An integrated composting-vermicomposting process has been developed for utilization of primary sewage sludge (PSS). Matured vermicompost was used as bulking material and a source of active microbial culture during aerobic activated composting (AAC). AAC resulted in sufficient enrichment of bulking material with organic matter after 20 cycles of recycling and mixing with PSS and produced materials acceptable for vermicomposting. Vermicomposting caused significant reduction in pH, volatile solids (VS), specific oxygen uptake rate (SOUR), total organic carbon (TOC), C/N ratio and pathogens and substantial increase in electrical conductivity (EC), total nitrogen (TN) and total phosphorous (TP) as compared to compost. Environmental conditions and stocking density have profound effects on vermicomposting. Temperature of 20 °C with high humidity is favorable environmental condition for vermicomposting employing Eisenia fetida. Favorable stocking density range for vermiculture is 0.5-2.0 kg m−2 (optimum: 0.5 kg m−2) and for vermicomposting is 2.0-4.0 kg m−2 (optimum: 3.0 kg m−2), respectively.  相似文献

Determination of thermal properties of composting bulking materials   总被引:2,自引:0,他引:2  
Thermal properties of compost bulking materials affect temperature and biodegradation during the composting process. Well determined thermal properties of compost feedstocks will therefore contribute to practical thermodynamic approaches. Thermal conductivity, thermal diffusivity, and volumetric heat capacity of 12 compost bulking materials were determined in this study. Thermal properties were determined at varying bulk densities (1, 1.3, 1.7, 2.5, and 5 times uncompacted bulk density), particle sizes (ground and bulk), and water contents (0, 20, 50, 80% of water holding capacity and saturated condition). For the water content at 80% of water holding capacity, saw dust, soil compost blend, beef manure, and turkey litter showed the highest thermal conductivity (K) and volumetric heat capacity (C) (K: 0.12–0.81 W/m °C and C: 1.36–4.08 MJ/m3 °C). Silage showed medium values at the same water content (K: 0.09–0.47 W/m °C and C: 0.93–3.09 MJ/m3 °C). Wheat straw, oat straw, soybean straw, cornstalks, alfalfa hay, and wood shavings produced the lowest K and C values (K: 0.03–0.30 W/m °C and C: 0.26–3.45 MJ/m3 °C). Thermal conductivity and volumetric heat capacity showed a linear relationship with moisture content and bulk density, while thermal diffusivity showed a nonlinear relationship. Since the water, air, and solid materials have their own specific thermal property values, thermal properties of compost bulking materials vary with the rate of those three components by changing water content, bulk density, and particle size. The degree of saturation was used to represent the interaction between volumes of water, air, and solids under the various combinations of moisture content, bulk density, and particle size. The first order regression models developed in this paper represent the relationship between degree of saturation and volumetric heat capacity (r = 0.95–0.99) and thermal conductivity (r = 0.84–0.99) well. Improved knowledge of the thermal properties of compost bulking materials can contribute to improved thermodynamic modeling and heat management of composting processes.  相似文献

The aim was to investigate pathogen survival during composting of pig manure solids with and without bulking agents in two trials of 56 days duration, each with four treatments. Salmonella was detected in the sawdust and straw bulking agents but was undetectable in the compost, except in one treatment at day 0. Enteric indicator organisms were reduced by day 7 (P<0.001) and were undetectable in the final compost, except for coliform which were present at 3.66-4.43 log?? CFU/g. Yeasts and moulds were reduced and aerobic spore-formers remained stable in one trial but both increased in the other (P<0.001). Bacillus licheniformis and Clostridium sporogenes were the predominant culturable spore-forming bacteria recovered. Microbial counts were influenced by the bulking agent but only at particular time points (P<0.05). Overall, the pig manure-derived compost complied with EU regulations for processed manure products, as E. coli and Enterococcus were below limits and it was Salmonella-free.  相似文献

Thermal (70–190 °C) and thermo-chemical (pH = 10 and 12, 25 °C and 90–190 °C) treatments were investigated in order to maximise the production of methane from pig manure. Methane production from treated and raw manure was assessed from batch mesophilic biochemical methane potential tests. Methane potential of manure soluble fraction increased with the temperature of thermal treatments whereas temperatures higher than 135 °C were necessary to improve the methane potential of the total fraction. Oxytetracycline (OTC) and chlortetracycline (CTC) are broad-spectrum antibiotics used in livestock production. Although laboratory-scale studies have shown that extractable concentrations of these compounds decrease over time within treated and untreated manures and soils, there is relatively little information from farm-scale experiments. The objective of this study was to determine the effect of different levels of management on manure pile temperature profiles and on the fate of OTC and CTC in manure from therapeutically treated calves. Four treatments were designed to span a range of management options – from simply piling up the manure to amending it with straw to increase aeration and adding insulating layers of straw. Replicate samples of antibiotic-containing calf manure were held at ambient temperature or placed in three locations within replicate 3 m3 piles of beef manure. During the 28-day incubation period, concentrations of buffer-extractable OTC and CTC/ECTC (the summed concentrations of CTC and its epimer 4-epi-chlortetracycline (ECTC)) in manure samples incubated at ambient temperature (11–24 °C) decreased 75% (from 18 to 4.6 mg kg−1 dry weight (DW)) and 90% (from 192 to 16 mg kg−1 DW), respectively. Concentrations of the CTC metabolite iso-chlortetracycline (ICTC) decreased 90% (from 37 to 3 mg kg−1 DW). OTC and CTC/ECTC concentrations in samples incubated for 28 days within a non-amended manure pile decreased 91% and >99%, respectively. During that period, the manure pile temperature ranged from 36 °C to 45 °C. Manure piles insulated with a blanket of straw and/or amended with straw (3:1, v/v) attained temperatures up to 70 °C and contained very low levels of OTC, CTC/ECTC, and ICTC (ranging from <0.1 to 0.4 mg kg−1 DW) after 28 days.  相似文献

Modelling N mineralization from bovine manure and sewage sludge composts   总被引:1,自引:0,他引:1  
Nitrogen mineralization kinetics were compared in three different soils (pH values: 5.2, 7.1 and 8.6) when treated with bovine manure (BM) and sewage sludge (SS) composts. The soil-compost mixtures were kept at a controlled moisture content of 60% of their water holding capacity (WHC) and were incubated in the dark at 25 °C for 2 years. Five mathematical models were compared (simple exponential, double exponential, special model, hyperbolic and parabolic), using as experimental data the mineralized N accumulated during 360 and 720 days of incubation. The results showed that the best fit for describing the mineralization of organic N from the compost after 1 year of experimentation was obtained with the simple exponential model. However, the special model showed the best fit for data from 2 years of incubation and thus better reflected organic N mineralization over a longer time-span. This suggested that the organic N in the two composts was made up of two organic pools of different degrees of stability.  相似文献

Nitrification under changing salinities (0-9%), temperatures (6-50 °C), ammonia (0-5 g N L−1) and nitrite concentrations (0-0.4 g N L−1) was investigated in fixed-bed reactors. For all conditions ammonia oxidation rates (AOR) were lower than nitrite oxidation rates (NOR). AORs and NORs increased from 12.5 to 40 °C and were very low at 6 °C and almost zero at 50 °C. No recovery of nitrification was obtained after incubation at 50 °C, whereas nitrification was restorable after incubation at 6 °C. Ammonia concentrations of 5 g N L−1 or nitrite concentrations up to 0.125 g N L−1 decreased AOR to almost zero. AORs and NORs recovered if ammonia or nitrite was removed. At concentrations of 1 and 5 g N L−1 ammonia AOR and NOR were inhibited by 50%, whereas 27 mg N/L nitrite inhibited AOR by 50%.  相似文献

The effect of C source on N losses by volatilization during composting was measured using four bulking agents, each at three humidity levels and composted in duplicate under passive and active aeration. The bulking agents were pine shavings alone and corrected with soybean, chopped grass hay alone and corrected with urea, long (unchopped) wheat straw and chopped oat straw. The readily available C of each bulking agent was determined by analyzing for BOD5. In 105 l laboratory vessels, the bulking agents were mixed with liquid swine manure and tap water for a C/N of 20 and three humidity levels of 60%, 65% and 70%. While being aerated actively or passively, the mixtures were composted for 21 days. Their initial and final C and N contents were measured to conduct a mass balance analysis and calculate C and N losses. C and N losses were compared to bulking agent BOD5. N losses were compared to C losses. The humidity level and aeration regime had no effect on C and N losses but the N losses were correlated to C losses and only the C losses could be correlated to the BOD5 of the bulking agent. Thus, the N losses are related not only to the availability of C but also to the extent of composting. A relationship established between N and C losses indicated that 85% of the initial total N of the compost was available for microbial degradation and that 70% of the available C was lost as CO2 during the immobilization process.  相似文献

The effect of IIF in Pacific oyster oocytes was studied using cryo and transmission electron microscopy (TEM). The viability of oocytes at each step of a published cryopreservation protocol was assessed in an initial experiment. Two major viability losses were identified; one when oocytes were cooled to −35 °C and the other when oocytes were plunged in liquid nitrogen. Although the cryomicroscope showed no evidence of IIF in oocytes cooled with this protocol, TEM revealed that these oocytes contained ice crystals and were at two developmental stages when frozen, prophase and metaphase I. To reduce IIF, the effect of seven cooling programmes involving cooling to −35 or −60 °C at 0.1 or 0.3 °C min−1 and holding for 0 or 30 min at −35 or −60 °C was evaluated on post-thaw fertilization rate of oocytes. Regardless of the cooling rate or holding time, the fertilization rate of oocytes cooled to −60 °C was significantly lower than that of oocytes cooled to −35 °C. The overall results indicated that observations of IIF obtained from cryomicroscopy are limited to detection of larger amounts of ice within the cells. Although the amount of cellular ice may have been reduced by one of the programmes, fertilization was reduced significantly; suggesting that there is no correlation between the presence of intracellular ice and post-thaw fertilization rate. Therefore, oyster oocytes may be more susceptible to the effect of high solute concentrations and cell shrinkage than intracellular ice under the studied conditions.  相似文献

Cellulosic plant and waste materials are potential resources for fermentative hydrogen production. In this study, hydrogen producing, cellulolytic cultures were enriched from compost material at 52, 60 and 70 °C. Highest cellulose degradation and highest H2 yield were 57% and 1.4 mol-H2 mol-hexose−1 (2.4 mol-H2 mol-hexose-degraded−1), respectively, obtained at 52 °C with the heat-treated (80 °C for 20 min) enrichment culture. Heat-treatments as well as the sequential enrichments decreased the diversity of microbial communities. The enrichments contained mainly bacteria from families Thermoanaerobacteriaceae and Clostridiaceae, from which a bacterium closely related to Thermoanaerobium thermosaccharolyticum was mainly responsible for hydrogen production and bacteria closely related to Clostridium cellulosi and Clostridium stercorarium were responsible for cellulose degradation.  相似文献

The evolution of sporulating bacteria in full-scale composting facilities with online temperature monitoring has been poorly studied, although organic matter recycling increases. We analysed Clostridium perfringens and sulphite-reducing clostridia (SRC) by cultivation, and streptomycetes by real-time PCR in five full-scale, temperature-monitored and aerated composting processes, and two pilot-scale drum composters. Facilities composted woodchips, sawdust, peat, or bark amended sludge or source-separated biowaste. Streptomycetes genes of 0.21-110 × 107 copies/g feed increased fast to 0.019-33 × 109 copies/g, and then were equal or decreased. SRC of 0.06-2.2 × 107 cfu/g feed decreased to 0-600 cfu/g, with re-growth in two facilities. End products were clean of C. perfringens, detected in sludge composts. Although processes contained large quantities of spore-forming bacteria, in the best facilities end products had the high quality. Temperature (>55 °C, >2d) was not related to the end compost quality, but relations between waste and bulking agent qualities, aeration, and processing time should be better controlled.  相似文献

The Greenshell™ mussel (Perna canaliculus) is the main shellfish species farmed in New Zealand. The aim of this study was to evaluate the effects of cryoprotectant concentration, loading and unloading strategy as well as freezing and thawing method in order to develop a protocol for cryopreservation of trochophore larvae (16–20 h old). Toxicity tests showed that levels of 10–15% ethylene glycol (EG) were not toxic to larvae and could be loaded and unloaded in a single step. Through cryopreservation experiments, we designed a cryopreservation protocol that enabled 40–60% of trochophores to develop to D-larvae when normalized to controls. The protocol involved: holding at 0 °C for 5 min, then cooling at 1 °C min−1 to −10 °C, holding for a further 5 min, then cooling at 0.5 °C min−1 to −35 °C followed by a 5 min hold and then plunging into liquid nitrogen. A final larval rearing experiment of 18 days was conducted to assess the ability of these frozen larvae to develop further. Results showed that only 2.8% of the frozen trochophores were able to develop to competent pediveligers.  相似文献

Long-term manure application is recognized as an efficient management practice to enhance soil organic carbon (SOC) accumulation and nitrogen (N) mineralization capacity. A field study was established in 1979 to understand the impact of long-term manure and/or chemical fertilizer application on soil fertility in a continuous maize cropping system. Soil samples were collected from field plots in 2012 from 9 fertilization treatments (M0CK, M0N, M0NPK, M30CK, M30N, M30NPK, M60CK, M60N, and M60NPK) where M0, M30, and M60 refer to manure applied at rates of 0, 30, and 60 t ha−1 yr−1, respectively; CK indicates no fertilizer; N and NPK refer to chemical fertilizer in the forms of either N or N plus phosphorus (P) and potassium (K). Soils were separated into three particle-size fractions (2000–250, 250–53, and <53 μm) by dry- and wet-sieving. A laboratory incubation study of these separated particle-size fractions was used to evaluate the effect of long-term manure, in combination with/without chemical fertilization application, on the accumulation and mineralization of SOC and total N in each fraction. Results showed that long-term manure application significantly increased SOC and total N content and enhanced C and N mineralization in the three particle-size fractions. The content of SOC and total N followed the order 2000–250 μm > 250–53μm > 53 μm fraction, whereas the amount of C and N mineralization followed the reverse order. In the <53 μm fraction, the M60NPK treatment significantly increased the amount of C and N mineralized (7.0 and 10.1 times, respectively) compared to the M0CK treatment. Long-term manure application, especially when combined with chemical fertilizers, resulted in increased soil microbial biomass C and N, and a decreased microbial metabolic quotient. Consequently, long-term manure fertilization was beneficial to both soil C and N turnover and microbial activity, and had significant effect on the microbial metabolic quotient.  相似文献

To examine the synergism of high temperature and sulfide on two dominant tropical seagrass species, a large-scale mesocosm experiment was conducted in which sulfide accumulation rates (SAR) were increased by adding labile carbon (glucose) to intact seagrass sediment cores across a range of temperatures. During the initial 10 d of the 38 d experiment, porewater SAR in cores increased 2- to 3-fold from 44 and 136 μmol L− 1 d− 1 at 28-29 °C to 80 and 308 μmol L− 1 d− 1 at 34-35 °C in Halodule wrightii and Thalassia testudinum cores, respectively. Labile C additions to the sediment resulted in SAR of 443 and 601 μmol L− 1 d− 1 at 28-29 °C and 758 to 1,557 μmol L− 1 d− 1 at 34-35 °C in H. wrightii and T. testudinum cores, respectively. Both T. testudinum and H. wrightii were highly thermal tolerant, demonstrating their tropical affinities and potential to adapt to high temperatures. While plants survived the 38 d temperature treatments, there was a clear thermal threshold above 33 °C where T. testudinum growth declined and leaf quantum efficiencies (Fv/Fm) fell below 0.7. At this threshold temperature, H. wrightii maintained shoot densities and leaf quantum efficiencies. Although H. wrightii showed a greater tolerance to high temperature, T. testudinum had a greater capacity to sustain biomass and short shoots under thermal stress with labile C enrichment, regardless of the fact that sulfide levels in the T. testudinum cores were 2 times higher than in the H. wrightii cores. Tropical seagrass tolerance to elevated temperatures, predicted in the future with global warming, should be considered in the context of the sediment-plant complex which incorporates the synergism of plant physiological responses and shifts in sulfur biogeochemistry leading to increased plant exposure to sulfides, a known toxin.  相似文献

An anaerobic digestion technique was applied to textile dye wastewater aiming at the colour and COD removal. Pet bottles of 5 L capacity were used as reactor which contains methanogenic sludge of half a liter capacity which was used for the treatment of combined synthetic textile dye and starch wastewater at different mixing ratios of 20:80, 30:70, 40:60, 50:50 and 60:40 with initial COD concentrations as 3520, 3440, 3360, 3264 and 3144 mg L−1, respectively. The reactor was maintained at room temperature (30 ± 3 °C) with initial pH of 7. The maximum COD and colour removal were 81.0% and 87.3% at an optimum mixing ratio of 30:70 of textile dye and starch wastewaters. Both Monod’s and Haldane’s models were adopted in this study. The kinetic constants of cell growth under Haldane’s model were satisfactory when compared to Monod’s model. The kinetic constants obtained by Haldane’s model were found to be in the range of μmax = 0.037-0.146 h−1, Ks = 651.04-1372.88 mg L−1 and Ki = 5681.81-18727.59 mg L−1.  相似文献

For composting, passive aeration can save energy costs while being just as efficient as forced or active aeration. Passive aeration requires the proper design of aeration ducts, and thus, the proper prediction of the convective airflow rates created by the temperature differential between the compost and the ambient air. To establish such relationship, the temperature and convective air flow regimes of composts were investigated using three bulking agents (wood shavings, hay and straw), each at three moisture contents (MC-60%, 65% and 70%) spanning the normal values. All bulking agent and aeration treatments were aerated in duplicate under passive and active regimes. Laboratory vessels of 105 L were used for all treatments. Passive aeration treatments produced temperatures above 57 degrees C, as did the treatments actively aerated at 4 mg of air s(-1) kg(-1) of initial dry compost material. Compost MC had an effect only on the peak compost temperature, occurring between day 2 and 6. After 6 days of composting, MC no longer had any effect on temperature regime because of the loss of moisture by each mixture. A relationship was established between the Grasholf number (Gr-ratio of buoyancy to viscous forces) and the convective airflow rates, to size the aeration ducts for passive aeration. In general, convective airflow rates ranged from 1.5 to 0.7 mg of dry air s(-1) kg(-1) of initial compost dry matter, from day 0 to day 20, respectively, and for all compost treatments. This airflow rate sizes the aeration ducts installed under compost piles for passive aeration. As compared to straw where airflow rate dropped over a given level of Gr, wood shavings and hay were found to be more effective as bulking agents, as their airflow rate increased constantly with Gr.  相似文献

Kinetics of the reaction of octacarbonyl dicobalt with ethyl diazoacetate leading to [μ2-{ethoxycarbonyl(methylene)}-μ2-(carbonyl)-bis(tricarbonyl-cobalt)] (Co-Co) (1), dinitrogen, and carbon monoxide were investigated at 10 °C in heptane solution. The initial rate of the reaction was measured by following both the gas evolution and the decrease of the octacarbonyl dicobalt concentration. The rate is first order with respect to octacarbonyl dicobalt and a complex order with respect to ethyl diazoacetate and carbon monoxide depending on the ratio of their concentrations. This is in accord with the formation of a heptacarbonyl dicobalt reactive intermediate (k1 (10 °C) = (1.22 ± 0.06) × 10−3 s−1) for which carbon monoxide and ethyl diazoacetate compete (k−1/k2 (10 °C) = 1.34 ± 0.07).  相似文献

This study investigated the optimum thermochemical liquefaction (TCL) operating conditions for producing biocrude from Spirulina platensis. TCL experiments were performed at various temperatures (200-380 °C), holding times (0-120 min), and solids concentrations (10-50%). TCL conversion at 350 °C, 60 min holding time and 20% solids concentration produced the highest biocrude yield of 39.9% representing 98.3% carbon conversion efficiency. Light fraction biocrude (B1) appeared at 300 °C or higher temperatures and represented 50-63% of the total biocrude. Biocrude obtained at 350-380 °C had similar fuel properties to that of petroleum crude with energy density of 34.7-39.9 MJ kg−1 compared to 42.9 MJ kg−1 for petroleum crude. Biocrude from conversion at 300 °C or above had 71-77% elemental carbon, and 0.6-11.6% elemental oxygen and viscosities in the range 40-68 cP. GC/MS of biocrude reported higher hydrocarbons (C16-C17), phenolics, carboxylic acids, esters, aldehydes, amines, and amides.  相似文献

As residual bones in mortality compost negatively impact subsequent tillage, two studies were performed. For the first study, windrows of mature cattle or calves were placed on a base of barley straw and covered with beef manure. Windrows were divided into two sections and turned at 3-month intervals. Approximately 5000 kg of finished compost per windrow was passed through a 6 mm trommel screen, with bones collected and weighed. Bone weight was 0.66% of mature cattle compost and 0.38% of calf compost on a dry matter basis, but did not differ after adjustment for weights of compost ingredients. In a subsequent study, four windrows were constructed containing mortalities, straw and beef manure (STATC) or straw, manure and slaughter waste (STATW). Also, straw, beef manure and slaughter waste was added to an 850 L rolling drum composter (DRUMW). Fresh bovine long-bones from calves were collected, weighed and embedded in the compost. Bones were retrieved and weighed when windrows were turned, or with DRUMW, after 8 weeks. Temperatures achieved followed the order STATW > STATC > DRUMW (p < 0.05). Rate of bone disappearance followed a pattern identical to temperature, with the weight of bones in STATW declining by 53.7% during 7 weeks of composting. For STATC, temperatures were uniform over three composting periods, but bone disappearance was improved (p < 0.05) when compost dry matter was lower (46%), as compared to 58%. Using a ratio of five parts manure to one part mortalities, results of this study demonstrated that residual bone was <1% of cured cattle compost and may be reduced by maintaining a high compost temperature and moisture content.  相似文献

F1-ATPase, a water-soluble portion of the enzyme ATP synthase, is a rotary molecular motor driven by ATP hydrolysis. To learn how the kinetics of rotation are regulated, we have investigated the rotational characteristics of a thermophilic F1-ATPase over the temperature range 4-50°C by attaching a polystyrene bead (or bead duplex) to the rotor subunit and observing its rotation under a microscope. The apparent rate of ATP binding estimated at low ATP concentrations increased from 1.2 × 106 M−1 s−1 at 4°C to 4.3 × 107 M−1 s−1 at 40°C, whereas the torque estimated at 2 mM ATP remained around 40 pN·nm over 4-50°C. The rotation was stepwise at 4°C, even at the saturating ATP concentration of 2 mM, indicating the presence of a hitherto unresolved rate-limiting reaction that occurs at ATP-waiting angles. We also measured the ATP hydrolysis activity in bulk solution at 4-65°C. F1-ATPase tends to be inactivated by binding ADP tightly. Both the inactivation and reactivation rates were found to rise sharply with temperature, and above 30°C, equilibrium between the active and inactive forms was reached within 2 s, the majority being inactive. Rapid inactivation at high temperatures is consistent with the physiological role of this enzyme, ATP synthesis, in the thermophile.  相似文献
