Whereas the transfer of Listeria from surfaces to foods and vice versa has been well documented, little is known about the mechanism of bacterial transfer. The objective of this work is to gain a better understanding of the forces involved in listerial biofilms adhesion using atomic force microscopy (AFM). L. monocytogenes Scott A was grown as biofilms on stainless steel surfaces by inoculating stainless steel coupons with Listeria and incubating the coupons for 48 h at 32 °C with a diluted 1:20 tryptic soy broth. After growth, biofilms were equilibrated over saturated salt solutions at a constant relative humidity (%RH) before measurement of adhesion forces using AFM. The effects of contact time, loading force, and biofilm relative humidity (%RH) suggested that neither contact time, loading force nor biofilm %RH had a significant effect on biofilm adhesiveness at a cellular level (P > 0.05). In a second set of experiments, the influence of material type on biofilm adhesiveness was evaluated using two different colloidal probes (SiO2 and polyethylene). Results showed that the maximum pull-off force and retraction work needed to retract the cantilever for glass (−85.42 nN and 1.610−15 J, respectively) were significantly lower than those of polyethylene (−113.38 nN and 2.7 × 10–15 J, respectively; P < 0.001). The results of this study suggest that Listeria biofilms adhere more strongly to hydrophobic surfaces than hydrophilic surfaces when measured at a cellular level. These results provide important insights that could lead to new ways to remediate and avoid listerial biofilm formation in the food industry. 相似文献

We investigate the mechanical response of PC12 neurites subjected to a drag force imposed by a laminar flow perpendicular to the neurite axis. The curvature of the catenary shape acquired by an initially straight neurite under the action of the drag force provides information on both elongation and tension of the neurite. This method allows us to measure the rest tension and viscoelastic parameters of PC12 neurites and active behavior of neurites. Measurement of oscillations in the strain rate of neurites at constant flow rate provides insight on the response of molecular motors and additional support for the presence of a negative strain-rate sensitivity region in the global mechanical response of PC12 neurites. 相似文献

S ummary : The effect of relative humidity (RH) and inositol on the lethal action of artificial sunlight has been studied. The lethality of these radiations is shown to be strongly dependent on RH provided the intensity of the light is not too great. A critical region between 55 and 65% RH exists in which there is an abrupt change in the sensitivity of airborne cells to all wavelengths tested. Not only does the death rate change suddenly in this RH region but so also does the rate of mutant production. This critical RH region corresponds to the RH level at which bound water is removed from the nucleic acid bases and it is suggested that these water molecules are able to prevent lethal excitations or migration of excitation energy. Inositol is shown to prevent radiation damage and, because of its structure, it is assumed to replace water in the dried cells. Some ramifications of these findings with respect to the maintenance of cell integrity are discussed. 相似文献

Individual blastospores of Candida albicans were deposited on the surface of 50-mm membranes (Millipore Corp.) and placed within sealed glass chambers at various relative humidities (RH). After 48 hr, virtually all cells maintained at 100 and 10% RH had survived, but 84% of the cells maintained at 60% RH failed to develop into colonies when transferred to Sabouraud medium. No morphological abnormalities could be observed in cells surviving low RH values, but their initial rate of multiplication after transfer to Sabouraud medium was greatly reduced, compared to that demonstrated by cells maintained at 100% RH. At 60% RH, the exposure time required to kill 50% of the blastospores was 2 to 3.5 days. The inimical effect of 60% RH was confirmed in a total of 21 isolates of C. albicans. No deleterious effect was noted when 12 other species of yeasts were subjected to 10, 60, and 100% RH. The single isolate of Candida brumptii and 1 out of the 20 isolates of Cryptococcus neoformans tested also failed to grow after blastospores had been exposed to 60% RH for 4 days. 相似文献

The predatory gall midge Feltiella acarisuga (Vallot) (Diptera: Cecidomyiidae) is a biological control agent for twospotted spider mites on greenhouse vegetable crops. Effects of temperature and relative humidity (RH) on development of immatures, reproduction, and prey capture were determined in order to confirm the suitability of F. acarisuga for use in greenhouses. Developmental time ranged from 10 days at 27°C to 34 days at 15°C. At 20°C, developmental time was significantly shorter at 96% RH than at 84% RH. There was very poor survival of immatures at 64% RH and none at 36%. Lifespan of adult females decreased with increasing temperature, but temperature had no significant effect on number of eggs laid. At 20°C, lifespan was longer at 84 and 96% RH than at 64 or 36% RH. The number of spider mites attacked by 3-day-old larvae over 8 h increased with increasing temperature from 15 to 27°C. The number of mites attacked also increased with increasing RH at 27°C. We conclude that F. acarisuga will complete its life cycle and reproduce under conditions typically found in vegetable greenhouses in northern temperate climates. However, extended periods of low RH (<60% RH) could reduce reproduction and survivorship sufficiently to impair the predator's action against spider mite populations. 相似文献

Airborne Pasteurella pestis (A-1122) at low humidities [20 to 50% relative humidity (RH)] exhibited exponential decay when either 1% peptone or Heart Infusion Broth (HIB) was used as the diluent in the viable assay system. At higher RH values (65 and 87%), however, the 1% peptone diluent adversely affected the viability assay. In contrast, HIB as diluent was remarkably effective in demonstrating a higher number of viable cells in aerosols held at high RH values. Similarly, with HIB as diluent, aerosols were shown to contain viable cells during 90 min of observation; with 1% peptone, viability was not detectable after 20 min in the airborne state. 相似文献

The stabilization of slopes by vegetation has been a topical issue for many years. Root mechanical characteristics significantly influence soil reinforcement; therefore it is necessary to research into the indicators of root tensile properties. In this study, we explored the influence of root moisture content on tensile resistance and strength with different root diameters and for different tree species. Betula platyphylla, Quercus mongolica, Pinus tabulaeformis, and Larix gmelinii, the most popular tree species used for slope stabilization in the rocky mountainous areas of northern China, were used in this study. A tensile test was conducted after root samples were grouped by diameter and moisture content. The results showedthat:1) root moisture content had a significant influence on tensile properties; 2) slightly loss of root moisture content could enhance tensile strength, but too much loss of water resulted in weaker capacity for root elongation, and consequently reduced tensile strength; 3) root diameter had a strong positive correlation with tensile resistance; and4) the roots of Betula platyphylla had the best tensile properties when both diameter and moisture content being controlled. These findings improve our understanding of root tensile properties with root size and moisture, and could be useful for slope stabilization using vegetation. 相似文献

Three strains of the Columbia SK (Col-SK) group of viruses [Mengo, Maus Elberfeld (ME), and Col-SK viruses] have been studied in the airborne state. All three strains were found to give identical aerosol decay patterns at 16 or 26 C, when held at the same relative humidity (RH). During the first 5 min of aerosol storage time at 16 C, virus inactivation was RH-dependent, with survival maximal at either high (greater than 80%) or low (less than 5%) RH. After 5 min at 16 C, further inactivation, regardless of RH, was insignificant. At 26 C, the effect on survival of RH between 40 and 60% was even more pronounced than at 16 C, and continued after 5 min through 6 hr. Results of this study indicated that the inactivation of airborne Col-SK group viruses was similar to that of other ribonucleic acid (RNA) viruses, particularly poliovirus. Since members of the Col-SK group are picornaviruses, they may well serve as an aerosol model representative of small, ether-resistant, single-stranded RNA viruses. 相似文献

不同含水量(MC7.1%~1.2%)的洋葱种子贮藏在35℃、室温、15℃和5℃条件下1~3年,适度超干处理能延长种子的贮藏寿命;种子的贮藏寿命与种子含水量和贮藏温度密切相关。种子贮藏的最适含水量随温度的改变而发生相应的变化,35℃时MC为3.4%;室温时为3.4%~4.5%;15℃时为4.5%~5.1%。MC≤2.2%不利于延长种子寿命。在室温自然条件下贮藏1~3年,适度超干种子(MC3.4%)内MDA和H2O2含量、O-*2产生速率和LOX活性明显地低于未超干种子(MC7.1%)和高度超干种子(MC1.2%),而抗氧化酶AsA-POD、CAT和SOD的活性显著地高于未超干种子(MC7.1%)和高度超干种子(MC1.2%)。据此认为对脂质过氧化的抑制作用是适度超干种子耐贮藏的生理原因之一。 相似文献

Effect of Temperature and Relative Humidity on the Survival of Foodborne Viruses during Food Storage

Millions of people suffer from foodborne diseases throughout the world every year, and the importance of food safety has grown worldwide in recent years. The aim of this study was to investigate the survival of hepatitis A virus (HAV) and viral surrogates of human norovirus (HuNoV) (bacteriophage MS2 and murine norovirus [MNV]) in food over time. HAV, MNV, and MS2 were inoculated onto either the digestive gland of oysters or the surface of fresh peppers, and their survival on these food matrices was measured under various temperature (4°C, 15°C, 25°C, and 40°C) and relative humidity (RH) (50% and 70%) conditions. Inoculated viruses were recovered from food samples and quantified by a plaque assay at predetermined time points over 2 weeks (0, 1, 3, 7, 10, and 14 days). Virus survival was influenced primarily by temperature. On peppers at 40°C and at 50% RH, >4- and 6-log reductions of MNV and HAV, respectively, occurred within 1 day. All three viruses survived better on oysters. In addition, HAV survived better at 70% RH than at 50% RH. The survival data for HAV, MS2, and MNV were fit to three different mathematical models (linear, Weibull, and biphasic models). Among them, the biphasic model was optimum in terms of goodness of fit. The results of this study suggest that major foodborne viruses such as HAV and HuNoV can survive over prolonged periods of time with a limited reduction in numbers. Human leukocyte migration inhibitory factor (LIF) appears to be a serine esterase and protease exhibiting specific affinity towards arginine esters and amides. On the basis of indirect evidence that an amide of the oligopeptide benzoyl-phenylalanyl-valyl-arginine might have high and selective affinity for LIF, we prepared an ester of this oligopeptide, benzoyl-phenylalanyl-valyl-arginine (3H) methyl ester (3H-BPVAME) for the direct measurement of LIF activity in a double-phase radio-enzyme assay. The hydrolysis of 3H-BPVAME followed enzyme-substrate kinetics in that the reaction was time-, temperature-, pH-, and concentration-dependent. 3H-BPVAME rpoved to be more selective and approximately 20 times more sensitive as a substrate for LIF than previously used radiolabeled substrates such as 3H-BAEE and 3H-TAME. Moreover, hydrolysis of 3H-BPVAME by partially purified LIF preparations was significantly inhibited by 10(-8) to 10 (-5) M of guanosine 3',5'-cyclic monophosphate (cGMP), further supporting the hypothesis that cGMP acts as a regulator of LIF activity. Inhibition of LIF-induced esterolysis was also provided by dibutyryl cGMP, but only at concentrations 10(-7) to 10(-5) M; 8-bromo cGMP and adenosine 3',5'-cyclic monophosphate were both ineffective. These results provide support for the use of 3H-BPVAME as a more selective substrate to detect esterase activity in LIF preparations than heretofore described and the possible development of a biochemical assay for the measurement of this lymphokine. 相似文献

Seeds of Hancornia speciosa germinated best at a temperatureof 2030 °C. The viability of the seeds during storagewas short and the best storage conditions for viability entailedkeeping the seeds in polyethylene bags. Seed viability was maintainedonly when the seeds were stored at a moisture content above30%; storage conditions which allowed dehydration resulted ina rapid loss of viability (the seeds showed recalcitrant behaviour). Low temperature during storage did not improve longevity. Most methods of dietary reconstruction are limited in their applicability to either extant or extinct taxa. We apply and discuss a method in which dietary information can be reconstructed from chips in the tooth enamel of both living and fossil primates. Such chips can be used to indicate the presence of large hard foods in the diet, and also to provide an estimate of the bite force that was used when the chip was created. Furthermore, the equations derived from this method allow an estimate of maximum bite force to be obtained from a simple measurement of tooth size. We use this method to investigate dietary differences in nonhuman great apes (Pongo, Gorilla, Pan). The high frequency of chips on teeth of Pongo indicate that they frequently use high forces to process hard foods such as seeds and nuts. Gorilla can generate even higher bite forces, but their low incidence of tooth chips suggests that they do so when consuming soft but tough foods. Tooth chips provide a lasting dietary signal that is not easily masked or erased, making them particularly useful for the study of rarely eaten items such as some fallback foods. 相似文献

Very little is known regarding the ecology of Nitrosospira sp. strain AF-like bacteria, a unique group of ammonia oxidizers within the Betaproteobacteria. We studied the response of Nitrosospira sp. strain AF-like ammonia oxidizers to changing environmental conditions by applying molecular methods and physiological measurements to Californian grassland soil manipulated in the laboratory. This soil is naturally high in Nitrosospira sp. strain AF-like bacteria relative to the much-better-studied Nitrosospira multiformis-like ammonia-oxidizing bacteria. Increases in temperature, soil moisture, and fertilizer interacted to reduce the relative abundance of Nitrosospira sp. strain AF-like bacteria, although they remained numerically dominant. The overall abundance of ammonia-oxidizing bacteria increased with increasing soil moisture and decreased with increasing temperature. Potential nitrification activity was altered by interactions among temperature, soil moisture, and fertilizer, with activity tending to be higher when soil moisture and temperature were increased. The increase in potential nitrification activity with increased temperature was surprising, given that the overall abundance of ammonia-oxidizing bacteria decreased significantly under these conditions. This observation suggests that (i) Nitrosospira sp. strain AF-like bacteria may respond to increased temperature with an increase in activity, despite a decrease in abundance, or (ii) that potential nitrification activity in these soils may be due to organisms other than bacteria (e.g., archaeal ammonia oxidizers), at least under conditions of increased temperature. 相似文献

Four inoculation techniques were compared for initiation of growth on cake surfaces: spot, air cabinet, spray (atomizer), and talc addition methods. Molds were isolated from commercial cakes and were identified as Aspergillus sydowii, Aspergillus ochraceus, Penicillium funiculosum, and Eurotium herbariorum. Cake surfaces were inoculated with mold spores and incubated under three equilibrium relative humidity (ERH) levels: 97, 85, and 75%. Random contamination by spores in a ventilated air cabinet was the simplest method of inoculation, but standard deviations in the inoculation rates (20% on a relative scale) were almost twice those observed with the other methods. The spot method was the most reproducible. Cake samples inoculated in the air cabinet had colony counts 10 times lower than those obtained for potato dextrose agar plates at 97% ERH, which was not the case with the spray and talc methods. Growth of molds was much slower in the samples incubated in 75% relative humidity, with all methods. Colony counts were generally similar in systems adjusted at 85 to 97% ERH but were lower for samples incubated at 75% ERH. In comparisons of the shelf life estimates obtained by the various inoculation methods, a correlation coefficient (r2) of 0.70 was obtained between the spot method and the other methods of inoculation, while talc, air cabinet, and spray shelf life data were correlated better (r2 ≈ 0.97). The spot method appeared to be the method of choice in consideration of ease of use, precision, and the ability to enable the study of the effects of the environment on mold-free shelf life as well as on the rate of growth of molds on cakes. 相似文献
