Summary Studies of bovine carotid artery smooth muscle cells, during long-term in vitro subcultivation (up to 100 population doublings),
have revealed phenotypic heterogeneity among cells, as characterized by differences in proliferative behavoir, cell morphology,
and contractile-cytoskeletal protein profiles. In vivo, smooth muscle cells were spindle-shaped and expressed desmin and alpha-smooth
muscle actin (50% of total actin) as their predominant cytoskeletal and contractile proteins. Within 24 h of culture, vimentin
rather than desmin was the predominant intermediate filament protein, with little change in alpha-actin content. Upon initial
subcultivation, all cells were flattened and fibroblastic in appearance with a concommitant fivefold reduction in alpha-actin
content, whereas the beta and gamma nonmuscle actins predominated. In three out of four cell lines studied, fluctuations in
proliferative activity were observed during the life span of the culture. These spontaneous fluctuations in proliferation
were accompanied by coordinated changes in morphology and contractile-cytoskeletal protein profiles. During periods of enhanced
proliferation a significant proportion of cells reverted to their original spindle-shaped morphology with a simultaneous increase
in alpha-actin content (20 to 30% of total actin). These results suggest that in long-term culture smooth muscle cells undergo
spontaneous modulations in cell phenotype and may serve as a useful model for studying the regulation of intracellular protein
expression.

Vasoconstrictors activate theNa+-K+-2Cl cotransporter NKCC1 inrat aortic smooth muscle, but the mechanism is unknown. Efflux of86Rb+ from rat aorta in response tophenylephrine (PE) was measured in the absence and presence ofbumetanide, a specific inhibitor of NKCC1. Removal of extracellularCa2+ completely abolished the activation of NKCC1 by PE.This was not due to inhibition of Ca2+-dependentK+ channels since blocking these channels withBa2+ in Ca2+-replete solution did not preventactivation of NKCC1 by PE. Stimulation of NKCC1 by PE was inhibited70% by 75 µM ML-9, 97% by 2 µM wortmannin, and 70% by 2 mM2,3-butanedione monoxime, each of which inhibited isometric forcegeneration in aortic rings. Bumetanide-insensitive Rb+efflux, an indication of Ca2+-dependent K+channel activity, was reduced by ML-9 but not by the other inhibitors. Stretching of aortic rings on tubing to increase lumen diameter to120% of normal almost completely blocked the stimulation of NKCC1 byPE without inhibiting the stimulation by hypertonic shrinkage. Weconclude that activation of theNa+-K+-2Cl cotransporter by PE isthe direct result of smooth muscle contraction throughCa2+-dependent activation of myosin light chain kinase.This indicates that theNa+-K+-2Cl cotransporter isregulated by the contractile state of vascular smooth muscle. 相似文献

Previous studies indicate involvement of the multifunctional Ca2+/calmodulin-dependent protein kinase II (CaMKII) in vascular smooth muscle (VSM) cell migration. In the present study, molecular loss-of-function studies were used specifically to assess the role of the predominant CaMKII delta2 isoform on VSM cell migration using a scratch wound healing assay. Targeted CaMKII delta2 knockdown using siRNA or inhibition of activity by overexpressing a kinase-negative mutant resulted in attenuation of VSM cell migration. Temporal and spatial assessments of kinase autophosphorylation indicated rapid and transient activation in response to wounding, in addition to a sustained activation in the leading edge of migrating and spreading cells. Furthermore, siRNA-mediated suppression of CaMKII delta2 resulted in the inhibition of wound-induced Rac activation and Golgi reorganization, and disruption of leading edge morphology, indicating an important function for CaMKII delta2 in regulating VSM cell polarization. Numerous previous reports link activation of CaMKII to ERK1/2 signaling in VSM. Wound-induced ERK1/2 activation was also found to be dependent on CaMKII; however, ERK activity did not account for effects of CaMKII in regulating Golgi polarization, indicating alternative mechanisms by which CaMKII affects the complex events involved in cell migration. Wounding a VSM cell monolayer results in CaMKII delta2 activation, which positively regulates VSM cell polarization and downstream signaling, including Rac and ERK1/2 activation, leading to cell migration. 相似文献

We have developed a novel cell culturesystem that supports the shortening of smooth muscle cells. Primary ratairway smooth muscle cells were plated on an ethanol-fixed, confluentmonolayer of homologous smooth muscle cells (homologous cell substrate, HCS). Cells grown on HCS exhibited morphological and functional characteristics consistent with a differentiated phenotype. Cells onHCS were spindle shaped with a well-defined long axis, whereas cellsgrown on glass were larger and irregularly shaped. Smooth muscle-specific -actin immunostained diffusely in cells on HCS, whereas it appeared as stress fibers in cells on glass. Agonists recruited a greater fraction of HCS cells to contract, resulting ingreater changes in cell area or length on average, but the maximalcapacity of shortening of individual cells was similar between thegroups. Unlike cells on glass, cells on HCS shortened to methacholine.HCS was reversible and persisted over several passages. Agonistsstimulated intracellular Ca2+ oscillations in cells on HCS,whereas they elicited biphasic peak and plateau transients in cells onglass. Summary To study mechanisms controlling growth and phenotype in human vascular smooth muscle cells, we established culture conditions
under which these cells proliferate rapidly and achieve life-spans of 50–60 population doublings. In medium containing heparin
and heparin-binding growth factors, growth rate and life-span of human vascular smooth muscle cells increased more than 50%
relative to cultures with neither supplement, and more than 20% compared to cultures supplemented only with heparin-binding
growth factors. In contrast to observations made in rat vascular smooth muscle cells, smooth muscle-specific α-actin in the
human cells was expressed only in the presence of heparin and colocalized with β/γ nonmuscle actins in stress fibers, not
in adhesion plaques. Heparin, in the presence of heparin-binding growth factors, also caused more than 170% stimulation of
tracer glucosamine incorporation into hyaluronic acid and a 7.5-fold increase in hyaluronic acid accumulation. In comparison,
total sulfate incorporation into sulfated glycosaminoglycans increased by less than 40%. In light of our previous findings
that heparin suppresses collagen gene expression, we conclude that heparin induces human vascular smooth muscle cells exposed
to heparin-binding growth factors to remodel their extracellular matrix by altering the relative rates of hyaluronic acid
(HA) and collagen synthesis. The resulting hyaluronic-acid-rich, collagen-poor matrix may enhance infiltration of CD44/hyaluronate-receptor-bearing

The effect on phenotypic expression of rabbit vascular smooth muscle cells (SMC) of the interstitial matrix proteins collagen I and fibronectin, the basal lamina proteins collagen IV and laminin, and the serum adhesion protein vitronectin was examined in culture. Experiments were performed in foetal calf serum stripped of fibronectin and vitronectin to eliminate their confounding effects. All the proteins promoted adhesion to the plastic culture dish (in a concentration dependent manner) of SMC freshly isolated from the artery wall. These cells had a high volume density of myofilaments (Vvmyo) in their cytoplasm. Laminin was best at maintaining SMC with a high Vvmyo (Vvmyo = 49.8%) followed by collagen IV (41.7%). Cells plated on vitronectin showed the lowest Vvmyo (31.3%). The results support the concept that the SMC basal lamina has a role in maintaining cells in the high Vvmyo phenotype. 相似文献

Cells receive signals from the extracellular matrix through receptor-dependent interactions, but they are also influenced by the mechanical properties of the matrix. Although bulk properties of substrates have been shown to affect cell behavior, we show here that nanoscale properties of collagen fibrils also play a significant role in determining cell phenotype. Type I collagen fibrils assembled into thin films provide excellent viewing of cells interacting with individual fibrils. Cells can be observed to extensively manipulate the fibrils, and this behavior seems to result in an incompletely spread stellate morphology and a nonproliferative phenotype that is typical of these cells in collagen gels. We show here that thin films of collagen fibrils can be dehydrated, and when seeded on these dehydrated fibrils, smooth muscle cells spread and proliferate extensively. The dehydrated collagen fibrils appear to be similar to the fully hydrated collagen fibrils in topology and in presentation of β1 integrin ligation sites, but they are mechanically stiffer. This decrease in compliance of dehydrated fibrils is seen by a failure of cell movement of dehydrated fibrils compared to their ability to rearrange fully hydrated fibrils and from direct measurements by nanoindentation and quantitative atomic force measurements. We suggest that increase in the nanoscale rigidity of collagen fibrils can cause these cells to assume a proliferative phenotype. 相似文献

Prelamin A accumulation and persistent DNA damage response (DDR) are hallmarks of vascular smooth muscle cell (VSMC) ageing and dysfunction. Although prelamin A is proposed to interfere with DNA repair, our understanding of the crosstalk between prelamin A and the repair process remains limited. The extracellular signal-regulated kinases 1 and 2 (ERK1/2) have emerged as key players in the DDR and are known to enhance ataxia telangiectasia-mutated protein (ATM) activity at DNA lesions, and in this study, we identified a novel relationship between prelamin A accumulation and ERK1/2 nuclear compartmentalisation during VSMC ageing. We show both prelamin A accumulation and increased DNA damage occur concomitantly, before VSMC replicative senescence, and induce the localisation of ERK1/2 to promyelocytic leukaemia protein nuclear bodies (PML NBs) at the sites of DNA damage via nesprin-2 and lamin A interactions. Importantly, VSMCs treated with DNA damaging agents also displayed prelamin A accumulation and ERK compartmentalisation at PML NBs, suggesting that prelamin A and nesprin-2 are novel components of the DDR. In support of this, disruption of ERK compartmentalisation at PML NBs, by either depletion of nesprin-2 or lamins A/C, resulted in the loss of ATM from DNA lesions. However, ATM signalling and DNA repair remained intact after lamins A/C depletion, whereas nesprin-2 disruption ablated downstream Chk2 activation and induced genomic instability. We conclude that lamins A/C and PML act as scaffolds to organise DNA-repair foci and compartmentalise nesprin-2/ERK signalling. However, nesprin-2/ERK signalling fidelity, but not their compartmentalisation at PML NBs, is essential for efficient DDR in VSMCs.DNA damage is a major driving force during cellular ageing, and it has been implicated in hastening the development of cardiovascular diseases, including atherosclerosis where the accumulation of senescent cells has been shown to accelerate disease.1 Normally, DNA damage is efficiently repaired by the DNA damage response (DDR), a complex signalling cascade of proteins that include sensors (NBS1/MRE11), transducers (ataxia telangiectasia-mutated protein (ATM)/ataxia telangiectasia- and Rad3-related protein (ATR)) and effectors (p53/p21). However, if damage is overwhelming or repair is inefficient, the accumulation of unrepaired DNA damage leads to persistent DNA damage signalling and premature senescence.2 Recently, the nuclear lamina has been implicated in the DDR, and its disruption is associated with accelerated cardiovascular ageing.3The nuclear lamina is composed of A-type (lamins A/C) and B-type (lamins B1/B2) lamins that underlie the nuclear envelope (NE) and extend throughout the nucleoplasm4 to form a scaffold that is essential for the compartmentalisation and the integrity of nuclear signalling.4 The pathological accumulation of lamin A precursors such as prelamin A or progerin causes Hutchinson Gilford Progeria Syndrome (HGPS), a disease of accelerated ageing where patients develop severe early-onset arteriosclerosis characterised by vascular smooth muscle cell (VSMC) attrition.4 During the DDR, lamin A interacts with Ku70 and γH2AX, and forms a framework that is essential for the positional stability of repair foci.5, 6 Prelamin A accumulation interferes with DNA-repair processes and has been shown to delay the recruitment of DNA-repair proteins such as 53BP1 to double strand breaks (DSBs)7 and cause mislocalisation of DNA PKcs.8 Fibroblasts and induced pluripotent stem (iPS) cells derived from HGPS patients show persistent DNA damage signalling and premature senescence.8 Importantly, aged VSMCs, both in vitro and in vivo, also accumulate prelamin A and activate persistent DDR, suggesting a key role for the nuclear lamina in vascular ageing.2The nesprin family of spectrin-repeat (SR) proteins were first identified as NE lamin A-binding proteins. However, both nesprin-1 and nesprin-2 show extensive alternate splicing, and variants have been shown to localise to multiple nuclear and cytoplasmic compartments.9 One such variant, nesprin-2βΔKASH1, retains a lamin A-binding region and localises to promyelocytic leukaemia protein nuclear bodies (PML NBs) in VSMCs.10 Previously, we demonstrated that nesprin-2βΔKASH1 scaffolds extracellular signal-regulated kinases 1 and 2 (ERK1/2) at PML NBs and acts to regulate nuclear ERK1/2 activity and downstream VSMC proliferation.10 Importantly, both ERK and PML NBs have also been implicated in the DDR; PML and ERK1/2 localise at DNA lesions where ERK1/2 enhance ATM- and ATR-mediated repair.11, 12, 13 In addition, PML and ERK1/2 are essential for regulating the cell cycle in response to DNA damage; PML forms nucleolar cap structures that sequester MDM2 and activate DNA damage-mediated p53 signalling, while ERK1/2 are essential for efficient G2/M checkpoint activation.14, 15, 16 Similar to nesprin-2, PML and ERK1/2 have also been shown to associate with the nuclear lamina suggesting that the nuclear lamina, potentially via nesprin-2βΔKASH1, may regulate ERK compartmentalisation during the DDR.17, 18In this study, we identify a novel signalling complex that regulates compartmentalisation of ERK1/2 during the DDR in VSMCs. We show that the nuclear lamina tethers PML NBs and spatially organises nuclear signalling events. Disruption of this organisation results in ATM mislocalisation from DNA-repair foci and impairs downstream DNA-repair signalling, ultimately leading to genomic instability. 相似文献

During vascular disease and following injury, vascular smooth muscle cells (VSMC) proliferate and produce inflammation-promoting cytokines and chemokines. Similar phenotypic changes can be elicited in vitro by activation of Toll-like receptors (TLR) within VSMC. TLR-activated VSMC also produce IL-1 alpha, but it is unknown whether endogenous IL-1 alpha stimulates VSMC in an autocrine manner. Here we tested the hypothesis that endogenous IL-1 alpha contributes to TLR-induced proliferation and chemokine release in human VSMC by using RNA interference to knock down IL-1 alpha expression. Knockdown of IL-1 alpha abolished TLR-induced proliferation and suppressed TLR4-induced release of monocyte chemoattractant protein-1 (MCP-1) by VSMC, indicating that endogenous IL-1 alpha plays a crucial role in both responses. Serum, PDGF, FGF-2, and EGF each increased cellular IL-1 alpha concentrations, and IL-1 alpha knockdown inhibited serum- and PDGF-induced DNA synthesis, further indicating that endogenous IL-1 alpha also contributed to VSMC responses to growth factors. IL-1 receptor antagonist, a competitive inhibitor of IL-1 receptor I (IL-1RI), also attenuated TLR-induced proliferation and both basal and TLR-induced MCP-1 expression, indicating at least a partial role of the IL-1RI in mediating these responses. The results support the hypothesis that autocrine actions of endogenous IL-1 alpha, mediated at least in part via IL-1RI signaling, contribute to a proproliferative and proinflammatory phenotypic shift in TLR-activated human VSMC, which might play a pathogenic role in vascular disorders. 相似文献

Vascular smooth muscle cells (VSMC) at capacitance arteries of hypertensive individuals and animals undergo dramatic polyploidization that contributes toward their hypertrophic phenotype. We report here the identification of a defective mitotic spindle cell cycle checkpoint in VSMC isolated from capacitance arteries of pre-hypertensive rats. These cells demonstrated a high predisposition to polyploidization in culture and failed to maintain cyclin B protein levels in response to colcemid, a mitotic inhibitor. Furthermore, this altered mitotic spindle checkpoint status was associated with the overexpression of Cks1, a Cdc2 adapter protein that promotes cyclin B degradation. Cks1 up-regulation, cyclin B down-regulation, and VSMC polyploidization were evidenced at the smooth muscle of capacitance arteries of genetically hypertensive and Goldblatt-operated rats. In addition, angiotensin II infusion dramatically increased Cks1 protein levels at capacitance arteries of normotensive rats, and angiotensin II treatment of isolated VSMC abrogated their ability to down-regulate Cks1 and maintain cyclin B protein expression in response to colcemid. Finally, transduction of VSMC from normotensive animals with a retrovirus that drives the expression of Cks1 was sufficient to alter their mitotic spindle cell cycle checkpoint status and promote unscheduled cyclin B metabolism, cell cycle re-entry, and polyploidization. These data demonstrate that Cks1 regulates cyclin B metabolism and ploidy in VSMC and may contribute to the understanding of the phenomena of VSMC polyploidization during hypertension. 相似文献

The aim of the present work was to study the morphological implications between the elastin and the phenotypic expression of the vascular smooth muscle cells. For this purpose, sixty human tortuous arteries from different territories have been studied. We have measured the morphometric indexes Intimal Thickening Index and Elastolyse Index and they have been quantified with computer system analysis, image-colour corresponding to the orcein and Verho?ff reactions for detecting elastin and the alpha-actin in the smooth muscle cells. We compared both territorial arteries from the cranial and from abdominal origin. The elastin concentration was similar in both territories, but not its morphology according to its spatial distribution. We have observed a relationship between the elastin structural organisation from the media of arteries and of the internal elastic lamina in these territories and the variation of reactivity to the smooth muscle alpha-actin as a marker of the phenotypic state. Our results confirm the hypothesis that elastin, besides intervening in the architecture of the arterial wall, is a factor implicated in the phenotypic variability of the smooth muscle cells and in the development and evolution of the intimal thickenings in human atherosclerosis. 相似文献

There is accumulating evidence that Ca2+-dependent signaling pathways regulate proliferation and migration of vascular smooth muscle (VSM) cells, contributing to the intimal accumulation of VSM that is a hallmark of many vascular diseases. In this study we investigated the role of the multifunctional serine/threonine kinase, calmodulin (CaM)-dependent protein kinase II (CaMKII), as a mediator of Ca2+ signals regulating VSM cell proliferation. Differentiated VSM cells acutely isolated from rat aortic media express primarily CaMKII gene products, whereas passaged primary cultures of de-differentiated VSM cells express primarily CaMKII2, a splice variant of the gene. Experiments examining the time course of CaMKII isoform modulation revealed the process was rapid in onset following initial dispersion and primary culture of aortic VSM with a significant increase in CaMKII2 protein and a significant decrease in CaMKII protein within 30 h, coinciding with the onset of DNA synthesis and cell proliferation. Attenuating the initial upregulation of CaMKII2 in primary cultured cells using small-interfering RNA (siRNA) resulted in decreased serum-stimulated DNA synthesis and cell proliferation in primary culture. In passaged VSM cells, suppression of CaMKII2 activity by overexpression of a kinase-negative mutant, or suppression of endogenous CaMKII content using multiple siRNAs, significantly attenuated serum-stimulated DNA synthesis and cell proliferation. Cell cycle analysis following either inhibitory approach indicated decreased proportion of cells in G1, an increase in proportion of cells in G2/M, and an increase in polyploidy, corresponding with accumulation of multinucleated cells. Platelet-derived growth factor BB (PDGF-BB) induced cyclin A expression and CDK2 activity in vascular smooth muscle cells (VSMC). Inhibition of nuclear factors of activated T cell (NFAT) activation by cyclosporin A (CsA) and VIVIT suppressed PDGF-BB-induced cyclin A expression and CDK2 activity, resulting in blockade of VSMC in the G(1) phase. In addition, CsA- and VIVIT-mediated inhibition of NFATs and small interfering RNA-targeted down-regulation of cyclin A levels suppressed PDGF-BB-induced VSMC DNA synthesis. PDGF-BB also induced cyclin A mRNA levels in VSMC in an NFAT-dependent manner. Cloning and bioinformatic analysis of rat cyclin A promoter revealed the presence of NFAT-binding elements, and PDGF-BB induced the binding of NFATs to these regulatory sequences in a CsA- and VIVIT-sensitive manner. Chromatin immunoprecipitation analysis showed that NFATc1 binds to the cyclin A promoter in response to PDGF-BB in a VIVIT-sensitive manner. Furthermore, PDGF-BB induced cyclin A promoter-luciferase reporter gene activity in VSMC, and it was inhibited by both CsA and VIVIT. Balloon injury induced cyclin A expression and CDK2 activity in rat carotid arteries, and these responses were also blocked by VIVIT. In addition, VIVIT attenuated balloon injury-induced SMC proliferation, resulting in reduced restenosis. Down-regulation of NFATc1 by its small interfering RNA inhibited PDGF-BB-induced cyclin A expression and DNA synthesis both in rat and human VSMC. Together, these findings demonstrate that the cyclin A-CDK2 complex may be a potential effector of NFATs, specifically NFATc1, in mediating SMC multiplication leading to neointima formation. Therefore, NFATs may be used as target molecules for the development of therapeutic agents against vascular diseases such as restenosis. 相似文献

Rat liver microsomal membranes have been shown to contain a biosynthetic pathway of UDP-glucose. In addition, they are able to bind UDP-glucose in a reversible manner, As UDP-glucose is also metabolized in these membranes, the study of the binding has been performed with a microsomal Triton X 100 extract. This reversible binding depends on pH (maximum at pH 8.1) and manganous ions, and disappears at pH 6.5. It exhibits a high affinity (K-diss equals 3 mu M), and a narrow specifity for UDP-glucose. Proteolytic digestion inhibits the binding up to 90%, showing that the UDP-glucose receptor has a proteic nature. These binding characteristics have been also found in the membranes themselves, indicating that the detergent solubilization does not destroy the protein binding capacity. 相似文献
