Experiments were conducted to measure optical backscatter and beam transmission of suspensions of 180, 150 and 90 μm sand, and 40 μm clay, in a recirculation tank designed to house an optical backscatterance sensor (O.B.S.) and a beam transmissometer. Particle size was determined using gravimetric techniques and Coulter counter. By contriving known sediment distributions from the fractionated sediment samples, it was found that both the O.B.S. and beam transmissometer responded approximately linearly to narrow band and broad band particle suspensions. The beam transmissometer showed greater sensitivity to the fine-grain fraction of a poly-disperse suspension than the O.B.S.  相似文献

The purpose of this research was to develop liposomal dry powder aerosols for protein delivery. The delivery of stable protein formulations is essential for protein subunit vaccine delivery, which requires local delivery to macrophages in the lungs. β-Glucuronidase (GUS) was used as a model protein to evaluate dry powder liposomes as inhaled delivery vehicles. Dimyristoyl phosphatylcholine:cholesterol (7∶3) was selected as the liposome composition. The lyophilization of liposomes, micronization of the powders, aerosolization using a dry powder inhaler (DPI), and in vitro aerodynamic fine particle fraction upon collection in a twinstage liquid impinger were evaluated. After lyophilization and jet-milling, the total amount of GUS and its activity, representing encapsulation efficiency and stability, were evaluated. The GUS amount and activity were measured and compared with freshly-prepared liposomes in the presence of mannitol, 43% of initial GUS amount, 29% of GUS activity after lyophilization and 36% of GUS amount, 22% of activity after micronization were obtained. Emitted doses from dry powder inhaler were 53%, 58%, 66%, and 73% for liposome powder:mannitol carrier ratios of 1∶0, 1∶4, 1∶9, and 1∶19. Fifteen percent of the liposome particles were less than 6.4 μm in aerodynamic diameter. The results demonstrate that milled liposome powders containing protein molecules can be aerosolized effectively at a fixed flow rate. Influences of different cryoprotectants on lyophilization of protein liposome formulations are reported. The feasibility of using liposomal dry powder aerosols for protein delivery has been demonstrated but further optimization is required in the context of specific therapeutic proteins. Hairy roots and suspended cells of transformedNicotiana tabacum were used to produce full length murine IgG1 monoclonal antibody. The maximum amount of antibody accumulated per g dry weight in the hairy root cultures was 6.5 times that in the suspension cultures. Up to 48% of the antibody in the suspension cultures was found extracellularly, while a maximum of only 17% was recovered from the hairy root medium. The amount of assembled antibody in the root and suspension cultures was significantly reduced by intracellular and/or extracellular antibody degradation soon after the end of the exponential growth phase. Bacitracin, a polypeptide antibiotic, has been shown in previous work to prevent degradation of peptides and hormones in plant and mammalian systems. Treatment of hairy roots and cell suspensions with 100 μg/mL bacitracin was not sufficient to prevent loss of antibody from the cultures, but improved the specific growth rates by up to 53%. At concentrations of 250 μg/mL and above, bacitracin had a toxic effect on hairy roots, which may limit the application of this peptide in plant tissue culture.  相似文献

Methods already worked out for the preparation and testing of aqueous D.D.T. suspensions against Tribolium castaneum , by a dipping method, have been applied to Oryzaephilus surinamensis , giving similar results, i.e. toxicity increases with increase in particle size.
In the same way, the precipitation of rotenone by exchange of solvents leads to the formation of simple aqueous suspensions. The theory of precipitation is described and methods are given of preparing five types of suspension: colloidal rotenone, a suspension of small elongated plates, a suspension containing small hexagonal plates in aggregates, and two suspensions containing hexagonal plate-shaped crystals of different sizes.
These were tested, by dipping, against Oryzaephilus surinamensis L. Within the range of crystal sizes up to about 150μ, toxicity is inversely related to the size of particle in suspension. The variation in median lethal concentration obtained in this way is of the order of 600 times. Crystal shape seems to be unimportant. Similar results were obtained with fine suspensions, using a spraying method.
The variation of mortality with time was also studied, using D.D.T. against Tribolium castaneum and rotenone against Oryzaephilus surinamensis. In the former case, both colloidal and crystalline D.D.T. show progressively increasing kills with the passage of time. Crystalline rotenone behaves similarly, but colloidal rotenone gives an initial paralytic effect, followed by recovery of the insects.  相似文献

Aqueous DDT suspensions containing particles of different sizes gave different temperature coefficients of mortality in dipping tests against adult Oryzaephilus surinamensis and Tribolium castaneum. In the range 12–30°C., colloidal DDT showed a large negative coefficient, but the coefficient for a suspension of DDT crystals of 400μ was smaller ( T. castaneum ) or negligible ( O. surinamensis ). Thus, although a suspension of crystalline DDT was more toxic than colloidal DDT to warm insects, it was less toxic than the colloid to cool insects.
A test was made of rotenone suspensions against adult milkweed bugs ( Oncopeltus fasciatus Dall.) by application of measured droplets under the wings. Colloidal rotenone was more toxic than a crystalline suspension. Milkweed bugs could not be killed in this way by DDT suspensions.
Tests were also made by injecting suspensions of rotenone, DDT and its fluorine analogue (DFDT) into milkweed bugs. A suspension of rotenone crystals was equitoxic with colloidal rotenone against insects kept for 3 days at 27°C. after treatment. The same was true of crystalline and colloidal DDT suspensions. If the bugs were kept at 10°C., colloidal rotenone was much more toxic than crystalline rotenone 2 days after treatment. But if the insects were kept for about 3 weeks and inspected at intervals, the first apparent difference in toxicity grew smaller, and in the end it nearly disappeared. With DDT the time for this to occur was about 10 days; and suspensions of colloidal and crystalline DFDT were equitoxic after only 2 days, even at 10°C.
In the application test, the difference between colloidal and crystalline rotenone was a real difference in toxicity. It did not disappear with the lapse of time; but in the injection tests the two forms of each poison differed only in speed of action, and not in ultimate toxicity.  相似文献

A procedure is outlined for the establishment of a proliferating cell suspension culture and subsequent plant regeneration of the latex-producing plant,Calotropis gigantea (Linn.) R. Br. Friable calluses were obtained by culturing hypocotyl explants on modified Murashige and Skoog medium with 2.69 μM α-naphthaleneacetic acid and 4.44 μM 6-benzyl-aminopurine. Friable calluses were transferred to modified Murashige and Skoog liquid medium containing 500 mg l−1 casein hydrolysate, 5% (v/v) coconut water and 5% (w/v) sucrose to initiate suspension cultures. Suspensions were subcultured every 10–12 days and supplemented with 13.56 μM 2,4-dichlorophenoxyacetic acid (2,4-D). After 3–4 subcultures, suspensions consisted of small, highly cytoplasmic cell clumps and large vacuolate cells. Plating the suspension clumps on medium containing 4.52 μM 2,4-dichlorophenoxyacetic acid and culturing in darkness produced macroscopic calluses, which subsequently produced a high number of shoots when placed in light and supplemented with 2.22 μM 6-benzyl-aminopurine and 0.45 μM 2,4-dichlorophenoxyacetic acid. Shoots were rooted using Bonner's solution containing 0.49 μM indole-3-butyric acid, and the plants successfully transferred to soil. This study examined the use of focused beam reflectance measurement (FBRM) for qualitative and quantitative analysis of pharmaceutical suspensions with particular application to toxicology supply preparations for use in preclinical studies. Aqueous suspensions of ibuprofen were used as prototype formulations. Initial experiments were conducted to examine the effects of operational conditions including FBRM probe angle, probe location, and mixing (method and rate of mixing) on the FBRM analysis. Once experimental conditions were optimized, the homogeneity and sedimentation-redispersion of particles in the suspensions were assessed. Ibuprofen suspension under continuous agitation was monitored using FBRM for 60 h to study particle size change over time. Another study was performed to determine if particle count rates obtained by FBRM could be correlated to suspension concentration. The location and the angle of the FBRM probe relative to the beaker contents, and the rate and the method of mixing the suspension were found to be sensitive parameters during FBRM analysis. FBRM was able to monitor the process of particle sedimentation in the suspension. The attrition of ibuprofen particles was detectable by FBRM during prolonged stirring with an increase in the number of smaller particles and decrease in the number of larger particles. A strong correlation was observed between particle count rate by FBRM and ibuprofen concentration in the suspension. Also, change in content uniformity in the suspension at different locations of the beaker was represented by FBRM particle count. Overall, FBRM has potential to be a useful tool for qualitative and quantitative analysis of pharmaceutical suspensions.  相似文献

The purpose of this research was to investigate the measurement and in vitro delivery implications of multimodal distributions, occurring near or in the respirable range, emitted from pressurized metered-dose inhalers (pMDIs). Particle size distributions of solution pMDIs containing hydrofluoroalkane-134a (HFA-134a) and ethanol were evaluated using 2 complementary particle-sizing methods: laser diffraction (LD) and cascade impaction (CI). Solution pMDIs were formulated from mixtures of HFA-134a (50%–97.5% wt/wt) and ethanol. A range of propellant concentrations was selected for a range of vapor pressures. The fluorescent probe, Rhodamine B, was included for chemical analysis. The complementary nature of LD and CI allowed identification of 2 dominant particle size modes at 1 and 10 μm or greater. Increasing propellant concentrations resulted in increases in the proportion of the size distributions at the 1-μm mode and also reduced the particle size of the larger droplet population. Despite significant spatial differences and time scales of measurement between the particle-sizing techniques, the fine particle fractions obtained from LD and CI were practically identical. This was consistent with LD experiments, which showed that particle sizes did not decrease with increasing measurement distance, and may be explained by the absence of significant evaporation/disintegration of larger droplets. The fine particle fractions (FPFs) emitted from HFA-134a/ethanol solution pMDI can be predicted on the basis of formulation parameters and is independent of measurement technique. These results highlight the importance of presenting particle size distribution data from complementary particle size techniques.  相似文献

Human serum albumin (HSA) nanometer or micron particles represent promising drug-carrier systems. The azimuthal and radial variations of a linear polarization-sensitive backscattering Mueller matrix were experimentally studied in two cases: the scattering particle was smaller or larger in size to the probing wavelength of 780 nm. The results show that the twofold and fourfold structures are characteristic of small particle size suspension, whereas the eightfold structure is characteristic of large particle size suspension. Moreover, for both particle size suspensions, the element patterns have strong radial dependence when the suspension concentration and the incident power of laser change. In addition, for both particle size suspensions, the rotational symmetry of each element is lost in the case of oblique incidence but the multifold structure is maintained. Some suggestions for applications of Mueller matrix imaging in biomedical optics are provided.  相似文献

Growth of two laboratory strains ofChlamydia trachomatis (Gambia 17 and UW 5) was compared in McCoy cell monolayers 1–3 days old and McCoy cells inoculated while in suspension (day 0). Both cell preparations were treated with cytochalasin B. Each chlamydial strain produced more inclusions in the cell suspension preparations than in monolayers. Efforts to inoculate cycloheximide-treated cells in suspension were unsuccessful because of the toxic effect of this chemical. When patient specimens were tested in cell suspensions and in monolayers, all positives were detected comparably in both systems, although bacterial contamination was more pronounced in the cell suspensions. The results indicate that cell suspensions can be used as a convenient and rapid supplement to preformed monolayers for chlamydial culture tests.  相似文献

The purpose of this research was to prepare a dry powder vaccine formulation containing whole inactivated influenza virus (VIIV) and a mucoadhesive compound suitable for nasal delivery. Powders containing WIIV and either lactose or trehalose were produced by lyophilization. A micro-ball mill was used to reduce the lyophilized cake to sizes suitable for nasal delivery. Chitosan flakes were reduced in size using a cryo-milling technique. Milled powders were sieved between 45 and 125 μm aggregate sizes and characterized for particle size and distribution, morphology, and flow properties. Powders were blended in the micro-ball mill without the ball. Lyophilization followed by milling produced irregularly shaped, polydisperse particles with a median primary particle diameter of ≈21 μm and a yield of ≈37% of particles in the 45 to 125 μm particle size range. Flow properties of lactose and trehalose powders after lyophilization followed by milling and sieving were similar. Cryo-milling produced a small yield of particles in the desired size range (<10%). Lyophilization followed by milling and sieving produced particles suitable for nasal delivery with different physicochemical properties as a function of processing conditions and components of the formulation. Further optimization of particle size and morphology is required for these powders to be suitable for clinical evaluation. Published: March 10, 2006  相似文献

The purpose of this study was to evaluate the hypothesis that spacer devices have limited effect on the in vitro fine particle dose emitted from solution metered dose inhalers containing different proportions of HFA134a [1,1,1,2-tetrafluoroethane] propellant. Two solution formulations (80% and 97.5% wt/wt HFA134a) were tested across the actuator alone, actuator plus Aerochamber, and Ace holding chamber. Particle size distributions were determined using laser diffraction (LD) and cascade impaction (CI). Multimodal particle size distributions were identified using LD. CI analyses were characterized by a major mode located at ≈0.5 μm. The fine particle dose emitted from the inhaler spacer combinations containing 97.5% HFA134a was independent of the device setup used. Fine particle doses were influenced by spacer setup in 80% HFA134a formulations, indicating different plume dynamics of low vapor pressure formulations. Sampling inlet deposition was approximately O when spacer devices were used with either formulation. When spacers were not used, sampling inlet deposition was increased significantly. However, inlet deposition with the 97.5% HFA134a formulation was significantly less than that of the 80% HFA134a formulation (≈25% of emitted dose compared with 69% respectively). Thus, high propellant concentration formulations appear to have more robust in vitro performance. This is particularly important given the preponderance of poor patient compliance that is associated with spacer use. High propellant concentrations had the advantage of fine particle doses that were independent of the device setup and significantly lowered sampling inlet deposition when no spacer was used.  相似文献

The quantitative uptake of suspended particles has been studied in 14 species of ciliated protozoa in terms of the maximum rate of water cleared at low particle concentrations and of the maximum ingestion rate at high particle concentrations. The results, supported by data from the literature, show that ciliates which feed on larger particles (> 1–5m) compare favorably with metazoan suspension feeders with respect to the ability to concentrate dilute suspensions of particles. Species specialized on smaller food particles (0.2–1m), the size range of most bacteria in natural environments, require a higher concentration of particles. Bacterial population densities which can sustain ciliate growth are found in sediments, waters rich in organic material, and in the early successional stages of decomposing organic material. This is not the case in open waters in general where bacterivorous ciliates cannot play a role as grazers of bacteria.  相似文献

A comparison was made of total aflatoxins (AFs) in 43 samples of chilies collected during winter and 42 in summer to determine the effect of season on contamination. The samples were analyzed by HPLC with fluorescence detection. The limits of detection and quantification for AFB1 and AFG1 were 0.05 μg/kg and 0.50 μg/kg, whilst for AFG2 and AFB2 they were 0.10 μg/kg and 0.60 μg/kg. In the winter samples, AFs were detected in 18 (72%) whole and 14 (60%) ground chilies, with concentration ranges 0.00-52.30 μg/kg and 0.00-74.60 μg/kg respectively. In the summer samples, 17 (64%) whole and 12 (76%) ground chilies were contaminated with AFs at concentrations 0.00-61.50 μg/kg and 0.00-95.90 μg/kg respectively. The percentage of samples greater than the European Union statutory limit for AFB1 and total AF for whole chilies were 48 and 36%, compared with ground chili values of 50 and 45%, respectively, in the winter season. In the summer season, the samples greater than the European Union limit for AFB1 and total AF in whole chilies were 52 and 38%, compared with values of 54 and 49% in ground chilies respectively. AF contamination was found to be higher in summer chili samples and hence winter chilies may provide a better quality product with respect to AF contamination. The ability to undertake this analysis in Pakistan will enhance greatly the ability to improve chili production in that country, as described herein.  相似文献

A procedure is outlined for the establishment of a proliferating cell suspension culture of East Indian rosewood (Dalbergia latifolia Roxb.) and efficient plant regeneration from callus derived from such cultures. Callus was induced from hypocotyl segments derived from 1-week-old axenic seedlings on Murashige and Skoog (1962) medium (MS) containing 10.8 μM naphthaleneacetic acid (NAA) and 2.2 μM benzyladenine (BA). Calli were increased by subculturing on MS supplemented with same growth regulators and 10% coconut water (CW). Friable calli were used to initiate cell suspension cultures. Optimum cell proliferation occurred in MS containing 10.8 μM NAA, 2.2 μM BA and 10% CW, using an initial inoculum cell density of 2%. Cell clumps composed of 20–25 cells harvested from suspension cultures at the exponential growth phase readily formed callus within 3 weeks following plating on the semi-solid MS as above. High-frequency shoot-bud differentiation was induced in these calli on MS containing 2.7 μM NAA and 13.3 μM BA. The regeneration frequency declined at higher BA concentrations. The organogenic potential of the cell suspensions was influenced by the age of the culture. Regenerated shoots were rooted on half-strength MS containing 5.7 μM indole-3-acetic acid, 4.9 μM indole-3-butyric acid and 5.3 μM indole-3-propionic acid. A rapid and sensitive assay for measuring myoblast fusion in suspension cultures of embryonic chick pectoral myoblasts is described. Fusion-competent cells are generated by growth in suspension using a low calcium medium. Fusion-promoting levels of calcium are added, and the suspensions incubated for 1–6 h. The cells are then trypsinized to disperse cellular aggregates and sized in a Coulter particle counter. This assay minimizes many of the artifacts inherent in measurements of fusion in monolayer cultures, and is designed for the rapid screening of agents for their effects on fusion.  相似文献

A comparative study was performed of interphase in situ hybridization (ISH) to deparaffinized 4-m tissue sections and nuclear suspensions from eight prostatic adenocarcinomas, as well as one normal prostatic control. Whole nuclear suspensions were derived from the same tumor areas to evaluate differences of ISH to truncated versus whole nuclei. DNA probes specific for the centromeres of chromosome 1, 7, 8, 10, and Y were used for detection of numerical chromosomal changes and aneuploidy. In six adenocarcinomas chromosome aberrations (+7, +8, –8, –10, –Y) were seen. However, ISH to sections revealed focal aberrations (–10, –Y) in four cases that could not be distinguished in the suspensions. Chromosomal alterations occurring in larger tumor areas were also detected in the nuclear suspensions. Chromosome copy number changes, especially gains, were better discriminated in the nuclear suspensions. The rate of ISH aneuploidy seen in nuclear suspensions corresponded with that observed in the tissue sections (P<0.01). Ploidy patterns as assessed by ISH to sections and nuclear suspensions were in concordance with DNA flow cytometry (bothP<0.001). We conclude that both section and suspension ISH were able to accurately detect aneuploidy and numerical chromosomal aberrations occurring in larger histological areas. However, section ISH was also capable of revealing (small) focal cytogenetic abnormalities, due to a precise analysis of only target cells. Focal abnormalities were not detected by suspension ISH, probably due to an admixture of non-aberrant tumor cells and stromal elements.  相似文献

Thymidine and leucine incorporation into macromolecules of soil bacteria extracted by homogenization-centrifugation were measured after size-fractionation of the bacterial suspension through different sized filters (1.0, 0.8, 0.6, 0.4 m). The specific thymidine incorporation rate was highest for the unfiltered and 1.0 m filtered suspensions (approximately 10 × 10–21 mol thymidine bacteria–1 h–1), but decreased to 1.39 × 10–21 mol bacteria–1 h–1 for bacteria passing the 0.4 m filter. The proportion of culturable bacteria (percent colony forming units/acridine orange direct counts) also decreased with bacterial cell size from 5.0% for the unfiltered bacterial suspension to 0.8% in the 0.4 µm filtrate. A strong linear correlation (r 2 = 0.995) was found between the specific thymidine incorporation rate and the proportion of culturable bacteria. Leucine incorporation gave similar results to the thymidine incorporation. No effects of cell size on the degree of isotope dilution or unspecific labeling of other macromolecules were found either for the thymidine or the leucine incorporation technique. These data indicate that small bacteria, although more numerous than larger ones, not only constitute a smaller proportion of the soil bacterial biomass than larger bacteria, but also contribute to a lesser degree to carbon transformations in soil.  相似文献

Plant suspension cultures are highly aggregated, preventing the direct application of flow cytometry for the study of population dynamics. The utility of single cells to accurately represent aggregated suspension cultures was tested through the analysis of total protein content. Specifically, protein content of two Taxus cuspidata suspension culture lines was studied using the Bradford assay for aggregated suspension cultures, and flow cytometry with fluorescein isothiocyanate staining for protoplasts and single cells. Taxus protein levels were measured at 75–160 mg per gram dry weight via the Bradford assay. Aggregated suspension cultures, protoplasts, and single cells predicted the same trend of protein content over the culture period (21 days). Normalized protein content of isolated single cells was statistically equivalent to aggregated suspensions for both cell lines. However, normalized protein content of isolated protoplasts showed significant differences from aggregated suspensions for one of the two cell lines. Elicitation with methyl jasmonate (MJ) is commonly utilized to increase paclitaxel accumulation in suspension cultures, and therefore the effect of MJ elicitation on protein content in aggregated suspensions, isolated single cells and protoplasts was assessed. Aggregated suspension cultures, protoplasts, and single cells did not show any change in total protein content following elicitation with MJ at 200 M on day 7. This study illustrates the usefulness of flow cytometry for obtaining culture population information and the value of using intact single cells for the study of plant metabolism.  相似文献
