Abstract

Genetic variation at 33 loci in three species of Leiopelma was examined by cellulose acetate electrophoresis of liver enzymes. Heterozygosity is apparently comparable to levels detected in other amphibians. Genetic differentiation between species is extensive. L. hochstetteri is greatly divergent from both L. archeyi and L. hamiltoni, but these two are relatively similar genetically, in agreement with morphological, ecological, and developmental patterns. We tentatively estimate that the divergence of hochstetteri from the other two species occurred during the Miocene, whereas that of archeyi and hamiltoni occurred during the Pliocene. Implications of the genetic data for conservation of these rare species are discussed.  相似文献

Abstract

Albumin evolution is examined within the anuran genus Leiopelma using the quantitative immunological technique of micro-complement fixation (MC'F). The albumins of L. archeyi and L. hamiltoni are relatively similar to each other, yet both differ greatly from the albumin of L. hochstetteri. Using albumin as a molecular clock, we estimate that L. hochstetteri diverged from the lineage leading to L. archeyi and L. hamiltoni during the Miocene, about 15 million years ago. The divergence of L. archeyi and L. hamiltoni appears to have been a Pliocene event (about 3 million years ago).  相似文献

Endemic New Zealand frogs of the genus Leiopelma are from a basal lineage of extant anurans that release defensive secretions onto their skin when disturbed. Here, we characterize the gross anatomy and microscopic structure of the skin of L. archeyi, L. hochstetteri, and L. pakeka using stereoscopic, light and transmission electron microscopy. The terrestrial L. archeyi and L. pakeka possess dimorphic granular glands, categorized as type I and II, based on their frequency and morphological traits, whereas the semi‐aquatic L. hochstetteri lacks type I glands. This is the first report of differential dimorphism in anurans of the same genus. This dimorphism could be interpreted as an adaptation to different physiological or ecological needs of these species. However, species within this ancient genus share similar general gland morphology with other anurans, namely, a secretory unit containing storage granules ensheathed by myoepithelial cells. Type I glands are ellipsoid, large and contain a homogeneous mass of electron‐dense granules (1.8 ± 0.08 μm in diameter). Type II glands are round and contain larger heterogeneous granules (4.06 ± 0.16 μm) of varying densities. Exposure to noradrenaline causes the contraction of myoepithelial cells, resulting in bulk discharge of type I glands through the epidermal duct onto the skin surface. Differential release of secretions from dimorphic glands may be indicative of their functional specialisation in antipredatory or regulative roles. Mass spectrometric techniques were used to de novo sequence peptides present in the skin secretions of Leiopelma species. A total of 30 previously undescribed peptides from Leiopelma species were fully or partially sequenced. These peptides exhibited no similarity to any known compounds. J. Abstract

A new population of terrestrial Leiopel‐matid frog was discovered in the Whareorino Forest, northern King Country, New Zealand, in 1991. Searches were carried out from June 1991 to December 1993 to determine the species present and to document variation in external morphology, habitat, and local distribution. These confirmed that a terrestrial frog resembling L. archeyi is present in the area, as well as Hochstetter's frog Leiopelma hochstetteri and the introduced Australian hylid frog Litoria aurea.

In Whareorino Forest, the terrestrial Leiopelma was mostly above 500 m altitude and L. hochstetteri above 350 m. The terrestrial Leiopelma occupies sites under rocks and logs in forest. It also occurs in vegetation, such as crown fern Blechnum discolor, tree fern Cyathea smithii, hook grass Uncinia uncinata, and rice grass Microlaena avenacea. Egg clusters of this frog were found in crown fern and tree fern, as well as under stones.

The terrestrial Leiopelma is susceptible to predation by Litoria aurea and rats. This is the first documented evidence of predation on Leiopelma on the New Zealand mainland. The future of this small remnant Leiopelma population is uncertain, and further investigation of the impact of anuran and mammalian predators is needed.  相似文献

Abstract

A reappraisal of the conservation status of the New Zealand frog fauna is presented using the 2008 version of the New Zealand Threat Classification System. Of New Zealand's four extant endemic species, three are judged to be ‘Threatened’ (Leiopelma hamiltoni being ‘Nationally Critical’, and L. pakeka and L. archeyi being ‘Nationally Vulnerable’) and one ‘At Risk’ (L. hochstetteri ‘Declining’). Three Leiopelma species are listed as extinct—they are known from bone deposits in caves throughout the country until some time in the last 1000 years. Three introduced and naturalised Litoria species are abundant in New Zealand although two (L. aurea and L. raniformis) are threatened in their country of origin (Australia). An additional unidentified frog taxon from northern Great Barrier Island is listed as ‘Data Deficient’.  相似文献

New Zealand's native frogs (genus Leiopelma) display a very high degree of endemism and belong to a distinctive and ancient evolutionary lineage. All four extant species are considered to be threatened or endangered, but the mechanisms behind their decline are poorly understood. Many of the potential factors causing population declines are confounded for the two species living on the mainland; in particular, habitat degradation and the presence of introduced mammalian pests have been difficult to disentangle in the past. Here we present data on a population of Hochstetter's Frog (Leiopelma hochstetteri) living in a pest‐free mainland sanctuary, the 3363‐ha Maungatautari Ecological Island. Survey data from this population shows a fourfold increase in numbers between 2009 and 2012 from a very low initial density. Our results suggest that for Hochstetter's Frog at least, and possibly the other mainland species of Leiopelma, introduced mammals are a major driving factor in population declines. Given that the frog population at Maungatautari was discovered by serendipity when planning for the pest‐free sanctuary was already well underway, this also highlights the conservation value of large pest‐free areas and ecosystem‐level management.  相似文献

Abstract

Leiopelma hochstetteri, the most widespread of New Zealand's native frogs, is recognised as threatened, and is fully protected by legislation. As a first step to characterise the diet and trophic level of L. hochstetteri within streams in the Waitakere Ranges, Auckland, stable carbon and nitrogen isotope analyses were undertaken on a variety of sympatric terrestrial and aquatic plant and animal species, including adult frogs. These results show that: (1) aquatic and terrestrial food webs are linked by terrestrial inputs into the stream; (2) invertebrate and vertebrate predators separate well into distinct trophic groups, and (3) L. hochstetteri occupies an intermediate trophic position among predators, with a diet, at least as an adult, comprising terrestrial invertebrates. Shortfin eels and banded kokopu are identified as potential predators of L. hochstetteri, but data for rats are inconclusive. These results have important implications for the conservation of New Zealand native frog species and riparian stream habitat.  相似文献

Leiopelma hochstetteri belongs to a singular anuran lineage endemic to New Zealand that diverged from other frogs about 200 million years ago. The species now is reduced to a series of isolated populations in the northern half of the North Island and Great Barrier Island. We have used mitochondrial and nuclear sequence data to examine the genetic affinities of extant populations of L. hochstetteri. Phylogenetic reconstructions reveal that populations are highly structured. Each of the geographically isolated populations harbours independent mtDNA lineages as well as different degrees of nuDNA differentiation. Moreover, molecular dating reveals that this structure originated in the early Pleistocene. This pattern of genetic structure likely results from unfavourable climatic conditions during the Pleistocene combined with the low dispersal ability of the species. Isolated populations in forested refugia existed even in the southern part of the distribution of the species during glacial cycles. Previously published variation in chromosome numbers and isozyme data are consistent with the new evidence. We identified 13 evolutionary significant units (ESUs) that should serve as the focus for future management and conservation of this species.  相似文献

Intraspecific color variation has long fascinated evolutionary biologists. In species with bright warning coloration, phenotypic diversity is particularly compelling because many factors, including natural and sexual selection, contribute to intraspecific variation. To better understand the causes of dramatic phenotypic variation in Malagasy poison frogs, we quantified genetic structure and color and pattern variation across three closely related species, Mantella aurantiaca, Mantella crocea, and Mantella milotympanum. Although our restriction site‐associated DNA (RAD) sequencing approach identified clear genetic clusters, they do not align with current species designations, which has important conservation implications for these imperiled frogs. Moreover, our results suggest that levels of intraspecific color variation within this group have been overestimated, while species diversity has been underestimated. Within major genetic clusters, we observed distinct patterns of variation including: populations that are phenotypically similar yet genetically distinct, populations where phenotypic and genetic breaks coincide, and populations that are genetically similar but have high levels of within‐population phenotypic variation. We also detected admixture between two of the major genetic clusters. Our study suggests that several mechanisms—including hybridization, selection, and drift—are contributing to phenotypic diversity. Ultimately, our work underscores the need for a reevaluation of how polymorphic and polytypic populations and species are classified, especially in aposematic organisms.  相似文献

Abstract

A terrestrial endemic frog resembling Leiopelma archeyi was discovered in the Whareorino Forest, northern King Country, New Zealand, in 1991, where it is broadly sympatric with L. hochstetteri. To clarify its taxonomic status, allozyme electrophoresis of toe tissue was used to compare it genetically with four other populations of terrestrial Leiopelma (L. archeyi from Tapu and Tokatea, Coromandel; L. hamiltoni from Stephens Island; L. pakeka from Maud Island). Thirteen presumed genetic (allozyme) loci could be consistently scored for the five populations. At 11 loci, no genetic differences were found between the Whareorino frog and the two Coromandel L. archeyi populations. Allelic frequencies differed slightly at two loci. We therefore conclude that the terrestrial Whareorino frog represents a western population of L. archeyi. L. hamiltoni from Stephens Island is genetically closer to L. archeyi than is L. pakeka from Maud Island. The Whareorino L. archeyi population is morphologically similar to Coromandel L. archeyi populations, although multivariate analysis suggests subtle morphological differences, including the relative position of the nostril. Size comparisons between Whareorino and three Coromandel sites (Moehau, Tapu, Tokatea) show there were more larger frogs (35–38 mm snout‐vent length) at Whareorino and Tokatea compared with Moehau and Tapu, where maximum snout‐vent lengths were 34 and 36 mm, respectively.  相似文献

The frogs Ascaphus truei and Leiopelma hochstetteri are members of the most basal lineages of extant anurans. Their cranial muscles have not been previously described in full and are investigated here by dissection. Comparison of these taxa is used to review a controversy regarding the homologies of the jaw adductor muscles in Lissamphibia, to place these homologies in a wider gnathostome context, and to define features that may be useful for cladistic analysis of Anura. A new muscle is defined in Ascaphus and is designated m. levator anguli oris. The differences noted between Ascaphus and Leiopelma are in the penetration of the jaw adductor muscles by the mandibular nerve (V3). In the traditional view of this anatomy, the paths of the trigeminal nerve branches define homologous muscles. This scheme results in major differences among frogs, salamanders, and caecilians. The alternative view is that the topology of origins, insertions, and fiber directions are defining features, and the nerves penetrate the muscle mass in a variable way. The results given here support the latter view. A new model is proposed for Lissamphibia, whereby the adductor posterior (levator articularis) is a separate entity, and the rest of the adductor mass is configured around it as a folded sheet. This hypothesis is examined in other gnathostomes, including coelacanth and lungfish, and a possible sequence for the evolution of the jaw muscles is demonstrated. In this system, the main jaw adductor in teleost fish is not considered homologous with that of tetrapods. This hypothesis is consistent with available data on the domain of expression of the homeobox gene engrailed 2, which has previously not been considered indicative of homology. Summary Previous work showed that the mutations uvrD3, uvrE156, and recL152 were closely linked and increased UV-sensitivity. They were phenotypically distinguishable in that only the uvrD3 mutation significantly decreases host cell reactivation of UV-irradiated phage (Hcr-) and repair of methylmethane sulfonate (MMS)-induced damage, and only the uvrE156 mutation increased mutation rates (Mut-). MMS-resistant revertants of a uvrD3 mutant were still UV-sensitive and fell into two phenotypic classes, Hcr- Mut+ (non-mutator) and Hcr+ Mut-. In this work complementation tests were done by examining UV-and MMS-sensitivity and host cell reactivation in heterogenotes containing combinations of uvrD3, uvrE156, recL152, and the MMS-resistant mutations derived from uvrD3. The mutations could not complement each other in the repair of UV-damage, the one trait all had in common, indicating that they were in one gene. For the most part, the different mutations were able to complement each other in respect to traits in which one was deficient and the other had wild type activity.  相似文献

Abstract

Photosynthetic responses to light intensity were studied under laboratory conditions in seven bryophyte species from evergreen laurel forest, a threatened habitat, on Terceira island in the Azores. Four mosses (Andoa berthelotiana, Echinodium prolixum, Fissidens serrulatus, Myurium hochstetteri) and three liverworts (Bazzania azorica, Frullania tamarisci, Lepidozia cupressina) were selected to encompass a range of potential responses to variations in the forest light environment. Carbon dioxide exchange measurements were made, using an infra-red gas-analyser, at photosynthetic photon flux densities (PPFD) of 0-900 µmol m-2 s-1 and a mean temperature of 21°C in fully hydrated shoots. Most species achieved light saturation of photosynthesis below 30 µmol m-2 s-1, the lowest value being for A. berthelotiana (20 µmol m-2 s-1) and the highest for M. hochstetteri (68 µmol m-2 s-1). The liverwort F. tamarisci had the highest maximum photosynthetic rate (Pmax, 23 µmol CO2 g-1 h-1) whereas Pmax was lowest in the mosses E. prolixum and M. hochstetteri (10 µmol CO2 g-1 h-1). Dark respiration rate, a critical factor in toleration of shade by forest floor plants, was highest in the species with the highest values for Pmax. Compensation point was extremely low (7 µmol photons m-2 s-1) in Fissidens serrulatus, a species found in the deep shade of forest ravines and caves, and highest in M. hochstetteri a moss restricted to better illuminated habitats within and outside the forest. No photoinhibition was detected during the relatively short exposures to high irradiances. Comparison of these responses with data on the forest light environment indicates that, despite the possession of considerable shade adaptations, during winter in the evergreen laurel forest, low light levels may often limit photosynthetic rates of the bryophytes.  相似文献

Nineteen polymorphic microsatellite loci were characterized from the endangered takahe (Porphyrio hochstetteri). Like many of New Zealand's other native avian species, levels of polymorphism were low, with variation detected at only 19 of 110 (17.3%) loci, and most polymorphic loci (78.9%) were diallelic (mean number of alleles = 2.3). Despite these low levels of variation, the microsatellites developed here will be useful for parentage assignment for confirming pedigrees, and investigating relationships between genetic variation, pedigree‐based inbreeding and reproductive success in this highly endangered species.  相似文献

The New Zealand freshwater mussels are taxonomically revised and compared with key Australian and South American taxa using DNA sequence data. Three living species are recognised: Echyridella menziesii, Echyridella aucklandica and Echyridella onekaka. Cucumerunio websteri websteri and Cucumerunio websteri delli, are treated as junior synonyms of Unio aucklandica, which is transferred from Cucumerunio (and Hyridella) to Echyridella. Lectotypes are designated for Unio waikarensis and Unio hochstetteri, which are illustrated together with primary type specimens of other relevant taxa. The type locality of U. aucklandica is restricted to the catchment of the Kawakawa River.  相似文献

We sequenced the chloroplast rubisco large subunit (rbcL) gene in 236 samples of Porphyra sensu lato from the northeast Pacific. Comparisons of sequences within the study area as well as comparisons with published sequences revealed up to five cryptic species among the 22 named species: a species closely related to Porphyra abbottiae, a species previously identified as P. pseudolinearis, a species closely related to P. pseudolanceolata and previously identified as that species, a previously unknown species from the eastern Aleutian Islands, and a species closely related to P. schizophylla and previously identified as that species. All of these previously unrecognized species had high bootstrap values separating them from the other species. In addition, our wide geographic sampling allowed us to extend, curtail or clarify the geographic ranges of a number of the species. We also provide published sequences for P. gardneri and P. smithii for the first time. We compared amount of sequence divergence within species grouped on the basis of sexuality (monoecious, sectored into separate male and female “halves”, or dioecious), habitat (high, mid, or low intertidal/subtidal), and seasonality (winter, spring, or summer) using Tukey’s HSD t test, but we observed no significant differences between species grouped in this manner. Different species showed different levels of genetic variation in the rbcL gene apparently unrelated to these traits. Also, we observed no differences in the patterns of genetic variation in a species based on whether the specimens were collected from outside or from within the region covered by ice during Pleistocene glaciations.  相似文献

The genera Leontodon s.str. and Hedypnois are so far the only known sources of hydroxyhypocretenolides, a rare subclass of guaianolide type sesquiterpene lactones. In this study the three endemic species from the Azorean Archipelago, L. filii, L. hochstetteri, and L. rigens, were analyzed together with L. hispidus and L. × grassiorum, a hybrid originating from L. hispidus and L. hochstetteri. Flowering heads were analyzed by UHPLC-DAD-MS with regards to their phenolics' profiles, establishing qualitatively identical profiles for all taxa. The following phenolics were detected in flowering heads of all investigated taxa: caffeoyltartaric acid, cichoric acid, chlorogenic acid, 3,5-di-O-caffeoylquinic acid, luteolin, luteolin 7-O-β-d-glucopyranoside, luteolin 4′-O-β-d-glucopyranoside, and luteolin 7-O-β-d-glucuronide.In UHPLC-DAD-MS analyses of the rhizomes, no flavonoids were detected. In rhizomes, caffeoyltartaric acid was only detected in L. hispidus. However, in addition to caffeoylquinic acid derivatives already found in the flowering heads, 1,5-di-O-caffeoylquinic acid, 3,4-di-O-caffeoylquinic acid, and 4,5-di-O-caffeoylquinic acid were detected in rhizomes of all investigated taxa.The chemophenetically most interesting group of hydroxyhypocretenolides was detected in rhizomes of all investigated taxa. 11,13β-Dihydro-14-dihydroxyhypocretenolide was detected in L. filii and L. hochstetteri, while 11,13β-dihydro-14-hydroxyhypocretenolide-β-d-glucopyranoside was present in all Azorean taxa. 1,10-Epoxy-14-hydroxyhypocretenolide-β-d-glucopyranoside and 1,10-epoxy-14-hydroxyhypocretenolide-β-d-glucopyranoside-6′-O-p-hydroxyphenylacetic acid ester were restricted to the Azorean taxa and the hybrid L. × grassiorum, while the dimeric sesquiterpenoid 14-hydroxyhypocretenolide-β-d-glucopyranoside-4′,14″-hydroxyhypocretenoate ester was restricted to L. hispidus and L. × grassiorum.  相似文献

To understand how species adapt and evolve it is necessary to appreciate the relationship between genetic variation and the environment. Here, the fossil record and molecular data from different lineages of the marine Gastropoda are used to understand the evolution of genetic variations found in the nuclear gene for mitochondrial malate dehydrogenase (mMDH; EC 1.1.1.37) in the living intertidal Muricid snail, Nucella lapillus. Assuming a molecular clock, DNA sequences of mMDH indicate that two variants found in N. lapillus, mMDH9 and mMDH10, may have arisen as long as 144 MY (million years) ago and at least prior to the evolution of the Muricidae approximately 112–90 MY ago. The Muricidae contain by far the greatest majority of the Neogastropoda specialized for life in the intertidal habitat. In N. lapillus the mMDH9 and mMDH10 variants covary with variations in other biochemically defined loci, inherited phenotypic traits (shell shape and physiology) and karyotype frequencies to differentiate two distinct nuclear haplotypes that are associated with different temperature environments. The variations in shell shape that are associated with the haplotypes of N. lapillus represent adaptations to temperature stress and similar variations occur in other related intertidal molluscs whose lineages are much older than Nucella, which arose around 25 million years ago. It is suggested that the divergence of the mMDH variants found in N. lapillus may reflect an ancient genetic event, such as a chromosomal mutation, perhaps involving variation in other linked traits that together became important in the subsequent evolution of the marine Gastropoda.  相似文献

The D3 domain and its flanking regions of 28S rRNA of four pairs of closely related sexual species (Eupelops hirtus and E. torulosus; Oribatella calcarata and O. quadricornuta; Chamobates voigtsi and Ch. borealis; Liacarus coracinus and L. subterraneus) and four pairs of closely related parthenogenetic species (Nanhermannia nana and Na. coronata; Nothrus silvestris and No. palustris; Tectocepheus sarekensis and T. minor; Camisia spinifer and Ca. segnis) of oribatid mites were sequenced to investigate (1) if the D3 region can be used as a species marker and (2) if there is genetic variation among closely related species pairs and if its magnitude is related to reproductive mode. Furthermore, we investigated the world-wide genetic variation of the D3 region from the oribatid mite species Platynothrus peltifer. There was no intraspecific genetic variation in the D3 region in any of the species studied; it was even identical in two closely related parthenogenetic species (Na. nana and Na. coronata) and two closely related sexual species (E. hirtus and E. torulosus). The genetic differences of the other species pairs indicated that both parthenogenetic and sexual lineages have various ages. On average, however, the differences between the closely related parthenogenetic species were larger than those between closely related sexual species, indicating that parthenogenetic lineages exist historically and may radiate slower than sexual species. The findings of this study support the hypothesis that some of the parthenogenetic oribatid mite taxa (Tectocepheus, Nothrus) are ‘ancient asexuals’. The absence of intraspecific or intra-individual variation in the D3 region of parthenogenetic species is consistent with the presence of concerted evolution in the 28S rRNA gene. From this we infer the existence of a meiotic process, which is consistent with the automixy known from several other parthenogenetic oribatid species. Selected NaCl tolerant and unselected control lines ofHolcus lanatus L.,Lolium perenne L.,Dactylis glomerata L., andFestuca rubra L. were grown in sand culture at 0, 100, 200, 250, and/or 300 ml m-3NaCl for seven weeks. The tolerant lines of all four species produced significantly greater both shoot and root dry matter at all NaCl treatments compared with the unselected control lines. Na+, K+, Cl-, Ca2+, and Mg2+ contents of leaf, stalk, and roots of each species were determined. The tolerant lines ofH. lanatus contained less Na+ and less Ca2+ but higher K+ in shoots, compared with the unselected line. By contrast theL. perenne tolerant line had higher Na+ and Cl- contents at 250, and 300 mol m-3 NaCl in shoots than the unselected line suggesting a halophytic nature of the tolerant line.D. glomerata accumulated greater quantities of ions compared with the other species examined. The tolerant line contained significantly less Cl- but more K+ in its shoots than the unselected line. Na+, Cl-, and K+ contents in the shoots of the tolerant line ofF.rubra were higher than in the unselected line shoots. Therefore selection for NaCl tolerance may provide useful material for examining the basis of tolerance.  相似文献
