Analysis of the greenbug (Schizaphis graminum Rond.) — sorghum interaction system confirmed the hypothesis that rare insect virulence was related to reduced fitness. Greenbug clones from the Krasnodar population virulent to resistance genes Sgr5 and Sgr6 revealed lower fecundity in comparison with avirulent ones and were replaced in model populations during reproduction on a susceptible sorghum line. The main role of aphid fecundity was shown to provide higher fitness, reducing the frequency of virulent clones in natural populations.  相似文献

Abstract

Soybean aphid (Aphis glycines Matsumura) is the most important soybean [Glycine max (L.) Merr.] insect pest in the USA. The objectives of this study were to characterize the resistance expressed in five plant introductions (PIs) to four soybean aphid biotypes, determine the mode of resistance inheritance, and identify markers associated with genes controlling resistance in these accessions. Five soybean PIs, from an initial set of 3000 PIs, were tested for resistance against soybean aphid biotypes 1, 2, 3, and 4 in choice and no-choice tests. Of these five PIs, PI 587663, PI 587677, and PI 587685 expressed antibiosis against all four biotypes, while PI 587972 and PI 594592 expressed antibiosis against biotypes 1, 2, and 3. F2 populations derived from PI 587663 and PI 587972 were evaluated for resistance against soybean aphid biotype 1, and populations derived from PIs 587677, 587685, and 594592 were tested against biotype 3. In addition, F2:3 plants were tested against biotypes 2 and 3. Genomic DNA from F2 plants was screened with markers linked to Rag1, Rag2, Rag3, and rag4 soybean aphid-resistance genes. Results showed that PI 587663 and PI 594592 each had three genes with variable gene action located in the Rag1, Rag2, and Rag3 regions. PI 587677 had three genes with variable gene action located in the Rag1, Rag2 and rag4 regions. PI 587685 had one dominant gene located in the Rag1 region and an additive gene in the Rag2 region. PI 587972 had one dominant gene located in the Rag2 region controlling antixenosis- or antibiosis-type resistance to soybean aphid biotypes 1, 2, or 3. PIs 587663, 587677, and 587685 also showed antibiosis-type resistance against biotype 4. Information on multi-biotype aphid resistance and resistance gene markers will be useful for improving soybean aphid resistance in commercial soybean cultivars.

Amount of certain amino acids required by herbivores, and their availability in host plants are of crucial importance for insect growth, development, and life processes. Therefore, we carried out profiling and quantification of 17 amino acids in diverse sorghum genotypes, and on the Chilo partellus larvae reared on them, to understand the association and contribution of different amino acids in plant resistance to insects. Sorghum germplasm lines IS 2205 and IS 2123 had severe detrimental effects on the development and survival of C. partellus followed by varieties ICSV 700 and ICSV 708 in comparison to susceptible check, Swarna. Profiling of sorghum seedlings, seeds, and the C. partellus larvae fed on these genotypes for 17 amino acids revealed that Arginine, Glycine, Isoleucine, Leucine, Proline, and Valine in sorghum seedlings and the C. partellus larvae had significant and positive association, suggesting their role in the development and survival, while negative association of Cystine indicated its contribution in plant defense. Furthermore, C. partellus acquired less of cyclic and aliphatic amino acids per unit amount from the test resistant genotypes, while more from the susceptible check, than their presence in the seedlings. Present studies suggest that Alanine, Cystine, Glycine, and Proline contents in C. partellus larvae; Cystine and Proline contents in sorghum seedlings; and Methionine content in sorghum seeds, have significant and negative association, and contribute to explain?>93% and >96%, respectively, of the variability in antibiosis mechanism and overall resistance to C. partellus. These studies have implications for antibiosis and nutritional mechanism of host plant–insect interactions in sorghum against C. partellus.  相似文献

The soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), is a limiting factor in soybean production in the North Central region of the USA. The objectives of this work were to identify sources of resistance to A. glycines in 14 soybean genotypes, and also document changes in total protein, peroxidase, and chlorophyll in response to aphid feeding. A reduced number of A. glycines was observed on the genotypes UX 2569-159-2-01 and UX 2570-171- 04, indicating the presence of antixenosis and/or antibiosis. UX 2569-159-2-01 expressed the highest level of resistance; whereas, UX 2570-171-04 had moderate levels of resistance to A. glycines. Chlorophyll content was relatively unaffected by A. glycines, except for a reduction in UX 2569-159-2-01 infested plants at 5 and 15 days after infestation (DAI). No changes were detected in total protein content between infested and control plants for the genotypes analyzed; however, peroxidase activity was higher in infested UX 2570-171-04 at both 5 and 10 DAI. This improvement in peroxidase content in infested UX 2570-171-04 may be playing multiple roles in the plant tolerance.  相似文献

Abstract

Greenbug (Schizaphis graminum Rondani) and Hessian fly [Mayetiola destructor (Say)] are two major destructive insect pests of wheat (Triticum aestivum L.) throughout wheat production regions in the USA and worldwide. Greenbug and Hessian fly infestation can significantly reduce grain yield and quality. Breeding for resistance to these two pests using marker-assisted selection (MAS) is the most economical strategy to minimize losses. In this study, doubled haploid lines from the Synthetic W7984 × Opata M85 wheat reference population were used to construct linkage maps for the greenbug resistance gene Gb7 and the Hessian fly resistance gene H32 with genotyping-by-sequencing (GBS) and 90K array-based single nucleotide polymorphism (SNP) marker data. Flanking markers were closely linked to Gb7 and H32 and were located on chromosome 7DL and 3DL, respectively. Gb7-linked markers (synopGBS773 and synopGBS1141) and H32-linked markers (synopGBS901 and IWB65911) were converted into Kompetitive Allele Specific PCR (KASP) assays for MAS in wheat breeding. In addition, comparative mapping identified syntenic regions in Brachypodium distachyon, rice (Oryza sativa), and sorghum (Sorghum bicolor) for Gb7 and H32 that can be used for fine mapping and map-based cloning of the genes. The KASP markers developed in this study are the first set of SNPs tightly linked to Gb7 and H32 and will be very useful for MAS in wheat breeding programs and future genetic studies of greenbug and Hessian fly resistance.

Abstract

Soybean aphid, Aphis glycines Matsumura, is a highly destructive pest for soybean production. E08934, a soybean advanced breeding line derived from the wild soybean Glycine soja 85-32, has shown strong resistance to aphids. To dissect the genetic basis of aphid resistance in E08934, a mapping population (070020) consisting of 140 F3-derived lines was developed by crossing E08934 with an aphid-susceptible line E00003. This mapping population was evaluated for aphid resistance in a greenhouse trial in 2010 and three field trials in 2009, 2010, and 2011, respectively. The broad-sense heritability across the field trials was 0.84. In the mapping population 070020, two major quantitative trait loci (QTL) were detected as significantly associated with aphid resistance, and designated as Rag6 and Rag3c, respectively. Rag6 was mapped to a 10.5 centiMorgan (cM) interval between markers MSUSNP08-2 and Satt209 on chromosome 8, explaining 19.5–46.4% of the phenotypic variance in different trials. Rag3c was located at a 7.5 cM interval between markers MSUSNP16-10 and Sat_370 on chromosome 16, explaining 12.5–22.9% of the phenotypic variance in different trials. Rag3c had less resistance effect than Rag6 across all the trials. Furthermore, Rag6 and Rag3c were confirmed in two validation populations with different genetic backgrounds. No significant interaction was detected between Rag6 and Rag3c in either the mapping population or the validation populations. Both Rag6 and Rag3c were indicated as conferring antibiosis resistance to aphids by a no-choice test. The new aphid-resistance gene(s) derived from the wild germplasm G. soja 85-32 are valuable in improving soybeans for aphid resistance.

Combined inheritance of oligogenes (Sgr1, Sgr4, Sgr5, and Sgr6) and a weakly expressed resistance to infestation with virulent greenbug clones was studied in sorghum. Under these conditions, the resistance was shown to depend on the interaction between minor resistance genes of the host plant and the virulence genes of the pest rather than on the residual effect of oligogenes. The minor genes can be independent of or weakly linked to the major resistance gene. They differentially interact with phytophage genotypes and, contrary to Van der Plank's postulates, are not responsible for the long-term (horizontal) resistance. The possibility of rapidly overcoming the effect of minor genes was confirmed by observation of seasonal dynamics of a natural aphid population on a resistant variety.  相似文献

Abstract

Host plant selection for oviposition is an important determinant of progeny performance and survival for phytophagous insects. Specific cues from the plant influence insect oviposition behavior; but, to date, no set of host plant quantitative trait loci (QTLs) have been shown to have an effect on behavioral sequences leading to oviposition. Three QTLs in wheat (Triticum aestivum L.) have been identified as influencing resistance to the wheat stem sawfly (WSS) (Cephus cinctus Norton). Wheat near-isogenic lines (NILs) for each of the three QTLs were used to test whether foraging WSS were able to discriminate variation in plant cues resulting from allelic changes. A QTL on chromosome 3B (Qss-msub-3BL) previously associated with stem solidness and larval antibiosis was shown to affect WSS oviposition behavior, host preference, and field infestation. Decreased preference for oviposition was also related to a QTL allele on chromosome 2D (Qwss.msub-2D). A QTL on chromosome 4A (Qwss.msub-4A.1) affected host plant attractiveness to foraging females, but did not change oviposition preference after females landed on the stem. These findings show that oviposition decisions regarding potential plant hosts require WSS females to discriminate signals from the plant associated with allelic variation at host plant quantitative loci. Allele types in a host plant QTL associated with differential survival of immature progeny can affect maternal choices for oviposition. The multidisciplinary approach used here may lead to the identification of plant genes with important community consequences, and may complement the use of antibiosis due to solid stems to control the wheat stem sawfly in agroecosystems.

The soybean looper Chrysodeixis includens (Walker) (Lepidoptera: Noctuidae) is known as an important pest of leguminous plants worldwide. In Brazil, this pest species is gaining importance to producers of the common bean Phaseolus vulgaris L. (Fabaceae) because it limits field production of the crop. Chemical control is still the primary method of insect control. However, due to the possible harmful effects of pesticides to humans and the environment, alternative and less aggressive practices are being investigated. For this reason, the use of resistant plant genotypes represents a valuable tool in insect control. This study evaluated the biological aspects of larvae of C. includens confined to 14 bean genotypes under laboratory conditions (26 ± 2°C; 65 ± 10% RH; photoperiod of 14 h L:10 h D). The duration of the instars, total duration of the larval phase, consumption while in the larval phase, weight of the fifth instar larvae, larval viability, duration of the pre-pupal and pupal phases, pupal weight, pupal viability, pupal deformity, caterpillar-to-adult cycle, duration of the pre-oviposition and oviposition periods, and total number of viable eggs per female were evaluated. The genotypes “IAC Boreal,” “IAC Harmonia,” and “IAC Formoso” expressed antibiosis, prolonging the caterpillar-to-adult cycle and reducing the larval viability; however, each of these genotypes also experienced high leaf consumption. “IAC Jabola” expressed moderate levels of antibiosis and/or antixenosis (feeding), while the genotype “BRS Horizonte” expressed antixenosis (feeding). The data obtained with IAC Boreal, IAC Harmonia, IAC Formoso, IAC Jabola, and BRS Horizonte are promising and may help with the improvement of programs aimed at managing C. includens damage to this leguminous agricultural crop.  相似文献

Bemisia tabaci (Genn.) (Hemiptera: Aleyrodidae) has been recognized as an important pest of many agricultural systems including soybean [Glycine max (L.) Merrill] crops. As an alternative to chemical control, the use of resistant genotypes represents an important tool for integrated pest management (IPM). This study aimed to evaluate the biological development of Bemisia tabaci biotype B confined on 13 soybean genotypes under greenhouse conditions. Initially, the nymphal period, complete development period (egg–adult), and the viability of the silverleaf whitefly nymphs were evaluated in all genotypes. Then, four genotypes promising for resistance (‘Jackson,’ UX-2569-159, ‘P98Y11,’ and ‘TMG132 RR’) and a susceptible genotype (PI-227687) were selected for further assays, where two insect populations were compared: a first population from the initial rearing (cabbage plants) and another corresponding to insects previously reared out on the selected genotypes. In addition to the parameters evaluated in preliminary tests, we also determined the viability and incubation period of eggs. Moderate levels of resistance (antibiosis/antixenosis) to B. tabaci biotype B were found in three genotypes. ‘P98Y11’ and ‘TMG132 RR’ were less suitable for insect development, extending the development cycle, and UX-2569-159 caused high nymphal mortality. We did not observe a significant increase in the level of plant resistance by the use of previously stressed insects. This suggests that the evaluation of a single whitefly generation may be sufficient to make correct decisions on promising soybean genotypes.  相似文献

The two-spotted spider mite, Tetranychus uticae Koch (Acari: Tetranychidae), is globally one of the most devastating pests that feed on numerous crops, including common bean (Phaseolus vulgaris L.). This study was aimed to evaluate the effects of genotype and morphological attributes of common bean on T. uticae. Forty common bean accessions were used to investigate antixenosis and antibiosis through assessing mite feeding preference and reproduction under laboratory conditions. Three resistant (i.e., 56, 63, 238) and two susceptible (i.e., 182, 236) accessions, along with cultivars Naz (resistant) and Akhtar (susceptible), were used in a life-table study. Both antixenosis and antibiosis mechanism were observed in all of the accessions, albeit a negative correlation occurred. Significant differences were observed for all traits of T. urticae: developmental time of immature stages, reproduction, adult longevity and life-table parameters. Based on the intrinsic rate of increase, the accessions 56, 63, 182, 238, and cv. Naz impose high antibiotic effects on T. urticae. Although significant variation existed among accessions for morphological factors, only glandular trichomes correlated with mite fecundity and feeding preference.  相似文献

Viruses vectored by the aphid Amphorophora agathonica cause decline in black raspberry plant health resulting in a shortened life and poor fruit quality of the infected plantings. New aphid resistant cultivars could increase the longevity of plantings providing growers and processors with consistent fruit production. Recent exploration of the native range of black raspberry identified three sources of aphid resistance: Ag4 from Ontario (ON), Canada, Ag5 from Maine (ME), and a third source from Michigan (MI) with no formal designation. The objectives of this study were to assess segregation of these three sources of aphid resistance in populations with single and combined sources and develop markers that can identify each source of resistance. A genetic linkage map constructed for ORUS 4305 placed the ON aphid resistance locus on Rubus linkage group (RLG) 6. Segregation ratios in populations with single and combined sources, and linkage mapping in two populations (ORUS 4304 and ORUS 4812) segregating for the Ag5 and MI sources, respectively, indicated that these three sources of resistance are each conferred by single dominant genes/alleles that are linked on RLG6. Confirmation of marker association in 16 validation populations identified four markers that could be used to predict resistance; however, none could distinguish between the ON and MI sources. These four markers may be useful for screening populations to enrich the field-planted progeny for aphid resistance. Fine mapping of the resistance loci is needed to develop functional markers at each of the resistance loci to enable pyramiding and durable aphid resistance.  相似文献

Plants have co-evolved with a diverse array of pathogens and insect herbivores and so have evolved an extensive repertoire of constitutive and induced defence mechanisms activated through complex signalling pathways. OXI1 kinase is required for activation of mitogen-activated protein kinases (MAPKs) and is an essential part of the signal transduction pathway linking oxidative burst signals to diverse downstream responses. Furthermore, changes in the levels of OXI1 appear to be crucial for appropriate signalling. Callose deposition also plays a key role in defence. Here we demonstrate, for the first time, that OXI1 plays an important role in defence against aphids. The Arabidopsis mutant, oxi1-2, showed significant resistance both in terms of population build-up (p?<?0.001) and the rate of build-up (p?<?0.001). Arabidopsis mutants for β-1,3-glucanase, gns2 and gns3, showed partial aphid resistance, significantly delaying developmental rate, taking two-fold longer to reach adulthood. Whilst β-1,3-glucanase genes GNS1, GNS2, GNS3 and GNS5 were not induced in oxi1-2 in response to aphid feeding, GNS2 was expressed to high levels in the corresponding WT (Col-0) in response to aphid feeding. Callose synthase GSL5 was up-regulated in oxi1-2 in response to aphids. The results suggest that resistance in oxi1-2 mutants is through induction of callose deposition via MAPKs resulting in ROS induction as an early response. Furthermore, the results suggest that the β-1,3-glucanase genes, especially GNS2, play an important role in host plant susceptibility to aphids. Better understanding of signalling cascades underpinning tolerance to biotic stress will help inform future breeding programmes for enhancing crop resilience.  相似文献

The soybean aphid (Aphis glycines Matsumura), an invasive species, has posed a significant threat to soybean [Glycine max (L.) Merr.] production in North America since 2001. Use of resistant cultivars is an effective tactic to protect soybean yield. However, the variability and dynamics of aphid populations could limit the effectiveness of host-resistance gene(s). Gene pyramiding is a promising way to sustain host-plant resistance. The objectives of this study were to determine the prevalent aphid biotypes in Michigan and to assess the effectiveness of different combinations of aphid-resistance genes. A total of 11 soybean genotypes with known resistance gene(s) were used as indicator lines. Based on their responses, Biotype 3 was a major component of Michigan aphid populations during 2015–2016. The different performance of Rag-“Jackson” and Rag1-“Dowling” along with the breakdown of resistance in plant introductions (PIs) 567301B and 567324 may be explained by Biotype 3 or an unknown virulent biotype establishing in Michigan. With the assistance of flanking markers, 12 advanced breeding lines carrying different aphid-resistance gene(s) were developed and evaluated for effectiveness in five trials across 2015 to 2017. Lines with rag1c, Rag3d, Rag6, Rag3c?+?Rag6, rag1b?+?rag3, rag1c?+?rag4, rag1c?+?rag3?+?rag4, rag1c?+?Rag2?+?rag3?+?rag4, and rag1b?+?rag1c?+?rag3?+?rag4 demonstrated strong and consistent resistance. Due to the variability of virulent aphid populations, different combinations of Rag genes may perform differently across geographies. However, advanced breeding lines pyramided with three or four Rag genes likely will provide broader and more durable resistance to diverse and dynamic aphid populations.  相似文献

The whitebacked planthopper (WBPH), Sogatella furcifera Horvath, is one of the most destructive pests in rice (Oryza sativa L.) production. Host-plant resistance has been considered as an efficient and eco-friendly strategy to reduce yield losses caused by WBPH. In this study, we found that an indica rice cultivar IR54751-2-44-15-24-2 (IR54751) displayed high resistance to WBPH at both seedling and tillering stages. The resistance of IR54751 was mainly contributed by antixenosis and tolerance rather than antibiosis. An F2 population derived from a cross between IR54751 and a susceptible japonica cultivar 02428 was constructed to detect the quantitative trait loci (QTLs) conferring the resistance to WBPH. In total, four QTLs including qWBPH3.1, qWBPH3.2, qWBPH11, and qWBPH12 were identified and distributed on three different chromosomes. The four QTLs had LOD scores of 3.8, 8.2, 5.8, and 3.9, accounting for 8.2, 21.5, 13.9, and 10.4% of the phenotypic variation, respectively. Except for qWBPH3.1, the resistance alleles of the other three QTLs were all from IR54751. Further, a secondary population harboring only single qWBPH11 locus was developed from the F2 population by marker-assisted selection. Finally, qWBPH11 was delimited in a 450-kb region between markers DJ53973 and SNP56. The identification of WBPH resistance QTLs and the fine mapping of qWBPH11 will be helpful for cloning resistance genes and breeding resistant rice cultivars.  相似文献

Bemisia tabaci (Genn.) biotype B (Hemiptera: Aleyrodidae) is an important pest of vegetable crops, including collard greens Brassica oleracea var. acephala (Brassicaceae). The use of resistant genotypes is an interesting option to reduce insect populations and can be used as an important tool for integrated pest management (IPM). This study evaluated 32 genotypes of collard greens against the attack of silver leaf whitefly, with the aim to characterize antixenosis. Initially, a multiple-choice trial was conducted using all genotypes, in which the adult attractiveness was assessed on two leaves per genotype at 24 and 48 h after infestation. After 48 h, one leaf of each genotype was randomly selected for the determination of the number of eggs per square centimeter. From the results of the multiple-choice trial, 13 genotypes were selected for a no-choice oviposition test, following the same method of the previous test. Colorimetric analyses were also performed to establish possible correlations between leaf color and insect colonization. Genotypes HS-20, OE, and VA were less attractive, demonstrating antixenosis. Genotypes LG, VE, J, MG, MOP, HS-20, VA, and MT had less oviposition in the multiple-choice test, which indicated expression of antixenosis. In the no-choice test, genotypes VE, P1C, CCB, RI-919, H, and J had less oviposition, which also characterized antixenosis. Therefore, genotypes VE and J showed the highest resistance stability because both had less oviposition in both test modalities. Thus, the resistance to B. tabaci biotype B indicates the genotypes HS-20, OE, VA, VE, and J are promising for use in breeding programs to develop resistance to whitefly.  相似文献

This study examined the induction of the defence-related hormones jasmonic acid (JA), salicylic acid (SA) and abscisic acid (ABA) and the phytoalexin medicarpin in Medicago truncatula when challenged by the pea aphid Acyrthosiphon pisum. There was some induction of hormones in the compatible interaction between A. pisum clone N116 and M. truncatula cultivar DZA315, whereas JA, SA and medicarpin exhibited more significant increases in foliage concentration during the incompatible interaction between A. pisum clone PS01 and M. truncatula cultivar Jemalong A17. Foliar concentration of JA, SA and medicarpin exhibited a positive relationship with aphid density after 3-day feeding, whereas ABA was not affected by the presence of aphids. When aphids were restricted to a single leaf using plastic tubes, JA, SA and medicarpin displayed strong local induction, whereas there were no significant systemic increases in uninfested leaves. Medicarpin and SA appeared to increase with duration of aphid feeding, whereas JA showed a more transient increase in concentration 24 h after challenge commenced. Results suggest that increases in JA, SA and medicarpin are associated with M. truncatula resistance to particular clones of A. pisum. The variation in concentration of the defence-related compounds recorded with regard to aphid density, duration of challenge, genotypes of plant and aphids, and between locally challenged and distant leaves reinforces the need for consideration of these experimental factors when generalizing about the plant defence processes that occur during aphid–plant interactions.  相似文献

Climate change leads to phenology shifts of many species. However, not all species shift in parallel, which can desynchronize interspecific interactions. Within trophic cascades, herbivores can be top–down controlled by predators or bottom–up controlled by host plant quality and host symbionts, such as plant-associated micro-organisms. Synchronization of trophic levels is required to prevent insect herbivore (pest) outbreaks. In a common garden experiment, we simulated an earlier arrival time (~2 weeks) of the aphid Rhopalosiphum padi on its host grass Lolium perenne by enhancing the aphid abundance during the colonization period. L. perenne was either uninfected or infected with the endophytic fungus Epichloë festucae var. lolii. The plant symbiotic fungus produces insect deterring alkaloids within the host grass. Throughout the season, we tested the effects of enhanced aphid abundance in spring on aphid predators (top–down) and grass–endophyte (bottom–up) responses. Higher aphid population sizes earlier in the season lead to overall higher aphid abundances, as predator occurrence was independent of aphid abundances on the pots. Nonetheless, after predator occurrence, aphids were controlled within 2 weeks on all pots. Possible bottom–up control of aphids by increased endophyte concentrations occurred time delayed after high herbivore abundances. Endophyte-derived alkaloid concentrations were not significantly affected by enhanced aphid abundance but increased throughout the season. We conclude that phenology shifts in an herbivorous species can desynchronize predator–prey and plant–microorganism interactions and might enhance the probability of pest outbreaks with climate change.  相似文献
