Metabolism of ammonia (NH3) and hydroxylamine (NH2OH) by wild-type and a nitrite reductase (nirK) deficient mutant of Nitrosomonas europaea was investigated to clarify the role of NirK in the NH3 oxidation pathway. NirK-deficient N. europaea grew more slowly, consumed less NH3, had a lower rate of nitrite (NO2 ) production, and a significantly higher rate of nitrous oxide (N2O) production than the wild-type when incubated with NH3 under high O2 tension. In incubations with NH3 under low O2 tension, NirK-deficient N. europaea grew more slowly, but had only modest differences in NH3 oxidation and product formation rates relative to the wild-type. In contrast, the nirK mutant oxidized NH2OH to NO2 at consistently slower rates than the wild-type, especially under low O2 tension, and lost a significant pool of NH2OH–N to products other than NO2 and N2O. The rate of N2O production by the nirK mutant was ca. three times higher than the wild-type during hydrazine-dependent NO2 reduction under both high and low O2 tension. Together, the results indicate that NirK activity supports growth of N. europaea by supporting the oxidation of NH3 to NO2 via NH2OH, and stimulation of hydrazine-dependent NO2 reduction by NirK-deficient N. europaea indicated the presence of an alternative, enzymatic pathway for N2O production.  相似文献

The impacts of various nitrogen sources, i.e. NO 3, NH4 + or NH4NO3 in combination with gaseous NH3, on nitrogen-, carbon- and water-use efficiency and 13C discrimination (δ13C) by plants of the C3 species Triticum aestivum L. (wheat) and the C4 species Zea mays L. (maize) were studied. Triticum aestivum and Z. mays were hydroponically grown with 2 mol · m−3 of N supplied as NO 3, NH4 + or NH4NO3 for 21 and 18 d, respectively, and thereafter exposed to gaseous NH3 at 320 μg · m−3 or to ambient air for 7 d. In T. aestivum and Z. mays over a 7-d growth period, nitrogen-use efficiency (NUE) values were influenced by N-sources in the decreasing order NH4NO3-N > NO 3-N > NH4 +-N and NO 3-N > NH4NO3-N > NH4 +-N, respectively. Fumigation with NH3 decreased the NUE values of plants grown with any of the N-forms. During 28- and 7-d growth periods, N-sources affected water-use efficiency (WUE) values in the decreasing order of NH4 +-N > NO 3-N≈NH4NO3-N in non-fumigated T. aestivum, while fumigation with NH3 increased the WUE of NO 3-grown plants. There were insignificant effects of N-sources on WUE values of Z. mays over 25- and 7-d growth periods. Furthermore, δ13C values in plant tissues (leaves, stubble and roots) were higher (less negative) in NH4 +-grown plants of T. aestivum and Z. mays than in those supplied with NH4NO3 or NO 3. Regardless of the N-form supplied to the roots of the plant species, exposure to NH3 caused more-positive δ13C values in the plant tissues. These results indicate that the variations in N-source were associated with small but significant variations in δ13C values in plants of T. aestivum and Z. mays. These differences in δ13C values are in the direction expected from differences in WUE values over long or short growth periods and with differences in the extent of non-Rubisco (ribulose-1,5-bisphosphate carboxylase-oxygenase, EC 4.1.1.39) carboxylate contribution to net C acquisition, as a function of N-source. Sampling spatial and temporal variation in soil nitrogen availability   总被引:18,自引:0,他引:18  
There are few studies in natural ecosystems on how spatial maps of soil attributes change within a growing season. In part, this is due to methodological difficulties associated with sampling the same spatial locations repeatedly over time. We describe the use of ion exchange membrane spikes, a relatively nondestructive way to measure how soil resources at a given point in space fluctuate over time. We used this method to examine spatial patterns of soil ammonium (NH+ 4) and nitrate (NO 3) availability in a mid-successional coastal dune for four periods of time during the growing season. For a single point in time, we also measured soil NH+ 4 and NO 3 concentrations from soil cores collected from the mid-successional dune and from an early and a late successional dune. Soil nitrogen concentrations were low and highly variable in dunes of all ages. Mean NH+ 4 and NO 3 concentrations increased with the age of the dune, whereas coefficients of variation for NH+ 4 and NO 3 concentrations decreased with the age of the dune. Soil NO 3 concentration showed strong spatial structure, but soil NH+ 4 concentration was not spatially structured. Plant-available NH+ 4 and NO 3 showed relatively little spatial structure: only NO 3 availability in the second sampling period had significant patch structure. Spatial maps of NH+ 4 and NO 3 availability changed greatly over time, and there were few significant correlations among soil nitrogen availability at different points in time. NO 3 availability in the second sampling period was highly correlated (r = 0.90) with the initial soil NO 3 concentrations, providing some evidence that patches of plant-available NO 3 may reappear at the same spatial locations at irregular points in time. Nitrite (NO2 ) is an intermediate in a variety of soil N cycling processes. However, NO2 dynamics are often not included in studies that explore the N cycle in soil. Within the presented study, nitrite dynamics were investigated in a Nothofagus betuloides forest on an Andisol in southern Chile. We carried out a 15N tracing study with six 15N labeling treatments, including combinations of NO3 , NH4 + and NO2 . Gross N transformation rates were quantified with a 15N tracing model in combination with a Markov chain Monte Carlo optimization routine. Our results indicate the occurrence of functional links between (1) NH4 + oxidation, the main process for NO2 production (nitritation), and NO2 reduction, and (2) oxidation of soil organic N, the dominant NO3 production process in this soil, and dissimilatory NO3 reduction to NH4 + (DNRA). The production of NH4 + via DNRA was approximately ten times higher than direct mineralization from recalcitrant soil organic matter. Moreover, the rate of DNRA was several magnitudes higher than the rate of other NO3 reducing processes, indicating that DNRA is able to outcompete denitrification, which is most likely not an important process in this ecosystem. These functional links are most likely adaptations of the microbial community to the prevailing pedo-climatic conditions of this Nothofagus ecosystem.  相似文献

The loss of nitrogen (N) from field-applied animal manure through ammonia (NH3) volatilisation and nitrous oxide (N2O) emission is of major environmental concern. Both lime and dicyandiamide (DCD) have been suggested as amendments that can mitigate N2O emissions, but simultaneously increase the risk of NH3 volatilisation. This study evaluated the impact of lime and DCD on NH3 and N2O emissions following application of liquid hog manure. Hydrated lime (Ca(OH)2) was added to an acidic soil to achieve three pH levels (4.7, 6.3 and 7.4). Soil samples (100 g) were then placed in 500 ml screw-top Mason-jars and de-ionised water was added to bring the samples to 50, 70 and 90% water-filled pore space (WFPS). Slurry was applied at a rate equivalent to 116,000 l ha−1, while DCD was applied at 30% of the NH4-N rate applied. Jars were sealed and incubated at 21°C for 21 d. Ammonia volatilisation was quantified using boric acid traps, while N2O gas concentration was analysed using gas chromatography. Dicyandiamide had no effect (P>0.05) on either NH3 or N2O emissions. Both NH3 and N2O emissions increased (P<0.05) as WFPS increased, with emissions ranging from 0.9 to 1.4 kg NH3-N ha−1 and 123 to 353 g N2O-N ha−1, respectively. Liming decreased (P<0.01) N2O emissions from 547 to 46 g N2O-N ha−1, but increased (p<0.01) NH3 volatilisation from 0.36 to 1.92 kg NH3-N ha−1. Results suggest that liming to a pH ≥6.3 can reduce N2O emissions, however, this reduction will be accompanied by a substantial loss of NH3. Section Editor: H. Lambers  相似文献

Controlled cultivation of marine macroalgal biomass such as Ulva species, notably Ulva lactuca, is currently studied for production of biofuels or functional food ingredients. In a eutrophic environment, this macrophyte is exposed to varying types of nutrient supply, including different and fluctuating levels of nitrogen sources. Our understanding of the influences of this varying condition on the uptake and growth responses of U. lactuca is limited. In this present work, we examined the growth response of U. lactuca exposed to different sources of nitrogen (NH4+; NO3; and the combination NH4NO3) by using photo-scanning technology for monitoring the growth kinetics of U. lactuca. The images revealed differential increases of the surface area of U. lactuca disks with time in response to different N-nutrient enrichments. The results showed a favorable growth response to ammonium as the nitrogen source. The NH4Cl and NaNO3 rich media (50 μM of N) accelerated U. lactuca growth to a maximum specific growth rate of 16.4 ± 0.18% day−1 and 9.4 ± 0.72% day−1, respectively. The highest biomass production rate obtained was 22.5 ± 0.24 mg DW m−2·day−1. The presence of ammonium apparently discriminated the nitrate uptake by U. lactuca when exposed to NH4NO3. Apart from showing the significant differential growth response of U. lactuca to different nitrogen sources, the work exhibits the applicability of a photo-scanning approach for acquiring precise quantitative growth data for U. lactuca as exemplified by assessment of the growth response to two different N-sources.  相似文献

Animal waste causes environmental problems like eutrophication of ground and surface water or the pollution of the atmosphere because of its high NH4 + content. The aim of our study was to fix the nitrogen of swine waste as biomass. Therefore, an isolated alga, Chlorella sp., and bacteria naturally living in liquid manure were grown in batch cultures (containing diluted swine waste supplied with a nutrient solution) and continuous cultures (undiluted liquid manure) to achieve reduction of NH4 + and total organic carbon (TOC) contents. For continuous cultivation, a photobioreactor of our own design was used. The batch cultivation of Chlorella sp. and bacteria in swine waste resulted in good growth of both groups of organisms and in a reduction of 25% NH4 + and 80% TOC. In the continuous cultivation a steady state was not achieved owing to a change in the composition of the bacterial population. NH4 + was totally removed, but NO2 (up to 100 mM) was transiently released. NO3 was not detected. These effects might be explained by the presence of heterotrophic nitrifiers, which are able to oxidize NH4 + to NO2 and to reduce NO2 to gaseous compounds. Long-term exposure of native vegetation to elevated atmospheric CO2 concentrations is expected to increase C inputs to the soil and, in ecosystems with seasonally dry periods, to increase soil moisture. We tested the hypothesis that these indirect effects of elevated CO2 (600 μl l−1 vs 350 μl l−1) would improve conditions for microbial activity and stimulate emissions of nitrous oxide (N2O), a very potent and long-lived greenhouse gas. After two growing seasons, the mean N2O efflux from monoliths of calcareous grassland maintained at elevated CO2 was twice as high as that measured from monoliths maintained at current ambient CO2 (70 ± 9 vs 37 ± 4 μg N2O m−2 h−1 in October, 27 ± 5 vs 13 ± 3 μg N2O m−2 h−1 in November after aboveground harvest). The higher N2O emission rates at elevated CO2 were associated with increases in soil moisture, soil heterotrophic respiration, and plant biomass production, but appear to be mainly attributable to higher soil moisture. Our results suggest that rising atmospheric CO2 may contribute more to the total greenhouse effect than is currently estimated because of its plant-mediated effects on soil processes which may ultimately lead to increased N2O emissions from native grasslands. Aerobic N2O production was studied in nitrifying humus from urea-fertilized pine forest soil. Acetylene and nitrapyrin inhibited both NH4+ oxidation and N2O production, indicating that N2O production was closely associated with autotrophic NH4+ oxidation. N2O production was enhanced by low soil pH; it was negligible above pH 4.7. When soil pH decreased from 4.7 to 4.1, the relative amount of N2O-N produced from NH4+-N oxidized increased exponentially to 20%. There was also some evidence that N2O formation was stimulated by salts (potassium sulfate and sodium phosphates). The maximum rate of N2O-N production was 0.17 μg of N2O-N per g of soil per h. When humus was treated with NO2, N2O evolved immediately, indicating chemical formation, but no N2O was formed on the addition of NO3. The amount of N2O-N evolved was 0.6 to 4.6% of NO2-N added. A high concentration of NO2 and low soil pH enhanced chemical production of N2O. There was no accumulation of NO2 during nitrification. The calculations indicated that chemical formation of N2O was not the main source of N2O during NH4+ oxidation. After the addition of inhibitors of NH4+ oxidation the soils contained NO3, but no N2O was produced. The results suggest that enhanced autotrophic NH4+ oxidation is a potential source of N2O in fertilized acid forest soil.  相似文献

Increases in the supply of atmospheric CO2 and N are expected to alter the carbon cycle, including CH4 emissions, in boreal peatlands. These effects were studied in a glasshouse experiment with peat monoliths cored from an oligotrophic pine fen. The cores with living plants were kept in 720 ppmv and 360 ppmv CO2 atmospheres for about 6 months under imitated natural temperature cycle. Fertilisation with NH4NO3 (3 g m−2 for 25 weeks) was applied to 18 of the 36 monoliths. The rate of CH4 flux was non-linearly dependent on the number of Eriophorum vaginatum shoots growing in the monoliths, probably due to the gas transport properties of the aerenchyma. The average CH4 efflux rate, standardised by the number of shoots, was increased by a maximum of 10–20% in response to the raised CO2 level. In the raised-NH4NO3 treatment, the increase in CH4 release was lower. The effect of combined CO2+NH4NO3 on CH4 release was negligible and even lower than in the single treatments. Both potential CH4 production and oxidation rates at 5, 15 and 25°C were higher near the surface than at the bottom of the core. As expected, the rates clearly depended on the incubation temperature, but the different treatments did not cause any consistent differences in either CH4 production or oxidation. The determination of potential CH4 production and oxidation in the laboratory is evidently too crude a method of differentiating substrate-induced differences in CH4 production and oxidation in vivo. These results indicate that an increase in atmospheric CO2 or N supply alone, at least in the short term, slightly enhances CH4 effluxes from boreal peatlands; but together their effect may even be restrictive. Massive anthropogenic acceleration of the global nitrogen (N) cycle has stimulated interest in understanding the fate of excess N loading to aquatic ecosystems. Nitrate (NO3 ) is traditionally thought to be removed mainly by microbial respiratory denitrification coupled to carbon (C) oxidation, or through biomass assimilation. Alternatively, chemolithoautotrophic bacterial metabolism may remove NO3 by coupling its reduction with the oxidation of sulfide to sulfate (SO4 2−). The NO3 may be reduced to N2 or to NH4 +, a form of dissimilatory nitrate reduction to ammonium (DNRA). The objectives of this study were to investigate the importance of S oxidation as a NO3 removal process across diverse freshwater streams, lakes, and wetlands in southwestern Michigan (USA). Simultaneous NO3 removal and SO4 2− production were observed in situ using modified “push-pull” methods in nine streams, nine wetlands, and three lakes. The measured SO4 2− production can account for a significant fraction (25–40%) of the overall NO3 removal. Addition of 15NO3 and measurement of 15NH4 + production using the push–pull method revealed that DNRA was a potentially important process of NO3 removal, particularly in wetland sediments. Enrichment cultures suggest that Thiomicrospira denitrificans may be one of the organisms responsible for this metabolism. These results indicate that NO3 -driven SO4 2− production could be widespread and biogeochemically important in freshwater sediments. Removal of NO3 by DNRA may not ameliorate problems such as eutrophication because the N remains bio-available. Additionally, if sulfur (S) pollution enhances NO3 removal in freshwaters, then controls on N processing in landscapes subject to S and N pollution are more complex than previously appreciated. Ethanol-precipitated substances after fermentation of various agro-industrial wastes by Aureobasidium pullulans were examined for their pullulan content. Grape skin pulp extract, starch waste, olive oil waste effluents and molasses served as substrates for the fermentation. A glucose-based defined medium was used for comparison purposes. Samples were analysed by an enzyme-coupled assay method and by high-performance anion-exchange chromatography with pulsed amperometric detection after enzymic hydrolysis with pullulanase. Fermentation of grape skin pulp extract gave 22.3 g l−1 ethanol precipitate, which was relatively pure pullulan (97.4% w/w) as assessed by the coupled-enzyme assay. Hydrolysed starch gave only 12.9 g l−1 ethanol precipitate, which increased to 30.8 g l−1 when the medium was supplemented with NH4NO3 and K2HPO4; this again was relatively pure pullulan (88.6% w/w). Molasses and olive oil wastes produced heterogeneous ethanol-precipitated substances containing small amounts of pullulan, even when supplemented with nitrogen and phosphate. Overall, grape skin pulp should be considered as the best substrate for pullulan production. Starch waste requires several hydrolyis steps to provide a usable carbon source, which reduces its economic attraction as an industrial process. The influence of (NH4)2SO4 concentration and dilution rate (D) on actual and potential H2 photoproduction has been studied in ammonium-limited chemostat cultures of Rhodobacter capsulatus B10. The actual H2 production in a photobioreactor was maximal (approx. 80 ml h−1 l−1) at D = 0.06 h−1 and 4 mM (NH4)2SO4. However, it was lower than the potential H2 evolution (calculated from hydrogen evolution rates in incubation vials), which amounted to 100–120 ml h−1 l−1 at D = 0.03–0.08 h−1. Taking into account the fact that H2 production in the photobioreactor under these conditions was not limited by light or lactate, another limiting (inhibiting) factor should be sought. One possibility is an inhibition of H2 production by the H2 accumulated in the gas phase. This is apparent from the non-linear kinetics of H2 evolution in the vials or from its inhibition by the addition of H2; initial rates were restored in both cases after the vials had been refilled with argon. The actual H2 production in the photobioreactor at D = 0.06 h−1 was shown to increase from approximately 80 ml h−1 l−1 to approximately 100 ml h−1 l−1 under an argon flow at 100 ml min−1. Under maximal H2 production rates in the photobioreactor, up to 30% of the lactate feedstock was utilised for H2 production and 50% for biomass synthesis. The influence of ammonia on the anaerobic degradation of peptone by mesophilic and thermophilic populations of biowaste was investigated. For peptone concentrations from 5 g l−1 to 20 g l−1 the mesophilic population revealed a higher rate of deamination than the thermophilic population, e.g. 552 mg l−1 day−1 compared to 320 mg l−1 day−1 at 10 g l−1 peptone. The final degree of deamination of the thermophilic population was, however, higher: 102 compared to 87 mg NH3/g peptone in the mesophilic cultures. If 0.5–6.5 g l−1 ammonia was added to the mesophilic biowaste cultures, deamination of peptone, degradation of its chemical oxygen demand (COD) and formation of biogas were increasingly inhibited, but no hydrogen was formed. The thermophilic biowaste cultures were most active if around 1 g ammonia l−1 was present. Deamination, COD degradation and biogas production decreased at lower and higher ammonia concentrations and hydrogen was formed in addition to methane. Studies of the inhibition by ammonia of peptone deamination, COD degradation and methane formation revealed a K i (50%) for NH3 of 92, 95 and 88 mg l−1 at 37 °C and 251, 274 and 297 mg l−1 at 55 °C respectively. This indicated that the thermophilic flora tolerated significantly more NH3 than the mesophilic flora. In the mesophilic reactor effluent 4.6 × 108 peptone-degrading colony-forming units (cfu)/ml were culturable, whereas in the thermophilic reactor effluent growth of only 5.6 × 107 cfu/ml was observed. Natural 15N abundance measurements of ecosystem nitrogen (N) pools and 15N pool dilution assays of gross N transformation rates were applied to investigate the potential of δ15N signatures of soil N pools to reflect the dynamics in the forest soil N cycle. Intact soil cores were collected from pure spruce (Picea abies (L.) Karst.) and mixed spruce-beech (Fagus sylvatica L.) stands on stagnic gleysol in Austria. Soil δ15N values of both forest sites increased with depth to 50 cm, but then decreased below this zone. δ15N values of microbial biomass (mixed stand: 4.7 ± 0.8‰, spruce stand: 5.9 ± 0.9‰) and of dissolved organic N (DON; mixed stand: 5.3 ± 1.7‰, spruce stand: 2.6 ± 3.3‰) were not significantly different; these pools were most enriched in 15N of all soil N pools. Denitrification represented the main N2O-producing process in the mixed forest stand as we detected a significant 15N enrichment of its substrate NO3 (3.6 ± 4.5‰) compared to NH4+ (−4.6 ± 2.6‰) and its product N2O (−11.8 ± 3.2‰). In a 15N-labelling experiment in the spruce stand, nitrification contributed more to N2O production than denitrification. Moreover, in natural abundance measurements the NH4+ pool was slightly 15N-enriched (−0.4 ± 2.0 ‰) compared to NO3 (−3.0 ± 0.6 ‰) and N2O (−2.1 ± 1.1 ‰) in the spruce stand, indicating nitrification and denitrification operated in parallel to produce N2O. The more positive δ15N values of N2O in the spruce stand than in the mixed stand point to extensive microbial N2O reduction in the spruce stand. Combining natural 15N abundance and 15N tracer experiments provided a more complete picture of soil N dynamics than possible with either measurement done separately.  相似文献

The aim of this research was to test whether NH4 + and NO3 affect the growth, P demand, cell composition and N2 fixation of Cylindrospermopsis raciborskii under P limitation. Experiments were carried out in P-limited (200 μg l−1 PO4-P) chemostat cultures of C. raciborskii using an inflowing medium containing either 4,000 μg l−1 NH4-N, 4,000 μg l−1 NO3-N or no combined N. The results showed the cellular N:P and C:P ratios of C. raciborskii decreased towards the Redfield ratio with increasing dilution rate (D) due to the alleviation of P limitation. The cellular C:N and carotenoids:chlorophyll-a ratios also decreased with D, predominantly as a result of an increase in the chlorophyll-a and N content. The NH4 + and NO3 supply reduced the P maintenance cell quota of C. raciborskii. Consequently, the biomass yield of the N2-grown culture was significantly lower. The maximum specific growth rate of N2-grown culture was also the lowest observed. It is suggested that these differences in growth parameters were caused by the P and energy requirement for heterocyte formation, nitrogenase synthesis and N2 fixation. N2 fixation was partially inhibited by NO3 and completely inhibited by NH4 +. It was probably repressed through the high N content of cells at high dissolved N concentrations. These results indicate that C. raciborskii is able to grow faster and maintain a higher biomass under P limitation where a sufficient supply of NH4 + or NO3 is maintained. Information gained about the species-specific nutrient and pigment stoichiometry of C. raciborskii could help to access the degree of nutrient limitation in water bodies. Methyl fluoride (CH3F) and dimethyl ether (DME) inhibited nitrification in washed-cell suspensions of Nitrosomonas europaea and in a variety of oxygenated soils and sediments. Headspace additions of CH3F (10% [vol/vol]) and DME (25% [vol/vol]) fully inhibited NO2- and N2O production from NH4+ in incubations of N. europaea, while lower concentrations of these gases resulted in partial inhibition. Oxidation of hydroxylamine (NH2OH) by N. europaea and oxidation of NO2- by a Nitrobacter sp. were unaffected by CH3F or DME. In nitrifying soils, CH3F and DME inhibited N2O production. In field experiments with surface flux chambers and intact cores, CH3F reduced the release of N2O from soils to the atmosphere by 20- to 30-fold. Inhibition by CH3F also resulted in decreased NO3- + NO2- levels and increased NH4+ levels in soils. CH3F did not affect patterns of dissimilatory nitrate reduction to ammonia in cell suspensions of a nitrate-respiring bacterium, nor did it affect N2O metabolism in denitrifying soils. CH3F and DME will be useful in discriminating N2O production via nitrification and denitrification when both processes occur and in decoupling these processes by blocking NO2- and NO3- production.  相似文献

Withania somnifera is an important medicinal plant that contains withanolides and withaferins, both bioactive compounds. We have tested the effects of macroelements and nitrogen source in W. somnifera cell suspension cultures with the aim of optimizing the production of biomass and withanolide A. The effects of the macroelements NH4NO3, KNO3, CaCl2, MgSO4 and KH2PO4 at concentrations of 0.0, 0.5, 1.0, 1.5 and 2.0× strength and of the nitrogen source [NH4 +/NO3 (mM/mM) ratio of: 0.00/18.80, 7.19/18.80, 14.38/18.80, 21.57/18.80, 28.75/18.80, 14.38/0.00, 14.38/9.40, 14.38/18.80, 14.38/28.20, and 14.38/37.60 (mM)] in Murashige and Skoog medium were tested for biomass and withanolide A production. The highest accumulation of biomass [147.81 g l−1 fresh weight (FW) and 14.02 g l−1 (dry weight (DW)] was recorded in the medium containing a 0.5× concentration of NH4NO3, and the highest production of withanolide A content was recorded in the medium with 2.0× KNO3 (4.36 mg g−1 DW). The NH4 +/NO3 ratio also influenced cell growth and withanolide A production, with both parameters being larger when the NO3 concentration was higher than that of NH4 +. Maximum biomass growth (110.45 g l−1 FW and 9.29 g l−1 DW) was achieved at an NH4 +/NO3 ratio of 7.19/18.80, while withanolide A production was greatest (3.96 mg g−1 DW) when the NH4 +/NO3 ratio was 14.38/37.60 mM.  相似文献

 A method is presented to isolate mutants of Chlorella sorokiniana with defects in NO3 metabolism. Three nitrite-reductase (NIR; E.C.1.7.7.1)-deficient mutants were obtained from 500 pinpoint-colony-forming clones. The final screening was performed using NO3 , NO2 or NH+ 4 as N-source. The mutants isolated absorb NO3 with rates close to those measured for the wild type and they excrete NO2 into the medium. The ratio between NO3 uptake and NO2 excretion was 1:1. The sensitivity of NO3 uptake to NH+ 4 was reduced in the mutant strains as it was in the N-starved wild type of Chlorella. Nitrate reductase (NR; EC 1.6.6.1) expression and NR activity were slightly reduced compared to the wild type due to feedback regulation in the mutant strains. No NIR protein was found in the three mutants. However, NIR activity was obtained (50% of the wild-type) for one mutant strain. The NIR-deficient mutants and the already available NR-deficient mutants will be promising tools for investigations of the nitrate assimilation pathway on the molecular level and for studies searching for signaling of C and N metabolism by inorganic N-compounds. Wetlands are often highly effective nitrogen (N) sinks. In the Lake Waco Wetland (LWW), near Waco, Texas, USA, nitrate (NO3) concentrations are reduced by more than 90% in the first 500 m downstream of the inflow, creating a distinct gradient in NO3 concentration along the flow path of water. The relative importance of sediment denitrification (DNF), dissimilatory NO3 reduction to ammonium (DNRA), and N2 fixation were examined along the NO3 concentration gradient in the LWW. “Potential DNF” (hereafter potDNF) was observed in all months and ranged from 54 to 278 μmol N m−2 h−1. “Potential DNRA” (hereafter potDNRA) was observed only in summer months and ranged from 1.3 to 33 μmol N m−2 h−1. Net N2 flux ranged from 184 (net denitrification) to −270 (net N2 fixation) μmol N m−2 h−1. Nitrogen fixation was variable, ranging from 0 to 426 μmol N m−2 h−1, but high rates ranked among the highest reported for aquatic sediments. On average, summer potDNRA comprised only 5% (±2% SE) of total NO3 loss through dissimilatory pathways, but was as high as 36% at one site where potDNF was consistently low. Potential DNRA was higher in sediments with higher sediment oxygen demand (r 2 = 0.84), and was related to NO3 concentration in overlying water in one summer (r 2 = 0.81). Sediments were a NO3 sink and accounted for 50% of wetland NO3 removal (r 2 = 0.90). Sediments were an NH4+ source, but the wetland was often a net NH4+ sink. Although DNRA rates in freshwater wetlands may rival those observed in estuarine systems, the importance of DNRA in freshwater sediments appears to be minor relative to DNF. Furthermore, sediment N2 fixation can be extremely high when NO3 in overlying water is consistently low. The data suggest that newly fixed N can support sustained N transformation processes such as DNF and DNRA when surface water inorganic N supply rates are low.  相似文献
