Twenty-seven properties considered to be required for good bakery products were examined in 56 industrial and 2 laboratory yeast strains. The data obtained were applied to principal-component analysis, one of the multivariate statistical analyses. The first and second principal components together were extracted, and these accounted for 77.7% of the variance. The first principal component was interpreted as the glycolytic activity of yeast in dough, and the second one was interpreted as the balance of leavening abilities in sweet and flour doughs from the factor loadings. The scattergram on the two principal components was effective in grouping the 58 yeast strains used.  相似文献

This study examines the interactions that occur between Saccharomyces cerevisiae and Oenococcus oeni strains during the process of winemaking. Various yeast/bacteria pairs were studied by applying a sequential fermentation strategy which simulated the natural winemaking process. First, four yeast strains were tested in the presence of one bacterial strain leading to the inhibition of the bacterial component. The extent of inhibition varied widely from one pair to another and closely depended on the specific yeast strain chosen. Inhibition was correlated to weak bacterial growth rather than a reduction in the bacterial malolactic activity. Three of the four yeast strains were then grown with another bacteria strain. Contrary to the first results, this led to the bacterial stimulation, thus highlighting the importance of the bacteria strain. The biochemical profile of the four yeast fermented media exhibited slight variations in ethanol, SO(2) and fatty acids produced as well as assimilable consumed nitrogen. These parameters were not the only factors responsible for the malolactic fermentation inhibition observed with the first bacteria strain. The stimulation of the second has not been reported before in such conditions and remains unexplained.  相似文献

The data on cranial measurements performed in silver foxes indicate that there are differences in sizes measured between the farm foxes--bred population and the population selected for domestication. A method of principal components was used to analyse the cranial measurements and their changes under domestication. The first component covers about 50% of cranial diversity, which is interpreted as variation in the total skull size. This component clearly separates the two sexes, but not different populations. The second component presumably reflects the growth rate allometry between the skull length and width. The third and fourth components are measurements of skull width; the fifth one reflects the sizes of brain skull. None of these components clearly separate the foxes from farm--bred and domesticated populations. However, some differences in distribution are observed.  相似文献

Multiple-group principal component analysis was used to investigate morphological differences between skulls of male and female hooded seals. The scores on the first principal axis (PCI), calculated from the variance-covariance matrix of log-transformed data, were highly correlated with age and the component was interpreted as a general growth axis. The other components had correlations between dimensions and component scores of different signs and magnitude and were interpreted as shape axes. The standardized component scores were subjected to analyses of variance and discriminant analyses. Sexual dimorphism was disclosed on eight of the components including PCI, and was functionally related mostly to the hood of the males.  相似文献

Principal component maps of the gene arrangement frequencies of 108 natural populations in Europe, North Africa and the Middle East were prepared to investigate the evolutionary forces shaping the geographic variation of inversion frequencies. Principal component maps were also prepared from ten climate variables at 347 localities of the same region. The first inversion principal component (18% of total variation) showed a N-S cline strikingly similar to the pattern exhibited by the first principal component of climatic variables. This resemblance is interpreted as showing the outcome of a selective process, which favors the increase in frequency of the Standard gene arrangements when moving to the north. This interpretation is corroborated by the fact that such clines were formed in South and North America, following recent colonization by this species. Patterns shown by the second (12% of total variance) and third (8% of total variance) principal components are interpreted as related to historical events, the migrational advance to the north after the end of the last glaciation and the locations of the species refugia at that time.  相似文献

Multiple group principal component analysis was used to investigate the morphological relationships between feral American mink from three areas of Southern Norway based on 15 skull dimensions. The first principal axis calculated from the variance-covariance matrix of log-transformed data is usually interpreted as a growth axis. In the present analysis, the scores of the first principal axis of males were found to be uncorrelated with estimated age for specimens younger than seven months old, and was interpreted as a growth-free size axis. However, the second component of males was found to reflect mainly growth in the postorbital area. The first component of females was thought to reflect a mixing of growth-out and growth-in size variation. One of the reasons for separating size and size-free variation in the present work was to test whether growth-dependent size influenced the assessment of geographic variation within each sex. The scores on the components reflecting size variation (growthout and growth-in) were, however, not found to deviate significantly between the areas. Discriminant analyses run on the component scores, both with size variation included and excluded, gave the same result. In mean, the mink from Western Norway deviated significantly from the mink from Trondelag and Eastern Norway, while the mink from the two latter areas did not deviate morphometrically. The deviation found was in shape rather than in size of both sexes.  相似文献

Summary Production of phenylacetylcarbinol (PAC) was measured in various yeast species. The yeast strains tested were cultivated under submerged conditions in a medium containing corn steep and sucrose as the main components; sucrose, acetaldehyde and benzaldehyde were added to the grown cultures. In a first series of experiments the initial rate of PAC production, i.e. the PAC production determined 30 min after the addition of benzaldehyde was determined in 38 yeast strains, mostly of the generaSaccharomyces andCandida. The amount of PAC produced varied from zero (12 strains) to 1.24 mg ml–1. In a second series of experiments, 15 strains, which in the first series had shown a higher PAC production, were further tested. Sucrose, acetaldehyde and benzaldehyde were added to the cultures until the PAC production ceased. The highest PAC production (6.3 mg ml–1) was reached in the strainSaccharomyces carlsbergensis Budvar; the production was slightly lower in 4 strains of the generaSaccharomyces, Candida andHansenula.  相似文献

Korean traditional rice wines yakju and takju are generally brewed with nuruk as the source of the saccharogenic enzymes by natural fermentation. To improve the quality of Korean rice wine, the microorganisms in the nuruk need to be studied. The objective of this research was to improve the quality of Korean wine with the wild-type yeast strains isolated from the fermentation starter, nuruk. Only strain YA-6 showed high activity in 20% ethanol. Precipitation of Y89-5-3 was similar to that of very flocculent yeast (?80%) at 75.95%. Using 18S rRNA sequencing, all 10 strains were identified as Saccharomyces cerevisiae. Volatile compounds present in yakju were analyzed by gas chromatography-mass selective detector. The principal component analysis (PCA) of the volatile compounds grouped long-chain esters on the right side of the first principal component, PC1; these compounds were found in yakju that was made with strains YA-6, Y89-5-3, Y89-5- 2, Y90-9, and Y89-1-1. On the other side of PC1 were short-chain esters; these compounds were found in wines that were brewed with strains Y183-2, Y268-3, Y54-3, Y98-4, and Y88-4. Overall, the results indicated that using different wild-type yeast strains in the fermentation process significantly affects the chemical characteristics of the glutinous rice wine.  相似文献

Ten different versions of the D1/D2 divergent domain of the large-subunit ribosomal DNA were identified among interbreeding members of the yeast species Clavispora lusitaniae. One major polymorphism, located in a 90-bp structural motif of the D2 domain, exists in two versions that differ by 32 base substitutions. Three other polymorphisms consist of a two-base substitution, a two-base deletion, and a single-base deletion, respectively. The polymorphisms are independent of one another and of the two mating types, indicating that the strains studied belong to a single, sexually active Mendelian population. Several strains were heterogeneous for one or more of the polymorphisms, and one strain was found to be automictic and capable of producing asci on its own by isogamous conjugation or by bud-parent autogamy. These observations suggest circumspection in the use of sequence divergence as the principal criterion for delimiting yeast species.  相似文献

The nutritional value of isogenic yeast strains and two microalgal species for gnotobiotically grown Artemia was examined. Yeast cell wall mutants were always better feed for Artemia than their respective wild type. Yeast cells harbouring null mutants for enzymes involved early in the biochemical pathway for cell wall mannoproteins synthesis performed best as feed for Artemia. Yeast cells defective in chitin or β-glucan production were scored in second order. The mnn6 isogenic yeast mutant, harbouring a null mutation for mannoprotein phosphorylation, performed poorly as feed for Artemia, although with good growth. These results suggest that any mutation affecting the yeast cell wall scaffolding by reducing the amount of covalent links between the major components of yeast cell wall, namely mannoproteins, β-glucans and chitin, is sufficient to improve the digestibility for Artemia. The results with microalgae indicated that within one species, strains can have different nutritional value under gnotobiotic conditions. The growth phase was another parameter influencing feed quality, although here it was not possible to reveal the exact cause. It is anticipated that the standard Artemia gnotobiotic growth test is an excellent tool to study the mode of action of bacteria, with a probiotic as well as with a pathogenic character.  相似文献

Summary The operation of packed bed fermenters for ethanol production using the EX-FERM technique with two cycles of 24 h each is described. Twelve Saccharomyces strains were tested with sugarcane particles which had been previously dried and stored as the substrate. All the strains showed acceptable sugar consumption, in some cases above 97%, and ethanol yield coefficients. Sugar consumption values differed, significantly among yeast strains for both cycles. Specific initial rates of ethanol production for the strains ranged from 0.75 to 0.82 g/g· h. Sugar extraction from the particles influenced the first 4 h period of each cycle; final sugar extraction was about 96%. The initial yeast biomass figures were low, within 2–4 g/l, and the final distribution of yeast between solids and circulating liquid varied according to the yeast strain employed. Hydrolysis of sucrose into its components was demonstrated for selected yeast strains during the EX-FERM concurrent extraction and fermentation. The results of the present study support the validity of the operation of packed bed fermenters in cycles for the EX-FERM technique, and suggest employment of smaller cane particles.  相似文献

The morphology of 175 specimens of Ornithogalum belonging to 12 species were analyzed on the basis of 31 characters. The methods used were cluster analysis (agglomerative clustering using Ward's average), principal components analysis (using correlation coefficients) and oligothetic characterization of clusters and species. The splitting level at which seven clusters were separated is the best possible one according to the criteria as used. The separation of nine clusters yields more information on the distribution of the species. Both the clusters and the species are separated from each other in the attribute space performed by the first, second and third principal component. The species of the umbellatum-angustifolium complex, i.e. O. umbellatum, O. angustifoliwn, O. refractum, O. monticolum, O. baeticum and O. algeriense , are clearly separated. A. parallel evolution of the pollen-sterile strains of the bulbilliferous species O. umbellatum and O. angustifolium could be traced. A dichotomous key for all the species involved is proposed.  相似文献

Discriminant coordinates analysis is an adequate technique for analyzing the linear relationships between a number of new variates (i.e. environmental or functional attributes) and a set of vegetational attributes already summarized in the form of a classification. It displays the principal differences among classes in relation to the new variates considered. The procedure and its rationale are equivalent to a special case of principal components analysis.A case study on radiometer satellite data is presented. Two discriminant coordinates displayed the main differences in the seasonal dynamics of the NDVI (an index of standing green biomass) among broad phytogeo-graphic units in the Patagonia region. The first coordinate can be interpreted as an index of height and convexity of the NDVI seasonal curve. It suggests that the principal difference among regions was the total seasonal growth. The adaptive evolution of Saccharomyces cerevisiae to repeated vacuum fermentations was investigated by metabolomic analysis using gas chromatography coupled to time-of-flight mass spectrometry. The first round (VFI, 30 cycles) and second round (VFII, 10 cycles) of repeated fermentations could be clearly distinguished by principal components analysis on intracellular metabolites, indicating that significant difference of metabolic states occurred between them. Further investigation revealed that higher levels of glycerol, trehalose, myo-inositol and glutamate might be involved in response to vacuum stress during initial cycles, while the decreases in their levels indicated that yeast cells adapted to vacuum condition as the fermentation progressed. Furthermore, lower levels of glycerol, myo-inositol, trehalose and glutamate during VFII indicated that the adapted yeast represented better vacuum tolerance. Additionally, glycolysis and TCA cycle intermediates were enhanced whereas glycerol biosynthesis was depressed by vacuum. The decreases of most amino acids might be related to increases in intermediates of glycolysis and TCA cycle as VFI progressed. These findings provided new insights into underlying mechanisms in adaptive evolution of yeast under vacuum condition.  相似文献

Multiple-group principal component analysis and discriminant analysis were used to investigate the morphological differences between adult skulls of male and female minks, badgers and otters from Norway. The first principal component axis, calculated from the variance-covariance matrix of log-transformed data, was interpreted as a growth-free size axis in all three species, while the other components were interpreted as representing shape. Having largely separated size and shape variation, these two aspects of sexual dimorphism could be studied. The standardized component scores were subjected to an analysis of variance and discriminant analyses were performed on size-in and size-out data. Sexual dimorphism was disclosed on eight of the 12 components in minks and on seven of the 12 components in badgers and otters. In mink the multivariate differences were more due to size than to shape, whereas in badgers and otters most of the multivariate differences were due to shape, but the differences in size were also significant. The shape dimorphism was shown to be functionally related to jaw and neck muscles. The results were discussed in relation to recent theories to explain the evolutionary significance of sexual dimorphism in body size of mustelids. It was concluded that these theories do not fully explain the dimorphism found in the skulls of the moderately dimorphic badger and otter.  相似文献

The aim of this research was to identify the Saccharomyces spp. associated with Žilavka grapes and to evaluate their enzymatic activities, H2S production and micro-fermentation performance. For this purpose, a total of 143 yeast strains isolated from three production areas of the Mostar wine region (Bosnia and Herzegovina) were studied and analysed. Firstly, yeasts were identified to genus level by growth on WL nutrient agar and the test of assimilation of lysine. Later, molecular identification at species level was carried out with RFLP analysis of 18S rDNA + ITS region, and at strain level with microsatellite-primed PCR (MSP-PCR). At physiological level yeast strains were grouped into different clusters by means of the Joining-Tree-Clustering-Method. All yeasts tested were identified as S. cerevisiae, resulting a total of 18 different strains. All of the investigated strains produced hydrogen sulphide, 89% were able to complete the fermentation, and none of them was able to synthesize killer toxins. Since enzymes play a very important role in wine aroma development, it was very encouraging that 33% of the strains were able to synthesize pectinolytic enzyme but only one produced β-glucosidase. In the second part of the selection process two indigenous strains were compared with commercial yeast in a microvinification and Žilavka wines with different profiles of volatiles were obtained. This research represents a first step in the selection of indigenous yeast strains from the Mostar region with the goal of maintaining the specific organoleptic characteristics of Žilavka wine.  相似文献

Sixteen strains of the new yeast species Candida sergipensis have been isolated from frozen pulps of the tropical fruits umbú ( Spondias tuberosa Avr. Cam.) and mangaba ( Hancornia speciosa Gom.). Candida sergipensis was one of the prevalent species in the yeast community of these substrates. The new asexual ascomycetous yeast is phylogenetically related to Candida spandovensis and Candida sorbophila, species belonging to the Wickerhamiella clade, as evidenced by the sequences of the D1/D2 domains of their large subunit ribosomal DNAs. The species C. sergipensis and C. spandovensis can be separated on the basis of growth on 50% glucose agar, xylose and succinate, negative for the first species and positive for the second. We tested the attraction of Triatoma dimidiata and T. pallidipennis to traps baited with yeast volatiles. Two traps were simultaneously presented in opposite sides of an experimental arena. One trap presented a yeast culture in sucrose solution, while the other contained sucrose solution as control. A first experimental series was done without offering a central refuge for bugs. In a second series, one shelter where the insects could hide was offered and the traps were presented afterwards. In the first series, yeast baited traps attracted significantly more insects than control ones for both species. In the second series, T. pallidipennis was significantly attracted to yeast, whereas T. dimidiata was not attracted. The potential use of yeast baited traps for capturing these vectors of Chagas disease is discussed.  相似文献

Sexual selection for divergent female preferences has been proposed to stimulate speciation. We tested this basic model by selecting for divergence in the courtship repertoire of the house fly Musca domestica L. Specifically, we subjected replicate strains to artificial selection for differentiation along the first two principal components of the phenotypic intercorrelation structure of five courtship traits. Highly significant differentiation in courtship repertoire resulted, and the magnitude of the selection response was highest along the first principal component (representing the ‘size’, or general intensity, of courtship). Videotaped matings of the crosses between divergent lines (i.e. males of one strain mating with females from a different line) showed that the selection responses in the intensities of male performances were due to shifts in female preferences. In particular, the males were able to accommodate the demands of ‘foreign’ females (as well as their own) in the no-choice situation (i.e. only one male and one female per mating chamber). In contrast to this plasticity of the males, the females were consistent in their differential resistance responses, regardless of the type of male involved in the courtship. Multiple-choice mate choice tests revealed significant reproductive isolation among some lines, although the effect was asymmetrical. The patterns of nonrandom mating were largely due to females from more genetically healthy lines (i.e. with overall high mating propensity) discriminating against males from populations with more inbreeding depression. We suggest that the inability to achieve true (symmetrical) reproductive isolation could have been due to low evolutionary potential in the ‘shape’ of courtship, as defined by the second principal component. The behaviour of cells of Rhodococcus erythropolis DCL14, Xanthobacter Py2, Arthrobacter simplex and Mycobacterium sp. NRRL B-3805, in biphasic systems containing different organic solvents was evaluated and compared. The data, obtained mainly by fluorescence microscopy and image analysis, was interpreted using principal components analysis (PCA). With this technique, the variability of the data could be summarised in 7 components, representing 75.8% of the variance of the data. Over a third of the variance could be explained by the first two principal components which represent solvent toxicity. Apparently this is the major factor influencing cell behaviour in an organic:aqueous system. However, factors such as substrate concentration, cell adaptation ability (resulting in morphological changes and aggregation or separation of cells) and membrane composition (specific to each strain) also play an important role in cell resistance to solvent toxicity. The results regarding cell shape indicate that loss of viability occurs, in the tested bacterial strains, after incorporation of molecules of solvent in the cellular membrane. This should result in an increase in membrane fluidity, and thus, in an alteration of cell shape. The ability to form “self-defence” clusters was observed to be different amongst the four strains. X. Py2 showed, in general, a low tendency to form aggregates under the tested conditions; A. simplex and R. erythropolis aggregated mainly in the presence of low log P solvents; and Mycobacterium. sp. cells showed a high ability to aggregate.  相似文献
