Background and aims Cress-seed (Lepidium sativum) exudate exerts an allelochemical effect, promoting excessive hypocotyl elongation and inhibiting root growth in neighbouring Amaranthus caudatus seedlings. We investigated acidic disaccharides present in cress-seed exudate, testing the proposal that the allelochemical is an oligosaccharin—lepidimoic acid (LMA; 4-deoxy-β-l-threo-hex-4-enopyranuronosyl-(1→2)-l-rhamnose).Methods Cress-seed exudate was variously treated [heating, ethanolic precipitation, solvent partitioning, high-voltage paper electrophoresis and gel-permeation chromatography (GPC)], and the products were bioassayed for effects on dark-grown Amaranthus seedlings. Two acidic disaccharides, including LMA, were isolated and characterized by electrophoresis, thin-layer chromatography (TLC) and nuclear magnetic resonance (NMR) spectroscopy, and then bioassayed.Key Results Cress-seed exudate contained low-Mr, hydrophilic, heat-stable material that strongly promoted Amaranthus hypocotyl elongation and inhibited root growth, but that separated from LMA on electrophoresis and GPC. Cress-seed exudate contained ∼250 µm LMA, whose TLC and electrophoretic mobilities, susceptibility to mild acid hydrolysis and NMR spectra are reported. A second acidic disaccharide, present at ∼120 µm, was similarly characterized, and shown to be β-d-xylopyranosyl-(1→3)-d-galacturonic acid (Xyl→GalA), a repeat unit of xylogalacturonan. Purified LMA and Xyl→GalA when applied at 360 and 740 µm, respectively, only slightly promoted Amaranthus hypocotyl growth, but equally promoted root growth and thus had no effect on the hypocotyl:root ratio, unlike total cress-seed exudate.Conclusions LMA is present in cress seeds, probably formed by rhamnogalacturonan lyase action on rhamnogalacturonan-I during seed development. Our results contradict the hypothesis that LMA is a cress allelochemical that appreciably perturbs the growth of potentially competing seedlings. Since LMA and Xyl→GalA slightly promoted both hypocotyl and root elongation, their effect could be nutritional. We conclude that rhamnogalacturonan-I and xylogalacturonan (pectin domains) are not sources of oligosaccharins with allelochemical activity, and the biological roles (if any) of the disaccharides derived from them are unknown. The main allelochemical principle in cress-seed exudate remains to be identified.  相似文献

Lepidimoide promotes growth of the cockscomb hypocotyl. It is exuded from germinating seeds of various plant species into their culture environment. The mode of exudation of lepidimoide from seeds into the culture solution, using sunflower and buckwheat seeds, was studied in relation to seed germination. In the dry seeds, about 75% of the lepidimoide was found in the seed coat and about 25% in the kernel. Upon water imbibition it was released into the culture solution. However, the quantity of lepidimoide detected in the seed exudate was more than three times the total amount in dry and imbibed seeds, suggesting that lepidimoide was also produced de novo in the seeds and subsequently released. When seed coats or kernels were imbibed separately, the quantity of lepidimoide released from the seed coats into the culture solution was much the same as that in the dry seeds, but the amount of lepidimoide detected in the exudate of kernels was about 16 times that in the dry kernels. These results suggest that lepidimoide, already present in dry seeds, as well as that newly produced in the kernels following imbibition, was released into the environment.  相似文献

Summary Comparisons have been made of the amount and composition of seed and seedling exudates in barley, wheat, cucumber, and bean. Except in the case of wheat a greater proportion of the total nitrogen content in the seed exudates was formed by protein and peptide nitrogen than by nitrogen of free amino acids. In contrast, the greater part of the total nitrogen in the exudates of seedlings was formed by free amino acid nitrogen, except in the case of barley. Peptides represented 8 to 26 per cent of the protein and peptide fraction in seed exudates, the highest amount being found in bean. On an equal weight basis, the spectrum of amino acids released from seeds and seedlings differed little between barley, wheat and cucumber; greater differences were observed in bean exudates. Seedlings exuded reducing substances to a considerably greater extent than seeds. The spectrum of reducing sugars in seed and root exudates differed greatly, especially in the case of keto sugars. Differences in the organic acid spectra were small, except for bean plants, the seedling exudate of which contained more organic acids with a richer spectrum than seed exudate. Both seeds and seedlings of cucumber exuded a small quantity and a poor spectrum of organic acids.  相似文献

A new allelopathic substance that promoted the shoot growth of different plant species but inhibited the root growth was isolated as an amorphous powder from mucilage of germinated cress (Lepidium sativum L.) seeds. This substance was identified as sodium 2-O-rhamnopyranosyl-4-deoxy-threo-hex-4-enopyranosiduronate (designated lepidimoide) from the mass and the nuclear magnetic resonance and infrared spectra coupled with some chemical evidence. Lepidimoide promoted the hypocotyl growth of etiolated Amaranthus caudatus L. at concentrations higher than 3 μm and inhibited the root growth at concentrations higher than 100 μm. The growth-promoting activity in hypocotyls was 20 or 30 times as much as that of gibberellic acid.  相似文献

Hyperaccumulator species of the genera Alyssum can accumulate 100 times more Ni than normal crops and are therefore used for phytomining and phytoextraction of nickel contaminated soils. Basic studies on the physiology and metal uptake mechanisms of these plants are needed to increase efficiency and uptake capacity of Nickel (Ni) by hyperaccumulators. Recent attempts to disclose if those hyperaccumulator species require higher Ni level than normal plants failed because of the high Ni content in the seeds (7000-9000 microg g(-1)). In this study, we attempted to use chelator buffered nutrient solution to deplete Ni from the seed/seed coat and to obtain low Ni seedlings of Alyssum cultivars to be used in physiology studies. HEDTA-buffered nutrient solution did not deplete Ni from the seeds, perhaps because Ni was mainly localized within the seedling embryonic tissues with greatest Ni enrichment in the cotyledons and hypocotyls. We could not observe any positive correlation between seed fitness and germination capacity with seed Ni content. Investigation of nickel localization in Alyssum seeds using synchrotron X-ray microfluorescence (micro-SXRF) showed that nickel is localized in the embryonic tissues with greatest Ni enrichment observed in the cotyledons and hypocotyl.  相似文献

以大白菜、萝卜、番茄和黄瓜种子为受体,采用实验室培养皿种子发芽生物测试法研究了黄瓜种子浸提液、种子萌发、胚根和芽苗分泌物、芽苗腐解物和芽苗浸提液的化感效应。结果表明:(1)黄瓜种子浸提液对大白菜、萝卜、番茄和黄瓜种子萌发均有化感抑制作用,即黄瓜种子内含有某些化感抑制物质。(2)在水浸提过的黄瓜种子萌发过程中,它不仅对其近邻套种的大白菜、萝卜和番茄种子萌发产生化感抑制作用,而且其胚根和芽苗分泌物对后茬播种的4种蔬菜种子发芽也表现出不同程度的化感抑制作用;黄瓜芽苗腐解物和芽苗水浸提液也对各受体蔬菜种子发芽与生长产生不同程度的化感抑制作用,且随着腐解芽苗量的增加或浸提液浓度的升高,各受体蔬菜种子的发芽指标值、化感效应指数值和综合效应值随之降低。(3)黄瓜种子浸提液及芽苗各器官的化感物质对黄瓜种子的萌发与生长产生了自毒作用,且黄瓜芽苗腐解物、芽苗浸提液、胚根及芽苗分泌物对受体黄瓜的自毒作用均为最大。研究发现,黄瓜种子浸提液、种子萌发时期以及芽苗各器官的化感物质主要是通过抑制受体胚根的生长而起化感抑制作用,即受体蔬菜种子胚根对化感效应最为敏感;因黄瓜种子及萌发期释放化感物质的途径有所不同,导致受体大白菜、萝卜、黄瓜和番茄的化感响应也不相同;在黄瓜种子萌发和芽苗生长的早期,化感物质即开始在芽苗体内进行合成与积累,一部分可通过胚根和芽苗分泌途径释放到环境中,另一部分可通过芽苗腐解途径释放化感物质,并对受体蔬菜种子萌发与生长表现出较强的化感抑制作用。  相似文献

Abstract. A system is described whereby seedling development can be analysed in terms of growth rates of specific 1 mm regions of the hypocotyl. The technique involves time-lapse photography of marked hypocotyls in a specially designed chamber which accommodates seedlings in various orientations with respect to gravity, and under irradiation regimes differing in light quality, quantity and direction. The results of a preliminary study of the upward growth of etiolated or green cress seedlings in darkness or overhead while light are reported. Highest growth rates in etiolated seedlings were observed in zones in the upper one-third of ihe hypocotyl. In green seedlings, growth was more prominent within the subapical zones. Light further restricted growth of the median and basal zones in both types of seedling. However, in their immediate responses to the onset of irradiation, green and etiolated seedlings differed markedly. In etiolated seedlings, recovery of growth at the apex was accompanied by the development of inhibition in the median-basal regions; green seedlings showed a transient inhibition of growth in the apical zone together with a strong immediate inhibition in the median-basal regions.  相似文献

Summary Amino acids, organic acids, and sugars make up the bulk of 14C-assimilates that leach from freshly excised sections of subterranean hypocotyls of squash plants allowed to fix 14CO2 photosynthetically. Based on kinetics of leakage, it appears that metabolites are present in significant levels in intercellular regions of squash hypocotyls and that they may be the major source of hypocotyl exudates. Although the anatomical relations of epidermal cells hinder excessive metabolite losses by subterranean portions of hypocotyls, avenues of diffusion are continuous along cell walls and middle lamellae between cortical tissue and the hypocotyl surface. re]19751115  相似文献

Germinating seeds tend to release a variety of proteins into their surrounding surfaces; some of which have an inhibitory action against plant pathogens. The aim of this study was to investigate and identify defence proteins present in the exudates from water-imbibed and chitosan-imbibed (0.1% w/v) seeds of chickpea (Cicer arietinum L). Chickpea seeds imbibed in chitosan released a higher amount of proteins in the exudate when compared to the seeds imbibed in water. The obtained exudates were analysed in regard to specific protein activities by enzymatic assays and SDS-PAGE analysis. Results showed that the exude obtained from chickpea seeds imbibed in chitosan solution exhibited a new isoform of chitinase, chitosanase and protease inhibitors. These exudates also have an “in vitro” inhibitory effect on the growth of the fungus, Fusarium oxysporum f.sp. ciceri. Our results suggest that seed exudates protect seeds during their germination from soil pathogens.  相似文献

Rice seedlings release momilactone B into the environment   总被引:7,自引:0,他引:7  
Since the growth inhibitor momilactone B was found recently in root exudates of rice (Oryza sativa L.), 3-day-old rice seedlings were transferred to hydroponic culture and the level of momilactone B released into the environment from the seedlings was measured. At day 15 after transfer, the level of momilactone B in the culture solution was 1.8 nmol per seedling compared with endogenous levels of 0.32 and 0.63 nmol per root and shoot, respectively, suggesting that rice seedlings actively releases momilactone B into the culture solution. This release must occur from the roots because only rice roots were immersed in the culture solution. Momilactone B inhibited the growth of ten cress (Lepidium sativum L.) seedlings at concentrations greater than 3 microM. Ten rice seedlings were incubated with ten cress seeds in a Petri dish containing 1 ml of medium, the medium contained 18 nmol of momilactone B, which came to 18 microM. This level of momilactone B was enough to reveal growth inhibition of the cress seedlings. Release level of momilactone B and its effectiveness as a growth inhibitor suggest that it may play an important role in rice allelopathy.  相似文献

Diamine oxidase (DAO, EC 1.4.3.6.) activity and polyamine content were measured in the shoot apex, leaves, epicotyl, cotyledons, hypocotyl and roots of light-grown bean ( Phaseolus vulgaris L. cv. Lingot) and soybean ( Glycine max L. cv. Sakai) seedlings at 3 different stages of germination (5, 8 and 14 days) as well as in embryos and cotyledons from soaked seeds. No DAO activity was detected in embryos and cotyledons of either plants. In bean seedlings DAO activity was only detectable in the shoot apex, primary leaves and cotyledons, while in soybean the activity was only detectable in the hypocotyl and roots. During seedling growth, in both plants, a different pattern of DAO activity was observed. In both species spermidine and spermine were the most abundant polyamines in embryos and cotyledons. Cadaverine, absent in bean, was only detected in soybean embryos. In the seedlings of both plants, increasing gradients of putrescine, spermidine and spermine from base to shoot apex were found. A high concentration of cadaverine was present in soybean hypocotyls and roots. A possible correlation between DAO activity and the endogenous content of the preferential substrate is discussed in relation to the possible involvement of the enzyme in regulating the cellular level of polyamines.  相似文献

The hypocotyls or roots of many seed plants contract during seedling growth. Anatomical evidence is here reported for the first time that G-fibers (gelatinous or tension wood fibers) may cause contraction of roots and hypocotyls in dicotyledonous seedlings long after germination. To document repositioning of seedling buds, selected perennials (20 dicotyledons and one cycad) native to the fire-prone pine rocklands of subtropical South Florida were germinated and measured for 4-5 mo. The height of cotyledonary nodes above the soil decreased because of axis contraction or bending in eight species. Anatomy suggested that two mechanisms operate: (1) previously well-documented collapse of parenchyma cells in two species (Convolvulaceae and Zamiaceae) and (2) newly documented production of G-fibers in six species (all Fabaceae). Contraction or bending of the hypocotyl and/or taproot moved the cotyledonary and later buds of the seedling closer to the soil surface or buried them. Bud repositioning by these mechanisms may protect the lateral buds from injury by fire or other environmental stresses and allow resprouting.  相似文献

A few rice (Oryza sativa L.) varieties or rice straw produce and release allelochemicals into soil in which interfere with the growth of neighboring or successive plants. Allelopathic rice PI312777 and Huagan-1 at their early growth stages released momilactone B, 3-isopropyl-5-acetoxycyclohexene-2-one-1, and 5,7,4′-trihydroxy-3′,5′-dimethoxyflavone into soil at phytotoxic levels, but non-allelopathic rice Huajingxian did not. Both allelopathic and non-allelopathic rice residues released momilactone B and lignin-related phenolic acids (p-hydroxybenzoic, p-coumaric, ferulic, syringic and vanillic acids) into the soil during residue decomposition to inhibit successive plants. The results indicated that allelochemicals involved in rice allelopathy from living and dead plants are substantially different. Interestingly, the concentrations of the allelochemicals released from the allelopathic rice seedlings in soil increased dramatically when they were surrounded with Echinochloa crus-galli. The concentrations of the allelochemicals were over 3-fold higher in the presence of E. crus-galli than in the absence of E. crus-galli. However, the same case did not occur in non-allelopathic Huajingxian seedlings surrounded with E. crus-galli. In addition to allelochemical exudation being promoted by the presence of E. crus-galli, allelopathic rice seedlings also increased allelochemical exudation in response to exudates of germinated E. crus-galli seeds or lepidimoide, an uronic acid derivative exuded from E. crus-galli seeds. These results imply that allelopathic rice seedlings can sense certain allelochemicals released by E. crus-galli into the soil, and respond by increased production of allelochemicals inhibitory to E. crus-galli. This study suggests that rice residues of both allelopathic and non-allelopathic varieties release similar concentrations and types of allelochemicals to inhibit successive plants. In contrast, living rice plants of certain allelopathic varieties appear to be able to detect the presence of interspecific neighbors and respond by increased allelochemicals.  相似文献

Abstract. Growth in various regions of the hypocotyls of dark-grown cress seedlings before and after exposure lo continuous white light has been analysed by time-lapse photography. In the dark, growth in the hook was minimal, the upward growth of the seedling being sustained by extension of the shank, especially the uppermost zones. Following irradiation, the hook and the remainder of the hypocotyl showed dissimilar growth responses. Previously growing regions of the shank were inhibited while zones within the hook, especially the apical end of the inner (concave) side, showed marked growth stimulation. These changes in growth rates commenced within 1 h from exposure to the light stimulus and thus considerably preceded any observable changes in hook angle.  相似文献

Summary The flowering of pin (long-styled) and thrum (short-styled) plants in populations of Lithospermum caroliniense (Walt.) McMill. was synchronized and both morphs had an extended period of flowering. The plants produced a large number of flowers that did not set seeds — hence the seed production per plant was very low. The seeds showed strong dormancy. Even under natural conditions, only 19% of the seeds produced seedlings. However, seedling survival was relatively high probably due to rapid root growth to deeper layers where moisture was not limiting. The probability of survival of seedlings increased with age; survival of seedlings, juveniles and adults after 2 years was 39, 68 and 98%, respectively. The dry weight of roots increased from 10% in June to 42% in September whereas the proportion of biomass contained in leaves, stems and hypocotyls decreased with age.  相似文献

In the dark, plant seedlings follow the skotomorphogenetic developmental program, which results in hypocotyl cell elongation. When the seedlings are exposed to light, a switch to photomorphogenetic development occurs, and hypocotyl cell elongation is inhibited. We have manipulated the expression of the AtPGP1 (for Arabidopsis thaliana P glycoprotein1) gene in transgenic Arabidopsis plants by using sense and antisense constructs. We show that within a certain light fluence rate window, overexpression of the AtPGP1 gene under the control of the cauliflower mosaic virus 35S promoter causes plants to develop longer hypocotyls, whereas expression of the gene in antisense orientation results in hypocotyls shorter than those occurring in the wild type. In the dark, hypocotyls of transgenic and wild-type plants are indistinguishable. Because the AtPGP1 gene encodes a member of the superfamily of ATP binding cassette-containing (ABC) transporters, these results imply that a transport process is involved in a hypocotyl cell elongation pathway active in the light. The AtPGP1 transporter is localized in the plasmalemma, as indicated by immunohistochemical techniques and biochemical membrane separation methods. Analysis of the AtPGP1 expression pattern by using reporter gene constructs and in situ hybridization shows that in wild-type seedlings, AtPGP1 is expressed in both the root and shoot apices.  相似文献

在实验室人工控制条件下 ,研究了不同浓度 1,2 ,4 三氯苯胁迫对大豆种子萌发和幼苗生长的影响 .结果表明 ,5 0~ 2 0 0 μg·g-1的 1,2 ,4 三氯苯胁迫对大豆种子的发芽率影响甚小 ,但延迟大豆种子的出苗速率 ;30 0 μg·g-1的 1,2 ,3 三氯苯胁迫使种子停止发芽 .在正常条件下 ,大豆种子播种后第 4天其下胚轴超氧化物歧化酶 (SOD)活性达到高峰 ,之后趋于稳定 .不同浓度 1,2 ,4 三氯苯处理使大豆下胚轴SOD活性下降 ,丙二醛 (MDA)、蛋白质含量及下胚轴直径增加 ;幼苗的生长受抑制 ,干重、鲜重有一定程度的下降 ;而鲜重 /干重的比值变化不大 .受害程度随着 1,2 ,4 三氯苯浓度的增加而加重 .初生根比下胚轴对 1,2 ,4 三氯苯胁迫较为敏感 .  相似文献

Adventitious buds were formed on the hypocotyls of decapitated flax seedlings. Scanning electron and light microscopic examinations of hypocotyls showed that epidermal cells divided to produce meristematic spots from which several leaf primordia were formed. Between leaf primordia and the original vascular tissues of hypocotyls, new xylem cells were formed which connected them. About 10, 30 and 60% of adventitious buds were formed on upper, middle and basal parts of hypocotyls of decapitated seedlings, respectively. Removal of apical meristem together with longer hypocotyl zero to four cm long below the apical meristem) induced higher percentage of adventitious bud formation in the remaining hypocotyl. When the entire hypocotyl was cut into 16 segments (0.25 cm each) and these segments were cultured on MS medium containing 3% sucrose and 0.8% agar, adventitious buds were mainly formed in the lowest five segments. These results suggested that there was a gradient of inhibitory factor(s) from apical to basal part of hypocotyl with respect to adventitious bud formation. Auxin transport inhibitors, morphactin and TIBA induced adventitious bud formation on intact seedlings by suppressing the basipetal movement of auxin.  相似文献

Summary The occurrence of elongation growth-related osmiophilic particles (OPs) was investigated in hypocotyls of sunflower, bean, and spruce as well as in pea epicotyls and in cress roots of intact seedlings. In all analyzed species, OPs were found to occur specifically within the periplasmic space between plasma membrane and the outer epidermal cell walls of elongating parts of hypocotyls, epicotyls, and roots, whereas cells of nonelongating parts were devoid of OPs. Auxin (IAA) markedly increased the number of OPs in epicotyl and hypocotyl segments. Treatment of pea epicotyl segments with the lectin concanavalin A inhibited their elongation growth in the presence of IAA. At a subcellular level this effect was characterized by the occurrence of a pronounced osmiophilic layer in the periplasmic space of the outer periclinal and the outer part of the anticlinal epidermal cell walls. Treatment of IAA-incubated segments with the secretion inhibitor brefeldin A inhibited both elongation growth and periplasmic occurrence of OPs. This effect was accompanied by complementary accumulation of OPs in the peripheral cytoplasm of epidermal cells. Together the results indicate that IAA-induced epidermis-specific secretion of OPs is closely related to cell elongation growth not only in organs of monocotyledonous species, but also in dicotyledonous angiosperms as well as in gymnosperms.Abbreviations OPs osmiophilic particles - ConA concanavalin A - BFA brefeldin A - IAA -indolyl acetic acid  相似文献
