Three varieties of Phaseolus vulgaris which differ in their sensitivity to ozone were examined for changes in some physiological and structural plasma membrane characteristics. Plasma membrane vesicles were prepared from control and ozone-treated (0.2 to 0.5 microliters per liter ozone for 5 hours) leaf tissue, and the (K+ + Mg2+)-ATPase activity determined and compared. No major changes were observed in the resistant varieties. The sensitive variety showed a severe inhibition of ATPase activity which was largely due to a decrease in the K+-stimulated component. This inhibition was completely reversed by the addition of sulfhydryl compounds.

Ozone-induced plasma membrane permeability changes may be effected by damage to membrane proteins, perhaps by oxidation of amino acid sulfhydryl groups to disulfide and sulfenic moieties.

The influence of ozone on Ca2+ transport in plant membranes from pinto bean (Phaseolus vulgaris L. var Pinto) leaves was investigated in vitro by means of a filtration method using purified vesicles. Two transport mechanisms located at the plasma membrane are involved in a response to ozone: (a) passive Ca2+ influx into the cell and (b) active Ca2+ efflux driven by an ATP-dependent system, which has two components: a primary Ca2+ transport directly linked to ATP which is partially activated by calmodulin and a H+/Ca2+ antiport coupled to activity of a H+-ATPase. The passive Ca2+ permeability is increased by ozone. A triangular pulse of ozone stimulates a higher influx of Ca2+ than does a square wave, even though the total dose was the same (0.6 microliter per liter × hour). Leaves exposed to a square wave did not exhibit visible injury and were still able to recover from oxidant stress by activation of calmodulin-dependent Ca2+ extrusion mechanisms. On the other hand, leaves exposed to a triangular wave of ozone, exhibit visible injury and lost the ability of extruding Ca2+ out of the cell.  相似文献

Gibberellic acid was tested for its effect on tumor induction by Agrobacterium tumefaciens in primary pinto bean (Phaseolus vulgaris) leaves in various stages of development. The hormone was found to promote tumor induction in partially aged leaves but did not effect tumor induction in very young leaves or in fully matured leaves. It is suggested that the natural loss of susceptibility to tumor induction in maturing pinto bean leaves is associated with a concomitant loss of endogenous gibberellins and/or a sensitivity to gibberellins.  相似文献

Acid-soluble nucleotides of unifoliate leaves of Pinto bean plants (Phaseolus vulgaris L.) were determined at young, mature, and senescent stages of development. At least 25 components could be distinguished on the basis of inorganic phosphorus determinations and 37 or more fractions on the basis of 32P labeling, with adenosine di- and triphosphates accounting for 60% of the total moles of nucleotide. The total nucleotide P and inorganic P, on a fresh weight basis, decreased about 44% between each stage of leaf development, but decrements in the levels of individual nucleotides varied from this over-all pattern.  相似文献

Crown-gall tumor initiation by Agrobacterium tumefaciens (SMITHand TOWN.) CONN, strain B6, on Phaseolus vulgaris L. var. "Pinto"was found to be sensitive to the addition of various salts tothe freshly inoculated primary leaves. Calcium, magnesium, zincand ammonium sulfates, cobalt and nickel chlorides and potassiumnitrate increased the number of tumors on inoculated leavesby 100 to 200 percent. Maximal promotions were obtained at concentrationsof 10–7 to 10–5 M with all of these salts exceptpotassium nitrate which was most active at 10–2M. Sodium,potassium, calcium, magnesium and aluminum chlorides, and sodium,potassium and nickel sulfates had little or no effect on tumorinitiation. The combination of sodium sulfate with magnesiumchloride gave promotions comparable to those obtained with magnesiumsulfate, indicating that both the magnesium and sulfate ionswere necessary for the promotion obtained with this latter salt.Combining any two of the active salts at their optimum concentrationsfor promotion resulted in a reciprocal inhibition of the promotionobtained with either salt alone, suggesting these salts to beactive by different and potentially antagonistic mechanisms.The possible nature of these promotions is discussed, but theyremain too obscure to warrant a specific proposal for theirmode of action. 1Present adress: Department of Botany, University of Khartoum,Khartoum, Sudan.  相似文献

Extracts from various parts of Phaseolus vulgaris L. var. Pinto plants were found to exhibit cytokinin activity with the highest levels present in extracts of the fruit tissue. To separate the cytokinins present in the fruit, extracts were chromatographed in several solvent systems. Bioassays of chromatograms indicated the presence of active factors in those regions of migration associated with several known N-6-substituted aminopurines.  相似文献

Precise oxygen equilibrium curves of hemoglobin stripped of phosphates were determined at pH 7.4 and five different temperatures. The data were thermodynamically analyzed according to Adair's stepwise oxygenation theory and the allosteric model of Monod etal. Heat of oxygenation of Hb(O2)3 was significantly larger than that of Hb, indicating that the shape of oxygen equilibrium curve is not invariant with the change of temperature. The results do not support the idea that the cooperative effects are essentially entropic in nature, suggesting that the allosteric transition from the unliganded T-state to the unliganded R-state is an endothermic process, during which the hemoglobin molecule gains entropy.  相似文献

It was shown that raising pod seedlings by the hydroponics method on KH2PO4 solutions at concentrations between 10(-7) and 10(-5) M leads to an increase in the rate of oxygen release (delta O2/delta t), with the chlorophyll content in leaves being unchanged. The values of the parameters FM/FT of slow fluorescence induction and B/A of photoinduced changes in ESR1 signals from pod leaves correlate with the delta O2/delta t value.  相似文献

The importance of the absolute and relative concentrations of monovalent and divalent cations and centrifugal speed (pressure) in the dissociation of mung bean 80S ribosomes has been examined. In the absence of Mg2+ ions, ribosome monomers yield 47S and 34S particles. Fixation with glutaraldehyde, however, indicates that this dissociation pattern is largely dependent upon high pressures developed during centrifugation and that in the absence of such artifacts the immediate product of Mg-free conditions is a 74S particle. Since 74S particles rapidly revert to the 80S form when Mg is replaced, this would appear to be a conformational change. Ribosomes were also dissociated in the presence of Mg2+ ions if the K+ ion concentration was raised. Three major particles were produced, 38S and 49S from the small ribosomal sub-unit and 60S from the large sub-unit. A proportion of the 80S monomer population is more resistant to dissociation. Experiments with puromycin indicate that the more resistant fraction probably represents ribosomes completed with nascent polypeptide resulting from polysome breakdown.  相似文献

Sucrose gradient analyses were made on free ribosomes extracted from Azuki bean seedling leaves subjected to mannitol induced water stress. Comparisons were made of the effect of duration of stress and the effect of urea on the free ribosomes. The capacities of the ribosomal fractions to incorporate amino-acids were also studied. Water stress reduced total free ribosomes and proportion of polysomes in the ribosomes as well as the capacity of the ribosomes to incorporate amino-acids into proteins. Polysomes which were broken down by the water stress also appeared to be more susceptible to denaturation and synthesized different proteins than polysomes which remained in the leaves after water stress.  相似文献

A crystalline protein-proteinase inhibitor has been isolated from seeds of Pinto bean (Phaseolus vulgaris cultvar. Pinto). It has an average molecular weight of 19 000 as estimated by gel filtration. This crystalline inhibitor is highly active against both bovine pancreatic trypsin and alpha-chymotrypsin. Complexes of both trypsin-inhibitor and alpha-chymotrypsin-inhibitor have been isolated. The inhibitor which was derived from the dissociated trypsin-inhibitor complex was only 62% as effective as the original compound against either enzyme. In contrast, the inhibitor obtained from alpha-chymotrypsin-inhibitor complex retained its full original inhibitory activity for trypsin, but only 25% of its original activity against alpha-chymotrypsin. The dissociated inhibitor from alpha-chymotrypsin-inhibitor compex, despite its full inhibitory activity, had been modified to such an extent that it could no longer form any precipitable complex with trypsin. The crystalline protein-proteinase inhibitor is not homogeneous and has been resolved into two distinct inhibitors in terms of their physical and chemical properties. These two inhibitors are designated as Pinto bean proteinase inhibitor I and II and their respective minimum molecular weights are 9100 and 10 000. They differ most strikingly in their amino acid composition in that inhibitor II is void of both valine and methionine.  相似文献

A new leaf disc chamber allows measurements of chlorophyll fluorescence and CO2 and H2O vapor exchanges during infusion of solution into the cut edge of the disc. Polyethylene glycol (molecular weight, 6000) was used to apply a mild external osmotic stress to the leaf disc within this chamber. This stress rapidly caused a temporary increase in transpiration. This increase was reversed (5-6 minutes later) and after 20 to 25 min, the stomates nearly completely closed. Internal CO2 (calculated) and leaf temperature followed the transpiration measurements. However, chlorophyll fluorescence (small rise) followed internal CO2 (small rise). This complete sequence of events resembles those caused by exposure of leaves to certain air pollutants which have been seen to cause such a transient increase followed by a decrease in stomatal closure.  相似文献

Light (about 3,000 foot-candles) neither increased nor decreased the sink strength of young, rapidly expanding leaves of Phaseolus vulgaris L. cv. Black Valentine, as measured by the comparative rates of import of 14C-labeled photosynthates by sink leaves in the light versus dark in short term experiments. Although irradiated sink leaves accumulated more 14C activity, the difference was fully accounted for by photosynthetic reabsorption of respiratory CO2 derived from substrates translocated to the sink leaves.

Maximum sink strength was attained when the sink leaf reached 7 to 8 cm2 in area (9 to 10% of its fully expanded size). Thereafter sink strength declined rapidly and asymptotically to a near zero value at about 45% final area. During this period, however, the rapid decline in translocation was offset by a rapid rise in the photosynthetic rate of the sink leaf, maintaining a near constant relative rate of dry weight increase until the sink leaf had expanded to about 17% of its final area. Although the increasing photosynthetic capacity was associated with a decreasing import capacity, suggesting that the rate of translocation to the sink leaf was controlled by the developing capacity of the sink leaf for photosynthesis, it was not possible to vary the total (true) translocation rate to the sink leaf by varying the photosynthetic rate of the sink leaf in short term light-dark experiments. Despite a high ratio of source to sink in these experiments, no evidence accrued that translocation into young bean leaves was ever sink-limited.

To compare the effects of ozone and peroxyacetyl nitrate (PAN) on leaf lipids, fatty acids and malondialdehyde (MDA), morning glory (Pharbitis nil Choisy cv Scarlet O'Hara) and kidney bean (Phaseolus vulgaris L. cv Gintebo) plants were exposed to either ozone (0.15 microliter per liter for 8 hours) or PAN (0.10 microliter per liter for up to 8 hours). Ozone increased phospholipids in morning glory and decreased in kidney bean at the initial stage (2-4 hours) of exposure, while it scarcely changed glycolipids, the unsaturated fatty acids, and MDA in both plants. A large reduction of glycolipids occurred 1 day after ozone exposure in both plants. PAN caused marked drops in phospholipids and glycolipids in kidney bean at relatively late stage (6-8 hours) of exposure, while it increased phosphatidic acid and decreased the unsaturated fatty acids, an increase which was accompanied by a large increase in MDA. These results suggest that ozone may not directly oxidize unsaturated fatty acids at the initial stage of exposure, but may alter polar lipid metabolism, particularly phospholipids. On the other hand, PAN may abruptly and considerably degrade phospholipids and glycolipids by peroxidation or hydrolysis at the late stage of exposure. The present study shows that ozone and PAN affect polar lipids in different manners.  相似文献

A highly purified preparation of carboanhydrase from the leaves of the bean Vicia faba var. major (Harz) F. Janthina was obtained. The enzyme was homogeneous during analytical disc-electrophoresis in polyacrylamide gel and analytical ultracentrifugation. The molecular weight of the enzyme is 270,000. The enzyme molecule contains six Zn atoms, has a quaternary structure and is made up of six subunits with molecular weights of 45,000. Some properties of the enzymes from bean leaves and pea leaves were compared. The enzymes differ in molecular weights and behaviour during DEAE-cellulose chromatography and gel filtration through Sephadex G-200.  相似文献

Quantitative studies on ferredoxin in greening bean leaves   总被引:8,自引:2,他引:6  
Two methods of measuring small amounts of the iron-sulphur protein ferredoxin are described. One involves measurements of the signal at g=1.96 produced by reduced ferredoxin in an e.p.r. (electron-paramagnetic-resonance) spectrometer; the other depends on the rate of ferredoxin-dependent electron transport in a chloroplast bioassay measured in an O(2) electrode. These methods of measurement were used to examine the development of ferredoxin during the greening of etiolated bean leaves. Ferredoxin is present in low concentrations in the leaves and cotyledons of 14-day-old etiolated beans (Phaseolus vulgaris L. var. Canadian Wonder), and develops in a linear manner with time when the leaves are illuminated. This synthesis appears to be independent of chlorophyll synthesis during the early stages of greening. However, the chlorophyll/ferredoxin ratio reaches a final value of approx. 360 irrespective of the light intensity, indicating the existence of a control mechanism operative in deciding the stoicheiometry of these components in the mature chloroplast. The ferredoxin synthesis appears to be light-dependent, and red light is the most effective in its promotion. The effect of red illumination is not reversed by far-red light, indicating the absence of a phytochrome control of ferredoxin synthesis. From experiments using specific inhibitors of chloroplast protein synthesis, it is concluded that ferredoxin is synthesized on cytoplasmic ribosomes.  相似文献

由于城市化的加剧导致近地面臭氧(O3)浓度日益增加,对植物生长和生态系统的功能产生了显著影响,因此准确评估近地层O3浓度升高对植物的影响具有重要意义。本文利用开顶式气室(OTCs),系统探讨了模拟O3胁迫下大豆抗氧化系统抗坏血酸(AsA)-谷胱甘肽(GSH)循环清除活性氧(ROS)的机制及其对植株生长发育的影响。结果表明,在整个生育期内,与对照相比, 80?10 nL?L-1和110?10 nL?L-1 O3可以使大豆叶片丙二醛(MDA)含量、相对电导率增大,超氧阴离子(O2 )产生速率、过氧化氢(H2O2)含量升高,超氧化物歧化酶(SOD)活性减弱; AsA-GSH循环中的AsA、GSH含量减少,脱氢抗坏血酸(DHA)、氧化型谷胱甘肽(GSSG)含量增加,过氧化物酶(APX)、单脱氢抗坏血酸还原酶(MDHAR)、谷胱甘肽还原酶(GR)活性呈现出前期增强后期减弱趋势,而脱氢抗坏血酸还原酶(DHAR)活性呈现出增强-减弱-增强的趋势。以上结果说明,O3浓度升高促进了大豆叶片ROS的代谢速率,降低了AsA-GSH循环效率,表明抗氧化系统不能长时间忍受高浓度O3带来的氧化伤害,从而使膜脂过氧化程度加重,对大豆表现为伤害效应。  相似文献
