The objectives of this experiment were to compare estrous synchronization responses and AI pregnancy rates of beef heifers using protocols that included either CIDR or MGA as the progestin source. The hypotheses tested were that: (1) estrous synchronization responses after (a) progestin removal, and (b) PGF(2alpha); and, (2) AI pregnancy rates, do not differ between heifers synchronized with either progestin source. At the start of the experiment (Day 0) in both years, heifers were assigned randomly to receive, MGA supplement for 14 days (MGA-treated; n=79) or CIDR for 14 days (CIDR-treated; n=77). On Day 14 progestin was removed and heifers were observed for estrus up to and after PGF(2alpha) on Days 31 and 33 for CIDR-treated and MGA-treated heifers, respectively. Heifers that exhibited estrus within 60h after PGF(2alpha) were inseminated by AI 12h later; the remaining heifers were inseminated at 72h after PGF(2alpha) and given GnRH (100mug). More (P<0.05) CIDR-treated heifers exhibited estrus within 120h after progestin removal than MGA-treated heifers. Intervals to estrus after progestin removal were shorter (P<0.05) for CIDR-treated heifers than MGA-treated heifers. More (P<0.05) CIDR-treated heifers exhibited estrus and were inseminated within 60h after PGF(2alpha) than MGA-treated heifers. Pregnancy rates did not differ (P>0.10) between MGA-treated (66%) and CIDR-treated (62%) heifers. In conclusion, the use of CIDR as a progestin source in a 14-day progestin, PGF(2alpha), and timed AI and GnRH estrous synchronization protocol was as effective as the use of MGA to synchronize estrus and generate AI pregnancies in beef heifers.  相似文献

In Experiment 1, all cattle were fed MGA (0.5 mg/head/d) for 7 d (designated Days 0 to 6) and given PGF on Day 6. One-half were administered estradiol valerate (EV; 5 mg, im) on Day 0. At Location 1, a higher proportion (P < 0.005) of EV-treated heifers were detected in estrus and bred by AI between Days 7 and 13 than control heifers not receiving EV (27 of 33 versus 15 of 32), but the number of pregnancies (12 vs 10) was not significantly different. Eighty-three of 104 EV-treated and 89 of 106 control cows were inseminated, resulting in 50 and 45 pregnancies, respectively (not significant). At Location 2, cattle were similarly treated and exposed to bulls on Days 7 to 49. Fall pregnancy rate was higher (P < 0.015) for EV-treated than control heifers (44 of 48 vs 33 of 46), but was not significantly different for cows (22 of 26 vs 19 of 23). In Experiment 2, estradiol 17beta (E17beta; 5 mg, im) and progesterone (100 mg, im) were administered on Day 0 (instead of EV). In a third group (designated the PGF group), cattle were bred on Days 0 to 6, and PGF was administered on Day 6 to those not yet bred. For 213 cows, the percentage pregnant to a synchronized estrus was greater in the PGF group (72%) than in either the control group treated with MGA (49%; P = 0.005) or the group receiving MGA and E17beta (54%; P < 0.025). Fall pregnancy rates were 91, 89, and 96% for the 213 cows (not significant) and 89, 93, and 98% for 131 heifers (not significant) in the PGF, MGA and E17beta groups, respectively. In cattle without a functional CL, the average diameter of the largest follicle at Day 6 was 1 to 2 mm smaller in the E17beta + MGA group than in the MGA group (difference significant only in cows at Location 1). Combined for both locations, the synchronized pregnancy rate in heifers without a functional CL on Day 6 was higher (P < 0.05) in the E17beta + MGA group than in the MGA group (11 of 21, 52% versus 4 of 20, 20%). Estrogen treatment caused regression of ovarian follicles with emergence of a new follicular wave. Including estrogen in an estrus synchronization program utilizing MGA and PGF significantly increased fall pregnancy rate in heifers (at 1 location) and the synchronized pregnancy rate of heifers without a functional CL at the time of PGF treatment (combined for both locations).  相似文献

The objective was to determine the efficacy of a previously used CIDR or melengestrol acetate (MGA; 0.5mg/head/day) for resynchronization of estrus in beef heifers not pregnant to timed-AI (TAI). In three experiments and a field trial, heifers were reinseminated 6-12 h after first detection of estrus. Pregnancy diagnosis was done from approximately 25-43 days after either TAI or reinsemination. In Experiment 1, 79 heifers received a once-used CIDR from 13 to 20 days after TAI and 80 heifers were untreated controls. For these two groups, there were 34 and 35 heifers, respectively, not pregnant to TAI; median +/- S.E. intervals from TAI to onset of estrus were 22 +/- 0.2 days versus 20 +/- 0.6 days (P < 0.001); estrus rates were 70.6% versus 85.7% (P = 0.1); conception rates were 62.5% versus 76.7% (P < 0.3); and pregnancy rates were 44.1% versus 65.7% (P = 0.07), for CIDR and untreated (control) groups, respectively. In Experiment 2, heifers (n = 651) were TAI (Day 0) and 13 days later randomly assigned to one of seven groups (n = 93 per group) to receive a once-used CIDR (three groups; Days 13-20), MGA (three groups; Days 13-19), or no treatment (control group). Groups given a CIDR or MGA also received: no further treatment (CIDR or MGA alone); 1.5mg estradiol-17beta (E-17beta) and 50 mg progesterone (P4) in 2 mL canola oil on Day 13; or E-17beta and P4 on Day 13 and 0.5 mg E-17beta on Day 21 (24 h after CIDR removal or 48 h after the last feeding of MGA). Pregnancy rate to TAI was lowest (P < 0.05) for the group given a CIDR plus E-17beta and P4 on Day 13 and E-17beta on Day 21. Variability in return to estrus was greater (P < 0.001) in the control and MGA groups than in CIDR groups. Conception and pregnancy rates in heifers given a CIDR (65.1 and 61.4%) were higher (P<0.01) than those fed MGA (49.6 and 40.4%), but not different from controls (62.2 and 54.9%, respectively). In Experiment 3, 616 heifers received a once- or twice-used CIDR for 7 days, beginning 13+/-1 days after TAI, with or without a concurrent injection of 150 mg of P4 (2 x 2 factorial design). Pregnancy rate to TAI was 47.2%. In heifers that returned to estrus, there was no significant difference between a once- or twice-used CIDR for rates of estrus (68.8%, P < 0.3), conception (65.9%, P < 0.6) and pregnancy (45.3%, P < 0.8). Injecting progesterone at CIDR insertion increased the median interval from CIDR removal to onset of estrus (P < 0.05) and reduced rates of estrus (63.8% versus 73.8%, P<0.05), conception (60.5% versus 70.6%, P = 0.1) and pregnancy (38.6% versus 52.2%, P < 0.02). In a field trial, 983 heifers received a once-used CIDR for 7 days, beginning 13 +/- 1 days after TAI. Pregnancy rate to TAI was 55.2%. The median (and mode) of the interval from CIDR removal to estrus was 2.5 days. Estrus, conception and pregnancy rates were 78.2, 70.3 and 55.0% (overall pregnancy rate to TAI and rebreeding, 78.7%). In summary, a once- or twice-used CIDR for 7 days, starting 13 +/- 1 days after TAI resulted in the majority of nonpregnant heifers detected in estrus over a 4-day interval, with acceptable conception rates; however, injecting progesterone at CIDR insertion significantly reduced both estrus and pregnancy rates, and estradiol treatment after CIDR removal was associated with a decreased pregnancy rate to TAI. Zebu cattle are notorious for poor fertility characterized by late maturity and long intercalving intervals attributed to a variety of factors, including genetic, nutritional and climatic. The aim of the present investigation, therefore, was to induce fertile estrus in acyclic pubertal heifers and postpartum anestrous Zebu cows by hormonal intervention. Pubertal Hariana and Sahiwal anestrous heifers (n=51) and postpartum cows (n=55) were either assigned a placebo (controls, N=6 for each breed and parity) or treated with 10-d norgestomet (3 mg) subcutaneous ear implants, with an initial injection of 3 mg, im norgestomet + 5 mg estradiol valerate, followed by 500 IU eCG at implant withdrawal (NOR-treated groups). Jugular venous plasma samples were obtained from a total of 28 animals (controls : 4 heifers and 4 cows; NOR-treated : 12 heifers and 8 cows) on Days 0 (implant insertion), 3, 7, 9 and Day 10 (implant withdrawal), every 12 h on Days 11 and 12, and then once daily on Days 17, 24 and 31. All the samples were assayed for progesterone. Almost all (97%) heifers and 81% cows were induced to estrus, the majority (92% heifers and 79% cows) within 120 h of implant removal. Synchrony of the induced estrus was better in cows, but interval to estrus and estrus duration were significantly longer in heifers (P<0.05). Post-treatment fertility, based on Day 28 nonretum rate, first service, and overall conception rates, was better in heifers (78.9, 60.5 and 73.7%, respectively) than cows (77.1, 48.6 and 62.9%, respectively), but the differences were significant only for the overall pregnancy rate (71.8% for heifers and 51.2% for cows; P<0.05). Low pre-treatment plasma progesterone values (<0.5ng/mL) were consistent with ovarian inactivity, confirming the true anestrus status of experimental animals. Controls failed to exhibit estrus and maintained low progesterone concentrations throughout the study. In treated animals, high progesterone values from Day 17 onwards suggested ovulatory estrus. These early luteal phase progesterone concentrations in nonpregnant (P=0.06) and nonpregnant, nonretum (P<0.05) animals were low in comparison with those of pregnant animals. Good fertility resulting from breeding according to estrus, inspite of variable intervals to estrus and estrus duration, advocates its advantage over fixed-time insemination in norgestomet-treated anestrous Zebu cattle.  相似文献

Two experiments were conducted to determine the conception rates of heifers time-inseminated following melengestrol acetate/prostaglandin F(2alpha) (MGA/PG) estrous synchronization treatment. In Experiment 1, timed insemination of heifers at 72 h after the PG injection, without regard for behavioral estrus, tended to improve (P < 0.15) the percentage of heifers pregnant to artificial insemination (AI) compared with that of synchronized heifers bred 12 h after they were first detected in estrus. In the timed-insemination treatment, heifers exhibiting behavioral estrus 48 to 72 h after PG tended to have an increased (P < 0.15) conception rate to AI compared with heifers exhibiting estrus within 48 h of PG administration. In Experiment 2, the number of heifers conceiving to AI following the MGA/PG estrous synchronization regimen was increased by mass insemination of all heifers not exhibiting estrus by 72 h after PG. The pregnancy rate to AI was higher in heifers with serum progesterone (P(4)) concentrations higher than 1 ng/ml compared with that of heifers with concentrations lower than 1 ng/ml. Of heifers with serum P(4) greater than 1 ng/ml, the pregnancy rate to AI tended to be higher when concentrations exceeded 2 ng/ml than when concentrations were 1 to 2 ng/ml. In cyclic heifers, timed insemination can increase the percentage of heifers pregnant after being synchronized with MGA/PG.  相似文献

Three hundred and ten yearling heifers of various breeds were used in five trials to compare two estrus synchronization treatments. Treatment 1 consisted of Melengestrol Acetate-Prostaglandin F(2)alpha (MGA-PGF(2)alpha). Heifers were fed 0.5 mg MGA/head/d for 14 to 16 d. Sixteen or 17 d after the final MGA feeding, heifers were injected i.m. with 25 mg PGF(2)alpha. Treatment 2 consisted of Syncro-Mate B (SMB). Heifers were given a 9-d norgestomet implant plus an injection containing 3 mg norgestomet and 5 mg estradiol valerate i.m. at implant insertion. Heifers were observed for estrus at 6-h intervals for 120 h after the end of treatments and were artificially inseminated 12 to 18 h after observed estrus. Heifers synchronized with MGA-PGF(2)alpha and SMB had a similar (P > 0.10) estrous response (83.4 vs 90.2%) and a similar (P > 0.10) degree of synchrony (71.8 vs 79.0%) following treatment. However, the synchronized conception rate (68.7 vs 40.6%) and the synchronized pregnancy rate (57.3 vs 36.6%) were higher (P < 0.01) in MGA-PGF(2)alpha than SMB heifers. Breeding season pregnancy rates were similar in both treatment groups. Heifers in both groups that were classified as cycling prior to initiation of treatment had improved reproductive performance following synchronization compared with those classified as noncycling. Based on higher synchronized conception and pregnancy rates and lower labor requirements and drug costs, the MGA-PGF(2)alpha system appears to be a better method to synchronize estrus in beef heifers than the SMB system.  相似文献

A 2 x 3 factorial experiment was conducted to determine the effects of an implant (trenbolene acetate/estradiol or no implant) and method of estrus suppression (immunization against GnRH, melengestrol acetate, or no suppression) on growth performance and carcass characteristics of heifers fed for slaughter. At the start of a 21-d feed adaption phase, crossbred beef heifers (n = 144, 390+/-2.8 kg) were given their first dose of an anti-GnRH vaccine or started on melengestrol acetate (MGA). Thereafter, heifers were fed a high-concentrate diet (78% barley grain) for 84 d (Days 0 to 83), received implants on Day 0, a second vaccination on Day 21, and were slaughtered on Days 84 or 85. Implanting increased average daily gain (1.72 vs 1.50 kg/d, P < 0.01), feed efficiency (6.02 vs 6.75 kg dry matter intake/kg gain, P < 0.01), preslaughter weight (532 vs 513 kg, P < 0.01), carcass weight (301 vs 289 kg, P < 0.01), and ribeye area (88.6 vs 85.9 cm2, P < 0.05), but had no affect (P > 0.05) on dry matter intake, grade fat thickness, marbling score, or lean yield. Compared to heifers fed MGA, those immunized against GnRH had a greater ribeye area (90.0 vs 84.6 cm2) and lean yield (63 vs 61%), and had thinner grade fat (7.5 vs 8.6 mm; P < 0.05 for each). Furthermore, immunized heifers had lower (P < 0.001) plasma progesterone concentrations than control heifers on Days 42, 63 and 83. Heifers fed MGA had less estrus mounting activity (P < 0.05) and lower plasma progesterone concentrations (P < 0.001) than the remaining heifers. Method of estrus suppression did not affect (P > 0.05) preslaughter weight, average daily gain, dry matter intake, feed efficiency, carcass weight, or marbling score. In conclusion, implanting significantly increased growth performance and preslaughter and carcass weights. Two progestin-based protocols for estrus synchronization in replacement beef heifers were compared on the basis of estrous response, interval to and synchrony of estrus, and pregnancy rate. The objective was to determine, whether addition of GnRH to a melengestrol acetate (MGA)-prostaglandin F2alpha (PGF2alpha) estrus synchronization protocol would improve synchrony of estrus without compromising fertility in yearling beef heifers. Heifers at two locations (Location 1, n = 60 and Location 2, n = 64) were assigned randomly to one of two treatments by breed and pubertal status. Heifers were defined as, pubertal when concentrations of progesterone in serum were elevated (> or = 1 ng/mL) in either one of two samples obtained 10 and 1 day prior to treatment initiation. Prior to MGA administration, 18/60 (30%) and 36/64 (56%) of the heifers at Locations 1 and 2, respectively, were pubertal. Heifers in both treatments were fed MGA (0.5 mg/head/day in 1.8 kg/head/day supplement) for 14 days followed by 25 mg of PGF2alpha i.m. (MGA-PGF2alpha) 19 days after MGA withdrawal (Day 33 of treatment). One-half of the heifers at each location received 100 microg of GnRH i.m. 12 days after MGA withdrawal (Day 26 of treatment; MGA Select). The control group received only MGA-PGF2alpha. Heifers were observed for signs of behavioral estrus continuously during daylight hours for 7 days beginning on the day PGF2alpha was administered. Heifers were inseminated 12 h after observed estrus. There was a treatment by location by pubertal status interaction (P < 0.05) for interval to estrus. Compared to the respective control treatment at each location, prepubertal heifers assigned to the MGA Select protocol at Location 1 had longer intervals to estrus, whereas at Location 2, prepubertal heifers assigned to the MGA-PGF2alpha protocol had longer intervals to estrus. The higher number of pubertal heifers at Location 2 was associated with a reduced variance in the interval to estrus among MGA Select treated heifers. Total estrous response and synchronized conception rates were similar between treatments at both locations. These data suggest that addition of GnRH to the MGA-PGF2alpha protocol may improve synchrony of estrus, however, the degree of synchrony may be influenced by pubertal status of heifers at the time treatments are imposed. Further studies are needed to define production systems in which the MGA Select protocol is warranted for use in beef heifers.  相似文献

The use of exogenous progestagens for estrus synchronization in cattle can result in a persistent dominant follicle which is associated with reduced fertility. We examined whether the LHRH agonist, deslorelin, would prevent the formation of a persistent follicle in heifers synchronized with norgestomet. The estrous cycles of heifers were synchronized with cloprostenol, and on Day 7 of the ensuing cycle the heifers received one of the following treatments for 10 d: Group C (n = 5), untreated control; Group N (n = 6), injection of a luteolytic dose of cloprostenol on Days 7 and 8 and implant of norgestomet from Day 7 to Day 17 (i.e. typical 10-day norgestomet implant period); Group D (n = 6), injection of cloprostenol on Days 7 and 8 and implants of deslorelin from Day 7 to Day 17; Group ND (n = 6), injections of cloprostenol and both norgestomet and deslorelin implants as above. Follicle growth was monitored using ultrasonography. Group-N heifers showed continued follicle growth and had larger follicles on Day 17 of the cycle than Group-C heifers (16.8 +/- 1.6 and 10.4 +/- 1.6 mm). Follicle growth for Group-D and ND heifers was similar and variable, and seemed to depend on follicle status at the initiation of treatment. Heifers with follicles of 5 to 10 mm (n = 9) in diameter either showed no follicle growth (2 9 ) or developed large follicles (7 9 ), while heifers with follicles approximately 12 mm (n = 3) in diameter showed follicle atresia with no further significant growth. On Day 17, size of the largest follicle was similar for Group-ND (14.3 +/- 2.9) and Group-D (16.8 +/- 1.6) heifers. Heifers in Group N showed estrous behavior 1.8 +/- 0.2 d after treatment, whereas heifers in Groups D and ND did not show estrus for 2 to 4 wk. The results show that combined treatment with progestagen and an LHRH agonist does not consistently prevent the development of a persistent dominant follicle and that return to estrus can be delayed after treatment with an LHRH agonist.  相似文献

Our objective was to determine whether extending the interval from 17 to 19 d between removal of melengestrol acetate (MGA) feed and administration of PGF2 alpha would alter conception rates, pregnancy rates and the degree of synchrony in replacement beef heifers. A commercial heifer operation in north-central Kansas purchased 591 Angus x Hereford heifers from 12 sources. Prior to the spring breeding season, 14% of the heifers were culled. The remaining heifers were assigned randomly to 2 MGA-PGF2 alpha synchronization systems. All heifers were fed MGA (0.5 mg/head/d) for 14 d, and PGF2 alpha was administered either 17 or 19 d after the completion of MGA feeding. Heifers were inseminated artificially for 30 d followed by 30 d of natural mating. Based on each source, first-service conception rates ranged from 66 to 90%, whereas overall pregnancy rates ranged from 91 to 100%. Heifers given PGF2 alpha on Day 17 after MGA had first-service conception rates of 75.9% compared with 81.4% for heifers receiving PGF2 alpha on Day 19. In response to the PGF2 alpha injection, 99% of the Day 19 heifers that were detected in estrus were inseminated artificially by 72 h after the PGF2 alpha injection, whereas 74% of the heifers in the Day 17 treatment were inseminated by that time. Average interval to artificial insemination (AI) after PGF2 alpha was greater (P < 0.01) for the Day 17 heifers (73.1 +/- 1.1 h) than for the Day 19 heifers (56.2 +/- 1.1 h). No differences in conception rates or overall pregnancy rates occurred; however, heifers receiving PGF2 alpha on Day 19 after MGA had shorter intervals to estrus, and a greater proportion was inseminated within 72 h after PGF2 alpha, thus possibly facilitating successful timed insemination of the remaining heifers not yet inseminated by that time.  相似文献

Four experiments were conducted to investigate modifications to gonadotropin releasing hormone (GnRH)-based fixed-time Al protocols in beef cattle. In Experiment 1, the effect of reducing the interval from GnRH treatment to prostaglandin (PGF) was examined. Lactating beef cows (n = 111) were given 100 mg gonadorelin (GnRH) on Day 0 (start of treatment) and either 500 microg cloprostenol (PGF) on Day 6 with Al and 100 microg GnRH 60 h later, or PGF on Day 7 with Al and GnRH 48 h later (6- or 7-day Co-Synch regimens). Pregnancy rates were 32/61 (53.3%) versus 26/50 (52.0%), respectively (P = 0.96). In Experiment 2. cattle (n = 196) were synchronized with a 7-day Co-Synch regimen and received either no further treatment or a CIDR-B device (Days 0-7). Pregnancy rates were 32/71 (45.1%) versus 33/77 (42.9%) in cows (P < 0.8), and 9/23 (39.1 %) versus 17/25 (68.0%) in heifers (P < 0.05). In Experiment 3, 49 beef heifers were randomly assigned to receive 12.5 mg pLH on Day 0, PGF on Day 7 and 12.5 mg of pLH on Day 9 with Al 12 h later (pLH Ovsynch), or similar treatment plus a CIDR-B device from Days 0 to 7 (pLH Ovsynch + CIDR-B), or 1 mg estradiol benzoate (EB) and 100 mg progesterone on Day 0, a CIDR-B device from Days 0 to 7 (EB/ P4 + CIDR-B), PGF on Day 7 (at the time of CIDR-B removal) and 1 mg i.m. EB on Day 8 with AI on Day 9 (52 h after PGF). Pregnancy rate in the EB/P4 + CIDR-B group (75.0%) was higher (P < 0.04) than in the pLH Ovsynch group (37.5%): the pLH Ovsynch + CIDR-B group was intermediate (64.7%). In Experiment 4, 266 non-lactating cows were allocated to a 7-day Co-Synch protocol (Co-Synch), a 7-day Co-Synch plus 0.6 mg per head per day melengestrol acetate (MGA) from Days 0 to 6 inclusive (Co-Synch + MGA) or MGA (Days 0-6) plus 2 mg EB and 50 mg progesterone on Day 0. 500 microg PGF on Day 7, 1 mg EB on Day 8 and fixed-time Al 28 h later (EB/ P4 + MGA). Pregnancy rates (P < 0.25) were 44.8% (39/87: Co-Synch), 47.8% (43/90; Co-Synch + MGA), and 60.7% (54/89: EB/P4 + MGA). In conclusion, a 6- or 7-day interval from GnRH to PGF in a Co-Synch regimen resulted in similar pregnancy rates in cows. The addition of a progestin to a Co-Synch or Ovsynch regimen significantly improved pregnancy rates in heifers but not in cows. Progestin-based regimens that included EB consistently resulted in high pregnancy rates to fixed-time Al.  相似文献

Dairy heifers were used to compared the effects of two timed AI + controlled internal drug release (CIDR) protocols (5-day vs. a modified 7-day) on: (1) luteal regression to initiate a new ovarian follicular wave; (2) ovarian response to the initial GnRH injection; and (3) pregnancy outcomes. Holstein heifers (N = 543) were assigned randomly to two treatments: (1) 25 mg PGF (im) and a CIDR insert on Day −7 followed by 100 μg of GnRH (GnRH-1) on Day −5 and 25 mg PGF (im) at CIDR insert removal (7-day [7D]) on Day 0; or (2) 100 μg GnRH (GnRH-1) and insertion of a CIDR on Day −5 and 25 mg PGF (im) at CIDR removal (5-day [5D]) on Day 0. Insemination with frozen-thawed conventional or gender-biased semen occurred after detected estrus from Days 0 to 2 or by appointment at 72 h after PGF when a second 100-μg dose of GnRH was given. Blood was collected on Days −7, −5, 0, and 3 to determine concentrations of progesterone and incidence of luteolysis. Ovaries were scanned on Days −5 and 0. Luteolysis in the 7D treatment by 48 h after the initial PGF was greater (P < 0.01) than what occurred spontaneously in the 5D treatment (36.2% vs. 19.7%, respectively). Incidence of ovulation after GnRH-1 on Day −5 was greater (P < 0.05) for 7D than for 5D heifers, but the proportion of heifers with an induced CL on Day 0 did not differ between treatments. Heifers inseminated after detected estrus (166/543, 30.6%) on Days 0, 1, and 2 had greater (P < 0.05) pregnancy per AI (P/AI) at 32 days post AI than after timed AI (38.2% vs. 28.3%) on Day 3. Pregnancy P/AI, however, was greater (P < 0.05) for 7D heifers inseminated at estrus (46.5%) than for 7D heifers receiving the timed AI (26.8%) and differed (P < 0.05) from all 5D heifers regardless of insemination time at estrus (30.5%) or at timed AI at 72 h (29.9%). At the Florida location in which conventional and sexed semen were used during two breeding clusters, P/AI using sexed semen (43.9%, N = 56) did not differ from that of conventional semen (21.2%, N = 50). Remaining replicates of sexed semen produced similar P/AI at the other two locations (sexed = 27.6%, N = 71; and sexed = 31.9%, N = 215). We concluded that the modified 7-day CO-Synch + CIDR program produced more P/AI in heifers inseminated at estrus than a standard 5-day CO-Synch + CIDR program, but when timed AI occurred at 72 h after PGF and CIDR insert removal, P/AI did not differ between programs.  相似文献

Yearling Bos indicus × Bos taurus heifers (n = 410) from three locations, were synchronized with either the Select Synch/CIDR+timed-AI (SSC+TAI) or 7-11+timed-AI (7-11+TAI) treatments. On Day 0 of the experiment, within each location, heifers were equally distributed to treatments by reproductive tract score (RTS; Scale 1-5: 1 = immature, 5 = estrous cycling) and body condition score. The 7-11+TAI treatment consisted of melengestrol acetate (0.5 mg/head/d) from Days 0 to 7, with PGF (25 mg im) on Day 7, GnRH (100 μg im) on Day 11, and PGF (25 mg im) on Day 18. The SSC+TAI heifers received the same carrier supplement (without MGA) from Days 0 to 7, and on Day 11 they were given 100 μg GnRH and an intravaginal CIDR (containing 1.38 g progesterone). The CIDR were removed on Day 18, concurrent with 25 mg PGF im For both treatments, estrus was visually detected for 1 h twice daily (0700 and 1600 h) for 72 h after PGF, with AI done 6 to 12 h after a detected estrus. Non-responders were timed-AI and received GnRH (100 μg im) 72 to 76 h post PGF. The 7-11+TAI heifers had a greater (P < 0.05) estrous response (55.2 vs 41.9%), conception rate (47.0 vs 31.3%), and synchronized pregnancy rate (33.5 vs 24.8%) compared to SSC+TAI heifers, respectively. Heifers exhibiting estrus at 60 h (61.7%) had a greater (P < 0.05) conception rate compared to heifers that exhibited estrus at ≤ 36 (35.3%), 48 (31.6%), and 72 h (36.2%), which were similar (P > 0.05) to each other. As RTS increased from ≤ 2 to ≥ 3, estrous response, conception rate, synchronized pregnancy rate, and 30 d pregnancy rate all increased (P < 0.05), irrespective of synchronization treatment. In conclusion, the 7-11+TAI treatment yielded greater synchronized pregnancy rates compared to SSC+TAI treatment in yearling Bos indicus × Bos taurus heifers.  相似文献

The effects on estrus and fertility of 3 estrus synchronization protocols were studied in Brahman beef heifers. In Treatment 1 (PGF protocol; n=234), heifers received 7.5 mg, i.m. prostianol on Day 0 and were inseminated after observed estrus until Day 5. Treatment 2 (10-d NOR protocol; n = 220) consisted of norgestomet (NOR; 3 mg, s.c. implant and 3 mg, i.m.) and estradiol valerate (5 mg, i.m.) treatment on Day -10, NOR implant removal and 400 IU, i.m. PMSG on Day 0, and AI after observed estrus through to Day 5. Treatment 3 (14-d NOR+PGF protocol; n = 168) constituted a NOR implant (3 mg, sc) on Day -14, NOR implant removal on Day 0, PGF on Day 16, and AI after observed estrus through to Day 21. All heifers were examined for return to estrus at the next cycle and inseminated after observed estrus. The heifers were then exposed to bulls for at least 21 d. During the period of estrus observation (5 d) after treatment, those heifers treated with the PGF protocol had a lower (P<0.01) rate of estrual response (58%) than heifers treated with the 10-d NOR (87%) or 14-d NOR+PGF (88%) protocol. Heifers treated with the 10-d NOR protocol displayed estrus earlier and had a closer synchrony of estrus than heifers treated with either the PGF or the 14-d NOR+PGF protocol. Heifers treated with the 14-d NOR+PGF protocol had higher (P<0.05) conception and calving rates (51 and 46%) to AI at the induced estrus than heifers treated with the PGF (45 and 27%) or the 10-d NOR (38 and 33%) protocol. Calving rate to 2 rounds of AI was greater (P<0.05) for heifers treated with the 14-d NOR-PGF (50%) protocol than heifers treated with the 10-d NOR (38%) but not the PGF (43%) protocol. Breeding season calving rates were similar among the 3 protocols. The results show that the 14-d NOR+PGF estrus synchronization protocol induced a high incidence of estrus with comparatively high fertility in Brahman heifers.  相似文献

Two trials were conducted to examine the effects of estrus synchronization scheme, gonadotropin injection protocol and presence of a large ovarian follicle on response to superstimulation of follicular development and the ensuing superovulation. Estrus was synchronized with either a progestin compound (MGA) or by the use of a luteolytic agent (PGF). Superstimulation was induced with 280 mg equivalents of pFSH administered either by a single subcutaneous injection or by a series of 8 intramuscular injections over 4 d. Follicular development was followed for 5 d with real-time ultrasound, and the heifers were retrospectively classified as to the presence or absence of a large follicle (> or = 8 mm; morphologically dominant follicle) at the start of superstimulation. The 2 trials differed by season of the year and genetic origin of the heifers. In Trial I (20 heifers), the ovulation rate was influenced by the 3-way interaction of the synchronization scheme, injection protocol and morphologically dominant follicle (P = 0.05). The number of large follicles on Day 5 (Day 0 = day of start of superstimulation) and ovarian score (scale 1 to 5 based on extent of follicular development; 1 = least, 5 = most) on Day 5 were significantly correlated (P < 0.05) with ovulation rate. In Trial II (20 heifers), the ovulation rate, number of embryos recovered, number of transferable embryos and ovarian weights were all greater (P < 0.05 to P < 0.01) with the 8-injection protocol than the 1-injection protocol. The number of medium follicles (5 to 7 mm) on Days 2 and 3, number of large follicles (> or = 8 mm) on Days 3, 4 and 5 and ovarian scores on Days 4 and 5 were all significantly correlated (P < 0.05) with ovulation rate. In both trials, differences in follicle populations were not seen until Day 3 of the superstimulation procedure. Collectively, these trials do not provide strong support for a single injection of FSH, as used here, nor does it indicate a clear advantage for either MGA or PGF as a means of enhancing the ovulation rate or embryo quality.  相似文献

Two experiments evaluated a modified delivery of prostaglandin F2alpha (PGF2alpha) after a melengestrol acetate (MGA) treatment in Angus and Bos indicus x Bos taurus (BI) heifers. Experiment 1 was replicated three times with yearling BI heifers (n = 695). Heifers received MGA (0.5 mg head(-1) day(-1)) for 14 days. In Replications 1 and 2, heifers received either 25 mg of PGF2alpha im 19 days after MGA (single) or 12.5 mg of PGF2alpha im 19 and 20 days after MGA (split). In Replication 3, heifers received the same treatments, with PGF2alpha initiated either 18 or 19 days after MGA. Estrus was detected for 72 h after PGF2alpha, with AI commencing 8-12 h after a detected estrus. Heifers not observed in estrus by 72 h were timed-AI concomitant with GnRH (100 microg im). Heifers from Replication 2 (n = 146) had blood samples collected at the initial PGF2alpha and at timed-AI to determine corpus luteum (CL) regression by evaluating plasma progesterone concentrations. The interval from MGA withdrawal to PGF2alpha did not have a significant effect on any variable in Replication 3 and there were no treatment by replication effects for any variables, therefore data were pooled. Modifying the PGF2alpha treatment from a single treatment to two treatments on consecutive days increased (P < 0.05) 72 h estrous response (43.2% versus 50.1%), timed-AI (23.9% versus 33.5%) and total-AI pregnancy rates (34.5% versus 42.5%), and CL regression (79.1% versus 92.5%), respectively. In Experiment 2, yearling Angus (n = 66) and 2-year-old BI (n = 68) heifers were synchronized as per Experiment 1 (with the initial PGF2alpha 19 days after MGA). Neither breed nor PGF2alpha treatment effected (P > 0.05) 72 h estrous response, total-AI pregnancy rate, or CL regression rate. In conclusion, treating yearling BI heifers with split treatments of PGF2alpha (given on two consecutive days) improved estrous response and pregnancy rates by increasing PGF2alpha-induced luteolysis.  相似文献

This study was conducted to determine the efficacy of feeding melengestrol acetate (MGA) for 14 days and administering prostaglandin F(2)alpha (PGF) 17 days after MGA to synchronize or induce estrus in yearling beef heifers. The study involved 56 Angus (n = 19), Hereford (n = 15) and Simmental (n = 22) heifers that were assigned by breed and pubertal status to either MGA+PGF or to control groups. Heifers in the synchronized group were fed 0.5 mg MGA per head per day for 14 days from a grain carrier and were injected with 25 mg, i.m. PGF 17 days after the last daily feeding of MGA. Control heifers were fed from a grain carrier without MGA and were not treated with PGF. Heifers were classified as pubertal when concentrations of progesterono in the serum exceeded 1 ng/ml in 1 of 2 samples collected prior to the initiation of treatments. Blood samples were collected 7 days before and on the day that treatment with MGA or carrier began and 7 days before and on the day that PGF was administered. Progesterone concentrations in the serum were elevated ( > 1 ng/ml) in 61% (17 28 ) of the MGA+PGF-treated heifers and in 61% (17 28 ) of the control heifers prior to feeding MGA. However, concentrations of progesterone in the serum at the time PGF was administered differed (P<0.05) between MGA+PGF and control groups. Concentrations of progesterone in the serum exceeded 1 ng/ml in 100% (28 28 ) of the MGA+PGF-treated heifers and in 71% (20 28 ) of control heifers at the time PGF was administered (P<0.05). All heifers were inseminated 12 hours after the first detected estrus. Twenty-two of 28 (79%) of the MGA+PGF-treated heifers exhibited estrus within 6 days after PGF compared with 9 of 28 (32%) of control heifers (P<0.05). The conception rate at first service did not differ between MGA+PGF and control groups (64% and 67%, respectively). Synchronized pregnancy rates were higher (P<0.05) for MGA+PGF-treated heifers than for control heifers (14 28 , 50% vs 6 28 , 21%). Increased concentrations of progesterone in serum at the time PGF was administered and higher pregnancy rates during the synchronized period among MGA+PGF-treated heifers demonstrate the efficacy of this treatment for use in estrus synchronization. Moreover, this treatment may have a potential effect on inducing puberty in breeding age heifers.  相似文献

This study was carried out to evaluate the luteotrophic influence of early (before Day 7 as well as after Day 7; Day 0=estrus) bovine embryos and the relationship between plasma progesterone (P4) concentrations and embryo survival. Virgin Holstein dairy heifers (n=325) from a single herd were randomly allocated to be nonbred, bred by artificial insemination (AI) or by embryo transfer (ET). Bred heifers were either treated with 1500 IU human chorionic gonadotrophin (hCG) on Day 7 of the estrous cycle or received no hCG treatment. Plasma P4 concentrations on Days 0, 5, 7, 10, 13, 15, 17, 19 and 21 were similar in pregnant AI- and ET-bred heifers and, this was observed in both hCG-treated and untreated females. Nonbred, AI- and ET-bred nonpregnant heifers (both hCG-treated and untreated) presented similar plasma P4 concentrations. Plasma P4 concentrations of pregnant heifers significantly deviated from those of nonpregnant and nonbred heifers on Day 17. In hCG-treated heifers, plasma P4 concentrations and Day 28 pregnancy rate were significantly higher in females with an induced accessory corpus luteum (CL) than in those females without an induced accessory CL. Treatment with hCG, although inducing the formation of accessory CL and significantly increasing plasma P4 concentrations had no significant effect on Day 28 pregnancy rate. In conclusion, this study does not support the existence of any peripherally detectable luteotrophic influence from early embryos (Days 5-7). Plasma P4 was only significantly related to embryo survival on Day 17, the time of expected onset of luteolysis.  相似文献

Growth and ovarian function of crossbred beef heifers grazing low and high endophyte-infected tall fescue pastures were studied for 2 successive years. In April of each year, 20 weanling and 20 yearling heifers were included in the study. All heifers were weighed at 28-d intervals for 112 d. Blood samples were collected from each heifer on Day 0 and +7 of each of five 28-d periods and analyzed for progesterone concentration. Heifers with progesterone concentrations >/= 1.5 ng/ml on either or both Day 0 and +7 were classified as having normal cyclic ovaries. High endophyte-infected fescue pastures adversely altered the ovarian activity (P < 0.05) of weanling heifers in both years. In each trial, average weight gains were lower (P < 0.05) in yearling and weanling heifers grazing the high endophyte-infected pastures than in heifers grazing low endophyte-infected pastures. In 1992, heifers were synchronized with PGF(2alpha) administered on Days 101 and 112. Blood samples were collected on 0, 4, 8 and 12 d after the second PGF(2alpha) injection for progesterone analysis. Heifers grazing high and low endophyte-infected pastures were pastured separately with 4 bulls each and were given heatmount detectors. At 96 h, less estrus activity was observed (P < 0.10; power=0.63) in weanling heifers grazing the high vs. low endophyte pastures although pregnancy rates were similar for all groups. Progesterone concentrations suggested that weanling heifers on the high endophyte pastures had a higher incidence of luteal dysfunction after PGF(2alpha) synchronization. In summary, high endophyte-infected pastures decreased growth in both weanling and yearling heifers, ovarian activity and luteal function were adversely altered in weanling heifers with subsequent decreased estrus response to estrus synchronization.  相似文献

A study was conducted to determine whether concurrent administration of vaccines for reproductive diseases and an anthelmintic, given immediately prior to estrus synchronization with a progestogen, would affect subsequent estrous response and fertility in beef heifers. Two hundred ninety-five yearling crossbred beef heifers on 3 ranches in Oregon and Kansas were assigned to 1 of 2 treatments. Control heifers were vaccinated and dewormed 30 d prior to the anticipated breeding. Treated heifers received the same vaccination and deworming products 9 d prior to breeding. All heifers in both groups were treated with norgestomet (Syncro-Mate-B) for 9 d to synchronize estrus. The administered vaccine consisted of modified live virus for infectious bovine rhinotracheitis, parainfluenza and bovine viral diarrhea, with 5-way bacterin for leptospirosis and bacterin for vibriosis. Heifers were also administered ivermectin or fenbendazole for deworming. Heifers exhibiting synchronized estrus were artificially inseminated and subsequently exposed to fertile bulls. Pregnancy status was determined at a later date by palpation per rectum. Vaccination and deworming at the time of treatment with progestogen compared with immunizing and deworming 30 d prior to breeding did not impair the expression of behavioral estrus. The percentage of treated heifers conceiving to AI or natural service did not differ from that of the respective control heifers. These data suggest that vaccination, deworming and synchronization of estrus with norgestomet can be performed simultaneously without any detrimental effects upon occurrence of behavioral estrus or fertility.  相似文献
