Dry lettuce seeds (Lactuca sativa L. cv. Grand Rapids), whichreceived 5 min far-red light (FR) 0.5 h after the onset of waterimbibition, showed 17% and 50% germination without and withacid immersion treatment (pH 0.1) for 1 h and rinsing with water,respectively. The acid treatment caused only 6% germinationor less in FR-treated seeds held for 10 to 30 d in dark storage.The 10 to 30 d skotodormant seeds did not respond to red light(R) or gibberellin A3 (GA3) singly, but showed 84% or higherpercentage germination if 1 h acid immersion was given beforeR or GA3. The 20 d skotodormant seeds, which received R treatmentat day 10 but remained dormant showed 89% germination with onlyacid treatment. Similar values were obtained with 30 d skotodormantseeds which received one or two R treatments at day 10 or 20,i.e. the only requirement for these R-treated dormant seedswas an acid immersion. This releases the skotodormancy and rendersthe seeds more sensitive to R or GA3, but the skotodormancywas initiated again if no light or hormone treatments were givenimmediately. The repetitive R or GA3 treatments, which did notcause skotodormant seeds to germinate, lessened the degree ofskotodormancy. The germination of these skotodormant seeds canonly be induced by the synergistic action of R and GA3. In thisstudy, GA3 caused higher germination percentages in R-treatedskotodormant seeds than R stimulated in GA3-treated seeds. Itis suggested that (i) repetitive R or Ga3 treatments maintaina high endogenous level of the far-red-absorbing form of phytochrome(Pfr) and GA activity, respectively, (ii) the accumulated stableintermediates of phytochrome persist in fully-imbibed skotodormantseeds for up to 20 d, without phytochrome expressing its functionuntil the seeds are acidified and (iii) a model is formulatedto interpret the results of acidification, growth promotersand R effects on germination of light-sensitive lettuce seeds. Ten-30 d imbibed skotodormant lettuce seeds (Lactuca sativaL. cv. Grand Rapids) showed no germination with water alone.However, following a single treatment of red light (R), gibberellinA3 (GA3) or 1 h acid immersion (pH 0–1) plus water rinse,7% germinated. These imbibed skotodormant seeds germinated 85%or higher if acid immersion was carried out before R or GA3.Similar values were obtained with imbibed skotodormant seedsunder acid immersion plus drying treatment applied at day 10or 20 plus R or GA3 treatment applied 10 d later. One or twodrying treatments alone reduced the degree of skotodormancyand made seeds more responsive to R but not GA3. Seeds withone R plus drying treatment at day 10 or 20 germinated about50% with or without an additional R, and 80% or more with GA3on day 20 or 30. The 20 or 30 d skotodormant seeds having R(with or without drying) or acid plus R and drying treatmenton day 10 or 20 and additional dark incubation in water for10d showed 85 to 100% germination with only acid immersion.The skotodormancy was eliminated by the acid immersion but itwas initiated again if R or GA3 treatment was not given immediately.It is concluded that the drying treatment, after eliminationof skotodormancy by acid or acid + R pretreatment, preventsthese seeds re-entering skotodormancy and maintains a high germinationpotential under dark storage for up to 20 d. A 10-min irradiation with red light (R) of ‘Grand Rapids’lettuce (Lactuca sativa L.) seeds increased respiratory rates(QO2 and QCO2) over those of far red (FR)-irradiated seeds within2 to 3 hr. Differences in respiration between R- and FR-irradiatedseeds became more pronounced with time. The respiration, indarkness at 25?C, of seeds air dried after germination promotingimpregnation treatments with 1.0 mM GA3+0.5 mM kinetin in acetonewas stimulated slightly by the 6th hour after planting and markedlyby the 12th and 24th hour. Results with seeds in GA3+kinetindissolved in water were similar. ABA, which inhibits germinationand seedling growth, prevented stimulation in respiration oflight-induced seeds. ABA also repressed respiration when usedwith GA3+kinetin. The germination and growth retardant, isopropylN-(3-chlorophenyl) carbamate (CIPC), did not affect the respirationof either R- or FR-treated seeds. The effects of phytochrome/light and other regulatory agentson the swelling of protoplasts from the primary leaves of etiolatedoat seedlings have been investigated. The protoplasts did notswell in darkness. Red (R) light immediately followed by far-red(FR) or FR light treatment alone for 4 h induced swelling slowly.In comparison, the protoplasts treated with R or FR-R swellmore rapidly and with a shorter lag period. The effect of redlight on the protoplast swelling was photoreversible by FR,suggesting the involvement of endogenous phytochrome. Exogenous gibberellic acid (GA3) or dibutyryl cAMP (DBcAMP)stimulated the effect of R irradiation on the protoplast swelling.As with R irradiation, these agents were sufficient to causethe swelling of protoplasts in the dark with a shorter lag periodthan those maintained in darkness or a 5 min FR. The combinationof a 5 min R irradiation and GA3 showed a synergistic effecton the enlargement of protoplast size. Two fractionation procedures were used to study the phenomenonof phytochrome pelletability or binding to a particulate fractionof maize coleoptiles. Using a revised procedure, we were unableto show an increase of phytochrome pelletability during darkincubation of red irradiated plant tissue, reported by Manabeand Furuya for pea seedlings (6), and the Pfr-enhanced affinityfor Pr in R/FR irradiated tissue as reported by Quail et al.(11). However, we were able to match these reported observationsusing a procedure which we have regarded as standard. In thestandard procedure, the irradiated tissue is homogenized andthe brei permitted to incubate in the dark at 0?C before fractionationby differential centrifugation prior to measurements of phytochromepelletability. In the revised procedure this incubation is eliminatedand fractionation and measurement follow directly on tissuehomogenization. A progressive decrease of particulate phytochromewas observed during dark incubation at 0?C of the brei fromred irradiated tissue, but no substantial decrease was observedduring dark incubation of the red irradiated tissue at 0?C.The decrease was not dependent on in vitro PfrPr reversion.In the case of R/FR irradiated tissues, phytochrome pelletabilitywas found to decrease during dark incubation of both the irradiatedtissue and its brei at 0?C. With these results and a recognitionof the tendency of phytochrome to dissociate from the particulatefraction in vitro, we have thus rationalized the results ofQuail et al. Germination responses to light were studied in the upper andlower seeds of cocklebur (Xanthium pennsylvanicum Wallr.). Thelower seed was dark-germinating and negatively photoblastic;the upper one had a red-light (R) requirement and was positivelyphotoblastic. Germination of the lower seeds was inhibited bya prolonged single irradiation with R, blue (B) or far-red (FR)light applied during imbibition. The maximal inhibitory effectof a single irradiation occurred 9 h and 13 h after the startof soaking at 33 °C and 23 °C, respectively. However,the inhibitory effect of R differed from that of B and FR, byonly delaying germination. A single exposure to B or FR lightcould be replaced by intermittent B or FR irradiation, and theireffects were repeatedly reversible by the following R irradiation.If the upper seeds were not exposed to R during imbibition,they failed to germinate even at 33 °C which was optimalfor germination, and the promotive effect of R increased withdelay of its application time. The photoperceptive locus incocklebur seeds was the axial tissue for all B, R and FR. Lightreceived by the cotyledonary tissue had little effect. Germinationdimorphism in response to light is discussed with respect tothe phytochrome content and the ageing of axial tissues. Effects of ethylene, CO2 and O2 on the germination of after-ripenedupper cocklebur (Xanthium pennsylvanicum Wallr.) seeds wereexamined in relation to pre-irradiation by red (R) or far-red(FR) light In order to remove the pre-existing Pfr, seeds weresoaked in the dark for various periods prior to light irradiationand gas treatments. Regardless of light, 0.3 Pa C2H4 promotedgermination at 23 ?C, but it strongly inhibited germinationwhen applied at 33 ?C, the optimal temperature for the germinationof this seed. However, delayed application of C2H4 during 33?C incubation stimulated germination independently of lightin a similar manner to that seen at 23 ?C. It is, therefore,suggested that the germination-regulating action of C2H4 iscompletely independent of phytochrome. In contrast, the germination-promoting effect of 3–0 kPaCO2 was pronounced only when the seeds were previously irradiatedby R, regardless of temperature, suggesting that CO2 actionto promote germination depends upon Pfr. A synergism betweenCO2 and C2H4 at 23 ?C was observed only in the germination ofseeds pre-irradiated by R, while at 33 ?C an antagonism occurredindependently of light. The stimulation of C2H4 production byCO2 was most striking in the cotyledonary tissue pre-irradiatedby R. However, the R-dependent enhancement of CO2-stimulatedC2H4 production was negated by the subsequent FR and it wasnot found in the presence of 1-aminocyclopropane-1-carboxylicacid (ACC). Moreover, the R dependency of the germination-promotingCO2 effect disappeared in the presence of C2H4. The R-dependentC2H4 production enhanced by CO2 may thus be involved, at leastpartially, in some step of conversion from methionine to ACC. The germination-promoting effect of C2H4, but not CO2, was enhancedby O2 enrichment regardless of light. It has previously been demonstrated that far-red irradiation of dry Lactuca sativa L. seeds results in inhibition of subsequent germination. Although red has no effect on dry seeds, a red irradiation following a farred irradiation reverses the effect of far-red. This phenomenon is most noticeable in seeds with artificially raised levels of phytochrome in the far-red absorbing form. Qualitatively similar results have been found for the seeds of Plantago major L., Sinapis arvensis L., and Bromus sterilis L. Action spectra studies on Plantago seeds show that the action peaks for promotion and inhibition of germination of hydrated seeds are at 660 and 730 nanometers, respectively. The action spectrum for inhibition of subsequent germination following irradiation of dry seeds is qualitatively and quantitatively similar to that for hydrated seeds, with an action peak at 730 nanometers, indicating absorption by phytochrome in the far-red absorbing form. However, the action spectrum for the reversal of this far-red effect on dry seeds has a broad peak at 680 nanometers and subsidiary peaks at 650 and 600 nanometers. It is proposed that this effect is due to light absorption by the phytochrome intermediate complex meta-Fa, and that the action spectrum reflects the in vivo absorption properties of this intermediate.  相似文献

One short red (R) irradiation increases the ATP content of Kalanchoë blossfeldiana Poelln. cv Feuerblüte seeds before onset of germination. Phytochrome control is demonstrated by the full R/far-red light (FR) reversibility of the effect in water imbibed seeds. In seeds imbibed in the presence of gibberellin A3 (GA3, one short R exposure already increases the ATP content when given 2h after start of imbibition, showing phytochrome control at the energy-metabolic level when one R pulse cannot yet induce germination. After longer imbibition periods in the presence of GA3, one short FR irradiation also increases the ATP content of ungerminated Kalanchoë seeds. The time course of the ATP levels after a R or FR germination inducing irradiation shows an initial increase that clearly preceeds germination. A second increase starts about 15 h after irradiation and is most probably the consequence of the germination itself. The results suggest that, in Kalanchoë seeds, the increase in ATP levels, induced by irradiation(s) and preceding germination, is a phytochrome-mediated process, supplying energy, required for germination.  相似文献

A short period (15–30 min) at 30° C promotes germination of seeds of Lactuca sativa L. cv. Repolhuda in darkness. Far-red light reverses this stimulation, and the escape curves for phytochrome and high-temperature action are quite similar, indicating that the two factors act at a common point in the chain of events leading to germination. Seeds of paper birch (Betula papyrifera Marsh.) were induced to germinate by prechilling at 3 C or by red light. The light requirement was mediated by phytochrome and the action of phytochrome during prechilling was investigated. Red irradiation (R) prior to prechilling markedly enhanced the effectiveness of the prechilling treatment in inducing subsequent germination at 18 C. Reversal of this enhancement by far-red irradiation (FR) was more effective when FR was supplied after a 1-week prechill treatment than after a 2-week treatment. The R enhancement effect exhibited a sharp drop as prechilling temperature was increased from 5 to 7 C. This decline is consistent with a membrane phase transition at about 7 C where Pfr action is diminished by a loss in sensitivity of its receptor sites. Although phytochrome action was observed during prechilling treatments, the seeds failed to germinate at prechilling temperatures. Therefore, it was concluded that while potentiation of germination by Pfr occurred during prechilling, some other reaction(s) leading to radicle protrusion requires higher temperatures. In one seed source loss of germination potential was observed with protracted storage at 3 C. This was prevented by R supplied during the prechilling treatment. Taken collectively the data suggest that action of phytochrome during prechilling is accentuated in these seeds by two factors: (a) an increase in the sensitivity (or number) of Pfr receptor sites; and (b) preservation of Pfr by deferment of thermal reversion.  相似文献

The elongation rate of cowpea epicotyls from whole cowpea (Vigna sinensis) seedlings and derooted and debladed plants (explants) increased after the main light period (8-hour duration) was extended with either continuous low intensity tungsten light or brief (5 minutes) far-red (FR) irradiation. This end-of-day FR effect was reversed by red (R) irradiation suggesting the involvement of phytochrome. These results confirm and extend those obtained previously with other species. Localization studies indicate the epicotyl to be the site of the photoreceptor. Treatment of cowpea seedlings with paclobutrazol, a gibberellin (GA) biosynthetic inhibitor, abolished the FR promoted epicotyl elongation, indicating a role for GAs in this process. There was no significant difference in epicotyl elongation rates of R plus FR irradiated explants treated with GA1 or GA20 and R irradiated explants treated with GA1. However, R irradiation inhibited subsequent epicotyl elongation of GA20 treated explants. Moreover, the observation, using GC-MS, that GA1 and GA20 are native GAs in cowpea lends support to the concept that phytochrome may control the conversion of endogenous GA20 to GA1 in cowpea.  相似文献

Summary In henbit (Lamium amplexicaule L.) seeds, micromolar levels of gibberellic acid (GA3) introduced at the onset of imbibition markedly stimulated germination. Sensitivity to GA3 stimulation is rapidly lost if the seeds are pretreated in darkness at 15° for 1 day or more on a water substrate. Loss in responsiveness to GA3 is hastened by pretreatment in far-red or by dark pretreatment at higher (25°) temperatures and is reduced by pretreatments at 5°. Germination of certain dry seeds (achenes) of Lactuca sativa L. cv. Grand Rapids was increased to ca. 75% after irradiation with 665 nm red light (R; 1x103 J m-2); this response was eliminated by far-red light (FR) following the R. The response of dry seeds required an order of magnitude more light than that of wetted seeds, and was not maximal until 48 h after irradiation. Other seeds, which could not be stimulated by R in dry state, showed a partial response after 10 min hydration. Irradiation of dry seeds (or seeds wetted 1 h) with FR (1x103 J m-2) reduced dark germination from 26% to 2%. Seeds dehydrated in an oven (60°C, 90 min) showed a decrease in germination if irradiated with R (1x105 J m-2) before wetting. The results show that phytochrome is present in dry lettuce seeds (and functional in some seed lots) prior to wetting; and that in other seed lots the molecule becomes functional within minutes after wetting the seeds. Transformation of the FR absorbing from of phytochrome (PFR) to the inactive from (PR) occurs at lower seed moisture content than the reverse reaction. It appears that dormancy in seeds ripened in sunlight might be assured during seed drying and maturation by the more effective transformation of PFR to PR than vice versa as phytochrome is dehydrated.Abbreviations FR far-red - R red - CAL seeds from California - NC seeds from North Carolina (see text)  相似文献

Combinations of far-red light (FR) (4 min) and gibberellic acid (GA3), given at the beginning of a daily 12-h dark period in a growth room, were used to study floral induction in four maturity genotypes of the milo group of sorghum (Sorghum bicolor (L.) Moench). The 12-h dark period without GA3 application or FR induced flowering in only the early genotype; FR hastened initiation in the early genotype, while GA3 hastened floral initiation in the two intermidiate-flowering genotypes. GA3 and FR together had a strong synergistic effect, hastening floral initiation by 30 to more than 80 d in the early and intermediate genotypes. Red light (R) did not hasten flowering; FR preceded by R gave the same effect as FR alone. GA3 promoted stem elongation equally whether floral initiation occurred or not; thus, its effect on stem elongation was independent of floral initiation. Plant-derived smoke extracts mimics the effect of red light on germination in light-sensitive lettuce seeds and partially overcomes the inhibitory effect of far-red light. Interaction between a smoke extract and gibberellins, cytokinins, abscisic acid and ethephon was investigated. Smoke acted synergistically with GA3 and increased the sensitivity of the lettuce seeds to ABA. Germination of Rumex obtusifolius L. seeds (nutlets) is low in darkness at 25° C. Germination is stimulated by exposure to 10 min red light (R) and also by a 10-min elevation of temperature to 35° C. A 10-min exposure to far-red light (FR) can reverse the effect of both R (indicating phytochrome control) and 35° C treatment. Fluence-response curves for this reversal of the effect of R and 35° C treatments are quantitatively identical. The effects of red light, far-red light, Gibberellin A3, andethephon were studied on the germination of lettuce seeds cv.Grand Rapids harvested at different stages of development. Seeds did not become capable of germination until 8 days afteranthesis. Red light promoted seed germination from the age of8–9 days following anthesis up to the newly mature stage.Ten or 11 days following anthesis, a large percentage of seedsbecame capable of germination in the dark and therefore couldbe considered not dormant. They were affected by far-red light,but less so than the mature seeds. The effect of light on the germination of developing seeds appearedto be similar to the known light effect on mature lettuce seedgermination. Measuring the ratio of the number of photooriented chloroplaststo the total number of chloroplasts, we found that photoorientationof chloroplasts in protonemata of the fern Adiantum capillus-veneriscould be induced by brief irradiation with polarized red light.After irradiation with red light (R) of 3 or 10 min, orientationalmovement was detected as early as 10 min after the irradiation;it continued during the subsequent dark period for 30–60min, after which chloroplasts gradually dispersed again. WhenR-treated protonemata were irradiated briefly with a second10-min pulse of R, 60 min after the onset of the first irradiation,the orientational response of chloroplasts was again observed.Typical red/far-red photoreversibility was apparent in the response,indicating the involvement of phytochrome. By contrast, irradiationwith polarized blue light for 10 min was ineffective, whileirradiation with blue light (B) at the same fluence for a longerperiod of time clearly induced the photoorientation of chloroplasts.It is likely that longterm irradiation is necessary for theresponse mediated by a blue-light receptor. When protonemata were irradiated with far-red light (FR) immediatelyafter R or after a subsequent dark period of 10 min, the magnitudeof the orientational response was smaller and chloroplasts dispersedmore quickly than those exposed to R alone. When FR was appliedat 50 min, when the response to R had reached the maximum level,chloroplasts again dispersed rapidly to their dark positions.These results indicate that PFR not only induces the photoorientationmovement of chloroplasts but also fixes the chloroplasts atthe sites to which they have moved as a result of photoorientation.
