Summary Neuronal circuits in the brain and retrocerebral complex of the cockroach Diploptera punctata have been mapped immunocytochemically with antisera directed against the extended enkephalin, Met-enkephalin-Arg6-Gly7-Leu8 (Met-8). The pathways link median and lateral neurosecretory cells with the corpus cardiacum/corpus allatum complex. In females, nerve fibres penetrate the corpora allata and varicosities or terminals, immunoreactive to Met-8, surround the glandular cells. Males differ in having almost no Met-8 immunoreactivity in the corpora allata. The corpora cardiaca of both males and females are richly supplied with Met-8 immunoreactive material, in particular in the cap regions immediately adjacent to the corpora allata. A similarity in the amino-acid sequences of Met-8 and the C-terminus of the recently characterised allatostatins of D. punctata suggests that the pathways identified with the Met-8 antisera may be the same as those by which the allatostatins are transported from the brain to the corpus allatum. In comparative studies on the blowfly Calliphora vomitoria, similar neuronal pathways have been identified except that no sexual dimophism with respect to amounts of immunoreactive material within the corpus allatum has been observed. These results suggest a possible homology in the neuropeptide regulation of the gland.  相似文献

Summary A monoclonal antibody against allatostatin I was used to demonstrate the allatostatin-immunoreactive pathways between the brain and the corpus cardiacum-corpus allatum complex in the adult cockroach Diploptera punctata. The antibody was two to three orders of magnitude more sensitive to allatostatin I than to the other four known members of the allatostatin family. Whole and sectioned brains in which immunoreactivity was localized with horseradish peroxidase-H2O2-diaminobenzidine reaction showed strongly immunoreactive cells in the pars lateralis of the brain with axons leading to and arborizing in the corpus cardiacum and the corpus allatum. Although many neurosecretory cells of the pars intercerebralis project to the corpora allata only, four strongly immunoreactive cells were evident here (two pairs on either side), and these did not project to the corpus cardiacum and corpus allatum but rather terminated within the protocerebrum in areas in which lateral cells also formed arborizations. Immunoreactivity was found in many other cells in the brain, especially in the tritocerebrum.  相似文献

The neurosecretory system and retrocerebral endocrine glands of Nezara viridula Linn. have been described on the basis of in situ preparations and histological sections employing the paraldehyde fuchsin (PF) and performic acid-victoria blue (PAVB) techniques. In the brain of N. viridula, there are two medial groups–each consisting of five neurosecretory cells which belong to A-type. The lateral neurosecretory cells are absent. The axons of the two groups of medial neurosecretory cells (MNC) compose the two bundles of neurosecretory pathways (NSP) that decussate in the anterodorsal part of the protocerebrum. The two pathways, after the cross-over, run deep into the protocerebrum and deutocerebrum and emerge as NCC-I from the tritocerebrum. The nervi corporis cardiaci-I (NCC-I) of each side which are heavily loaded with NSM terminate in the aorta wall. Thus, the neurosecretory material (NSM), elaborated in the medial neurosecretory cells of the brain, is stored in the aortic wall and nervi corporis cardiaci-I (NCC-I). The NCC-II are very short nerves that originate from the tritocerebrum and terminate in the corpora cardiaca (CC) of their side. Below the aorta, but dorsal to the oesophagus, lie two oval or spherical corpora cardiaca. A corpus allatum (CA) lies posterior to the corpora cardiaca (CC). The corpora cardiaca do not contain NSM; only the intrinsic secretion of their cells has been occasionally observed which stains orange or green with PF staining method. The corpus allatum sometimes exhibits PF positive granules of cerebral origin. A new connection between the corpus allatum and aorta has been recorded. The suboesophageal ganglion contains two neurosecretory cells of A-type which, in structure and staining behaviour, are similar to the medial neurosecretory cells of the brain. The course and termination of axons of suboesophageal ganglion neurosecretory cells, and the storage organ for the secretion of these cells have been reported. It is suggested that the aortic wall and NCC-I axons function as neurohaemal organ for cerebral and suboesophageal secretions.  相似文献

Summary

Corpora cardiaca of Locusta migratoria, contain the axon endings of the neurosecretory cells of the brain and store in neurosecretory granules a variety of mostly unidentified neurohormones. Homogenates of corpora cardiaca served to generate a battery of monoclonal antibodies screened by their immunoreactivity to antigenic determinants present in the neurosecretory cells of the pars intercerebralis in the brain. The results are illustrated with three selected monoclonal antibodies which recognize antigens located within the neurosecretory granules of the pericarya of the pars intercerebralis, the cerebro-cardiac axon tracts and the axonic endings in the neurohaemal part of the corpora cardiaca. The apparent molecular weights of these antigenes were determined by Western blotting. We discuss the potential of these monoclonal antibodies for the isolation and structure determination of neuropeptides.  相似文献

Summary Insulin, glucagon and adipokinetic hormone antisera were applied to the corpora cardiaca, perisympathetic organs, neurohemal areas and peripheral neurosecretory cells of three insect species, the locust Locusta migratoria, the cockroach Periplaneta americana, and the stick insect Carausius morosus. The neurohemal part of the corpora cardiaca was shown to be immunoreactive to both insulin and glucagon antisera while the glandular cells reacted to adipokinetic hormone antisera. The perisympathetic organs seem to be devoid of these three substances, but certain peripheral neurohemal areas contained AKH and glucagon immunoreactive products. The latter were found to originate in the peripheral neurosecretory cells.  相似文献

The neurosecretory system of the earwig, Euborellia annulipes, contained material similar to that of FMRFamide, as shown by immunocytochemistry. Within the brain were two pairs of darkly staining perikarya in the medial protocerebrum, and up to four pairs of immunoreactive cells in the lateral protocerebrum. The corpora allata appeared immunoreactive in 10-day females, but not in 2-day-old adults. Additionally, immunoreactive material was detected in midgut endocrine cells of both 2- and 10-day-old females. FMRFamide at 1 to 100 nM did not inhibit juvenile hormone production by earwig corpora allata in vitro. This was true of glands of low activity from 2-day cat food-fed or starved virgin females, 10-day starved females, and those of relatively high activity from 10-day-old, cat food-fed females. In contrast, FMRFamide at 50 and 100 (but not at 1) nM stimulated gut motility in vitro in distended guts from 2-day fed females. Preparations from starved females and those from 10-day fed females (in which feeding behavior is on the decline) did not respond to exogenous FMRFamide with enhanced rates of contraction. Lastly, preparations from females starved for 7 days and subsequently fed for 3 days responded to 10 nM FMRFamide with increases in gut motility.  相似文献

Immunoreactivity against peptides of the allatostatin family having a typical YXFGL-NH2 C-terminus has been localized in different areas of the central nervous system, stomatogastric nervous system and gut of the cockroach Blattella germanica. In the protocerebrum, the most characteristic immunoreactive perikarya are situated in the lateral and median neurosecretory cell groups. Immunoreactive median neurosecretory cells send their axons around the circumesophageal connectives to form arborizations in the anterior neuropil of the tritocerebrum. A group of cells in the lateral aspect of the tritocerebrum project to the antennal lobes in the deutocerebrum, where immunoreactive arborizations can be seen in the periphery of individual glomeruli. Nerve terminals were shown in the corpora allata. These terminals come from perikarya situated in the lateral neurosecretory cells in the pars lateralis and in the subesophageal ganglion. Immunoreactive axons from median neurosecretory cells and from cells positioned in the anteriormost part of the tritocerebrum enter together in the stomatogastric nervous system and innervate foregut and midgut, especially the crop and the valve between the crop and the midgut. The hindgut is innervated by neurons whose perikarya are located in the last abdominal ganglion. Besides immunoreactivity in neurons, allatostatin-immunoreactive material is present in endocrine cells distributed within the whole midgut epithelium. The distribution of FMRFamide immunoreactivity in the brain-retrocerebral complex of adult female Diploptera punctata was examined. Immunoreactivity was observed in the brain and corpus allatum as well as in the corpus cardiacum. Immunoreactivity co-localized with allatostatin immunoreactivity within several lateral neurosecretory cells of the brain and in their endings within the corpus allatum. By in vitro radiochemical assay of juvenile hormone release, the effect of two native D. punctata RFamides, an FLRFamide (Leucomyosuppressin) and an FIRFamide were examined. The latter, for which the sequence (SKPANFIRFamide) is reported here, stimulated juvenile hormone release but acted only on corpora allata from females at the end of vitellogenesis (day 6). The interaction of these two RFamides and three D. punctata allatostatins, Dippu-AST 2, 5, and 7 were similarly examined. Only Dippu-AST 2 stimulated release of RFamides from the corpora allata and only on day 6 whereas both RFamides were able to attenuate the inhibitory activity of Dippu-AST 2.  相似文献

The neurosecretory system of Labedura riparia has been described from sections and whole mounts using a variety of techniques. The pars intercerebralis contains two clusters of medial neurosecretory cells (MNC), each cluster consisting of 8 to 10 A-cells and occasional B-cells. The lateral sides of the brain have a few B-cells. The axons of the median neurosecretory cells terminate in the cephalic aorta (AO), whereas the axons of the lateral neurosecretory cells (LNC) terminate in the corpora cardiaca (CC). It appears that the neurosecretory material (NSM) elaborated in the MNC is stored in the cephalic aorta and that elaborated in the LNC is stored in the corpora cardiaca, which are two oval or elongate bodies composed of large chromophobe and small chromophil cells. Posteriorly there is the oval or elongate corpus allatum (CA) attached to the CC by thick nerves. The CA consists of one cell type only. Both CC and CA contain no A-cell neurosecretory material. It has been suggested that the neurosecretory system of L. riparia is composed of two complexes. One is formed by the medial neurosecretory cells for which the aorta functions as a neurohaemal organ, and the other is formed by lateral neurosecretory cells-lateral neurosecretory pathways-nervi corporis cardiaci-II in which the corpora cardiaca function as a neurohaemal organ.  相似文献

The ultrastructure of the retrocerebral endocrine-aortal complex of the earwig, Euborellia annulipes has been studied. The space between the inner and outer stromal layers of the aorta is occupied by numerous axon terminals and pre-terminals containing large electron dense granules (NS-I) of approximately 100 to 220 nm and a few axon terminals having small granules (NS-II) of approximately 40 to 90 nm; the former appear to belong to medial neurosecretory A-cells, and the latter to the B-cells of the brain. The corpora cardiaca consist of intrinsic cells with mitochondria and multivesicular bodies. Granules of type NS-II and NS-III are observed in the axon terminals and pre-terminals in the corpora cardiaca. The NS-II are identical to those found in the aorta and are probably the secretions of the lateral B-cells. Granules of type NS-III are 40 to 120 nm and electron dense, and are intrinsic in origin. Similar granules occur in the intrinsic cells of the corpora cardiaca. E M studies have confirmed the rôle of the aorta as a neurohaemal organ for the medial neurosecretory cells, and the corpora cardiaca for the lateral neurosecretory cells of the brain. The corpora cardiaca also act as a reservoir for the intrinsic secretion. The corpus allatum is a solid body consisting of parenchymal cells with prominent nuclei, mitochondria, and endoplasmic reticulum. In between its cells are occasional glial cells and also neurosecretory as well as non-neurosecretory axons. The gland is devoid of A-cell NSM.  相似文献

Summary In the brain of the adult worker bee (Apis mellifica) prolactin-like (PRL) immunoreactive cells were localized in the lateral neurosecretory cell region and the subesophageal ganglion by means of the PAP procedure. These cells emit nerve fibres which pass through the neuropile of the brain to the corpora cardiaca where a great number of immunoreactive axon terminals is present. Tests with antisera against rat pituitary prolactin and human luteinizing hormone were negative. These results indicate that hPRL material is produced in neurosecretory cells of the bee brain and transferred via axons to the corpora cardiaca for storage and subsequent release into haemolymph.This work is part of the Ph. Brain-retrocerebral complexes of female crickets,Gryllus bimaculatus andAcheta domesticus, treated with antibody to allatostatin-1 from a cockroach,Diploptera punctata, show extensive immunoreactivity. The results suggest that allatostatins or allatostatin-like molecules are produced in neurosecretory cells of the brain and are delivered to the corpora allata through nervous connections and/or via haemolymph. Radiochemical measurements of juvenile hormone III biosynthesis by isolated corpora cardiaca-corpora allata complexes from adultG. bimaculatus have been used to demonstrate an in vitro sensitivity of these glands to allatostatin-1 fromD. punctata. Allatostatin-1 is a relatively potent inhibitor of juvenile hormone III biosynthesis in corpora allata of both young adult females and males. In glands taken from 3-day virgin females, 50% inhibition of hormone biosynthesis is reached at ca. 3 nmol·l-1 allatostatin-1. The inhibitory action of allatostatin-1 is rapid, dose-dependent and reversible. Addition of 200 mol·l-1 farnesol to the incubation medium prevents inhibition of juvenile hormone III biosynthesis by allatostatin-1. The implantation of active corpora allata into intact Locusta females during growth accelerates pre-vitellogenic oöcyte growth and vitellogenesis. Localised stimulation of yolk deposition follows the implantation of active corpora allata between the ovarioles demonstrating a gonadotrophic rôle for the corpus allatum hormone. Electrocoagulation of the median neurosecretory cells of the brain prevents vitellogenesis whilst pre-vitellogenic oöcyte growth occurs normally. Implantation of active corpora allata into females with ablated cerebral neurosecretory cells promotes vitellogenesis in a proportion of test animals although mature oöcytes are never produced.It is suggested that the rôle of the median neurosecretory cells during egg development in Locusta is primarily concerned with the activation and maintenance of activity of the corpora allata. The corpus allatum hormone acts both metabolically and gonadotrophically.  相似文献

In the bean bug, Riptortus clavatus, allatectomy suppressed reproduction in adults reared under nondiapause-inducing long-day conditions, and transection of the nervi corporis allati induced reproduction in adults reared under diapause-inducing short-day conditions. These effects of allatectomy and denervation were observed both in the morphology of reproductive organs and in the electrophoresis pattern of hemolymph proteins in both sexes. These results indicate that, in diapause adults, the brain suppresses the activity of the corpus allatum to secrete juvenile hormone through nervous pathways. The removal of the corpora cardiaca–corpus allatum complex in females not only inhibited ovarian development, as allatectomy did, but also prevented mature eggs in the oviduct from being laid. Although the mealworm Tenebrio molitor inhabits very dry environments, it has at least two diuretic peptides, which increase fluid secretion by the free portions of the Malpighian tubules. Unlike other insect corticotropin-releasing factor (CRF)-related peptides isolated to date, these are non-amidated peptides. The immunocytochemical localization of Tenmo-DH(37) was investigated using antisera raised against this hormone. Immunoreactive neurosecretory cells were found in the brain and abdominal ganglia with immunoreactive processes projecting to the peripheral nervous system. Intense staining of the neurohaemal release site, the corpora cardiaca, was observed. In addition, neurosecretory cells immunoreactive to Tenmo-DH(37) were found in the posterior midgut and a network of immunoreactive nerve processes extended over the surface of the midgut. Tenmo-DH(37) is widely distributed and its staining pattern resembles that found for other, amidated CRF-related diuretic peptides.  相似文献

Summary Separate antisera were raised to the N- and C-terminal half of the diuretic hormone from Manduca sexta. Antisera against the two halves of this peptide recognized the same cells in M. sexta, and preabsorption of the antisera with the peptides used as antigens abolished the immunoreactivity, confirming their specificity. The antisera reacted with two median neurosecretory cells on each side of the protocerebral groove in larvae, and with a group of about 80 small median neurosecretory cells in the adult, as well as their axons to, and their axon terminals in, the corpora cardiaca. During the early pupal stages, small cells, which are possibly derived from a common neuroblast, differentiate into immunoreactive neurosecretory cells, which explains the large increase in cell numbers in the adult. In the sleepy sulphur butterfly, Eurema nicippe, homologous median neurosecretory cells in the adult were immunoreactive with both antisera.  相似文献

The corpora cardiaca of Schistocerca consist of a neurohaemal part containing mainly extrinsic neurosecretory axons coming from the brain, and of a glandular part consisting mainly of intrinsic neurosecretory cells. Some extrinsic axons penetrate into the glandular region, and innervate intrinsic corpus cardiacum neurosecretory cells. The fine structure of the latter has been examined and related to other neurosecretory cell types. Secretion occurs by exocytosis. Omega-profiles are more frequently observed in corpora cardiaca stimulated electrically or by acetylcholine than in the controls.  相似文献

Aqueous extracts of brain, thoracic ganglion or corpora cardiaca of female Glossina morsitans were shown to contain a substance which inhibited the synthesis of lipid from l[U-14C] leucine by fat cells incubated in vitro. The highest concentration of this substance was found in the corpora cardiaca; approximately 1 × 10?6 gland pairs μl?1 were required for maximum inhibition. At concentrations greater than 1 × 10?4 gland pairs μl?1 the lipid synthesis inhibiting factor (hereafter referred to as the LSIF) was inactivated by the presence of a substance which could be removed by gel filtration. The concentration of LSIF in the corpora cardiaca and midbrain varied throughout the reproductive cycle of the female. Net release of LSIF from the midbrain occurred between the 2nd and 7th day of the 9-day reproductive cycle. Net release from the corpora cardiaca began on day 5 and continued until the end of the interlarval period on day 9. Results are consistent with the hypothesis that LSIF is synthesised mainly in the medial neurosecretory cells of the midbrain whereas the corpora cardiaca are the site of storage and release into the haemolymph. LSIF was present in midbrain and corpora cardiaca extracts from male G. morsitans but at lower concentrations than in females. No variation in LSIF concentration could be correlated with the feeding cycle. LSIF activity was not detected in fresh haemolymph but was found at high concentration in boiled haemolymph, suggesting the presence of an inhibitor which was inactivated at high temperature. Preliminary investigations into the nature of LSIF have shown it to be inactivated by proteolytic enzymes and to be recoverable in a single peak from a Sephadex G15 column.Results support the view that LSIF is a peptide hormone which, in conjunction with an inhibitor, controls the lipid synthetic ability of the fat cells of the adult female tsetse fly throughout the reproductive cycle.  相似文献

This study also shows sites of release of stored extrinsic neurosecretory material and of products elaborated by intrinsic glandular cells of the corpora cardiaca.

The brain of Glossina morsitans Westwood contains four groups of neurosecretory cells which are stainable with chrome haematozylin and phloxin. The axons of these cells form a pair of nervi corporis cardiaci which pass posteriorly from the brain and innervate the corpora cardiaca and corpus allatum before uniting with a small ganglion posterior to the corpora cardiaca. This ganglion is considered to represent the fusion of the fusion of the hypocerebral and ventricular ganglia which remain separate in other insects.
There is no frontal ganglion in the adult Glossina and the recurrent nerve fuses with one of the nervi corporis cardiaci immediately behind the brain. The oesophageal nerves arising from the fused hypocerebral and ventricular ganglia innervate the oesophagus in the anterior part of the thorax, the proventriculus and the posterior extension of the oesophagus close to the crop. These nerves possess both sensory and motor nerve endings. The differences which exist between Glossina and other cyclorrhaphous Diptera with respect to their neuroendocrine/stomatogastric system are noted and considered in terms of the control of neuroendocrine function.  相似文献
