Larvae ofPhyllopertha horticola L. (Coleoptera: Scarabaeidae) cause increasing problems on sports fields and lawns in NW-Europe. A biological control programme using insect parasitic nematodes is being developed. This paper contains the results of bioassays with various species and isolates of the nematode generaHeterorhabditis andSteinernema. In bioassays in small pots with moist sand, most of the nematode isolates gave 30–60% mortality against each of the three larval stages. The susceptibility of the grubs for nematode infection generally increased with larval development.H. bacteriophora, H. heliothidis, H. megidis, a DutchHeterorhabditis isolate NLH-E87.3 andS. glaseri 326 showed the highest mortality rates, with nearly 100% mortality of third instar grubs. The DutchHeterorhabditis isolate NLH-E87.3 andS. glaseri 326 were selected as candidates for further studies on their potential as biological control agents forP. horticola grubs in the field.  相似文献

Rearing methods for two coleopterous predators,Thanasimus dubius andTemnochila virescens, imported into Australia for the biological control ofIps grandicollis, were developed. Bionomic data obtained from laboratory rearings between 1982–1987 showed thatT. dubius eggs took about 7 days to hatch and that duration of the larval stage was about 42 days. Observations showed thatT. dubius had a prolonged prepupal stage (x=56.4 days, range 14–274 days), which was probably non-diapausal in nature. Mean adult longevity was 50 days (range 1–358 days).Temnochila virescens eggs took almost 9 days to hatch, and a lengthy larval stage (x=155.4 days, range 73–333 days) was observed. Mean duration of the pupal stage was 14 days (range 7–28 days). A long preoviposition period (x=141 days, range 47–206 days) was observed, and adults were very long-lived (x=232.7 days, range 14–667 days). Capacity for increase (rc) calculated from rearing data suggested that numbers ofT. dubius could be increased faster thanT. virescens. Mortality between 1982–1987 averaged about 70% for both species. However, mortality ofT. dubius in 1987 increased significantly, suggesting that inbreeding or other methodological factors could be responsible. A mass-rearing method usingIps-infested pine billets was developed as a cheaper alternative to laboratory rearing, and was shown to be effective in producing large numbers of insects for release.  相似文献

Parasitism of the stem and branch-boring mothColeophora parthenica Meyrick [Lepidoptera: Coleophoridae], introduced into California for the biological control of Russian thistle,Salsola australis R. Brown [Chenopodiaceae] was studied in the Coachella Valley of southern California during 1985–1986. Eight parasitoid species were reared from overwintering larvae ofC. parthenica, but none from the F1 larvae, and just 2 individuals of one species from the F2 summer generation. The level of parasitism of overwintering larvae was positively correlated with branch diameter, and ranged from 2% in the primary (main) branches to 45% in the tertiary branches in the spring 1985 sample, and from 2% to 19% in the spring 1986 sample, respectively. Rates of parasitism>20% were only found at sites with higher plant cover and chenopod diversity, but no other plant source or alternate hosts of the parasitoids ofC. parthenica were found. The 2 dominant species, the solitary, hymenopterous ectoparasitoids,Norbanus perplexus (Ashmead) [Pteromalidae] andEurytoma strigosa Bugbee [Eurytomidae], are both congeners of native parasitoids ofC. parthenica in Pakistan. The 2 other species of parasitoids ofC. parthenica in southern California for which other hosts are known are polyphagous and external on the larvae. No specialized endoparasitoid Braconidae, like those which dominate the native parasitoid complex in Pakistan and the U.S.S.R., have transferred toC. parthenica during its first 10 years in southern California.   相似文献

The efficacies of several entomopathogenic nematodes ofSteinernema andHeterorhabditis spp. were examined against tobacco cutworm,Spodoptera litura Fabricius.H. bacteriophora HY showed 100% mortality after 20 h against 2nd instar of tobacco cutworm. In the case of 3–4th instar,S. carpocapsae PC.,H. bacteriophora HY andS. monticola CR showed 100% mortality after 47 h. In the case of 5–6th instar,S. carpocapsae PC proved more effective than the others. Generally, the number of nematodes harvested increased as their size decreased. Also, the highest number of nematodes was obtained in the 5–6th instar ofS. litura byH. bacteriophora HY, showing about 1.3×106 nematodes per larva.In vitro culturedS. carpocapsae PG showed 100% mortality after 73 h against 5–6th instar tobacco cutworm, indicating that nematodes producedin vitro can be potentially used for the biological control ofS. litura instead of nematodesin vivo.  相似文献

AdultGlossina morsitans morsitans exposed to wet conidia ofBeauveria bassiana andMetarhizium anisopliae suffered high mortalities ranging from 90 to 100% by 2 weeks post-exposure. Infected ♂ ♂ maintained in the same cages with non-infected ♀♀ throughout the experimental period transmitted the fungal infection to the ♀♀ resulting in mortalities of 65% withB. bassiana and 55% withM. anisopliae. Likewise, infected ♀♀ maintained together with non-infected ♂♂ transmitted the infection to the ♂♂ resulting in mortalities of 75% withB. bassiana and 45% withM. anisopliae. Female tsetse flies infected withB. bassiana andM. anisopliae and maintained in the same cages with non-infected ♀♀ also transmitted infection to the non-infected tsetse resulting in mortalities of 62% and 48% withB. bassiana andM. anisopliae respectively. Infected tsetse exposed to non-infected tsetse of the opposite sex for only 30 min were also able to transmit the fungal infection. Pupae produced by female tsetse infected withB. bassiana andM. anisopliae exhibited higher pupal mortality than those produced by non-infected ♀♀. However, pupae exposed directly to dry spores ofB. bassiana andM. anisopliae had no increase in pupal mortality but adults emerging from theB. bassiana-exposed pupae had markedly reduced longevity.   相似文献

The biology and behaviour ofTetrastichus sesamiae Risbec, a pupal endoparasitoid ofMaruca testulalis Geyer, were studied under laboratory conditions. Most adults emerged from the host pupae between 08.00 h and 09.00 h and mating and oviposition started almost immediately. Both ♂♂ and ♀♀ mated repeatedly, and each ♀ could lay eggs for up to 6 days and in up to 5 host pupae. Progeny production ranged from 0–263 offspring per ♀ and adult longevity was from 4.3–13.9 days. The quality of food available to the adults was a major factor influencing progeny production, and longevity. The species was capable of parasitizing and completing development on pupae of such other major Lepidopteran crop pests asChilo partellus Swinhoe,Busseola fusca Fuller,Eldana saccharina Wlk andSpodoptera exempta Wlk, an important finding for biological control of these pests under intercropped agro ecosystems. Apart from parasitism, ♀♀ ofT. sesamiae also caused considerable mortality by stinging, and presumably paralyzing, host pupae without ovipositing.   相似文献

A mutation in theCENTRORADIALIS (CEN) gene ofAntirrhinum and in theTERMINAL FLOWER 1 (TFL1) gene ofArabidopsis causes their indeterminate inflorescence to determinate. We clonedCEN/TFL1 homologs fromNicotiana tabacum, the wild-type of which has a determinate inflorescence. TheCEN gene was expressed in the inflorescnece meristem and kept its inflorescence meristem identity, whereas the tobacco homolog (NCH) was expressed at a low level throughout the plant’s development. AlthoughCEN andNCH are highly homologous genes, they may have been recruited to different developmental functions during their evolution. TwoNCH genes are derived from amphidiploidN. tabacum, but both of them hybridized with its diploid parents,N. sylvestris andN. tomentosiformis. Southern blotting, and the genomic organization ofTFL1 inArabidopsis revealed that anotherCEN homolog exists in the genome ofArabidopsis. These results suggest that there are two copies of theCEN homolog per diploid plant. The gregarious parasitoidCotesia flavipes (Hymenoptera: Braconidae) attacks larvae of pyralid and noctuid stemborers by entering the stemborer tunnel. The short-range foraging behavior of femaleC. flavipes was studied on stemborerinfested plants, in patches with host-related products and in artificial transparent tunnels. In addition, the longevity under specific conditions and the potential and realized fecundity of femaleC. flavipes were determined. Larval frass, caterpillar regurgitate, and holes in the stem are used in host location byC. flavipes. The response to host products byC. flavipes seems not to be host species specific. FemaleC. flavipes respond to frass from four stemborer species and one leaf feeder. No differences are found in the behavior ofC. flavipes on maize plants infested with the suitable host,Chilo partellus (Lepidoptera: Pyralidae), or the unsuitable host,Busseola fusca (Lepidoptera: Noctuidae). Attacking a stemborer larva inside the stem is risky for the parasitoid. The mortality rate of the parasitoids inside the stem is high: 30–40% of the parasitoids are killed by the spitting and biting stemborer larva.C. flavipes is relatively shortlived: without food the parasitoids die within 2 days; with food and under high-humidity conditions they die within 5–6 days.C. flavipes is proovigenic and has about 150 eggs available for oviposition. A relatively large proportion of the available egg load (20–25%) is allocated to each host, so femaleC. flavipes are egg depleted after parasitizing only five or six hosts.  相似文献

We examined the genomic occurrence of the transposable elementpogo in over 120 strains ofDrosophila melanogaster, from around the world and from different eras. All had multiple copies of a 2.1 kilobase (kb)pogo element, and multiple copies of several size classes between 1.0 and 1.8 kb. There were differences between strains in intensities or presences of deletion-derivative size classes, suggesting current or recent mobility in the species. We were unable to find anypogo-hybridization in eight other species in the genus, in three subgenera, or in the relatedScaptomyza pallida. Thepogo element may be a ‘middle-aged’ element in the genome ofD. melanogaster, having entered the species since its divergence from its sibling species, but long before theP andhobo elements.  相似文献

During the period 1986–1988 the parasitoids of smaller poplar borer,Saperda populnea L. (Coleoptera, Cerambycidae) along the Danube in Bulgaria were studied. A total of 898 smaller poplar borer galls were investigated—289 in 1986, 404 in 1987, and 205 in 1988. In the particular years the parasitoids of smaller poplar borer namelyBillaea irrorata Meig.(Diptera, Tachinidae), Dolichomitus populneus Ratz. (Hym.: Ichneumonidae), andIphiaulax impostor Scop. (Hym.: Braconidae) destroyed from 13.1 up to 28.0% of the pest larvae, thus decreasing their frequency together with the other limiting factors by 56.1% up to 81.2%. The tachinid flyB. irrorata decreased the smaller poplar borer larvae frequency by 9.3% up to 18.9%, while the ichneumonoidD. populneus, and the braconoidI. impostor destroyed insignificant percentages of these larvae—0.5% up to 1.7% and 0.5% up to 2.5% resp. The flight period of the pest preceded or almost coincided with the ones ofD. populneus, andI. impostor. The flyB. irrorata always appeared last and its flight period was expanded.   相似文献

The compartmentation and metabolism of indole-3-acetic acid (IAA) was examined in protoplasts derived from needles ofPinus sylvestris L., leaves of normal plants ofNicotiana tabacum L., leaves ofN. tabacum plants carrying the T-DNA gene 1 (rG1 plants) and leaves ofN. tabacum plants carrying the T-DNA gene 2 (rG2 plants) by using a rapid cell-fractionation method. In all tissues, 30%–40% of the IAA pool was located in the chloroplast, while the remainder was found in the cytosol. Quantitative analysis of indole-3-ethanol (IEt) showed that in bothPinus andNicotiana the IEt pool was located exclusively in the cytosol. The only plant that contained endogenous indoleacetamide (IAAm) was therG1-mutant ofN. tabacum, expressing theAgrobacterium tumefaciens T-DNA gene 1. Cellular fractionation of protoplasts from this transgenic plant showed that the entire IAAm pool was located in the cytosol. Feeding experiments utilizing [5-3H]tryptophan, [5-3H]IEt, [1′-14C] and [2′-14C]IAA demonstrated that the biosynthesis and catabolism of IAA occurred in the cytosol in bothPinus and in the wild type and the different mutants ofNicotiana. Furthermore, the biosynthesis of IAAm in therG1 plants was also shown to be localized in the cytosol.  相似文献

In order to assess the role of parasitoids in the regulation of non-outbreaking populations of Epirrita autumnata, a geometrid lepidopteran with outbreaking populations in northern Europe, we examined the temporal and spatial variation of larval parasitism in southwestern Finland during 6 successive years. The study was carried out on two spatial scales, among trees within sites of about 1 ha and among sites separated by distances of 2–10 km, using experimental and observational approaches respectively. The overall percent parasitism was independent of host density on both spatial scales, while temporally it fluctuated only little. Of the two main parasitoids, the commoner one, Protapanteles immunis, showed a variable response to host density on the larger spatial scale and negative density dependence on the smaller scale. Temporally, parasitism caused by this species was independent of host density. Another parasitoid, Phobocampe bicingulata, showed positive density dependence on the smaller spatial scale and had a variable response on the larger scale, but exhibited negative density dependence over time. The results of this study caution against drawing conclusions concerning population regulation on the grounds of spatial density dependence alone. Larval parasitoids apparently do not maintain low densities in the E. autumnata populations studied. However, they may suppress E. autumnata densities to a level low enough for density-dependent mortality factor(s) to become regulating. Larval tobacco hornworms,Manduca sexta (L.), of 2 different colonies were exposed to parasitism by the gregarious endoparasitoid,Cotesia congregata (Say). A comparison was made of parasitoid larval, pre-pupal, and pupal mortality, female and male dry weight and larval development time. In general, “Maryland” hornworms were more suitable hosts than “North Carolina” hornworms. Although the presence of dietary nicotine increased parasitoid mortality in individuals reared from hornworms of both colonies, the effect was more severe among individuals parasitizing the North Carolina hornworms. We conducted inundative release experiments withTrichogramma nubilale (Hymenoptera: Trichogrammatidae) to suppressOstrinia nubilalis (Lepidoptera: Pyralidae) in sweet corn (Zea mays): two experiment duringO. nubilalis first generation and three experiments during second generation. Five measurements of ear and stalk damage were used to assess.O. nubilalis control in treated and untreated plots within each experimental field. In one experiment during second generation, naturalO. nubilalis populations were sufficiently high to demonstrate that the parasitoids (three releases totaling 4.4 million parasitoids per ha) parasitized an estimated 57.4% of the placedO. nubilalis egg masses and reduced the mean number ofO. nubilalis larvae per ear by 97.4% the number of tunnels per stalk by 92.9%, and the number of larvae per stalk by 94.3% in the release plot. Ear damage in this experiment was suppressed to meet acceptable standards for use in cut-corn commercial processing. It has recently become apparent that many strains ofE. coli contain nucleases encoded by themcrA andmcrB loci that, recognize the modified base 5-methylcytosine in DNA. Plant DNAs have particularly high levels of this modification and the activity of these 5-methylcytosine-specific nucleases is particularly relevant to cloning plant genomic DNAs. We show here that for preparing libraries in a λ replacement vector, the use of suitablemcr hosts andmcr packaging mixes can increase the efficiency of cloning of plant genomic DNAs by at least two orders of magnitude. We also provide evidence that the activity of themcr nucleases is probably a significant source of bias in the representation of sequences in plant genomic libraries.  相似文献

Laboratory studies showed that ♀♀ ofTrichogramma embryophagum (Hartig) andT. semblidis (Aurivillius) when offered an alternative of young and old eggs of the grain moth,Sitotroga cerealella Oliv., prefer ovipositing into young eggs. The fecundity of ♀♀ that are simultaneously offered both young and old eggs was lower in comparison to ♀♀ offered only young eggs (inspite of the fact that in both variants young eggs were in excess). We investigated the ability of entomopathogenic nematodes to infect diamondback moth (DBM),Plutella xylostella (L.) (Lepidoptera: Plutellidae) on a leaf surface. In a leaf disk assay, mortality of late stage DBM larvae ranged from <7% caused bySteinernema kushidai Mamiya to >95% caused byS. carpocapsae (Weiser) All strain. LC50 values forS. carpocapsae, S. riobravis Cabanillas, Poinar & Raulston, andHeterorhabditis bacteriophora Poinar NC1 strain were 14.6, 15.4, and 65.4 nematodes/larva, respectively.S. carpocapsae, S. riobravis, andH. bacteriophora caused 29%, 33%, and 14% mortality of DBM pupae, respectively. DBM mortality caused byS. carpocapsae on radish declined at low (<76%) to moderate (76–90%) RH, because nematode survival and infectivity declined at low (<76%) to moderate (76–90%) RH. However, DBM mortality caused byS. riobravis did not decline with RH.S. riobravis survival declined with RH, but infectivity did not. Overall, nematode survival and infectivity to DBM larvae were lower forS. riobravis than forS. carpocapsae. In addition, DBM mortality was higher on radish plants (pubescent leaves) than on cabbage plants (glaborous leaves).  相似文献

Under laboratory conditions (22±2°C, 60%±5% H.R.; and 16L.: 8D. photoperiod), the rearing of the 2 main parasitoids ofPhloeotribus scarabaeoides (Bernard, 1788) in Southern Spain:Cheiropachus quadrum (Fabricius, 1787) andRaphitelus maculatus Walker, 1834 (Hym.: Pteromalidae) has been carried out. Several densities of population of the parasitoids have been tested in order to determine their reproductive behaviour, and this is significant if the output of a rearing is to be increased. A great competition among the individuals of the same sex has been observed; in the case of the ΦΦ, a potential negative correlation with the increase of density of population, has been noted. The biotic potential belonging to the 2 principal parasitoids species with and without competition, are calculated, and the aspects concerning the parasitoid colonization of the olive tree beetle infestation spots, are discussed.   相似文献

Mycorrhizal association ofTricholoma matsutake withPinus densiflora was studied. A naturally establishedP. densiflora stand (age: ca. 45 yr) where occurrences ofT. matsutake sporocarps had been confirmed was studied in lbaraki Prefecture, Japan. Pine root systems connected withT. matsutake sporocarps via the fungal white mycelia were sampled in October 1997. The sampled pine roots were covered overall with mycelia. Under a dissecting microscope, the mycelia were confirmed to form fungal sheaths on the lateral roots. Under a light microscope, transverse and longitudinal sections of these roots showed the presence of both fungal sheaths and Hartig nets, which are typical of ectomycorrhizas. The fungal sheath was ca. 1.5–20 μm. in thickness, and felt prosenchymatous in texture. Hartig nets developed continuously at the cortex and extended to the boundary between cortical cells and endodermal cells. The same ectomycorrhizal morphotype on the pine was also recovered from inside the same mycelial colony (i.e., “shiro”) ofT. matsutake from winter to summer. These results suggest thatT. matsutake has a perennial ectomycorrhizal association withP. densiflora.  相似文献

The defence chemicals and behavioural adaptations (gregariousness and active defensive behaviour) of pine sawfly larvae may be effective against ant predation. However, previous studies have tested their defences against very few species of ants, and few experiments have explored ant predation in nature. We studied how larval group size (groups of 5 and 20 in Neodiprion sertifer and 10, 20 and 40 in Diprion pini) and variation in levels of defence chemicals in the host tree (Scots pine, Pinus sylvestris) affect the survival of sawfly larvae. Food preference experiments showed that ants do eat sawfly larvae, although they are not their most preferred food item. According to our results, ant predation significantly increases the mortality rate of sawfly larvae. Larval mortality was minor on pine tree branches where ant traffic was excluded. We also found that a high resin acid concentration in the host tree significantly decreased the mortality of D. pini larvae when ants were present. However, there was no such relationship between the chemical concentrations of the host tree and larval mortality for N. sertifer. Surprisingly, grouping did not help sawfly larvae against ant predation. Mortality risk was the same for all group sizes. The results of the study seemingly contradict previous understanding of the effectiveness of defence mechanisms of pine sawfly against ant predation, and suggest that ants (Formica exsecta in particular) are effective predators of sawfly larvae.An erratum to this article can be found at  相似文献
