Production of oilseed rape, Brassica napus L., is affected by various insect pests. The cabbage stem weevil, Ceutorhynchus pallidactylus (Mrsh.) (Col.: Curculionidae), is one of the most damaging pests in Northern and Central Europe that requires regular control measures. Host plant resistance is a key factor in integrated pest management systems. To evaluate a large number of genotypes for their susceptibility to infestation by C. pallidactylus, new screening techniques were developed for testing both, the amount of feeding and the number of eggs deposited by adult C. pallidactylus on accessions of Brassicaceae under controlled conditions. In no‐choice screening tests, the leaf area consumed by adult cabbage stem weevil was quantified on a wide spectrum of 107 brassicaceous genotypes (B. napus, Brassica rapa L. and Brassica oleracea L. cultivars, breeding lines, resynthesized rapeseed lines and wild Brassicaceae). In comparison to feeding on the standard cultivar ‘Express’, the average leaf area consumed by C. pallidactylus on nine oilseed rape cultivars, four resynthesized rapeseed lines and five other accessions [B. oleracea, Camelina alyssum (Mill.) and Lunaria annua L.] was significantly reduced by 44–90%. In dual‐choice screening tests for the evaluation of oviposition preferences on 42 genotypes, female C. pallidactylus laid significantly fewer eggs into plants of two oilseed rape cultivars, five resynthesized rapeseeds and three accessions of B. oleracea and Brassica fruticulosa Cyrillo, respectively, than into plants of the standard cv ‘Express’. Results of both laboratory screening tests were confirmed by results of additional field testing.  相似文献

The cabbage stem flea beetle (CSFB), Psylliodes chrysocephala L. (Coleoptera: Chrysomelidae), is one of the most important pests in European winter oilseed rape production. Adult beetles feed on young leaves whereas larvae mine within the petioles and stems. Larval infestation can cause significant crop damage. In this study, the host quality for CSFB of four oilseed rape (Brassica napus L.) cultivars and seven other brassicaceous species with different glucosinolate (GSL) profiles was assessed under controlled conditions. Larval instar weights and mortality were measured after 14 and 21 days of feeding in the petioles of test plants. To study the impact of GSL on the performance of larvae, the GSL contents in petioles from non-infested and infested plants were analysed before, and 21 days after, the start of larval infestation. Larval performance was not significantly different between the four cultivars of oilseed rape, but differed considerably among the other brassicaceous species tested. In comparison to the weight of larvae in the standard B. napus cv. Robust, the larval weight was higher in turnip rape (Brassica rapa L. var. silvestris) and significantly reduced in white mustard (Sinapis alba L.), oil radish (Raphanus sativa L. var. oleiformis), and cabbage (Brassica oleracea L. convar. capitata var. alba). The duration of larval development increased in white mustard and oilseed radish. The GSL profiles of the petioles showed little difference between non-infested and infested plants of oilseed rape whereas the content of aliphatic GSL increased in the infested turnip rape plants. In contrast, the aliphatic and benzenic GSL decreased in infested Indian rape (B. rapa subsp. dichotoma Roxb.). Larval weight was not correlated with the total GSL content of plants, neither before infestation nor 21 days after. Larval weight was positively correlated with progoitrin and 4-hydroxyglucobrassicin. White mustard, which provides inferior host quality for larval development, has the potential to introduce insect resistance into high-yielding oilseed rape cultivars in breeding programmes.  相似文献

Pollen beetle, Meligethes aeneus (Fabricius) (Coleoptera: Nitidulidae), and cabbage seed weevil, Ceutorhynchus assimilis (Paykull) (Coleoptera: Curculionidae), are important pests in the production of European winter oilseed rape, Brassica napus L. (Brassicaceae), which is grown on several million hectares in Europe. Insecticide treatments are common to control both pests once they exceed economic damage thresholds; however, not many chemical classes are available for their control in European oilseed rape. Particularly pollen beetles recently developed high levels of pyrethroid resistance impairing field control at recommended rates in many countries, whereas no resistance is yet reported to another important insecticide, thiacloprid. The major objective of this study was to investigate the spatio‐temporal susceptibility status of pollen beetle against the recently introduced insecticide thiacloprid. From 2009 to 2012 more than 630 populations of pollen beetle collected in 13 countries were monitored for resistance to thiacloprid by using an adult vial test. No shifting to lower susceptibility of pollen beetle to thiacloprid has been observed between 2009 and 2012. Furthermore, we were able to show that pollen beetle larvae are extremely susceptible to thiacloprid, whereas within strains larvae are significantly more resistant than adults to pyrethroids such as lambda‐cyhalothrin. Dose‐response data for thiacloprid against cabbage seed weevil populations collected in 2011 in Germany, Sweden, and Ukraine showed a 10‐fold higher intrinsic sensitivity compared to pollen beetle, and showed only a low variation in response. In addition, we also tested 17 cabbage seed weevil populations collected in five countries against lambda‐cyhalothrin with low variation in response (three‐fold), suggesting full baseline susceptibility and no resistance to pyrethroids. The implications of the data presented for resistance management in coleopteran pests in winter oilseed rape are discussed.  相似文献

The cabbage seed weevil, Ceutorhynchus assimilis Payk. [syn. Ceutorhynchus obstrictus (Marsham)] (Coleoptera: Curculionidae), a crucifer-feeding insect, is a pest of oilseed rape (Brassica napus L.). It is known to be attracted by isothiocyanates, crucifer-specific volatiles that are metabolites of the glucosinolates. The responses of this insect to other electrophysiologically-active volatiles from rape were tested in a linear track olfactometer. Attraction was demonstrated to nitriles (phenylacetonitrile, 4-pentenenitrile and 5-hexenenitrile), which are also glucosinolate metabolites, and to volatiles emitted by a wider spectrum of plant families ((Z)-3-hexen-1-ol and methyl salicylate). Combination of an isothiocyanate mixture with phenylacetonitrile increased attraction, but there was no such increase when the isothiocyanate mixture was combined with methyl salicylate. A mixture of 23 volatiles, emulating an attractive air-entrainment extract of oilseed rape, was not significantly attractive, although a high proportion of weevils (60%) turned towards it. The potential of these volatiles for inclusion into an isothiocyanate-based monitoring system is discussed.  相似文献

Abstract The cabbage seed weevil ( Ceutorhynchus assimilis Payk.) lays eggs singly into pods of oilseed rape ( Brassica napus L.) through punctures bored with the mouthparts, preferring pods not recently used for oviposition. A simple new choice test has been used to test individual components of egg-laying behaviour for their effect on oviposition site selection. It is confirmed that an oviposition-deterring pheromone (ODP) is deposited during abdomen brushing of the pod which follows egg-laying. Neither pin punctures, weevil feeding punctures, oviposition punctures nor eggs had any deterrent effect. Pods walked on by female weevils were not avoided by those laying eggs. Observations suggest that the ODP is sensed by contact chemoreceptors on the antennae. The deterrent effect lasted only 1–2 h. The implications of these findings on the adaptive significance of the pheromone and its possible use in pest control are discussed.  相似文献

Concerns about the negative effects of chemical control of oilseed rape (Brassica napus L.) pests on non-target species, human safety, and development of insecticide resistance, require alternative control strategies such as the use of trap crops and biocontrol to be developed. Psylliodes chrysocephala(L.) (Coleoptera: Chrysomelidae) (cabbage stem flea beetle) and Ceutorhynchus pallidactylus (Marsh.) (Coleoptera: Curculionidae) (cabbage stem weevil) are two major stem-mining pests of oilseed rape. This study investigated the phenology of these pests and their main parasitoids in the UK, the potential use of turnip rape (Brassica rapa L.) as a trap crop to reduce oilseed rape infestation, and the effects of insecticide treatment on pest incidence and larval parasitism. Water trap samples, plant dissections and pest larval dissections were done to determine: the incidence of adult pests and their parasitoids, the level of plant infestation by the pests and percentage larval parasitism, respectively. The turnip rape trap crop borders reduced P. chrysocephalabut not C. pallidactylus infestation of oilseed rape plots. Treatment of the trap crop with insecticide had little effect on either pest or parasitoid incidence in the oilseed rape. TersilochusmicrogasterSzép. andT. obscurator Aub. (Hymenoptera: Ichneumonidae) were the main larval parasitoids of P. chrysocephalaand C. pallidactylus, respectively. Tersilochus microgasteris reported for the first time in the UK. The implications for integrated pest management are discussed.  相似文献

Diacylglycerol acyltransferase (EC 2.3.1.20) activity was assayed during the maturation of seeds of oilseed rape (Brassica napus L.) and safflower (Carthamus tinctorius L.). Developmental studies were also conducted with microspore-derived embryos of oilseed rape (B. napus L. cv Topas) and an embryogenic microspore-derived cell-suspension culture of winter oilseed rape (B. napus L. cv Jet Neuf). In the maturing seeds, diacylglycerol acyltransferase activity increased to a maximum during rapid accumulation of lipid and declined, thereafter, with seed maturity. In microspore-derived embryos of oilseed rape (cv Topas), high levels of diacylglycerol acyltransferase activity were found throughout the early torpedo to late cotyledonary developmental stages with maximum enzyme specific activity associated with the mid-cotyledonary developmental stage. The cell-suspension culture of winter oilseed rape (cv Jet Neuf) contained 3 to 4% triacylglycerol on a dry weight basis and represented about half of the total lipid. The fatty acid profile of total lipid and triacylglycerol in the cell-suspension culture was similar in samples taken during a 1-year period. The Jet Neuf culture contained diacylglycerol acyltransferase with specific activity similar to that of Topas microspore-derived embryos. Jet Neuf diacylglycerol acyltransferase also displayed an enhanced specificity for erucoyl-CoA over oleoyl-CoA when assayed with 14 [mu]M acyl-coenzyme A in the reaction mixture. The specific activity of diacylglycerol acyltransferase in homogenates prepared from the Jet Neuf culture ranged from 5 to 15 pmol of triacylglycerol min-1 mg-1 of protein when assayed at intervals during a period of 1 year. Thus, the cell-suspension culture may represent an attractive tissue source for purification and characterization of triacyl-glycerol biosynthetic enzymes.  相似文献

The endogenous levels of ABA were measured in Agrobacterium rhizogenes A4 Tl-DNA transformed oilseed rape (Brassica napus L. var. oleifera cv . Brutor and cv. Drakkar), cabbage (Brassica oleracea) . A4 transformed tobacco ( Nicotiana tabacum cv. Xanthi) and their normal counterparts, using high performance liquid chromatography and enzyme-liked immunosorbent assay. Measurements were made on different plant tissues (i. e. floral stem, terminal bud, young leaf, mature leaf, root and root tips) and ABA levels were compared in unstressed and osmotically stressed oilseed rape plants (cv. Brutor). In unstressed Plants. in each of the 5 independent transformation events studied, a significant reduction (about 65% of control) in ABA concentration was observed in all transformed plants. When subjected to an osmotic stress, TL transformed Brutor plants showed a higher ABA accumulation than untransformed plants. The change in ABA content as a consequence of TL-DNA transformation is discussed with regard to phenotype, drought resistance and adaptability.  相似文献

Quantitative trait loci (QTLs) contributing to yellow seed colour and acid detergent fibre (ADF) were localized and compared in 3 mapping populations developed from 2 crosses (designated 'YE1' and 'YE2') between 2 distinct sources of true-breeding yellow-seeded oilseed rape (Brassica napus) and 2 different black-seeded genotypes. A clear correlation was observed between seed colour and ADF content in both crosses. In all 3 populations, a major QTL, with a large effect on both seed colour and ADF in multiple environments, was detected at the same position on chromosome N18. In YE1, a second minor QTL, with a small effect on seed colour but not on ADF content, was localized on chromosome N1. In YE2, no QTL was observed on N1; however, 2 minor seed-colour loci were localized to N15 and N5. A second major QTL for ADF was localized in YE1 on N13; in YE2, no other QTLs for ADF were detected. Combined QTL and segregation data for seed colour and ADF content in the different populations suggest that a partially dominant B. napus gene for seed colour on N18 contributes to a reduction in fibre content in different yellow-seeded B. napus genotypes. The other QTLs that were identified appear to represent different genes in the 2 yellow-seeded rapeseed sources, which, in each case, affect only fibre content or seed colour, respectively. Potential candidate genes and implications for marker-assisted breeding of oilseed rape with reduced seed dietary fibre content are discussed.  相似文献

Unlike most studies on hybridisation between oilseed rape and Brassica rapa, this study focused on hybridisation with oilseed rape as the maternal parent. This is a key cross because, assuming that plastids are inherited maternally, F(1)-hybrid production with maternal oilseed rape (B. napus) is the only transgene escape route from transplastomic oilseed rape. We investigated such F(1)-hybrid production in winter oilseed rape co-cultivated with weedy B. rapa at three plant densities each with two proportions of the different species. The paternity of the progeny produced on oilseed rape was assessed, and several fitness parameters were determined in oilseed rape mother plants in order to correlate hybridisation and plant competition. At higher density, the vegetative fitness per mother plant decreased significantly, but the density only affected the frequency of F(1)-hybrids significantly (a decrease) in the treatment with equal proportions of each species. As to the proportions, at higher B. napus frequencies, there were fewer F(1)-hybrids per mother plant and a significant increase in most biomass components. Thus, B. rapa was the stronger competitor in its effect on both the vegetative and reproductive fitness in B. napus, and the hybridisation frequency. In conclusion, the relative frequency of the two species was a more influential parameter than the density. Hybridisation with B. napus as the female will be most likely at current field densities of B. napus and when B. rapa is an abundant weed.  相似文献

The impact of osmotic stress on growth, physiology, and metabolism of winter oilseed rape(Brassica napus L.)was investigated by detailed analysis of biomass traits,hormone metabolites and osmolytes in two genetically unrelated drought-tolerant genotypes and two unrelated drought-sensitive genotypes. Seedlings were grown in vitro under controlled conditions and osmotic stress was simulated by applying a gradual treatment with polyethylene glycol(PEG6000), followed by hypo-osmotic treatment of variants used for metabolite determination. The results provide a basis for the identification of reliable selection criteria for drought resistance in oilseed rape. The in vitro cultivation system established during this study enabled effective discrimination of early osmotic stress responses between drought-resistant and -susceptible oilseed rape genotypes that also show large differences in relative seed yield under drought conditions in the field. Clear physiological and metabolic differences were observed between the drought-resistant and drought-sensitive genotypes, suggesting that osmotic adjustment is a key component of drought response in oilseed rape. Unexpected ly, however, the drought-resistant genotypes did not show typical hormonal adjustment and osmolyte accumulation suggesting that they possess alternative physiological mech anisms enabling avoidance of stress symptoms.  相似文献

1 The spatio‐temporal distributions of predatory carabid beetles (Coleoptera: Carabidae) and their potential prey, the larvae of three coleopterous pests, Meligethes aeneus (Fabricius) and Ceutorhynchus spp. [Ceutorhynchus pallidactylus (Marsham), the cabbage stem weevil, and Ceutorhynchus assimilis (Paykull), the cabbage seed weevil], were studied within a crop of winter oilseed rape. The distributions of Collembola were recorded as potential alternative prey. Insect distributions were analysed and compared using Spatial Analysis by Distance Indices. 2 Mature larvae of the pests dropped from the crop canopy to the soil for pupation in temporal succession from May to early July. Their distributions within the crop were irregular and differed with species. 3 Adults of seven species or genera of carabid were abundant and active within the crop during May and June: Nebria brevicollis (Fabricius), Anchomenus dorsalis (Pontoppidan), Loricera pilicornis (Fabricius), Amara similata (Gyllenhal), Asaphidion spp., Pterostichus madidus (Fabricius) and Pterostichus melanarius (Illiger). 4 During May, N. brevicollis was spatially associated with peak numbers of M. aeneus larvae and with Collembola. Anchomenus dorsalis was spatially associated with Ceutorhynchus spp. larvae during two peaks in the abundance of the latter in early and late June. Nebria brevicollis and A. dorsalis coincided in both time and space with larvae of the three coleopterous pests when they were most vulnerable to predation by epigeal predators and are therefore good candidates for conservation biocontrol. 5 The importance of carabid beeetles in the natural enemy complex in winter oilseed rape and their potential for biocontrol of spring and summer pests are discussed in relation to husbandry practices for the crop and its adjacent areas which could be manipulated to promote carabid survival for integrated pest management.  相似文献

以10个甘蓝型油菜、11个芥菜型油菜和7个白菜型油菜品种为材料,选用已报道的14对白菜EST-SSR引物和8对油菜EST-SSR引物,探索其在3种类型油菜中的通用性,并利用筛选出的在3类油菜中有通用性和多态性的EST-SSR标记对供试油菜品种进行聚类分析.结果表明:(1)选用的14对白菜EST-SSR引物和8对油菜EST-SSR引物,在供试的3种类型油菜中都有扩增,完全可用.(2)在白菜型、甘蓝型和芥菜型油菜品种中扩增显示出多态性的引物数分别为18、16 和14对,其中有10对引物在3种类型油菜品种间扩增产物具有多态性.(3)利用这10对扩增多态性EST-SSR引物对供试油菜品种聚类分析,结果显示,在遗传相似系数为0.67时,3种类型油菜品种分别独自聚为一大类,表明开发和建立不同类型油菜间可通用的EST-SSR标记是可行且有应用价值的.  相似文献

A range of crops have been transformed with delta-endotoxin genes from Bacillus thuringiensis (Bt) to produce transgenic plants with high levels of resistance to lepidopteran pests. Parasitoids are important natural enemies of lepidopteran larvae and the effects of Bt plants on these non-target insects have to be investigated to avoid unnecessary disruption of biological control. This study investigated the effects of Cry1Ac-expressing transgenic oilseed rape (Brassica napus) on the solitary braconid endoparasitoid Cotesia plutellae in small-scale laboratory experiments. C. plutellae is an important natural enemy of the diamondback moth (Plutella xylostella), the most important pest of brassica crops world-wide. Bt oilseed rape caused 100% mortality of a Bt-susceptible P. xylostella strain but no mortality of the Bt-resistant P. xylostella strain NO-QA. C. plutellae eggs laid in Bt-susceptible hosts feeding on Bt leaves hatched but premature host mortality did not allow C. plutellae larvae to complete their development. In contrast, C. plutellae developed to maturity in Bt-resistant hosts fed on Bt oilseed rape leaves and there was no effect of Bt plants on percentage parasitism, time to emergence from hosts, time to adult emergence and percentage adult emergence from cocoons. Weights of female progeny after development in Bt-resistant hosts did not differ between plant types but male progeny was significantly heavier on wildtype plants in one of two experiments. The proportion of female progeny was significantly higher on Bt plants in the first experiment with Bt-resistant hosts but this effect was not observed again when the experiment was repeated.  相似文献

In darkness, dormancy was imposed on seeds of lettuce (Lactuca sativa L. cv. Grand rapids) by high temperature and on seeds of oilseed rape (Brassica napus L. cv. Apex) by osmotic stress using polyethylene glycol (PEG 8000). In both cases, dormancy was broken by incubating the seeds in aqueous extracts of combustion products from Salix viminalis wood chips or Themeda triandra leaves. Dormancy of rapeseed, but not lettuce, was also broken by a solution of smoke from burnt straw of Triticum aestivum. The greatest stimulation from burnt vegetation was achieved with an aqueous extract of pyrolysed willow wood chips, which had been subjected to temperatures of up to 800 °C during combustion in a down-draught gasifier. This suggests that some biologically active substances obtained from combustion of plant tissues are highly heat-stable.  相似文献

A RFLP map of Brassica napus, consisting of 277 loci arranged in 19 linkage groups, was produced from genetic segregation in a combined population of 174 doubled-haploid microspore-derived lines. The integration of this map with a B. napus map derived from a resynthesized B. napus x oilseed rape cross allowed the 10 linkage groups of the B. napus A genome and the 9 linkage groups of the C genome to be identified. Collinear patterns of marker loci on different linkage groups suggested potential partial homoeologues. RFLP patterns consistent with aberrant chromosomes were observed in 9 of the 174 doubled-haploid lines. At least 4 of these lines carried nonreciprocal, homoeologous translocations. These translocations were probably the result of homoeologous recombination in the amphidiploid genome of oilseed rape, suggesting that domesticated B. napus is unable to control chromosome pairing completely. Evidence for genome homogenization in oilseed rape is presented and its implications on genetic mapping in amphidiploid species is discussed. The level of polymorphism in the A genome was higher than that in the C genome and this might be a general property of oilseed rape crosses.  相似文献

Replicated small plot field experiments were done at two sites growing winter oilseed rape (ADAS Boxworth, Cambridgeshire and ADAS High Mowthorpe, North Yorkshire) and two sites growing spring oilseed rape (ADAS Bridgets, Hampshire and ADAS Rosemaund, Herefordshire) to investigate the effect of cabbage aphid (Brevicoryne brassicae) on crop yield and quality. All four sites were included in the first 2 yr of the experiment in 1994 and 1995 but only those with winter oilseed rape were continued into the final year in 1996. Plots were artificially inoculated with cabbage aphids at either five aphid 4 m-2 or 5 aphids 16 m-2 or left uninoculated to become naturally infested. In 1995 and 1996 the naturally infested treatment was omitted. Sprays of the aphicide pirimicarb at GS 3.3, 3.7, 4.5, 4.9 and 5.5 were used to manipulate aphid populations. Once a plot had been treated at a target growth stage it was sprayed on all subsequent occasions to prevent recolonisation. Aphid numbers were assessed prior to each spray date and their effect on the crop measured in terms of yield of seed and oil and glucosinolate content. Artificial inoculation of aphids was often successful in establishing different populations of the pest at a range of growth stages. Results showed that cabbage aphid sometimes reduced both crop yield and quality. Yield responses to insecticide treatment tended to be larger in spring oilseed rape than in winter oilseed rape mainly because it became more heavily infested at an early growth stage. Tentative thresholds are proposed for control of the pest in both winter and spring oilseed rape. It is stressed that cabbage aphid is a sporadic pest and rarely likely to reach these threshold levels in field crops.  相似文献

A thorough understanding of the relationships between plants and pathogens is essential if we are to continue to meet the agricultural needs of the world's growing population. The identification of genes underlying important quantitative trait loci is extremely challenging in complex genomes such as Brassica napus (canola, oilseed rape or rapeseed). However, recent advances in next-generation sequencing (NGS) enable much quicker identification of candidate genes for traits of interest. Here, we demonstrate this with the identification of candidate disease resistance genes from B.?napus for its most devastating fungal pathogen, Leptosphaeria maculans (blackleg fungus). These two species are locked in an evolutionary arms race whereby a gene-for-gene interaction confers either resistance or susceptibility in the plant depending on the genotype of the plant and pathogen. Preliminary analysis of the complete genome sequence of Brassica rapa, the diploid progenitor of B.?napus, identified numerous candidate genes with disease resistance characteristics, several of which were clustered around a region syntenic with a major locus (Rlm4) for blackleg resistance on A7 of B.?napus. Molecular analyses of the candidate genes using B.?napus NGS data are presented, and the difficulties associated with identifying functional gene copies within the highly duplicated Brassica genome are discussed.  相似文献
