The changes in isozyme profiles of catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), and glutathione reductase (GR) during severe deactivation of total CAT activity by aminotriazole (AT) treatment were investigated in the leaves ofArabidopsis thaliana (Columbia ecotype) in relation to H2O2-mediated oxidative stress. In spite of striking deactivation of total CAT activity by 0.1 mM AT, there were no significant differences in H2O2 levels or total leaf soluble protein contents including a Rubisco in both the control and AT-treated leaves. On the other hand, one specific protein band (molecular mass, 66 kD) was observed on the SDS-gel from leaf soluble proteins whose staining intensity was strikingly enhanced by AT treatment for 6 h. However, this band disappeared at 12 h. In the native-gel assays of CAT, POD, APX and GR isozymes, AT remarkably inhibited the expression of the CAT1 isozyme with no effects on CAT2 and CAT3, and generally had no effect on POD isozyme profiles. However, AT stimulated the intensity of activities of pre-existing APX1 and GR1 isozymes. In particular, it induced a new synthesis of one GR isozyme. Therefore, these results collectively suggest that a striking deactivation of total CAT activity by AT inA. thaliana leaves largely results from the suppression of CAT1 isozyme, and that APX1, GR1, and a newly synthesized GR isozyme could complement the role of CAT1 to metabolize H2O2 into non-toxic water.  相似文献

Thermotolerance and related antioxidant enzyme activities induced by both heat acclimation and exogenous salicylic acid (SA) application were studied in grapevine (Vitis vinifera L. cv. Jingxiu). Heat acclimation and exogenous SA application induced comparable changes in thermotolerance, ascorbic acid (AsA), glutathione (GSH), and hydrogen peroxide (H2O2) concentrations, and in activities of the antioxidant enzymes superoxide dismutase (SOD), peroxidase (POD), glutathione reductase (GR), ascorbic peroxidase (APX) and catalase (CAT) in grape leaves. Within 1 h at 38 °C, free SA concentration in leaves rose from 3.1 μg g−1 FW to 19.1 μg g−1 FW, then sharply declined. SA application and heat acclimation induced thermotolerance were related to changes of antioxidant enzyme activities and antioxidant concentration, indicating a role for endogenous SA in heat acclimation in grape leaves.  相似文献

Following leaf application of salicylic acid (SA), calcium chloride, hydrogen peroxide and 6-benzylaminopurine (BA), Manila grass (Zoysia matrella) plants were exposed to day/night temperature of 7/2 °C for 120 h in a growth chamber. The lower content of malondialdehyde (MDA) and H2O2 and higher activities of ascorbate peroxidase (APX), guaiacol peroxidase (POD) and catalase (CAT) during exposure to low temperature in pre-treated plants in comparison with control plants demonstrated that these compounds improved the chilling tolerance of Manila grass.  相似文献

The effects of exogenously applied hydrogen peroxide on the antioxidant system of pea plants were investigated. Ten-day-old pea seedlings were sprayed with 2.5 mM H2O2 and 24 h later with 0.2 mM PQ. Samples were taken 0, 2 and 5 h after the start of illumination. The protective effect of H2O2 was evaluated by monitoring of parameters related to the damage caused by PQ. The treatment with PQ led to a severe leakage of electrolytes from leaf tissues. Malondialdehyde level increased in PQ treated plants, but remained unchanged in H2O2 pre-treated ones after 5 h of illumination. Increased catalase and glutathione-S-transferase activity was observed in pea plants treated with H2O2 and PQ. Ascorbate peroxidase activity decreased significantly after paraquat application, but pre-treatment with H2O2 prevented ascorbate peroxidase inhibition to some extent. Increased guaiacol peroxidase activity was detected after H2O2 application. PQ application caused a drastic decline in the levels of thiol-group bearing compounds, reduced glutathione and ascorbate, while the quantity of oxidized glutathione and dehydroascorbate were increased. The results presented on changes in enzymatic and nonenzymatic antioxidants suggest that preliminary H2O2 application to pea plants treated with PQ, alleviates the toxic effects of the herbicide.  相似文献

Effects of exogenous salicylic acid (SA) on plant growth, contents of Na, K, Ca and Mg, activities of superoxide dismutase (SOD), guaiacol peroxidase (GPX), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), glutathione reductase (GR) and catalase (CAT), and contents of ascorbate and glutathione were investigated in tomato (Lycopersicon esculentum L.) plants treated with 100 mM NaCl. NaCl treatment significantly increased H2O2 content and lipid peroxidation indicated by accumulation of thiobarbituric acid reactive substances (TBARS). A foliar spray of 1 mM SA significantly decreased lipid peroxidation caused by NaCl and improved the plant growth. This alleviation of NaCl toxicity by SA was related to decreases in Na contents, increases in K and Mg contents in shoots and roots, and increases in the activities of SOD, CAT, GPX and DHAR and the contents of ascorbate and glutathione.  相似文献

In the present investigation, we studied the possible potentiating effect of salicylic acid (SA) under Cd toxicity in Oryza sativa L. leaves. Cd treatments for 24 h reduced the shoot length, dry biomass and total chlorophyll content followed by high Cd accumulation in shoots. About 16 h presoaking with SA resulted in partial protection against Cd, as observed by minor changes in length, biomass and total chlorophyll. SA priming resulted in low Cd accumulation. Enhanced thiobarbituric acid reactive substances (TBARS), hydrogen peroxide (H2O2) and superoxide anion (O2 ) content were seen when Cd was applied alone, while under SA priming the extent of TBARS, H2O2 and O2 were significantly low, suggesting SA-regulated protection against oxidative stress. The antioxidant enzymes like Catalase (CAT), guaiacol peroxidase (GPx), glutathione reductase (GR) and superoxide dismutase (SOD) showed varied activities under Cd alone. CAT activity increased after Cd treatment, followed by a decline in GPX and GR activity. SOD also declined at the highest concentrations with an initial increase. Under SA-priming conditions, the efficiency of the antioxidant enzymes was significantly elevated. GPx and SOD activity showed significant increase in activity. The ascorbate activity increased after Cd treatment, followed by a decline in glutathione under SA-free condition. SA priming showed gradual increase in these non-enzymic antioxidants. Our results indicate that Cd-induced oxidative stress can be regulated by SA.  相似文献

Cd~(2+)胁迫对小桐子幼苗叶片抗氧化系统的影响   总被引:1,自引:0,他引:1  
以小桐子幼苗为材料,设置不同浓度CdCl_2处理,测定Cd~(2+)胁迫对小桐子幼苗叶片中可溶性蛋白、丙二醛(MDA)含量,以及5种抗氧化酶活性和2种抗氧化剂含量的变化,探讨镉胁迫对小桐子幼苗抗氧化系统的影响。结果表明:(1)Cd~(2+)胁迫导致小桐子幼苗叶片中可溶性蛋白含量降低、MDA含量增加;(2)随着镉胁迫时间的延长,幼苗叶片中愈创木酚过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、抗坏血酸专一性过氧化酶(APX)、谷胱甘肽还原酶(GR)等抗氧化酶活性表现出先升高然后降低的变化趋势;(3)幼苗叶片中还原型抗坏血酸(ASA)和还原型谷胱甘肽(GSH)含量随着胁迫时间延长而降低,但其中氧化型抗坏血酸(DHA)和氧化型谷胱甘肽(GSSG)含量则升高。研究表明,镉胁迫初期能诱导小桐子幼苗抗氧化系统活性显著增强,提高其抗氧化能力,但随着胁迫时间的延长,致使其抗氧化酶的活性和抗氧物质含量下降,植株遭受明显氧化胁迫,幼苗生长受到镉的严重毒害。  相似文献

The present study was performed to see the physiological role of cytosolic ascorbate peroxidase (APX) and its relationship to other enzymes involved in the H2O2 scavenging metabolism, and also to elucidate the regulation of APX expression in dark-grown radish (Raphanus sativus L. cv Taiwang) cotyledons. To do so, 3-amino-l,2,4-triazole (aminotriazole), a known specific inhibitor of catalase, was used to simulate a catalase-deficient phenomenon in cotyledons. Aminotriazole, in very low concetration (10-4 M), inhibited remarkably the development of catalase activity in cotyledons during dark germination. This inhibition of catalase by aminotriazole, however, did not result in any significant changes in the growth response and the H2O2 level of developing cotyledons. In addition, the development of guaiacol peroxidase (GPX) activity was also not significantly affected. Unlike GPX, cytosolic APX activity was induced rapidly and reached a 1.7-fold increase in aminotriazole treated cotyledons at day 7 after germination. However,in vitro incubation of cytosolic APX preparation from cotyledons with aminotriazole did not result in any significant change in activity. One cytosolic APX isozyme (APXa) band involved in this APX activation was predominantly intensified in a native polyacrylamide gel by activity staining assay. This means that this APXa isozyme seems to play a key role in the expression of cytosolic APX activity. On the other hand, 2-day-old control seedlings treated with exogenous 1 mM H2O2 for 1 h showed a significant increase of cytosolic APX acitivity even in the absence of aminotriazole. Also, 2 μM cycloheximide treatment substantially inhibited the increase of APX activity due to aminotriazole. Based on these results, we suggest that a radish cytosolic APX could probably be substituted for catalase in H2O2 removal and that the expression of APX seems to be regulated by a change of endogenous H2O2 level which couples to APX protein synthesis in a translation stage in cotyledons.  相似文献

The effect of varied zinc (Zn) supply on the activities of superoxide dismutase (SOD), ascorbate (AsA) peroxidase, glutathione (GSSG) reductase, catalase and guaiacol peroxidase was studied in leaves of bean (Phaseolus vulgaris) plants grown for 15 days in nutrient solution. Zinc deficiency severely decreased plant growth and the leaf concentrations of soluble protein and chlorophyll. Resupply of Zn to deficient plants for up to 72h restored protein concentrations more rapidly than chlorophyll and plant growth. With the exception of guaiacol peroxidase, the activities of all enzymes were significantly decreased by Zn deficiency, in particular GSSG reductase and SOD. Within 72h of resupplying Zn to deficient plants, the enzyme activities reached the level of the Zn sufficient plants. The results indicate severe impairment in the ability of Zn-deficient leaves to enzymically scavenge O2 - and H2O2. Consequences and reasons of this impairment are discussed in terms of photooxidation of chloroplast pigments and inhibition of the biosynthesis of the related scavenger enzyme proteins.  相似文献

The effect of acute ozone (O3) fumigation on isozyme patterns of superoxide dismutase (SOD), peroxidase (POD) and ascorbate peroxidase (APX) in mature (ML) and young leaves (YL) of two poplar clones, contrasting in O3-sensitivity was analysed. Untreated leaves of both the O3-sensitive (O3-S) clone Eridano of Populus deltoides×P. maximowiczii and the O3-resistant (O3-R) clone I-214 of P.×euramericana showed four distinct SOD isoforms with a relative mobility (Rf) of 0.54 (MnSOD), 0.60 (Cu/ZnSOD), 0.65 (unidentified), and 0.71 (Cu/ZnSOD). After O3-fumigation the activity of the SOD isoforms showed only quantitative variations with respect to control plants. In ML of untreated O3-R plants seven POD isoforms (Rf= 0.13, 0.19, 0.34, 0.59, 0.64, 0.70 and 0.75) were found, while in YL one isoform (Rf= 0.34) was undetected. Only three POD isoforms in both ML and YL of untreated O3-S plants were resolved. The electrophoretic pattern of POD in O3-S leaves was greatly modified by acute O3-fumigation with the appearance of new isoforms in both YL and ML and the disappearance of an isoform (Rf= 0.13) in YL. Additionally, O3-exposure induced the appearance of two APX isoforms in YL (Rf= 0.66 and 0.70), and one isoform in ML (Rf= 0.70) of the O3-S clone. By contrast, the activity of the three APX isoformes (Rf= 0.64, 0.70 and 0.76) detected in O3-R leaves showed only quantitative variation with respect to untreated plants. From these data it is concluded that: 1) in these poplar hybrids antioxidant enzyme activity is developmentally regulated and greatly affected by acute O3 stress treatments and 2) the different enzymes activity displayed by the two poplar clones, especially for POD and APX isoformes, could partly explain their distinct O3-sensitivity.  相似文献

The effects of Ca(NO3)2 stress on biomass production, oxidative damage, antioxidant enzymes activities and polyamine contents in roots of grafted and non-grafted tomato plants were investigated. Results showed that when exposed to 80 mM Ca(NO3)2 stress, the biomass production reduction in non-grafted plants was more significant than that of grafted plants. Under Ca(NO3)2 stress, superoxide anion radical (O2) producing rate, hydrogen peroxide (H2O2) and malondialdehyde (MDA) contents of non-grafted plants roots were significantly higher than those of grafted plants, however, nitrate (NO3 ), ammonium (NH4 +) and proline contents, superoxide dismutase (SOD, EC1.15.1.1), peroxidase (POD, EC1.11.1.7), catalase (CAT, EC1.11.1.6) and arginine decarboxylase (ADC, EC 4.1.1.19) activities of grafted plants roots were significantly higher than those of non-grafted plants. Regardless of stress, free, conjugated and bound polyamine contents in roots of grafted plants were significantly higher than those of non-grafted plants. The possible roles of antioxidant enzymes, prolines and polyamines in adaptive mechanism of tomato roots to Ca(NO3)2 stress were discussed. To access contributions of inductive responses of the antioxidant enzymes in the resistance to salt stress, activities of the enzymes were determined in the rice (Oryza sativaL. cv. Dongjin) plant. In the leaves of the rice plant, salt stress preferentially enhanced the content of H2O2 as well as the activities of the superoxide dismutase (SOD), ascorbate peroxidase (APX), and peroxidase specific to guaiacol, whereas it induced the decrease of catalase activity. On the other hand, salt stress had little effect on the activity levels of glutathione reductase (GR). In order to analyze the changes of antioxidant enzyme isoforms against salt stress, plant extracts were subjected to native PAGE. Leaves of the rice plant had two isoforms of Mn-SOD and five isoforms of Cu/Zn-SOD. Fe-SOD isoform was not observed in the activity gels. Expression of Cu/Zn-1, -2, and Mn-SOD-2 isoforms was preferentially enhanced by salt stress. Seven APX isoforms were presented in the leaves of the rice plants. The intensities of APX-4 to -7 were enhanced by salt stress, whereas those of APX-1 to -3 were minimally in changed response to salt stress. There were seven GR isoforms in the leaves of rice plants. Levels of activity for most GR isoforms did not change in the stressed plants compared to the control plants. On the other hand, the levels of activity for most antioxidant enzymes changed little in the roots of stressed plants compared to the control plants. These results collectively suggest that SOD leads to the overproduction of hydrogen peroxide in the leaves of rice plants subjected to salt stress: The overproduction of hydrogen peroxide functions as the signal of salt stress, which induces the induction of specific APX isoforms but not specific GR isoforms under catalase deactivation.  相似文献

The activities and isoenzyme pattern of catalase (CAT), superoxide dismutase (SOD) and peroxidase (POD) have been studied during germination of Chenopodium murale seeds. CAT and SOD activities were similar in dry seeds and during first 2 d of imbibition. CAT activity increased during radicle protrusion and early seedling development. The maximum SOD activity was found at final stages of germination and early seedling development. POD activity was not detected until the 6th day of germination, indicating POD involvement not until early seedling development. The effects of methyl jasmonate (MJ) and salicylic acid (SA) on changes of the activities of major antioxidant enzymes, superoxide anion accumulation (O2 ), ascorbate, total glutathione (TG), malondialdehyde (MDA) content and ginsenoside accumulation were investigated in ginseng roots (Panax ginseng L.) in 4 l (working volume) air lift bioreactors. Single treatment of 200 μM MJ and SA to P. ginseng roots enhanced ginsenoside accumulation compared to the control and harvested 3, 5, 7 and 9 days after treatment. MJ and SA treatment induced an oxidative stress in P. ginseng roots, as shown by an increase in lipid peroxidation due to rise in O2 accumulation. Activity of superoxide dismutase (SOD) was inhibited in MJ-treated roots, while the activities of monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), SOD, guaiacol peroxidase (G-POD), glutathione peroxidase (GPx) and glutathione reductase (GR) were induced in SA-treated roots. A strong decrease in the activity of catalase (CAT) was obtained in both MJ- and SA-treated roots. Activities of ascorbate peroxidase (APX) and glutathione S transferase (GST) were higher in MJ than SA while the contents of reduced ascorbate (ASC), redox state (ASC/(ASC+DHA)) and TG were higher in SA- than MJ-treated roots while oxidized ascorbate (DHA) decreased in both cases. The result of these analyses suggests that roots are better protected against the O2 stress, thus mitigating MJ and SA stress. The information obtained in this work is useful for efficient large-scale production of ginsenoside by plant-root cultures.  相似文献

Arsenic (As) toxicity and its biochemical effects have been mostly evaluated in ferns and a few higher plants. In this study, we investigated the effect of As (10.0 and 50.0 μM) on seedling growth, root anatomy, lipid peroxidation (malondialdehyde and conjugated dienes), electrolyte leakage, H2O2 content, root oxidizability and the activities of antioxidant enzymes in mung bean (Phaseolus aureus Roxb.). Arsenic significantly enhanced lipid peroxidation (by 52% at 50.0 μM As), electrolyte leakage and oxidizability in roots. However, there was no significant change in H2O2 content. Arsenic toxicity was associated with an increase in the activities of superoxide dismutase (SOD), guaiacol peroxidase (GPX) and glutathione reductase (GR). In response to 50.0 μM As, the activities of SOD and GR increased by over 60% and 90%, respectively. At 10.0 μM As, the activity of ascorbate peroxidase (APX) increased by 83%, whereas at 50.0 μM it declined significantly. The catalase (CAT) activity, on the other hand, decreased in response to As exposure, and it corresponded to the observed decrease in H2O2 content. We conclude that As causes a reduction in root elongation by inducing an oxidative stress that is related to enhanced lipid peroxidation, but not to H2O2 accumulation.  相似文献

When rice seedlings grown for 10 and 20 days were subjected to in vitro drought stress of −0.5 and −2.0 MPa for 24 h, an increase in the concentration of superoxide anion (O2.−), increased level of lipid peroxidation and a decrease in the concentration of total soluble protein and thiols was observed in stressed seedlings compared to controls. The concentration of H2O2 as well as ascorbic acid declined with imposition of drought stress, however glutathione (GSH) concentration declined only under severe drought stress. The activities of total superoxide dismutases (SODs) as well as ascorbate peroxidase (APX) showed consistent increases with increasing levels of drought stress, however catalase activity declined. Mild drought stressed plants had higher guaiacol peroxidase (GPX) and chloroplastic ascorbate peroxidase (c-APX) activity than control grown plants but the activity declined at the higher level of drought stress. The activities of enzymes involved in regeneration of ascorbate i.e. monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) were higher in drought stressed plants compared to controls. Results suggest that drought stress induces oxidative stress in rice plants and that besides SOD, the enzymes of ascorbate-glutathione cycle, which have not been studied in detail earlier under stressful conditions, appear to function as important component of antioxidative defense system under drought stress.  相似文献

The addition of 0.5 mM salicylic acid (SA) to the hydroponic growth solution of young maize (Zea mays L.) plants under normal growth conditions provided protection against subsequent low-temperature stress. This observation was confirmed by chlorophyll fluorescence parameters and electrolyte leakage measurements. In addition, 1 d of 0.5 mM SA pre-treatment decreased net photosynthesis, stomatal conductivity and transpiration at the growth temperature (22/20 °C). Since there was only a slight decrease in the ratio of variable to maximal fluorescence (Fv/Fm) the decrease in photosynthetic activity is not due to a depression in photosystem II. The analysis of antioxidant enzymes showed that whereas SA treatment did not cause any change in ascorbate peroxidase (EC 1.11.1.11) and superoxide dismutase (EC 1.15.1.1) activities, there was a decrease in catalase (EC 1.11.1.6) activity, and an increase in guaiacol peroxidase (EC 1.11.1.7) and glutathione reductase (EC 1.6.4.2) activities after the 1-d SA treatment at 22/20 °C. In native polyacrylamide gels there was, among the peroxidase isoenzymes, a band which could be seen only in SA-treated plants. It is suggested that the pre-treatment of maize plants with SA at normal growth temperature may induce antioxidant enzymes which lead to increased chilling tolerance. The effects of chemical and microbial elicitors such as β-aminobutyric acid (BABA), Salicylic acid (SA), and Pseudomonas fluorecens CHAO on hydrogen peroxide generation and activity of the enzymes related to its metabolism, i.e., superoxide dismutase (SOD), guaiacol peroxidase (GPOX), and catalase (CAT) were investigated in tomato roots infected with root-knot nematode (Meloidogyne javanica). Results of this study show that treating the tomato seedlings with the above elicitors significantly reduces the nematode infection level. Among the tested elicitors, BABA has reduced the nematode galls, number of egg masses per plant and number of eggs per individual egg mass more than the others. Additionally, the amount of H2O2, a product of oxidative stress, SOD and GPOX specific activities were significantly increased in the elicitor treated plants in comparison to control. Our observation shows that BABA also increases the H2O2 accumulation and the SOD and GPOX activities more as compared with the other tested elicitors. Such increases have occurred in two phases and maximum levels of them were observed at 5 days after treatment. In contrast with the increase in SOD and GPOX activities, the CAT activity doesnot show any significant increase in treated plants as compared with the control and other tested elicitors. It can be concluded that BABA, SA, and Pseudomonas fluorescens CHAO induce oxidative stress in tomato roots through generation of reactive oxygen species (ROS) and the enzymes related to their metabolism.  相似文献

Mountain environmental stresses result in increased formation of hydrogen peroxide (H2O2) and accumulation of malondialdehyde (MDA) in leaves of Polygonum viviparum. The activities of several antioxidative system enzymes such as superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), peroxidase (POD, EC 1.11.1.7), glutathione reductase (GR, EC 1.6.4.2), dehydroascorbate reductase (DHAR, EC 1.8.5.1) and the contents of several non-enzymatic antioxidants such as reduced form of ascorbate (ASC), dehydroascorbate (DHA), reduced glutathione (GSH), and oxidized glutathione (GSSG) were investigated in leaves of P. viviparum, which were collected from three altitudes (2,200, 3,200, and 3,900 m) of Tianshan Mountain in China. The activities of these four antioxidative enzymes were accompanied by increases of H2O2 levels from 2,200 to 3,200 m. However, the activities of CAT and POD were decreased, whereas the activities of SOD and GR continually increased at 3,900 m. Analyses of isoforms of SOD, CAT, POD, and GR showed that the leaves of P. viviparum exposed different altitude conditions are capable of differentially altering the intensity. Additionally, two new isoforms of SOD were detected at 3900 m. A continual increase in the ASC, ASC to DHA ratio, GSH and GSH/[GSH + GSSG] ratio, and the activity of DHAR were observed in leaves of P. viviparum with the elevation of altitude. These results suggest that the higher contents of ASC, GSH as well as an increase in reduced redox state may be essential to antioxidation processes in the leaves of P. viviparum, whereas antioxidant enzymes system is a cofactor in the processes.  相似文献

Effects of exogenous H2O2 application on vinblastine (VBL) and its precursors, vindoline (VIN), catharanthine (CAT) and α-3′,4′-anhydrovinblastine (AVBL), were measured in Catharanthus roseus seedlings in order to explore possible correlation of VBL formation with oxidative stress. VBL accumulation has previously been shown to be regulated by an in vitro H2O2-dependent peroxidase (POD)-like synthase. Experimental exposure of plants to different concentrations of H2O2 showed that endogenous H2O2 and alkaloid concentrations in leaves were positively elevated. The time-course variations of alkaloid concentrations and redox state, reflected by the concentrations of H2O2, ascorbic acid (AA), oxidative product of glutathione (GSSG) and POD activity, were significantly altered due to H2O2 application. The further correlation analysis between alkaloids and redox status indicated that VBL production was tightly correlated with redox status. These results provide a new link between VBL metabolisms and redox state in C. roseus.  相似文献
