chroomonas placoidea was characterized by a low proportion of 22:6n-3 (0.2% of total fatty acids), and a significant proportion of 22:5n-6 (4.5%), and the presence of 24-ethylcholesta-5,22E-dien-3beta-ol (35.5% of total sterols). The fatty acid composition of the rhodophyte Rhodosorus sp. was similar to those of the Cryptophyceae except for lower proportions of 18:4n-3 and lack of C21 and C22 PUFA. It is postulated that the primary endosymbiosis of a photosynthetic n-3 C18 PUFA-producing prokaryote and a eukaryotic host capable of chain elongation and desaturation of exogenous PUFA, resulted in the Rhodophyceae capable of producing n-3 C20 PUFA. The secondary endosymbiosis of a photosynthetic n-3 C20 PUFA-producing eukaryote (such as a Rhodosorus sp. like-rhodophyte) and a eukaryotic host capable of further chain elongation and desaturation, resulted in the Cryptophyceae being capable of producing n-3 C20 and C22 PUFA de novo. Selected isolates were examined further in feeding trials with juvenile Pacific oysters (Crassostrea gigas). Rhodomonas salina CS-24(containing elevated 22:6n-3) produced high growth rates in oysters; equivalent to the microalga commonly used in aquaculture, Isochrysis sp. (T.ISO).  相似文献

Two new steryl esters with a polyhydroxylated ergostane-type nucleus, 3beta,5alpha-dihydroxy-(22E,24R)-ergosta-7,22-dien-6beta-yl oleate (1) and 3beta,5alpha-dihydroxy-(22E,24R)-ergosta-22-en-7-one-6beta-yl oleate (2), were isolated from the fruiting bodies of the basidiomycete Tricholomopsis rutilans along with three known sterols (3, 4, and 5). The structures of compounds 1 and 2 were established on the basis of spectroscopic means and chemical methods.  相似文献
