20 populations of kale (B. oleracea var. acephala L.) selected from 127 populations for fresh consumption terms of yield and leaf quality characteristics as superior types using weight-based ranking method from the Black Sea Region of Turkey were evaluated at the DNA level using randomly amplified polymorphic DNA (RAPD) markers compared to some morphological characters. The 7 primers selected from 100 decamers used generated 110 bands, of which 60 (54.5%) were polymorphic. Jaccard's genetic distances were calculated and dendogram was generated using the UPGMA algorithm. The dendogram obtained were classified into three main groups and four subgroups. The accessions showed a limited clustering in compare to morphological characters such as the number of leaf, leaf intentation of the margin, leaf and midrib color and thickness of midrib than geographical characteristics. Leaf color and midrib thickness characters clustered in the same group as OR49 and G18 accessions; S20, G6 and OR37 accessions, respectively.  相似文献

A set of 30 accessions of five Curcuma species-C. latifolia, C. malabarica, C. manga and C. raktakanta and 13 morphotypes (identified on the basis of morphological markers) of C. longa conserved in the In Vitro Genebank at National Bureau of Plant Genetic Resources, New Delhi, were subjected to RAPD analysis. Of the 200 RAPD primers screened, 21 polymorphic primers were selected for further study. Mean genetic similarities based on Jaccard’s similarity coefficient ranged from 0.18 to 0.86 in accessions of cultivated species, i.e., C. longa and from 0.25 to 0.86 in wild species. The dendrogram derived from the RAPD data corroborated the morphological classification of the morphotypes. The efficiency of individual RAPD primers was also compared; primers OPC-20, OPO-06, OPC-01 and OPL-03 were adjudged highly informative in discriminating the germplasm of Curcuma.  相似文献

The DNA genetic diversity of 40 accessions of genus Leymus was analyzed by random amplified polymorphic DNA (RAPD) markers. A total of 352 products were amplified by 34 10-mer arbitrary primers, among which 337 products (95.74 %) were found to be polymorphic. 5–14 polymorphic bands were amplified by each polymorphic primer, with an average of 9.91 bands. The data of 352 RAPD bands were used to generate Jaccard’s similarity coefficients and to construct a dendrogram by means of UPGMA. Great genetic diversity in genus Leymus was observed, the genetic diversity among the different species more abundant than that of the different accessions, and the different accessions in a species or the species from the same areas were clustered together.  相似文献

The present study explores morphological, genetic and phytochemical composition of Bunium persicum populations belonging to high altitudinal areas of Indian Himalayan region. In total, 23 morphological traits (13 quantitative and 10 qualitative traits) and 32 Random Amplified Polymorphic DNA primers were employed to infer the population structure of the species. Of the fourteen populations, five genetically diverse populations were analyzed for phytochemical diversity. Among morphological traits, inflorescence, seed and branch traits were most significant in detecting variation. At molecular level, primers TIBMBA-06 and OPR-16 were found most polymorphic with respect to Polymorphism Information Content and Marker Index values. Dendrogram grouped all populations into two major clusters while population from Shong region out grouped separately showing its distantness from all other populations. STRUCTURE analysis was done by using Bayesian model, which characterised all populations into four clusters and some degree of admixture was also observed within individuals. Shong population showed distinct genetic makeup as also suggested by dendrogram. Phytochemical analysis showed the presence of 55 components, of which, 2-methyl-3-phenyl propanal, benzeneacetic acid, 1-phellandrene, γ-terpene, α-terpinolene, Δ0.3-carene and sabinene were major components in its essential oils. The present study revealed high genetic and phytochemical diversity in B. persicum accessions from north-western Himalayan regions. Specifically, accessions from Saptal regions were having higher quantity of essential oils and can be selected for cultivation to meet the commercial demand to some extent. Further, the diversity information provided herein can be useful in management and improvement of this species through future breeding programmes.  相似文献

Subgenus Cerasus species are useful genetic resources for cherry breeding programs. A total of 17 morphological traits together with 19 random amplified polymorphic DNA (RAPD) primers were used to study 39 accessions including 34 wild Cerasus subgenus genotypes belonging to Prunus avium L., P. cerasus L., P. mahaleb L., P. microcarpa Boiss., P. incana Pall., and P. brachypetala Boiss. species, along with an unknown wild Cerasus sample, two advanced cherry cultivars (‘Lambert’ and ‘Bulgar’), and two rootstocks (‘Colt’ and ‘Gisela 6’). Genotypes were separated into different groups according to their species and collection sites using cluster analysis performed by Ward’s clustering method based on morphological data. Nineteen RAPD primers from 60 screened produced 304 polymorphic reproducible bands (98.15% polymorphism). According to the similarity matrix, the lowest similarity was obtained between P. avium and P. microcarpa samples. A dendrogram was prepared by the unweighted pair-group method with arithmetic average (UPGMA), and the accessions were separated according to their species and geographic origin. In both morphological and molecular results, the advanced cultivars and rootstocks were separated from wild genotypes, and the unknown genotype was grouped with P. mahaleb accessions. Grouping by morphological characteristics was compared with the results of RAPD analysis, with no significant correlations between morphological and molecular data being found. This is the first report of molecular (RAPD) genetic diversity study in wild Cerasus subgenus genotypes from Iran, and the results demonstrate the high potential of RAPD analysis for discrimination of Cerasus subgenus genotypes.  相似文献

The aim of this study was to characterize 23 important Iranian sweet cherry (Prunus avium) cultivars collected from different provinces of Iran and 1 foreign cultivar, which was used as control, considered for breeding programs by using 21 microsatellite markers and 27 morphological traits. In sweet cherry (Prunus avium) accessions, leaf, fruit, and stone morphological characters were evaluated during two consecutive years. The study revealed a high variability in the set of evaluated sweet cherry accessions. The majority of important correlations were determined among variables representing fruit and leaf size and variables related to color. Cluster analysis distinguished sweet cherry accessions into two distinct groups. Principal component analysis (PCA) of qualitative and quantitative morphological parameters explained over 86.59% of total variability in the first seven axes. In PCA, leaf traits such as leaf length and width, and fruit traits such as length, width, and weight, and fruit flesh and juice color were predominant in the first two components, indicating that they were useful for the assessment of sweet cherry germplasm characterization. Out of 21 SSR markers, 16 were polymorphic, producing 177 alleles that varied from 4 to 16 alleles (9.35 on average) with a mean heterozygosity value of 0.82 that produced successful amplifications and revealed DNA polymorphisms. Allele size varied from 95 to 290 bp. Cluster analyses showed that the studied sweet cherry genotypes were classified into five main groups based mainly on their species characteristics and SSR data. In general, our results did not show a clear structuring of genetic variability within the Iranian diffusion area of sweet cherry, so it was not possible to draw any indications on regions of provenance delimitation. The results of this study contribute to a better understanding of sweet cherry genetic variations in Iran, thus making for more efficient programs aimed at preserving biodiversity and more rational planning of the management of reproductive material.  相似文献

Two species of the genus Galeopsis L., G. tetrahit L. and G. bifida Boenn. (family Lamiaceae), are problematic to distinguish often wrongly recognized, and treated by some taxonomists as a single species. Morphological diagnostical characters of these species are variable and partly overlap. Species independence of G. tetrahit and G. bifida was evaluated and their diagnostic characters verified using ISSR and RAPD markers. A total of 57 ISSR and 28 RAPD fragments were obtained providing distinct subdivision of the accessions examined into two groups. Analysis of molecular data using the neighbor-joining method showed that the accessions studied fell into two clades in the same way as demonstrated by the analysis of 20 morphological characters using single linkage method. These results suggest that G. tetrahit and G. bifida are distinct species. The morphological characters were found to be more variable compared to the molecular markers, although the combined of these characters provided differentiation of the species.  相似文献

Commiphora wightii is a medicinally important endangered species endemic to the Thar Desert of Rajasthan, India and adjoining areas of Pakistan. The populations of this species are declining sharply because of its extensive use as a natural herb. Random amplified polymorphic DNA analysis was conducted to find the genetic variation among 7 populations of C. wightii. Of the 100 random primers screened, 44 primers yielded 220 loci. Statistical analysis indicated low genetic diversity (H pop = 0.0958; I = 0.1498; mean polymorphic loci = 14.28%), and high genetic differentiation among the populations (G ST = 0.3990; AMOVA Φ ST of 0.3390; Bayesian θ (II) = 0.3002). The low genetic diversity may be due to geographic isolation and restricted gene flow (N m = 0.7533) between the fragmented populations. Unsustainable utilization of the plant has fragmented the population continuum which served the purpose of genetic exchange between populations. Mantel’s test was performed which revealed a highly significant positive correlation between genetic and geographic distance (r 2 = 0.614, P = 0.023) among the populations studied. Low variation can also be attributed to poor seed setting and the slow growth pattern of the species, which is also an apomict. In UPGMA dendrogram the Commiphora wightii samples were divided into two major and one minor cluster. These findings can serve as a guide to preserving the genetic resources of this medicinal plant species.  相似文献

A pea rust fungus, Uromyces viciae-fabae, has been classified into two varieties, var. viciae-fabae and var. orobi, based on differences in urediniospore wall thickness and putative host specificity in Japan. In principal component analyses, morphological features of urediniospores and teliospores of 94 rust specimens from Vicia, Lathyrus, and Pisum did not show definite host-specific morphological groups. In molecular analyses, 23 Uromyces specimens from Vicia, Lathyrus, and Pisum formed a single genetic clade based on D1/D2 and ITS regions. Four isolates of U. viciae-fabae from V. cracca and V. unijuga could infect and sporulate on P. sativum. Retrotransposons are retrovirus-related mobile sequences that have the potential to replicate via RNA intermediates and increase the genome size by insertion into new sites. The retroelement, Gret1, has been identified as playing a key role in generating fruit color variation in cultivated grape (Vitis vinifera L.) due to its insertion into the promoter of VvMybA1. Fruit color variation is an important distinguishing feature of cultivated grapes and virtually no fruit color variation is observed in wild grape species. The presence and relative copy number of Gret1 was assessed using quantitative PCR on 22 different Vitis species, only four of which (plus interspecific hybrids) are known to contain white accessions. Gret1 copy number was observed to vary by species as well as by color within species and was significantly higher in white-fruited accessions across all taxa tested. Additionally, genomic regions surrounding Gret1 insertion were sequenced in white V. vinifera, hybrid, V. labrusca, V. aestivalis, and V. riparia accessions.  相似文献

Twenty accessions of the sparsely cultivated diploid Musa cultivars Matti (AA) and Rasakadali (AB) were subjected to random amplified polymorphic DNA (RAPD) assay. A total of 14 random primers were used for the estimation of interand intracultivar variations. Out of 86 bands generated, 64 were polymorphic (74.4 % polymorphism). The cluster analysis grouped the cultivars into two major clusters: cluster I with 10 accessions of Matti and 2 of Rasakadali and cluster II comprising the remaining 8 accessions of Rasakadali. The coefficient of genetic similarity (GS) was from 0.73 to 0.99, suggesting low level of intercultivar variation. The accessions of Rasakadali with mean GS of 0.89 were genetically more diverse than those of Matti (GS = 0.93).  相似文献

Flemingia macrophylla (Willd.) Kuntze ex Merr., a multi-purpose legume with potential as dry-season forage crop, mainly occurs in subhumid to humid environments of tropical and subtropical Asia. Despite increasing interest in conservation of germplasm suitable for low-input production systems information on the genetic diversity of F. macrophylla is extremely scarce. The creation of baseline data is supposed to contribute to more efficient conservation management and to identify collecting strategies of novel germplasm. Random amplified polymorphic (RAPD) markers were used to investigate the genetic variation among 37 F. macrophylla accessions. Germplasm analysed in this study originated from Bac Kan province, Northeast Vietnam. Eight primers generated a total of 47 amplified RAPD loci of which 38 were polymorphic. Jaccard’s similarity coefficients among accessions ranged from 0.069 to 1 with a mean of 0.67. The UPGMA dendrogram revealed three clusters along with three outliers. No correspondence between geographic and genetic distance was found (Mantel test: R = 0.21; P = 0.016). Analysis of molecular variance (AMOVA) revealed significant (P < 0.001) differentiation between accessions collected in lowland and upland regions. Results of UPGMA clustering were confirmed by the pattern of principle coordinates analysis (PCO) plotting. Future collecting strategies should target populations at large distances and along the altitudinal range. Ex situ conservation should encompass those accessions that showed genetic divergence. In situ conservation may consist of establishing a system of interconnected population fragments to guarantee continuing genetic exchange via corridors and of rehabilitating degraded habitats.  相似文献

Galium catalinense (Rubiaceae) is a perennial shrub consisting of two subspecies endemic to California’s Channel Islands: Galium catalinense subsp. catalinense on Santa Catalina Island, and G. catalinense subsp. acrispum, a state-endangered taxon on San Clemente Island. A long history of overgrazing by introduced herbivores has contributed to population declines in G. catalinense subsp. acrispum. We surveyed 12 populations throughout the taxon’s range for genetic variation using eight polymorphic microsatellite loci to determine the genetic impact of this demographic bottleneck. At the taxon level, 65 alleles were identified with an average of 8.1 alleles per locus, although many alleles were rare; the effective number of alleles per locus averaged 2.6. Expected heterozygosity was 0.550. Individual populations had between six and eight polymorphic loci, with expected heterozygosities ranging from 0.36 to 0.60, and effective numbers of alleles ranging from 1.8 to 3.5 per locus. Populations fell into three or four genetic clusters, depending on type of analysis, which may represent refugia where the populations persisted during intense herbivory. There is little evidence of genetic bottlenecks or substantial inbreeding within populations. These findings, coupled with indications of recent migration between populations, suggest that G. catalinense subsp. acrispum is currently unlikely to be endangered by genetic factors, but small population sizes make the taxon vulnerable to future loss of genetic diversity. Management strategies based on these genetic data, population sizes, and the spatial distribution of populations are discussed.  相似文献

Okra is an important vegetable crop that provides a significant portion of vitamins and minerals for populations in several countries. Okra has been cultivated in Turkey for centuries, and was likely introduced by the Arabs from Africa in ancient times. In this study, we aimed to clarify the genetic variation within 35 Turkish okra germplasm, by comparing it against 25 different genotypes from India, Africa, and the United States, using 30 morphological characters and 19 sequence-related amplified polymorphism (SRAP) primer combinations. Fruit, leaf, and stem color were the primary characteristics to distinguish the okra accessions. Those features, among 30 individual phenotypic traits, explained 42 % of phenotypic variation in the first three axes of the principal component analysis (PCA) with leaf shape, flower size, cotyledon length, fruit-surface angularity, cotyledon width, and petal color. Phenotypic observation results showed that while 1051 Togo (10.76), Red Wonder (7.99), TR-05-1 (7.2), 1159 Togo (7.17), and Red Balady (7.15) were found to be more divergent accessions, Cajun Queen (5.06), Perkins Spineless (5.09), Jade (5.18), TR-01-1 (5.2), and DLGG (5.32) were the closest okra accessions. According to phenotypic data, Turkish okra accessions were located adjacent to the Indian, American, and African okra accessions in clusters three and four. However, marker data showed that African okra possessed a more distinct form compared to the other okra germplasms. Nineteen SRAP primer combinations produced 92 bands and 29 (31.5 %) of them were found to be polymorphic among okra accessions. 1051 Togo was found to be the most divergent accession in phenotypic observation.  相似文献

Cabbage (Brassica oleracea var. capitata L.) is one of the most popular cultivated vegetables worldwide. Cabbage has rich phenotypic diversity, including plant height, head shape, head color, leaf shape and leaf color. Leaf color plays an important role in cabbage growth and development. At present, there are few reports on fine mapping of leaf color mutants in B. oleracea. In this study, a naturally occurring yellow-green leaf cabbage mutant (YL-1), derived from the self-pollinated progenies of the hybrid ‘Hosom’, was used for inheritance analysis and gene mapping. Segregation populations including F2 and BC1 were generated from the cross of two inbred lines, YL-1 and 01–20. Genetic analysis with the F2 and BC1 populations demonstrated that the yellow-green leaf color was controlled by a single recessive nuclear gene, ygl-1. Insertion–deletion (InDel) markers, designed based on the parental re-sequencing data, were used for the preliminary mapping with BSA (bulked segregant analysis) method. A genetic map constructed with 15 InDels indicated that ygl-1 was located on chromosome C01. The ygl-1 gene is flanked by InDel markers ID2 and M8, with genetic distances of 0.4 cM and 0.35 cM, respectively. The interval distance between two markers is 167 kb. Thus, it enables us to locate the ygl-1 gene for the first time in B. oleracea. This study lays the foundation for candidate gene prediction and ygl-1gene cloning.  相似文献

We analyzed variability of morphological characters and genetic polymorphism of inter-simple sequence repeat (ISSR) markers in nine natural populations of three Lotus species from Eastern Europe, aiming to provide insights into the nature of the species L. ucrainicus. Nuclear ribosomal internal transcribed spacer (nrITS) was used as an additional molecular marker for a small subset of accessions. Analysis of variance, and principal coordinate and principal component analyses were applied for morphological data study. Cluster analysis [unweighted pair-group method with arithmetic mean (UPGMA)], principal coordinate analysis, and analysis of population genetic structure were used for ISSR pattern study. Morphological and genetic (ISSR, nrITS) evidence suggested hybrid origin of L. × ucrainicus as a result of hybridization between tetraploid species L. corniculatus and diploid species L. stepposus. We conclude that L. × ucrainicus may represent a case of hybrid speciation in statu nascendi, occurring before our very eyes.  相似文献

The genetic variation within and among seven Tunisian natural Hypericum humifusum L. populations belonging to three bioclimatic zones (sub-humid, upper semi-arid, and lower semi-arid) was assessed using random amplified polymorphic DNA markers. Eight selected primers produced a total of 166 bands, of which 153 were polymorphic. The genetic diversity within a population, based on Shannon’s index and percentage of polymorphic loci, was relatively high. The level of variation among populations did not differ significantly. However, the variation among populations grouped according to their bioclimates was significant.  相似文献

This research was conducted to study the genetic variation among eighteen genotypes of sesame (Sesamum indicum L.) collected from various agro-climatic regions of Iran along with six exotic genotypes from the Asian countries using both agro-morphological and ISSR marker traits. The results showed significant differences among genotypes for all agro-morphological traits and a relatively high genetic coefficient of variation observed for number of fruiting branches per plant, capsules per plant, plant height and seed yield per plant. Cluster analysis based on these traits grouped the genotypes into five separate clusters. Larger interthan intra cluster distances implies the presence of higher genetic variability between the genotypes of different groups. Genotypes of two clusters with a good amount of genetic divergence and desirable agronomic traits were detected as promising genotypes for hybridization programs. The 13 ISSR primers chosen for molecular analysis revealed 170 bands, of which 130 (76.47%) were polymorphic. The generated dendrogram based on ISSR profiles divided the genotypes into seven groups. A principal coordinate analysis confirmed the results of clustering. The agro-morphological traits and ISSR markers reflected different aspects of genetic variation among the genotypes as revealed by a non significant cophenetic correlation in the Mantel test. Therefore the complementary application of both types of information is recommended to maximize the efficiency of sesame breeding programs. The discordance among diversity patterns and geographical distribution of genotypes found in this investigation implies that the parental lines for hybridization should be selected based on genetic diversity rather than the geographical distribution.  相似文献

The aim of this study was to investigate natural serpentine and non-serpentine populations of Teucrium polium aggr. and to document the differences in their morphological traits, as well as estimate which characters are most likely contributing to differentiation of the populations. Nine populations distributed both on and off serpentine soils were investigated, and 12 different morphological features of 270 individuals were studied by univariate and multivariate statistical analyses. The results did not demonstrate clear delimitation of the serpentine from the non-serpentine populations. Using these ecotypes as classification factors in the discriminant analysis, it can be stated that stem height, leaf length in the fifth leaf pair, internode length between the fourth and the fifth leaf pairs, and corolla tube length are features with the greatest discriminant ability. The character variation was generally higher for the vegetative features, while reproductive features failed to demonstrate clear differences. The similarity groupings indicated by the cluster analysis were synchronous with groups distinguished by both the discriminant and PCA analyses. Significant differences were observed between sets of vegetative characters in all populations investigated.   相似文献
