Five different types of shoot tip explant from three-year-oldand 13-year-old Sitka spruce trees were established on a rangeof media and shoot growth was recorded. While Schenk and Hildebrandtmedium gave the greatest shoot elongation, Webb and Street mediumproduced the healthiest-looking shoots. Poorest growth was recordedfor the explants cultured without the crown or any vasculartissues from the previous year's growth. [14C]sucrose tracerstudies indicated that poor uptake was not responsible for thereduced growth and it was suggested that damage due to inadequatewater uptake may be responsible. Results demonstrated that peelingthe protective scales from resting buds and trimming the shoottissues below the crown to a wedge shape and excluding the barkwas a reliable method of explant preparation for establishingmature and juvenile Sitka spruce in culture. Summary Chloroplast DNA (cpDNA) restriction analysis was used to classify five reforestation seedlots as to species. The material included two Sitka spruce (Picea sitchensis (Bong.) Carr.), one white spruce (P. glauca (Moench) Voss) from interior British Columbia, and two putative hybrid seedlots from the coast-interior introgression zone in British Columbia. The cpDNA patterns generated by Bam-HI and Bc1-I from individual trees of Sitka spruce, white spruce, western white spruce (P. glauca var. albertiana (S. Brown)), and Engelmann spruce (P. engelmanni (Parry)) were species-specific. They were used as reference patterns for comparisons. In addition, two controlled crosses between white and Sitka spruce were analyzed to demonstrate the paternal inheritance of cpDNA in spruces. The cpDNA restriction patterns for the five seedlots were obtained from composite samples of seedlings from each lot and compared to the typical cpDNA patterns of each species. Restriction patterns for the two Sitka spruce seedlots agreed with those from the Sitka spruce tree, while patterns for the white spruce seedlots from British Columbia agreed with those from the white spruce tree, lacking evidence of any Engelmann spruce component in the sample. On the other hand, one putative hybrid seedlot showed cpDNA patterns similar to white spruce while the other showed fragments unique to both Sitka and white spruce, indicating that this was a hybrid seedlot. The analysis of cpDNA restriction polymorphism has proven to be an effective tool for classifying seedlots in regions of introgression. To our knowledge, these results provide the first demonstration of the use of cpDNA analysis for solving practical forestry problems.  相似文献

Two species of boreal tree seedlings, paper birch (Betula papyrifera Marsh.) and black spruce (Picea mariana (Mill.) B.S.P.), and the ericaceous shrub Kalmia angustifolia L. were grown in pots with humus from a birch-dominated site and two spruce-Kalmia sites. Root systems interacted with humus form in controlling soil-N cycling as well as energy and nutritional deficiencies of soil microorganisms. In general, Kalmia seedlings affected microbial dynamics and N cycling differently than birch and spruce seedlings did. Birch and spruce seedlings reduced gross N mineralization and immobilization rates, soil mineral-N pools and the amounts of NH –N accreted on buried cation exchange resins in all three soils. Compared to birch and spruce seedlings, the growth of Kalmia resulted in significantly higher gross N mineralization rates, soil mineral-N pools and resin-NH accretion in soil from the fertile birch site. Gross N immobilization rates in all soils were generally higher with Kalmia than with spruce or birch seedlings. All three species of seedlings acquired N from the birch site soil, whereas only Kalmia seedlings acquired N from the two spruce-Kalmia site soils. Relative to control treatments, the amount of N mineralized anaerobically increased in the birch-site soil and decreased in the poor spruce-Kalmia site soil with all three species of seedlings. All seedlings increased the microbial biomass in the birch-site soil. Kalmia humus and Kalmia root systems increased microbial energy-deficiency and decreased microbial nutritional deficiency compared to the other humus and seedlings used. Results are discussed in terms of each species' nutrient acquisition mechanism and its competitive ability during secondary succession.  相似文献

Ectomycorrhizal fungi (Paxillus involutus, Suillus grevillei and two unidentified basidiomycetes from excised Sitka spruce mycorrhizas) were isolated from stands of Sitka spruce either in monoculture or in a mixture with Japanese larch in an Irish conifer plantation. The growth of these fungi and their mycorrhizal formation in Sitka spruce and Japanese larch were examined after incubation in modified Melin-Norkrans medium containing either KH2PO4, Ca3(PO4)2 or Fe phytate as the phosphorus (P) source. P. involutus and S. grevillei utilized all three P sources. The unidentified basidiomycetes had limited ability to utilize Fe phytate. Basidiomycete 1 showed poor growth on KH2PO4 whereas growth of basidiomycete 2 was low on Ca3(PO4)2. Pure culture synthesis studies confirmed that P. involutus and the two basidiomycetes formed mycorrhizas with both tree species but S. grevillei was mycorrhizal only on Japanese larch. P. involutus formed more mycorrhizas in both conifers than the other fungi. Following inoculation with each of the four fungi, shoot and root dry mass of both Sitka spruce and Japanese larch seedlings was enhanced compared with uninoculated/nonmycorrhizal controls. On Fe phytate, Paxillus-inoculated Sitka spruce seedlings had the lowest primary root length and on KH2PO4, Suillus-inoculated Japanese larch had the greatest number of short roots. The only differences when Sitka spruce seedlings were grown in either monoculture or in a mixture with Japanese larch mycorrhizal with S. grevillei were primary root length and number of short roots after growth on media containing Fe phytate.  相似文献

Robust, polymorphic microsatellite DNA markers (simple sequence repeats—SSRs) are valuable tools for a range of tree conservation and breeding applications. SSRs are routinely used in the study of population genetic structure and diversity, pedigree reconstruction and genetic linkage mapping. Their abundance in the genome, co-dominant inheritance and potential for cross-species amplification make microsatellites highly prized markers. This paper characterises 22 novel genomic polymorphic microsatellite loci for Sitka spruce (Picea sitchensis (Bong.) Carr.). Amplification of DNA from Sitka spruce material was carried out both with a set of unrelated trees to obtain diversity statistics for each locus, and with the progeny of a full-sib family to test simple Mendelian inheritance. Observed heterozygosity ranged from 0.38 to 0.91 and allele number per locus ranged from 6 to 21, with a mean of 12.2. In addition, the primer pairs were tested with DNA from Norway spruce (P. abies) and white spruce (P. glauca) to investigate their potential for cross-species amplification and ten loci amplified in all three species. The results from these genomic microsatellites are compared to data generated from microsatellites derived from Picea EST libraries. In summary, this novel, highly polymorphic markers represent a significant addition to the rapidly expanding Picea genomics tool-box. 1 Green spruce aphid (Elatobium abietinum) is a serious pest of spruce (Picea spp.) in north‐west Europe, causing defoliation of one‐year‐old and older needles. 2 Relationships between population development of E. abietinum, needle loss and tree growth were compared for five pure genotypes of Sitka spruce and mixed‐genotype material of Sitka and Norway spruce, grown under high and low nutrient conditions. 3 Despite wide differences in flushing date between spruce genotypes, E. abietinum populations peaked on the same date on each genotype and on the mixed‐genotype material, irrespective of nutrient supply. 4 Larger aphid populations developed on trees grown under high nutrient conditions than under low nutrients. However, more needles were lost per aphid in the low nutrient treatment and overall defoliation rates in the two nutrient treatments were similar. 5 Total aphid numbers differed significantly between Sitka spruce genotypes within nutrient treatments, but not in relation to bud‐burst or needle terpene content. Reductions in height growth caused by infestation were greater (15–44%), and related to mean aphid densities and defoliation, in the low nutrient treatment, but were smaller (11–27%) and not related to aphid density and defoliation in the high nutrient treatment. 6 Development of E. abietinum populations was similar on Norway and Sitka spruce, but Norway spruce lost fewer needles. However, the effects of infestation on tree growth were more closely related to aphid density and were similar for Norway and Sitka spruce. 7 Infestation caused a decrease in total root dry weight of Norway and Sitka spruce in proportion to the reductions observed in above‐ground growth.  相似文献

Sitka spruce [Picea sitchensis (Bong.) Carr.] seedlings were grown for 3 years in an outside control plot or in ambient (355 mol mol-1) or elevated (ambient + 350 mol mol-1) atmospheric CO2 environments, within open top chambers (OTCs) at the Institute of Terrestrial Ecology, Edinburgh. Sequential harvests were carried out at the end of each growing season and throughout the 1991 growing season, five in all. Plants grown in elevated CO2 had, (i) 35 and 10% larger root/shoot ratios at the end of the first and third season, respectively, (ii) significantly higher summer leader extension relative growth rates, which declined more rapidly in early autumn than ambient grown plants, (iii) after three growing seasons a significantly increased mean annual relative growth rate, (iv) consistently lower foliar nutrient concentrations, and (v) after two growing seasons smaller total projected needle areas. Plants grown inside OTCs were taller, heavier and had a smaller root/shoot ratio than those grown outside the chambers. There was no effect of CO2 concentration on Sitka spruce leaf characteristics, although leaf area ratio, specific leaf area and leaf weight ratio all fell throughout the course of the 3 year experiment.  相似文献

Urea fertilizer labelled with 15N (2.5 atom %) was applied to a 20 year old Sitka spruce stand on a peaty gley at a rate equivalent to 160 kg N ha−1. The application of urea resulted in increased biomass and N concentration of needles and enhanced development of the crown. Differences in N concentrations of the amended trees were also observed for new wood and bark. Analysis of 15N in tree biomass showed a continued influence of fertilizer N in the second growing season following urea application. The overall recovery of fertilizer N in the trees was estimated to be about 10%.  相似文献

Sitka spruce planted on nutrient-poor soils in mixture with pine or larch, unlike pure spruce, does not become N deficient and does not require N fertilizer. To test the hypothesis that N availability in the soil is enhanced beneath mixed species, the seasonal changes in different N forms were compared in humus (L+F+H) and soil beneath 15-year-old Sitka spruce (SS) and mixed Sitka spruce-Scots pine (SS and SP) planted on a gleyed heathland soil. Amounts of mineral and organic N extracted from humus in spring were significantly (p < 0.05) higher in SS and SP than in SS. Larger amounts were measured in the underlying soil, which favoured the deeper-rooting spruce and pine in SS and SP plots. Annual net N mineralization, measured by in-situ incubation, was 32 and 47 kg N ha-1 in the surface 10 cm beneath SS and (SS and SP), respectively. In spring, readily mineralized organic N (waterlogged incubation at 30°C) was higher in humus and soil from (SS and SP) than from SS by 15 kg N ha-1. The larger N pools beneath (SS and SP) were consistent with the higher total N content of the humus beneath (SS and SP), 446 compared with 255 kg N ha-1 beneath SS. This indicated that beneath (SS and SP) N had been transferred from the underlying soil.  相似文献

An investigation of fine (< 1 mm in diameter) and small (1–2 mm in diameter) roots in the organic soil layer was carried out in a Norway spruce forest stand with different treatments of water and nutrients, including control (C); ammonium sulphate application (NS); nitrogen-free fertilization (V); irrigation with liquid fertilization (a complete nutrient solution) (IF); NS followed by artificial drought (ND); V followed by artificial drought (VD). In order to evaluate the vitality and function of the fine roots, the following approaches were used: i) classification of fine roots, based on morphological characteristics; ii) nutrient uptake bioassay, using 32P-phosphate and 35S-sulphate; iii) nutrient concentration in fine roots and its relation to nutrient uptake. The NS treatment showed effects on the fine and small roots, with a decrease in amount of living roots, and a decrease in the total amount of fine and small roots. The VD treatment resulted in increased amounts of living small roots, while the ND treatment showed the opposite, as compared with the V and NS treatments, respectively. The uptake of P was negatively related to the P supply, with a higher P uptake for C and NS fine roots than for IF and V fine roots. The specific root length (SRL, m g-1 DW) decreased for NS fine roots and increased for IF fine roots, indicating a further increase in uptake for NS roots and a decreased uptake for IF roots if calculated on a root length basis. So far, the NS and IF treatments maintain a considerable increase in above-ground biomass with a significantly reduced root biomass and standing crop.  相似文献

Relationships between tree parameters above ground and the biomass of the coarse root system were examined in six mixed spruce-beech stands in the Solling Mountain region in northwest Germany. The selected stands were located on comparable sites and covered an age range of 44 to 114 years. Coarse roots (d?\ge?2 mm) of 42 spruce and 27 beech trees were sampled by excavating the entire root system. A linear model with logarithmic transformation of the variables was developed to describe the relationship between the coarse root biomass (CRB, dry weight) and the corresponding tree diameter at breast height (DBH). The coefficients of determination (R 2) attained values between 0.92 for spruce and 0.94 for beech; the logarithmic standard deviation values were between 0.29 and 0.43. A significantly different effect of tree species on the model estimates could not be detected by an analysis of co-variance (ANCOVA). For spruce, the derived relationships were similar to those reported in previous studies, but not for beech. Biomass partitioning in the tree compartments above and below ground differs significantly between spruce (coarse root/shoot ratio 0.16±0.06) and beech (coarse root/shoot ratio 0.10±0.03) in the mixed stands. These results are similar to those given in other studies involving pure spruce and beech stands on comparable sites in the region, although the ratios of pure stands in other regions growing under different site conditions are somewhat higher. Comparing trees of the same DBH classes, root/shoot ratios of spruce are 1.2 to 3 times higher than those of beech. Dominant spruce trees (DBH>60 cm) attained the highest ratios, suppressed beech trees (DBH<10 cm) the lowest. Site conditions of varying climate and soils and interspecific tree competition are likely to affect root/shoot ratio and DBH-coarse root biomass relationships. The greater variability in beech compared with spruce indicates a high 'plasticity' and adaptability of beech carbon allocation. Thus, the derived equations are useful for biomass estimates of coarse roots involving trees of different ages in mixed stands of spruce and beech in the Solling Mountains. However, application of these relationships to stands in other regions would need further testing.  相似文献

Studies on biogeochemical cycling of elements are performed in even aged forest ecosystems of Norway spruce, Sitka spruce and beech at several locations in Denmark. Episodes with high sea salt deposition and unusual low water surplus percolation caused extreme salt concentrations in the soil solutions. The changes were smallest beneath beech stands and most pronounced at the most salt-affected Sitka spruce stands. Contemporary, the pH drops and the Al3+ concentration increased above 20 mg L-1 owing to cation exchange with Na+. The Cl- and Al3+ concentrations reached levels reported as toxic for Norway spruce, but not for Sitka spruce and beech. The changes in the soil water chemistry must be considered as important factors for the reduced vitality in Norway spruce ecosystems in Denmark.  相似文献

  1. A warming climate, as predicted under current climate change projections, is likely to influence the population dynamics of many forest insect species. Numerous bark beetle species in both Europe and North America have already responded to a warming climate by significantly expanding their geographical ranges.
  2. The aim of the current study was to investigate how populations of bark beetles within stands of Sitka spruce, a widely planted non-native commercial plantation tree species in the U.K., were likely to respond to a warming climate. Experimental plots were established in stands of Sitka spruce over elevational gradients in two commercial forest plantations, and the abundance and emergence times of key bark beetle species were assessed over a 3-year period using flight interception traps. The air temperature difference between the lowest and highest experimental plot in each forest was consistently >1°C throughout the 3-year period.
  3. In general, the abundance of the most dominant bark beetle species (e.g. Trypodendron, Dryocoetes, Hylastes spp.) was higher, and emergence times tended to be earlier in the year at the lower elevation plots, where temperatures were higher, although not all bark beetle species responded in the same manner.
  4. The results of the study indicated that, under the projected future climate warming scenarios, monoculture Sitka spruce stands at low elevations may potentially be more vulnerable to significant outbreak events from existing or invasive bark beetle species. Hence, consideration of establishing more resilient forests of Sitka spruce by diversifying the species composition and structure of Sitka spruce stands is discussed.

Abscisic acid at 104 M inhibited elongation and induced dormancyin roots of Sitka spruce cuttings growing in solution culture.The dormancy was characterized by superficial browning whichprogressed towards the root tip and by the development of alayer of lignified and suberized cells around the root apex.The dormant roots remained alive and resumed growth on transferto ABA-free solutions. The induction of dormancy was confinedto those roots actually bathed in the ABA solution and a rangeof conditions which affected shoot activity did not alter theresponse. By contrast, the time for which the roots had beenelongating strongly influenced root dormancy since it was notinduced in roots which had recently begun to elongate in solutionculture. The seasonal course of nitrogen uptake, incorporation and remobilization in different shoot components of winter oilseed rape (Brassica napus L.) was studied under field conditions including three rates of 15N labelled nitrogen application (0, 100 or 200 kg N ha-1) and two irrigation treatments (rainfed or watered at a deficit of 20 mm). The total amount of irrigation water applied was 260 mm, split over 13 occasions in a 7-week-period ranging from 1 week before onset of flowering until 4 weeks after flowering.Nitrogen application and irrigation increased plant growth and nitrogen accumulation. Irrespective of N and irrigation treatment more than 50% of total shoot N was present in the stem when flowering started. At the end of flowering, pod walls were the main N store containing about 30–40% of shoot N. The quantities of N remobilized from stems and pod walls amounted in all treatments to about 70% of the N present in these organs at mid-flowering. At harvest, stem and pod walls each contained about 10% of total shoot N, the remaining 80% being incorporated into seeds. The main component contributing to the response of seed N accumulation to nitrogen application and irrigation was pods in axillary racemes. Up to 20 kg N ha-1, corresponding to about 10% of final shoot N content, was lost from the plants by leaf drop.Irrigation increased the recovery at harvest of applied N from 30% to about 50%, while the level of N application did not affect the N recovery. 15N labelled (fertilizer derived) nitrogen constituted a greater proportion of the N content in old leaves than in young leaves and increased with age in the former, but not in the latter. Relative to soil N, fertilizer derived N also contributed more to the N content of vegetative than to that of reproductive shoot components. Small net changes in shoot N content after flowering reflected a balance between N import and export, leading to continuous dilution of 15N labelled N with unlabelled N.  相似文献

A rapid, one-step procedure has been developed for inducing direct organogenesis and somatic embryogenesis in cultures of Phaseolus coccineus L., P. acutifolius A., P. aureus L. [Vigna radiata L. Wilczek] and P. wrightii L. Development of somatic embryos and shoot buds occurred within 6–8 weeks of culture from intact seedlings raised on MS (Murashige and Skoog 1962) medium supplemented with N6-benzylaminopurine (BAP). Shoot buds or embryoids originated from subepidermal tissue of the regions adjacent to the shoot apex, hypocotyl and cotyledonary axils. While P. acutifolius and P. aureus were regenerated via shoot formation and P. wrightii by somatic embryogenesis, both embryogenesis and shoot regeneration were observed in P. coccineus. Relatively higher levels of BAP, 50–80 M, were found to be optimal for inducing regeneration while lower concentrations were ineffective. About 40–70 shoots and 70–250 somatic embryos were produced per responding seedling. Regenerated shoots and somatic embryos developed into whole plants on a basal medium or the one supplemented with 1 M naphthaleneacetic acid.  相似文献

Abstract Maximum germination of Sitka spruce seed was obtained at 22°C under continuous fluorescent light or following a single 10 min exposure to red light (RL) given after 48 h of dark imbibition. The levels of endogenous cytokinins and gibberellins were examined in seeds which were exposed to different light regimes. Cytokinin levels in Sitka spruce seeds increased approximately 1.7–fold during exposure to continuous white light, during dark stratification, or within 1 h or a 30 min exposure to RL. A single peak of activity could be eluted from the LH?20 Sephadex column which appeared in the same volume as authentic zeatin riboside. Gibber-ellin activity in Sitka spruce seeds increased rapidly during exposure to 30 min of RL and then declined during subsequent dark incubation. Similarly, under continuous white light, gibberellin activity was at least twice as high as in the dark. Two gibberellm-like substances were found in Sitka spruce seeds which appeared to be similar to GA7 and GA9 on the basis of their chromato-graphic properties. The zone of gibberellin activity directly associated with photo-excitation of Sitka spruce seeds contains conjugate gibberellins, possibly glucosyl esters, which when hydrolysed with β-glucosidase form two products which are similar to the free gibberellins which were detected. The light response in Sitka spruce seed is compared to a similar response in Scots pine (Pinus sylvestris L.) seeds.  相似文献

Cloned Sitka spruce grafts were subjected to NH4NO3 mist (1 mm day-1 equivalent) acidified with H2SO4, for 4 days a week, from April until November. Three N treatments at pH 5.0 used 0.01 mol m-3 15NH4 15NH3, 1.6 mol m-3 15NH4 NO3 or 1.6 mol m-3 NH4 15NO3. At pH 2.5, the treatments were15NH4 15NO3,15NH4NO3 and NH4 15NO3 all at 1.6 mol m-3. At the end of the growing season,15N was found in every part of the trees, even when NH4NO3 was supplied at 0.01 mol m-3. It was shown that both and could enter needles to a similar degree and be transported to the roots. No differences between pH treatments or clones could be demonstrated. Entry of labelled N via the roots, in those trees with unprotected compost, was reflected in higher fine root15N content, but no change in shoot15N levels. Per cent incorporation expressed as labelled N as a percentage of total N, was generally at its highest in bark, and in older needles in all treatments, values ranging from 5 to 11%. When15N content was expressed as total content for each tree part on a per tree basis, 25–36% was found in current year needles, with a further 12% in current year bark. The most reproducible data was that for dry weight of tree parts as per cent of the whole tree, where proportions compared closely between treatments and the two clones used. The implications of the results for cuticular transport mechanisms, N storage and internal cycling are discussed.  相似文献
