Abstract

The effect of sugars on growth of isolated cotyledons from germinating castor bean seeds. — The presence of sugars in the medium induces a number of biochemical and physiological changes in the cotyledons isolated from germinating castor bean seeds. In the present research, the extent and the mechanism of the effect of sugars on growth of the isolated cotyledons was investigated.

The results achieved may be summarized as follows:

  • Sucrose markedly stimulates the growth (as increase of fresh weight) of the isolated cotyledons; its action is already apparent when using a 10-2M concentration and reaches a maximum (stimulation by about 400%) for the concentration between 5.10-2M and 1,5.10-3M. The increase of sucrose induced growth is due in almost equal proportions to water uptake and to increase of dry weight.

  • The difference (Δ P.O.) between the osmotic pressure (P.O.) of the cell sap and that of the external medium is markedly higher for the cotyledons treated with sucrose (7–8 atm.) than the Δ P.O. of cotyledons incubated in water.

  • Analyses of cell contents in sugars show that sucrose is taken up by the cotyledon cells against a concentration gradient. The increase of the difference of osmotic pressure between cotyledon and the external medium is satisfactorily accounted for by this active accumulation of sugars.

  • When mannitol is added to the incubating medium in addition to sucrose, the active uptake of sugar is not disturbed; but the effect of sucrose on growth decreases, and it completly disappears when mannitol concentration in the medium is such, to make the value of Δ P.O. for the cotyledons in sucrose plus mannitol equal to the Δ P.O. for the cotyledons incubated in water.

  • Besides increasing the P.O. in the cotyledons, sucrose markedly accelerates the decrease of free aminoacid in the isolated cotyledons.

  • Auxin (β—indolacetic acid) does not stimulate, or stimulates weakly, growth of the cotyledon incubated in water; some stimulation can be observed only when sucrose is present. Gibberellic acid stimulates growth (though to a much lower extent than sucrose) in the cotyledons in water, while its action does no longer appear when sugar is present.

The decrease of free aminoacid together with other observations, indicates an effect of sucrose on protein metabolism; however, growth stimulation by sucrose seems to depend essentially on its active accumulation - against a concentration gradient - and thuson the increase of osmotic pressure. It is thus suggested that in these tissues osmotic pressure is the most important factor in limiting growth by cell extension.  相似文献

The Author studies the action of the presence of the ear and its parts in the growth of the stem and in the value of the water of traspiration and of assimilation in the higher leaf. The stet shows:

  1. A removed ear, not yet mature, exerts a depressing effect upon the development of the stem and ucon the leaf's functions.

  2. The threads and the pistils removed inhibit the enlongament of the stem and alter the functions of leaves.

  3. Maturing seeds removed cause trubles on the function of the vegetative organ.

Abstract

ACTION OF LIGHT ON SEEDLING GROWTH IN A YELLOW-GREEN MUTANT OF TRITICUM DURUM DESF. — Dry grains of Triticum durum cv. « Cappelli » and its radiation-induced yellow-green mutant were sown in sand in four environment, different for light conditions (daylight, artificial light, ultraviolet rays, darkness), having all the same temperature (23°C.). Seven days after sowing, the following data were registered: a) growth (in cm.) of the shoot and of the longest root; b) fresh weight (in gr.) of shoots and roots; c) water content of shoots and roots.

The following main results were obtained:
  1. the yellow-green mutant always shows less growth and less weight increase than normal « Cappelli », with the exception of the seedlings grown at daylight, which have shorter shoots and longer roots than the normal « Cappelli » but the same weight:

  2. artificial light, besides depressing the growth of the roots of the yellow-green mutant, which becomes green under these conditions, induces a remarkable decrease in the water content of the roots of the two wheats.

Summary

The Author studies with the weighing method the action of plant-eater insects on the alteration of the substances fresch and dry in the hust leaves and he comes to the following conclusions:

  1. The fresh substance stand less alterations in the leaves with the excavated border than in the sound leaves, the dry substance is less in the hust leaves than in those sound.

  2. The fresh substance shows little waverigs in the leaves notched on alone side of the border, but the dry substance is always inferior in the hust leaves than in the sound half and in those entire.

  3. The substances fresh and dry are less in the bored leaves than in the sound, the dry substance shows little alternations in those bored in one or the other half of the border.

This would indicate that the traumatic action of insects causes in the leaves a depresson of the cellular swuelling, whih determines a bigger mobilization of the dry substanc's elements than in the sound leaves.  相似文献

Abstract

The Influence of Short Periods of Lighting with Different Wavelengths on the Orcadian Changes of the Enzymatic Digestive Activities in Palaemon serratus (Crustacea, Natantia)

The effect of light on the circadian rhythms of digestive activities in the Shrimp Palaemon serratus have been studied.

  1. Continuous darkness leads in a few days to a disappearance of the variations of the circadian rhythms of digestive enzymes while these rhythms go on in continuous light.

  2. Short (1 or 2 hrs) and low intensity flashes of white light are effective in bringing on the reappearance of rhythmic variations in darkness.

  3. We have been able to establish an isoquantic spectrum of action of the light. Two values of wavelength appears to account for a maximum sensibility of the shrimp: one in ultraviolet light and an other one, more important, in the green (λ=544 nm).

  4. In green light it is possible to obtain the same effect of light by decreasing the time of stimulation to 5 or 10 mn and in increasing the total quantity of energy. Significant responses are obtained with total energy greater than 10000 pE. cm‐2.

Abstract

Preliminary observations on the enzymatic degradation of RNA in castor bean seeds. — Cocucci, Maggio, Monroy and Marrè have shown the decrease of RNA content during ripening in castor bean seeds, and its increase during germination. Furthermore, these Authors have demonstrated that in the dry ripe seeds the ribosomes are undetectable, and that they increase rapidly during germination. Two peaks of ribosomes are easily detected upon ultracentrifugal analysis in germinating seeds (Cocucci and Sturani). These observations were the basis for our investigations of the enzymes of RNA metabolism in castor bean seeds. This paper deals with our preliminary observations on RNA degrading enzymes in these tissues. We have been able to measure RNase activity, phosphodiesterase, 3′-,5′- and 2′-nucleotidases in castor bean seeds at different stages of development. RNase activity (measured in crude extracts) changes little during the ripening process, its rate corresponding to 40–50 μMoles of nucleotides liberated from RNA per hour and per gram of fresh weight. In the dry seeds, RNase activity is 30–40 μMoles of nucleotides/h.g.f.w., and it increases to about 60–70 μMoles/h/g.f.w. after 72 hours of germination.

Phosphodiesterase activity is about 4–5 μMoles/h.g.f.w.

The following rates have been found in seeds almost completely ripe seeds for 3′-, 5′- and 2′-nucleotidase activities, respectively 45–50 μMoles/h.g.f.w.; 6–7 μMoles/h.g.f.w.; 8 μMoles/h.g.f.w.; ATP-ase activity was of about 80–100 μMoles of phosphate liberated /h.g.f.w. - The high activity of 3′-nucleotidase, of the same order of that of RNase, suggests that these two enzymes are responsible for degradation of RNA to nucleosides and inorganic phosphate. Further investigations are being carried on to define the biochemical properties of castor bean RN-ase.  相似文献

Abstract

Nucleic Acids in Ripening of Castor Bean Endosperm. - I. Quantitative and Qualitative Changes of total RNA. — The ripe Castor Bean seed endosperm (Ricinus Communis var. Sanguinea) has a very low level of nucleic acids and particularly of ribosomal RNA. However, they rapidly increase during the germination of the seed (S. Cocucci et al., Acc. Naz. Lincei, 38, 545, 1965).

The behaviour of the nucleic acids, obtained by phenol extraction from Castor Bean seed endosperm at different ripening stages, is the following:

  1. From the moment when the developing seed has reached the maximal fresh weight to the moment of its complete ripening, the RNA content decreases from about 700 μg per seed to about 200 μg.

  2. During the same period the DNA level (about 13–15 μg per seed) remains nearly costant.

  3. Fractionation by sucrose density gradient of the phenol extracted RNA (H. L. Sanger and C. A. Knight, Biochem. Biophys. Reserch Commun., 13, 445, 1963) shows that during the ripening of the seed the high molecular weight RNA decreases much more than the low molecular weight RNA.

  4. Radioactive phosphate injected in the seeds is incorporated in the RNA at all the stages. Fractionation by sucrose density giadient shows that the phosphate is essentially incorporated in low molecular weight RNA and there is pratically no labelling of the heavier RNA after quite a long period (2 hr.).

These data suggest that the decrease of the RNA during the ripening of the seed can be due to a fall in the ability to synthesize high molecular RNA.  相似文献

Abstract

The enzymes acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are primary targets in attenuating the symptoms of neurodegenerative diseases. Their inhibition results in elevated concentrations of the neurotransmitter acetylcholine which supports communication among nerve cells. It was previously shown for trans-4/5-arylethenyloxazole compounds to have moderate AChE and BChE inhibitory properties. A preliminary docking study showed that elongating oxazole molecules and adding a new NH group could make them more prone to bind to the active site of both enzymes. Therefore, new trans-amino-4-/5-arylethenyl-oxazoles were designed and synthesised by the Buchwald-Hartwig amination of a previously synthesised trans-chloro-arylethenyloxazole derivative. Additionally, naphthoxazole benzylamine photoproducts were obtained by efficient photochemical electrocyclization reaction. Novel compounds were tested as inhibitors of both AChE and BChE. All of the compounds exhibited binding preference for BChE over AChE, especially for trans-amino-4-/5-arylethenyl-oxazole derivatives which inhibited BChE potently (IC50 in µM range) and AChE poorly (IC50?100?µM). Therefore, due to the selectivity of all of the tested compounds for binding to BChE, these compounds could be applied for further development of cholinesterase selective inhibitors.

Ecological risk actually refers to two separate things. First, risk to the environment as a result of human activity. Contaminated sites are an example. Second, risk to the biota—flora, fauna, and people—as a result of environmental hazards. Geophysical risk arising from natural hazards is an example. Risk is a combination of likelihoods and consequences. This article examines methods used to quantify the consequences. At the general level, such methods are linked to the methods used to quantify the likelihoods and thus to quantify the risks. It is possible to use the existing frameworks of risk management, health risk assessment, and ecological risk analysis to develop a risk management framework that is suitable for ecological risk assessment. The framework consists of the following steps:
  1. Determine concernsby using risk assessment techniques for various scenarios.

  2. Identify the consequences by systematically identifying hazards.

  3. Undertake calculations by using relevant models.

  4. Evaluate certainties, uncertainties, and probabilities involved in the calculations of the vulnerability and of the exposure.

  5. Compare with criteriato assess the need for further action.

  6. Determine and act on options to control, mitigate, and adapt to the risk.

  7. Communicatethe results to those who need to know.

Abstract

Select LAB, including Lactobacillus fermentum TISTR 950, Lactobacillus plantarum TISTR 2265 and Lactobacillus casei TISTR 1500 were investigated for their ability to enhance GABA, TPC and the antioxidant activity of perilla seed juice. L. casei TISTR 1500 produced higher GABA and TPC contents and presented higher antioxidant activity than other strains. Furthermore, the optimal fermentation condition to perilla seeds inoculated with L. casei TISTR 1500 to improve the GABA, TPC and antioxidant activity was performed using 33 full factorial design. The final optimal values for perilla fermentation was found at fermentation time of 4.82 days (4 days 19?h 40?min), initial substrate of 5% (w/v) and fermentation temperature of 30.07?°C. Under the optimal fermentation condition, an observed values of GABA, TPC, ABTS, DPPH and FRAP were 71.46 µg/g, 3175.00 µg GAE/g, 1991.40 µg TEAC/g, 9178.29 µg TEAC/g and 7753.34 µg TEAC/g, respectively, which was 3.3, 0.9, 2.9, 10.8 and 10.2 times higher than that of unfermented perilla seeds, and 2.1, 0.8, 0.9, 10 and 9.2 times of fermented perilla seeds before the optimization. These results may provide the foundation to further target in industrial application for the production of plant-based and develop functional perilla seed products containing GABA.

Abstract

This study evaluated remediation of the herbicide sulfentrazone in soils with three different mineralogies (kaolinite, hematite, and gibbsite) and three remediation sulfentrazone treatments (Canavalia ensiformis L., Crotalaria juncea L., and natural attenuation). This study was conducted in a factorial scheme, in triplicate with randomized block design. Sulfentrazone was applied at 0 and 400?g ha?1. We analyzed sulfentrazone residue in the soils by high-performance liquid chromatography and confirmed the results with bioassays of Pennisetum glaucum. Herbicide movement was greater in the kaolinitic soil without plant species. The retention of herbicide in the kaolinitic soil occurred in larger quantities in the 0–12?cm layer, with higher levels found in the treatments with plants. In the hematitic soil with C. juncea, all applied herbicides were concentrated in the 0–12?cm layer. In the other hematitic soil treatments, sulfentrazone was not detected by chemical analysis at any soil depth, although in many treatments, it was detected in the bioassay. Phytoremediation was more efficient with C. ensiformis grown in gibbsitic soil, reducing the sulfentrazone load by approximately 27%. Natural attenuation was more efficient than phytoremediation in oxidic soils due to soil pH and texture soils favored microbial degradation of the compound.

Summary

In order to study a side of the influence of ultra-violet light on plants, the A. has harvested, 10 days after fertilisation, 1040 plants of Triticum vulgare Vill. and made many groups of them; then he sheared differently the ears and screened them partially from light so as to obtain, from the same subject, treated and untreated grains. Such ears he treated for different times of exposure with the light of a quarzlamp giving almost only ultra-violet rays. For the present, are here reported only results concerning the influence of ultra-violet light upon both germinating energy and power:

  • ultra-violet light stimulates the germinating power of grains, born from subjects treated no longer than 240 hours at a distance of 1 m from the lamp; longer treatments abate germinating power;

  • ultra-violet light stimulates, the germinating energy of grains born from subjects treated no longer than 120 hours; treatments during 120–240 hours don't notably modify it; treatments of over 240 hours reduce it very much;

  • according to the conditions in which this work has been done, a 10 days-irradiation seems to be, under every point of view, for the best. A 71/2 — days irradiation seems to be insufficient, a 121/2 — days one excessive, and hence prejudicial;

  • glumes are strong screening-organ for ultra-violet light;

  • ultra-violet light gives late appearing effects.

Since great difficulties usually occur whenever tests such as the present one are undertaken, the A. proposes some leading ways to previously settle the U.V.-rays dosimetry fit for every plant, in order to avoid wast of time and to get data of general value.  相似文献

The following procedure for staining Negri bodies in sections is based on methods previously described by MacNeal, by Haynes, and by Richter:

Fixation:
  1. 1. Zenker's solution 4 hours at 37°C or Dominici's 3 hours.

  2. 2. 70% alcohol, 12 to 18 hours at room temperature.

  3. 3. 80% alcohol, about 5 to 6 hours.

  4. 4. 90% alcohol, about 4 to 6 hours.

  5. 5. Absolute alcohol about 16 hours.

  6. 6. Ether and absolute alcohol aa, about 8 hours.

  7. 7. 16 to 24 hours in the following mixture: celloidin 1 g., methyl salycilate 25 cc., abs. alcohol 25 cc., ether 25 cc.

  8. 8. Chloroform and paraffin, 2 to 3 hours.

  9. 10. Paraffin, 1 to 1 1/2 hours.

  10. 11. Embed.

staining:
  1. 1. Cut sections 4 to 5 μ.

  2. 2. Bring section to water and cover with Lugol's iodine for 10 minutes.

  3. 3. Decolorize with a 2% sodium thiosulfate (hypo).

  4. 4. Wash thoroly with water.

  5. 5. Cover with a mixture of equal parts of 0.5% phloxine and 1% eosin Y (National Aniline brand) and leave for 15 minutes.

  6. 6. Wash with water and stain 2 to 5 minutes in 0.1% azure B (National Aniline).

  7. 7. Wash with 96% alcohol and decolorize in a mixture of 2 parts absolute alcohol with 1 part clove oil, ordinarily for not more than 1/2 to 1 minute.

  8. 8. Dehydrate rapidly, clear, and mount in Yucatan Elemi.

Gallic acid acts as a precursor molecule to synthesize various tannin molecules. These are plant polyphenols and were proved to be good anti-oxidant, anti-cancerous, anti-inflammatory, anti-microbial compounds. In order to fully exploit prominent biological activities of specific tannins and to develop tannin-based new medicines, it is necessary to obtain their pure preparations with an aim of high yield and specificity. In the present study, gallic acid is synthesized by the hydrolysis of tannic acid using a microbial based transformation process. The microorganism was isolated and identified. The ability of the isolated microorganism to covert tannic acid into gallic acid was determined by HPLC and enzyme production.
