Given the ecological and commercial importance of the summer flounder (Paralichthys dentatus), there is a surprising paucity of information on the molecular genetics of this species. Some studies published to date
are concentrated on the reproduction biology. To address this shortcoming, a microsatellite-enriched genomic DNA library of
P. dentatus was generated and screened by sequencing. Twelve dinucleotide microsatellite loci were characterized by genotyping 24 samples.
The observed number of alleles ranged from three to thirteen with an average of 8.25, while the effective number of alleles
ranged from 2.21 to 8.28 with an average of 5.06. The observed and expected heterozygosities ranged from 0.0833 to 0.9583
and from 0.5594 to 0.8980, respectively. Significant deviations from Hardy–Weinberg expectations were detected at three loci
and linkage disequilibrium between two loci was significant after applying Bonferroni correction. In cross-species amplification,
three species showed at least two polymorphic loci. The 12 highly polymorphic microsatellite markers represent a powerful
molecular tool, which will allow for detailed population genetic analyses on this important marine fish. 相似文献

We developed a set of eight polymorphic microsatellite loci for pronghorn, Antilocapra americana. We screened 233 individuals from the National Bison Range in Moiese, MT, and found allele numbers from three to 11 and heterozygosity levels ranging from 0.142 to 0.807. These results suggest that these loci will be useful in paternity analysis and basic population genetics applications. 相似文献

An enrichment protocol was used to isolate and characterize microsatellite loci in Acer opalus, a Mediterranean tree species. Highly polymorphic microsatellite loci were required for paternity analyses in a population
of this species. Eight microsatellite loci were amplified and a total of 87 alleles were detected in a sample of 142 individuals
from one population, allowing the identification of each individual with a unique multilocus genotype. The paternity exclusion
probabilities varied from 0.261 for locus Aop820 to 0.806 for locus Aop450, and the parent-pair exclusion probabilities varied
from 0.433 for Aop820 to 0.940 for Aop450. The cumulative probabilities of exclusion for paternity and parentage of the eight
loci were both higher than 0.999, supporting the usefulness of these microsatellite loci for future paternity and parentage
analyses in A. opalus. Cross-species transferability was also assayed, supporting their potential use in other eight Acer species. 相似文献

We describe the first microsatellites for rockfishes in the diverse genus Sebastes. Clones containing microsatellites were isolated from the genomic library of a quillback rockfish, Sebastes maliger. Twelve microsatellites are characterized; six of these are polymorphic in quillback rockfish, and eight are polymorphic in
at least one rockfish species on which they were tested. The number of alleles per variable locus ranged from 4 to 15 and
averaged 6.8. The expected heterozygosities ranged from 0.38 to 0.79 and averaged 0.60 in these loci. These loci should prove
valuable in studies examining species identification, population genetics, hybridization, paternity, kinship, and microsatellite
evolution.

To obtain polymorphic molecular markers for population genetics and conservation studies in Bonelli's eagle populations, we screened a partial genomic library enriched for microsatellites with di‐ and tri‐nucleotide motifs [(GT) (CT) (AAC) and (GCC)]. A total of 15 polymorphic markers were obtained. The number of alleles ranged from two to eight. These markers will be very useful for paternity tests and population structure studies as well as for evaluating the outcome of conservation programs. 相似文献

Eight novel polymorphic microsatellite loci developed from a microsatellite enriched genomic library, are presented for the
Chinese pond turtle (Chinemys reevesii). We screened 30 individuals from three populations and detected high levels of polymorphism for all eight loci with the
number of alleles/locus ranging from 7 to 17 (average = 10.88). The values of expected and observed heterozygosities ranged
from 0.703 to 0.920 and 0.321 to 0.966, respectively. These highly variable loci will provide a powerful molecular toolkit

We report on the isolation and characterization of 11 microsatellite loci in the Sichuan snub-nosed monkey (Rhinopithecus roxellana) from AC-enriched genomic libraries. 20 microsatellite loci were screened from the libraries, of which 11 were polymorphic.
The number of observed alleles among 32 samples of snub-nosed monkey ranged from three to nine. The observed and expected
heterozygosity were 0.071–0.815 and 0.201–0.819, respectively. The high variability revealed in this study should be useful
tools for further study on social structure and population genetics of snub-nosed monkey in the future. 相似文献

Premise of the study: Microsatellite loci were isolated and developed as polymorphic markers for the New Zealand endemic root holoparasite Dactylanthus taylorii for use in population and conservation genetics studies. ? Methods and Results: Shotgun 454 pyrosequencing was performed on genomic DNA pooled from three individuals of D. taylorii. From 61709 individual sequence reads, primers for 753 microsatellite loci were developed in silico and 72 of these were tested for consistent amplification and variability. Ten microsatellite loci were found to be polymorphic and consistently scorable when screened in 44 individuals from five geographically distant populations. The number of alleles per locus ranged from four to 16 with an average of 9.7, and average observed heterozygosity per locus was between 0.182 and 0.634. ? Conclusions: These polymorphic microsatellite markers establish an important resource for ongoing conservation initiatives and planned population genetic studies of D. taylorii. 相似文献

Given the low substitution rate in plastomes, the polymorphic and codominant nature of chloroplast SSRs (cpSSRs) makes them ideal markers, complementing their nuclear counterpart. In Cupressaceae, cpSSRs are mostly paternally inherited, thus, they are useful in mating systems and pollen flow studies. Using e‐PCR, 92 SSR loci were identified across six Cupressaceae plastomes, and primers were designed for 26 loci with potential interspecific transferability. The 26 developed cpSSRs were polymorphic in four genera, Platycladus, Sabina, Juniperus, and Cupressus and are suitable for Cupressaceae molecular genetic studies and utilization. We genotyped 192 Platycladus orientalis samples from a core breeding population using 10 of the developed cpSSRs and 10 nuclear SSRs, and these individuals were identified with high confidence. The developed cpSSRs can be used in (1) a marker‐assisted breeding scheme, specifically when paternity identification is required, (2) population genetics investigations, and (3) biogeography of Cupressaceae and unraveling the genetic relationships between related species. 相似文献

We isolated seven novel polymorphic microsatellite DNA loci from the burrowing parrot (Cyanoliseus patagonus) and optimised them for future studies of population differentiation and genetic variation. The loci were screened for polymorphism
using 38 samples from wild individuals from three neighbouring colonies in Argentina. The primers amplified highly variable
loci characterised by 3–10 alleles per locus and their observed and expected heterozygosities ranged from 0.15 to 0.78 and
0.15 to 0.81, respectively. When we analysed 52 samples across Argentina and Chile, we found strong genetic differentiation
between the Chilean and the Argentinean subspecies as well as significant differentiation between two geographically separated
subspecies within Argentina. Our results indicate the suitability of these microsatellites for investigating further questions

Paternity exclusion studies provide useful information for testing certain theories of behavioral ecology and for the management
and conservation of both wild and captive populations of endangered species. This study used eight human nuclear microsatellite
loci, in the absence of species-specific PCR primers, to genetically identify the sires of 12 captive lowland gorillas (Gorilla gorilla gorilla) and 2 captive orangutans (Pongo pygmaeus pygmaeus andPongo p. abelii). Parentage assignments were confirmed by excluding all except a single potential sire for each offspring with the least
two loci. Sire-offspring relationships were verified in 12 of the 14 cases, and reassigned in the case of two gorilla offspring.
The orangutan paternity typing was supplemented by DNA fingerprinting. Additionally, five of the eight microsatellite loci,
in conjunction with behavioral data, were used for a non-exhaustive set of paternity exclusions for five wild mountain gorillas
(Gorilla g. beringei). The eight loci described in this study should be useful additions to the tools available for the study of genetics in the

Polymorphic di‐, tri‐ and tetranucleotide repeats were examined in Allonemobius to determine whether they could serve as useful markers in studies of sperm precedence, population genetics and hybrid zone structure. Ten microsatellite DNA loci were sufficiently polymorphic to be used for paternity tests and showed no evidence of linkage disequilibrium or deviations from Hardy–Weinberg equilibrium in Allonemobius socius. Nine of 10 of these microsatellites can be amplified from three other Allonemobius species, suggesting that these markers will have widespread utility in this ground cricket genus. 相似文献

We isolated microsatellite loci for the Trinidadian guppy Poecilia reticulata from the Oropuche and Caroni drainages in Trinidad, using a modified enrichment protocol that provided 93% enriched libraries. The eight loci presented here are polymorphic, and have between seven and 18 alleles in 33–48 individuals initially screened. Observed heterozygosities in this sample range from 0.28 to 0.88. These loci significantly increase the number of polymorphic microsatellite markers available for paternity and population analysis in this species. 相似文献

Practically no studies on the population genetics of the spinner dolphin (Stenella longirostris) exist. Seventeen pairs of DNA primers, cloned from an Mbo I digestion of S. longirostris liver DNA, were selected from a total of 288 sequences. Eight polymorphic microsatellite DNA markers were selected from the
17 primer pairs following amplification of DNA from skin samples of 65 spinner dolphins. Characterization of the polymorphisms
revealed between three and nine alleles per loci. The observed heterozygosity ranged from 0 to 0.6032, while the expected
heterozygosity ranged from 0.5834 to 0.73. Seven of the eight designed primer pairs amplified DNA from three other delphinid
species. There was a marked low observed heterozygosity in the spinner dolphin suggesting a high level of inbreeding within

Microsatellites were isolated from the damselfish Parma microlepis (Günther 1862) (Pomacentridae) and screened for 100 individuals. Seven of the eight loci tested were highly polymorphic, having 14–43 alleles with average heterozygosities between 0.86 and 0.97. Six new microsatellite DNA loci are isolated from a genomic library of the sphenodontid reptile tuatara (Sphenodon) and presented here as a tool for identifying individuals for future paternity and kinship studies. These loci, in combination
with four previously published loci, are sufficient to discriminate between clutch-mate siblings from Stephens Island and
Brothers Island populations. These populations represent high and low levels of genetic diversity in tuatara populations respectively.
An estimate of minimum number of fathers of each clutch found no evidence for multiple paternity in any clutch. These newly
isolated loci complete the development of an array of genetic tools for use in tuatara to enhance ongoing conservation and

Twenty-two nuclear microsatellite loci were isolated from a genomic DNA library derived from Madagascar’s Rousettus madagascariensis. Marker characteristics were determined from a single population (37 individuals) from Fort Dauphin (southeastern Madagascar).
Sixteen of the 22 loci were within Hardy–Weinberg expectations. These loci are highly informative with polymorphic information
content values ranging between 0.757 and 0.916. These loci will provide valuable information for the study of population genetics
and gene flow within this species of bats. Due to the dramatic reduction and alteration of their habitat, data generated utilizing
this marker suite will potentially provide additional information for the effective long-term management of this near-threatened
