  • 1 Four sets of experiments were devised to evaluate the absolute size of aedine larval populations. Three methods were used and compared: ‘capture-recapture’, ‘removal’ and ‘dipping’ methods.

A calibration method is described for measuring absolute radioautographic sensitivities under various experimental conditions. Sensitivities to 3H and 35S radiation, i.e. ratio of developed grains to radioactive decays in the specimen, were determined with Ilford L4 and Kodak NTE emulsions. The highest values obtained in monolayers of emulsion were ⅛ for 3H and 1/21 for 35S. The influence of various experimental parameters on sensitivity is described, and the possibilities for quantitative electron microscope radioautography are discussed.  相似文献

  • 1 Population density alone is unlikely to be a good predictor of the impacts of deer on their environment. The assessment of management requirements should therefore be based on assessment of deer impacts, alongside estimates of density.
  • 2 Both density and impacts need to be monitored at a landscape scale, and there is a need to develop appropriate methodologies that allow managers to consider the current and likely future impact of deer.
  • 3 The relevant scale for assessment (and management) varies both with deer species and context of impact, but should always encompass at least the estimated biological range of the population of deer present in an area. Some impacts (e.g. deer–vehicle collisions, and risks of disease transmission) may need to be assessed at a wider regional level.
  • 4 In this review we consider various approaches available for assessing: absolute or relative animal abundance; impacts of ungulates on agriculture, forestry, amenity woodlands and other conservation sites; impacts on public safety (e.g. through road traffic accidents) and on humans or livestock through potential spread of disease.
  • 5 In each case the advantages and disadvantages of a variety of methods are considered, before recommendations are made for methodologies which are sufficiently accurate, sufficiently robust and sufficiently practical to be favoured in a management context.
  • 6 To address impacts at the landscape scale requires management policies that integrate information on both positive and negative impacts of deer in order to ensure appropriate and holistic management. We present a decision‐support framework suitable for use within the UK, using inputs from a variety of possible impact types to assist managers and forewarn of situations where current management may need to be modified.

  • 1 At high densities, deer populations may have adverse effects upon and within their environment. In this review we explore published and unpublished information to derive density thresholds for deer species in relation to impacts upon agriculture, forestry, conservation habitats, road traffic, and human and livestock health in the UK. Impact levels are affected by many factors other than absolute density. We therefore seek to establish the range of densities within which negative impacts might start to occur and which should trigger objective monitoring of actual impacts.
  • 2 In commercial forestry, a threshold of 4 deer per 100ha has been suggested. Unfenced native woodlands seem to regenerate naturally if there are fewer than 4–5 large deer or fewer than 25 roe deer Capreolus capreolus per 100ha; open habitats may suffer only light or moderate impacts from red deer Cervus elaphus at landscape densities of 7–8 per 100ha.
  • 3 Woodland bird species may have declined where deer densities are high but absolute thresholds seem impossible to establish. One study suggests maximum diversity at about 8 white‐tailed deer Odocoileus virginianus per 100ha.
  • 4 Deer–vehicle collisions are affected by various factors in addition to deer density, but British and American studies suggest that accident frequencies decline at densities below 7–8 per 100ha.
  • 5 Fallow deer Dama dama populations may maintain bovine TB (bTB) infection at much lower densities (25/100ha) than red or roe deer (91/100ha and 200/100ha, respectively) assuming 100% prevalence. Even at 30% prevalence a density of 75 fallow deer per 100ha could maintain bTB within the population.
  • 6 We conclude that deer density alone is unlikely to be a good predictor of impact, and suggest that long‐term management should be based on assessment both of actual impacts and apparent density of deer.

The quantitative determination of sarcosine is of great importance in clinical chemistry, food and fermentation industries. Elevated sarcosine levels are associated with Alzheimer, dementia, prostate cancer, colorectal cancer, stomach cancer and sarcosinemia. This review summarizes the various methods for quantitative analysis of sarcosine with special emphasis on various strategies of biosensors and their analytical performance. The current bio sensing methods have overcome the drawbacks of conventional methods. Sarcosine biosensors work optimally at pH 7.0 to 8.0 in the linear range of 0.1 to 100?μM within 2 to 17?s and between 25 and 37?°C, within a limit of detection (LOD) between 0.008 and 500?mM. The formulated biosensors can be reused within a stability period of 3–180?days. Future research could be focused to modify existing sarcosine biosensors, leading to simple, reliable, and economical sensors ideally suited for point-of-care treatment.

  • 1 As the ephippia (chitinous shells enclosing diapausing eggs) of pelagic crustaceans of the genus Daphnia have been occasionally reported to float at the water surface, we considered that this might be an adaptation promoting their passive dispersal. We investigated the mechanisms by which ephippia appear at the water surface.
  • 2 While field surveys revealed that floating Daphnia ephippia are often numerous in various freshwater habitats, laboratory tests showed that newly formed ephippia are not buoyant initially. Once transferred to the surface by whatever means, however, they may remain there due either to surface tension or gas absorption.
  • 3 Video recordings showed that all ephippia at the water surface in laboratory vessels were shed there by ephippial females when moulting (despite the attendant risk of exposure to UV radiation). This implies that the moulting behaviour of female Daphnia may determine the fate of their dormant offspring, predetermining whether they remain in the natal environment (when the ephippium is released into the water column) or disperse (when it is deposited at the water surface).
  • 4 Our findings reveal a potential mechanism underlying the high dispersal capacity of freshwater cladocerans inhabiting island‐like aquatic habitats.

  1. Cyanobacteria are one of the oldest organisms on Earth and they originated at a time when damaging ultraviolet (UV) C radiation still reached the surface. Their long evolution led to several adaptations to avoid deleterious effects caused by exposure to solar UV radiation. Synthesis of sunscreen substances, such as mycosporine‐like amino acids (MAAs), allows them to photosynthesise with reduced risk of cell damage. The interplay of solar UV radiation and MAAs is well documented for cyanobacteria in the plankton realm, but little is known for those in the benthic realm, particularly of clear alpine lakes.
  2. Here, we assessed the temporal dynamics of MAAs in the benthic algal community of one clear alpine lake dominated by cyanobacteria during the ice‐free season and along a depth gradient using state‐of‐the‐art analytical methods (high‐performance liquid chromatography, nuclear magnetic resonance, liquid chromatography–mass spectrometry). We differentiated between the epilithic cyanobacterial community and the overlying loosely attached filamentous cyanobacteria, as we expected they will have an important shielding/shading effect on the former. We hypothesised that in contrast to the case of phytoplankton, benthic cyanobacteria will show less pronounced temporal changes in MAAs concentration in response to changes in solar UV exposure.
  3. Three UV‐absorbing substances were present in both types of communities, whereby all were unknown. The chemical structure of the dominant unknown substance (maximum absorption at 334 nm) resulted in the identification of a novel MAA that we named aplysiapalythine‐D for its similarity to the previously described aplysiapalythine‐C.
  4. Chlorophyll‐a‐specific MAA concentrations for epilithic and filamentous cyanobacteria showed a significant decrease with depth, although only traces were found in the former community. The temporal dynamics in MAA concentrations of filamentous cyanobacteria showed no significant variations during the ice‐free season.
  5. Our result on the low temporal MAA dynamics agrees with the reduced growth rates of benthic cyanobacteria reported for cold ecosystems. The permanent presence of this community, which is adapted to the high UV levels characteristic of clear alpine lakes, probably represents the most important primary producers of these ecosystems.

Assessing the spatial structure of abundance of a species is a basic requirement to carry out adequate conservation strategies. However, existing attempts to predict species abundance, particularly in absolute units and on large scales, are scarce and have led to weak results. In this work we present a scheme to obtain, in an affordable way, a predictive model of absolute animal abundance on large scales based on the modelling of data obtained from local ecological knowledge (LEK) and its calibration. To exemplify this scheme, we build and validate a predictive absolute abundance model of the endangered terrestrial tortoise Testudo graeca in Southeast Iberian Peninsula. For that purpose, we collected distribution and relative abundance data of T. graeca using a low cost methodology, such as LEK, by means of interviewing shepherds. The information from LEK was employed to build a predictive habitat-based model of relative abundance. The relative abundance model was transformed into an absolute abundance model by means of calibration with a classical absolute abundance sampling method such as distance sampling. The obtained absolute abundance model predicted the observed absolute abundances values well in independent locations when compared with other works (R 2 = 36%) and thus can offer a cost-effective predictive ability. Our results show that reliable habitat-based predictive maps of absolute species abundance on regional scales can be obtained starting from low cost sampling methods of relative abundance, such as LEK, and its calibration.  相似文献

  • 1 Previous work has shown that female Callosobruchus maculatus (F.) (Coleoptera: Bruchidae) can accurately assess the number of eggs on a seed and use this information to produce a near uniform distribution of egg-loads, so minimizing larval competition within hosts.
  • 2 The decision to lay on a particular seed may be based on one of two possible rules: (i) a‘relative’rule, which compares the number of eggs on the present seed with that on previously encountered seeds; and (ii) an‘absolute’rule, which takes into account the current egg-load alone.
  • 3 This paper describes an experimental study of oviposition behaviour which concludes that an absolute measure of egg-load probably determines egg-laying decisions. The implications of this result are discussed.

ATP methodology needs to be further standardized and improved in order to avoid the pitfalls that have sometimes hampered its application to biomass assays. The following steps have been reconsidered as far as the bacteriological applications is concerned:
  • destruction of free and somatic ATP: replacement of apyrase by mammalian ATPase, more readily accessible to specific inhibition;
  • extraction of bacterial ATP: protection of luciferase by lipids against inhibitory effect of cationic detergents with production of a constant light response.
New methods are proposed for the calibration of luminometers and for the matching of sample holders in multichannel instruments. The limit of sensitivity of ATP assays is discussed in the light of currently available reagents and instruments.  相似文献

  • 1 Ultraviolet radiation (UVR) damages early life stages of several fish species. Galaxias maculatus is a small catadromous fish, with landlocked forms occurring in many lakes within the Nahuel Huapi National Park (Patagonia, Argentina). In this work, the vulnerability of G. maculatus eggs exposed to both natural and artificial UVR was investigated in relation to water transparency.
  • 2 Field experiments were performed in two lakes differing in UVR attenuation. Galaxias maculatus eggs were exposed to in situ levels of UVR in quartz tubes incubated at various depths. For laboratory experiments, the eggs were exposed to five levels of artificial UVB radiation.
  • 3 Exposure to natural UVR causes various degrees of egg mortality depending on water transparency and incubation depth. In the less transparent lake (Kd320 = 3.08 m‐1), almost complete mortality was observed near the surface. At a depth of 43 cm the observed mortality was only 22%, but was still significantly different from the dark control. In the most transparent lake (Kd320 = 0.438 m‐1), almost total mortality was observed in tubes incubated at 2.56 m or shallower. A gradual decline in mortality was recorded from that depth to 3.78 m where the values approached those in the dark control treatments.
  • 4 A monotonic relationship between mortality and UV exposure could be observed both in field and laboratory experiments. Using the results from field incubations, a LD50 of 2.5 J cm‐2 nm‐1 was estimated. In a few mountain lakes, this value would be exceeded even if the eggs were laid at the maximum depth of the lake. Thus UVR seems a sufficient cause to explain the absence of G. maculatus populations in some mountain lakes. For most lakes, however, UVR is probably one of several important environmental factors, which together determine the habitat suitability.

  • 1 In this paper we investigate whether the technique of discriminant analysis can be used to estimate sampling biases for female tsetse.
  • 2 Discriminant analysis was first applied to laboratory samples of female tsetse, Glossina morshans morsitans Westwood, to test whether flies of known history could be assigned to the correct day of the pregnancy cycle on the basis of their fat, haematin and corrected residual dry weight.
  • 3 Following the satisfactory results from the laboratory samples, the same technique was applied to field samples of G.m. centralis Machado captured by electric traps and hand nets in Zambia and of G.palpalis palpalis (Robineau-Desvoidy) captured in biconical traps at five sites in Ivory Coast. The results show that flies on day 1 of the pregnancy cycle were most likely to be caught, with a second peak of day-6 and day-7 flies, while very few day-8 or day-9 flies were caught.
  • 4 These major peaks in fly trappability coincide with the known feeding habits of female tsetse, and indicate synchrony of feeding by many members of the population immediately after larviposition and again as the larva in utero moults from the second to third instar. G.palpalis is relatively more available at this later stage of its pregnancy cycle to the capture methods used than is G.morsitans. A third feed may be taken at a more variable point in the pregnancy cycle.
  • 5 This method of estimating the sampling biases of female tsetse could allow an estimate of total population size, as long as the absolute sampling efficiency of flies on any one day of the pregnancy cycle could be established by, for example, mark-release-recapture experiments.

ABSTRACT.
  • 1 All the insects visiting Tilia and Heracleum flowers showed diel patterns of activity, which could best be explained in terms of weather. Of the recorded parameters, solar radiation gave the best correlations with activity.
  • 2 Correlations between activity and radiation depended on the size and colour of particular groups of insects; small bright species visited fully insolated flowers, (r high), while large dark forms visited early and late in the day (r low or negative).
  • 3 Consequently mean reflectance and mean weight of foragers showed consistent trends through a day and predictable relations with radiation for both flowers, suggesting that thermal costs are a critical determinant of feeding times.
  • 4 By contrast, the nectar reward (as sugar amounts) in flowers was poorly correlated with timing of insect visits, indicating that caloric reward is not limiting; although nectar concentration (only measurable for Tilia) did correlate reasonably well with total visitation.
  • 5 The exception to this pattern was Bombus. This endothermic species visited early in the day when nectar was abundant. Thus visits correlated well with reward and negatively with radiation, showing a dependence on high rewards to offset thermoregulatory metabolic costs.
  • 6 For most insects foraging appeared to be largely determined by the visitors thermal requirements and the restrictions imposed by avoidance of overheating. Nectar rewards were not critical, and nectar concentration, itself dependent upon climatic parameters, was probably only incidentally correlated with diel patterning for all but the largest (endothermic) insects.

A kinetic model of the processes occurring in the plasma of a high-power low-pressure gas-discharge lamp is presented, and the output characteristics of the lamp are described. The lamp is excited by a longitudinal glow discharge and emits the I2(D′-A′) 342-nm and XeI(B-X) 253-nm bands and the 206.2-nm spectral line of atomic iodine. When the emitter operates in a sealed-off mode on the p(He): p(Xe): p(I2) = 400: 120: (100–200) Pa mixture, the fractions of the UV radiation power of iodine atoms, exciplex molecules of xenon iodide, and iodine molecules comprise 55, 10, and 35%, respectively. At the optimal partial pressure, the maximum total radiation power of the lamp reaches 37 W, the energy efficiency being about 15%.  相似文献
