The stoichiometry, stability constants and solution structure of the complexes formed in the reaction of copper(II) with hexapeptide NPTNLH, i.e. the Neobelliera Bullata Trypsin Modulating Oostatic Factor (Neb-TMOF), and its analogues DPTNLH, Ac-NPTNLH and Ac-DPTNLH have been determined by potentiometric, UV-visible, CD and EPR spectroscopic methods. Upon raising pH for Ac-NPTNLH and Ac-DPTNLH peptides, copper(II) coordination starts from the imidazole nitrogen of the His6; afterwards three deprotonated amide nitrogens are progressively involved in metal ions coordination. In a wide pH range of 4.5-8.5 for the NPTNLH and DPTNLH ligands the CuL complex dominates with the imidazole nitrogen of His6 coordinated to form a macrochelate. The N-terminal amino group of the NPTNLH and DPTNLH peptides takes part in the coordination of the metal ion in the CuL, CuH−1L and CuH−2L complexes. However, at pH above 9 the CuH−3L complex with the {NIm, 3N} coordination mode is formed. For the CuH−2L complex the spectroscopic data clearly indicate the 4N {NH2, CO or COO, 2N, NIm} bonding mode with the axial coordination of the N-terminal amine group to the metal ion.  相似文献

His-Val-His and His-Val-Gly-Asp are two naturally occurring peptide sequences, present at the active site of Cu,Zn-superoxide dismutase (Cu,Zn-SOD). We have already studied the interaction of His-Val-His=A (copper binding site) with Cu(II) and of His-Val-Gly-Asp=B (zinc binding site) with Zn(II). As a continuation of this work and for comparison purposes we have also studied the interaction of Zn(II) with His-Val-His and Cu(II) with His-Val-Gly-Asp using both potentiometric and spectroscopic methods (visible, EPR, NMR). The stoichiometry, stability constants and solution structure of the complexes formed have been determined. Histamine type of coordination is observed for/ZnAH/2+, /ZnA/+, /ZnA2H/+ and/ZnA2/ in acidic pH while deprotonation of coordinated water molecules is observed at higher pH. /CUB/ species is characterized by the formation of a macrochelate and histamine type coordination. Its stability results in the suppression of amide deprotonation which occurs at high pH resulting in the formation of the highly distorted from square planar geometry 4N complex/CuBH-3/3.  相似文献

The copper(II), nickel(II) and zinc(II) binding ability of the multi-histidine peptide N-acetyl-His-Pro-His-His-NH2 has been studied by combined pH-potentiometry and visible, CD and EPR spectroscopies. The internal proline residue, preventing the metal ion induced successive amide deprotonations, resulted in the shift of this process toward higher pH values as compared to other peptides. The metal ions in the parent [ML]2+ complexes are exclusively bound by the three imidazole side chains. In [CuH−1L]+, formed between pH 6-8, the side chains of the two adjacent histidines and the peptide nitrogen between them are involved in metal ion binding. The next deprotonation results in the proton loss of the coordinated water molecule (CuH−1L(OH)). The latter two species exert polyfunctional catalytic activity, since they possess superoxide dismutase-, catecholase- (the oxidation of 3,5-di-tert-butylcatechol) and phosphatase-like (transesterification of the activated phosphoester 2-hydroxypropyl-4-nitrophenyl phosphate) properties. On further increase of the pH rearrangement of the coordination sphere takes place leading to the [CuH−3L] species, the deprotonated amide nitrogen displaces a coordinated imidazole nitrogen from the equatorial position of the metal ion. The shapes of the visible and CD spectra reflect a distorted arrangement of the donor atoms around the metal ion. In presence of zinc(II) the species [ZnL]2+ forms only above pH 6, which is shortly followed by precipitation. On the other hand, the [NiL]2+ complex is stable over a wide pH range, its deprotonation takes place only above pH 8. At pH 10 an octahedral NiH−2L species is present at first, which transforms slowly to a yellow square planar complex.  相似文献

Formation (affinity) constants for 1:1 complexes of N-(2-acetamido)iminodiacetic acid (ADAH2) with Ca(II), Mg(II), Mn(II), Zn(II), Co(II), Ni(II), and Cu(II) have been determined. Probable structures of the various metal chelates existing in solution are discussed. Values for the deprotonation of the amide group in [Cu(ADA)] and subsequent hydroxo complex formation are also reported. The use of ADA as a buffer is considered in terms of metal buffers complexes which can be formed at physiological pH, i.e., at pH 7.0 there is essentially no free metal ion in 1:1 M2+ to ADA solutions.  相似文献

The crystal and molecular structures of the N-rac-isomer of the nickel(II) complex of 14-membered amide-containing macrocycle [NiL1] · 4H2O (H2L1=5,12-dioxo-1,4,8,11-tetraazacyclotetradecane) have been determined. Two deprotonated amide and two amine donors co-ordinate to the nickel(II) in nearly square planar manner with Ni-Namine bonds longer than Ni-Namide ones (1.930 vs. 1.898 Å). Water molecules do not co-ordinate and form hydrogen bond bridges between macrocyclic units in the crystal lattice. The analysis of 1H NMR data confirmed that the solid-state conformation of the macrocycle in N-rac[NiL1] is retained in aqueous solution though equilibrated with some amount of N-meso isomer. The comparison of the spectroscopic characteristics of the M(II) and M(III) complexes and the redox potentials of M(III/II) couples (M=Ni and Cu) for ML1 with those for ML2(H2L2=5,7-dioxo-1,4,8,11-tetraazacyclotetradecane) revealed a rather small influence of the trans- vs. cis-arrangement of amide donors in co-ordination spheres of the metal ions.  相似文献

The oligomerization of [CuII(Hx(tmdnTAA))]x+ (x = 0, 1, 2 and (tmdnTAA))2− is 2,4,9,11-tetramethyl-dinaphto[14]-2,4,6,9,11,13-hexaeneN4) was initiated in homogeneous solution via the reaction of this Cu(II) complex with pulse radiolytically generated radicals. The reaction produces Cu(III) intermediates which are rapidly converted to Cu(II) ligand-radical species. In contrast to the mechanism proposed for the electrochemical oligomerization, where the local concentration of radicals is probably high, the reaction kinetics in homogeneous solution is propagated by a process where the Cu(II) ligand-radical precursors react with [CuII(Hx(tmdnTAA))]x+.  相似文献

Electron spin resonance spectra of the first Cu(II) complexes of human serum albumin, dog serum albumin, l-aspartyl-l-histidine N-methylamide and glycyl-glycyl-l-histidine N-methylamide have been studied using isotopically pure 65Cu in its chloride form. At 77° K, the esr spectra of Cu(II) complex of human serum albumin exhibited only one form of esr signal between pH 6.5 and 11. No intermediate forms were detected. The presence of an equally spaced nine-line superhyperfine structure with spacing ~15 G indicated considerable covalent bonding between Cu(II) and four nitrogen atoms derived from the protein. The esr spectrum form of Cu(II) bound to human serum albumin detected at neutral pH would be consistent with the participation of four nitrogens from the α-NH2 group, two peptide groups, and the imidazole group of a histidine residue. In contrast, the esr spectra of Cu(II)-dog serum albumin complex showed a transition from a low pH form to a high pH form as the pH was increased to 9.5. These spectral changes were found to be reversible upon lowering the pH. Ligand superhyperfine splittings in the low pH form of the esr signal of Cu(II)-dog albumin were not resolved. The distinct pH dependence of the esr signals observed in human and dog serum albumin complexes could be correlated to their respective optical spectra changes as a function of pH. At room temperature and in the pH range between 6 and 11, the esr spectra of Cu(II) complexes of l-aspartyl-l-alanyl-l-histidine N-methylamide and glycyl-glycyl-l-histidine N-methylamide exhibited a well-resolved nine-line superhyperfine structure indicating metal coordination with four equivalent nitrogen atoms of peptide.  相似文献

A new, 14-membered, tetraza cyclic tetrapeptide containing histidine and lysine side-chains, c(β3homoLysdHisβ-AlaHis), was designed, synthesized and characterized; its copper(II) binding properties were investigated in dependence of pH by potentiometric and spectroscopic methods. In line with previous studies of similar systems, the progressive involvement of amide nitrogens in copper(II) coordination was evidenced for pH values greater than 6. At physiological pH the dominant species consists of a copper(II) center coordinated by two amide nitrogens, an imidazole nitrogen and a water molecule. In contrast, at pH values higher than 8.7, a copper(II) coordination environment consisting of four amide nitrogens in the equatorial plane and the axial imidazole ligands is formed as clearly indicated by spectroscopic data and theoretical calculations. The behavior of this 14-membered cyclic tetrapeptide is compared to that of its 12-membered cyclic analog, particular attention being paid to the effects of ring size on the respective copper(II) binding abilities.  相似文献

The tetradecapeptide containing the 10 aminoacid repeated sequence on the C-terminus of the Ni(II)-induced Cap43 protein, was analyzed for Ni(II) and Cu(II) binding. A combined pH-metric and spectroscopic UV-VIS, EPR, CD and NMR study of Ni(II) and Cu(II) binding to the blocked CH3CO-Thr-Arg-Ser-Arg-Ser-His-Thr-Ser-Glu-Gly-Thr-Arg-Ser-Arg-NH2 (Ac-TRSRSHTSEGTRSR-Am) peptide, modeling a part of the C-terminal sequence of the Cap43 protein, revealed the formation of octahedral complexes involving imidazole nitrogen of histidine, at pH 5.5 and pH 7 for Cu(II) and Ni(II), respectively; a major square planar 4N-Ni(II) complex (about 100% at pH 9, log K* = -28.16) involving imidazole nitrogen of histidine and three deprotonated amide nitrogens of the backbone of the peptide was revealed; a 3N-Cu(II) complex (maximum about 70% at pH 7, log K*=-13.91) and a series of 4N-Cu(II) complexes starting at pH 5.5 (maximum about 90% at pH 8.7, log K* = -21.39 for CuH(-3)L), were revealed. This work supports the existence of a metal binding site at the COOH-terminal part of the Cap43 peptide.  相似文献

The results are reported of a spectroscopic and potentiometric study of the copper(II) and nickel(II) complexes of the thyrotropin releasing factor (L-pyroglutamyl-L-histidyl-L-prolinamide, TRF) and some of its di- and tripeptide analogues Spectroscopic techniques used include absorption, circular dichroism and electron paramagnetic resonance spectroscopy TRF and pyroglutamyl-histidine behave similarly. At low pH the metal ions coordinate to the imidazole nitrogen and then cause the ionization of the amide protons of both the peptide linkage and the pyroglutamic acid with equal ease. Hence the concentration of MH?1 L species is always very low. The C-terminal proline amide residue plays an insignificant role in the complex formation Replacement of pyroglutamic acid with picolinic acid in the hormone molecule causes a major change in the structures of its complexes. The dipeptide analogue, Pic-His. forms dimeric species with Cu(II) that are not found in Cu(II) Pyr-His orCu(II) TRF solutions The introduction of tyrosine residue in the TRF sequence in place of histidine can, in some cases, lead to the direct involvement of proline amide in the binding of metal ions, e.g. , Ni(II) Pyr-Tyr-Pro-NH2  相似文献

Five heterocyclic benzothiazoline and -thiazole analogs have been synthesized and characterized by 1H NMR and IR spectroscopy. The analogs fall into two different classes, (a) those which contain one benzothiazoline group adjacent to the heterocyclic ring system (starting with 2-pyridinecarboxaldehyde, 2-thiophenecarboxaldehyde or 2-furaldehyde), and (b) those which have two benzothiazoline substituents (starting with 2,6-pyridinecarboxaldehyde and 2,5-thiohenecarboxaldehyde). In addition, the imine containing ligands, bis-2-[(pyridin-2-ylmethylene)-imino]-benzenethiol disulfide (PyIS)2 and bis-2-[(thiophen-2-ylmethylene)-imino]-benzenethiol disulfide(ThIS)2, were prepared starting with the disulfide of 2-aminothiophenol. Each species has been characterized by 1H NMR and IR spectroscopies. Ligation reactions with 2-(2-pyridyl)benzothiazoline (Py(Bt)) and Cu(OAc)2·1H2O resulted in the formation of a dinuclear species containing two copper ions, two ligand frames and two acetate groups, [Cu(PyAS)(OAc)]2 (1). Both copper ions are five-coordinate and bonded to one monodentate acetate, one ligand frame (NNS) and one bridging thiolate. Ligation reaction with 2-(2-pyridyl)benzothiazole (Py(oBt)) and CoCl2·xH2O or Cu(BF4)2·xH2O resulted in the formation of a six-coordinate, octahedral Co(II) complex, cis-[Co(Py(oBt)2Cl2] (2) and a five coordinate Cu(II) complex, [Cu(Py(oBt))2(OH2)](BF4) (3), respectively. As a basis for understanding the role of albumin in the transport of metal ions, detailed investigations have been carried out to elucidate the structure of Ni(II)- and Cu(II)-binding site of the peptide residue corresponding to the NH2-terminal peptide fragment 1-24 of human serum albumin by 1H and 13C NMR spectroscopy. These studies have been conducted in aqueous medium at different pH values and at different ligand/metal ratios. The results show the following: (i) Diamagnetic Ni(II) complex and paramagnetic Cu(II) complex are in slow exchange NMR time scale. (ii) Titration results of Ni(II)-bound form of peptide 1-24 show the presence of a 1:1 complex in the wide pH range (6.0-11.0), and the same stoichiometry is proposed for Cu(II) as well. (iii) Analysis of the spectra suggests that both Ni(II) and Cu(II) have one specific binding site at the NH2-terminal tripeptide segment (Asp-Ala-His...) involving the Asp alpha-NH2, His N(1) imidazole, two deprotonated peptide nitrogens (Ala NH and His NH), and the Asp COO- group. (iv) Complexation of Ni(II) and Cu(II) causes conformational change near the metal-binding site of the polypeptide chain, but there is no other binding group involved besides those in the first three residues.  相似文献

A novel ruthenium(II) complex of dipyridophenazine (DPPZ) with the ancillary ligand imidazole[4,5-f] [1,10]phenanthroline (IP), [Ru(IP)2(DPPZ)] (PF6)2, has been synthesized and characterized by elemental analysis, 1D and 2D 1H NMR, fast-atom bombardment mass spectra (FABMS), electronic spectroscopy and cyclic voltammetry. The DNA-binding properties of the complex were studied by spectroscopic methods. The intrinsic binding constant, K =2.1 × 107M−1, of the complex to calf thymus DNA has been determined by absorption titration in 5 mmol dm−3 Tris-HCl, 50 mmol dm−3 NaCl buffer (pH 7.0). The excited state lifetimes and luminescence quenching with [Fe(CN)6]4− as the quencher in the presence of DNA were also tested and mono-exponentiality was observed for the emission decay curves. Viscosity measurements together with the optical titrations unambiguously proved that the complex bound with DNA intercalatively and that the binding affinity to DNA was several times larger than that of the parent complex [Ru(bpy)2(DPPZ)]2+.  相似文献

The GH4C1 strain of hormone-producing rat pituitary cells has specific receptors for the tripeptide thyrotropin-releasing hormone (TRH). Membranes prepared from GH4C1 cells show intrinsic tryptophan fluorescence which was quenched by low concentrations (10–100 nM) of TRH and Nτ-methyl TRH but not by biologically inactive analogs of TRH. Membranes from GH4C1 cells were subjected to thermal denaturation. A conformational transition was noted above 40°C and an irreversible denaturation was observed at 52°C. TRH-induced quenching of intrinsic fluorescence was lost completely in membranes previously incubated for 10 min at 30°C while loss of [3H]-TRH binding was only about 20% at this temperature. Collisional quenching by iodide revealed that about 38% of the tryptophanyl residues in GH4C1 membranes were exposed to solvent. Quenching by TRH occurred with a shift in wavelength maximum from 336 to 342 nm suggesting that few of the tryptophanyl residues quenched by the tripeptide are totally exposed. Membranes prepared from cells preincubated with 20 nM TRH for 48 h, in which TRH receptors were decreased to 30% of control values, showed no quenching of tryptophan fluorescence in response to freshly added TRH. We conclude that the TRH-receptor interaction in GH4C1 cells is associated with a change in membrane conformation that can be measured by differential spectrofluorometry of intrinsic tryptophan fluorescence.  相似文献

Cobalt(II), nickel(II), copper(II) and zinc(II) complexes with 2-acetylthiophene benzoylhydrazone have been synthesized and characterized by elemental analyses, magnetic susceptibility measurements, electronic, IR, NMR and ESR spectral techniques. The molecular structures of ligand and its copper(II) complex have been determined by single crystal X-ray diffraction technique. The Cu(II) complex possesses a CuN2O2 chromophore with a considerable delocalization of charge. The structure of the complex is stabilized by intermolecular π–π stacking and C–H?π interactions. Hatbh acts as a monobasic bidentate ligand in all the complexes bonding through a deprotonated C–O and >CN groups. Electronic spectral studies indicate an octahedral geometry for the Ni(II) complex while square planar geometry for the Co(II) and Cu(II) complexes. ESR spectrum of the Cu(II) complex exhibits a square planar geometry in solid and in DMSO solution. The trend g|| > g > 2.0023 indicates the presence of an unpaired electron in the dx2-y2 orbital of Cu(II). The electro-chemical study of Cu(II) complex reveals a metal based reversible redox behavior. The Ni(II) complex shows exothermic multi-step decomposition pattern of the bonded ligand. The ligand and its most of the metal complexes show appreciable corrosion inhibition properties for mild steel in 1 M HCl medium. [Co(atbh)2] complex exhibited the greatest impact on corrosion inhibition among the other compounds.  相似文献

1H and 13C NMR studies of Zn(II) binding to oxidized glutathione (GSSG) in aqueous solution over the pH range 4-11 show that it forms a complex with a 1:1 Zn:GSSG stoichiometry. At pH values between 6 and 11 the metal ligands are the COO- and NH2 groups of the glutamate residues. Below pH 5 the glycine end of the molecule also binds to the metal ions. EPR and visible absorption spectra of Cu(II) GSSG solutions suggest that similar complexes are formed with Cu(II). The solid products obtained from these solutions are shown by analysis and EPR to be primarily binuclear with Cu2GSSG stoichiometry, although the structures depend on the pH and stoichiometry of the solution from which they were obtained.  相似文献

Interactions of inosine derivatives with copper(II) were studied in the pH range 1.4–13 in 50% H2O-50% DMSO solution. The distinct pH dependence of the optical spectra observed in copper(II)-inosine complexes are correlated to their respective EPR changes as a function of pH. It was concluded that a simple 1:1 complex of copper(II)-inosine is formed in the pH range 1.4–5.0 and bis complexes are present in the pH 5.0–6.2 region solutions of inosine and Cu(II). From pH 6.2 to 7.8 a diamagnetic, hydroxybridged complex dominates. At pH 7.8–9.2 an insoluble, oxybridged species is formed in addition to the soluble paramagnetic Cu(NI)4 complex. Starting from pH 9.1 the N-polymeric complex is formed which is stable up to pH 12.5, and above pH 12.5 the only species is the Cu(ribose)2 complex.  相似文献

The variations in the coordination environment of Co(II), Cu(II) and Zn(II) complexes with the neutral, tridentate ligand bis[1-(cyclohexylimino)ethyl]pyridine (BCIP) are reported. Analogous syntheses were carried out utilizing either the M(BF4)2 · xH2O or MCl2 · xH2O metal salts (where M = Co(II), Cu(II) or Zn(II)) with one equivalent of BCIP. When the hydrated metal starting material was used, cationic, octahedral complexes of the type [M(BCIP)2]2+ were isolated as the tetrafluoroborate salt (4, 5). Conversely, when the hydrated chloride metal salt was used as the starting material, only neutral, pentacoordinate [M(BCIP)Cl2] complexes (1-3) formed. All complexes were characterized by X-ray diffraction studies. The three complexes that are five coordinate have distortions due mainly to the pyridine di-imine bite angle. The [Cu(BCIP)Cl2] (2) also exhibits deviations in the Cu(II)-Cl bond distances with values of 2.4242(9) and 2.2505(9) Å, which are not seen in the analogous Zn(II) and Co(II) structures. Similarly, the two six coordinate complexes (5, 6) are also altered by the ligand frame bite angle giving rise to distorted octahedral geometries in each complex. The [Cu(BCIP)2](BF4)2 (6) also exhibits Cu(II)-Nimine bond lengths that are on average 0.14 Å longer than those found in the analogous 5 coordinate complex, [Cu(BCIP)Cl2]. In addition to X-ray analysis, all complexes were also characterized by UV/Vis and IR spectroscopy with 1H NMR spectroscopy being used for the analysis of the Zn(II) analogue (3).  相似文献

Combined pH-metric, UV-Vis, 1H NMR and EPR spectral investigations on the complex formation of M(II) ions (M=Co, Ni, Cu and Zn) with N-(2-benzimidazolyl)methyliminodiacetic acid (H2bzimida, hereafter H2L) in aqueous solution at a fixed ionic strength, I=10−1 mol dm−3, at 25 ± 1 °C indicate the formation of M(L), M(H−1L) and M2(H−1L)+ complexes. Proton-ligand and metal-ligand constants and the complex formation equilibria have been elucidated. Solid complexes, [M(L)(H2O)2] · nH2O (n=1 for M = Co and Zn, n=2 for M = Ni) and {Cu (μ-L) · 4H2O}n, have been isolated and characterized by elemental analysis, spectral, conductance and magnetic measurements and thermal studies. Structures of [Ni(L)(H2O)2] · 2H2O and {Cu(μ-L) · 4H2O}n have been determined by single crystal X-ray diffraction. The nickel(II) complex exists in a distorted octahedral environment in which the metal ion is coordinated by the two carboxylate O atoms, the amino-N atom of the iminodiacetate moiety and the pyridine type N-atom of the benzimidazole moiety. Two aqua O atoms function as fifth and sixth donor atoms. The copper(II) complex is made up of interpenetrating polymeric chains of antiferromagnetically coupled Cu(II) ions linked by carboxylato bridges in syn-anti (apical-equatorial) bonding mode and stabilized via interchain hydrogen bonds and π-π stacking interactions.  相似文献

Mixed ligand complexes of the type Cu(Z-aminoacidato)2(B2) (Z = benzyloxycarbonyl group, Z-aminoacidate = Z-glycinate (Zgly), Z-alaninate (Zala); Z-valinate (Zval), Z-leucinate (Zleu) ion, B = imidazole (Im), N-methylimidazole (MeIm)) were synthesized and characterized by means of electronic, infrared and EPR spectroscopies. For one of them, bis(Z-alaninato)bis(N-methylimidazole)copper(II) ethanol solvate, the crystal and molecular structure was also determined by the single crystal X-ray diffraction method. The complex crystallizes in the monoclinic space group P21/c, with cell dimensions a = 11.1119(6), b = 18.8398(7), c = 8.9652(5) Å, β = 105.380(2)° and Z = 2. The structure was solved by conventional Patterson and Fourier methods and refined by full-matrix least-squares to an R value of 0.045. The complex has square-planar coordination via two centrosymmetric carboxylic oxygens and two N-methylimidazole nitrogens. The second carboxylate oxygen is 2.731(5) Å from the copper atom in an ‘out of plane’ position. Packing is mainly determined by hydrogen bondings between amide nitrogen and amide carboxyl oxygen. Electronic, infrared and EPR spectra are consistent with this type of coordination geometry for anhydrous complexes, while for hydrate complexes are suggestive of tetragonal bipyramidal geometry.  相似文献
