In vitro experiments have demonstrated that the lactoperoxidase system produces a bactericidal effect on salmonellae and shigellae. The physiological concentrations of the components of this system, making it possible to obtain a pronounced bactericidal effect, have been established. Lactobacilli have been shown to potentiate the effect of the lactoperoxidase system. The possibility of realizing the bactericidal properties of the lactoperoxidase system with respect to salmonellae and shigellae in the preparations of immune lactosera, intended for passive enteral immunization against intestinal infections, has been suggested.  相似文献

Salmonella contamination of swine and morbidity rates among the workers of swine-breeding complexes and the members of their families, as well as among the population inhabiting the zone of possible influence rendered by such complexes on the environment, have been studied as exemplified by 4 complexes for large-scale swine breeding, differing in their technology of swine raising and fattening, their systems of the purification and utilization of manure-containing sewage. Besides, the influence of the raw meat (pork) and meat products, contaminated with salmonellae, sold in town, on salmonellosis morbidity among the urban population has been analyzed. The study has revealed that salmonellae are spread from swine-breeding complexes with the fluid and solid fractions of manure-containing sewage, as well as with the animals supplied to the meat-packing plant; the contamination of the workers of swine-breeding complexes with salmonellae becomes possible due to the low culture of the production and to the breach of sanitary and epidemic-control regulations and the contamination of the population occurs through meat and meat products.  相似文献

The action of T-activin on peritoneal macrophages of CBA mice after its introduction into the animals has been studied. In intact mice the phagocytic activity of macrophages and their resistance to the cytopathogenic action of Salmonella typhimurium live cells remains unchanged. The injection of corpuscular pertussis vaccine into mice leads to a decrease in the resistance of macrophages to the action of salmonellae. The simultaneous injection of T-activin into mice in doses of 0.1 and 1.0 microgram per animal abolishes the damaging action of the vaccine. The analysis of the in vitro action of T-activin on macrophages of intact mice revealed that the preliminary incubation of cells with the preparation sharply increases their resistance to the action of salmonellae, while its introduction simultaneously with bacteria or after them rapidly leads to the death of macrophages. The action of T-activin is supposed to be linked with triggering the biosynthetic processes mediating the resistance of macrophages to the cytopathogenic action of salmonellae.  相似文献

The comparative study of the incidence of the pathogenicity markers (DNAase, RNAase, phosphatase and hemolytic activity) in shigellae and salmonellae, acknowledged as the causative agents of intestinal infections, and in opportunistic bacteria isolated from the feces of patients with acute intestinal diseases and healthy persons has been made. The study has revealed that DNAase and RNAase activity occurs most frequently in Shigella flexneri, in salmonellae and in opportunistic enterobacteria isolated from the intestinal contents of patients with acute intestinal diseases. In this respect they essentially differ from the same species of opportunistic enterobacteria isolated from healthy persons.  相似文献

The growth phase of a bacterial (Salmonella typhimurium) culture was shown to have pronounced effects on the pathogenic properties of the harvested bacteria. Salmonellae obtained from a culture in primary (exponential) growth phase (PP) were more readily cleared from the blood and more readily killed by phagocytes than were salmonellae obtained from a more slowly growing secondary growth phase (SP) culture. PP salmonellae were observed to cause death of mice sooner than SP salmonellae. This appeared to be because the more rapid growth of PP, as compared to SP, salmonellae continued in the liver and spleen for several hours following intravenous injection, and more than compensated for their high in vivo death rate. As a result, within 4 h there were approximately 10-fold more live salmonellae in the spleens and livers of mice that had received PP, as compared to SP, salmonellae. This 10-fold difference was maintained until the death of the mice, indicating that after the first 4 h post-inoculation, the net in vivo growth of the salmonellae was the same regardless of their growth phase in the inoculating culture. This transition between PP and SP salmonellae occurred long before a dense stationary phase culture was obtained. Salmonellae grown in minimal media exhibited the biological properties of SP salmonellae and never entered as rapid a growth phase as did salmonellae in complete media.  相似文献

When injected intraperitoneally into mice in doses of 40.0-0.4 microgram, Cucumarioside, the preparation of triterpene glycosides obtained from sea cucumbers (Cusumaria japonica), enhanced the resistance of the animals to the subsequent challenge with Salmonella typhimurium. The study of the duration of the persistence of salmonellae in mice receiving the preparation in a dose of 0.001 microgram revealed a decrease in the contamination of their organs. The same dose of the preparation stimulated the phagocytic activity of peritoneal exudate cells with salmonellae showing decreased cytopathogenic action. This suggests that Cucumarioside enhances nonspecific protective factors, activates the macrophagal system and facilitates the development of complete phagocytosis.  相似文献

The authors studied the role played by Sac-plasmide of atypical S. newport strains inducing a more severe course of the disease in the development of the infectious process in man. In experiments with genetically-bound strains of salmonellae and E. coli K12, differing by the presence of Sac-plasmide, it was shown that the acquisition or the loss of the latter failed to effect significantly the capacity of the microbes to cause death of chick embryos infected on the membrane, to reproduce in the pulmonary tissue of mice infected intranasally, and affect the cells of the L1 strain. Genetic confirmation of the absence of any connection between the sucrose sign determined by plasmide and the capacity of the strains to cause dilatation of the ligated rabbit intestinal loop was presented. The data obtained indicated that high virulence of the S. newport strains under study was not associated with the presence of Sacplasmide in them.  相似文献

Single strains of Lactobacillus acidophilus and Lact. fermentum, isolated from chicken intestine, were used to study in vitro interactions with Salmonella enteritidis, Salm. pullorum or Salm. typhimurium in an ileal epithelial cell (IEC) radioactive assay. Exclusion, competition and displacement phenomena were investigated by respectively incubating (a) lactobacilli and IEC together, prior to addition of salmonellae, (b) lactobacilli, IEC and salmonellae together, and (c) salmonellae and IEC, followed by the lactobacilli. Lactobacilli were selected for study because of their strong ability to adhere to IEC and poor aggregation with salmonellae. The results demonstrated that Lact. acidophilus significantly reduced ( P < 0.05) the attachment of Salm. pullorum to IEC in the tests for exclusion and competition, but not in the displacement tests. Lactobacillus fermentum was found to have some ability to reduce the attachment of Salm. typhimurium to IEC under the conditions of exclusion ( P < 0.08), competition ( P < 0.09), but not displacement. However, both Lact. acidophilus and Lact. fermentum were unable to reduce the adherence of Salm. enteritidis to IEC under any of the conditions.  相似文献

In agreement with previous observations, mice dying of salmonellosis were found to have a relatively constant number of Salmonella cells in their carcasses (ca. 5 x 10(9)). This number was not the result of bacterial overgrowth in moribund animals and therefore appears to be related to lethality. Similar numbers of salmonellae were recovered from the carcasses of infected mice which had previously been rendered hyperreactive to endotoxin either by infection with M. tuberculosis BCG or by adrenalectomy. In BCG mice, desensitization to endotoxin did not occur during the infection and, therefore, at death these mice contained a number of bacteria which would be equivalent to 1,000 ld(50) of endotoxin. Although the number of bacteria recovered from normal mice is roughly equivalent to a lethal quantity of endotoxin, this is obviously not the case in hyperreactive mice. Therefore, the relationship between lethality and 5 x 10(9) salmonellae cannot be explained by their endotoxin content. Nevertheless, when hyperreactive BCG mice are challenged parenterally with Salmonella, the endotoxin content of the inoculum may markedly influence the course of the infection.  相似文献

Experimental Salmonella infection in rabbits was accompanied by immunomorphological transformation, starting in the ileum and the cervical lymph node on day 3, and in the popliteal lymph node on day 5 after inoculation. The intensity of immunomorphological reaction depended on the severity of the course of salmonellosis, and at the peak of the disease it depended on the degree of manifestation of intestinal pathology. The presence of salmonellae in the intestinal tissue, and particularly in the tissue of the ileum, facilitated the development of more pronounced and prolonged immunomorphological reaction in the mucous membrane of the ileum.  相似文献

To reveal the influence of cyclic adenosine monophosphate (cAMP) on the completion of the phagocytosis of salmonellae, the influence of insulin and isoproterenol on the phagocytic activity of peritoneal macrophages obtained from mice infected with S. typhimurium strains differing in virulence was studied in vitro. The study showed that isoproterenol, while increasing the intracellular content of cAMP, suppressed the bactericidal properties of macrophages with respect to salmonellae, whereas insulin decreased the level of cAMP in the cells and thus facilitated more rapid and complete digestion of ingested bacteria irrespective of their virulence.  相似文献

Samples of protein feed supplements produced by rendering plants were examined for salmonellae, total aerobic bacterial counts, coliform counts, and enterococci. Isolations of salmonellae were more frequent from products with high counts; however, 6% of the samples with total counts of less than 1,000 per gram and 14% of the samples with coliform counts of less than 1 per gram contained salmonellae. Serotypes of Escherichia coli which have been associated with disease in domestic animals and poultry were also isolated from products. Although the distribution of serotypes of salmonellae isolated from environmental swabs and flies was similar to that isolated from products, the isolation of several serotypes from flies which had not been isolated in plants suggested that flies may be a potential source of contamination.  相似文献

The bactericidal capability of murine peritoneal polymorphonuclear neutrophils against virulent and nonvirulent Salmonella typhimurium was examined in an in vitro system. Although preincubation of the bacteria in specific murine antiserum elicited greater chemiluminescence from phagocytizing neutrophils than did incubation in normal murine serum, antiserum did not enhance ingestion, as less than 5% of the challenge was taken up by neutrophils under any of the conditions studied. Nonvirulent salmonellae showed a transient decrease in viable numbers early during in vitro incubation with or without intact neutrophils. Virulent salmonellae, however, were able to multiply without a lag period except when these bacteria were pretreated with antiserum and incubated in association with intact murine neutrophils. Results of these in vitro studies suggest that the murine polymorphonuclear neutrophil and antisalmonella antibody must act together to effect neutrophil-associated bactericidal activity against virulent salmonellae, and thus, that the neutrophil alone does not play a major role in the protection of unvaccinated, sensitive mice from disease caused by S. typhimurium.  相似文献

The immunostimulating effect of corpuscular pertussis vaccine on the antigen-presenting and bactericidal functions of peritoneal and splenic macrophages in CBA and C57BL/6 mice, differing in the intensity of immune response to sheep red blood cells and Salmonella typhimurium, has been studied. The study has revealed that the injection of pertussis vaccine alters the functional activity of the cells under study, the effect depending on the immunizing dose, the strain of mice and the time elapsed from the moment of immunization. Pertussis vaccine enhances the low capacity of macrophages for antigen presentation in C57BL/6 mice with low responsiveness and alters the resistance of peritoneal and splenic macrophages to the cytopathic action of salmonellae.  相似文献

This study evaluated the potential for conversion of Class B to Class A biosolids with respect to salmonellae and fecal coliforms during solar drying in concrete lined drying beds. Anaerobically (8% solids) and aerobically (2% solids) digested Class B biosolids were pumped into field-scale drying beds, and microbial populations and environmental conditions were monitored. Numbers of fecal coliforms and salmonellae decreased as temperature and rate of desiccation increased. After 3 to 4 weeks, Class A requirements were achieved in both biosolids for the pathogens and the indicators. However, following rainfall events, significant increase in numbers was observed for both fecal coliforms and salmonellae. In laboratory studies, regrowth of fecal coliforms was observed in both biosolids and biosolid-amended soil, but the regrowth of salmonellae observed in the concrete-lined drying beds did not occur. These laboratory studies demonstrated that pathogens decreased in numbers when soil was amended with biosolids. Based on serotyping, the increased numbers of salmonellae seen in the concrete lined drying beds following rainfall events was most likely due to recolonization due to contamination from fecal matter introduced by animals and not from regrowth of salmonellae indigenous to biosolids. Overall, we conclude that the use of concrete-lined beds created a situation in which moisture added as rainfall accumulated in the beds, promoting the growth of fecal coliforms and salmonellae added from external sources.  相似文献

Autophagy has been demonstrated to be associated with the pathogenesis of cancer, but no consensus has been reached about its precise role. Therefore, we investigated whether autophagy in the intestinal epithelium is involved in the pathogenesis of intestinal tumors. To evaluate the relationship between autophagy and intestinal tumors, GFP-LC3-APC(min/+) mice were generated by mating GFP-LC3 transgenic mice with APC(min/+) mice. Autophagy was weakly induced in the intestinal polyp regions of the mice in comparison to their non-polyp regions. Under starved conditions, autophagy was not induced in the polyp regions, whereas it was observed in the non-polyp regions. Then, to examine whether a lack of autophagy in the intestinal epithelium enhances the induction of intestinal tumor, Atg7flox/flox:vil-cre-APC(min/+) mice, in which Atg7 had been conditionally deleted in the intestinal epithelium, were generated by mating Atg7flox/flox:vil-cre mice with APC(min/+) mice. However, there was no significant difference in the number of intestinal polyps between the Atg7flox/flox:vil-cre-APC(min/+) and the corresponding control Atg7flox/flox-APC(min/+) mice. These results indicate that autophagy in the intestinal epithelium is not involved in the pathogenesis of intestinal tumors, and future research should focus on regulating autophagy as a form of cancer therapy.  相似文献

The mouse Chromosome 1 locus Ity regulates the extent to which Salmonella typhimurium replicates within the reticuloendothelial cell system (RES) during the first days of infection. If animals are homozygous for the Itys susceptibility allele, the Gram-negative bacterium undergoes rapid net multiplication, and mice die of a typhoid fever-like disease by day 10 of infection. Animals that are homozygous or heterozygous for the resistance allele, Ityr, control net bacterial replication and survive the first phase of salmonellosis. Indirect studies have implicated the resident macrophage as the effector cell for regulation of early in vivo salmonellae growth. To verify this supposition and to evaluate the phenotypic expression of Ity, we developed an in vitro assay to compare kinetics of S. typhimurium growth within Ityr and Itys macrophages. Resident peritoneal and splenic macrophages were used from inbred Ityr and Itys mice and from Ity congeneic mice. With these mice and through the use of radiolabeled S. typhimurium and an avirulent temperature-sensitive mutant of the bacterium, we found that: phagocytosis of S. typhimurium by Ityr and by Itys macrophages was the same; S. typhimurium grew to a greater extent in Itys peritoneal and splenic macrophages than in Ityr cells; Ityr macrophages killed intracellular salmonellae more efficiently than did Itys macrophages. Thus, we have demonstrated directly that Ity is expressed by the macrophage and have shown for the first time with Ity congeneic mice that the basis for differential net growth of virulent S. typhimurium in Ityr and Itys macrophages is a variation in the degree of bacterial kill.  相似文献

A high correlation of the salmonellae and enterococci indices with the distance from the sources of contamination (effluents) at calm weather and reduced correlation at storm was revealed in examination of coastal waters of Baltic sea for salmonellae and indicator microbes in the biotypes of two populated localities. Monotypical character of the isolated salmonellae served as the peculiarity of the results obtained: S. typhimurium prevailed (96.3%): S. give were isolated in 3.7% of the cases. The latter were isolated chiefly in the biotypes of one of the populated localities (B); their relative content showed a sharp elevation during storm.  相似文献

Lethally irradiated conventional mice were killed at daily intervals after irradiation. A high correlation was found between the occurrence of bacterial over-growth in the digestive tract and invasion into the regional lymph nodes the spleen and the blood. By oral contamination of mice with quite high doses of either an exogenous or an endogenous biotype of an Enterobacteriaceae species at day 4 after irradiation, it was attempted to induce such condition of intestinal over-growth. In all animals such an abnormal colonization of the contaminant was indeed achieved in this way and the mice died from an infection caused by the biotype used for the contamination.The interval between contamination and death was different when different biotypes were used for contamination and occurred sooner after irradiation when an endogenous biotype was used for (re)contamination. It is concluded that oral contamination with exogenous as well as with endogenous potentially pathogenic bacteria during a period of severely decreased resistance to infections must be avoided. The colonization resistance of the digestive tract of these animals should be maintained as high as possible.  相似文献
