When grown in the absence of added sulfate, cocultures of Desulfovibrio desulfuricans or Desulfovibrio vulgaris with Methanobrevibacter smithii (Methanobacterium ruminantium), which uses H2 and CO2 for methanogenesis, degraded lactate, with the production of acetate and CH4. When D. desulfuricans or D. vulgaris was grown in the absence of added sulfate in coculture with Methanosarcina barkeri (type strain), which uses both H2-CO2 and acetate for methanogenesis, lactate was stoichiometrically degraded to CH4 and presumably to CO2. During the first 12 days of incubation of the D. desulfuricans-M. barkeri coculture, lactate was completely degraded, with almost stoichiometric production of acetate and CH4. Later, acetate was degraded to CH4 and presumably to CO2. In experiments in which 20 mM acetate and 0 to 20 mM lactate were added to D. desulfuricans-M. barkeri cocultures, no detectable degradation of acetate occurred until the lactate was catabolized. The ultimate rate of acetate utilization for methanogenesis was greater for those cocultures receiving the highest levels of lactate. A small amount of H2 was detected in cocultures which contained D. desulfuricans and M. barkeri until after all lactate was degraded. The addition of H2, but not of lactate, to the growth medium inhibited acetate degradation by pure cultures of M. barkeri. Pure cultures of M. barkeri produced CH4 from acetate at a rate equivalent to that observed for cocultures containing M. barkeri. Inocula of M. barkeri grown with H2-CO2 as the methanogenic substrate produced CH4 from acetate at a rate equivalent to that observed for acetate-grown inocula when grown in a rumen fluid-vitamin-based medium but not when grown in a yeast extract-based medium. The results suggest that H2 produced by the Desulfovibrio species during growth with lactate inhibited acetate degradation by M. barkeri.  相似文献

Hydrogenotrophic methanogens can use gaseous substrates, such as H2 and CO2, in CH4 production. H2 gas is used to reduce CO2. We have successfully operated a hollow-fiber membrane biofilm reactor (Hf-MBfR) for stable and continuous CH4 production from CO2 and H2. CO2 and H2 were diffused into the culture medium through the membrane without bubble formation in the Hf-MBfR, which was operated at pH 4.5–5.5 over 70 days. Focusing on the presence of hydrogenotrophic methanogens, we analyzed the structure of the microbial community in the reactor. Denaturing gradient gel electrophoresis (DGGE) was conducted with bacterial and archaeal 16S rDNA primers. Real-time qPCR was used to track changes in the community composition of methanogens over the course of operation. Finally, the microbial community and its diversity at the time of maximum CH4 production were analyzed by pyrosequencing methods. Genus Methanobacterium, related to hydrogenotrophic methanogens, dominated the microbial community, but acetate consumption by bacteria, such as unclassified Clostridium sp., restricted the development of acetoclastic methanogens in the acidic CH4 production process. The results show that acidic operation of a CH4 production reactor without any pH adjustment inhibited acetogenic growth and enriched the hydrogenotrophic methanogens, decreasing the growth of acetoclastic methanogens.  相似文献

The anaerobic pathway of chitin decomposition by chitinoclastic bacteria was examined with an emphasis on end product coupling to other salt marsh bacteria. Actively growing chitinoclastic bacterial isolates produced primarily acetate, H2, and CO2 in broth culture. No sulfate-reducing or methanogenic isolates grew on chitin as sole carbon source or produced any measurable degradation products. Mixed cultures of chitin degraders with sulfate reducers resulted in positive sulfide production. Mixed cultures of chitin-degrading isolates with methanogens resulted in the production of CH4 with reductions in headspace CO2 and H2. The combination of all three metabolic types resulted in the simultaneous production of methane and sulfide, with more methane being produced in mixed cultures containing CO2-reducing methanogens and acetoclastic sulfate reducers because of less interspecific H2 competition.  相似文献

The activity of and potential substrates for methane-producing bacteria and sulfate-reducing bacteria were examined in marsh, estuary, and beach intertidal sediments. Slow rates of methane production were detected in all sediments, although rates of sulfate reduction were 100- to 1,000-fold higher. After sulfate was depleted in sediments, the rates of methane production sharply increased. The addition of methylamine stimulated methanogenesis in the presence of sulfate, and [14C]methylamine was rapidly converted to 14CH4 and 14CO2 in freshly collected marsh sediment. Acetate, hydrogen, or methionine additions did not stimulate methanogenesis. [methyl-14C]methionine and [2-14C]acetate were converted to 14CO2 and not to 14CH4 in fresh sediment. No reduction of 14CO2 to 14CH4 occurred in fresh sediment. Molybdate, an inhibitor of sulfate reduction, inhibited [2-14C]acetate metabolism by 98.5%. Fluoracetate, an inhibitor of acetate metabolism, inhibited sulfate reduction by 61%. These results suggest that acetate is a major electron donor for sulfate reduction in marine sediments. In the presence of high concentrations of sulfate, methane may be derived from novel substrates such as methylamine.  相似文献

A bacterial consortium capable of sucrose degradation primarily to CH4 and CO2 was constructed, with acetate as the key methanogenic precursor. In addition, the effect of agar immobilization on the activity of the consortium was determined. The primary fermentative organism, Escherichia coli, produced acetate, formate, H2, and CO2 (known substrates for methanogens), as well as ethanol and lactate, compounds that are not substrates for methanogens. Oxidation of the nonmethanogenic substrates, lactate and ethanol, to acetate was mediated by the addition of Acetobacterium woodii and Desulfovibrio vulgaris. The methanogenic stage was accomplished by the addition of the acetophilic methanogen Methanosarcina barkeri and the hydrogenophilic methanogen Methanobacterium formicicum. Results of studies with low substrate concentrations (0.05 to 0.2% [wt/vol]), a growth-limiting medium, and the five-component consortium indicated efficient conversion (40%) of sucrose carbon to CH4. Significant decreases in yields of CH4 and rates of CH4 production were observed if any component of the consortium was omitted. Approximately 70% of the CH4 generated occurred via acetate. Agar-immobilized cells of the consortium exhibited yields of CH4 and rates of CH4 production from sucrose similar to those of nonimmobilized cells. The rate of CH4 production decreased by 25% when cysteine was omitted from reaction conditions and by 40% when the immobilized consortium was stored for 1 week at 4°C.  相似文献

Methanosarcina barkeri strain Fusaro was found to grow on pyruvate as sole carbon and energy source after an incubation period of 10–12 weeks in the presence of high pyruvate concentrations (100 mM). Growth studies, cell suspension experiments and enzymatic investigations were performed with pyruvate-utilizing M. barkeri. For comparison acetate-adapted cells of M. barkeri were analyzed.
  1. Pyruvate-utilizing M. barkeri grew on pyruvate (100 mM) with an initial doubling time of about 25 h (37 °C, pH 6.5) up to cell densities of about 0.8 g cell dry weight/l. The specific growth rate was linearily dependent on the pyruvate concentration up to 100 mM indicating that pyruvate was taken up by passive diffusion. Only CO2 and CH4 were detected as fermentation products. As calculated from fermentation balances pyruvate was converted to CH4 and CO2 according to following equation: Pyruvate-+H++0.5 H2O » 1.25 CH4+1.75 CO2. The molar growth yield (Ych 4) was about 14 g dry weight cells/mol CH4. In contrast the growth yield (Ych 4) of M. barkeri during growth on acctate (Acetate-+H+ » CH4+CO2) was about 3 g/mol CH4.
  2. Cell suspensions of pyruvate-grown M. barkeri catalyzed the conversion of pyruvate to CH4, CO2 and H2 (5–15 nmol pyruvate consumed/min x mg protein). At low cell concentrations (0.5 mg protein/ml) 1 mol pyruvate was converted to 1 mol CH4, 2 mol CO2 and 1 mol H2. At higher cell concentration less H2 and CO2 and more CH4 were formed due to CH4 formation from H2/CO2. The rate of pyruvate conversion was linearily dependent on the pyruvate concentration up to about 30 mM. Cell suspensions of acetate-grown M. barkeri also catalyzed the conversion of 1 mol pyruvate to 1 mol CH4, 2 mol CO2 and 1 mol H2 at similar rates and with similar affinity for pyruvate as pyruvate-grown cells.
  3. Cell extracts of both pyruvate-grown and acetate-grown M. barkeri contained pyruvate: ferredoxin oxidoreductase. The specific activity in pyruvate-grown cells (0.8 U/mg) was 8-fold higher than in acetate-grown cells (0.1 U/mg). Coenzyme F420 was excluded as primary electron acceptor of pyruvate oxidoreductase. Cell extracts of pyruvate-grown M. barkeri contained carbon monoxide dehydrogenase activity and hydrogenase activity catalyzing the reduction by carbon monoxide and hydrogen of both methylviologen and ferredoxin (from Clostridium).
This is the first report on growth of a methanogen on pyruvate as sole carbon and energy source, i.e. on a substrate more complex than acetate.  相似文献

Microbial processes influencing methane emission from rice fields   总被引:7,自引:0,他引:7  
Irrigated rice fields are an important source of atmospheric methane. In order to improve our understanding of the controlling processes, we measured in situ CH4 emission and CH4 oxidation in an Italian rice field in 1998 and 1999, and studied CH4 production in soil and root samples. The CH4 emission rates were correlated with diurnal temperature variations and showed pronounced seasonal and interannual variations. The contribution of CH4 oxidation to total CH4 flux, determined by specific inhibition with difluoromethane, decreased from 40% at the beginning to zero at the end of the season. The stable carbon isotopic composition of the emitted CH4 also decreased. The CH4‐oxidizing bacteria probably became limited by nitrogen as indicated by the seasonal decrease of NH4+. Thus, CH4 oxidation had little effect on CH4 emission. Methane production on rice roots was relatively constant over the season. Methane production in soil slowly increased after flooding and was highest in the middle of the season. Pore water concentrations of CH4 showed a similar seasonal pattern. In 1999, CH4 production increased later in the season and reached lower rates than in 1998. An additional drainage in 1999 resulted in higher ferric iron concentrations, higher soil redox potentials and lower acetate concentrations. As a result, acetate‐utilizing methanogens were probably out‐competed by iron‐reducers so that a larger percentage of [2–14C]acetate was converted to 14CO2 instead of 14CH4. The residual CH4 production was relatively low and was mainly due to H2/CO2‐dependent methanogenesis. Experiments with radioactive bicarbonate and with methyl fluoride as specific inhibitor showed that the theoretical ratio of 7:3 of methanogenesis from acetate vs. H2/CO2 was only reached later in the season when total CH4 production was at the maximum. In conclusion, our results give a mechanistic explanation for the intraseasonal and interannual differences in CH4 emission.  相似文献

Processes involved in formation and emission of methane in rice paddies   总被引:40,自引:9,他引:31  
The seasonal change of the rates of production and emission of methane were determined under in-situ conditions in an Italian rice paddy in 1985 and 1986. The contribution to total emission of CH4 of plant-mediated transport, ebullition, and diffusion through the flooding water was quantified by cutting the plants and by trapping emerging gas bubbles with funnels. Both production and emission of CH4 increased during the season and reached a maximum in August. However, the numbers of methanogenic bacteria did not change. As the rice plants grew and the contribution of plant-mediated CH4 emission increased, the percentage of the produced CH4 which was reoxidized and thus, was not emitted, also increased. At its maximum, about 300 ml CH4 were produced per m2 per hour. However, only about 6% were emitted and this was by about 96% via plant-mediated transport. Radiotracer experiments showed that CH, was produced from H2/CO2. (30–50%) and from acetate. The pool concentration of acetate was in the range of 6–10 mM. The turnover time of acetate was 12–16 h. A representative of the photosynthetic non-sulfur purple bacteria (Athiorhodaceae) capable of using simple alcohols has been isolated in pure culture. By means of quantitative analysis of cultures at different stages of development it has been shown that this organism converts isopropanol quantitatively into acetone, simultaneously reducing CO2 in the light. The data can be represented by the equation 2 CH3CHOHCH3 + CO2 → 2 CH3COCH3 + (CH2O) + H2O. Manometric experiments with suspensions of resting cells have fully corroborated the results obtained with growing cultures. The experiments have conclusively proved that an organic substrate may fulfill exclusively the function of hydrogen donor for the photochemical CO2-reduction in purple bacteria photosynthesis.  相似文献

Gut microbiota from Reticulitermes flavipes termites catalyzed an H2-dependent total synthesis of acetate from CO2. Rates of H2-CO2 acetogenesis in vitro were 1.11 ± 0.37 μmol of acetate g (fresh weight)−1 h−1 (equivalent to 4.44 ± 1.47 nmol termite−1 h−1) and could account for approximately 1/3 of all the acetate produced during the hindgut fermentation. Formate was also produced from H2 + CO2, as were small amounts of propionate, butyrate, and lactate-succinate. However, H2-CO2 formicogenesis seemed largely unrelated to acetogenesis and was believed not to be a significant reaction in situ. Little or no CH4 was formed from H2 + CO2 or from acetate. H2-CO2 acetogenesis was inhibited by O2, KCN, CHCl3, and iodopropane and could be abolished by prefeeding R. flavipes with antibacterial drugs. By contrast, prefeeding R. flavipes with starch resulted in almost complete defaunation but had little effect on H2-CO2 acetogenesis, suggesting that bacteria were the acetogenic agents in the gut. H2-CO2 acetogenesis was also observed with gut microbiota from Prorhinotermes simplex, Zootermopsis angusticollis, Nasutitermes costalis, and N. nigriceps; from the wood-eating cockroach Cryptocercus punctulatus; and from the American cockroach Periplaneta americana. Pure cultures of H2-CO2-acetogenic bacteria were isolated from N. nigriceps, and a preliminary account of their morphological and physiological properties is presented. Results indicate that in termites, CO2 reduction to acetate, rather than to CH4, represents the main electron sink reaction of the hindgut fermentation and can provide the insects with a significant fraction (ca. 1/3) of their principal oxidizable energy source, acetate.  相似文献

Emission rates of CH4 were measured in microcosms of submerged soil which were planted with rice. Drainage of the rice microcosms for 48 h resulted in drastically decreased CH4 emission rates which only slowly recovered to the rates of the undrained controls. Drainage also resulted in drastically increased sulphate concentrations which only slowly decreased to nearly zero background values after the microcosms were submerged again. The mechanisms responsible for the decrease of CH4 production by aeration were investigated in slurries of a loamy and a sandy Italian rice soil. Incubation of the soil slurries under anoxic conditions resulted first in the reduction of nitrate, sulphate and ferric iron before CH4 production started. Incubation of the soil slurries for 48 h under air resulted in immediate and complete inhibition of CH4 production. Although the soil slurries were then again incubated under anoxic conditions (N2 atmosphere), the inhibition of CH4 production persisted for more than 30 days. The redox potential of the soil increased after the aeration but returned within 15 days to the low values typical for CH4 production. However, the concentrations of sulphate and of ferric iron increased dramatically after the aeration and stayed at elevated levels for the period during which CH4 production was inhibited. These observations show that even brief exposure of the soil to O2 allowed the production of sulphate and ferric iron from their reduced precursors. Elevated sulphate and ferric iron concentrations allowed sulphate-reducing and ferric iron-reducing bacteria to outcompete methanogenic bacteria on H2 as common substrate. Indeed, concentrations of H2 were decreased as long as sulphate and ferric iron were high so that the Gibbs free energy of CH4 production from H2/CO2 was also increased (less exergonic). On the other hand, concentrations of acetate, the more important precursor for CH4, were not much affected by the short aeration of the soil slurries, and the Gibbs free energy of CH4 production from acetate was highly exergonic suggesting that acetotrophic methanogens were not outcompeted but were otherwise inhibited. Aeration also resulted in increased rates of CO2 production and in a short-term increase of N2O production. However, these increases were < 10% of the decreased production of CH4 and did not represent a trade-off in terms of CO2 equivalents. Hence, short-term drainage and aeration of submerged paddy fields may be a useful mitigation option for decreasing the emission of greenhouse gases.  相似文献

Many feeding trials have been conducted to quantify enteric methane (CH4) production in ruminants. Although a relationship between diet composition, rumen fermentation and CH4 production is generally accepted, the efforts to quantify this relationship within the same experiment remain scarce. In the present study, a data set was compiled from the results of three intensive respiration chamber trials with lactating rumen and intestinal fistulated Holstein cows, including measurements of rumen and intestinal digestion, rumen fermentation parameters and CH4 production. Two approaches were used to calculate CH4 from observations: (1) a rumen organic matter (OM) balance was derived from OM intake and duodenal organic matter flow (DOM) distinguishing various nutrients and (2) a rumen carbon balance was derived from carbon intake and duodenal carbon flow (DCARB). Duodenal flow was corrected for endogenous matter, and contribution of fermentation in the large intestine was accounted for. Hydrogen (H2) arising from fermentation was calculated using the fermentation pattern measured in rumen fluid. CH4 was calculated from H2 production corrected for H2 use with biohydrogenation of fatty acids. The DOM model overestimated CH4/kg dry matter intake (DMI) by 6.1% (R2=0.36) and the DCARB model underestimated CH4/kg DMI by 0.4% (R2=0.43). A stepwise regression of the difference between measured and calculated daily CH4 production was conducted to examine explanations for the deviance. Dietary carbohydrate composition and rumen carbohydrate digestion were the main sources of inaccuracies for both models. Furthermore, differences were related to rumen ammonia concentration with the DOM model and to rumen pH and dietary fat with the DCARB model. Adding these parameters to the models and performing a multiple regression against observed daily CH4 production resulted in R2 of 0.66 and 0.72 for DOM and DCARB models, respectively. The diurnal pattern of CH4 production followed that of rumen volatile fatty acid (VFA) concentration and the CH4 to CO2 production ratio, but was inverse to rumen pH and the rumen hydrogen balance calculated from 4×(acetate+butyrate)/2×(propionate+valerate). In conclusion, the amount of feed fermented was the most important factor determining variations in CH4 production between animals, diets and during the day. Interactions between feed components, VFA absorption rates and variation between animals seemed to be factors that were complicating the accurate prediction of CH4. Using a ruminal carbon balance appeared to predict CH4 production just as well as calculations based on rumen digestion of individual nutrients.  相似文献

Rumen bacteria retained methanogenic activity when stored at ?60° under H2. This activity, which resides in Methanobacterium ruminantium and Methanobacterium mobilis, is not lost when the cells are broken, as has been suggested. Unlike in Methanosarcina barkerii and Methanobacterium M.o.H., in rumen bacteria methanogenic enzymes are not soluble but readily precipitated at 15,000 g. Methane was synthesized from tetrahydrofolate derivatives but at slower rates than from CO2. From the data, it was not possible to determine if methyl- and methylene tetrahydrofolate were oxidized to CO2 prior to reduction to CH4. In room light, CH3-B12 was reduced to CH4 non-enzymatically in the presence of protein. When the reactions were carried out in the dark, very little CH4 was formed from CH3-B12 by rumen bacterial enzymes. The cell-free particulate fraction did not require added ATP for methanogenesis but showed an absolute requirement for H2.  相似文献

The metabolic pathways involved in hydrogen (H2) production, utilization and the activity of methanogens are the important factors that should be considered in controlling methane (CH4) emissions by ruminants. H2 as one of the major substrate for CH4 production is therefore should be controlled. One of the strategies on reducing CH4 is through the use of hydrogenotrophic microorganisms such as fumarate reducing bacteria. This study determined the effect of fumarate reducing bacteria, Mitsuokella jalaludinii, supplementation on in vitro rumen fermentation, CH4 production, diversity and quantity. M. jalaludinii significantly reduced CH4 at 48 and 72 h of incubation and significantly increased succinate at 24 h. Although not significantly different, propionate was found to be highest in treatment containing M. jalaludinii at 12 and 48 h of incubation. These results suggest that supplementation of fumarate reducing bacteria to ruminal fermentation reduces CH4 production and quantity, increases succinate and changes the rumen microbial diversity.  相似文献

Flooded rice fields, which are an important source of the atmospheric methane, have become a model system for the study of interactions between various microbial processes. We used a combination of stable carbon isotope measurements and application of specific inhibitors in order to investigate the importance of various methanogenic pathways and of CH4 oxidation for controlling CH4 emission. The fraction of CH4 produced from acetate and H2/CO2 was calculated from the isotopic signatures of acetate, carbon dioxide (CO2) and methane (CH4) measured in porewater, gas bubbles, in the aerenchyma of the plants and/or in incubation experiments. The calculated ratio between both pathways reflected well the ratio determined by application of methyl fluoride (CH3F) as specific inhibitor of acetate‐dependent methanogenesis. Only at the end of the season, the theoretical ratio of acetate: H2 = 2 : 1 was reached, whereas at the beginning H2/CO2‐dependent methanogenesis dominated. The isotope discrimination was different between rooted surface soil and unrooted deep soil. Root‐associated CH4 production was mainly driven by H2/CO2. Porewater CH4 was found to be a poor proxy for produced CH4. The fraction of CH4 oxidised was calculated from the isotopic signature of CH4 produced in vitro compared to CH4 emitted in situ, corrected for the fractionation during the passage from the aerenchyma to the atmosphere. Isotope mass balances and in situ inhibition experiments with difluoromethane (CH2F2) as specific inhibitor of methanotrophic bacteria agreed that CH4 oxidation was quantitatively important at the beginning of the season, but decreased later. The seasonal pattern was consistent with the change of potential CH4 oxidation rates measured in vitro. At the end of the season, isotope techniques detected an increase of oxidation activity that was too small to be measured with the flux‐based inhibitor technique. If porewater CH4 was used as a proxy of produced CH4, neither magnitude nor seasonal pattern of in situ CH4 oxidation could be reproduced. An oxidation signal was also found in the isotopic signature of CH4 from gas bubbles that were released by natural ebullition. In contrast, bubbles stirred up from the bulk soil had preserved the isotopic signature of the originally produced CH4.  相似文献

A simple coupled ecosystem-climate model is described that canpredict levels of atmospheric CH4, CO2, and H2during the Late Archean, given observed constraints on Earth'ssurface temperature. We find that methanogenic bacteria shouldhave converted most of the available atmospheric H2 intoCH4, and that CH4 may have been equal in importance to CO2 as a greenhouse gas. Photolysis of this CH4 may have produced a hydrocarbon smog layer that would have shielded the surface from solar UV radiation. Methanotrophic bacteria would have consumed some of the atmospheric CH4,but they would have been incapable of reducing CH4 to modern levels. The rise of O2 around 2.3 Ga would have drastically reduced the atmospheric CH4 concentrationand may thereby have triggered the Huronian glaciation.  相似文献

Methane produced from formate is one of the important methanogensis pathways in the rumen. However, quantitative information of CH4 production from formate has been rarely reported. The aim of this study was to characterize the conversion rate (CR) of formic acid into CH4 and CO2 by rumen microorganisms. Ground lucerne hay was incubated with buffered ruminal fluid for 6, 12, 24 and 48 h. Before the incubation, 13C-labeled H13COOH was also supplied into the incubation bottle at a dose of 0, 1.5, 2.2 or 2.9 mg/g of DM substrate. There were no interactions (P>0.05) between dose and incubation time for all variables evaluated. When expressed as an absolute amount (ml in gas sample) or a relative CR (%), both 13CH4 and 13CO2 production quadratically increased (P<0.01) with the addition of H13COOH. The total 13C (13CH4 and 13CO2) CR was also quadratically increased (P<0.01) when H13COOH was added. Moreover, formate addition linearly decreased (P<0.031) the concentrations of NH3-N, total and individual volatile fatty acids (acetate, propionate and butyrate), and quadratically decreased (P<0.014) the populations of protozoa, total methanogens, Methanosphaera stadtmanae, Methanobrevibacter ruminantium M1, Methanobrevibacter smithii and Methanosarcina barkeri. In summary, formate affects ruminal fermentation and methanogenesis, as well as the rumen microbiome, in particular microorganisms which are directly or indirectly involved in ruminal methanogenesis. This study provides quantitative verification for the rapid dissimilation of formate into CH4 and CO2 by rumen microorganisms.  相似文献

Agriculture has marked impacts on the production of carbon dioxide (CO2) and consumption of methane (CH4) by microbial communities in upland soils—Earth''s largest biological sink for atmospheric CH4. To determine whether the diversity of microbes that catalyze the flux of these greenhouse gases is related to the magnitude and stability of these ecosystem-level processes, we conducted molecular surveys of CH4-oxidizing bacteria (methanotrophs) and total bacterial diversity across a range of land uses and measured the in situ flux of CH4 and CO2 at a site in the upper United States Midwest. Conversion of native lands to row-crop agriculture led to a sevenfold reduction in CH4 consumption and a proportionate decrease in methanotroph diversity. Sites with the greatest stability in CH4 consumption harbored the most methanotroph diversity. In fields abandoned from agriculture, the rate of CH4 consumption increased with time along with the diversity of methanotrophs. Conversely, estimates of total bacterial diversity in soil were not related to the rate or stability of CO2 emission. These combined results are consistent with the expectation that microbial diversity is a better predictor of the magnitude and stability of processes catalyzed by organisms with highly specialized metabolisms, like CH4 oxidation, as compared with processes driven by widely distributed metabolic processes, like CO2 production in heterotrophs. The data also suggest that managing lands to conserve or restore methanotroph diversity could mitigate the atmospheric concentrations of this potent greenhouse gas.  相似文献

The number of microorganisms of major metabolic groups and the rates of sulfate reduction and methanogenesis processes in the formation waters of the high-temperature horizons of Dagang oil field have been determined. Using cultural methods, it was shown that the microbial community contained aerobic bacteria oxidizing crude oil, anaerobic fermentative bacteria, sulfate-reducing bacteria, and methanogens. Using cultural methods, the possibility of methane production from a mixture of hydrogen and carbon dioxide (H2 + CO2) and from acetate was established, and this result was confirmed by radioisotope methods involving NaH14CO3 and 14CH3COONa. Analysis of enrichment cultures 16S rDNA of methanogens demonstrated that these microorganisms belong to Methanothermobacter sp. (M. thermautotrophicus), which consumes hydrogen and carbon dioxide as basic substrates. The genes of acetate-utilizing bacteria were not revealed. Phylotypes of the representatives of Thermococcus spp. were found among archaeal 16S rDNA. 16S rRNA genes of bacterial clones belong to the orders Thermoanaerobacteriales (Thermoanaerobacter, Thermovenabulum, Thermacetogenium, and Coprothermobacter spp.), Thermotogales, Nitrospirales (Thermodesulfovibrio sp.) and Planctomycetales. 16S rDNA of a bacterium capable of oxidizing acetate in the course of syntrophic growth with H2-utilizing methanogens was found in high-temperature petroleum reservoirs for the first time. These results provide further insight into the composition of microbial communities of high-temperature petroleum reservoirs, indicating that syntrophic processes play an important part in acetate degradation accompanied by methane production.  相似文献
