Cd~(2+)胁迫对小桐子幼苗叶片抗氧化系统的影响   总被引:1,自引:0,他引:1  
以小桐子幼苗为材料,设置不同浓度CdCl_2处理,测定Cd~(2+)胁迫对小桐子幼苗叶片中可溶性蛋白、丙二醛(MDA)含量,以及5种抗氧化酶活性和2种抗氧化剂含量的变化,探讨镉胁迫对小桐子幼苗抗氧化系统的影响。结果表明:(1)Cd~(2+)胁迫导致小桐子幼苗叶片中可溶性蛋白含量降低、MDA含量增加;(2)随着镉胁迫时间的延长,幼苗叶片中愈创木酚过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、抗坏血酸专一性过氧化酶(APX)、谷胱甘肽还原酶(GR)等抗氧化酶活性表现出先升高然后降低的变化趋势;(3)幼苗叶片中还原型抗坏血酸(ASA)和还原型谷胱甘肽(GSH)含量随着胁迫时间延长而降低,但其中氧化型抗坏血酸(DHA)和氧化型谷胱甘肽(GSSG)含量则升高。研究表明,镉胁迫初期能诱导小桐子幼苗抗氧化系统活性显著增强,提高其抗氧化能力,但随着胁迫时间的延长,致使其抗氧化酶的活性和抗氧物质含量下降,植株遭受明显氧化胁迫,幼苗生长受到镉的严重毒害。  相似文献

Sunflower (Helianthus annuus L.) seeds were germinated and grown in the presence of 50, 100 and 200 μM CdCl2. The lower concentration (50 μM) of Cd2 ions produced slight decrease in reduced glutathione (GSH) content and overall increase (except superoxide dismutase) in antioxidant enzyme activities, and in H2O2 concentration. Chlorophyll content, lipid peroxidation and protein oxidation were not affected under 50 μM CdCl2. GSH content was diminished under 100 and 200 μM CdCl2, and except for superoxide dismutase, which activity remained unaltered, overall decreases in the antioxidant enzyme activities (catalase, ascorbate peroxidase, dehydroascorbate peroxidase, glutathione reductase) and in guaiacol peroxidase were observed. These Cd2 concentrations caused a decrease in chlorophyll content as well as an increase in lipid peroxidation, protein oxidation and H2O2 concentration. All the observed effects were more evident with the highest concentration of cadmium chloride used. The effect of calcium (Ca2+) on Trifolium repens L. seedlings subjected to cadmium (Cd2+) stress was studied by investigating plant growth and changes in activity of antioxidative enzymes. Physiological analysis was carried out on seedlings cultured for 2 weeks on half-strength Hoagland medium with Cd2+ concentrations of 0, 400 and 600 μM, and on corresponding medium supplied with CaCl2 (5 mM). Exposure to increasing Cd2+ reduced the fresh weight of the upper part (stems + leaves) of the seedlings more strongly than that of the root system. In both parts of T. repens seedlings H2O2 level and lipid peroxidation increased. In the upper part, Cd2+ exposure led to a significant decrease in the activity of superoxide dismutase, catalase and glutathione peroxidase and an increase in ascorbate peroxidase activity. In contrast, the roots showed an increase in the activity of antioxidative enzymes under Cd2+ stress. Ca2+ addition to medium reduced the Cd2+ accumulation, and considerably reversed the Cd2+-induced decrease in fresh mass as well as the changes in lipid peroxidation in the both parts of T. repens seedlings. Ca2+ application diminished the Cd2+ effect on the activity of antioxidative enzymes in the upper part, even though it did not significantly affect these enzymes in the roots. So the possible mechanisms for the action of Ca2+ in Cd2+ stress were considered to reduce Cd2+ accumulation, alleviate lipid peroxidation and promote activity of antioxidative enzymes.  相似文献

Soybean (Glycine max L. ev. Columbus) seedlings grown in culture solution were treated with cadmium as CdSO4. Final concentrations of cadmium (Cd2+) in the solution were 0, 0.45, 0.90, and 1.35 μM. Soybean leaves, analyzed 10 days after Cd2+ was added to the culture solution, showed increased respiration rate and activities of malate dehydrogenase, acid phosphatase, ribonuclease, deoxyribonuclease, and peroxidase but decreased activity of carbonic anhydrase. Increased activity of hydrolytic enzymes and peroxidase reflects a general senescence response while the carbonic anhydrase decrease is consistent with an antagonism between cadmium and endogenous zinc. Chlorosis, epinasty, abscission of leaves, and decreased growth rate occurred in seedlings treated with 1.35 μM Cd2+.  相似文献

Functioning of antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APO), and guaiacol peroxidases (GPO)) and low-molecular organic ROS scavengers (proline and phenolic compounds) in various organs (roots, cotyledons, stem, and needle) of 6-week-old seedlings of pine (Pinus sylvestris L.) developing in the chronic presence of ZnSO4 (50, 100, and 150 μM). Pine seedlings were grown in water culture in the climate-controlled chamber at an irradiance of 37.6 W/m2 with a 16-h photoperiod, an air temperature of 23 ± 1/15 ± 1°C (day/night), and a relative humidity of 55/70% (day/night). Endogenous Zn content was a key factor determining SOD activity decomposing superoxide into H2O2 and O2. Hydrogen peroxide produced is efficiently destroyed by CAT and also by APO and GPO. At the same time, the content of proline increased (especially at 150 μM ZnSO4), but the content of phenolic compounds remained unchanged. All these processes help to maintain stable intracellular levels of O2⊙− and H2O2 at elevated zinc concentrations and to prevent generation of hydroxyl radical and development of oxidative stress.  相似文献

Indian mustard (Brassica juncea L. cv. Vitasso) plants exposed to 10, 30, 50 and 100 μM of Cd for 5 d in hydroponic culture were analysed with reference to the distribution of Cd2+, the accumulation of biomass and antioxidants and antioxidative enzymes in leaves. Cd induced a decrease in plant biomass. The maximum accumulation of Cd occurred in roots followed by stems and leaves. Cd induced a decrease in catalase (CAT) and guiacol peroxidase (GPX) activities but an increase in ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDHAR) activities. Enhancement in dehydroascorbate reductase (DHAR) activity was also at 10 μM Cd. Glutathione reductase (GR) activity showed pronounced stimulation after all treatments, but glutathione S-transferase (GST) and glutathione peroxidase (GPOX) activities decreased. The effectiveness of ascorbate-glutathione cycle (AGC) was determined by the ratio of ascorbate to H2O2. This ratio decreased in the Cd-treated leaves which indicated that the cycle was disordered.  相似文献

Excess cadmium (Cd2+) in the soil environment is taken up by plants and can cause phytotoxicity. Elevated temperatures also lead to deleterious effects on plants. Plants are very often exposed to a combination of stresses rather than a single stress. The effect of Cd2+ and heat stress (HS) on the growth, root ultrastructure, lipid peroxidation (MDA), hydrogen peroxide accumulation and the activities of antioxidant enzymes peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) of rice roots from sensitive cv. DR-92 and tolerant cv. Bh-1 were investigated at 10 and 20 day of growth under controlled conditions. At day 10 under all Cd2+ treatments, the Cd2+ content between the two rice cultivars were almost similar. Application of 500 μM Cd2+ significantly increased metal concentrations at day 20 in the roots of rice seedlings resulting in a maximum accumulation of 44.25 μg Cd2+ g-1 dry wt in cv. DR-92 and 30 μg Cd2+ g-1 dry wt in cv. Bh-1 with a ~25 % decline in Relative Growth Index (RGI) in cv. DR-92. TEM studies revealed slight disorganization with cell wall ingrowths in root tissues from cv. DR-92 grown in 100 μM Cd2+ + HS. Uptake and accumulation of Cd2+ increased upon heat treatment in parenchyma, vacuoles and vascular cylinder of root tissues. Peroxidase primarily located in cell walls, the intensity being higher in sensitive cv. DR-92. Under Cd2+ stress alone, plants of sensitive cv. DR-92 significantly increased the H2O2 and MDA levels together with increased activities of the enzymes POD, CAT and APX at day 10 but remained almost stable at day 20. A strong increase in MDA levels was noted at day 20 in tolerant cv. Bh-1. Cd2+ + HS treatments in tolerant cv.Bh-1 led to a decreased H2O2 and MDA levels and decreased activities of the enzymes POD, CAT and APX. Results suggest stimulation of root antioxidant system under combination of two stresses and that heat stress seem to have a direct protective role by mitigating the effect of mild Cd2+ toxicity largely by enhanced Cd2+-MT formation contributing thereby towards the management of Cd2+ toxicity at cellular level that confers Cd2+ tolerance to rice cv. Hydroponic experiments were carried out to study the role of alginate-derived oligosaccharides (ADO) in enhancing wheat (Triticum aestivum L.) tolerance to cadmium stress. Data were collected on plant biomass, chlorophyll content, photosynthetic rate, antioxidant enzyme activity and malondialdehyde (MDA) content. Under 100 μM Cd stress, plant growth was significantly inhibited. Shoot length, root length, fresh and dry weight were sharply reduced by 24.21, 34.59, 22.1 and 14.7%, respectively of the control after 10 day of Cd exposure. Superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) activities were increased and MDA content increased. Wheat seeds were soaked for 5 h in 1,000 mg L−1 ADO solution before cadmium stress. ADO pretreatment alleviated cadmium toxicity symptoms, which were reflected by increasing root and shoot lengths, fresh and dry weight, chlorophyll content and photosynthetic rate (P n ). Furthermore, ADO pretreatment significantly increased antioxidant enzyme (SOD, CAT and POD) activities and reduced MDA content in leaves and roots. The results indicated that ADO pretreatment partially protected the seedlings from cadmium toxicity during the following growth period.  相似文献

A hydroponics experiment was conducted to test the effects of sodium nitroprusside (SNP, a donor of NO) supplied with different concentrations on copper (Cu) toxicity in ryegrass seedlings (Lolium perenne L.). Excess Cu (200 µM) reduced chlorophyll content, resulting a decrease in photosynthesis. Cu stress induced the production of hydrogen peroxide (H2O2) and superoxide anion (O2? ?), leading to malondialdehyde (MDA) accumulation. Furthermore, activities of antioxidant enzymes in Cu-treated seedlings such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were decreased. In addition, Cu stress inhibited the uptake of K, Mg, Fe, and Zn and increased Ca content in roots. Moreover, in leaves of Cu-stressed seedlings, K, Fe, and Zn contents were decreased and the contents of Ca and Mg were not affected significantly. In Cu-treated seedlings, Cu concentration in roots was higher than in leaves. Addition of 50, 100, 200 µM SNP in Cu-mediated solutions increased chlorophyll content and photosynthesis, improved antioxidant enzyme activities, reduced Cu-induced oxidative damages, kept intracellular ion equilibrium under Cu stress, increased Cu concentration in roots and inhibited Cu accumulation in leaves. In particular, addition of 100 µM SNP had the best effect on promoting growth of ryegrass seedlings under Cu stress. However, the application of 400 µM SNP had no obvious alleviating effect on Cu toxicity in ryegrass seedlings.  相似文献

Cadmium accumulation, the relative content of different chemical forms of Cd, as well as the toxic effect of Cd on nutrient element uptake, physiological parameters, and ultrastructure of Sagittaria sagittifolia L. seedlings were determined after the seedlings were exposed to different Cd concentrations for 4 days. The results showed that S. sagittifolia had the ability to accumulate large amounts of Cd. In the root, stem, and bulb, the predominant chemical Cd forms were NaCl extractable. With an increase in the Cd2+ concentration, the chlorophyll content, the relative membrane penetrability (RMP) of root cells, peroxidase (POD) activity, superoxide dismutase (SOD) activity in leaves, malondiadehyde (MDA) content and the superoxide anion (O2) generation rate in roots all decreased following an initial increase. On the other hand, catalase (CAT) activity, SOD activity in roots, MDA content, and the generation rate of O2 in leaves all increased gradually. The toxic effect of Cd2+ was more severe on roots than on leaves at the same concentration. Cadmium affected the mineral nutrition balance; mainly, it promoted the uptake of Ca, Cu, Mn, and Fe, while inhibited Mg, Na, and K uptake. The physiological toxic effect of Cd2+ was close to the ultrastructural damage induced by Cd contamination. A significant correspondence was observed between the Cd dose and its toxic effect. Cadmium could destroy the normal ultrastructure, disturb the ion balance, and interfere with cell metabolism.  相似文献

Abscisic acid (ABA), a widely known phytohormone involved in the plant response to abiotic stress, plays a vital role in mitigating Cd2+ toxicity in herbaceous species. However, the role of ABA in ameliorating Cd2+ toxicity in woody species is largely unknown. In the present study, we investigated ABA restriction on Cd2+ uptake and the relevance to Cd2+ stress alleviation in Cd2+-hypersensitive Populus euphratica. ABA (5 μM) markedly improved cell viability and growth but reduced membrane permeability in CdCl2 (100 μM)-stressed P. euphratica cells. Moreover, ABA significantly increased the activity of the antioxidant enzymes catalase (CAT), superoxide dismutase (SOD), and ascorbate peroxidase (APX), contributing to the scavenging of Cd2+-elicited H2O2 within P. euphratica cells during the period of CdCl2 exposure (100 μM, 24–72 h). ABA alleviation of Cd2+ toxicity was mainly the result of ABA restriction of Cd2+ uptake under Cd2+ stress. Steady-state and transient flux recordings showed that ABA inhibited Cd2+ entry into Cd2+-shocked (100 μM, 30 min) and short-term-stressed P. euphratica cells (100 μM, 24–72 h). Non-invasive micro-test technique data showed that H2O2 (3 mM) stimulated the Cd2+-elicited Cd2+ influx but that the plasma membrane (PM) Ca2+ channel inhibitor LaCl3 blocked it, suggesting that the Cd2+ influx was through PM Ca2+-permeable channels. These results suggested that ABA up-regulated antioxidant enzyme activity in Cd2+-stressed P. euphratica and that these enzymes scavenged the Cd2+-elicited H2O2 within cells. The entry of Cd2+ through the H2O2-mediated Ca2+-permeable channels was subsequently restricted; thus, Cd2+ buildup and toxicity were reduced in the Cd2+-hypersensitive species, P. euphratica.

Phragmites (Phragmites australis Cav. (Trin.) ex Steud) plants exposed to a high concentration of CdSO4 (50 μM) for 21 d were analysed with respect to the distribution of metal, its effects on antioxidants, the antioxidant enzymes and the redox status in leaves, roots and stolons. The highest accumulation of Cd2+ occurred in roots followed by leaves, and it was not significant in the stolons when compared with the control plants. In particular, in roots from Cd-treated plants, both the high amount of GSH and the parallel increase of glutathione-S-transferase (EC 2.5.1.18; GST) activity seemed to be associated with an induction of the detoxification processes in response to the high cadmium concentration. Superoxide dismutase (EC 1.15.1.1; SOD), ascorbate peroxidase (EC 1.11.1.11; APX), glutathione reductase (EC 1.6.4.2; GR) and catalase (EC 1.11.1.6; CAT) activities as well as reduced and oxidised glutathione contents in all samples of leaves, roots and stolons were increased in the presence of Cd2+ when compared to control plants. Despite the fact that Cd2+ has a redox characteristic not compatible with the Fenton-type chemistry that produces active oxygen species, the antioxidant response is widespread and generic. Increased activities of antioxidant enzymes in Cd-treated plants suggest that metal tolerance in Phragmites plants might be associated to the efficiency of these mechanisms.  相似文献

When seedlings of two rice (Oryza sativa L.) cultivars Ratna and Jaya were raised under 100 and 500 μM cadmium nitrate in the medium, a high proline content was noted in Cd2+ stressed seedlings compared to controls. Seedlings grown under 500 μM Cd(NO3)2 maintained increased proline level compared to non-stressed seedlings. Kinetic properties of RNase extracted from control grown and Cd2+ stressed seedlings showed a marked alteration in Km due to Cd2+ treatment. The RNase isoforms were purified from 15-d-old rice seedlings with a total purification of 22.25 fold and 74.75 % yield using conventional biochemical techniques. Three RNase isoforms, namely I, II and III were eluted from DEAE-Sephacel column. The isoform RNase II had Km value of 3.2 mg(RNA) cm-3. The in vitro osmotic stress created by incorporation of PEG in the enzyme assay medium led to decreased affinity of enzyme towards its substrate with increase in Km. This loss in affinity was partially restored by the addition of 1 M proline in the assay medium, suggesting the possible protective role of proline on RNase under osmotic stress. Because of its prolific growth, oilseed rape (Brassica napus L.) can be grown advantageously for phytoremediation of the lands contaminated by industrial wastes. Therefore, toxic effect of cadmium on the germination of oilseed rape, the capability of plants for cadmium phytoextraction, and the effect of exogenous application of plant growth regulators to mitigate phytotoxicity of cadmium were investigated. For the lab study of seedlings at early stage, seeds were grown on filter papers soaked in different solutions of Cd2+ (0, 10, 50, 100, 200 and 400 μM). In greenhouse study, seedlings were grown in soil for 8 weeks, transferred to hydroponic pots for another 6 weeks growth, and then treated with plant growth regulators and cadmium. Four plant growth regulators viz. jasmonic acid (12.5 μM), abscisic acid (10 μM), gibberellin (50 μM) and salicylic acid (50 μM); and three levels of Cd2+ (0, 50 and 100 μM) were applied. Data indicated that lower concentration of Cd2+ (10 μM) promoted the root growth, whereas the severe stresses (200 or 400 μM) had negative effect on the establishment of germinating seedlings. Plants treated with any of the tested plant growth regulators alleviated cadmium toxicity symptoms, which were reflected by more fresh weight, less malondialdehyde concentration in leaves and lower antioxidant enzyme activities. The application of abscisic acid to the plants cultivated in the medium containing 100 μM Cd2+ resulted in significantly lower plant internal cadmium accumulation. Two chickpea cultivars PBG-1 and PDG-3 along with a wild species Cicer judaicum were investigated to compare the activities of their antioxidant enzymes in mature seeds and roots, as well as shoots and cotyledons of seedlings germinated under dark and continuous illumination of 40 μmol m−2 s−1 photosynthetically active radiation (PAR). Seedling biomass of C. judaicum was lower as compared to cultivars of PBG-1 and PDG-3 both under dark and light conditions. Light reduced the biomass of seedlings. Activities of glutathione reductase (GR) and ascorbate peroxidase (APX) were higher in shoots and roots of C. judaicum compared to the cultivars PBG-1 and PDG-3. In mature seeds, the activities of GR and APX were higher in the cultivated genotypes whereas catalase (CAT) and peroxidase were higher in C. judaicum. Under illumination, a general upregulation of CAT in both shoots and cotyledons and of GR in shoots was observed in all the three genotypes. However, superoxide dismutase (SOD) increased in C. judaicum and APX in PBG-1 and PDG-3. The differences in antioxidant enzyme system between wild and cultivated genotypes possibly contribute to better tolerance of wild Cicer species against abiotic and biotic stresses.  相似文献

Effects of Cd2+ on growth and Cd-binding complex formation in roots were examined with various seedlings of mono- and dicotyledonous plants. Maize, oat, barley and rice exhibited the greater tolerance to Cd2+ (100 μM) than did azuki bean, cucumber, lettuce, pea, radish, sesame and tomato (10–30 μM). Azuki bean was the most sensitive to Cd2+ (<10 μM). Under these Cd-treatments, cereal roots accumulated Cd2+ in the cytoplasmic fractions and transported Cd2+ into the same fractions of shoot tissues, to larger extents than did dicotyledonous roots. Cereal roots synthesized a Cd-binding complex containing phytochelatins in the cytoplasmic fractions, depending upon Cd2+ concentrations applied (30–100 μM). Such a complex was not detected from the same fractions of dicotyledonous roots treated with Cd2+. These results suggest that the Cd-binding complex formation has an important role in the tolerance of cereal roots against Cd2+.  相似文献

In the present study Prosopis juliflora plants grown in hydroponics solution were exposed to 50,100 and 1000 μM CdCl2. The cadmium uptake, transport and toxicity on the photosynthetic activities in the plants were measured at 48 h after starting cadmium treatments. The results showed that the concentration of Cd2+ in P. juliflora tended to increase with addition of Cd2+ to hydroponics solution. However, the increase of Cd2+ in roots and leaves varied largely. In this sense, the accumulation of Cd2+ in P. juliflora roots increased significantly in proportion with the addition of this metal. In contrast a relatively low level of Cd2+ transportation index, and bioaccumulation factor were found in P. juliflora at 48 h after of treatments. On the other hand the maximum photochemical efficiency of photosystem II (Fv/Fm) and the activity of photosystem II (Fv/Fo) ratios in P. juliflora leaf treated with Cd2+ not showed significantly changes during the experiment. These results suggested that the photosynthetic apparatus of P. juliflora was not the primary target of the Cd2+ action. Further studies will be focused in understanding the participation of the root system in Prosopis plants with the rhizosphere activation and root adsorption to soil Cd2+ under natural conditions.  相似文献
