Sporulation of the rust fungus Uromyees phaseoli was checked on three different Brasilian bean cultivars on the upper and lower leaf surface of primary leaves in a growth chamber at 21°C. Although the sporulating area of the pustules was greater on the upper leaf surface, the pustules on the lower leaf surface produced nearly two times more spores on all three cultivars. The total number of spores produced per pustule was 41,600/81,000 spores (upper/ lower leaf surface) for the susceptible cultivar Rosinha G-2/C-21 and for the cultivars which possess horizontal resistance: Carioca/C-224 29,600/49,500 spores (upper/lower leaf surface), Roxo/C-743 32,964/50,700 spores (upper/lower leaf surface). The two cultivars with horizontal resistance produced nearly a third less spores than the susceptible cultivar.  相似文献

Ochratoxin A is a toxic and carcinogenic fungal secondary metabolite; its presence in foods is increasingly regulated. Various fungi are known to produce ochratoxins, but it is not known which species produce ochratoxins consistently and which species cause ochratoxin contamination of various crops. We isolated fungi in the Aspergillus ochraceus group (section Circumdati) and Aspergillus alliaceus from tree nut orchards, nuts, and figs in California. A total of 72 isolates were grown in potato dextrose broth and yeast extract-sucrose broth for 10 days at 30°C and tested for production of ochratoxin A in vitro by high-pressure liquid chromatography. Among isolates from California figs, tree nuts, and orchards, A. ochraceus and Aspergillus melleus were the most common species. No field isolates of A. ochraceus or A. melleus produced ochratoxin A above the level of detection (0.01 μg/ml). All A. alliaceus isolates produced ochratoxin A, up to 30 μg/ml. We examined 50,000 figs for fungal infections and measured ochratoxin content in figs with visible fungal colonies. Pooled figs infected with A. alliaceus contained ochratoxin A, figs infected with the A. ochraceus group had little or none, and figs infected with Penicillium had none. These results suggest that the little-known species A. alliaceus is an important ochratoxin-producing fungus in California and that it may be responsible for the ochratoxin contamination occasionally observed in figs.  相似文献

Biosynthesis of ochratoxin A by Aspergillus ochraceus Wilh. was investigated by radiolabeling experiments in which phenylalanine-1-14C and sodium acetate-2-14C were supplied to the fungus in sucrose-yeast extract medium. Results showed that phenylalanine was incorporated unaltered into the phenylalanine moiety of ochratoxin A, whereas the isocoumarin moiety of ochratoxin A was mostly derived via acetate condensation.  相似文献

Occurrence of ochratoxin A in herbal drugs of Indian origin — a report   总被引:1,自引:0,他引:1  
This paper contains a report of occurrence of ochratoxin A in some common herbal medicines collected from different store-houses and shop-keepers of Bihar, India. Of 129 samples of 9 plants, 55 were found to be contaminated with various levels of ochratoxin A. The level of ochratoxin A was found maximal in barks ofHolarrhena antidysenterica (1.14 – 2.34 μg/g) whereas it was minimal in rhizomes ofTacca aspera (0.3 – 0.74 μg/g).Aspergillus ochraceus, A sulphureus and Penicillium viridicatum isolates obtained from drug samples were also examined for their toxigenic potentials. 19 isolates ofA ochraceus, 13 ofA sulphureus and 37 isolates ofP viridicatum were found to be toxigenic out of 67, 33, and 107 isolates, respectively. The ochratoxin A produced by Aochraceus was in the range of 0.09 to 2.44 μg/mL, byA sulphureus 0.1 to 1.76 μg/mL, and byP viridicatum 0.14 to 2.78 μg/mL of the culture filtrate.  相似文献

The maize weevil Sitophilus zeamais Motsch. is an important pest of maize that attacks the grain both in the field and during storage. The damage caused by the maize weevil S. zeamais on maize landraces, Amarelo Antigo, Asteca, Caiano, Carioca, and Ferrinho, was evaluated by no-choice tests under laboratory conditions. The commercial varieties Sol da Manh?, BR 106, BR 451, and the synthetics PC 0203 and PC 9903 were evaluated for comparisons with the maize landraces. The parameters evaluated were susceptibility index, number of weevil progeny, development time, weevil progeny dry weight, and grain dry weight loss. The landraces were more susceptible to the maize weevil as compared to the commercial varieties. Based on the cluster analysis, two groups of susceptibility to the maize weevil were observed: one of more susceptible populations formed by local landraces and BR 451, and another less susceptible, with commercial varieties, synthetics, and the landrace Amarelo.  相似文献

Ochratoxin A is a potent nephrotoxin and a possible human carcinogen that can contaminate various agricultural products, including grapes and wine. The capabilities of species other than Aspergillus carbonarius within Aspergillus section Nigri to produce ochratoxin A from grapes are uncertain, since strain identification is based primarily on morphological traits. We used amplified fragment length polymorphisms (AFLPs) and genomic DNA sequences (rRNA, calmodulin, and β-tubulin genes) to identify 77 black aspergilli isolated from grape berries collected in a 2-year survey in 16 vineyards throughout Italy. Four main clusters were distinguished, and they shared an AFLP similarity of <25%. Twenty-two of 23 strains of A. carbonarius produced ochratoxin A (6 to 7,500 μg/liter), 5 of 20 strains of A. tubingensis produced ochratoxin A (4 to 130 μg/liter), 3 of 15 strains of A. niger produced ochratoxin A (250 to 360 μg/liter), and none of the 19 strains of Aspergillus “uniseriate” produced ochratoxin A above the level of detection (4 μg/liter). These findings indicate that A. tubingensis is able to produce ochratoxin and that, together with A. carbonarius and A. niger, it may be responsible for the ochratoxin contamination of wine in Italy.  相似文献

Ochratoxin A (OTA), a mycotoxin produced by Aspergillus and Penicillium species, is composed of a dihydroisocoumarin ring linked to phenylalanine, and its biosynthetic pathway has not yet been completely elucidated. Most of the knowledge regarding the genetic and enzymatic aspects of OTA biosynthesis has been elucidated in Penicillium species. In Aspergillus species, only pks genes involved in the initial steps of the pathway have been partially characterized. In our study, the inactivation of a gene encoding a nonribosomal peptide synthetase (NRPS) in OTA-producing A. carbonarius ITEM 5010 has eliminated the ability of this fungus to produce OTA. This is the first report on the involvement of an nrps gene product in OTA biosynthetic pathway in an Aspergillus species. The absence of OTA and ochratoxin α, the isocoumaric derivative of OTA, and the concomitant increase of ochratoxin β, the dechloro analog of ochratoxin α, were observed in the liquid culture of transformed strain. The data provide the first evidence that the enzymatic step adding phenylalanine to polyketide dihydroisocoumarin precedes the chlorination step to form OTA in A. carbonarius and that ochratoxin α is a product of hydrolysis of OTA, giving an interesting new insight into the biosynthetic pathway of the toxin.  相似文献

Adaptive strategies of desmids in two tropical monomictic lakes were studied in relation to morphometric differences. The study aimed at evaluating desmids' strategies during inter-annual succession patterns in two tropical lakes (Dom Helvécio and Carioca) in southeast Brazil. From monthly samples of freshwater algae collected during five consecutive years (2002–2006) from the two lakes, the desmids were classified according to size, life form, and life strategies (competitive, stress tolerant, and ruderal). Our results indicated that Lake Carioca (19º45′20″S, 42º37′12″W), a small, shallow, turbid, and mesotrophic system showed dominance of invasive/opportunistic taxa (C) and R-strategists (Staurastrum species <20 μm), whereas Lake Dom Helvécio (19º45′–19º48′45″S, 42º33′45″W), a large, deep, and clear system was dominated by ruderal (R) species (Closterium aciculare and Pleurotaenium trabecula >50 μm) and tolerant to stress (S) species (Staurastrum and Staurodesmus species >20 μm). A significant number of desmids from the lakes had low MLD (≤20 μm), low volume (<103), and high SV?1 ratio (>1). Consequently, C-strategist species were abundant and formed a group having great adaptive value in the lakes, with higher resilience and capacity to change composition after the turnover period. Stratification stability and atelomixis were the main driving forces to sustain high desmid biomass in these two lakes, despite the greater macrophyte richness and great water column depth at Lake Dom Helvécio and the main factor influencing richness and contribution of complex forms.  相似文献

Wheat (moisture content: 26%) was autoclaved or left untreated, inoculated with conidia ofPenicillium viridicatum and stored at 10°C. The fungus grew on both substrates and was the dominant mould on the non-autoclaved grain. Autoclaving resulted in an earlier onset of ergosterol, ochratoxin A, and citrinin production due to accelerated mould growth. Yield of ochratoxin A increased while citrinin slightly decreased in autoclaved wheat.  相似文献

The effect of zinc, copper, and iron levels on production of ochratoxin A by Aspergillus ochraceus Wilhelm in a synthetic medium in a shake culture was investigated. Optimal concentrations of ZnSO4, CuSO4, and FeCl3 for ochratoxin A production were 0.055 to 2.2 mg/liter, 0.004 to 0.04 mg/liter, and 1.2 to 24 mg/liter, respectively. Zinc and copper levels greater than optimum reduced the rate of ochratoxin accumulation without altering either glutamate or sucrose utilization. Ochratoxin A production was correlated with rapid utilization of sucrose by the fungus and decreasing pH of the medium. Most of the glutamic acid was removed from the medium prior to ochratoxin production. There was no correlation between mycelial dry weight and ochratoxin A production.  相似文献

A method is described for the isolation and purification of ochratoxin A, ochratoxin B, ochratoxin ß mellein, 4-hydroxymellein and penicillic acid produced byAspergillus ochraceus in a synthetic liquid medium. Ochratoxin α, which was not found in the culture medium, was obtained by acid hydrolysis of ochratoxin A. The southern state of Brazil is responsible for 90 % of the wine produced in the country; however, the mycobiota and the occurrence of ochratoxin A (OTA) on wine grapes produced in this region has never been fully characterized. To evaluate the fungal and OTA contamination in wine grapes of two varieties cultivated in southern Brazil, a survey was conducted in three regions with slightly different climates. From each region, samples were obtained at early ripeness and at harvest stage. Eight genera were isolated; Alternaria and Trichoderma were the most prevalent. Within the Aspergillus genus, the section Nigri was predominant. One uniseriate black Aspergillus was able to produce OTA under the conditions tested but no detectable levels of OTA were found in the grape samples. The low overall incidence of black Aspergillus and the absence of OTA on the grapes may be explained by the meteorological conditions at the analyzed regions, and characterizes a high quality of this product regarding mycotoxigenic contamination. This is the first study to fully describe the mycological biota of two wine grape varieties produced at three different regions of Rio Grande do Sul state.  相似文献

Bean plants Phaseolus vulgaris L. (cv. Carioca and Negro Huasteco) and Vigna unguiculata L. Walp (cv. Epace-10) were grown in a growth chamber with a photosynthetic photon flux density of 200 mol m–2 s–1 at leaf level and air temperature of 25+1 °C. Fully expanded, first pair leaves of 12-d-old plants were submitted for 90 min to high temperature (25, 30, 35, 40, 45, and 48 °C). Chlorophyll a fluorescence parameters (ETR, qP, qN, and F0) were investigated using a modulated fluorimeter at 25 °C during recovery considered here as 48 h after stress induction period. An accentuated decrease in qP and an increase in qN at 48 °C in Carioca and Negro Huasteco was not observed in Epace-10. In response to excitation irradiance a great potential for ETR was found in Negro Huasteco at 25 °C, also demonstrated by net photosynthetic rate. At 48 °C ETR was high for Epace-10 while it was equal to zero for Carioca and Negro Huasteco. Tolerance to high temperature observed in Epace-10 provided important information about the adaptative characteristics of Vigna cultivars to warm climates.  相似文献

Natural feed ingredients (corn, barley and wheat bran) and compound feed (manufactured pellet) are two types of fodder used for animal feeding, especially camel in Saudi Arabia. Twenty samples of each type of fodder were collected from seven different regions and screened for the presence of fungi, aflatoxins, ochratoxin and zearalenone. Fungal isolation of natural feed ingredients yielded 10 genera and 38 species of different fungi. Compound fodder samples were contaminated with 16 genera and 32 species of fungi. Total counts of Aspergillus, Penicillium and Fusarium in the animal feed samples were ranged from 54 to 223 × 103, 31.9 to 60 × 103 and 18 to 29 × 103 CFU/g, respectively. These isolates when tested for aflatoxin, ochratoxin and zearalenone producing ability, revealed this property in only four isolate, identified as Aspergillus flavus, A. parasiticus, A. ochraceus and Fusarium graminaerum. The percentage of toxigenic fungi was ranged from 5.5% to 30% for natural feed ingredients and from 4.5% to 20% for compound feed. The incidence of aflatoxins (AFT) in samples of natural feed ingredients was found to be ranged from 1 to 24.8 ppb, ochratoxin A (OTA) ranged from 1 to 44 ppb and zearalenone (ZON) ranged from 1 to 23 ppb. Contamination of compound feed with aflatoxin and ochratoxin A was ranged from 1 to 6.4 ppb and 1 to 4.7 ppb, respectively. All samples collected were found contaminated with fungi or their toxins and natural feed samples were more contaminated compared to compound feed samples. The concentrations detected were in the allowed limit (<20 ppb) except four samples of natural feed ingredients which were above the allowed limit of the tested mycotoxins. In conclusion, feed samples were contaminated with fungi and some toxigenic isolates which were responsible about mycotoxin production. Some samples had exceeded amount of AFT, OTA and ZON and may be contaminated with other mycotoxins which mean implication of fungi in camel health problems and death in Saudi Arabia.  相似文献

During bean seed storage, yield can be lost due to infestations of Acanthoscelides obtectus Say, the bean weevil. The use of resistant varieties has shown promising results in fighting these insects, reducing infestation levels and eliminating chemical residues from the beans. The expression of resistance to A. obtectus in bean varieties is frequently attributed to the presence of phytohemagglutinins, protease inhibitors and alpha-amylase, and especially to variants of the protein arcelin, which reduce the larval viability of these insects. To evaluate the effect of bean seed storage time on the resistance expression of bean varieties to A. obtectus , tests with seeds of three ages (freshly-harvested, 4-month-old, and 8-month-old) were conducted in the laboratory, using four commercial varieties: Carioca Pitoco, Ipa 6, Porrillo 70, ônix; four improved varieties containing arcelin protein: Arc.1, Arc.2, Arc. 3, Arc.4; and three wild varieties also containing arcelin protein: Arc.1S, Arc.3S, and Arc. 5S. The Arc.5S, Arc.1S, and Arc.2 varieties expressed high antibiosis levels against the weevil; Arc.1 and Arcs expressed the same mechanism, but at lower levels. The occurrence of oviposition non-preference was also observed in Arc.5S and Arc.1S. The Arc.3 and Arc. 4 varieties expressed low feeding non-preference levels against A. obtectus. The expression of resistance in arcelin-bearing, wild or improved varieties was affected during the storage of seeds, and was high under some parameters but low in others. The results showed that addition of chemical resistance factors such as protein arcelin via genetic breeding may be beneficial in improving the performance of bean crops.  相似文献

The occurrence of toxigenic fungi producing aflatoxins and ochratoxin A in foodstuffs was studied in the Czech Republic. Twenty five commodities were collected at twelve collection places in the Czech Republic (300 food samples). The presence of potentially toxigenicAspergillus flavus was observed in 28% of the sampled foods (black pepper, caraway seeds, fruit tea, black tea, oat flakes, fine flour, rolled oat flakes and semolina) in the year 1999, and in 25% of the sampled foods (black pepper, black tea, fine flour) in the year 2000.A tamarii (aflatoxins producer) was found in 3 black pepper samples (25%) in both years. Aflatoxins were detected in black pepper and caraway seed samples in the year 1999 and in sweet red pepper in the year 2000.A parasiticus andA nomius were not isolated. Aspergillus section Nigri (potential producer of ochratoxin A) was detected in some foodstuffs. A defined minimal medium has been developed which is either restrictive or permissive for the production of ochratoxin A byPenicillium verrucosum simply by changing the nitrogen and carbon source. The combination of ammonium/glycerin promoted, whereas the combination nitrate/glucose repressed ochratoxin production.In parallel mutants ofP verrucosum have been constructed by restriction endonuclease mediated integration, which were not able to produce ochratoxin. Different types of transformants, which either produce no detectable amounts of ochratoxin A but ochratoxin α, strains which produce only ochratoxin B and strains which produces none of these secondary metabolites, have been observed.  相似文献

Income breeding animals support reproduction by the intake of energy and molecular building blocks during adulthood. Capital breeders, in contrast, fuel reproduction by the intake of materials during the larval stage. There exists, however, some controversy as to whether adult feeding in capital breeders actually increases fitness. We tested whether individuals feed as adults in the weevil Zabrotes subfasciatus Boheman, whether they equally accept carbohydrate- or nitrogen-rich food (sugar or yeast), whether food ingestion varies with the available host type and whether feeding as adults alters life history traits. The hosts tested were three varieties of Phaseolus vulgaris, Carioca, Jalo, and Preto; Vigna unguiculata Frade; and Vicia faba. We also tested whether sugar ingestion affects life history traits differently in the presence of different host types. We found that Z. subfasciatus feed as adults and equally accept sugar and yeast, and food ingestion depends on host type. Sugar and yeast significantly promote oviposition of larger eggs on Carioca seeds and enhance adult emergence, and sugar ingestion increases longevity. Moreover, the insects avoid feeding when on V. faba seeds, on which they did not oviposit. Mean fitness values showed the following hierarchy: Carioca = Jalo > Frade > Preto > V. faba. These results show that the capital breeder Z. subfasciatus feed as adults, which increases life history traits and suggests the existence of a hierarchy of performance on different hosts, with the P. vulgaris varieties Carioca and Jalo offering the best conditions.  相似文献
