Tropomyosin represents a major allergen of decapod crustaceans such as shrimps and crabs, and its highly conserved amino acid sequence (>90% identity) is a molecular basis of the immunoglobulin E (IgE) cross-reactivity among decapods. At present, however, little information is available about allergens in edible crustaceans other than decapods. In this study, the major allergen in two species of edible crustaceans, Antarctic krill Euphausia superba and mantis shrimp Oratosquilla oratoria that are taxonomically distinct from decapods, was demonstrated to be tropomyosin by IgE-immunoblotting using patient sera. The cross-reactivity of the tropomyosins from both species with decapod tropomyosins was also confirmed by inhibition IgE immunoblotting. Sequences of the tropomyosins from both species were determined by complementary deoxyribonucleic acid cloning. The mantis shrimp tropomyosin has high sequence identity (>90% identity) with decapod tropomyosins, especially with fast-type tropomyosins. On the other hand, the Antarctic krill tropomyosin is characterized by diverse alterations in region 13–42, the amino acid sequence of which is highly conserved for decapod tropomyosins, and hence, it shares somewhat lower sequence identity (82.4–89.8% identity) with decapod tropomyosins than the mantis shrimp tropomyosin. Quantification by enzyme-linked immunosorbent assay revealed that Antarctic krill contains tropomyosin at almost the same level as decapods, suggesting that its allergenicity is equivalent to decapods. However, mantis shrimp was assumed to be substantially not allergenic because of the extremely low content of tropomyosin.  相似文献

Decapod crustaceans are relatively widespread in Jurassic deposits of Europe and a small number have been collected from the north-east of France. A new fossil of a macrurous decapod assigned to Eryma burgundiaca sp. nov., an erymid lobster, is described from this area and comparisons are made with closely allied species from Europe. Probable pagurid anomurous decapods are represented by Orhomalus magnificus sp. nov., Palaeopagurus acutus sp. nov. and Palaeopagurus neraudeaui sp. nov. No brachyurous decapods are presently known from the fauna. Based on the associated ammonites, the material is considered to be Callovian in age.  相似文献

The acorn barnacles (Cirripedia, Thoracica, Balanomorpha) are a diverse group of crustaceans found in virtually all marine and estuarine habitats. Barnacles are important model species in various biological researches, including evolution, intertidal ecology, larval biology and antifouling. However, there remains a lack of a thorough understanding of the phylogeny for this group of animals, particularly at higher taxonomic levels. In this study, we attempt to determine the phylogenetic relationships among balanomorphan families based on analysis of complete mitochondrial genome from various barnacle families and investigate the evolution of mitogenome in barnacles. Whole mitogenomes of six barnacles were newly sequenced, including Acasta sulcata (Archaeobalanidae), Armatobalanus allium (Archaeobalanidae), Chelonibia testudinaria (Chelonibiidae), Octomeris sp. (Chthamalidae), Savignium biporata (Pyrgomatidae) and Tetraclitella divisa (Tetraclitidae), which exhibit five different gene arrangements. Phylogenetic analysis on 15 complete mitochondrial genome sequences from major barnacle families supported Chthamalidae, Pyrgomatidae and Tetraclitidae formed monophyletic units, but suggested polyphyly of both Archaeobalanidae and Balanidae. Chthamalidae was the earliest diverged lineage in Balanomorpha. Chelonibiidae + Tetraclitidae formed the sister taxon to the monophyletic superfamily Balanoidea (Archaeobalanidae + Balanidae + Pyrgomatidae). The members of Archaeobalanidae and Balanidae intermingled in the inferred topology with the monophyletic Pyrgomatidae deeply nested within. Two Megabalanus species from the family Balanidae and A. sulcata from the family Archaeobalanidae share the same six‐gene‐cluster inversion as compared to the other ten balanomorphan barnacles, providing further evidence for the non‐monophyly for the two families. We showed here that the informativeness of the complete mitogenome sequence and rare genomic events in resolving various questions concerning Balanomorpha relationships. The non‐monophyletic status of Archaeobalanidae and Balanidae falsified many previous hypotheses concerning the complex evolution of Balanomorpha and call for further investigations and careful revision on the taxonomy of the group. Future study focusing on the enigmatic and tentatively basal lineages, for example, Chionelasmatoidea Pachylasmatoidea and Catophragmidae, would be most helpful in fully resolving the phylogeny and mitogenome evolutionary history of acorn barnacles.  相似文献

The ingestion or inhalation of some proteins may lead to adverse immune reactions. Allergens may trigger allergic reactions in genetically predisposed individuals when they are absorbed through the skin or make contact with mucous membranes. An allergic disease often deteriorates the quality of life and may sometimes be life-threatening due to anaphylactic shock. A number of allergens have been characterized from various multicellular organisms to date. It is thought to be reasonable to pay a special attention to the substance which is highly cross-reactive and which causes adverse responses in the molecules that are not sensitized but similar to the sensitized allergen. Tropomyosin has been described as an important food allergen in shrimp, lobster, crab, oysters, squid, and other invertebrates. Allergic reactions to shellfish and mollusks are often cross-reactive, which may be explained by the highly conserved amino acid sequences of tropomyosins among invertebrates, but vertebrate tropomyosins are not known to be allergenic. Several tropomyosins from domestic arthropods have been reported to be allergenic. Recently, it was suggested that an infection of helminthic parasites might lead to sensitization to tropomyosin and elicit allergic reactions to other invertebrates. Much effort has been made to characterize these allergenic tropomyosins from various sources. We will discuss the physicochemical characteristics and the potential application of tropomyosin for the diagnosis and therapeutics of allergic disorders.  相似文献

Decapod crustaceans synthesize highly active proteolytic enzymes in the midgut gland and release at least a part of them into the stomach where they facilitate the first step in peptide hydrolysis. The most common proteinases in the gastric fluid characterized so far are serine proteinases, that is, trypsin and chymotrypsin. These enzymes show highest activities at neutral or slightly alkaline conditions. The presence of acid proteinases, as they prevail in vertebrates, has been discussed contradictorily yet in invertebrates. In this study, we show that acid aspartic proteinases appear in the gastric fluid of several decapods. Lobsters Homarus gammarus showed the highest activity with a maximum at pH 3. These activities were almost entirely inhibited by pepstatin A, which indicates a high share of aspartic proteinases. In other species (Panulirus interruptus, Cancer pagurus, Callinectes arcuatus and Callinectes bellicosus), proteolytic activities were present at acid conditions but were distinctly lower than in H. gammarus. Zymograms at pH 3 showed in each of the studied species at least one, but mostly two-four bands of activity. The apparent molecular weight of the enzymes ranged from 17.8 to 38.6 kDa. Two distinct bands were identified which were inhibited by pepstatin A. Acid aspartic proteinases may play an important role in the process of extracellular digestion in decapod crustaceans. Activities were significantly higher in clawed lobster than in spiny lobster and three species of brachyurans. Therefore, it may be suggested that the expression of acid proteinases is favored in certain groups and reduced in others.  相似文献

The origin and early diversification of decapod crustaceans and their expansion from marine to continental environments are key events in arthropod evolution. Rare fossil decapods are known from the Palaeozoic, and the earliest eumalacostracans with undoubted decapod affinities are the Late Devonian Palaeopalaemon and Aciculopoda, found in offshore marine deposits. Here, we describe a new species of the shrimp Tealliocaris found in floodplain and temporary pond deposits from the Famennian (Late Devonian) of Belgium, together with a rare Palaeozoic assemblage of other crustaceans (conchostracans, notostracans and anostracans) and chelicerates (eurypterids). Tealliocaris walloniensis sp. nov. documents the earliest occurrence of continental decapod crustaceans and indicates that decapods have been part of continental ecosystems at least since the Late Devonian.  相似文献

Exposure to wave action and other environmental factors can alter the morphology of intertidal barnacles. We tested several hypotheses on the causes of morphological variation in the cirri and penises of the barnacle Tetraclita stalactifera at sites differing in wave exposure, at different heights in the intertidal zone, and at different levels of population density. Unlike many other acorn barnacle species, cirrus and penis characteristics did not correspond to differences in wave exposure or crowding. However, barnacles from higher tidal elevations had thicker cirri and thicker penises than those from lower elevations. Because of reduced time submerged at higher elevations, increased thickness may be a means of compensating for reduced feeding and mating opportunity by allowing for continued feeding and mating attempts during periods of greater wave action. Our observations of differences in cirrus and penis morphology suggest that phenotypic plasticity in penis and cirrus characteristics are adaptations shared by the species T. stalactifera and other acorn barnacles, but that T. stalactifera responds differently to environmental stimuli than do other species.  相似文献

Wave-exposure influences the form of many organisms. Curiously, the impact of flow extremes on feeding structures has received little attention. Barnacles extend feather-like legs to feed, and prior work revealed a highly precise association between leg length and water velocity in one species. To assess the generality of this flow-dependence, we quantified variation in four leg traits (ramus length, ramus diameter, seta length, and intersetal spacing) in four intertidal barnacles (Balanus glandula, Chthamalus dalli, Semibalanus cariosus, Pollicipes polymerus) over a wave-exposure gradient in the North-Eastern Pacific. All species exhibited a negative allometric relation between leg length and body mass. Proportionally longer feeding legs may permit smaller barnacles to avoid lower flow and particle flux associated with boundary layers. Although coefficients of allometry did not vary with wave-exposure, form differences among wave-exposures were substantial. Depending on the species, acorn barnacles of the same size from protected shores had feeding legs that were 37-80% longer and 18-25% thinner, and setae that were 36-50% longer and up to 25% more closely spaced, than those from exposed shores. Differences were less pronounced for the gooseneck barnacle, P. polymerus. Moreover, in situ water velocity explained an impressive percentage of overall leg-length variation: 92% in B. glandula, 67% in C. dalli, 91% in S. cariosus, and 92% in P. polymerus. Clearly, both size and shape of barnacle feeding legs respond to local flow conditions. This response appears widespread--across two orders of thoracican barnacles, Pedunculata and Sessilia, and two superfamilies of acorn barnacles (Balanoidea and Chthamaloidea)--and likely adaptive. Longer rami and setae would yield a larger feeding area in low flow, whereas shorter, stouter rami with shorter setae would be less vulnerable to damage in high flow. Finally, the proportionally most variable species was abundant in the widest range of habitats, suggesting that increased plasticity may permit a wider niche breadth.  相似文献

Several different clones which contain sequences complementary to the mRNA encoding tropomyosin were isolated from cDNA libraries prepared from locust RNA. Based on the sequence analysis of available clones, the complete primary structure of the locust tropomyosin was explored. The deduced protein sequence showed a repeating pattern of amino acid residues characteristic of a coiled-coil structure. The amino acid sequence of locust tropomyosin contains domains of complete homology but also regions of pronounced variability when compared with tropomyosins of other species. Northern blot as well as Western blot analysis revealed that different forms of tropomyosins are expressed in locust muscles.  相似文献

Tropomyosin is an alpha-helical coiled-coil protein with structural similarities to the streptococcal M protein. In order to show serologic cross-reactivity between streptococcal M proteins and tropomyosin, we selected from a panel of murine mAb those which reacted with M proteins and tropomyosins in the ELISA. Western blots were used to study the reactions of each mAb with human and rabbit cardiac and rabbit skeletal tropomyosins. The antibodies were further characterized for their reactions with the additional autoantigens myosin, actin, keratin, and DNA. Five mAb were found which reacted with either PepM5 or ColiM6 protein and tropomyosin in Western blots or ELISA. Two of the tropomyosin positive mAb were also antinuclear antibodies and were inhibited with DNA. In Western blots of cardiac tropomyosins, the mAb reacted with either the 70-kDa dimer of tropomyosin, the 35-kDa monomer, or both. Some differences were observed in the reactions of the mAb with the different tropomyosins in Western blots. The heart cross-reactive epitopes shared between M proteins and tropomyosin were in most instances shared with cardiac myosin. Differences were observed among the reactions of the mAb with the different tropomyosins. This report constitutes the first evidence of serologic cross-reactivity between streptococcal M proteins and tropomyosins.  相似文献

Tropomyosins can be classified as belonging to an alpha-like or a beta-like family depending on the absence or presence, respectively in their protease-V8 digestion pattern of two peptides with an apparent molecular mass of 21 kDa. Chicken cardiac tropomyosin and the 43 kDa component from gizzard tropomyosin are accordingly classified as alpha-like tropomyosins, while the 33 kDa gizzard tropomyosin component is a beta-like tropomyosin. The 21 kDa peptides have an overall charge which is more positive than that of the intact tropomyosin or any other tropomyosin peptide and probably contain the -NH2 half of the molecule.  相似文献

Neurogenesis persists throughout life in the olfactory pathway of many decapod crustaceans. However, the relationships between precursor cells and the temporal characteristics of mitotic events in these midbrain regions have not been examined. We have conducted studies aimed at characterizing the sequence of proliferative events that leads to the production of new deutocerebral projection neurons in embryos of the American lobster, Homarus americanus. In vivo bromodeoxyuridine (BrdU) labeling patterns show that three distinct cell types are involved in neurogenesis in this region. Quantitative and temporal analyses suggest that the clearing time for BrdU is 2-3 days in lobster embryos, and that the sequence of proliferative events in the midbrain is significantly different from the stereotypical pattern for the generation of neurons in the ventral nerve cord ganglia of insects and crustaceans. The unusual pattern of proliferation in the crustacean midbrain may be related to the persistence of neurogenesis throughout life in these regions.  相似文献

Parasitic dinoflagellates of the genus Hematodinium infect several commercially important decapod crustaceans. Different species of Hematodinium have different levels of virulence in their respective hosts. Enzyme activities were studied from two species of Hematodinium, one isolated from the Norway lobster (Nephrops norvegicus) and the other from the American blue crab (Callinectes sapidus). We report the identification of differences in secretion of acid phosphatase (AP) and leucine arylamidase from two parasite species. Leucine arylamidase was only contained and secreted by the species infecting the blue crab. Both parasite species contained AP, but only the species infecting the Norway lobster secreted this enzyme. In this species, AP activity was predominantly in the soluble fraction (69.5%). AP activity was localized to cytoplasmic granules and on the membranes surrounding the cell nucleus. In addition to providing information on the cellular metabolism of the parasite, the pattern of activities of these enzymes may also be useful in distinguishing among different species of Hematodinium.  相似文献

Barnacles have evolved a number of specialized features peculiar for crustaceans: they produce a calcified, external shell; they exhibit sexual strategies involving dioecy and androdioecy; and some have become internal parasites of other Crustacea. The thoroughly sessile habit of adults also belies the highly mobile and complex nature of their larval stages. Given these and other remarkable innovations in their natural history, it is perhaps not surprising that barnacles present a spectrum of opportunities for study. This symposium integrates research on barnacles in the areas of larval biology, biofouling, reproduction, biogeography, speciation, population genetics, ecological genomics, and phylogenetics. Pioneering comparisons are presented of metamorphosis among barnacles from three major lineages. Biofouling is investigated from the perspectives of biochemical and biomechanical mechanisms. Tradeoffs in reproductive specializations are scrutinized through theoretical modeling and empirical validation. Patterns of endemism and diversity are delineated in Australia and intricate species boundaries in the genus Chthamalus are elucidated for the Indo-Pacific. General methodological concerns with population expansion studies in crustaceans are highlighted using barnacle models. Data from the first, draft barnacle genome are employed to examine location-specific selection. Lastly, barnacle evolution is framed in a deep phylogenetic context and hypothetical origins of defined characters are outlined and tested.  相似文献

In many decapod crustaceans, escape tailflips are triggered by lateral giant (LG) and medial giant (MG) interneurons, which connect to motor giant (MoG) abdominal flexor neurons. Several decapods have lost some or all of these giant neurons, however. Because escape-related giant neurons have not been documented in palinurans, I examined tailflipping and abdominal nerve cords for giant neurons in two scyllarid lobster species, Ibacus peronii and Ibacus alticrenatus. Unlike decapods with giant neurons, Ibacus do not tailflip in response to sudden taps. Ibacus can perform non-giant tailflipping: the frequency of tailflips during swimming is adjusted by altering the gap between each individual tailflip. Abdominal nerve cord sections show no LG or MG interneurons. Backfilling nerve 3 of abdominal ganglia revealed no MoG neurons, and the fast flexor motor neuron population is otherwise identical to that described for crayfish. The loss of giant neurons in Ibacus represents an independent deletion of these cells compared to other reptantian decapods known to have lost these giant neurons. This loss is correlated with the normal posture in scyllarids, in which the last two abdominal segments are flexed, and an alternative defensive strategy, concealment by digging into sand.  相似文献

Tropomyosin was isolated from bovine brain using mild conditions thereby avoiding heat precipitation. Separation by DEAE ion exchange chromatography yielded a 33 kDa tropomyosin and a mixture of 30 and 32 kDa tropomyosin. Binding of the tropomyosins to actin filaments was measured by a newly developed method. The binding was assayed by the retarding effect of tropomyosin on actin polymerization. The 33 kDa tropomyosin was found to bind to actin filaments with considerably higher affinity than the 30 and 32 kDa tropomyosin.  相似文献
