The compact conventional six-period torsatron L-V with an aspect ratio of A=3 has been considered as a starting point for further computational optimization of the collisionless confinement of the guiding center orbits of fusion α particles. The configurations obtained possess improved neoclassical transport together with an equilibrium β limit at a level of 〈β〉≈10%. Formate dehydrogenases (FDH) are useful for the regeneration of NADH, which is required for asymmetric reduction by several dehydrogenases and reductases. FDHs have relatively low activity and are labile, especially to -haloketones, thus FDH cannot be applied to the industrial manufacture of optically active -haloalcohols. To stabilize a FDH from Mycobacterium vaccae (McFDH) against the -haloketone ethyl 4-chloroacetoacetate (ECAA), a set of cysteine-mutant enzymes was constructed. Sensitivity to ECAA of mutant C6S was similar to that of the wild-type enzyme, and mutants C249S and C355S showed little activity. In contrast, mutant C256S exhibited remarkable tolerance to ECAA. Surprisingly, mutant C146S was activated by several organic compounds such as ethyl acetate. An optimized mutant, C6A/C146S/C256V (McFDH-26), was obtained by combining several effective mutations. Ethyl (S)-4-chloro-3-hydroxybutanoate [(S)-ECHB] was synthesized from ECAA to 49.9 g/l with an optical purity of more than 99% e.e. using recombinant Escherichia coli cells coexpressing McFDH-26 and a carbonyl reductase (KaCR1) from Kluyveromyces aestuarii.  相似文献

The incidence and range of endemic malaria caused by Plasmodium vivax has expanded during the past 30 years. This parasite forms hypnozoites in the liver, creating a persistent reservoir of infection. Primaquine (PQ), introduced 50 years ago, is the only drug available to eliminate hypnozoites. However, lengthy treatment courses and follow-up periods are not conducive to assessing the effectiveness of this drug in preventing relapses. Resistance to standard therapy could be widespread. Studies are urgently needed to gauge this problem and to determine the safety, tolerability and efficacy of shorter courses and higher doses of PQ.  相似文献

Recent studies have shown that nitroxyl (HNO) (1HNO/3NO?), which is the one-electron-reduced form of nitric oxide (NO), has unique biological activities, especially in the cardiovascular system, and HNO-releasing agents may have therapeutic potential. Since few HNO donors are available for use under physiological conditions, we synthesized and evaluated a series of Piloty’s acid (PA) derivatives and evaluated their HNO-releasing activity under physiological conditions. N-Hydroxy-2-nitrobenzenesulfonamide (17) was the most efficient HNO donor among our synthesized PA derivatives, including the lead compound, 2-bromo-N-hydroxybenzenesulfonamide (2). The high HNO-releasing activity is suggested to be due to electronic and steric effects. Compound 17 may be a useful tool for biological experiments.  相似文献

Breeding Brünnich's guillemots Uria lomvia show stepwise mass loss at the time of hatch. This mass loss has usually been explained as an adaptation to reduce the cost of flight during the chick‐rearing period because flight time increases during that period. It is possible, however, that mass loss also increases dive performance during the chick‐rearing period because time spent diving also increases during that period. Reduced mass could reduce basal metabolic rate or costs associated with buoyancy and therefore increase aerobic dive limit. To examine the role of mass loss in dive behavior, we attached time‐depth‐temperature recorders for 24–48 h to chick‐rearing and incubating Brünnich's guillemots at Coats Island, Nunavut (2005: n=45, 2006: n=40), and recorded body mass before and after each deployment. There was no relationship between mass and dive duration during either incubation or chick‐rearing. Seventeen of the birds we sampled during incubation were resampled during chick‐rearing. For this group, dive duration increased with mass loss between incubation and chick‐rearing (r2=0.67–0.75). Mass loss occurred through reductions in metabolically‐active tissues (liver, bladder) and buoyant tissues (lipids) although muscle and gut mass did not change. Despite the large change in lipids, buoyancy only changed by 0.1%, and mass loss therefore did not have much effect on costs associated with buoyancy. Nonetheless, surface pause duration for a given dive depth decreased during chick‐rearing, supporting the idea that reduced mass led to increased aerobic dive limit through reduced metabolic rate and inertial costs; oxygen stores did not increase. We also attached neutrally (n=9) and negatively (n=11) buoyant handicaps to the legs of adults to assess the effect of artificial mass increases on time budgets. Artificially increasing mass decreased total time spent diving but did not change time spent flying. There was no change in shift length between incubation and chick‐rearing, and therefore no support for the idea that mass loss reflected a change in fasting endurance requirements. An energetic model suggested that the observed mass reduction reduced dive costs by 5–8% and flight costs by 3%. We concluded that mass loss may be as important for increasing dive performance as increasing flight performance.  相似文献

Abstract

Enzymes are indispensable biocatalysts required in various steps of textile processing to minimize various chemical-induced hazards. The present work focuses on the applications of the truncated α-amylase in textile industry for desizing of fabrics by starch hydrolysis. The multiple sequence alignment was performed to find homology and the possible truncation region in Bacillus subtilis MTCC 121 α-amylase with same bacilli family α-amylase. Two constructs were generated for α-amylase gene of Bacillus subtilis MTCC 121 (Amy_F, full-length and Amy_T, C-terminal truncated) were cloned, overexpressed, purified, and characterized. Results revealed that activity of Amy_T was found to be 2.87-fold better than Amy_F. Further, the optimum temperature of Amy_F and Amy_T was obtained at 45?°C and 55?°C, respectively, whereas optimum pH was recorded at pH 7 and pH 8, respectively. Improved thermostability of Amy_T was further confirmed through thermal shift assay. Subsequently, starch-coated fabrics were tested for starch removal using the α-amylases. Comparative analysis revealed that Amy_T performed better in starch removal from polystyrene (85%), silk (75%), and cotton (70%) fabrics. The removal of starch from the fabrics was further confirmed by FESEM. Conclusively, this work presents one truncated α-amylase as an improved candidate over its full-length counterpart for textile desizing.  相似文献

Diarrhea is one of the main causes of children's death. Due to the importance of health in children under 5 years, this study has been conducted to determine the influence of the use of improved sanitation facilities (UISF) and the use of improved drinking-water sources (UIDWS) on the diarrhea-associated deaths per 1000 live births in this age group. An ecologic study was conducted in 75 countries, and the data on UIDWS and UISF information have been collected by World Health Organization (WHO) under a program named “Global Analysis and Assessment of Sanitation and Drinking-Water” (GLAAS). To determine the correlation between UIDWS and UISF with the diarrhea-associated deaths in children, Pearson correlation was applied. Besides, a linear regression analysis was used to investigate the relationship between UIDWS and UISF with the diarrhea-associated deaths in children. According to the findings, there was a significant statistical relationship between UIDWS and UISF with the diarrhea-associated deaths in children (p < 0.001). The diarrhea-associated deaths in children under 5 years are influenced by factors such as UIDWS and UISF, and this rate can be reduced making UISF and UIDWS available for people and especially people of rural areas.  相似文献

We previously described a series of 314-helical β-peptides that bind the hDM2 protein and inhibit its interaction with a p53-derived peptide in vitro. Here we present a detailed characterization of the interaction of these peptides with hDM2 and report two new β-peptides in which non-natural side chains have been substituted into the hDM2-recognition epitope. These peptides feature both improved affinity and inhibitory potency in fluorescence polarization and ELISA assays. Additionally, one of the new β-peptides also binds the hDM2-related protein, hDMX, which has been identified as another key therapeutic target for activation of the p53 pathway in tumors.  相似文献

Summary This paper describes a flow-injection analysis (FIA) system for measuring -amylase in cultivation media. It is based on measuring the decolorization of an iodine-starch complex. The FIA-system is optimized with respect to stability and reproducibility. β-Glucosidase (βG) can relieve the product inhibition of cellobiose in the cellulosic ethanol production by converting cellobiose into glucose. For the potential recycled uses, βG was immobilized and stabilized in the form of enzyme coating on polymer nanofibers. The βG coating (EC-βG) was fabricated by crosslinking additional βG molecules onto covalently attached βG molecules (CA-βG) via glutaraldehyde treatment. The initial activity of EC-βG was 36 times higher than that of CA-βG. After 20 days of incubation under shaking, CA-βG and EC-βG retained 33 and 91% of each initial activity, respectively. Magnetic nanofibers were also used for easy recovery and recycled uses of βG coating. It is anticipated that the recycled uses of highly active and stable βG coating can improve the economics of cellulosic ethanol production so long as economical materials are employed as a host of enzyme immobilization.  相似文献

β-Galactosidases (EC 3.2.1.23) constitute a large family of proteins that are known to catalyze both hydrolytic and transgalactosylation reactions. The hydrolytic activity has been applied in the food industry for decades for reducing the lactose content in milk, while the transgalactosylation activity has been used to synthesize galacto-oligosaccharides and galactose containing chemicals in recent years. The main focus of this review is on the expression and production of Aspergillus niger, Kluyveromyces lactis and bacterial β-galactosidases in different microbial hosts. Furthermore, emphasis is given on the reported applications of the recombinant enzymes. Current developments on novel β-galactosidases, derived from newly identified microbial sources or by protein engineering means, together with the use of efficient recombinant microbial production systems are converting this enzyme into a relevant synthetic tool. Thermostable β-galactosidases (cold-adapted or thermophilic) in addition to the growing market for functional foods will likely redouble its industrial interest.  相似文献

Monitoring therapeutic efficacy of antimalarial drugs is important because treatment failure rates are the primary basis for changing antimalarial treatment policy. An important aspect of efficacy studies is the use of PCR genotyping to distinguish recrudescent from new infections. The conclusions reached using this technique might be misleading if there is insufficient parasite diversity or a non-uniform haplotype frequency distribution in the study area. Statistical techniques can be used to overcome this problem, but only when data describing the haplotype frequency distribution are available. Therefore, assessing haplotype frequency and distribution should form an integral part of all studies investigating the therapeutic efficacy of antimalarial treatment regimes.  相似文献

Here, we report the synthesis and in depth characterization of a second generation β-lactone derived virulence inhibitors. Based on initial results that emphasized the intriguing possibility to disarm bacteria in their virulence the present study develops this concept further and analyses the potential of this strategy for drug development. We were able to expand the collection of bioactive compounds via an efficient synthetic route. Testing of all compounds revealed several hits with anti-virulence activity. Moreover, we demonstrated that these molecules act solely by reducing virulence but not killing bacteria which is an important prerequisite for preserving the useful microbiome. Finally, incubation of lactones with eukaryotic cell lines indicated a tolerable cytotoxicity which is essential for entering animal studies.  相似文献

Background

Clinical trials show that asthma can be controlled in the majority of patients, but poorly controlled asthma still imposes a considerable burden. The level of asthma control achieved reflects the behaviour of both healthcare professionals and patients. A key challenge for healthcare professionals is to help patients to engage in self-management behaviours with optimal adherence to appropriate treatment. These issues are particularly relevant in primary care, where most asthma is managed. An international panel of experts invited by the International Primary Care Respiratory Group considered the evidence and discussed the implications for primary care practice.

Discussion

Causes of poor control Clinical factors such as exposure to triggers and concomitant rhinitis are important but so are patient behavioural factors. Behaviours such as smoking and nonadherence may reduce the efficacy of treatment and patients' perceptions influence these behaviours. Perceptual barriers to adherence include doubting the need for treatment when symptoms are absent and concerns about potential adverse effects. Under-treatment may also be related to patients' underestimation of the significance of symptoms, and lack of awareness of achievable control. Implications Three key implications for healthcare professionals emerged from the debate. First, the need for simple tools to assess asthma control. Two approaches considered were the monitoring of biometric markers of control and questionnaires to record patient-reported outcomes. Second, to understand the reasons for poor control for individual patients, identifying both clinical (e.g. rhinitis) and behavioural factors (e.g. smoking and nonadherence to treatment). Third was the need to incorporate, within asthma review, an assessment of patient perspectives including their goals and aspirations and to elicit their beliefs and concerns about asthma and its treatment. This can be used as a basis for agreement between the healthcare professional and patient on a predefined target regarding asthma control and a treatment plan to achieve this.

Summary

Optimum review of asthma is essential to improve control. A key priority is the development of simple and effective tools for identifying poor control for individual patients coupled with a tailored approach to treatment to enable patients to set and achieve realistic goals for asthma control.  相似文献
