Sucrose-agarose gels and sucrose liquid diets were used to study the phenol oxidising enzymes in the salivary secretions of the grain aphid, Sitobion avenae (Fabricius). Activity indicating the presence of two oxidoreductases, polyphenol oxidase (PPO) and peroxidase (Px), was found. Both enzymes were present in the aphids stylet sheath (gelling saliva) but only polyphenol oxidase activity was found in the halos around sheaths and thus in watery saliva. Electrical penetration graphs (EPG) revealed that the secretion of these enzymes into the gels, by an individual aphid, was associated with its probing activity observed during penetration of the epidermal and mesophyll tissues. The grain aphids PPO, secreted in its saliva reacted with a range of phenolic compounds. As most of these phenolics occur naturally in cereals, the grain aphid could modify its host-plants phenolic composition. The importance of the grain aphids polyphenol oxidase and peroxidase in detoxifying cereal phenolics is discussed.  相似文献

In order to study the effect of glycosylation on its biological activities and to develop IL-1 with less deleterious effects, N-acetylneuraminic acid (NeuAc) with C9 spacer was chemically coupled to human recombinant IL-1. NeuAc-coupled IL-1 (NeuAc-IL-1) exhibited reduced activities in vitro and receptor-binding affinities by about ten times compared to IL-1. In this study, we examined a variety of IL-1 activities in vivo. NeuAc-IL-1 exhibited a marked reduction in the activity to up-regulate serum IL-6, moderate reduction in the activities to up-regulate serum amyloid A and NOx. However, it exhibited comparable activities as IL-1 to down-regulate serum glucose and to improve the recovery of peripheral white blood cells from myelosuppression in 5-fluorouracil-treated mice. In addition, tissue level of NeuAc-IL-1 was high compared to IL-1. These results indicate that coupling with NeuAc enabled us to develop neo-IL-1 with selective activities in vivo and enhanced tissue level.  相似文献

Overexpression of the MDR protein, or p-glycoprotein (p-GP), in cells leads to decreased initial rates of accumulation and altered intracellular retention of chemotherapeutic drugs and a variety of other compounds. Thus, increased expression of the protein is related to increased drug resistance. Since several homologues of the MDR protein (CRP, ltpGPA, PDR5, sapABCDF) are also involved in conferring drug resistance phenomena in microorganisms, elucidating the function of the MDR protein at a molecular level will have important general applications. Although MDR protein function has been studied for nearly 20 years, interpretation of most data is complicated by the drug-selection conditions used to create model MDR cell lines. Precisely what level of resistance to particular drugs is conferred by a given amount of MDR protein, as well as a variety of other critical issues, are not yet resolved. Data from a number of laboratories has been gathered in support of at least four different models for the MDR protein. One model is that the protein uses the energy released from ATP hydrolysis to directly translocate drugs out of cells in some fashion. Another is that MDR protein overexpression perturbs electrical membrane potential () and/or intracellular pH (pHi) and therebyindirectly alters translocation and intracellular retention of hydrophobic drugs that are cationic, weakly basic, and/or that react with intracellular targets in a pHi, or -dependent manner. A third model proposes that the protein alternates between drug pump and Cl channel (or channel regulator) conformations, implying that both direct and indirect mechanisms of altered drug translocation may be catalyzed by MDR protein. A fourth is that the protein acts as an ATP channel. Our recent work has tested predictions of these models via kinetic analysis of drug transport and single-cell photometry analysis of pHi, , and volume regulation in novel MDR and CFTR transfectants that have not been exposed to chemotherapeutic drugs prior to analysis. This paper reviews these data and previous work from other laboratories, as well as relevant transport physiology concepts, and summarizes how they either support or contradict the different models for MDR protein function.  相似文献

The relationship is examined between vegetation and climate using climatic variables collected from 644 meteorological stations located throughout China. Multivariate methods are applied directly to the raw climatic data in order to define climatic clusters and to examine the relationship between the clusters and vegetation types. This approach is based on the concept of multidimensional climatic space defined by the combination of climatic variables. Phytoclimatic classes are defined on the basis of the distribution of vegetation types in climatic clusters and a new phytoclimatic classification of China is proposed. Patterns of climatic changes between neighbouring phytoclimatic classes are described. Two indexes of the influence of climate on vegetation are proposed based on discriminant analysis.  相似文献

Summary 3 families with hereditary angioedema in 9 members are described. The serum concentrations of complement factors C3, C4, and C-inhibitor (CINH) were determined in all available family members. C inhibiting activity was characteristically low or absent in sera from patients. CINH and C4 concentrations were also low, whereas C3 was present in normal concentrations in patients and in unaffected family members. In all 3 families fatal attacks of laryngeal edema had occured. 4 individuals, 2 children and 2 adults, had low concentrations of CINH and C4 but had never experienced symptoms of angioedema.
Zusammenfassung 3 Familien mit hereditärem Angioödem bei insgesamt 9 Mitgliedern werden beschrieben. Die Serumkonzentrationen der Komplementfaktoren C3, C4 und des C-Inhibitors (CINH) wurden bei allen erreichbaren Familienmitgliedern bestimmt. Die C-Hemmaktivität und die Konzentrationen von CINH und C4 waren in den Patientenseren niedrig. Der C3-Serumspiegel war jedoch bei Patienten und gesunden Familienmitgliedern normal. In allen 3 Familien sind tödlich verlaufende Anfälle von Larynxödem vorgekommen. 4 Familienmitglieder, 2 Kinder und 2 Erwachsene, waren trotz niedriger Serumkonzentrationen von C4 und CINH asymptomatisch.

Ribonuclease activity at pH 7.1 (alkaline ribonuclease) was determined in homogenates of rat superior cervical ganglion up to 5 days after postganglionic nerve injury under optimal conditions of assay. Measurements were performed in the presence and absence of the sulfhydryl blocking agent, N-ethylmaleimide, to assess the proportion of alkaline ribonuclease apparently bound to endogenous inhibitor. Total ribonuclease activity per ganglion was stimulated 1.3 fold by 1 day after injury and remained elevated over the 5 day period. Free ribonuclease activity accounted for about 60% of the observed increase in total activity at day 1, but had returned to control level by day 3. At day 3 the entire 90% increase in total activity was attributable to ribonuclease bound to endogenous inhibitor (i.e. latent activity). These changes are occurring at times after nerve injury when marked alterations in RNA turnover have been observed, implicating alkaline ribonucleases in the control of RNA metabolism during nerve regeneration.  相似文献

A study was made of the fluorescence induction curves from gently-broken spinach chloroplasts inhibited with DCMU. It was found that there were four kinetically different phases associated with such curves of which only the fastest did not appear to follow exponential kinetics. A comparison of the effects of various concentrations of DCMU on the rate of oxygen evolution and on the fluorescence induction curve did not support the hypothesis that any of the kinetic phases was simply an artefact caused by incomplete inhibition of electron transport. It was also found that 5 min of dark incubation did not maximally oxidize the electron acceptors to photosystem 2 since some acceptors were only oxidized following far-red illumination, suggesting a heterogeneity among these acceptors with respect to their re-oxidation properties. This article will contextualize ethnographic and clinicalfeatures that distinguish one particular alternativehealing method (Self-Acceptance Training) frommainstreampsychotherapeutic procedures. Factors common to manypsychotherapies are listed and a series of contrasts andcomparisons made by examining definitions of: (1) presentingproblems, (2) inciting events, (3) phasic development, (4)taxonomic classifications, (5) therapeutic interventions,and (6) prognostic formulations. In acid-sensitive watersheds of the northeastern US, decreases in SO2 emissions and atmospheric deposition of sulfur have not been accompanied by marked changes in pH and acid neutralizing capacity (ANC). To better understand this phenomenon, we investigated the long-term trends in soil solution (1984–1998) and stream water (1982–2000) chemistry along a natural soil catena at the Hubbard Brook Experimental Forest, New Hampshire, USA. Significant declines in strong acid anion concentrations were accompanied by declines in base cation concentrations in soil solutions draining the Oa and Bs soil horizons at all elevations. The magnitude of change varied with position in the landscape. Recovery, as indicated by increasing ANC (mean 2.38µEqL–1year–1) and decreasing concentrations of inorganic monomeric Al (mean 1.03µmolL–1year–1), was confined to solutions draining the Bs horizon at mid-to-higher elevations. However, persistently low Ca2+/Ali ratios (<1) in Bs soil solutions at these sites may be evidence of continuing Al stress to trees. In Bs soil solution at a lower elevation site and in Oa soil solutions at all sites, declines in base cations (mean 3.71µEqL–1year–1) were either similar to or exceeded declines in strong acid anions (mean 3.25µEqL–1year–1) resulting in no change in ANC. Changes in the chemistry of stream water reflected changes in soil solutions, with the greatest improvement in ANC occurring at high elevation and the rate of increase decreasing with decreases in elevation. The pH of soil solutions and stream waters either declined or did not change significantly. Therefore pH-buffering processes, including hydrolysis of Al and possibly the deprotonation of organic acids, have prevented increases in drainage water pH despite considerable reductions in inputs of strong acids.  相似文献

2,3-Di-O-phytanyl-1-O-glucopyranosylglycerol and polar derivatives of its 6-glucose moiety have been synthesized. The target molecule contains the diphytanyl-sn-glycerol moiety which is -linked to glucose. The key step in its synthesis involves the coupling of phytanyl bromide and isopropylidene threitol. We also demonstrated that the 6-hydroxyl group of glycolipids can be functionalized without protection of the sugar moiety.  相似文献

Summary The intracellular distributions of major muscle proteins, myosin, actin, tropomyosin, -actinin, and desmin, in smooth muscle cells of chicken gizzard at various stages of embryogenesis were investigated by immunofluorescence-labeling of enzyme-dispersed cells cultured up to three hours. These muscle proteins, except some part of myosin, were organized into fibrous structures as soon as synthesis and accumulation of proteins started. As for myosin, a considerable amount of it was dispersed in soluble cytoplasm as well. On the other hand, Ca++-dependent contractility was detected with detergent-extracted myoblasts and glycerinated tissue from embryos older than 7 days. Although the nascent myofibrils bear a resemblance to stress fibers, the former could be distinguished from the latter by their high stability in dispersed, spherical cells. The above findings, therefore, show that the synthesis of contractile proteins is followed by immediate assembly of them into functional myofibrils without undergoing any intermediate structure. Based on these findings, the mechanism of myofibril formation in developing smooth muscle cells is discussed.  相似文献

Several pieces of research suggest that leaf liquid-path conductance (K1) may be smaller in evergreen species (EG) than in deciduous species (DC), coexisting in tropical dry forests. This was investigated in four DC and two EG in a tropical dry forest in Venezuela. The K1 (kg m-2 MPa-1 s-1) were consistently lower in EG (8.4 × 10-5) than in DC (12 to 20 × 10-5). Differences in water potential (W) inside the leaves were calculated using K1 values in connection with maximum transpiration rates measured in the field. It was found that W was about 1.9 MPa in EG, intermediate for one DC (Pithecellobium dulce, 1.7 MPa) and minimum for other DC (1.0 to 1.3 MPa).  相似文献

Vegetation responses to climatic change can be studied retrospectively by utilizing the Quaternary fossil record. There has been controversy over the extent to which major changes in vegetation patterns at the continental scale lag behind the climatic changes that drive them, and to what extent vegetation can ever be said to be in equilibrium with climate. The equilibrium question has no single answer. The predominant mode of vegetation response to climatic change depends on the space and time frame and resolution of the data set in which the response is observed.Vegetation (as observed on particular space and time scales) can be in dynamic equilibrium with climate if its response time is sufficiently fast in relation to the rate of climatic change to which it is observed to be responding. Several processes can be involved in the response: successional, migrational, edaphic, and evolutionary. Successional response times can be deduced from forest succession models. The other processes are less well understood and different ideas exist concerning their rates. According to one hypothesis, migrational lags caused delays of thousands of years in the postglacial dynamics of forest composition. The alternative hypothesis explains these changes as dynamic equilibrium responses to changes in climatic seasonality and climatic anomaly patterns. Neither hypothesis need be universally true; gradient analysis and forest succession models are among the techniques that can be used in inferential tests of these hypotheses for particular space-time regions.Dynamic equilibrium may often be a reasonable approximation for the responses of the broadest continental-scale forest patterns to orbitally induced climatic changes. But as spatial and temporal frames of observation are diminished and resolution increased, biotic processes must eventually come to dominate. At sufficiently fine scales the main observable phenomena are successional responses to natural disturbance events. Summary The genomic stability of tissue culture regenerants of sugarcane (Saccharum spp. hybrids, cvs CP721210, CP68-1067 and B43-62) was analyzed by DNA restriction fragment length polymorphism (RFLP). Plants regenerated from calli, cell suspensions, cryopreserved cell suspensions and protoplasts were used. Total DNA isolated from 19 different sources was digested with EcoRI, HindIII, BamHI, BamHI, EcoRI and PstI and probed with six known maize mitochondrial genes (coxI, coxII, atpA, atp6, atp9 and rrn18-rrn5), three random maize mitochondrial cosmid clones, two random maize chloroplast cosmid clones and a wheat Nor locus clone. Hybridization patterns indicated that the variation observed was minor and appeared only in the secondcycle regenerants. No differences were observed among the three cultivars and the regenerants from calli, suspension culture, cryopreserved suspension culture and protoplasts. Mitochondrial DNA (mtDNA) isolated from CP72-1210 plants and its embryogenic cell suspensions, and bulk samples from all CP72-1210 regenerants pooled together were digested with EcoRI, HindIII, PstI, BamHI and SalI and probed with three recombinationally active wheat mtDNA clones, K, K3 and X2. No variation in the mtDNA restriction patterns was observed between the CP72-1210 plants and its regenerants. However, restriction pattern variation was observed only from EcoRI digestion, and hybridization patterns of K3, K and X2 revealed minor variations in the mtDNA of cell suspensions when compared with the DNA of the CP72-1210 plant. Except for a qualitative variation detected by the X2 probe and minor stoichiometric variations detected by the K3 probe, sugarcane DNAs were found to be stable after plant regeneration.Florida Agriculture Experiment Station Journal Series No. R-02703  相似文献

Summary The expression of wild type CFTR facilitates the efflux of neutral amino acids (Rotoli et al., Biochem. Biophys. Res. Commun. 204: 653–658, 1994); as a result, after an extensive depletion of intracellular amino acid pool obtained through an incubation in saline solution, the intracellular leucine levels were lower in murine C127 cells transfected with the wild type CF gene (C127 CFTRw/t) than in cells transfected with either mutant CF (C127 CFTRF508 cells) or mock vector only. No change in amino acid efflux was detected when C127 CFTRw/t and C127 CFTRw/t and C127 CFTRF508 cells were studied under conditions known to activate protein kinase A. Upon an incubation in Cl free medium, a permeant analogue of cAMP caused a marked cell depolarization of C127 CFTRw/t cells but not of C127 CFTRF508 cells, thus showing a functional expression of CFTR protein in the former cell line. However, we found that, upon a Cl free incubation and in the absence of exogenous cAMP, C127 CFTRw/t cells developed a marked hyperpolarization that was not detected in C127 CFTRF508 cells. A microsomal vesicle fraction (GV) markedly enriched by the Golgi marker enzyme latent inosine diphosphatase (IDPase) has been isolated from photoautotrophic suspension-cell protoplasts ofChenopodium rubrum L. Addition of ATP creates a substantial pH gradient across the GV membrane as measured by accumulation of acridine orange. The GV showed a density of 1.14 g·cm-3 by equilibrium density centrifugation on sucrose gradients. Coincidence of acridine-orange accumulation and IDPase activity was confirmed on Percoll gradients. Formation of the pH gradient half-saturates at 0.3 mM MgATP, peaks at pH 7, and is competitively inhibited by ADP (k i0.1 mM), but not by Pi; it is hardly inhibited by orthovanadate, quickly dissipated by monensink 2=18 nM), nigericin (k 1/2=25 nM), and sluggishly by N-ethylmaleimide (k 1/235 M). Inhibition by KNO3 (k 1/26.7 mM) is incomplete (60%). Uridine 5-diphosphate (UDP)-glucose, UDP-galactose, but not UDP-mannose and the pertinent sugars, dissipate the ATP-generated pH gradient (k 1/210–20 mM UDP-glucose; optimum pH at 7.8). This UDP-glucose activity is accompanied by release of Pi, but not of glucose or sucrose. UDP-glucoseinduced Pi release from the GV saturates (k 1/2=1 mM UDP-glucose; optimum pH at 7) and is completely inhibited by the anion-channel blocker 4,4-diisothiocyano-2,2-stilbene disulfonic acid (DIDS;k 1/2=140 M). The GV incorporates UDP-[U-14C]glucose into an acid-labile, alkaline-stable macromolecular compound; this process is like-wise inhibited by DIDS. The terminal steps of oxidative phosphorylation include transport of phosphate and ADP into the mitochondrial matrix, synthesis of ATP in the matrix, and transport of the product ATP into the cytosol where it can be utilized to perform cellular work. Three nuclear genome encoded membrane proteins, namely, the phosphate carrier (PHC), the adenine nucleotide carrier (ANT), and the ATP synthase complex, consisting of at least 13 individual subunits, catalyze these reactions. The locations of the and subunits of the mitochondrial ATP synthase complex and the mitochondrial phosphate carrier, PHC, on human chromosomes were determined using cloned rat liver cDNA as probes. Human homologues of the subunit are on chromosomes 9 and 18, the subunit are on chromosomes 10 and 14, and the PHC was localized to chromosome 12.  相似文献

A highly generalised (five classes) grouping of Holdridge life zones, has been used to derive predicted natural ecosystems for the present day climate using temperature and precipitation derived from two experiments undertaken with the NCAR Community Climate Model. These predictions differ from one another and both differ significantly from the prescribed classification groupings of ecosystem complexes used with a state-of-the-art land surface parameterization submodel, the Biosphere-Atmosphere Transfer Scheme. The highly generalised groupings show relatively little sensitivity to the temperature changes induced by doubling atmospheric CO2 but greater response when precipitation is also modified. All the doubled-CO2 scenarios predict increased desert areas although these future climatically-induced changes to the global-scale ecology are very much smaller than the extensive disturbances already caused by mankind's land clearance and poor agriculture. Land-use change rendered 13% of the Earth's land surface desert whereas the most pessimistic doubled-CO2 result gives rise to only a 2% increase in desert area.  相似文献

Summary While reported interactions of elongation factor-1 (EF-1) with various other molecules involved in protein biosynthesis are abundant, its interactions with major cytoskeletal proteins have not been as extensively examined. Major roles for EF-1 in cytoskeletal organization emerge from a review of such interactions within species as diverse as slime molds and mammals, sea urchins and higher plants. The regeneration potential of shoot tip, stem, leaf, cotyledon and root explants of two papaya cultivars (Carica papaya cv. Solo and cv. Sunrise) were studed. Callus induction of these two cultivars of papaya showed that the shoot tips and stems are most suitable for forming callus, while leaves, cotyledons and roots are comparatively difficult to induce callus. Callus induction also varied with the varities. Somatic embryogenesis was obtained from 3-month-old root cultures. A medium containing half strength of MS inorganic salts, 160 mg/l adenine sulfate, 1.0 mg/1 NAA, 0.5 mg/1 kinetin and 1.0 mg/1 GA3 was optimal for embryogenesis. The callus maintained high regenerative capacity after two years of culture on this medium.
