The aerial parts of Centaurea grisebachii ssp. grisebachii, a Greek endemic species belonging to section Acrolophus (Cass.) DC., were investigated for the occurrence of sesquiterpene lactones. The eudesmanolide, 4-epi-malacitenolide was the major compound. In addition to the sesquiterpene lactones, five flavonoid aglycons, two lignans, one nor-isoprenoid and one sterol were found.  相似文献

Relationships between two plant communities in the south-western Alps, high-altitude manured meadows (Triseto-Polygonion) and lower tall grass prairies (Adenostylion) have been examined by means of the flavonoid analysis of specimens belonging to six species common to those two associations. The species, collected at eight stations of the Ligurian Alps and the Ubaye valley, are: Trollius europaeus (Ranunculaceae), Chaerophyllum aureum (Umbellifereae), Silene cucubalus (Caryophyllaceae), Polygonum bistorta) (Polygonaceae), Alchemilla vulgaris (Rosaceae) and Centaurea montana (Compositae). In every species, factorial analysis of correspondence was applied to the distribution of flavonoid glycosides. This led to the partition of the populations into two groups according to the two sectors (Ligurian Alphs Ubaye valley) of the collection area. Secondly, populations of two species, Alchemilla vulgaris and Centaurea montana, exhibited further correlation of their flavonoid pattern with their phytosociological origin.  相似文献

(Asteraceae) 25 compounds (two pentacyclic triterpenes, one sterol glucoside, five quinic acid derivatives, one phenolic acid, and 16 flavonoids) were isolated and characterized. Aspartic proteinases are present in a variety of organisms including plants. Common features of aspartic proteases include an active site cleft that contains two catalytic aspartic residues, acid pH optima for enzymatic activity, inhibition by pepstatin A. Plant aspartic proteinases occur in seeds and may be involved in the processing of storage proteins. Many of them have been purified and characterized. The presence of aspartic proteases in seeds of Centaurea calcitrapa during germination was investigated by measuring the activity on enzyme extracts. The aspartic proteases are present mainly in the beginning of seed germination suggesting that they could initiate the degradation of protein reserves in germinating seeds.

These proteases were purified by salt precipitation followed by anion-exchange chromatography. Purified aspartic proteases have an optimal pH between 3.5 and 4.5, using FTC-hemoglobin as substrate and an optimal temperature at 52 °C. The ability of seed extracts for milk clotting was tested and the clotting time that was achieved is in the same range found for flower extracts appropriated for special cheeses in which weak clotting agents are required.  相似文献

Field studies were conducted to determine the competitive interactions between introduced biological control agents that attack the seed heads of spotted knapweed (Centaurea stoebe ssp. micranthos) and diffuse knapweed (Centaurea diffusa). Two weevils, Bangasternus fausti and Larinus minutus (Coleoptera: Curculionidae), were each paired with the previously established fly, Urophora affinis (Diptera: Tephritidae). Each species was released either alone or in pair-wise combinations inside screen cages placed over existing knapweed plants at six field sites in Montana and one in Oregon. Larinus minutus produced almost three times as many progeny on diffuse knapweed as on spotted knapweed. Larinus minutus reproduction was not affected by competition with U. affinis, but U. affinis reproduction was reduced by the presence of L. minutus (by 71% on spotted and 77% on diffuse knapweed). Bangasternus fausti reproduction generally was not affected by competition with U. affinis, nor was U. affinis affected by B. fausti on either host plant. There were extremely few cases of successful production of both weevil and fly in the same capitulum, which was probably because weevil larvae consume the developing flies. Both weevils increased the total proportion of seed heads infested on diffuse knapweed, and B. fausti increased it on spotted knapweed. However, the release of either weevil did not significantly further reduce seed production on either plant. The results and experimental design are discussed in light of the subsequent establishment and impact of these agents.  相似文献

Genetic engineering has improved the product yield of a variety of compounds by overexpressing, inactivating, or introducing new genes in microbial systems. The production of flavor-enhancing ester compounds is an emerging area of heterologous gene expression for desired product yield in Escherichia coli. Isoamyl acetate, butyl acetate, ethyl acetate, and butyl butyrate are reported here to be produced by expressing Saccharomyces cerevisiae genes ATF1 or ATF2 and the strawberry gene SAAT in E. coli when the appropriate substrates are provided. Increasing the concentration of alcohol added to the reaction generally resulted in increased ester production. ATF1 expression was found to produce more isoamyl acetate and butyl acetate than ATF2 expression or SAAT expression in the strains and culture conditions examined. Additionally, SAAT expression resulted in greater isoamyl acetate and butyl acetate production than ATF2 expression. Butyl butyrate is produced by cell-free extracts of E. coli harboring SAAT but not ATF1 or ATF2.  相似文献

Species of Colletotrichum cause diseases on a wide range of hosts, frequently infecting plants in the Agavaceae (monocotyledons: Liliales). Three species of Colletotrichum restricted to the Agavaceae were detected through morphological studies of specimens and molecular sequence analyses of the LSU of the nu-rDNA and the ITS region of the nu-rDNA from cultures. Colletotrichum agaves on Agave is fully described and illustrated. Colletotrichum dracaenophilum is described as a new species for isolates having long conidia and occurring on Dracaena sanderiana from China. Colletotrichum phormii and Glomerella phormii are determined to be the correct scientific names for the asexual and sexual states, respectively, of a species commonly referred to as C. rhodocyclum and G. phacidiomorpha occurring mainly on Phormium. In addition, C. gloeosporioides and C. boninense were isolated from plants in the Agavaceae. All species of Colletotrichum described on Agavaceae were evaluated based on type specimens. A key to the five species of Colletotrichum on Agavaceae is included. This paper includes one new species, Colletotrichum dracaenophilum, and three new combinations, Colletotrichum phormii, Glomerella phormii, and Phaeosphaeriopsis phacidiomorpha.  相似文献

The flavonoid profiles of Astilbe (four taxa studied) and Rodgersia (two taxa studied) are based on simple flavonol glycosides. Astilbe has 3-O-mono-, 3-O-di-, and 3-O-triglycosides of kaempferol, quercetin, and myricetin, while Rodgersia has only mono- and diglycosides of kaempferol and quercetin. Astilbe×arendsii was also shown to accumulate dihydrochalcone glycosides. The flavonoid profile of Rodgersia is the simplest recorded so far in the herbaceous Saxifragaceae. The flavonoids of two species of Aruncus were shown to be based upon kaempferol and quercetin 3-O-mono- and 3-O-diglycosides. One of the species also exhibited an eriodictyol glycoside. The triglycoside differences were not considered important, but the differences in myricetin occurrences were taken as evidence against derivation of Saxifragaceae from an Aruncus-like ancestor. Should such an event be proposed, however, serious consideration would have to be given to the current pattern of myricetin occurrence in the two families.  相似文献

Ray flower and leaf flavonoids were investigated for the three species of Helianthus series Microcephali. Ray flowers of all species contain coreopsin, sulphurein, and quercetin 7-O-glucoside; those of H. microcephalus also contain quercetin 3-O-glucoside. A mixture of flavonoid aglycones, mostly methoxylated flavones, occurs in leaves of H. microcephalus, but not in H. glaucophyllus or H. laevigatus which also lack the glandular trichomes that in Helianthus are typically associated with flavonoid aglycones. The presence of compounds with the 6,8,4′ pattern of methoxylation in H. microcephalus suggests that the series is more similar in flavonoids to series Angustifolii than to series Corona-solis.  相似文献

Leaf and ray flower flavonoids were investigated for the seven species of Helianthus series Angustifolii. Flavone aglycones occur in small glandular trichomes located on leaf undersurfaces of H. angustifolius, H. floridanus and H. simulans. Other species lacked both glandular trichomes and flavone aglycones. Flavonol glycosides occur in low concentrations in leaves of all species but were not characterized. Anthochlors (chalkones) occur in leaves of H. heterophyllus and H. longifolius. Ray flower flavonoids include anthochlor and flavonol glycosides and occur in the basal region of the ligule producing a band of UV A around the head. Anthochlors are the predominant ray flower flavonoids in H. angustifolius, H. heterophyllus and H. longifolius, whereas they are absent and quercetin 7-glucoside is present in H. carnosus and H. floridanus. Cladistic analysis of flavonoid and morphological characters indicates that evolution in the series has been a radiation from ancestral types rather than a linear sequence of progressively more derived species.  相似文献

The Teloschistaceae is a widespread family with considerable morphological and ecological heterogeneity across genera and species groups. In order to provide a comprehensive molecular phylogeny for this family, phylogenetic analyses were carried out on sequences from the nuclear ribosomal ITS region obtained from 114 individuals that represent virtually all main lineages of Teloschistaceae. Our study confirmed the polyphyly of Caloplaca, Fulgensia and Xanthoria, and revealed that Teloschistes is probably non-monophyletic. We also confirm here that species traditionally included in Caloplaca subgenus Gasparrinia do not form a monophyletic entity. Caloplaca aurantia, C. carphinea and C. saxicola s. str. groups were recovered as monophyletic. The subgenera Caloplaca and Pyrenodesmia were also polyphyletic. In the subgenus Caloplaca, the traditionally recognized C. cerina group was recovered as monophyletic. Because this study is based solely on ITS, to maximize taxon sampling, the inclusion of phylogenetic signal from ambiguously aligned regions in MP (recoded INAASE and arc characters) resulted in the most highly supported phylogenetic reconstruction, compared with Bayesian inference restricted to alignable sites.  相似文献

A DNA fragment was identified and cloned from Staphylococcus epidermidis (Se) using femA from S. aureus (Sa) as a heterologous hybridization probe. DNA sequence analysis of a portion of this clone revealed two complete ORFs highly related to femA and femB of Sa. The genomic arrangement of the Se femA/B complex was nearly identical to that observed in Sa. Intra- and interspecies relatedness of these genes and conservation of genomic organization were consistent with gene duplication of one of these genes in an ancestral organism. Recombinant FEMA, produced in Escherichia coli (Ec), was purified to near homogeneity. Identity of the purified protein was verified by N-terminal amino acid (aa) sequence analysis.  相似文献

The deleterious effects of temperature-induced coral bleaching, a process by which corals lose their endosymbiotic algae (zooxanthellae; genus Symbiodinium) primarily at temperatures above mean yearly maximums, has not been well described for alcyonacean soft corals (Coelenterata, Octocorallia). The study of Symbiodinium cells lost from Sarcophyton ehrenbergi, Sinularia sp., and Xenia sp., which have not been compared in bleaching studies, indicate that the soft coral S. ehrenbergi released the greatest number of symbiont cells, however, it was less susceptible to heat stress surviving temperatures of 34 °C for >39 h. Sinularia sp. showed intermediate levels of bleaching tolerance to elevated temperatures, surviving prolonged exposures at 32 °C, but dying within 24 h at 34 °C. Xenia sp., however, was the most vulnerable to high heat stress maximally releasing Symbiodinium at temperatures ≤30 °C. This evidence indicates that Xenia sp. is even more susceptible to elevated temperatures than Acropora spp., previously reported to be the most vulnerable coral species to elevated temperature-induced bleaching.

Molecular analysis showed that the more resistant soft coral species (S. ehrenbergi) had the same type of Symbiodinium (clade C) as less resistant soft corals (Xenia sp.). In comparison to scleractinian corals collected from the same region that show similar bleaching resistance to high temperatures (e.g. Porities solida—more robust; Favites complanata—moderate resistance; Acropora hyacinthus—less robust), all scleractinian corals were symbiotic with Symbiodinium from clade C. A. hyacinthus, however, was found to possess multiple symbionts (clades B and C), and this represents a first report of Clade B in any Acropora species.  相似文献

The objective of this work was to study the partial consumption of malic acid in a semi-synthetic medium by Schizosaccharomyces yeast under wine-making conditions. The aim was to control the extent of deacidification by stopping the activity of Schizosaccharomyces by the addition of Saccharomyces yeast at different delayed intervals of time. This was thought possible because the Schizosaccharomyces yeasts are much more slower than the Saccharomyces yeasts and may be inhibited by the latter. In fact, the opposite was observed. Schizosaccharomyces exhibited a amensal effect against Saccharomyces: the Saccharomyces growth was inhibited by Schizosaccharomyces, this inhibition being proportional to the Schizosaccharomyces concentration, reversible, and sensitive to temperature. Such interactions similar to a killer effect have not been described before. At the same time a competition towards sugar was noted, being negative for Schizosaccharomyces since these yeasts have slower metabolic rates. Nevertheless, total demalication was nearly always possible because sufficient concentrations of Schizosaccharomyces were always reached. The delayed inoculation of Saccharomyces after Schizosaccharomyces was not a good solution to obtain partial deacidification.  相似文献

1. The absorption spectrum of chlorophyll b in vivo at 77°K is presented as the difference spectrum between preparations of spinach and chlorophyll b-free Vischeria stellata chloroplasts.

2. A shoulder on this spectrum around 662 nm is due to a component different from chlorophyll b. This component may well be identical with the chlorophyll a form, chlorophyll a (665).

3. The 77°K chlorophyll b absorption spectra in the nonfractionated photosyn-thetic pigment apparatus and in fractions mainly representing Photosystems 1 or 2 are not significantly different.

4. The aerobic irreversible photobleaching of chlorophyll b was studied in the intact pigment complex as well as in fractions mainly consisting of Photosystem 1 or 2. A two-step photobleaching was observed in all cases. The time-course of this bleaching was not significantly different for chlorophyll b in both fractions.

A laboratory experiment at 20±5℃ shows that the water extract of fresh Nerium indicum had an obvious effect on killing Oncomelania hupensis. Treated with 0.1% water extract for four days, the mortality of O. hupensis was up to 100%. The effect of different tissues of N. indicum on O. hupensis was in order of stem phloem>leaf>root phloem>flower. The effect of N. indicum on O. hupensis was about ten times higher than that of Pterocarya stenoptera and Rumex japonicus, and was equal to that of 1×10-3mg·L-1 niclosamidum.  相似文献

Immunoperoxidase staining of muscle infected with Trichinella spiralis for murine collagen types I and IV provided both qualitative and quantitative evidence of extensive synthesis of both types of collagen by fibroblasts in infected muscle compared to that seen uninfected muscle. Moreover, fibroblasts in muscle infected with T. pseudospiralis, a nonencapsulating species, showed significantly less staining for both types of collagen compared to muscle from mice infected with T. spiralis. Analysis of collagen composition of isolated nurse cells using an ELISA specific for either type I or type IV murine collagen suggested that of these 2 types of collagen, only type IV basement membrane collagen is found in Trichinella capsular collagen. Excretory/secretory products of T. spiralis and T. pseudospiralis induced extensive synthesis of exclusively type IV collagen by 3T3 murine fibroblasts in vitro.  相似文献
