While anther culture or pollinations with Hordeum bulbosum have provided suitable methods for haploid production in bread wheat, they have been largely unsuccessful in durum wheat. Pollinations with maize were used in an attempt to produce haploid seedlings and, from these, fertile doubled haploids of durum wheats. Moreover, the effect of various concentrations and combinations of a synthetic auxin, 2, 4-dichlorophenoxyacetic acid (2,4-D), kinetin, and an ethylene inhibitor, silver nitrate (AgNO3), on embryo recovery were also investigated. Haploid seedlings were recovered from Triticum turgidum ssp. turgidum cv Rampton Rivet pollinated with maize following in-vivo treatment of ovaries with 2,4-D for 2 weeks and subsequent embryo culture. The recovery of haploid seedlings from T. turgidum ssp. durum cv. Wakona pollinated with maize necessitated the addition of AgNO3, to the 2,4-D treatment. Overall, haploid seedlings were produced in 1.7% and 3.3% of pollinated florets for Rampton Rivet and Wakona respectively. The success of the present work represents a significant breakthrough for haploid production in durum wheats. Wide hybridization with maize followed by in-vivo treatment of ovaries with 2,4-D alone, or in combination with AgNO3, may provide a widely-applicable method of haploid production in tetraploid wheats.  相似文献

Summary Meiotic chromosome pairing and Giemsa C-banding analyses in crosses of several European blue-grained wheat strains with Chinese Spring double ditelosomic and other aneuploid lines showed that Triticum aestivum Blaukorn strains Berlin, Probstdorf, Tschermak, and Weihenstephan are chromosome substitutions, in which the complete wheat chromosome 4A pair is replaced, whereas the strains Brünn and Moskau are 4B substitutions. The alien chromosome pair in all of these strains is an A genome chromosome (4A) from diploid Triticum monococcum or T. boeoticum not present in common tetraploid and hexaploid cultivated wheats. The Blaukorn strain Weihenstephan W 70a86 possesses, in addition to a rye chromosome pair 5R compensating for the loss of part of chromosome 5D, a 4A/5DL translocation replacing chromosome pair 4B of wheat.  相似文献

Summary The progeny of 551 regenerants of the hexaploid wheat cultivar Millewa were analysed for somaclonal mutants at the threeAdh-1 loci in hexaploid wheat. Seventeen regenerants gave rise to progeny having altered ADH1 zymograms. Progeny with altered zymograms in 13 of these regenerants were aneuploid. The remaining 4 regenerants gave rise to euploid progeny with altered ADH1 zymograms. The genetics of three of these somaclonal mutants is described in detail. These regenerants were interpreted to possess a 4A isochromosome, a 3BS/4A translocation and a 7BS/4A translocation, respectively.This research was partly supported by the Rockefeller Foundation and a Reserve Bank of Australia Rural Credit Development Fund research grant  相似文献

Summary A complete set of isogenic wheat/rye D-genome substitutions were produced by crossing an inbred line of spring rye Secale cereale L. cv. Prolific to a tetraploid wheat, the A-and B-genomes of which had previously been extracted from hexaploid wheat, Triticum aestivum L. em Thell. cv. Thatcher. After chromosome doubling, the derived hexaploid triticale (x Triticosecale Wittmack) was backcrossed to 6x Thatcher and selection for wheat/rye substitution lines was carried out in BCF3 to BCF6 families by using Giemsa C-banding. Five fertile disomic wheat/rye D-genome substitution lines were obtained and their chromosomal constitution was determined to be 1D/1R, 2D/2R, 7D/4R, 6D/6R, 7D/7R. The two remaining 3R and 5R substitutions are at the moment in a monosomic condition. Another 1D/7R substitution was detected but this plant was very weak and sterile, indicating that only substitutions between homoeologous chromosomes result in fertile, vigorous plants. Furthermore, many rye telocentrics as well as rye-rye and rye-wheat translocations were selected. Since all lines selected in this program share the same genetic background of Thatcher wheat, genetic heterogeneity is excluded. The material is very useful, therefore, for analyzing the effects of different rye chromosomes or chromosome segments in an otherwise homozygous background.Contribution No. 797  相似文献

The inheritance of alpha-amylases was studied insix F2 populations of hexaploid oats (Avenasativa, A. byzantina, A. fatua, A. sterillis) usingpolyacrylamide gel electrophoresis. A total of 22 lociwas identified and described. Three main linkagesof four or five loci each and an additional two pairs oflinked loci were detected. It seems likely that thethree main linkage groups represent homeologous chromosomes. Matching of alpha-amylase profilesof hexaploid (AACCDD), tetraploid (AACC), and diploid(AA) species was made to assign the linkage groups toparticular subgenomes in the hexaploid oat. It was proposed that Linkage 1 (Amy12-Amy10-1-Amy4-Amy13-Amy11) belongs to the D-subgenome; Linkage2 (Amy10-2, Amy9-Amy8-Amy6) belongs to the A-subgenome;and Linkage 3 (Amy7-Amy3-Amy5-Amy2) belongs to the C-subgenome of the hexaploid oat. Themalt and greenalpha-amylases in hexaploid and tetraploid oats havebeen identified. Isozymes of greenalpha-amylase were slower in electrophoretic mobility than other isozymesand were governed by loci assigned to the A- andD-subgenomes.  相似文献

Shoot tips of Triticum aestivum L. cvs. Turbo and Nandu, both summer wheat varieties, were excised from 4 and 10 day-old seedlings, and used for induction of embryogenic callus. A modified L3 medium, supplemented with 10 M 2,4-dichlorophenoxyacetic acid (2,4-d) for culture initiation, and 5 M 2,4-d for subculturing, was optimal; 90% of 4 day-old Turbo seedlings formed embryogenic callus. Optimal plant regeneration was achieved from callus incubated on a modified MS medium without 2,4-d, but supplemented with 2.22 M 6-benzylaminopurine and 0.27 M naphthaleneacetic acid. Plantlets formed via embryogenesis from all embryogenic Turbo calli initiated from 4 day-old explants, with a mean number of 8 regenerants per explant. Regeneration occured via embryogenesis only. Summary Twenty two basidiomycetes, mostly white rot fungi, were grown on wheat straw. Lignin-, cellulose-, and hemicellulose-degradation was recorded in order to find a species growing on lignin preferably. The oyster-mushroomPleurotus sp. florida showed fastest delignification of all tested fungi.Straw pretreated by this fungus was fermented anaerobically to biogas. Summary The common wheat selection 79-4045 was identified as a wheat-rye 1B/1R chromosome translocation line, by means of C-banding patterns and test cross with Chinese Spring double-ditelosomic line. Summary Wheat accessions lacking some of the - and -gliadin components encoded by the Gli-1 loci on the short arm of chromosome 1D in bread wheat and chromosome 1A in durum wheat were studied by two-dimensional polyacrylamide gel electrophoresis and restriction fragment analysis. Digested genomic DNAs of normal and null forms were probed with a cDNA clone related to -/-gliadins and with a genomic clone encoding an LMW subunit of glutenin. The hybridisation patterns with the -/-gliadin probe were similar to those of cvs Chinese Spring and Langdon used as standards for bread and durum wheats, respectively, but several restriction fragments located on the 1D chromosome of bread wheat and the 1A chromosome of durum wheat were absent in the null forms. In addition, specific LMW glutenin fragments encoded by the same chromosomes were also absent in the null forms, suggesting that simultaneous deletions of blocks of genes for both -/-gliadins and LMW glutenins had occurred. Comparisons of the protein and RFLP patterns enabled some proteins to be mapped to specific restriction fragments.  相似文献

Summary Amphiplasty in hexaploid triticale, the artificial amphiploid of tetraploid wheat and diploid rye, is analyzed for the first time using a modified, highly reproducible, silver-staining procedure. A comparative analysis of metaphase somatic cells by phase contrast, C-banding and silver-staining of the hexaploid triticale cv. Cachirulo and its parents, namely, the tetraploid durum wheat cv. Enano de Andujar and the diploid rye cv. Petkus has been made. Two silver-stained nucleolar organizer regions (Ag-NORs) (the chromosome pair 1 R) are observed in all rye plants analyzed, whereas four Ag-NORs (chromosome pairs 1 B and 6 B) are found both in the tetraploid wheat parent and in the triticale. The rye Ag-NORs are absent in the triticale. Since the Agstaining reaction of NORs can be considered as an indication for genetic activity, the silver procedure can be used to visualize gene functionality at the rDNA sites with conventional light microscopy and, consequently, the modified Ag-staining method described can be very useful in analyzing the amphiplasty phenomenon in natural or artificial hybrid combinations and derivatives in the Triticum group and its relatives.  相似文献

Summary Electrophoretical analyses of the gliadin fraction extracted from seeds of the intervarietal substitution lines of T. aestivum ssp. spelta in the T. aestivum ssp. vulgare cv Chinese Spring for the homoeologous groups 1 and 6 and substitution lines of 6D chromosome of Chinese Spring in the durum wheat cv Langdon allowed the identification of seeds without gliadin proteins controlled by genes on chromosome 6A and 6B. A gliadin component of Chinese Spring, not previously assigned to any specific chromosome, is controlled by chromosome 6D in the 6D (6A) and 6D (6B) disomic substitution lines of Langdon. Additional genes controlling the synthesis of this component may be present on other chromosomes, very likely 6A and 6B, since the analysis of the Chinese Spring compensating nullisomic-tetrasomics involving the 6D chromosome does not show the loss of this component or any apparent change in staining intensity. Chromosomal location data and two-dimensional gliadin maps reveal close homologies between the two hexaploid wheats, Chinese Spring (T. aestivum ssp. vulgare) and T. aestivum ssp. spelta, belonging to different subspecies in the hexaploid group of genomic formula AABBDD. The comparison of gliadin electrophoretic patterns aiding in the identification of evolutionary pathways in wheat is stressed.  相似文献

Summary The low molecular weight heat shock protein (HSP) profiles of the hexaploid wheat cultivar Chinese Spring and its ditelosomic series were characterized by isoelectric focusing polyacrylamide gel electrophoresis of denatured in vivo radiolabeled proteins. Comparisons of the ditelosomics (DTs) to the euploid Chinese Spring enabled the assignment of genes controlling 9 of the 13 targeted HSPs to seven chromosome arms. There did not appear to be a genome-specific action in the regulation of expression of these HSPs. There did appear to be a higher frequency of controlling genes within homoeologous DT lines 3, 4 and 7. Significant variation in protein quantity was evident among the DT lines for some HSPs, while other HSPs were remarkably stable in their expression across all DTs examined. The results are useful in identifying specific DT lines for the investigation of HSP functions in hexaploid wheat.  相似文献

Aegilops tauschii (2n=2 x=14, DD) is a rich source of genetic variability for hexaploid wheat (Triticum aestivum, 2n=6 x=42, AABBDD) improvement. This variability can be accessed through utilizing synthetic hexaploid wheat lines, which contain genomes from Ae. tauschii and T. turgidum (2n=4 x=28, AABB). Numerous desirable characteristics can and have been introgressed into common hexaploid wheat with this germplasm. In this work, the genetic variability in the two puroindoline genes (a and b) contained on the D genome, and the relationship that sequence polymorphisms in these genes have on endosperm texture among a population of 75 CIMMYT synthetic hexaploid accessions is described. Kernel texture was evaluated using the single kernel characterization system (SKCS). Kernel texture differed significantly (P0.0001) among the synthetic hexaploid accessions (range 2.6–40.9) and the parent types, durum or Ae. tauschii. The interaction term between parent types was also a significant effect (P0.0001). In addition to the wild-type protein sequences of the puroindoline genes (those present in Chinese Spring and all other soft wheats), three other translated sequences were identified in puroindoline a and two others in puroindoline b. These protein sequences were associated with significantly (P0.0001) softer endosperm textures than the wild-type protein sequences. As the softer alleles are expressed in a hexaploid background, they are immediately available to wheat breeding programs.  相似文献

Crop residues protect soils from erosion, reduce soil water evaporation and increase soil organic matter. Yet management of stubbles for cropping can be difficult. Surface-retained residue can act as a mechanical barrier to slow emergence and reduce seedling biomass. Longer coleoptiles improve seedling emergence with deep sowing and may assist where stubble load is large. In a glasshouse study, six wheat and barley genotypes were sown at 30 and 50mm depth into pots containing pasteurised soil. Unweathered sorghum, canola and wheat stubble were added at 0, 3 and 6t/ha equivalents to the soil surface and pots watered above or below the stubble. Stubble species and watering regime had little effect on seedling growth. However, deeper sowing and increased stubble mass adversely affected most seedling characteristics particularly slowing seedling emergence and reducing tiller number to decrease plant biomass (environmental correlations (re) of –0.98** and 0.88**, respectively). Shorter coleoptile Rht-B1b wheats Banks and Janz, and barley Beecher emerged slower and abnormally with thicker stubble, and had more sterile tillers to reduce total tiller number and biomass. Deeper crowns for these genotypes also resulted in proportionally less biomass located above the stubble. The converse was true of long coleoptile Vigour 18, Halberd, and its Rht8 progeny, HM14bS which were less affected by stubble mass and sowing depth. In a corresponding field study, increasing wheat stubble mass from 0 to 3 and 6 t/ha delayed seedling emergence and decreased plant number to reduce biomass. Short coleoptile wheat genotypes Hartog and Janz emerged slower and produced less biomass at 3 and 6t/ha of stubble than long coleoptile wheat genotypes Halberd and HM14bS. Emergence of seedlings sown at 50mm depth with 6t/ha overlying stubble was similar to that sown at 120mm with no stubble, reflecting the similar impact of retained residues to deep sowing. Genetic variation for coleoptile length and availability of gibberellin-responsive dwarfing genes such as Rht8 will allow development of long coleoptile wheats for deep sowing or where stubble retention is practiced.  相似文献

Summary Two double heterozygous 0/0 thalassemic sibs of Mexican descent were studied. The father had a 0/0 genotype, while the mother, one sib and several maternal relatives were 0/0 heterozygotes. Parental consanguinity and an apparently low frequency of thalassemia among Mexicans suggested a possible common origin of both 0 and 0 genes. A hypothesis to explain such a possibility is proposed on the basis of a partial mispairing between 0 and genes followed by a crossing-over which would results in a 0 recombinant gene. This hypothesis could also be extended to explain the 22 gluala, 22 alaglu and 116 arghis Hb variants as recombinants from double crossing-over between and mispaired genes for which the name interstitial-Lepore is proposed.  相似文献

Zusammenfassung Harzkonservierte Fossilien ermöglichen bei Anwendung adäquater Methoden die morphologische Analyse der Feinmerkmale bis zur Auflösungsgrenze des Lichtmikroskops, Beobachtung in verschiedenen Ebenen und Richtungen, und somit konkrete Rückschlüsse auf die Wirkung und Bedeutung der Einzelelemente und des Gesamtgefüges.Eine so eingehende funktionsmorphologische Analyse mit Berücksichtigung der Positionsvariation (graduell verschiedene Gestaltung in gesetzmäßiger Abhängigkeit von der Lage innerhalb der Gesamtfeder) der Einzelelemente wie Abzweigungs-, Knick-, Neigungswinkel, Krümmung, Länge, Dicke, Querschnitt, Dichte, Differenzierungsgrad der verschiedenen Abschnitte von Rhachis, Rami, Radii inklusive Häkchen und Cirren wird erstmals für fossile Vogelfedern geliefert (hier als Abriß zu einer dokumentarisch und thematisch ausführlicheren Darstellung in Stuttgarter Beiträge zur Naturkunde).Diese Federn entstammen der untersten Unterkreide und sind damit nur relativ wenig jünger alsArchaeopteryx. Sie weisen extrem differenzierten Aufbau auf, der auf hohe flugtechnische und wärmeisolierende Leistungsfähigkeit schließen läßt.Die hier vorgelegten funktionsmorphologischen Ermittlungen an fossilen Körperkonturfedern mögen auch zu einer intensiveren Analyse der bis jetzt stark vernachlässigten Untersuchung ganz normaler Körperfedern rezenter Vögel anregen. Erst dann, nach umfassender Kenntnis ihrer Ausgestaltung innerhalb der verschiedensten rezenten Vogelgruppen, läßt sich überzeugend begründen, ob und wieweit die hier vorgelegten Federn dieses Unterkreide-Vogels noch ursprüngliche Elemente (Plesiomorphien) oder ihnen eigene Sonderbildungen (Autapomorphien) aufweisen; das gilt sowohl für morphologische wie für funktionelle Elemente der Gesamtstruktur.
Resin-preserved fossil bird's feathers from the Lowermost Cretaceous
Summary Parts of some feathers, originating from a single bird, were discovered in our collections of Lower Cretaceous amber from the Lebanon mountains — which, in general, contains the oldest terrestrial microfossils preserved with all morphological details.These contour feathers of the trunk, which are nearly as old as Archaeopteryx (Lowermost Cretaceous: Neocomian/Uppermost Jurassic: Kimmeridigian) were studied with magnifications of 500–900 in several levels by a special technique. (In normal fossils, i.e., impressions, the granulation of the sediment and the fossil's bulky carbon remainders cause a blurred image even at a magnification of merely 100).Special emphasis was laid on the study of the individual elements' gradual variation, depending on the respective position within the total feather (position variation). Where appropriate, an analysis of lengths, quantity, degree of differentiation, angle of inclination, break, and branching, cross-sectional view, curvature, etc. of the rhachis, rami, distal and proximal radii, barbicles, hooklets, etc. were undertaken. [Through measurements of the depth of details the effects caused by a sloping position (apparent variation) may be precisely separated from the real variation.]On the basis of such a detailed knowledge of structure and relative position a thorough functional analysis of the single elements as well as the total system is given.Principal features: The production of plain stability in the feather's center, and of flexibility in its apical and lateral rims; dispersion of forces in case of pressure or a pulling load; function of the hooklets (which donot serve as an interlocking mechanism while the feather is in the normal resting position, but function with increasing braking action only when a neighboring ramus diverges to a precisely defined extent from its resting position) including the mechanism of their unhooking; devices for the avoidance of harmful hooking into contacted parts of other feathers; production of maximal stability by minimal air resistance, and of minute chambers (<0,00001 mm3) with still air for optimal heat isolation.Apart from this abstract, further information, accompanied by numerous figures, will be given in a later paper in Stuttgarter Beiträge zur Naturkunde.

Summary Embryo and endosperm peroxidases from dry mature seeds of three subspecies of tetraploid wheat (Triticum turgidum L.) were subjected to genetic analysis. The inheritance of eight isozymes (embryo isozymes a2, d1, d2, e and f; and endosperm isozymes b, d and 4) were studied in F2's obtained from different wheat accessions. Simple monogenic inheritance producing three banded: one null segregation and two epistatic segregations (97 and 151) were found. In the case of isozymes b, d and 4, monogenic or epistatic segregation depended on the F2 analyzed. Segregation data indicated that at least 9 different loci would determine the peroxidase isozymes of tetraploid wheat seed, all the loci studied containing null alleles. Furthermore, several loci determining embryo peroxidases were noticed to be mutually linked. All these data are discussed in context of the inheritance of seed peroxidases in hexaploid wheat and rye.  相似文献

Summary Ethidium bromide (EB) and ultraviolet light (UV) in combination are known to produce a synergistic induction of petite mutants in yeast. Two other agents were combined with EB, 3-Carbethoxypsoralene (3 CPs) activated by 365 nm light or rays. EB in combination with 3 CPs also resulted in an enhanced production of petite mutants. After the photoaddition of 3 CPs in exponential phase cells, recovery of the petite mutation during dark liquid holding was inhibited by the presence of EB producing an enhanced number of petite mutants. The behavior of mitochondrial antibiotic resistance markers after individual and combined treatments with EB and 3 CPs indicates a random loss of markers after EB and a preferential loss of a certain region for the 3 CPs photoaddition. The combination of the two agents leads to an additivity of total drug marker losses rather than a synergistic loss. The combination of EB with rays produced no enhancement in petite induction. A combination of UV and 3 CPs showed a synergistic interaction for petite induction. These results indicate that the three agents, EB, UV and 3 CPs photoaddition may share a common repair step for mitochondrial lesions.  相似文献

Summary Progenies of a tetraploid 1BL/1RS wheat-rye translocation line, CV 256, selected from the cross Cando x Veery, were analyzed by means of Giemsa C-banding. CV 256 is cytologically stable for the presence of the 1BL/1RS translocation but still segregating for A- and B-genome chromosomes of Cando and Veery. In CV 256, nucleolar activity of the 1RS NOR locus is suppressed, as judged by the absence of a secondary constriction in that rye segment and the capability of organizing nucleoli. PAGE analysis of prolamins confirmed the presence of two 1RS secalins in all single seeds analyzed. SDS-PAGE analysis of reduced glutenins of single seeds indicated that some seeds contained the Cando Glu-B1 locus (subunits 6+8), some contained the Veery Glu-B1 locus (subunits 7+9) while others contained all four subunits, indicating that the material was heterozygous. Pm8 resistance is expressed in the tetraploid 1BL/1RS translocation line based on the reactions of six well-defined powdery mildew isolates. However, Pm8 resistance is not expressed in the hexaploid wheat cultivars Olymp, Heinrich and Florida, which also contain the 1BL/1RS translocation. Obviously, the existence of the 1BL/1RS translocation is not a proof for the expression of the associated genes. PAGE results did not show a clear linkage between powdery mildew resistance and the presence of 1RS secalins.  相似文献

Three structural classes of (13)--d-glucans are encountered in some important soil-dwelling, plant-associated or human pathogenic bacteria. Linear (13)--glucans and side-chain-branched (13,12)--glucans are major constituents of capsular materials, with roles in bacterial aggregation, virulence and carbohydrate storage. Cyclic (13,16)--glucans are predominantly periplasmic, serving in osmotic adaptation. Curdlan, the linear (13)--glucan from Agrobacterium, has unique rheological and thermal gelling properties, with applications in the food industry and other sectors. This review includes information on the structure, properties and molecular genetics of the bacterial (13)--glucans, together with an overview of the physiology and biotechnology of curdlan production and applications of this biopolymer and its derivatives.  相似文献
