There is no data or literature on the effects of supplementing infants with yeast selenium, although its intestinal absorption and bioavailability are higher in adults compared with other selenium compounds. The aim of the present investigation was to study the impact of selenium enriched yeast on the serum selenium concentration of preterm infants living in a low selenium area (Hungary). Twenty-eight preterm infants with mean ± SD birth weight of 962 ± 129 g and gestational age 27 ± 1 wk were randomized into two groups at birth with respect to selenium supplementation. In the supplemented group (n = 14) infants received 4.8 mg yeast selenium containing 5 μg selenium daily via nasogastric drip during the first 14 postnatal days. The nonsupplemented infants were used as a reference group. In the supplemented group, the serum selenium concentration increased from 32.1 ± 8.5 μg/L to 41.5 ± 6.5 μg/L and in the nonsupplemented group it decreased from 25.9 ± 6.8 μg/L to 18.2 ± 6.4 μg/L from birth in two weeks time. Compared with previous studies, our results suggest that the bioavailability of selenium in the form of yeast selenium is higher than that of other selenium compounds used for preterm infants. We did not observe any complications or side-effects owing to enterai yeast selenium supplementation. We conclude that selenium enriched yeast is a safe and an effective form of short-term enterai selenium supplementation for infants.  相似文献

The knowledge about Se bioavailability from animal food is sparse. This study was therefore initiated in order to evaluate the bioavailability of Se from pork meat in humans. Twelve male volunteers (age 21–30 years) participated in a randomised crossover study with strictly controlled diet containing 170 g pig meat/10 MJ per day and 106 ± 13 g Se/d for 3 × 3 weeks. Complete faecal and urinary collections were made during the last week of each period. Bioavailability was evaluated from absorption and retention of Se and changes in plasma Se concentration and blood glutathione peroxidase (GSHPx) activity. The apparent absorption of Se was 94 ± 2% (100 ± 13 g/d). Faecal and urinary excretion were 7 ± 1 g/d and 39 ± 21 g/d, respectively, resulting in a retention of 61 ± 24 g/d. The diet intervention did not affect plasma Se concentration and GSHPx activity. Absorption and retention of Se from pig meat were high suggesting a high availability. However, the availability of pig meat Se for blood Se protein appears to be low.  相似文献

? Few studies have investigated plant-plant interactions involving hyperaccumulator plants. Here, we investigated the effect of selenium (Se) hyperaccumulation on neighboring plants. ? Soil and litter Se concentrations were determined around the hyperaccumulators Astragalus bisulcatus and Stanleya pinnata and around the nonhyperaccumulators Medicago sativa and Helianthus pumilus. We also compared surrounding vegetative cover, species composition and Se concentration in two plant species (Artemisia ludoviciana and Symphyotrichum ericoides) growing either close to or far from Se hyperaccumulators. Then, Arabidopsis thaliana germination and growth were compared on soils collected next to the hyperaccumulators and the nonhyperaccumulators. ? Soil collected around hyperaccumulators contained more Se (up to 266 mg Se kg(-1) ) than soil collected around nonhyperaccumulators. Vegetative ground cover was 10% lower around Se hyperaccumulators compared with nonhyperaccumulators. The Se concentration was higher in neighboring species A. ludoviciana and S. ericoides when growing close to, compared with far from, Se hyperaccumulators. A. thaliana showed reduced germination and growth, and higher Se accumulation, when grown on soil collected around Se hyperaccumulators compared with soil collected around nonaccumulators. ? In conclusion, Se hyperaccumulators may increase the surrounding soil Se concentration (phytoenrichment). The enhanced soil Se contents around hyperaccumulators can impair the growth of Se-sensitive plant species, pointing to a possible role of Se hyperaccumulation in elemental allelopathy.  相似文献

Neighbors of Se hyperaccumulators Stanleya pinnata and Astragalus bisulcatus were found earlier to have elevated Se levels. Here we investigate whether Se hyperaccumulators affect Se localization and speciation in surrounding soil and neighboring plants. X-ray fluorescence mapping and X-ray absorption near-edge structure spectroscopy were used to analyze Se localization and speciation in leaves of Artemisia ludoviciana, Symphyotrichum ericoides and Chenopodium album growing next to Se hyperaccumulators or non-accumulators at a seleniferous site. Regardless of neighbors, A. ludoviciana, S. ericoides and C. album accumulated predominantly (73–92%) reduced selenocompounds with XANES spectra similar to the C-Se-C compounds selenomethionine and methyl-selenocysteine. Preliminary data indicate that the largest Se fraction (65–75%), both in soil next to hyperaccumulator S. pinnata and next to nonaccumulator species was reduced Se with spectra similar to C-Se-C standards. These same C-Se-C forms are found in hyperaccumulators. Thus, hyperaccumulator litter may be a source of organic soil Se, but soil microorganisms may also contribute. These findings are relevant for phytoremediation and biofortification since organic Se is more readily accumulated by plants, and more effective for dietary Se supplementation.  相似文献

A preliminary study was carried out in order to compare the selenium concentration in breast cancer patients and healthy subjects (controls) in Israel. Blood serum samples were obtained from 32 breast cancer patients and 36 controls and were analyzed for selenium by the XRF method. A weighted mean of 0.076±0.014 ppm Se in the blood serum of breast cancer patients, as compared to 0.119±0.023 ppm Se for controls, was obtained. These results indicate that the concentration of selenium in breast cancer patients is significantly lower than in controls. The relationship between selenium concentration and malignancy stage shows an inverse dependence, i.e., the concentration decreases with stage number.  相似文献

Mitochondria from Trigonella foenum-graecum seedlings grown independently in the presence of either selenium (0.75 ppm) or mimosine (0.1 mM) exhibited respiration-stimulated energy-dependent uptake of Ca2+. Uptake studies were carried out independently at a series of Ca2+ concentrations at two different levels: (1) 1–20 μM and (2) 25–1500 μM. Levels of uptake were 50–100% higher in the mitochondria of seedlings of both the Se and mimosine groups. Detailed kinetic analyses revealed negative cooperative effects operative during uptake of Ca2+ at 25–1500 μM given in the medium. Hill coefficients for Ca2+ uptake by the mitochondria of different groups remained unchanged (nH, 0.75). Biphasic Scatchard plots were concave upward, suggestive of two classes of binding sites. High-affinity binding sites were estimated to be 16 nmol/mg protein with dissociation constant (K Ca) of 2.5×109 L/mol. In contrast, graphical analyses of the uptake of Ca2+ in the range 1–20 μM in the medium revealed cooperative effects of positive nature. Selenium has been increasingly recognized as an important element in biological systems, which participates in numerous biochemical processes in organisms, notably in enzyme reactions. Selenium can substitute sulfur of cysteine and methionine to form their selenium analogues, selenocysteine (Sec) and selenomethionine (SeM). The nature of amino acid pockets in proteins is dependent on their composition and thus different non-covalent forces determine the interactions between selenium of Sec or SeM and other functional groups, resulting in specific biophysical behavior. The discrimination of selenium toward sulfur has been reported. In order to elucidate the difference between the nature of S-π and Se-π interactions, we performed extensive DFT calculations of dispersive and electrostatic contributions of Se-π interactions in substituted benzenes/hydrogen selenide (H2Se) complexes. The results are compared with our earlier reported S-π calculations, as well as with available experimental data. Our results show a larger contribution of dispersive interactions in Se-π systems than in S-π ones, which mainly originate from the attraction between Se and substituent groups. We found that selenium exhibits a strong interaction with aromatic systems and may thus play a significant role in stabilizing protein folds and protein–inhibitor complexes. Our findings can also provide molecular insights for understanding enzymatic specificity discrimination between single selenium versus a sulfur atom, notwithstanding their very similar chemical properties.  相似文献

Kashin–Beck disease (KBD) is a special type of endemic osteoarthritis. It has been suggested that alterations in selenium metabolism and apoptosis play a role in KBD. However, the underlying molecular mechanism remains largely unclear. We performed a microarray analysis using RNA isolated from cartilages of KBD patients and healthy controls, through Significance Analysis of Microarray (SAM) software. Functional gene networks and crucial molecules associated with differentially expressed genes were investigated via Ingenuity Pathway Analysis (IPA) and hub gene analysis. Quantitative real-time PCR was used to check the validation of chip test. We identified 52 up-regulated apoptosis-related genes and 26 down-regulated selenium-related genes between KBD and controls, and these genes associated with the “MYC-mediated apoptosis signaling pathway”. We confirmed the results from array studies with quantitative real-time PCR analysis. Our results suggest that abnormal regulation of selenium metabolism and apoptosis through the MYC mediated signaling pathway contributes to the pathogenesis of KBD, but the relationship between apoptosis gene and selenium gene was not found.  相似文献

The real significance of the level of trace elements copper, zinc and selenium in erythrocytes is still a matter of debate. This review is an evaluation of the potential value of copper, zinc and selenium in red blood cells in physiology, nutrition and pathology, taking into account the other commonly used indices and the analytical difficulty encountered for erythrocyte determinations. The main analytical methods and reference values in erythrocytes are also presented. The literature of the last 3 decades was analyzed. On the basis of these papers, the role of erythrocyte copper in various pathologies should be re-investigated, especially when they include inflammatory processes or hormonal changes. The main interest of zinc determination in red blood cells concerns thyroid pathology. In general, the value of selenium in erythrocytes agrees with selenium in serum and its determination is often unnecessary.  相似文献

Selenium (Se) is essential for human health. Despite evidence that Se intake affects inflammatory responses, the mechanisms by which Se and the selenoproteins modulate inflammatory signalling, especially in the gut, are not yet defined. The aim of this work was to assess effects of altered Se supply and knock-down of individual selenoproteins on NF-κB activation in gut epithelial cells. Caco-2 cells were stably transfected with gene constructs expressing luciferase linked either to three upstream NF-κB response elements and a TATA box or only a TATA box. TNFα and flagellin activated NF-κB-dependent luciferase activity and increased IL-8 expression. Se depletion decreased expression of glutathione peroxidase1 (GPX1) and selenoproteins H and W and increased TNFα-stimulated luciferase activity, endogenous IL-8 expression and reactive oxygen species (ROS) production. These effects were not mimicked by independent knock-down of either GPX1, selenoprotein H or W; indeed, GPX1 knock-down lowered TNFα-induced NF-κB activation and did not affect ROS levels. GPX4 knock-down decreased NF-κB activation by flagellin but not by TNFα. We hypothesise that Se depletion alters the pattern of expression of multiple selenoproteins that in turn increases ROS and modulates NF-κB activation in epithelial cells, but that the effect of GPX1 knock-down is ROS-independent.  相似文献

Grassland canopy management (spring burn, mowing and residue removal in late-summer, or no management) and native tallgrass species composition (cool season mixture, warm season mixture, or combined cool and warm mixture) effects on C and N in aboveground biomass and soil were investigated at Brookings SD on a previously-plowed Barnes clay loam (fine-loamy, superactive, frigid Calcic Hapludoll). During the last 2 yr of the 9-yr experiment, shoot biomass was affected by canopy management with the burn (2,730 kg ha?1) and mow (3,421 kg ha?1) treatments containing less than no management (4,655 kg ha?1). Burn treatment biomass contained 1,189 kg ha?1 and 25 kg ha?1 of C and N, mow contained 1,433 kg ha?1 and 33 kg ha?1 of C and N, while no management contained 2,014 kg ha?1 and 39 kg ha?1 of C and N, respectively. Soil C accumulation was independent of grass species composition. Soil C accumulation rates, which increased in strong linear fashion (r 2 of 0.89 to 0.92) after initial grass establishment, were 387 kg C ha?1 yr?1, 503 kg C ha?1 yr?1, and 711 kg C ha?1 yr?1 for burn, mow, and no management treatments, respectively. Thus, grassland management methods used after conversion of cropland to grassland have important effects on grass biomass and soil C accumulation.  相似文献

Long-term sulfate, selenate and molybdate accumulation and translocation were investigated in two ecotypes of Stanleya pinnata and non-hyperaccumulator Brassica juncea under different levels of applied sulfate and selenate. Morphological differences were observed between the ecotypes of S. pinnata, but few differences in selenium (Se) and sulfur (S) accumulation were measured. Se-to-S ratios were nearly identical between the ecotypes under all treatments. When compared with B. juncea, several unique trends were observed in the hyperaccumulators. While both S. pinnata ecotypes showed no significant effect on Se content of young leaves when the supplied sulfate in the growth medium was increased tenfold (from 0.5 to 5 mM), the Se levels in B. juncea decreased 4- to 12-fold with increased sulfate in the growth medium. Furthermore, S. pinnata’s S levels decreased slightly with high levels of supplied Se, suggesting competitive inhibition of uptake, while B. juncea showed higher S levels with increasing Se, possibly due to up-regulation of sulfate transporters. Both ecotypes of S. pinnata showed much larger Se concentrations in young leaves, while B. juncea showed slightly higher levels of Se in older leaves relative to young. Molybdenum (Mo) levels significantly decreased in S. pinnata with increasing sulfate and selenate in the medium; B. juncea did not show the same trends. These findings support the hypothesis that S. pinnata contains a modified sulfate transporter with a higher specificity for selenate.  相似文献

The aim of this double-blind, placebo controlled clinical trial was to assess the effects of a combination of selenium and silymarin in men with lower urinary tract symptoms, benign prostatic hyperplasia and a prostate specific antigen (PSA) ≤2.5 ng/ml. The volunteers were randomized to two groups: the first one (n = 26) received 240 μg selenium (in the form of yeast l-selenomethionine) plus 570 mg silymarin daily for 6 months and the second (n = 29) received placebo. Outcome measures were changes in the International Prostate Symptom Score (IPSS), bladder volume (V), urinary flow rate, ultrasound estimated postvoid residual urine volume (RV), serum PSA, testosterone and selenium levels, safety clinical biochemistry, hematology and oxidative stress parameters at baseline and on day 180. The results showed statistically significant differences (p < 0.05) between treatment and control groups for the following parameters: IPSS score, urodynamic parameters: maximal rate of urine flow (Qmax), average flow (Qave), V and RV, total PSA value and serum selenium levels. There was a significant reduction in PSA in the selenium–silymarin group but no effect on blood testosterone level. Overall the treatment was well-tolerated with no adverse effects.  相似文献

We aimed to verify the levels of IGFBP2 and SOCS3 in cartilage and chondrocytes of Kashin–Beck disease (KBD) patients and the effects of different selenium concentrations on the protein expression levels. Chondrocytes were cultured with sodium selenite in vitro. Immunohistochemistry and western blotting were used to verify the protein expressions. IGFBP2 and SOCS3 were up-regulated in KBD chondrocytes and decreased with increasing selenium concentrations. IGFBP2 expressed highest in the middle zone of KBD cartilage, SOCS3 expressed higher in the middle and deep zone. IGFBP2 and SOCS3 may be the biomarkers for KBD diagnosis and evaluating the effect of selenium supplement.  相似文献

Previous studies have shown the pathogenic effects of grains cultivated in the endemic areas of Keshan disease and selenium is effective in the prevention of this disease. In this study, liver damages induced by feeding grains from an endemic area (endemic diet), and the effects of selenium and -tocopherol supplement were examined. After 3 months on the endemic diet, the amounts of serum enzymes were significantly increased when compared to controls (animals receiving diet from a non-endemic area). Liver enzymes (alkaline phosphatase and choline esterase) were also found to be altered in the serum, further suggesting liver damages in animals on an endemic diet. Supplement of the endemic diet with selenium or -tocopherol reversed the changes in serum enzymes. Increase in lipid peroxidation in the liver of animals on the endemic diet was observed when compared to that in control animals. Selenium and -tocopherol supplements prevented the increase in lipid peroxidation in the liver by the endemic diet. Semi-quantitative histochemical analysis of glutamate dehydrogenase and succinate dehydrogenase in liver tissue showed that the livers of animals on an endemic diet were more sensitive to ischemic damagesin vitro. Supplementation of the endemic diet with either selenium or -tocopherol reduced the sensitivity to ischemic damages. The results suggest that increased lipid peroxidation in the liver of rats on an endemic diet may be responsible for liver damages and elevation of serum enzymes. Restoration of glutathione peroxidase activity by selenium supplement or an increase in the content of -tocopherol in the liver can prevent lipid peroxidation in animals on an endemic diet and thus provide the protective effects against liver damages.  相似文献

Low concentrations of selenium (Se) predict mortality and cardiovascular diseases in some populations. The effect of Se on in vivo indicators of oxidative stress and inflammation, two important features of atherosclerosis, in human populations is largely unexplored. This study investigated the longitudinal association between serum selenium (s–Se) and a golden standard indicator of oxidative stress in vivo (8-iso-prostaglandin F, a major F2-isoprostane), an indicator of cyclooxygenase (COX)-mediated inflammation (prostaglandin F), high sensitive C-reactive protein (hsCRP), interleukin-6 (IL-6) and serum amyloid A protein (SAA) in a follow-up study of 27 years. The s–Se was measured in 615 Swedish men at 50 years of age in a health investigation. The status of oxidative stress and inflammation was evaluated in a re-investigation 27 years later by quantification of urinary 8-iso-PGF and 15-keto-dihydro- PGF (a major metabolite of PGF) and serum hsCRP, SAA and IL-6. Men in the highest quartile of s–Se at age 50 had decreased levels of 8-iso-PGF compared to all lower quartiles and decreased levels of PGF compared to all lower quartiles at follow-up. These associations were independent of BMI, diabetes, hyperlipidemia, hypertension, smoking, α-tocopherol and β-carotene at baseline. The s–Se was not associated with hsCRP, SAA or IL-6 at follow-up. In conclusion, high concentrations of s–Se predict reduced levels of oxidative stress and subclinical COX-mediated (but not cytokine-mediated) inflammation in a male population. The associations between Se, oxidative stress and inflammation, respectively, might be related to the proposed cardiovascular protective property of Se.  相似文献

Biological volatilization of selenium (Se) in contaminated areas represents an environmentally friendly phytoremediation approach. Implementation of phytovolatilization technology for the remediation of Se-contaminated soils or sediments is oftentimes limited by its low remediation efficiency under field conditions. This greenhouse study determined the feasibility of manipulating soil organic content and hydraulic conditions in a soil–pickleweed (Salicornia bigelovii) system for the enhancement of Se volatilization. Based on annual shoot biomass production rate under field conditions (approximately 1.5 kg m−2), the addition of pickleweed shoot tissues to the soil surface resulted in 2.2-fold more biogenic volatile Se than the control, up to 251.6 ± 140.5 μg m−2 d−1. Selenium volatilization was significantly reduced at a soil water potential of −25 kPa, but substantially increased after re-irrigation to 0 kPa. In a 42-day experiment, the rate of Se volatilization was significantly correlated with soil water potential (P < 0.0001). Findings from this study demonstrate that Se volatilization be substantially enhanced by amending soil with pickleweed residues and by creating wetting and drying cycles that can be monitored with soil water potential probes in the field.  相似文献

Chagas disease, a neglected tropical disease discovered over 100 years ago, is caused by the intracellular parasite Trypanosoma cruzi and is most frequently associated with chronic cardiomyopathy and digestive disorders. Initial invasion of cells is followed by progressive inflammatory destruction of heart, muscles, nerves, and gastrointestinal (GI) tract tissue. Approximately 30% of patients progress to a chronic cardiomyopathy associated with increased morbidity and mortality. Seven to 10% of patients develop megasyndromes involving the GI tract, in particular, the esophagus and the colon. Results from several studies suggest that selenium (Se) deficiency could be an important factor in the pathogenesis of Chagas disease. In this opinion article, Se supplementation is proposed as an adjuvant therapy for treatment of chronic Chagas disease.  相似文献
